Abstract:The development of protocols for the construction of congested quaternary centers is highly sought-after. Herein, we report a method for the cross-coupling of C(sp3) tertiary Grignard reagents with C(sp2) styrenyl bromides using readily available nickel precatalysts. We identified conditions that afford the products in practical yield for several combinations of electrophiles and nucleophiles, including sensitive α-magnesiated Grignard reagents. Dependent upon the nature of their substituents, regiodivergency was observed when α-vinyl bromides were employed.

Abstract:A nickel-catalyzed Kumada coupling of aniline derivatives was developed by selective cleavage of aryl C-N bonds under mild reaction conditions. Without preinstallation of an ortho directing group on anilines, the cross-coupling reactions of Boc-protected aromatic amines with aryl Grignard reagents afforded unsymmetric biaryls. Mechanistic studies by DFT calculations revealed that the nickel-mediated C-N bond cleavage is the rate-limiting step.

Abstract:Palladium-catalyzed Suzuki-Miyaura cross-coupling or aryl halides is widely employed in the synthesis of many important molecules in synthetic chemistry, including pharmaceuticals, polymers and functional materials. Herein, we disclose the first palladium-catalyzed decarbonylative Suzuki-Miyaura cross-coupling of amides for the synthesis of biaryls through the selective activation of the N-C(O) bond of amides. This new method relies on the precise sequence engineering of the catalytic cycle, wherein decarbonylation occurs prior to the transmetallation step. The reaction is compatible with a wide range of boronic acids and amides, providing valuable biaryls in high yields (>60 examples). DFT studies support a mechanism involving oxidative addition, decarbonylation and transmetallation and provide insight into high N-C(O) bond activation selectivity. Most crucially, the reaction establishes the use of palladium catalysis in the biaryl Suzuki-Miyaura cross-coupling of the amide bond and should enable the design of a wide variety of cross-coupling methods in which palladium rivals the traditional biaryl synthesis from aryl halides and pseudohalides.

Abstract:The Suzuki-Miyaura coupling is among the most important C-C bond-forming reactions available due to its reliability, chemoselectivity, and diversity. Aryl halides and pseudohalides such as iodides, bromides, and triflates are traditionally used as the electrophilic coupling partner. The expansion of the reaction scope to nontraditional electrophiles is an ongoing challenge to enable an even greater number of useful products to be made from simple starting materials. Herein, we present how an NHC-based Pd catalyst can enable Suzuki-Miyaura coupling where the C(acyl)-O bond of aryl esters takes on the role of electrophile, allowing the synthesis of various ketone-containing products. This contrasts known reactions of similar esters that provide biaryls via nickel catalysis. The underlying cause of this mechanistic divergence is investigated by DFT calculations, and the robustness of esters compared to more electrophilic acylative coupling partners is analyzed.

Abstract:In this study, we present a first comprehensive palladium-catalyzed Suzuki–Miyaura C(sp2)-C(sp2) cross-coupling reaction, employing newly derived di-bromo-boracycles as pivotal reagents for the synthesis of ortho arylated N-aryl amides. Our research hinges on the utilization of di-bromo-boracycles, which act as temporary reservoirs of boron, facilitating subsequent transformations in cross-coupling reaction. The di-bromo-boracycle offers a unique opportuni-ty for regioselective carbon-carbon bond formation to the ortho position of N-aryl amides. This mild, site-specific, and scalable protocol is compatible with a wide array of easily accessible functionalized anilides and aryl iodides, as demonstrated by 55 examples, yielding products in the range of 30%-95%. Furthermore, the methodology trans-cends conventional practices, enabling the construction of previously unattainable diagonal aryl systems, highlighting the exceptional potential of our approach for accessing intricate molecular architectures.

Abstract:We described the scope and limitations of palladium‐catalyzed Suzuki‐Miyaura cross‐coupling reactions of (hetero)aryl halides with (hetero)arylboronic acids with our optimized phosphorus imidazo[1,5‐a]pyridine ligand JagPhos I. Successful reactions were obtained with a series of (hetero)aryl halides with (hetero)arylboronic acids to furnish sterically‐hindered biaryls and biheterocycles. We also presented the scope and limitations of palladium‐catalyzed Buchwald‐Hartwig amination reactions of (hetero)aryl halides with anilines, secondary amines, and primary amines with our optimized phosphorus imidazo[1,2‐a]pyridine ligand JagPhos II. Sucessful reactions were obtained with a series of (hetero)aryl halides with various anilines and secondary amines. Palladium‐catalyzed Suzuki‐Miyaura and Buchwald‐Hartwig cross‐coupling reactions have become indispensable tools in the synthesis of otherwise inaccessible compounds because of the efficient, catalytic nature of these styles of reactions. The development of novel monophosphine ligands to achieve the cross‐coupling of substrates that are generally unreactive under standard conditions has developed into an extremely important area of research in the field of organometallic chemistry. Herein, we show the use of an imidazopyridine monophosphine ligand JagPhos I in the Suzuki‐Miyaura palladium‐catalyzed cross‐coupling reaction to deliver sterically‐hindered biaryls and unsymmetrical biheterocycles. We also report the scope and limitations of imidazopyridine monophosphine ligand JagPhos II in the Buchwald‐Hartwig amination reactions of (hetero)aryl halides with anilines, secondary amines, and primary amines.

