Abstract:Elaborating the environmental behavior of mefentrifluconazole, a novel triazole fungicide, in stereoselective level is of paramount importance for the application of the pesticide in agriculture. In this study, the enantioselective bioactivity, acute toxicity and stereoselective bioaccumulation of mefentrifluconazole in earthworm (Eisertia fetida) were investigated. Bioactivity tests against four pathogens revealed that R-( - )-mefentrifluconazole exhibited approximately 11-113 times higher bioactivity than its S-( + )-mefentrifluconazole. However, the LC50 of S-( + )-, rac- and R-( - )mefentrifluconazole to earthworm was measured to be 4.1, 11.4 and 7.3 mu g/cm(2), respectively, indicating active ingredient R-( - )-mefentrifluconazole is less toxic than its racemate and S-form. Accumulation of mefentrifluconazole in earthworms was non-enantioselective and negatively related to its adsorption onto soils. The concentration of mefentrilluconazole in in situ pore water (C-IPW) and CaCl2 extraction (C-CaCl2) was closely related to its accumulation in earthworms, suggesting that C-IPW and C-CaCl2 could be appropriate indicators for estimation of the bioavailability of mefentrilluconazole in soil. Conclusively, our study provides necessary information for the risk assessment of mefentrifluconazole in agriculture.
Influence of Soil Factors on the Stereoselective Fate of a Novel Chiral Insecticide, Paichongding, in Flooded Paddy Soils.

Abstract:In this study, the fate of paichongding was investigated in three soils with contrasting soil properties. In general, low soil pH has the potential to retard the mineralization and promote the dissipation of paichongding and the formation of its primary transformation product and to accelerate the formation of bound residue. The dissipation of paichongding stereoisomers was very fast and diastereoselective. This selectivity was found only between diastereomers and not between enantiomers and was observed to be soil dependent. In the acidic soil, the enantiomers (5R,7R)- and (5S,7S)-paichongding were degraded more quickly than (5R,7S)- and (5S,7R)-paichongding, whereas a contrary trend was observed in the neutral soil, and such selectivity did not occur in the alkaline soil. The OM and clay contents also played important roles in the fate of paichongding. This effect of soil properties should be considered in risk assessment of chiral pesticides and their application in the field.
Nonstereoselective Behavior of Novel Chiral Organophosphorus Pesticide Dufulin in Cherry Radish by Different Absorption Methods

Abstract:Dufulin is a biologically derived antiviral agent chemically synthesized by α-phosphoramidate in sheep and is effective against viral diseases in plants such as tobacco, rice, cucumber and tomato. However, the environmental behaviors and fate of Dufulin under different cultivation systems remain unknown. This study investigates the absorption, translocation and accumulation of 14C-Dufulin stereoisomers introduced by pesticide leaf daubing and by mixing the pesticide with soil in different tissues of cherry radish. We particularly focused on whether the behaviors of Dufulin enantiomers in plants were stereoselective. In the leaf uptake experiments, S-Dufulin and R-Dufulin were transported both up and down, while more than 93% of the pesticide remained in the labeled leaves. During the radicular absorption experiments, both enantiomers of Dufulin were taken up by radish roots and moved to the upper part of the plant, while less than 0.2% Dufulin was absorbed from the soil. Hence, it was easier for Dufulin to enter plants through the leaf surface than through the roots. However, we found in this trial that the stereoisomers of Dufulin underwent nonstereoselective absorption and translocation, which implies that rac-Dufulin and its metabolites should be a major research priority. Overall, our results provide a relatively accurate prediction of the risk assessment of Dufulin, which will help guide its rational use in the environment as well as ensure eco-environmental safety and human health.
Deposition and Dissipation of Difenoconazole in Pepper and Soil and Its Reduced Application to Control Pepper Anthracnose

