Abstract:Introduction: Soil salinization poses a worldwide challenge that hampers agricultural productivity. Methods: Employing high-throughput sequencing technology, we conducted an investigation to examine the impact of compost on the diversity of bacterial communities in saline soils. Our study focused on exploring the diversity of bacterial communities in the inter-root soil of plants following composting and the subsequent addition of compost to saline soils. Results: Compared to the initial composting stage, Alpha diversity results showed a greater diversity of bacteria during the rot stage. The germination index reaches 90% and the compost reaches maturity. The main bacterial genera in compost maturation stage are Flavobacterium , Saccharomonospora , Luteimonas and Streptomyces . Proteobacteria, Firmicutes, and Actinobacteria were the dominant phyla in the soil after the addition of compost. The application of compost has increased the abundance of Actinobacteria and Chloroflexi by 7.6 and 6.6%, respectively, but decreased the abundance of Firmicutes from 25.12 to 18.77%. Redundancy analysis revealed that soil factors pH, solid urease, organic matter, and total nitrogen were closely related to bacterial communities. Discussion: The addition of compost effectively reduced soil pH and increased soil enzyme activity and organic matter content. An analysis of this study provides theoretical support for compost's use as a saline soil amendment.

Abstract:The influence of bio-compost on the diversity, composition and structure of soil microbial communities is less understood. Here, Illumina MiSeq sequencing and a network analysis were used to comprehensively characterize the effects of 25 years of bio-compost application on the microbial diversity of soil and community composition. High dosages of bio-compost significantly increased the bacterial and fungal richness. The compositions of bacterial and fungal communities were significantly altered by bio-compost addition. Bio-compost addition enriched the relative abundance of beneficial microorganisms (such as Sphingomonas, Acidibacter, Nocardioides, etc.) and reduced the relative abundance of harmful microorganisms (such as Stachybotrys and Aspergillus). Electrical conductivity, soil organic matter and total phosphorus were the key factors in shaping soil microbial community composition. The bacterial network was more complex than fungal network, and bacteria were more sensitive to changes in environmental factors than fungi. Positive interactions dominated both the bacterial and fungal networks, with stronger positive interactions found in the bacterial network. Functional prediction suggested that bio-composts altered the soil bacterial-community metabolic function with respect to carbon, nitrogen and phosphorus cycles and fungal community trophic modes. In conclusion, suitable bio-compost addition is beneficial to the improvement of soil health and crop quality and therefore the sustainability of agriculture.

Remediation of organic amendments on soil salinization: Focusing on the relationship between soil salts and microbial communities

Abstract:Soil salinization has emerged as a major factor with an adverse influence on agricultural green development worldwide. It is necessary to develop high-efficiency and ecologically beneficial management measures for alleviating soil salinization. The experiment of application for cow manure (CM), biochar (BC), and bio-organic fertilizer (BIO) in soil with light salinity was conducted to investigate the remediation of organic materials on soil salinization with melon (Cucumis melo L.) by reducing the availability of saline ions and shifting the soil microbial community. Results showed that BC treatment significantly decreased the EC values of the soil and soil solution by 19.23% and 27.02% and the concentrations of Na+, K+, and Cl- by 13.28%, 13.08%, and 15.21%, respectively, followed by CM and BIO treatments. High-throughput sequencing identified that organic amendments significantly improved the richness of the soil bacterial community and increased the relative abundances of Acidobacteria and Firmicutes by 33.11% and 111.2%, respectively, and the beneficial salt-tolerant bacterial genera Flavobacterium, Bacillus and Arthrobacter by 32.04%, 38.92% and 35.67%, respectively. Additionally, soil Na+, Ca2+, K+ and Cl- were significantly negatively correlated with Acidobacteria and Flavobacterium and were also the most important factors driving the changes in the structure of the soil bacterial communities. The bacterial networks were more complex in the organic amendments treatments than in CK, reflecting through more nodes and links and a higher average clustering coefficient, density and modularity. This study provided a comprehensive understanding of the application of organic amendments in alleviating soil salinization and improving soil bacterial and fungal communities and provides scientific support for agriculture green development.

