Abstract:High performance liquid chromatography-mass spectrometry is one of the most commonly used strategies for lipid analysis. The development of versatile chromatographic stationary phases to meet the increasing demands for separation of complex lipids is very important. Styrene-maleic acid (SMA) copolymer is an amphiphilic polymer, which has been proven to have the ability to solubilize lipid molecules of various structures. In this study, styrene-maleic anhydride copolymer coated silica was first prepared by the thiol-ene click reaction. With L-cysteine hydrochloride or dodecanol as the post-modification reagents, Sil-SMA-amino acid and Sil-SMA-dodecanol stationary phase materials were further successfully fabricated via nucleophilic ring-opening reaction. The Fourier-transform infrared, thermogravimetric analysis, and elemental analysis results confirmed the two stationary phase materials were successfully prepared. Furthermore, both the Sil-SMA-dodecanol column and the Sil-SMA-amino acid column possessed reversed-phase/hydrophilic interaction/ion exchange mixed-mode retention mechanisms. The column efficiency of the Sil-SMA-derivatives columns reached 77,300 N/m. Based on the mixed-mode retention characteristics, the Sil-SMA-derivatives columns achieved both the lipid classes and species separation via a single column. The Sil-SMA-amino acid column was further successfully used to separate lipid extract from gastric cancer cell membrane. All these results demonstrated that the SMA-based stationary phase materials have a good potential for use in lipid separation.

Abstract:Styrene-maleic acid (SMA) copolymer has received much attention for its excellent solubilization characteristics. In this work, SMA copolymer brush-based chromatographic stationary phases were exploited and developed for the first time. First, SMA copolymer brush was in situ grown on the surface of spherical silica via living/controlled reversible addition-fragmentation chain transfer (RAFT) polymerization method. Subsequently, as a proof-of-concept demonstration, the copolymer was esterified by diethylene glycol mono-2-ethylhexyl ether (DGME) and 2-(2-ethylhexyloxy) ethanol (EHOE), respectively. The obtained Sil-SMA-DGME and Sil-SMA-EHOE copolymer-brush chromatographic stationary phases were characterized by transmission electron microscopy, Fourier transform infrared spectrometer, X-ray photoelectron spectroscopy, and thermogravimetric analysis, respectively. The chromatographic retention mechanism indicated that both the two packed columns exhibited hydrophilic/reverse mixed-mode retention modes. The maximum column efficiency was up to 71,000 N/m. The chromatographic separation performance evaluation indicated that the novel kind of stationary phases had excellent separation capabilities for hydrophilic, hydrophobic compounds and phospholipid standards. In addition, by combination with mass spectrometry identification, the Sil-SMA-DGME column was further exploited for separation and identification of phospholipids in human lung cancer cells. Totally, 9 classes including 186 phospholipid species were successfully identified. The results demonstrated the promising application prospects of the novel kind of SMA copolymer-brush chromatographic stationary phases.
Polymerized phosphonium ionic liquid functionalized silica microspheres as mixed-mode stationary phase for liquid chromatographic separation of phospholipids

Abstract:There is a large and growing demand for the vigorous development of new high performance liquid chromatography stationary phases in order to solve complex phospholipids separation. Herein, phosphonium-based ionic liquid trioctyl(allyl)phosphonium bromide ([P888Allyl]Br) was first synthesized with trioctylphosphine and allyl bromide. With [P888Allyl]Br as the polymerizable monomer, polymerized phosphonium ionic liquid functionalized silica microsphere (PIL@SiO2) was further synthetized via click chemistry reaction. Significantly, based on the inherent amphiphilic nature of the introduced [P888Allyl]Br, the packed PIL@SiO2 column displayed hydrophilic/hydrophobic mixed-mode retention mechanisms. The PIL@SiO2 column can achieve separation of nucleic acid bases and nucleosides, sulfonamides, amides and anilines with excellent selectivity in a shorter separation time. The column efficiency reached 109,700 N/m for 2-iodoacetamide. One of the important characteristics of the PIL@SiO2 column is that both phospholipid classes and species can be efficiently separated via the same column, outperforming that of the commercial amino column. Furthermore, the application potential of the PIL@SiO2 column was further verified via separation of phospholipids extracted from soy lecithin. The proposed PIL@SiO2 column provides a promising candidate for separation of complex phospholipid samples.