Abstract:The initial deposition amount, dissipation dynamics, retention rate, and field control efficacy of difenoconazole in pepper-soil system were studied with different application dosages, planting regions and patterns. The initial deposition amount of difenoconazole under the same application dosage showed the following order: fruits < cultivated soils < lower stems < upper stems < lower leaves < upper leaves, open field < greenhouse, and Changjiang < Cixi < Hefei < Langfang, respectively, which increased with increasing application dosage. The dissipation rates in leaves, stems, fruits and cultivated soils exhibited an initially fast and then slow trend, while the retention rates displayed a tendency of first increasing and then stabilizing with increasing application dosages. After 7 d of difenoconazole application, the retention rates at five concentrations were 10.3%-39.1%, and the field efficacy mostly reached the minimum effective dose. These results suggested that difenoconazole could be reduced by 25% based on the minimum recommended dose meeting the requirements of field control efficacy for controlling pepper anthracnose.

Abstract:The deposition, dissipation, and metabolism of difenoconazole as well as reduced application dose were studied in the open field and greenhouse pepper system. The deposition amount of difenoconazole gradually increased with the increasing dosages, which followed an order with upper leaf > lower leaf > upper stem > lower stem > cultivated soil > fruit. The dissipation half-lives of difenoconazole in different parts of pepper plant and cultivated soil were higher in the greenhouse than those in the open field. Difenoconazole was transformed to metabolite I (C16H13Cl2N3O2) in pepper plant, but metabolite II (C13H14ClN3O3) in cultivated soil. Additionally, the field control efficacy of difenoconazole at the 0.75-fold minimum recommended dosage could meet the basic control requirement after the first application for 7 d. The results revealed deposition distribution and metabolism characteristics of difenoconazole and the application dose may be effectively decreased in the open field and greenhouse pepper-planted ecosystem.

Abstract:The enantioselective environmental behavior of difenoconazole, a widely utilized triazole fungicide commonly detected in agricultural soils, has yet to be comprehensively explored within the earthworm-soil system. To address this research gap, we investigated the bioaccumulation and elimination kinetics, degradation pathways, biotransformation mechanisms, spatial distribution, and toxicity of chiral difenoconazole. The four stereoisomers of difenoconazole were baseline separated and analyzed using SFC-MS/MS. Pronounced enantioselectivity was observed during the uptake phase, with earthworms exhibiting a preference for (2 R ,4 R )-difenoconazole and (2 R ,4 S )-difenoconazole. A total of five transformation products (TPs) were detected and identified using UHPLC-QTOF/MS in the earthworm-soil system. Four of the TPs were detected in both earthworm and soil, and one TP was produced only in eaerthwroms. Hydrolysis and hydroxylation were the primary transformation pathways of difenoconazole in both earthworms and soil. Furthermore, a chiral TP, 3-chloro, 4-hydroxy difenoconazole, was generated with significant enantioselectivity, and molecular docking results indicate the greater catalytic bioactivity of (2 R ,4 R )- and (2 R ,4 S )-difenoconazole, leading to the preferential formation of their corresponding hydroxylated TPs. Furthermore, Mass Spectrometry Imaging (MSI) was applied for the first time to explore the spatial distribution of difenoconazole and the TPs in earthworms, and the "secretory zone" was found to be the dominant region to uptake and biodegrade difenoconazole. ECOSAR predictions highlighted the potentially hazardous impact of most difenoconazole TPs on aquatic ecosystems. These findings are important for understanding the environmental fate of difenoconazole, evaluating environmental risks, and offering valuable insights for guiding scientific bioremediation efforts.

