Abstract:This study was conducted to evaluate 16 early Indian cauliflower genotypes for agro-morphological traits, bioactive compounds and antioxidant activity. Analysis of variance, cluster analyses, and principal components analysis (PCA) elucidated patterns of variation among the genotypes. The genotype CC 12 exhibited highest marketable curd weight (450 g). Curd sinigrin varied from 3.29 to 16.37 µmol 100g−1 FW with maximum in DC 41–5. Total antioxidant capacity (CUPRAC and FRAP) ranged from 8.87 to 24.24 mg GAE 100g−1 and 11.71 to 34.00 mg GAE 100g−1 FW, respectively. The Phenotypic coefficient of variation (PCV) was higher than the Genotypic coefficient of variation (GCV) for all the characters with highest GCV, PCV and high heritability value for sinigrin content in curd. D2 statistics classified the genotypes into three clusters where genotypes in cluster I had high sinigrin both in curd and leaf. The PCA revealed that first principal component (PC1) contributed 47.86% of total variation whereas, second principal component (PC2) contributed 35.14%. The genotypes Pusa Deepali, Selection 71, and CC 13 could be used in breeding programme for higher yield and bioactive compounds.
Identification of elite genotypes through morpho-biochemical and molecular characterization of turmeric ( Curcuma longa L.)

Abstract:Turmeric is recognized as "the golden spice" and "spice of life". The abundant medicinal values of turmeric make it an important industrial crop. The genetic diversity analysis of this species is important for further improvement in its productivity through selective breeding to cap the continuous demand for turmeric. The variance analysis imparted notable differences among genotypes for 17 studied traits under study. High genotypic and phenotypic coefficient of variations were noticed for leaf oil percentage followed by rhizome oil, fresh finger yield, curcumin, dry rhizome per plant, secondary rhizomes per plant and leaves per plant. The genetic divergence assessed by Mahalanobis D 2 statistic, evident broad genetic variability among turmeric genotypes as accommodated by seven clusters. Out of 25 simple sequence repeat (SSR) markers, 15 were polymorphic. The average polymorphic information content (PIC) varied between 0.474 and 0.306 with an average of 0.408. The SSR marker analysis and diversity analysis, grouped 24 genotypes of turmeric in four major groups namely A, B, C and D with 13, 7, 3 and 1 genotypes, respectively. The genetic similarity among the genotypes varied from 0.07 to 0.82 (ATG-1/ATG-2) and the average similarity coefficient was (0.23). Marker cuMisat-22, cuMisat-7 and cuMisat-19 were found most suitable for diversity analysis. Based on mean performance and diversity analysis, accession ATG-17 (local collection from Anand) was superior for both fresh and dry rhizome yield per plant. Genotype Kasturi (ATG-21) and ATG-4 4 (local collection from Navsari) can be an important raw materials for the pharmaceutical and cosmetics industries as they were richer in curcumin and rhizome oil content respectively.

Abstract:Curcuma longa L. (Family- Zingiberaceae), commonly called turmeric, is one of the most important commercial spices. Being a center of turmeric's diversity, India boasts of more than fifty cultivars with variable agronomic characters and curcumin content. However, the quest for calibrating their genetic variability remains debatable as popular molecular marker-based studies have yielded inconsistent results. A promising PCR-based approach, known as the Start Codon Targeted (SCoT)-PCR system has been established to be more efficient than random DNA marker systems because this technique can generate more information on genetic polymorphism patterns. Hence, the present study proposes to use Start Codon Targeted (SCoT) markers to assess inter-cultivar genetic diversity of turmeric. We report an orthogonal L 16 (4 5 ) design-based approach to investigate the influence of five crucial factors (dNTPs, Taq DNA polymerase, primer, template DNA and annealing temperature of primer) on the efficiency of the polymerase chain reaction (PCR) for development of an optimized Start Codon Targeted (SCoT) amplification protocol using twenty-seven different cultivars of C. longa . The optimized PCR protocol yields best results with a 20 uL reaction mixture, containing 10×PCR Buffer (with 15 mM MgCl 2 ), 2.5 mM dNTPs, 10 mM SCoT primer, Taq polymerase (2 U), template DNA (50 ng), which was verified by using twenty-five SCoT primers. This standardized rapid protocol of SCoT-PCR reported here was employed to examine genetic diversity among twenty-seven Indian cultivars of turmeric, revealing high range of polymorphism ranging from 81.25 % to 100 %. Unweighted pair group method of arithmetic averages (UPGMA) cluster analysis of the SCoT marker data segregated the twenty-seven cultivars into two main clusters. The first cluster comprised of nine cultivars (36 % of the cultivars) which was further divided into three sub-clusters and the second one consisted of eighteen cultivars (64 %) which further divided into four sub-clusters. From the results of this study, it can be inferred that the employment of SCoT markers can be an effective tool for analysing genetic variability, phylogenetic relationships and develop a rich genetic resource management system for C. longa cultivars.