Abstract:A family of 2-(9H-carbazol-9-yl)phenyl-based phosphine ligands were synthesized and their efficacy in promoting the steric hindered Buchwald–Hartwig amination was evaluated. In the presence of Pd(OAc)2 (0.03–1.0 mol%) associated with the newly developed a carbazolyl-derived phosphine ligand, the synthesis of tetra-ortho-substituted diarylamines proceeded smoothly with excellent product yields (up to 99%). A remarkable result was obtained even for the coupling of highly sterically congested 2,6-diisopropylaniline and hindered 2-chloro-1,3,5-triisopropylbenzene (96% isolated yield). A possible decomposition pathway for the anthracenyl C–N coupling product is also reported.

Abstract:A palladium/norbornene cooperative catalyzed selective C-H bond amination of aryl iodides was explored, providing an efficient tool for constructing benzocyclic molecules. When <i>ortho</i>-substituted iodobenzene was involved, the C-H bond amination and following Heck cyclization efficiently delivered a 3-methyl-indole scaffold. On the other hand, we realized the controllable synthesis of monoaminated benzo-cyclobutanyl scaffold. The possible coordination of an installed terminal alkenyl group with palladium and steric hindrance were proposed to be responsible for the monoamination selectivity.

Abstract:A palladium-catalyzed Negishi cross-coupling reaction of ethyl bromodifluoroacetate with aryl bromides or aryl triflates to construct C(sp2)-CF2 bonds is described. The reaction was conducted under mild reaction conditions, and no preparation of organozinc reagents is required. This is the first report encompassing the conversion of aryl triflates into products containing C-CF2 bonds. In addition, the construction of C(sp2)-CHF bonds was achieved under mild conditions via a cross-coupling of aryl iodides with ethyl bromofluoroacetate.

Abstract:With the readily available Gorlos-Phos as a ligand, halogenated diarylamines were prepared efficiently from Pd-catalyzed monoamination of bromophenyl chlorides and primary arylamines, tolerating functional groups such as aldehyde, ketone, nitrile, fluorine, sulphonyl and trifluoromethyl groups. The formation of the amination of the C-CI bond and diamination products were not observed.

Abstract:Suzuki–Miyaura coupling is a practical and attractive carbon−carbon bond formation reaction due to its high efficiency and wide functional group compatibility, but its industrial applications are limited because it is typically catalysed by expensive palladium-containing transition-metal complexes. Here we show a robust and chemoselective organocatalytic Suzuki−Miyaura-type coupling of aryl halides with arylboronic acids catalysed by amines. The utility and scope of this reaction were demonstrated by the synthesis of several commercially relevant small molecules and a selection of derivatives of pharmaceutical drugs. The coupling of aryl halides and arylboronic acids is generally performed by metal-catalysed Suzuki–Miyaura reactions while metal-free approaches remain elusive. Here an organocatalytic approach based on amine catalysts is introduced for the preparation of commercially relevant asymmetric biaryls.

Abstract:De novo catalytic syntheses of diarylamines from a palladium‐catalyzed reductive Buchwald‐Hartwig amination of nitroarenes with aryl (pseudo)halides is described. The exquisite use of upstream nitroarenes as arylamine surrogates, the judicious selection of bis(pinacolato)diboron (B2pin2) as a stoichiometric reducing agent, and wide substrate scope including (hetero)aryl halides (Cl, Br and I) and aryl triflates, constitute the striking features of the current protocol. Moreover, application of this technique to the syntheses of advanced intermediates and active pharmaceutical ingredients also proved successful, thus providing an alternative step‐economical approach to the syntheses of diarylamine‐incorporated molecules. Preliminary mechanistic investigation demonstrates that an amine and a nitrosoarene intermediates might be involved in this reductive event.

Abstract:Palladium-catalyzed aminocarbonylation of 3-iodochromone was studied in the presence of primary and secondary amines using atmospheric pressure of carbon monoxide as a carbonyl source.