Abstract:Elucidating the enantiomeric chemistry and enantioselective fate of the novel chiral triazole fungicide mefentrifluconazole is of vital importance for agroecosystem safety and human health. The absolute configuration of mefentrifluconazole was identified firstly as S-(+)-mefentrifluconazole and R-(−)-mefentrifluconazole on a cellulose tris(3-chloro-4-methylphenylcarbamate) chiral phase. A baseline resolution (<math>Rs,</math> 2.51), favorable retention (<math>RT</math> ≤ 2.24 min), and high sensitivity (LOQ, 0.5 μg/kg) of enantiomer pair were achieved by reversed-phase liquid chromatography tandem mass spectrometry combined with a 3D response surface strategy. Nationwide field trials were undertaken to clarify the enantiomer occurrence, enantioselective dissipation, terminal concentrations, and storage stability of S-mefentrifluconazole and R-mefentrifluconazole in watermelon across China. The original deposition of the sum of enantiomer pair was estimated to be 14.4–163.7 μg/kg, and terminally decreased to < LOQ–59.3 μg/kg 10 days after foliage application. S-mefentrifluconazole preferentially degraded (T1/2, 3.3–6.0 days), resulting in the relative enrichment of R-mefentrifluconazole (T1/2, 3.9–6.6 days) in watermelon. A probabilistic model is recommended for the dietary risk assessment, although both acute (%ARfD, 0.435–22.188%) and chronic (%ADI, 1.697–9.658%) risks are acceptable for associated population. The long-term exposures should be continuously emphasized given the increasing applications and persistent fate of mefentrifluconazole, especially for urban children.

Abstract:The chiral fungicide prothioconazole (PTZ) is extensively employed in agricultural practices, prompting serious concern due to its environmental impact. PTZ is prone to undergo metabolism, leading to the formation of chiral prothioconazole-desthio (dPTZ) in the environment. However, limited knowledge exists regarding its enantioselective behavior and toxicity towards invertebrate organisms in soil ecosystems. In this study, R-(-)- and S-(+)- PTZ enantiomers were individually synthesized, and their stereoselective toxicity effects on earthworms (E. foetida) were studied in artificial soil under environmentally relevant concentration exposures. The results showed a significant accumulation of dPTZ in earthworms, surpassing the levels of PTZ. Moreover, the concentration of S-(-)- dPTZ in earthworms was notably higher than that of R-(+)- dPTZ after exposure, reaching peak levels on day 14. Concurrently, oxidative stress induced by S-(+)- PTZ enantiomers in earthworms exhibited a substantial increase compared to R-(-)- enantiomers on day 14, indicating a higher ecological risk associated with the former in non-target organisms. Transcriptome analysis unveiled distinct impacts on earthworm physiology. S-(+)-PTZ exposure significantly affected energy metabolism, immune responses and digestive systems. In contrast, R-(-)-PTZ exposure influenced the synthesis of carbohydrates, proteins, and lipids. These insights contribute to understanding the complex interactions between PTZ enantiomers and soil-dwelling organisms, providing a scientific foundation for advancing the application of high efficiency, low toxicity PTZ monomer pesticides.

Abstract:AbstractThe enantioselective behaviors of chiral pesticides would affect the accuracy of risk assessment. This study evaluated the enantioselectivity of chiral triticonazole (a widely used fungicide) in three kinds of fruits. Firstly, the enantioseparation of triticonazole enantiomers was carried out within 1.2 min utilizing CHIRALPAK OJ-3 column with a mixture of CO2 and methanol (93:7, v/v) using SFC-MS/MS. Secondly, field trials were conducted to clarify the enantioselective degradation and residue of S-( +)-triticonazole and R-(–)-triticonazole in fruits. The initial concentrations of rac-triticonazole were 25.1–93.1 ng/g, and enantioselective degradation was observed in pear, peach, and jujube after 2 h, 10 days, and 3 days, respectively. The degradation of S-( +)-triticonazole was fastest in pear (T1/2, 2.01 days), while the T1/2 of R-(–)-triticonazole was 5.02 days. The residue concentrations of rac-triticonazole were less than the MRL set by EU (10 ng/g) on the 3rd and 21st day in pear and peach, respectively, which were lower than 10 ng/g in jujube on the 30th day (no MRL). Finally, we found that the dietary intake risks of rac-triticonazole in fruits were low for 2–7 age, 20–50 age/female, and 20–50 age/male. The current study could provide complimentary references for the rational usage, MRL formulation, and risk assessment of chiral triticonazole.

