Abstract:Acoustic levitation can provide significant benefits for many fundamental research questions. However, it is important to consider that the acoustic field influences the measurement environment. This work focuses on the dissolution of immobilised drops using acoustic levitation in liquid–liquid systems. Previous work demonstrated that the acoustic field of standing waves impacts mass transfer by affecting the spread of dissolved substances in the continuous phase in two distinct ways: (I) solutes may either pass through nodal planes of the standing waves or (II) not pass. The binary systems examined for case (I) are 1-hexanol–water and 1-butanol–water, and for case (II), n-butyl acetate–water and toluene–water. This work quantifies the intensification effect of acoustic levitation on dissolution for the two types of behaviour, by comparing them with reference measurements of mechanically attached dissolving drops. The system was designed to ensure minimal intensification. The minimum intensification of mass transfer for levitating drops in the used setup of case (I) was 25%, and for case (II), it was 65%, both increasing with decreasing surface-equivalent diameter. With this understanding, acoustic levitation can be used more accurately in the field of mass transfer studies.

Dialysate to plasma solute concentration (D/P) versus peritoneal transport parameters in CAPD.

Abstract:The relationship between dialysate to plasma solute equilibration ratio (D/P) and diffusive (diffusive mass transport coefficients, KBD) as well as convective (sieving coefficient, S, and net ultrafiltration) transport characteristics were studied in clinically stable CAPD patients and in patients with loss of ultrafiltration capacity (UFC). Forty-one 6-h single-dwell studies with standard glucose-based dialysis fluids containing 1.36% (n = 9), 2.27% (n = 9), and 3.86% (n = 23) anhydrous glucose were carried out in 33 clinically stable CAPD patients. Eleven patients with loss of UFC were studied with 3.86% glucose solution. Intraperitoneal dialysate volumes were calculated from the dilution of the tracer (131I-albumin) with a correction applied for its elimination from the peritoneal cavity. KBD and S were estimated using the Pyle-Popovich-Moncrief model with aqueous plasma concentrations. A theoretical D/P curve was derived with and without taking convective transport and peritoneal reabsorption into account. The three different glucose solutions yielded D/P curves which were similar for urea and potassium. For creatinine a slower equilibration and for glucose and sodium a faster decrease in dialysate concentration were observed with more hypertonic solutions. In general, there was a strong correlation which was best at 240 min between D/P (for glucose dialysate/initial dialysate concentration, D/Do) and KBD for all solutes (except sodium), whereas the correlation between D/P and convective transport parameters was much weaker. KBD for creatinine (with 3.86% glucose solution) could be estimated (r = 0.98) from aqueous D/P using the experimental formula: creatinine KBD = -1.8 - ln (1 - D240/P)/0.1. Patients with loss of UFC due to increased diffusive transport (n = 8) could be discriminated from the clinically stable patients using KBD and D/P (or D/D0) for creatinine and glucose or D/P for sodium. However, patients with loss of UFC associated with increased peritoneal reabsorption (n = 2) could not be identified using these parameters. Theoretically derived D/P curves were in excellent agreement with measured D/P for 1.36% glucose solution and simulations were satisfactory also for the 2.27% and 3.86% solutions provided that the effect of convective transport was taken into account. The standardized peritoneal equilibration tests (PET) as proposed by Twardowski et al. [17] seems to be appropriately designed as regards duration of the dwell and the choice of glucose and creatinine as investigated solutes. Thus, PET can be recommended as a sensitive routine investigation for the monitoring of normal/abnormal peritoneal transport behaviour in peritoneal dialysis patients.
[Botulinum toxin for the treatment of pain syndromes].