Abstract:Turmeric ( Curcuma longa L.) is an economically valuable spice crop with a wide range of applications. Crop improvement efforts in this crop had led to identification, selection, and release of several varieties with desirable traits for yield, curcumin content etc. Understanding of genetic variation is a pre-requisite for any crop improvement programme. In this study, we examined the genetic variation present in prominent 18 improved varieties of turmeric in India using morphological and microsatellite markers. We studied 12 morphological characters including 11 quantitative traits, and curcuminoid content following the DUS guidelines of turmeric. For molecular characterization, we used 56 molecular markers including SSRs, and ISSRs to determine the genetic polymorphism among the genotypes. PIC value of these markers ranged from 0 to 0.5. Our study thus confirms the reliability of these molecular markers to study the genetic variation of turmeric varieties. The combination of methods i.e., cluster analysis based on morphological characterization and the UPGMA dendrogram based on molecular characterization revealed both the phenotypic and molecular variation among the improved turmeric varieties. This has helped us to understand pattern of variation, the level of genetic similarity and the genetic divergence among them. It was found that the varieties Varna, Megha Turmeric and Suvarna were the most divergent while Punjab Haldi 1 and Sudarsana were the least divergent among the improved varieties. The results from this study can be used in designing breeding programmes in turmeric.

Abstract:The availability of genetic resource provides an opportunity for plant breeders to develop new cultivars with desirable characteristics and these characteristics encompass various traits preferred by farmers (high yield potential, larger seed size, etc) as well as traits favored by breeders (pests and disease resistance). Genetic resources are also essential for conservation, as they provide a basis for identifying threatened species and developing conservation strategies. Curcuma caesia Roxb. (Zingiberaceae) is an underutilized perennial herb with bluish-black tuberous rhizome having the characteristic of aromatic and medicinal properties. There is an ample genetic diversity of Curcuma plants with medicinal importance around the world. Authentication and ensuring the genuineness of Curcuma are crucial aspects for marker-based analysis. Therefore, genetic variation serves as the base for the identification and characterisation of the plants. Molecular marker technology is widely used in genetic diversity analysis, germplasm resource identification, and molecular marker-assisted breeding technique. Multiple molecular markers have been used for characterization and identification of species for its sustainable utilization and conservation. The molecular investigation, which involves techniques like RAPD, SCAR, ISSR, and AFLP for fingerprinting, plays a crucial role in establishing molecular markers to assess the authenticity and diversity of plant species. Various marker analyses have demonstrated a significant degree of genetic variation among the species and these patterns tend to differ due to environmental influences and genetic factors. This review facilitates a complete information about the molecularassisted genetic diversity study of Curcuma caesia Roxb. (Black turmeric) which may be used as a database while exploring the diversity analysis.
Genetic Diversity and Population Structure Analysis in Indian Mustard Germplasm Using Phenotypic Traits and SSR Markers