Abstract:Continued application of new chiral fungicide mefentrifluconazole (MFZ) increases its risk to soil ecosystem. However, the toxicity of MFZ enantiomers to soil fauna and whether stereoselectivity exists remains poorly elucidated. Based on multilevel toxicity endpoints and transcriptomics, we investigated the negative effects of racemic, R-(-)-, and S-(+)-MFZ on Eisenia fetida . After exposure to S-(+) configuration at 4 mg/kg for 28 day, its reactive oxygen species levels were elevated by 15.4% compared to R-(-) configuration, inducing enantiospecific oxidative stress and transcriptional aberrations. The S-(+) isomer induced more severe cell membrane damage and apoptosis than the R-(-) isomer, and notably, the selectivity of apoptosis is probably dominated by the mitochondrial pathway. Mechanistically, differential mitochondrial stress lies in: S-(+) isomer specifically up-regulated mitochondrial cellular component compared to R-(-) isomer and identified more serious mitochondrial fission. Furthermore, S-(+) conformation down-regulated biological processes associated with ATP synthesis and metabolism, with specific inhibition of mitochondrial respiratory electron transport chain complex I and IV activity resulting in more severe electron flow disturbances. These ultimately mediated enantioselective ontogenetic process disorders, which were supported at phenotypic (weight loss), genetic, and protein (reverse modulate TCTP and Sox2 expression) levels. Our findings offer an important reference for elucidating the enantioselective toxicological mechanism of MFZ in soil fauna.

Abstract:More attention is being paid to the enantioselective toxicity of chiral pesticides. However, limited investigations have been done to assess the ecological risks of chiral pesticides to soil community. Fenvalerate (FV), an extensively used synthetic pyrethroid, is a typical chiral pesticide. The most insecticidally active enantiomer of FV, esfenvalerate (ESFV), also has been marketed and widely used. In this study, the toxicological sensitivity and bioaccumulation of FV and ESFV in earthworms were assessed. The results showed that FV was less toxic than ESFV, but more accumulated in earthworms. ESFV was at least 4 times more toxic to earthworms than FV according to the filter paper contact toxicity test and the artificial soil test. Enantiospecific induction in oxidative stress was observed in earthworms exposed to FV and ESFV. The bioaccumulation of FV and ESFV in earthworm tissues was also enantioselective, preferentially accumulating FV. The uptake of ESFV by earthworms was lower than that of FV, so that the biota to soil accumulation factor (BSAF) value of ESFV was lower than that of FV. Our findings suggest that the enantioselective toxicity and bioaccumulation of chiral pesticides should be considered for evaluating ecological risks of these compounds to non-target organisms.

Abstract:For decades, pesticides have been widely used for agricultural activities around the world, and the environmental problems caused by these compounds have raised widespread concern. However, the different enantioselective behaviors of chiral pesticide enantiomers are often ignored. Here, the selective degradation patterns and mechanisms of chiral pesticide enantiomers were successfully investigated for the first time in the soils of three cultivation areas with different pH values. Beta-cypermethrin was chosen as the target analyte. We found that the degradation rates of the four isomers of beta-cypermethrin were different. We used stepwise regression equations between degradation rates and functional genes to quantitatively study their relationships. Quantitative response analysis revealed that different isomers have different equations even under identical conditions. The results of path analysis showed that a single functional gene can make different direct and indirect contributions to the degradation of different isomers. Finally, the high-throughput technology was used to analysis the genome of the three tested soils and then compared the main microbial communities in them. We have successfully devised a method to investigate the molecular biological mechanisms of the selective degradation behavior of chiral compounds, thus enabling us to better understand these mechanisms.