Abstract:Although botulinum toxin (BoNT) is being used for therapeutic purposes for more than 20 years, the list of potential new indications continues to increase and includes various pain syndromes. The pain relief experienced by patients with dystonia and spasticity from intramuscular BoNT injections suggested that other chronic skeletal-muscles pain conditions may also benefit. BoNT inhibits the release of acetylcholine at the neuromuscular junction thereby reducing striatal muscle contractions and the proposed analgesic property was initially attributed to muscular relaxation. A specific analgesic BoNT effect is difficult to conclude from studies where pain is conditioned by other associated symptoms like dystonia, muscle contraction or spasticity. One alternative is to critically appraise clinical trials where BoNT was studied as the active intervention and pain evaluated as an outcome. From this analysis there is convincing evidence for the effectiveness of BoNT in the treatment of pain associated with cervical dystonia. For all other pain syndromes there have been relatively few, small sized, placebo-controlled studies (myofascial pain syndrome, chronic neck and low back pain, piriformis syndrome and fibromyalgia) and the results of these studies have been contradictory or non conclusive. To establish the analgesic properties of BoNT there is a need for appropriately designed, exploratory randomized controlled studies in well accepted human models of nociceptive or neuropathic pain. This does not exclude the subsequent need to conduct pragmatic trials to evaluate the effectiveness of BoNT in conditions where the improvement of pain or any associated clinical sign or symptom may be of clinical relevance.

Abstract:Mobility manipulation of liquid droplets is an important task of digital imicrofluidics. Acoustic levitation has revolutionised the contactless manipulation of liquid droplets for various applications. Acoustic levitation technique can be effectively used to manipulate droplets to obtain their coalescence. This paper reports a unique, versatile, and material-independent approach for the vertical coalescence of the droplets suspended in an acoustic levitator. The acoustic power of the levitator is carefully engineered to obtain vertical coalescence of two liquid droplets. Water, 20% and 40% glycerol–water solutions are used as the working liquids. The results of the experiments revealed three outcomes during the coalescence. The outcomes are analysed and discussed.

Abstract:Large droplet transport in a line-focussed acoustic manipulator in terms of maximum droplet size is achieved by employing a driving voltage control mechanism. The maximum volume of the transported droplets in the order of few microliters is thereby increased by three orders of magnitude compared to the constant voltage case, widening the application field of this method significantly. A drop-on-demand droplet generator is used to supply the liquid droplets into the system. The ejected sequence of picoliter-size droplets is guided along trajectories by the acoustic field and accumulates at the selected pressure node, merging into a single large droplet. Droplet movement is achieved by varying the reflector height. This also changes the intensity of the radiation pressure during droplet movement, which in turn could atomise the droplet. The acoustic force is adjusted by regulating the driving voltage of the actuator to keep the liquid droplet suspended in air and to prevent atomisation. In the herein presented levitation concept, liquids with a wide range of surface tension (water and tetradecane were tested) can be transported over distances of several mm. The aspect ratio of the droplet in the acoustic field is shown to be a good indicator for radiation pressure intensity and is kept between 1.1 and 1.4 during droplet transport. Despite certain limitations with volatile liquids, the presented acoustic levitator concept has the potential to expand the range of analytical characterisation and manipulation methods in applications ranging from chemistry and biology.

Atomisation of an acoustically levitated droplet: Experimental observations of a myriad of complex phenomenon

Abstract:We report the dynamics of a droplet levitated in a single-axis acoustic levitator. The deformation and atomization behavior of the droplet in the acoustic field exhibits a myriad of complex phenomena, in sequences of steps. These include the primary breakup of the droplet through stable levitation, deformation, sheet formation, and equatorial atomization, followed by secondary breakup which could be umbrella breakup, bag breakup, bubble breakup or multistage breakup depending on the initial size of the droplet. The visualization of the interfacial instabilities on the surface of the liquid sheet using both side and top-view imaging is presented. The primary breakup of the droplet precedes with a stable levitation of the droplet. The thinning of the sheet is caused by the differential acceleration induced by the greater pressure difference between the poles and the equator. As the sheet thickness reduces to the order of a few microns, Faraday waves develop at the thinnest region (preceding to rim) which causes the generation of tiny-sized droplets perpendicular to the sheet. The corresponding, hole formation results in a perforated sheet that causes the detachment of the annular rim which breaks due to Rayleigh Plateau (RP) instability. The radial ligaments generated in the sheet possibly due to Rayleigh Taylor (RT) instability break into droplets of different sizes. The secondary breakup exhibits We dependency and includes umbrella, bag, bubble, or multistage, ultimately resulting in complete atomization of the droplets. Both the primary and the secondary breakup of the droplet admit interfacial instabilities such as Faraday instability, Kelvin Helmholtz (KH) instability, RT instability, and RP instability and are well described with visual evidence.