Abstract:Indian mustard is an economically important oilseed crop in India; therefore, exploring the genetic diversity of various germplasm collections is quite relevant for its genetic improvement. In the present study, we investigated genetic diversity and population structure among a panel of 145 Indian mustard germplasm accessions using 11 agro-morphological traits and 182 SSR markers. The result of variance analysis exhibited significant differences among the tested genotypes inferring the presence of a high degree of variability. The phenotypic coefficient of variation for seed yield (CV = 28.26%) was the highest among all the studied traits followed by biological yield (CV = 25.72%). Days to maturity ranged from 130 to 141 days (CV = 1.88%), which was the minimum variation exhibited by Indian mustard genotypes. Out of 235 SSR primer pairs evaluated, 182 (77.45%) SSRs resulted in polymorphic amplicons, while 53 (22.55%) SSRs amplified monomorphic loci. Allele number varied from 2 to 7 with 3.97 average number of alleles per SSR marker. PIC value varied from 0.03 (SJ1668I) to 0.71 (EJU4) with an average value of 0.39 per SSR marker. Gene diversity in the present study ranged from 0.03 (SJ1668I) to 0.75 (EJU4) with an average value of 0.46. Cluster analysis using morphological traits and SSR markers grouped all 145 accessions into two major clusters each, respectively. Population structure analysis grouped all the genotypes into three subpopulations with varying degrees of admixture genotypes. Analysis of molecular variance inferred that 88% of the total variation resides within the groups. The resulting estimates of genetic diversity and population structure may be useful for the effective management of Indian mustard germplasm resources and for future mustard breeding programs.

Abstract:Curcuma kwangsiensis S. K. Lee & C. F. Liang, a traditional herb in southern China, accumulates substantial amounts of active components, curcuminoids and volatile oil. However, the limited hereditary information and SSR markers have hindered its breeding program and genetic analysis. Here, we examined the transcriptome of C. kwangsiensis using next-generation sequencing (NGS) technology. A dataset with 8.17 Gb of raw reads was generated and assembled into 77,976 unigenes. Moreover, 11,678 EST-SSR markers were screened from transcriptome data. Of the 800 synthesized primer pairs, 486 (60.8%) exhibited successful amplification and 115 (23.7%) were polymorphic. A set of 24 selected markers showed high cross-species transferabilities among 13 Curcuma species. In total, 277 alleles (6–19 alleles per locus) were discovered, and the polymorphic information content (PIC) ranged from 0.496 to 0.905. Based on the results of cluster and structure analyses, the 75 accessions were classified into four major groups with some admixtures. Finally, a core collection (22 accessions) was identified, exhibiting Na, Ne, I, and PIC values with retention rates of 87.7%, 107.8%, 102.5% and 102%, respectively. All these unigenes and EST-SSRs will be useful for germplasm resource evaluation and for diversifying the potential of product derivatives from Curcuma.

Abstract:Cannabis sativa L. has been an important medicinal and recreational plant since ancient times due to the presence of cannabinoids. Recently, there has been resurgence in cannabis research on account of phytochemical cannabidiol (CBD) oweing to its immense medicinal properties. This study reports the morphological, anatomical, phytochemical and molecular characterization of C. sativa collected from different regions in North India for diversity analysis and high-CBD accessions. The plant height, stem diameter, leaf length, and number of primary branches significantly differed ( p < 0.05) among the accessions. There is considerable diversity in the size, shape and surface ornamentation of C. sativa seeds. However, the shape, size and annulus diameter of the pollen grains exhibited a narrow range in subtropical climates. The collenchymatous tissue revealed diversity among the accessions, producing wavy stems in most male accessions. The northern Indian cannabis populations are dominated by intermediate chemotypes, where both CBD and tetrahydrocannabinol (THC) are present. CBD-rich fibre-type accessions are rarely found in nature. We found one CBD-rich accession with 0.19% THC and 1.36% CBD content, which could be a potential parental line for future breeding programmes. Dendrogram clustering and population structure analysis grouped the accessions into two major subpopulations irrespective of chemotype and sex. Overall, this comprehensive study provides valuable insights into the diversity of Indian C. sativa accessions, laying a foundation for further breeding and conservation efforts for CBD-rich Indian Cannabis germplasms.