Abstract:Fenpropidin (FPD), a widely employed chiral fungicide, is frequently detected in diverse environments. In an in vitro rat liver microsomes cultivation (RLMs), the metabolism exhibited the order of R -FPD > S -FPD, with respective half-lives of 10.42 ± 0.11 and 12.06 ± 0.15 min, aligning with kinetic analysis results. CYP3A2 has been demonstrated to be the most significant oxidative enzyme through CYP450 enzyme inhibition experiments. Molecular dynamics simulations unveiled the enantioselective metabolic mechanism, demonstrating that R -FPD forms hydrogen bonds with the CYP3A2 protein, resulting in a higher binding affinity (-6.58 kcal mol -1 ) than S -FPD. Seven new metabolites were identified by Liquid chromatography time-of-flight high-resolution mass spectrometry, which were mainly generated through oxidation, reduction, hydroxylation, and N-dealkylation reactions. The toxicity of the major metabolites predicted by the TEST procedure was found to be stronger than the predicted toxicity of FPD. Moreover, the enantioselective fate of FPD was studied by examining its degradation in three soils with varying physical and chemical properties under aerobic, anaerobic, and sterile conditions. Enantioselective degradation of FPD occurred in soils without enantiomeric transformation, displaying a preference for R -FPD degradation. R -FPD is a low-risk stereoisomer both in the environment and in mammals. The research presented a systematic and comprehensive method for analyzing the metabolic and degradation system of FPD enantiomers. This approach aids in understanding the behavior of FPD in the environment and provides valuable insights into their potential risks to human health.

Abstract:Comment on the editorial by Edwards et al Smoking restrictions have been progressively introduced in enclosed public spaces in a growing number of nations as the evidence of the harms of passive smoking accumulates and public preference for smoke free air grows. Bans on smoking on public transport and airlines, and in cinemas, theatres, and elevators have long been taken for granted in many nations. Indoor workplaces are also increasingly becoming smoke free by virtue of either explicit legislation or management policy. Yet throughout the world, hospitality venues—particularly pubs and bars—have staunchly resisted this momentum. The editorial by Edwards et al in this issue1 describes a depressingly typical “Groundhog Day” scenario to anyone monitoring global developments in this area. The situation today is an object lesson in “half-pregnant” policy and in how public health law should not be made. Those least exposed—the relatively transient patrons of restaurants, public transport, cinemas, and shopping malls—have often full legal protection from passive smoking. Yet those most exposed—bar and casino staff—are the least protected. In effect, government inaction says to them “we’ll protect citizens from passive smoking even when their exposure is brief. But notwithstanding your unparalleled high exposure, we’ll offer you no protection”. It is unimaginable that workplace health protection could proceed in such a fashion in relation to toxic fumes, noise, or asbestos. Abstract:The enantioselective toxic effect and environmental behavior of chiral pesticides have attracted increasing research attention. In this study, the enantioselective toxicity and residues of hexaconazole (HEX) in earthworms (Eisenia fetida) were investigated. In the present study, significant enantioselective degradation characteristics were observed in artificial soil with the R-enantiomer preferentially degrading (p < 0.05); however, no significant enantioselective bioaccumulation was observed in the earthworms (p > 0.05). The acute toxicity of S-(+)-HEX was higher than that of R-(-)-HEX in earthworms, with 48-h LC50 values of 8.62 and 22.35 μg/cm2, respectively. At 25 mg/kg, enantiospecific induction of oxidative stress was observed in earthworms; moreover, S-(+)-HEX had a greater influence on the contents of malonaldehyde, cytochrome P450, and 8-hydroxy-2-deoxyguanosine than R-(-)-HEX. These results were consistent with those of the enrichment analysis of differentially expressed genes. The transcriptome sequencing results showed that S-(+)-HEX had a more significant influence on steroid biosynthesis, arachidonic acid metabolism, and cell cycle processes than R-(-)-HEX, leading to abnormal biological function activities. These results indicate that S-(+)-HEX may pose a higher risk to soil organisms than R-(-)-HEX. This study suggests that the environmental risk of chiral pesticides to nontarget organisms should be assessed at the enantiomeric level.