Abstract:The application of modified biochar has been proven to be a novel and promising strategy to improve saline-alkali soil. However, the effect of iron-modified biochar (FB) on the chemical properties of saline-alkali soil at different depths remains unclear. Therefore, we designed a soil column that divides the soil into three consecutive parts to explore the amelioration effect of iron-modified biochar on saline-alkali soil chemical properties and the response of bacterial communities along a depth gradient. The results showed that soil chemical properties were significantly improved with 1% FB application, while the amelioration effect of FB was different between topsoil and subsoil. A significant increment in activities of extracellular enzymes in topsoil and decrease in base cations in subsoil were observed in 1% FB treatment. Moreover, the abundances of halophilic taxa were higher in subsoil than in topsoil, especially for the phyla Bacteroidetes and Deinococcus. Furthermore, the abundance of beneficial bacteria (e.g., Alphaproteobacteria, Sphingomonas, and Pontibacter) in saline-alkali soil increased at 1% FB. Our results suggest the influence of iron-modified biochar on soil properties and bacterial communities along a soil depth gradient, providing a novel strategy for improving saline-alkali soil with biochar.

Abstract:Reclamation of saline soil plays an important role in supporting high population growth in China. To evaluate the effects of reclamation and sustainability of salt-affected land production, soil chemical properties, enzyme activities, microbial community structure and function in sites reclaimed in 1976, 1984 and 1996 were characterized. The 2009 site was left to succession fallow and chosen as a control site. Results showed that electrical conductivity (EC) and pH decreased rapidly after the soil forming process started. An accumulation of soil organic C as a result of farming was observed. In all sites, inorganic N and available P were increased within 33 years after reclamation and commencement of agriculture. As a result of reclamation, soil enzyme activity (β-glucosidase, phosphatase, urease and arylsulfatase) was increased. There were no significant differences in soil enzyme activity between the sites reclaimed in 1976 and 1984, with the exception of phosphatase. Carbon source utilization patterns were less diverse in control soil than in treatment sites. The activities of reclamation resulted in synthesis of new phospholipid fatty acids (14:0, 17:1 c9, 16:0 2OH, 17:0 10Me, i17:0, 20:4 ω6c). Principal component analyses showed that the sites reclaimed in 1976 and 1984 clustered together and were distinct from 1996. Taken together, reclamation showed significantly increased soil quality and microbial activity. EC was the main limiting soil quality characteristic, which showed a comparative steady state after a reclamation time of 33 years. Differences in soil enzyme activity and microbial community function after long-term reclamation have potential to be reflected in soil functional integrity and ecosystem service.
Long-Term Amelioration Practices Reshape the Soil Microbiome in a Coastal Saline Soil and Alter the Richness and Vertical Distribution Differently Among Bacterial, Archaeal, and Fungal Communities

Abstract:Globally soil salinity is one of the most devastating environmental stresses affecting agricultural systems and causes huge economic losses each year. High soil salinity causes osmotic stress, nutritional imbalance and ion toxicity to plants and severely affects crop productivity in farming systems. Freezing saline water irrigation and plastic mulching techniques were successfully developed in our previous study to desalinize costal saline soil. Understanding how microbial communities respond during saline soil amelioration is crucial, given the key roles soil microbes play in ecosystem succession. In the present study, the community composition, diversity, assembly and potential ecological functions of archaea, bacteria and fungi in coastal saline soil under amelioration practices of freezing saline water irrigation, plastic mulching and the combination of freezing saline water irrigation and plastic mulching were assessed through high-throughput sequencing. These amelioration practices decreased archaeal and increased bacterial richness while leaving fungal richness little changed in the surface soil. Functional prediction revealed that the amelioration practices, especially winter irrigation with saline water and film mulched in spring, promoted a community harboring heterotrophic features. β-null deviation analysis illustrated that amelioration practices weakened the deterministic processes in structuring coastal saline soil microbial communities. These results advanced our understanding of the responses of the soil microbiome to amelioration practices and provided useful information for developing microbe-based remediation approaches in coastal saline soils.