Abstract:Cannabis , also known as marijuana or hemp, has been utilized since ancient times for industrial, religious, recreational, and medical uses. However, regardless of the intended use, there are legal requirements for quantitative testing of cannabinoids across the supply chains. This investigation aimed to evaluate the genetic diversity of 54 Cannabis samples collected from tropical and subtropical regions in India. The research found a high genotypic coefficient of variation (GCV), phenotypic coefficient of variation (PCV), heritability, and genetic advance for total cannabidiol (CBD) content. The genotypic and phenotypic correlation among the morpho-chemical characters revealed strong positive correlations among most characters. Clustering and Principal component analysis identified three accessions in cluster II (CIM-CS-65, CIM-CS-189 & CIM-CS-64) with high CBD content that could be used for breeding and as sources of high CBD content in Cannabis . CIM-CS-64, with its high CBD content with 0.01%THC content, holds potential as a valuable parental line for utilization in hybridization programs and recombinant breeding. Furthermore, in accordance with the NDPC Act of 1985, CIM-CS-64 can be commercialized for medicinal purposes, making it a promising source for the development of medicinal CBD products.

Abstract:Abstract Background The genetic base of soybean cultivars in India has been reported to be extremely narrow, due to repeated use of few selected and elite genotypes as parents in the breeding programmes. This ultimately led to the reduction of genetic variability among existing soybean cultivars and stagnation in crop yield. Thus in order to enhance production and productivity of soybean, broadening of genetic base and exploring untapped valuable genetic diversity has become quite indispensable. This could be successfully accomplished through molecular characterization of soybean genotypes using various DNA based markers. Hence, an attempt was made to study the molecular divergence and relatedness among 29 genotypes of soybean using SSR markers. Methods and results A total of 35 SSR primers were deployed to study the genetic divergence among 29 genotypes of soybean. Among them, 14 primer pairs were found to be polymorphic producing a total of 34 polymorphic alleles; and the allele number for each locus ranged from two to four with an average of 2.43 alleles per primer pair. Polymorphic information content (PIC) values of SSRs ranged from 0.064 to 0.689 with an average of 0.331. The dendrogram constructed based on dissimilarity indices clustered the 29 genotypes into two major groups and four sub-groups. Similarly, principal coordinate analysis grouped the genotypes into four major groups that exactly corresponded to the clustering of genotypes among four sub-groups of dendrogram. Besides, the study has reported eight unique and two rare alleles that could be potentially utilized for genetic purity analysis and cultivar identification in soybean. Conclusion In the present investigation, two major clusters were reported and grouping of large number of genotypes in each cluster indicated high degree of genetic resemblance and narrow genetic base among the genotypes used in the study. With respect to the primers used in the study, the values of PIC and other related parameters revealed that the selected SSR markers are moderately informative and could be potentially utilized for diversity analysis of soybean. The clustering pattern of dendrogram constructed based on SSR loci profile displayed good agreement with the cultivar’s pedigree information. High level of genetic similarity observed among the genotypes from the present study necessitates the inclusion of wild relatives, land races and traditional cultivars in future soybean breeding programmes to widen the crop gene pool. Thus, hybridization among diverse gene pool could result in more heterotic combinations ultimately enhancing genetic gain, crop yield and resistance to various stress factors.

Evaluation of Genetic Diversity Based on Microsatellites and Phytochemical Markers of Core Collection of Cymbopogon winterianus Jowitt Germplasm

Abstract:Cymbopogon winterianus Jowitt is an industrially important crop due to its value in the aromatic, perfumery and pharmaceutical industries. In this study, 72 accessions of C. winterianus were selected for molecular diversity analysis using SSR markers. It revealed a total of 65 polymorphic alleles showing an average of 68.10% polymorphism. The best SSR primer with competency in discriminating the germplasm was 3CM0506 with PIC (0.69), MI (0.69) and Rp (3.12). Genetic variation was studied between Assam, Manipur, Meghalaya and Arunachal Pradesh populations. A dendrogram based on the Neighbour-Joining Method showed clustering of germplasm on the collection site. A total of six relevant genetic populations were identified through a structure harvester software analysis. Moreover, a dendrogram based on similarity, complete linkage and Euclidean distance was also elucidated differentiating the genotypes with respect to the major phytochemical constituents of the essential oil. GC-FID and GC-MS analyses of the essential oil of the 72 germplasms revealed citronellal content from 2.58–51.45%, citronellol from 0.00–26.39% and geraniol from 0.00–41.15%. This is the first molecular diversity report with 72 accessions of C. winterianus collected from the NE region using 28 SSR primers as well as their diversity based on phytochemical markers. This diversity computation will help with acquisition of the knowledge and relationship among each individual accession leading to the development of improved and essential oil component-rich cultivars.