Abstract:The chiral pesticide famoxadone is mainly applied to control fungal diseases on fruiting vegetables. The fungicidal activity, ecotoxicological effects, and degradation behavior of famoxadone enantiomers are less well known. In this study, a systemic assessment of the stereoselectivity of famoxadone was performed in cucurbits and soil. Famoxadone enantiomers presented distinct inhibitory activities among different fungal species. The bioactivities of R-(−)-famoxadone were 2.7–178 times higher than S-(+)-famoxadone toward five phytopathogens. Based on the obtained LC50 values, famoxadone was super toxic to Eisenia foetida (E. foetida). Moreover, the acute toxicity of R-(−)-famoxadone presented 167 times greater to E. foetida than that of S-(+)-famoxadone, indicating that R-(−)-famoxadone showed higher bioactivity toward target organisms and non-target organisms than S-(+)-famoxadone. In addition, a simple high-performance liquid chromatography (HPLC) method was established to determine the stereoselective degradation of famoxadone in two species of cucurbits (cucumber and chieh-qua) and in field soil. The half-life values of famoxadone degradation were from 5.4 to 14.1 days, indicating that famoxadone was easily degraded. Additionally, no stereoselective degradation was found in cucurbits and soil. The results may provide promising implications for comprehensive environmental and ecological risk assessments of famoxadone.

Abstract:As a chiral piperidine fungicide, fenpropidin has been widely used to control plant diseases. However, there are rare studies that have investigated fenpropidin at the enantiomer level. In this study, the single-factor analysis combined with a Box-Behnken design was used to obtain the optimal enantio-separation parameters of the fenpropidin enantiomers on ultra-performance liquid chromatography-tandem mass spectrometry. The absolute configuration of two fenpropidin enantiomers was confirmed for the first time using electron circular dichroism and optical activity. On the Lux cellulose-3 column, S-(-)-fenpropidin flowed out before R-(+)-fenpropidin. The enantio-separation mechanism was revealed by molecular docking. A modified QuEChERS method was developed for the trace determination of the fenpropidin enantiomers in seven food and environmental substrates. The average recoveries were 71.5–106.1% with the intra-day and inter-day relative standard deviations of 0.3–8.9% and 0.5–8.0%. The method was successfully verified by enantioselective dissipation of fenpropidin in soil under the field. R-(+)-fenpropidin dissipated faster than S-(-)-fenpropidin, and the half-lives were 19.8 d and 22.4 d. This study established a brand-new effective chiral analysis method for the fenpropidin enantiomers, providing a basis for accurate residue monitoring and the risk assessment of fenpropidin.

Different Degradation Patterns of Chiral Contaminant Enantiomers: Paclobutrazol As a Case Study

Abstract:Studies often neglect the different trends between enantiomers in soil chiral contaminants. In the present study, paclobutrazol was chosen as the target analyte and the selective degradation patterns of this chiral pesticide were investigated in soils. After 14 days of culture, the degradation kinetics results demonstrated the degradation process of the two enantiomers conforms to first-order degradation kinetics equation, and the degradation rates were significantly different (paired t-test, P < 0.05). The degradation rate of (2S,3S)-paclobutrazol (S-paclobutrazol) was higher than that of (2R,3R)-paclobutrazol (R-paclobutrazol), with the concentration decrease of 42.6 and 28.7%, respectively. Meanwhile, the enantiomer fraction (EF) was 0.43 at final sampling point, and the half-life (t1/2) of S-paclobutrazol and R-paclobutrazol were 17.3 and 28.9 days, which demonstrated significant enantioselective degradation. Results from this study served to enhance our understanding about the enantioselective degradation behaviors of chiral pesticides, which provided some scientific basis for the potential risk assessment of chiral pesticides in the environment.