[One-pot synthesis of a poly(styrene-acrylic acid) copolymer-modified silica stationary phase and its applications in mixed-mode liquid chromatography]

Abstract:As modified ligands with a wide range of sources, abundant functional groups, and good biocompatibility, polymers have been widely used in the development of silica-based chromatographic stationary phases. In this study, a poly(styrene-acrylic acid) copolymer-modified silica stationary phase (SiO2@P(St-b-AA)) was prepared via one-pot free-radical polymerization. In this stationary phase, styrene and acrylic acid were used as functional repeating units for polymerization and vinyltrimethoxylsilane (VTMS) was used as a silane coupling agent to link the copolymer and silica. Various characterization methods, such as Fourier transform infrared (FT-IR) spectroscopy, thermogravimetric analysis (TGA), scanning electron microscopy (SEM), N2 adsorption-desorption analysis, and Zeta potential analysis, confirmed the successful preparation of the SiO2@P(St-b-AA) stationary phase, which had a well-maintained uniform spherical and mesoporous structure. The retention mechanisms and separation performance of the SiO2@P(St-b-AA) stationary phase in multiple separation modes were then evaluated. Hydrophobic and hydrophilic analytes as well as ionic compounds were selected as probes for different separation modes, and changes in the retention of the analytes under various chromatographic conditions, including different methanol or acetonitrile contents and buffer pH values, were investigated. In reversed-phase liquid chromatography (RPLC) mode, the retention factors of alkyl benzenes and polycyclic aromatic hydrocarbons (PAHs) on the stationary phase decreased with increasing methanol content in the mobile phase. This finding could be attributed to the hydrophobic and π-π interactions between the benzene ring and analytes. The retention changes of alkyl benzenes and PAHs revealed that the SiO2@P(St-b-AA) stationary phase, similar to the C18 stationary phase, exhibited a typical reversed-phase retention behavior. In hydrophilic interaction liquid chromatography (HILIC) mode, as the acetonitrile content increased, the retention factors of hydrophilic analytes gradually increased, and a typical hydrophilic interaction retention mechanism was inferred. In addition to hydrophilic interaction, the stationary phase also demonstrated hydrogen-bonding and electrostatic interactions with the analytes. Compared with the C18 and Amide stationary phases prepared by our groups, the SiO2@P(St-b-AA) stationary phase exhibited excellent separation performance for the model analytes in the RPLC and HILIC modes. Owing to the presence of charged carboxylic acid groups in the SiO2@P(St-b-AA) stationary phase, exploring its retention mechanism in ionic exchange chromatography (IEC) mode is of great importance. The effect of the mobile phase pH on the retention time of organic bases and acids was further studied to explore the electrostatic interaction between the stationary phase and charged analytes. The results revealed that the stationary phase has weak cation exchange ability toward organic bases and electrostatically repels organic acids. Moreover, the retention of organic bases and acids on the stationary phase was influenced by the analyte structure and mobile phase. Thus, the SiO2@P(St-b-AA) stationary phase could provide multiple interactions, as demonstrated by the separation modes described above. The SiO2@P(St-b-AA) stationary phase showed excellent performance and reproducibility in the separation of mixed samples with different polar components, indicating that it has promising application potential in mixed-mode liquid chromatography. Further investigation of the proposed method confirmed its repeatability and stability. In summary, this study not only described a novel stationary phase that could be used in RPLC, HILIC, and IEC modes but also presented a facile "one-pot" preparation approach that could provide a new route for the development of novel polymer-modified silica stationary phases.
[Prosthetic replacement of condylar head for management of traumatic temporomandibular joint ankylosis].

Abstract:OBJECTIVE To preliminarily evaluate the method of using titanium condylar protheses in management of traumatic temporomandibular joint ankylosis. METHODS Between August 2006 and April 2008, titanium condylar replacement was performed in 5 male patients. Preoperative CT datum of patient was obtained and a resin model was then produced and used as a reference to select a suitable titanium condyle for reconstruction of the joint. In the operation, the bony ankylosis was removed. According to the preoperation planning, the selected prosthesis was inserted and fixed to the distal mandibular segment. The results were assessed by means of examinations and measurements postoperatively and during follow-up. RESULTS The 5 patients received alloplastic replacement of 6 condyles. Three patients were under follow-up and follow-up time ranged from 8 months to 2 years and 6 months. The mean mouth opening was 30 mm. Two patients had good occlusion and one patient developed a slight open bite. CONCLUSIONS Prosthetic condylar replacement can provide a choice for management of traumatic temporomandibular joint ankylosis.