Abstract:Sugarcane is susceptible to red rot disease caused by phytopathogenic fungus Colletotrichum falcatum Went which ultimately affect the economy of farmers as well as sugar based industry. One of the various ways to control this devastating disease is to develop disease resistance sugarcane cultivar and this requires the complete understanding of genetic makeup of pathogen. Although South Gujarat is well known sugarcane cultivating area, less published data can be found about PCR-based genetic diversity in prevalent C. falcatum accessions. So, present investigation aims at finding molecular variation among the ten accessions of C. falcatum using RAPD and ISSR molecular markers. A total of 35 RAPD and 39 ISSR primers were screened across 10 C. falcatum accessions, of which 15 RAPD and 21 ISSR primers have showed consistent amplification. Statistics related to genetic variation were estimated using NTSYS-PC by means of Dice's coefficient. The results revealed 80.6% and 68.07% polymorphism and similarity coefficient ranged from 0.43 to 0.91 and 0.73 to 0.93 in RPAD and ISSR analysis respectively. The dendrogram generated using RAPD, ISSR and combined RAPD-ISSR grouped accessions into different clusters which reveal considerable level molecular variation among the C. falcatum accessions. It is also evident from PCA plots that accessions are rather dispersed with tested marker systems indicating good genetic base. So, in nut shell, we found considerable genetic variation and relatedness within C. falcatum accessions collected from different areas of south Gujarat, India using RAPD and ISSR markers.
Assessment of diversity of Indian aromatic rice germplasm collections for morphological, agronomical, quality traits and molecular characters to identify a core set for crop improvement

Abstract:Abstract Background Besides the Basmati, the aromatic rice germplasm (ARG) accessions are treasured for quality, medicinal value and aroma. The demand for aromatic rice is ever increasing. Genetic diversity is the source of variability to identify superior alleles controlling morphological, agronomic and quality traits, and molecular attributes. This study reports on the characterization of traits in ARG to identify a core set for breeding high-yielding varieties. Methods The genetic diversity was measured on the distinctness, uniformity and stability (DUS) of 46 traits in 208 Indian ARG in field, greenhouse and laboratory tests. We performed individual and combined analysis of DUS traits and molecular data generated using 55 SSR markers. The genetic distances between genotypes were estimated using Mahalanobis D 2 analysis and clustering by standardized Euclidean 2 distances, Ward Minimum variance, Gowers’ similarity index and PowerMarker. The aim was to derive a core set of non-Basmati ARG using PowerCore to deploy in crop improvement. Results Eighty-two alleles were detected. Alleles per marker ranged from 2 (RM505) to 5 (RM276) with an average of 3.04 alleles. The markers are informative in analyzing the diversity as the PIC values estimated varied from 0.17 (RM577 on chromosome 1) to 0.72 (RM276 on chromosome 6) with an average of 0.54 per locus. RM276 with repeat motif of (AG)8A3(GA) 33 on chromosome 6 was the most informative (amplified 5 alleles). The combined analysis had shown genotypes in a few clusters to be more diverse than others. SSR markers RM289, RM505, RM577 and RM22866 were identified as genotype specific markers. With PowerCore, 46 genotypes (22%) were identified as a core set of ARG that represent all the alleles detected in the entire set investigated. 2-Acetyl-1-pyrroline is considered to impart aroma; it was not detected by GC–MS tests in many ARG. Conclusions Forty-six genotypes in the core set have different maturity periods, plant statures, grain types and grain quality traits. A parent can be selected from the core set to improve aromatic rice depending on the breeding objective. The olfactory sensing of strong aroma emitted by cooked kernels of all ARG was found more decisive than the costly GC–MS tests.