Abstract:Soil salinization has become a major challenge that severely threatens crop growth and influences the productivity of agriculture. It is urgent to develop effective management measures to improve saline–alkali soil. Thus, in this study, soil properties, microbial communities, and function under desulfurization gypsum (DE), soil amendment (SA), farm manure (FA), and co-application of desulfurization gypsum, soil amendment, and farm manure (TA) in a field experiment were examined by high-throughput sequencing. The results showed that the application of modified materials is an effective approach in improving saline–alkali soil, especially TA treatment significantly increased the content of available phosphorus (AP), available potassium (AK), soil organic matter (SOM), and alkaline hydrolysis nitrogen (AHN) and decreased pH, bulk density (BD), and electrical conductivity (EC). The application of modified materials resulted in notable enhancement in fungal diversity and altered the composition and structure of the fungal community. Conversely, the effect on the bacterial community was comparatively minor, with changes limited to the structure of the community. Regarding the fungal community composition, Ascomycota, Mortierellomycota, and Basidiomycota emerged as the dominant phyla across all treatments. At each taxonomic level, the community composition exhibited significant variations in response to different modified materials, resulting in divergent soil quality. The TA treatment led to a decrease in Mortierellomycota and an increase in Ascomycota, potentially enhancing the ability to decompose organic matter and facilitate soil nutrient cycling. Additionally, the sensitivity of fungal biomarkers to modified materials surpassed that of the bacterial community. The impact of modified materials on soil microbial communities primarily stemmed from alterations in soil EC, AP, AK, and SOM. FUNGuild analysis indicated that the saprotroph trophic mode group was the dominant component, and the application of modified materials notably increased the symbiotroph group. PICRUSt analysis revealed that metabolism was the most prevalent functional module observed at pathway level 1. Overall, the application of modified materials led to a decrease in soil EC and an increase in nutrient levels, resulting in more significant alterations in the soil fungal community, but it did not dramatically change the soil bacterial community. Our study provides new insights into the application of modified materials in increasing soil nutrients and altering soil microbial communities and functions and provides a better approach for improving saline–alkali soil of Hetao Plain.

Abstract:Addressing the challenges posed by Saline-Alkali land holds significant promise for optimizing agricultural resources and fostering sustainable agricultural development in affected areas. This study explores the efficacy of coal-based solid waste soil conditioner and vermicompost as mixed-component soil amendments in varying proportions. Through comprehensive field experiments, we investigate the impact of these amendments on soil physicochemical properties, microbial diversity, and the growth of L. chinensis, a grass species native to saline areas. Our findings reveal that the application of vermicompost-based soil amendments effectively reduced soil conductivity and led to noteworthy improvements in the pH of Saline-Alkali soil. Moreover, these amendments demonstrated a marked capacity to enhance soil organic matter and available nutrients, most notably available phosphorus. Concurrently, the introduced soil amendments exhibited a positive influence on soil microbial diversity. A correlation analysis underscores the pivotal roles of soil pH and organic matter in shifting soil microbial communities. In response to amendments, L. chinensis exhibited varying degrees of growth enhancement, with biomass increments ranging from 6.37% to 19.91%. In summary, vermicompost and soil conditioner can improve Saline-Alkali land by supplementing organic matter and effective nutrients, improving pH and conductivity, and regulating microbial community, so as to realize the restoration and improvement of Saline-Alkali land; the greatest improvement was achieved with soil conditioner and vermicompost application rates of 30,000 kg·ha−1 and 15,000 kg·ha−1, respectively. Our results provide robust support for advocating for the integration of vermicompost-based soil amendments in Saline-Alkali land management strategies.

Modifying soil bacterial communities in saline mudflats with organic acids and substrates

Abstract:Aims: The high salinity of soil, nutrient scarcity, and poor aggregate structure limit the exploitation and utilization of coastal mudflat resources and the sustainable development of saline soil agriculture. In this paper, the effects of applying exogenous organic acids combined with biological substrate on the composition and diversity of soil bacterial community were studied in moderately saline mudflats in Jiangsu Province. Methods: A combination of three exogenous organic acids (humic acid, fulvic acid, and citric acid) and four biological substrates (cottonseed hull, cow manure, grass charcoal, and pine needle) was set up set up on a coastal saline mudflat planted with a salt-tolerant forage grass, sweet sorghum. A total of 120 kg ha −1 of organic acids and 5,000 kg ha −1 of substrates were used, plus two treatments, CK without application of organic acids and substrates and CK 0 in bare ground, for a total of 14 treatments. Results: No significant difference was found in the alpha diversity of soil bacterial community among all treatments ( p ≥ 0.05), with the fulvic acid composite pine needle (FPN) treatment showing the largest increase in each index. The beta diversity differed significantly ( p < 0.05) among all treatments, and the difference between citric acid–grass charcoal (CGC) and CK treatments was greater than that of other treatments. All treatments were effective in increasing the number of bacterial ASVs and affecting the structural composition of the community. Citric acid–cow manure (CCM), FPN, and CGC treatments were found to be beneficial for increasing the relative abundance of Proteobacteria , Chloroflexi , and Actinobacteria , respectively. By contrast, all treatments triggered a decrease in the relative abundance of Acidobacteria . Conclusion: Among the 12 different combinations of exogenous organic acid composite biomass substrates applied to the coastal beach, the CGC treatment was more conducive to increasing the relative abundance of the salt-tolerant bacteria Proteobacteria , Chloroflexi and Actinobacteria , and improving the community structure of soil bacteria. The FPN treatment was more conducive to increase the species diversity of the soil bacterial community and adjust the species composition of the bacterial community.