Abstract:As green, less toxic, and abundant ligands with rich functional groups, natural products are widely used in synthesis of chromatographic stationary phases. In this work, dodecyl imide maleopimaric acid glycidyl methacrylate ester (C12-MPAGN) was prepared from maleopimaric acid through the imidization and ring-opening based esterification reaction. By using "thiol-ene" click chemistry, it was chemically bonded to the silica and (3-mercaptopropyl) trimethoxysilane (γ-MPS) was used as the coupling agent to obtain dodecyl imide maleopimaric acid glycidyl methacrylate ester bonded silica stationary phase (Sil-C12-MPAGN). Fourier transform infrared spectroscopy (FT-IR), thermogravimetric analysis (TGA), scanning electron microscopies (SEM), and elemental analysis (EA) were utilized to verify that the Sil-C12-MPAGN stationary phase was successfully prepared with C12-MPAGN immobilized on the silica surface. In order to evaluate the chromatographic performance and retention mechanisms of the Sil-C12-MPAGN column and compared with C18 column, a variety of compounds were used, including stander mixture of Tanaka, alkylbenzenes, polycyclic aromatic hydrocarbons (PAHs), phenols and flavonoids. Based on these multiple interactions, including hydrophobic, hydrogen-bonding, and π-π interactions, high selectivity and superior separation performance were demonstrated by the Sil-C12-MPAGN column for probe molecules what had previously been mentioned. In addition, the quantitative determination of paclitaxel content in Yew bark extract was conducted with this column, which was found that the concentration was 83.67 mg/L, respectively. In short, the present study proposes a new strategy for introducing rosin to liquid chromatography with high selectivity and separation performance.

Abstract:Two new hydrophilic-hydrophobic and acid-base balanced liquid chromatographic stationary phases were obtained by using two long-chain monomers including undecylenic acid (UA) and oleic acid (OA) combined with short-chain 4-vinyl pyridine (Py) in-situ polymerized on silica microspheres surfaces, respectively. These two stationary phases can be used for reversed-phase liquid chromatography (RPLC) and hydrophilic interaction liquid chromatography (HILIC) modes. Hydrophilic, hydrophobic, acidic and basic analytes can be separated on two new stationary phases in RPLC or HILIC mode. Compared with commercial hydrophobic C18 columns and other HILIC columns, two new stationary phases have excellent selectivity and separation performance in the separation of 9 polycyclic aromatic hydrocarbons, 10 aromatic acids, 10 anilines and 5 oxazolidinones, respectively. This novel polymer-functionalized silica design strategy has opened up new ideas for the future of mixed-mode stationary phases with multiple functions and specificity.

Abstract:Streptococcus pyogenes is an important cause of human morbidity and mortality worldwide. A wealth of genomic information related to this pathogen has facilitated exploration of the proteome, particularly in response to environmental conditions thought to mimic various aspects of pathogenesis. Proteomic approaches are also used to identify immunoreactive proteins for vaccine development and to identify proteins that may induce autoimmunity. These studies have revealed new mechanisms involved in regulating the S. pyogenes proteome, which has opened up new avenues in the study of S. pyogenes pathogenesis. This article describes the methods used, and progress being made towards characterizing the S. pyogenes proteome, including studies seeking to identify potential vaccine candidates.

Abstract:Siliceous mesostructured cellular foam (MCF) with highly interconnected porous structure, ultralarge pore size and relatively uniform particle size (3–5μm) was prepared to achieve the mixed-mode and efficient separation of intact proteins. And molecular sieving effect for the first time played an important role in protein separation using mesoporous silica materials as HPLC stationary phase. The spherical silica particles were synthesized via hydrothermal method and the pore size was easily regulated by adding NH4F as well as altering the aging time. After aminopropyl derivatization, the chromatographic performance of functionalized mesoporous silica particles was investigated in comparison with those without modification and commercial NH2 column, and their mixed-mode retention mechanisms were investigated in detail. The superior separation performance for the retention of proteins was obtained on our home-made column in comparison with commercial NH2 column. The influences of aminopropyl derivatization and mobile phase composition on the column property were also investigated. Moreover, the home-made column showed similar performance for separation of polar anilines and neutral PAHs with the commercial column, owing to mixed-mode retention mechanisms including p–π stacking, electron interaction, hydrophobic effect, π–π EDA interaction and hydrogen bonding. All these results indicated that the aminopropyl modified MCF would be promising in the mixed-mode and efficient separation of biomolecules in addition with small molecules.
A composite hydrogel modified silica stationary phase for mixed‐mode liquid chromatography