Abstract:The rhizomes of Curcuma species have a long medicinal history in Asia. In China, Curcuma species mainly be utilized to make pharmaceutical products, including C. phaecocaulis, C. aromatica, C. wenyujin, C. kwangsiensis and C. longa . In this study, twenty-four samples were selected to study the genetic and chemical variability among five Curcuma species. The ITS2 and trnK intron gene fragment were used to identify the five Curcuma species, the differences in chemical composition were computed using the Euclidean distance based on the data of HPLC characteristic peak areas and the content of six key components, and agronomic characteristics were analyzed including morphological and volatile oil characteristics. The ITS2 and trnK intron gene fragment could distinguish the five Curcuma species clearly. The genetic distance between Curcuma species ranged from 0.0085 to 0.0767 based on the data of ITS2 gene sequences with 32 variation sites, and the genetic distance between Curcuma species ranged from 0.0003 to 0.0194 based on the data of trnK intron gene sequences with 39 variation sites. Five Curcuma species showed otherness chemical composition characteristics, with the Euclidean distance ranging from 3.373 to 6.998. The C. longa showed the biggest variation compared with other species, with the Euclidean distance above 6.239. Among the samples of the original plants of Ezhu, the volatile oil yield of W1 was the highest, reached to 105.75 mL per single plant. Among all the samples, J6 showed the highest yield of volatile oil, reached to 149.42 mL per single plant. The results showed that chemical composition similarity of the medicinal plants was the primary proof for the selection of the original plants of the Curcuma medicinal materials. The genetic distance and chemical variability were important references for discovering new medicinal plant resources.

Abstract:The present investigation was intended to assess the extent of genetic diversity present among the selected genotypes of Oryza sativa (including TGMS lines) using SSR markers. The SSR markers used for the study gave average Polymorphism Information Content (PIC) value of 0.551. The primer RM 9 followed by RM 536 has given maximum PIC value and the primer RM 29 gave the minimum. It was inferred that the primers RM 9 and RM 536 had been the better choices in this study. The lines, TS09 24 and TS09 26 were in the same cluster as they derived from the same parents. The TGMS lines TS 29 and TNAU 27S are in separate cluster from other parents indicating that these lines are highly diversified and their crosses can give improved cultivars. Bangladesh J. Abstract:Abstract Background Various Asparagus species constitute the significant vegetable and medicinal genetic resource throughout the world. Asparagus species serve as important commodity of food and pharmaceutical industries in India. A diverse collection of Asparagus species from different localities of Northwest India was investigated for its genetic diversity using simple sequence repeat (SSR) markers. Results Polymorphic SSR markers revealed high genetic diversity. Primer SSR-15 amplified maximum of 8 fragments while 3 primers, namely, SSR-43, SSR-63, and AGA1 amplified minimum of 3 fragments. Collectively, 122 alleles were amplified in a range between 3 and 8 with an average of 5 alleles per marker. The size of the amplified alleles ranged between 90 and 680 base pairs. Polymorphism information content (PIC) value varied from a highest value of 0.499 in primer AGA1 to a lowest value of 0.231 in primer SSR-63 with a mean value of 0.376 showing considerable SSR polymorphism. Dendrogram developed on the basis of Jaccard’s similarity coefficient and neighbor-joining tree segregated all the studied Asparagus species into two discrete groups. Structure analysis based on Bayesian clustering allocated different accessions to two independent clusters and exhibited low level of individual admixture. Conclusions The genetic diversity analysis showed a conservative genetic background for maximum species of asparagus. Only Accessions of Asparagus adscendens were split into two diverse clusters suggesting a wide genetic base of this species as compared to other species. Overall genetic diversity was high, and this germplasm of Asparagus can be used in future improvement programs. The findings of current research on Asparagus germplasm can make a momentous contribution to initiatives of interbreeding, conservation, and improvement of Asparagus in future.