Abstract:Soil amendments have been extensively used to remediate heavy metal contaminated soils by immobilizing or altering edaphic properties to reduce the bioavailability of heavy metals. However, the potential influences of long-term soil amendments applications on microbial communities and polluted soil health are still in its infancy despite that have been applied for decades. We used amplicon sequencing and q-PCR array to characterize the root-associated microbial community compositions and rhizosphere functional genes in a five-year field experiment with consecutive application of four amendments (lime, biochar, pig manure, and a commercial Mg-Ca-Si conditioner). Compared with the control, soil amendments reduced the available Cd (CaCl2 extractable Cd) in soils and strongly affected bacterial community compositions in four root-associated niches. Five rare keystone bacterial species were found belonging to the family Gallionellaceae (1), Haliangiaceae (1), Anaerolineaceae (2), and Xanthobacteraceae (1), which significantly correlated with soil pH and the functional genes nifH and phoD. Random forest analysis showed that rhizosphere soil pH and microbial functions, and root-associated keystone bacterial community compositions mainly influenced the Cd concentrations in rice grains. Altogether, our field data revealed five-year consecutive application of soil amendments regulated root-associated microbial community assembly and enhanced microbial functions, thereby improved rhizosphere health of Cd-contaminated soils.

Abstract:Due to the high salt content and pH value, the structure of saline-sodic soil was deteriorated, resulting in decreased soil fertility and inhibited soil element cycling. This, in turn, caused significant negative impacts on crop growth, posing a major challenge to global agriculture and food security. Despite numerous studies aimed at reducing the loss of plant productivity in saline-sodic soils, the knowledge regarding shifts in soil microbial communities and carbon/nitrogen cycling during saline-sodic soil improvement remains incomplete. Consequently, we developed a composite soil amendment to explore its potential to alleviate salt stress and enhance soil quality. Our findings demonstrated that the application of this composite soil amendment effectively enhanced microbial salinity resistance, promotes soil carbon fixation and nitrogen cycling, thereby reducing HCO 3 − concentration and greenhouse gas emissions while improving physicochemical properties and enzyme activity in the soil. Additionally, the presence of CaSO 4 contributed to a decrease in water-soluble Na + content, resulting in reduced soil ESP and pH by 14.64 % and 7.42, respectively. Our research presents an innovative approach to rehabilitate saline-sodic soil and promote ecological restoration through the perspective of elements cycles.

Influence of Organic Amendments Based on Garden Waste for Microbial Community Growth in Coastal Saline Soil

Abstract:Garden waste compost (GWC) has been applied as an amendment to improve the desalination efficiency, nutrient availability and diversity of the microbial community in coastal saline soil. Understanding the response of the microbial community to garden waste compost application is of great significance in coastal ecological restoration. Four treatments were established: CK, nonamended control; T1, application of 68 kg·m−3 garden waste compost; T2, application of 15 kg·m−3 bentonite; and T3, a mixture of garden waste compost and bentonite. In addition, soil physicochemical properties, soil enzymes, microbial biomass carbon and the soil microbial community were measured. The results show that T3 had a more significant effect on increasing soil enzymes, as well as microbial biomass carbon and nitrogen, urease, sucrase and dehydrogenase activities. Based on the relative abundance, microbial diversity and linear discriminant effect size (LEfSe) analyses, the amendments can be seen to have increased the microbial abundance and alpha diversity of the bacterial structure and also altered the microbial community structure. RDA and Pearson correlation analysis at the phylum level indicated that available nitrogen, total porosity, hydraulic conductivity, bulk density and EC were the primary determinants of microbial communities associated with this amendment. In conclusion, the application of garden waste compost enables more microorganisms to participate in the soil material cycle, indicating that garden waste composting is beneficial to the restoration of coastal soils.