Abstract:A novel composite hydrogel functionalized silica core-shell stationary phase was prepared by the surface modification of silica sphere. The successful synthesis of the new stationary phase (T-Sil@PAM/SA/UiO-66-NH 2 ) was proven by scanning electron microscope (SEM), transmission electron microscope (TEM), X-ray photoelectron spectroscopy (XPS) and X-ray diffraction (XRD), etc. Due to the coexistence of amide, hydroxyl, long carbon chain and UiO-66-NH 2 in composite hydrogel shell, the obtained stationary phase can be used in hydrophilic/reversed-phase liquid chromatography with multiple retention mechanisms, such as hydrophilic, hydrophobic and π - π interactions. The chromatographic retention behavior of T-Sil@PAM/SA/UiO-66-NH 2 demonstrated that the new stationary phase showed excellent separation performance for both polar analytes (such as alkaloids, saccharides, etc.) and nonpolar analytes (such as substituted benzene and polycyclic aromatic hydrocarbon (PAHs), etc.). Furthermore, compared with NH 2 column and commercial C18 column, the T-Sil@PAM/SA/UiO-66-NH 2 exhibited a certain superiority. Moreover, the relative standard deviation (RSD) of PAHs' retention time with eight replicates consecutive elution was found to range from 0.03% to 0.17%. Therefore, the successful use of T-Sil@PAM/SA/UiO-66-NH 2 in mixed‐mode liquid chromatography expanded the potential applications of hydrogels in the field of separation.

Abstract:Sections from 21 tumours diagnosed as primary or metastatic rhabdomyosarcoma were stained for alpha and beta enolase. The cases were subdivided into embryonal and alveolar subtypes (38% and 62%, respectively). Positive cytoplasmic staining for alpha enolase was seen in all but one case, and cytoplasmic staining for beta enolase was seen in some cells in 18 of the 21 cases (86% of the total, 88% of the alveolar subgroup, and 85% of the embryonal subgroup). No cells stained positively for beta enolase in the control series of neuroblastomas, fibrosarcomas, Wilms' sarcomas, and an osteosarcoma. The results show that beta enolase is a sensitive marker of muscular differentiation in rhabdomyosarcoma.
Preparation, characterization, and separation mechanism of a dehydroabietic-acid-based shape-selective chromatographic stationary phase 1

Abstract:Chromatographic stationary phases with molecular-shape selectivity are advantageous for the separation and analysis of geometric isomers. Herein, dehydroabietic acid is bonded on the surface of silica microspheres via 3-glycidoxypropyltrimethoxysilane to form a monolayer dehydroabietic-acid stationary phase (Si-DOMM) with a racket-shaped structure. Various characterization techniques indicate that Si-DOMM is successfully prepared, and the separation performance of a Si-DOMM column is evaluated. The stationary phase has a low silanol activity and metal contamination and a high hydrophobicity and shape selectivity. The resolutions of lycopene, lutein, and capsaicin on the Si-DOMM column confirm that the stationary phase exhibits high shape selectivity. The elution order of n -alkyl benzene on the Si-DOMM column indicates its high hydrophobic selectivity and suggests that the separation is an enthalpy-driven process. Repeatability experiments reveal highly stable preparation processes of the stationary phase and column and indicate that the relative standard deviations of retention time, peak height, and peak area are less than 0.26%, 3.54%, and 3.48%, respectively. Density functional theory calculations using n -alkylbenzenes, polycyclic aromatic hydrocarbons, amines, and phenols as model solutes provide an intuitive and quantitative description of the multiple retention mechanisms. The Si-DOMM stationary phase exhibits superior retention and high selectivity for these compounds via multiple interactions. The bonding phase of the monolayer dehydroabietic acid stationary phase with a racket-shaped structure has a unique affinity for benzene, strong shape selectivity, and good separation performance for geometrical isomers with different molecular shapes.

Abstract:High-performance liquid chromatography is one of the most important analytical tools for the identification and separation of substances. The efficiency of this method is largely determined by the stationary phase of the columns. Although monodisperse mesoporous silica microspheres (MPSM) represent a commonly used material as stationary phase their tailored preparation remains challenging. Here we report on the synthesis of four MPSMs via the hard template method. Silica nanoparticles (SNPs) which form the silica network of the final MPSMs were generated in situ from tetraethyl orthosilicate (TEOS) in the presence of (3-aminopropyl) triethoxysilane (APTES) functionalized p(GMA-co-EDMA) as hard template. Methanol, ethanol, 2-propanol, and 1-butanol were applied as solvents to control the size of the SNPs in the hybrid beads (HB). After calcination, MPSMs with different sizes, morphology and pore properties were obtained and characterized by scanning electron microscopy, nitrogen adsorption and desorption measurements, thermogravimetric analysis, solid state NMR and DRIFT IR spectroscopy. Interestingly, the 29Si NMR spectra of the HBs show T and Q group species which suggests that there is no covalent linkage between the SNPs and the template. The MPSMs were functionalized with trimethoxy (octadecyl) silane and used as stationary phases in reversed-phase chromatography to separate a mixture of eleven different amino acids. The separation characteristics of the MPSMs strongly depend on their morphology and pore properties which are controlled by the solvent during the preparation of the MPSMs. Overall, the separation behavior of the best phases is comparable with those of commercially available columns. The phases even achieve faster separation of the amino acids without loss of quality.