Abstract:A study was undertaken during the rabi season (October, 2021 to March, 2022) at the crop research center Sardar Vallabhbhai Patel University of Agriculture and Technology Meerut, Uttar Pradesh, India to find out the genetic divergence in 40 genotypes of Indian mustard using the D2 statistical approach. Forty (40) diverse genotypes of Indian mustard were examined for genetic divergence using a randomized complete block design (RCBD) with three replications. Observations were recorded on various 14 characters viz; days to 50% flowering, days to maturity, plant height (cm), number of primary branches plant-1, number of secondary branches plant-1, siliquae on main axis, length of main axis (cm), siliqua length (cm), number of seeds siliqua-1, 1000 seed weight (g), biological yield plant-1 (g), harvest index (%), oil content (%) and seed yield plant-1 (g). Mahalanobis D2 statistics were applied to the analysis of genetic divergence. Based on D2 values 40 genotypes were grouped into 5 clusters, maximum genotypes eleven obtained in cluster IV after that cluster I with nine genotypes, cluster V with eight genotypes, cluster III with seven genotypes and cluster II with five genotypes. The maximum intra-cluster was in cluster IV. The maximum inter-cluster distance was revealed between cluster II and V. It is clearly indicated that the genotypes included in this cluster are having a wide spectrum of genetic diversity. According to the role of many traits under examination in the manifestation of genetic divergence, plant height had the biggest contribution to divergence.

Abstract:Cannabis has been used as a source of nutrition, medicine, and fiber. However, lack of genomic simple sequence repeat (SSR) markers had limited the genetic research on Cannabis species. In the present study, 92,409 motifs were identified, and 63,707 complementary SSR primer pairs were developed. The most abundant SSR motifs had six repeat units (36.60%). The most abundant type of motif was dinucleotides (70.90%), followed by trinucleotides, tetranucleotides, and pentanucleotides. We randomly selected 80 pairs of genomic SSR markers, of which 69 (86.25%) were amplified successfully; 59 (73.75%) of these were polymorphic. Genetic diversity and population structure were estimated using the 59 (72 loci) validated polymorphic SSRs and three phenotypic markers. Three hundred ten alleles were identified, and the major allele frequency ranged from 0.26 to 0.85 (average: 0.56), Nei's genetic diversity ranged from 0.28 to 0.82 (average: 0.56), and the expected heterozygosity ranged from 0.28 to 0.81 (average: 0.56). The polymorphism information content ranged from 0.25 to 0.79 (average: 0.50), the observed number of alleles ranged from 2 to 8 (average: 4.13), and the effective number of alleles ranged from 0.28 to 0.81 (average: 0.5). The Cannabis population did not show mutation-drift equilibrium following analysis via the infinite allele model. A cluster analysis was performed using the unweighted pair group method using arithmetic means based on genetic distances. Population structure analysis was used to divide the germplasms into two subgroups. These results provide guidance for the molecular breeding and further investigation of Cannabis.

Abstract:Turmeric, a globally cultivated spice, holds significance in medicine, and cosmetics, and is also a very popular ingredient in South Asian cuisine. A study involving 53 turmeric genotypes evaluated for rhizome yield and related traits at Spices Research Center, Bogura, Bangladesh over three years (2019–22). A randomized complete block design was followed with two replications. ANOVA revealed significant trait variations among genotypes. Genotype T0015 emerged as the highest yielder at 28.04 t/ha. High heritability (0.58–0.99) and genetic advance characterized plant height (PH), mother rhizome weight (WMR), primary and secondary finger weights (WPF and WSF), and yield per plant (YPP) across seasons. Genetic gain (GG) was prominent in these traits. Genotypic and phenotypic coefficient variations (GCV and PCV) (6.24–89.46 and 8.18–90.88, respectively) across three years highlighted mother rhizome weight's importance followed by numbers of primary finger (NPF), and WPF. Positive and significant correlations, especially with PH, WMR, WPF, and YPP, emphasized their relevance to fresh yield (FY). Multiple linear regression identified PH, number of mother rhizome (NMR) and WMR as key contributors, explaining 37–79% of FY variability. Cluster analysis grouped genotypes into five clusters with maximum distance observed between clusters II and III. The geometric adaptability index (GAI) assessed adaptability and superiority, revealing nine genotypes outperforming the best existing cultivar. Genotype T0117 as the top performer based on GAI, followed by T0103 and T0094. Mean rank analysis favoured T0121 as the best performer, succeeded by T0117, T0082 and T0106. The top ten genotypes (T0015, T0061, T0082, T0085, T0094, T0103, T0106, T0117, T0121 and T0129) were identified as superior based on yield and overall ranking, warranting further evaluation. These findings may induce a window for improving turmeric research and ultimately play a role in enhancing its cultivation and productivity.