Complex Dynamics of an Acoustically Levitated Fluid Droplet Captured by a Low-Order Immersed Boundary Method

Abstract:We present a novel immersed boundary method that implements acoustic perturbation theory to model an acoustically levitated droplet. Instead of resolving sound waves numerically, our hybrid method solves acoustic scattering semi-analytically and models the corresponding time-averaged acoustic forces on the droplet. This framework allows the droplet to be simulated on inertial timescales of interest, and thereby admit a much larger time resolution than traditional compressible flow solvers. To benchmark this technique and demonstrate its utility, we implement the hybrid IBM for a single droplet in a standing wave. Simulated droplet shape deformations and streaming profile agree with theoretical predictions. Our simulations also yield new insights on the streaming profiles for elliptical droplets, for which a comprehensive analytic solution does not exist.

Abstract:As a ground-based experimental method for simulating the containerless state in space, acoustic levitation provides excellent containerless and contact-free conditions for the study of droplet dynamics, including droplet evaporation and phase separation. Meanwhile, the nonlinear effects of the acoustic field, such as acoustic radiation pressure and acoustic streaming, bring novel characteristics to the processes of droplet evaporation and phase separation. In this work, the evaporation and phase separation of aqueous two-phase-system (ATPS) droplets composed of polyethylene glycol (PEG) and ammonium sulfate (AMS) were investigated by a single-axis acoustic levitator, combined with image acquisition and processing technique. During the evaporation of the ATPS droplet, the square of its equatorial diameter, d 2 , decreases linearly with time, and its aspect ratio, γ , increases linearly with time. The PEG-AMS droplets initially in the single-phase regime can cross into the two-phase regime as the water evaporates, resulting in phase separation. The phase separation of the acoustically levitated PEG-AMS ATPS droplets includes three stages: firstly, a large number of PEG-rich globules formed inside the ATPS droplets, and then these PEG-rich globules collided, coagulated and migrated outward, and finally a horizontal layered structure of the whole droplet came into being. The evaporation constant, and the evolution of the PEG-rich globules and the AMS-rich phase area, were analyzed for ATPS droplets with different initial aspect ratios and different initial compositions. It is concluded that the greater the initial aspect ratio and the smaller the volume fraction of the PEG-rich phase, the faster the evaporation rate of the droplet; the greater the initial aspect ratio and the lager the volume fraction of the PEG-rich phase, the faster the phase separation. Numerical simulations show that the acoustically levitated droplets with a large aspect ratio are subjected to greater acoustic radiation pressure on the surface, and the corresponding sound field is more intense and the acoustic streaming is stronger, which accelerates the evaporation and phase separation of the levitated droplets. These findings help to deepen our understanding of the motion characteristics, evaporation dynamics and phase separation of acoustically levitated droplets, and provide a basis for containerless preparation and processing of materials under acoustic levitation.

Abstract:Ultrasonic atomization has been studied and used in applications for decades, but a mechanistic understanding of droplet production does not exist at a quantitative level. Early work seemed to indicate that droplet size follows from the driving frequency according to Kelvin's equation. The underlying assumptions in this model do not hold at larger frequencies, and the model relies on a fitting parameter that has not been consistent across experimental studies. Additionally, modern experiments often indicate multi-modal droplet size distributions further complicating the physics. It is difficult to experimentally measure relevant quantities like capillary wavelength because of the time and length scales involved and the fact that the phenomenon occurs at a free liquid surface. We use a high speed digital holographic microscope, the only one of its kind in the world, to image the liquid surface—in full 3D and real time—fast enough to resolve capillary waves generated by 5–50 MHz ultrasound. We extract from this data the spectrum of capillary wavelengths and then compare these to droplet size distributions measured with laser diffraction particle sizing. Both thickness mode and surface acoustic wave lithium niobate transducers are included as well as a range of frequencies in each case. It is shown that droplet size does depend directly on capillary wavelength, but that capillary wavelength is not related to the driving frequency in a simple way.