Abstract:In this study, a mixed mode stationary phase for capillary liquid chromatography (cLC) was prepared by chemical modification of glycol diglycidyl ether (EGDE) and γ-aminobutyric acid (GABA) bonded to the silica, named as Sil–EGDE–GABA. The Sil–EGDE–GABA was characterized by Fourier transform infrared spectroscopy and elemental analysis. The retention of nucleotide bases at different acetonitrile levels in the mobile phase indicated that the stationary phase possessed both hydrophilic chromatography (HILIC) and reversed-phase chromatography (RPLC) characteristics. Positional isomers, aniline compounds and a mixture of polar–nonpolar analytes were separated in RPLC mode. The stationary phase exhibited different separation mechanisms in the separation of positional isomers, compared with a commercial ODS column. Sulfonamides and biogenic amines were successfully separated in HILIC mode. These results demonstrated the application possibilities of the prepared Sil–EGDE–GABA as packing materials in cLC.

Abstract:Polymer microspheres have received attention because of their excellent properties. In this work, a polymer matrix zwitterionic amino acid stationary phase was prepared. Polyglycidyl methacrylate divinylbenzene (PGMA-DVB) microspheres was used as the matrix and successful modification with L-phenylalanine by ring-opening reaction of epoxy groups on the surface of PGMA-DVB microsphere matrix. The stationary phase was characterized by scanning electron microscopy, Fourier-transform infrared spectra, and elemental analysis. The phenylalanine-modified stationary phase was used in RPLC/HILIC mixed-mode chromatography for the separation of alkylbenzenes, polycyclic aromatic hydrocarbons, phenols, nucleosides and nucleic acids and etc. as probes, respectively. The good spatial selectivity of the stationary phase was demonstrated by the separation of biphenyl isomers. The prepared stationary phase showed good stability in alkaline condition (pH = 10) and low swelling in organic solvent mobile phase. Meanwhile, the stationary phase also showed good performance in the separation of different vitamins (Rs > 5.3), and the separation and detection of PAHs in river water.

Abstract:Transmembrane protein (TMP)-functionalized materials have resulted in powerful new methods in chemical analysis. Of particular interest is the development of high-throughput, TMP-functionalized stationary phases for affinity chromatography of complex mixtures of analytes. Several natural and synthetic phospholipids and lipid mimics have been used for TMP reconstitution, although the resulting membranes often lack the requisite chemical and temporal stability for long-term use, a problem that is exacerbated in flowing separation systems. Polymerizable lipids with markedly increased membrane stability and TMP functionality have been developed over the past two decades. More recently, these lipids have been incorporated into a range of analytical methods, including separation techniques, and are now poised to have a significant impact on TMP-based separations. Here, we describe current methods for preparing TMP-containing stationary phases and examine the potential utility of polymerizable lipids in TMP affinity chromatography.

Abstract:Zwitterions are a promising choice to prepare separation materials because of their hydrophilicity and biocompatibility. We described the preparation of two zwitterionic polymer functionalized stationary phases and evaluation under mixed-mode chromatography. A zwitterionic monomer, S-(4-vinylbenzyl) cysteine (SVC), was synthesized and bonded to silica via reversible addition fragmentation chain transfer (RAFT) polymerization to afford a zwitterionic stationary phase, Sil-SVC. A hydrophobic monomer, N-(4-phenylbutan-2-yl) acrylamide (NPA), was copolymerized with SVC onto the stationary phase (Sil-SVC-NPA) for comparison. The stationary phases were characterized with FT-IR, TGA, EA, and zeta-potential measurements. Mobile phase composition (ACN content, pH and salt concentration) was varied to study the retention property. Linear solvation energy relationship and Van't Hoff plot were used to investigate the retention mechanism and how chromatographic conditions influenced it. Both stationary phases showed a mixed-mode of RPLC/HILIC/IEC and satisfactory performance in separating hydrophobic analytes (alkylbenzenes and polycyclic aromatic hydrocarbons), hydrophilic nucleotide and bases, and anions, high column efficiency of 60000 plates · m −1 was achieved. In summary, zwitterionic polymers are attractive options to prepare stationary phases and the retention property can be easily regulated by copolymer.