Abstract:Two family are described affected by a mild myotonic syndrome, characterized by almost exclusively presence of myotonic phenomenon, in absence of symptoms as muscular weakness, hypotrophy or hypertrophy ("myotonia levior"). The nosographic classification of this disease in the congenital myotonias is today uncertain, and it could be a case of a low expressivity variant of the dominant form (Thomsen disease), or the heterozygotic phenotype of the recessive form (Becker disease). Family history, clinical and electrophysiological data obtained in the patients seem to support first hypothesis, providing an useful element for a correct genetic counselling in these cases.

Experimental: A system consisting of 12-hydroxysteric acid (HSA) soaps mixed with different ratios of monoethanolamine (MEA) and choline hydroxide, was prepared. Microliter droplets of the samples were acoustically levitated and monitored with a camera, while exposed to CO2 to modify the pH through diffusion at the air-liquid interface and inside the droplet. The phase transition and water mobility in the levitated droplets were evaluated through X-ray scattering (SAXS/WAXS) and magnetic resonance studies, in real-time. Finally, the droplets were collected and examined under the microscope. Findings: The introduction of CO2 gas induced a phase transition from micelles to multi-lamellar tubes, resulting in a gel-like behavior both in the bulk and at the interface. The high stability of the acoustic levitator allowed the investigation of this dynamic phenomenon, in real-time, in a contact-free environment. This study showcases the suitability of acoustic levitation as a tool to investigate complex chemical processes at interfaces.

Abstract:A digital holographic particle tracking velocimetry system is applied to quantitatively study the drop atomization induced by capillary waves, and the breakup caused by increased sound pressure levels. A wavelet-based algorithm is used for particle detection and autofocusing with a wide size range of 20 mu m-2 mm. To eliminate the influence of large particles on small particles, a two-step detection method is adopted. Large drops are first characterized and simulated by a diffraction-based model. Then the contributions of the drops are subtracted from the original hologram followed by the detection of small droplets. Finally, the velocity and size distribution of the secondary droplets are obtained from the experimental holograms. The results demonstrate the validity of the digital in-line holographic technique for the atomization and breakup study of acoustically levitated drops. (C) 2014 Optical Society of America
Ultrasonic spectroscopy of sessile droplets coupled to optomechanical sensors

Abstract:We describe a system for interrogating the acoustic properties of sub-nanoliter liquid samples within an open microfluidics platform. Sessile droplets were deposited onto integrated optomechanical sensors, which possess ambient-medium-noise-limited sensitivity and can thus passively sense the thermally driven acoustic spectrum of the droplets. The droplet acoustic breathing modes manifest as resonant features in the thermomechanical noise spectrum of the sensor, in some cases hybridized with the sensor's own vibrational modes. Excellent agreement is found between experimental observations and theoretical predictions, over the entire ~ 0 - 40 MHz operating range of our sensors. With suitable control over droplet size and morphology, this technique has the potential for precision acoustic sensing of small-volume biological and chemical samples.

Suspension Dynamics of Droplets in Acoustic and Gravitational Fields

Abstract:In the field of acoustic suspension or levitation of droplets against gravity, the application of Gorkov's acoustic radiation force for small particles (within the Rayleigh limit) or its extensions to larger ones (beyond the Rayleigh limit) is limited to predicting the suspension position of the droplet. Since this approach treats the droplet as a rigid particle, it fails to capture the fluid dynamics of the droplet and is also unsuitable for studying interfacial phenomena such as droplet deformation, splitting, or coalescence. In this work, we employ our recently developed acoustic body force in Eulerian form, which models the droplet as a fluid, to theoretically investigate the suspension dynamics of droplet subjected to standing waves through the interaction between acoustic, interfacial, and gravitational forces. Our theory predicts that when interfacial forces are dominant, the presence of positive and negative acoustic force regions within droplets exceeding the Rayleigh limit reduces the net acoustic force counteracting gravity. As a result, the suspension dynamics become highly dependent on droplet size, in contrast to droplets within the Rayleigh limit, where the dynamics remain size-independent. Thus, beyond the Rayleigh limit, as the droplet size to wavelength ratio increases, the critical acoustic energy density ($E_{cr}$) required to suspend the droplet initially rises sharply, which agrees with recent experimental results. After $E_{cr}$ reaches a local maximum at $d/\lambda \approx 0.65$, it exhibits a pattern of alternating decreases and increases, with each successive peak surpassing the previous one. Remarkably, our study reveals a size-dependent shifting of the suspension position between nodes and antinodes for droplets beyond the Rayleigh limit, whereas droplets within this limit maintain a consistent suspension position regardless of size.

Abstract:Evaporation of cyclohexane and ethanol binary drops under acoustic levitation was investigated. The aim was to understand the effect of acoustic levitation on the evaporation dynamics and involved physical processes of the binary drops. We report the occurrence of liquid-phase separation of the binary drop during evaporation under acoustic levitation. Through systematic experiments, it was found that the enhanced evaporation rate of the drop under acoustic levitation led to a significant temperature decrease. In addition, driven by external acoustic streaming, water vapor tended to be enriched and condensed on the drop surface. Because ethanol is extremely soluble in water, tiny cyclohexane droplets were extracted from ethanol owing to water condensation. In addition, driven by the internal flow of the acoustically levitated drop, the extracted cyclohexane droplets coalesced, eventually resulting in macro-segregation in the drop. These findings provide new insights into the evaporation dynamics of acoustically levitated drops, thus, shedding light on industrial purification and separation of volatile liquids with opposite water solubilities, such as cyclohexane and ethanol.

Stereo Measurement of Internal Flow Structure in Volatile Droplets on the Acoustic Levitation

Abstract:This paper describes experimental investigation on the three-dimensional internal flow structures of the acoustic levitated volatile droplets by stereo PIV. The acoustic levitation is one of the effective techniques to levitate and manipulate droplets in air. In order to apply the acoustic levitation to various fields such as the material science, analytical chemistry, drug discovery and so on, it is essential to understand the flow structures and transport phenomena on the acoustically levitated droplet. Our observation shows the internal flow structure is changed with the increase of the saturated vapor pressure of the levitated droplets. In the case of decane, nonane, octane, heptane, hexane and pentane droplets, the rotational motion is generated in each droplet. In the case of ethanol and methanol droplets, there is the flow from the equator toward the center of the droplet in the horizontal plane. In addition, the two or four vortices are generated in the horizontal plane with the increase of the droplet diameter.

Droplet displacement, internal mixing or drop splitting, are amongst the elementary operations that SAW can achieve, which are useful on lab-on-chip microfluidics benches. On the purpose to understand the underlying physical mechanisms involved during these operations, we study experimentally the droplet dynamics varying different physical parameters. Here in particular, the influence of liquid viscosity and acoustic frequency is investigated: it is indeed predicted that both quantities should play a role in the acoustic-hydrodynamic coupling involved in the dynamics. The key point is to compare the relative magnitude of the attenuation length, i.e. the scale within which the acoustic wave decays in the fluid, and the size of the drop. This relative magnitude governs the relative importance of acoustic streaming and acoustic radiation pressure, which are both involved in the droplet dynamics. Direct measurement of forces in air-based acoustic levitation systems

Abstract:Acoustic levitation is frequently used for non-contact manipulation of objects and to study the impact of microgravity on physical and biological processes. While the force field produced by sound pressure lifts particles against gravity (primary acoustic force), multiple levitating objects in the same acoustic cavity interact via forces that arise from scattered sound (secondary acoustic forces). Current experimental techniques for obtaining these force fields are not well-suited for mapping the primary force field at high spatial resolution and cannot directly measure the secondary scattering force. Here we introduce a method that can measure both acoustic forces in situ, including secondary forces in the near-field limit between arbitrarily shaped, closely spaced objects. Operating similarly to an atomic force microscope, the method inserts into the acoustic cavity a suitably shaped probe tip at the end of a long, flexible cantilever and optically detects its deflection. This makes it possible to measure forces with a resolution better than 50 nN, and also to apply stress or strain in a controlled manner to manipulate levitated objects. We demonstrate this by extracting the acoustic potential present in a levitation cavity, directly measuring the acoustic scattering force between two objects, and applying tension to a levitated granular raft of acoustically-bound particles in order to obtain the force-displacement curve for its deformation.