Abstract:Soil organic amendments in agricultural production can benefit crop production and a wide range of soil properties, including soil aggregation. Soil aggregate formation is largely driven by microbial activities, and can in-turn influence microbial communities by generating distinct microbial habitats, as well as associated impacts on water and nutrient dynamics. We investigated the long-term effects of two fertilizer management strategies (poultry manure compost vs. mineral fertilizer) and biochar amendment (0 vs. 10 t ha−1 walnut shell biochar, 900 °C pyrolysis temperature, by-product of gasification) on soil aggregation, soil organic C, and microbial community dynamics in water-stable aggregate fractions in corn-tomato rotations. Using wet-sieving, soils (0–15 cm) were divided into four size fractions: large macroaggregates (2000–8000 μm), small macroaggregates (250–2000 μm), microaggregates (53–250 μm) and silt and clay (<53 μm) for calculation of mean weight diameter in both 2014 and 2018. The total C and microbial community composition and abundance within each fraction were evaluated in 2018. Across all treatments, six years of continuous compost application maintained soil aggregate stability and C storage by increasing soil microbial biomass and associated dissolved organic C. Bacterial and fungal populations under compost treatments were significantly higher than under mineral fertilizer treatments based on 16S rRNA gene copy number and internal transcribed spacer (ITS) abundance, which likely contributed to the formation and maintenance of macroaggregates in compost treatments. Interestingly, continuous application of manure compost may increase microbial available C sources by increasing the abundance of bacteria with the potential to degrade aromatic C as predicted from 16S sequences. Soil under the mineral fertilizer treatment showed decreases in the proportion of large macroaggregates, bulk soil C, and aggregate-associated C storage compared to the compost treatment. The application of highly recalcitrant walnut shell biochar had limited long-term impacts on soil aggregation and C dynamics, likely due to its lack of microbially-available C and limited interaction with the soil environment. Our results indicate that continuous compost inputs maintained soil structure and associated physical stabilization of SOM by enlarging soil microbial available C pool, higher soil microbial biomass, and increasing aggregate formation. The soil aggregate structure, in-turn, generated diverse habitats and altered soil microbial communities. Compost inputs, in addition to or in partial replacement of mineral fertilizer inputs, can provide valuable microbial-driven ecosystem services, such as carbon storage and soil structure, while still providing fertility for crop growth.

Abstract:Saline–alkali soils exhibit ionic toxicities associated with neutral salinity, as well as a high pH that hinders the exclusion of sodium ions and absorption of vital nutrients; thus, obstructing the development of coastal shelterbelts. A three-year field experiment using a high-soil-pH site was conducted for this study to investigate the influences of five prospective amendments on the soil microenvironments of different soil layers compared to a control. Firstly, the bacterial phyla Proteobacteria, Firmicutes, and Actinobacteria were found to be the most predominant in the samples. As for the fungi phylum, Ascomycota was identified as the most abundant. Similar to Module 1’s findings, the relative abundances of Ascomycota varied across treatments. Additionally, differences in the ACE index were primarily observed in the deeper soil layers, where all five soil amendments increased the bacterial ACE index compared to the CK (no additive). Only the BA (biochar mixed with arbuscular mycorrhizal fungi) and AM (arbuscular mycorrhizal fungi on its own) treatments significantly increased the fungal ACE index. In the 20–40 cm soil layer, the pH value of the control group was significantly higher than that of all other treatments, except for the AM treatment. However, the AM treatment induced significantly higher soil enzyme activities and available nutrients compared to the CK. Moreover, the Mantel test showed significant correlations between the Module 1 community, the generalist (microbial species that serve as module hubs and connectors, primarily for Acidobacteria) community and soil pH, electrical conductivity, enzyme activities, as well as bacterial and fungal ACE indices. Pearson’s correlation revealed a significantly positive association between enzyme activities and available nutrients. Our findings suggested that keystone microbial species have the potential to improve the availability of soil nutrients through the regulation of microbial diversity and stimulation of soil enzyme activities, to ultimately ameliorate saline–alkali soil. Furthermore, the application of AM in combination with an appropriate amount of biochar is a preferred strategy for the improvement of saline–alkali soils.

Abstract:Saline-alkali soil poses significant chanllenges to sustainable development of agriculture. Although biochar is commonly used as a soil organic amendment, its microbial remediation mechanism on saline-alkali soil requires further confirmation. To address this, we conducted a pot experiment using cotton seedlings to explore the potential remediation mechanism of rice straw biochar (BC) at three different levels on saline-alkaline soil. The results showed that adding of 2% biochar greatly improved the quality of saline-alkaline soil by reducing pH levels, electrical conductivity (EC), and water-soluble ions. Moreover, biochar increased the soil organic matter (SOM), nutrient availability and extracellular enzyme activity. Interestingly, it also reduced soil salinity and salt content in various cotton plant tissues. Additionally, biochar had a notable impact on the composition of the microbial community , causing changes in soil metabolic pathways. Notably, the addition of biochar promoted the growth and metabolism of dominant salt-tolerant bacteria, such as Proteobacteria, Bacteroidota, Acidobacteriota, and Actinobacteriota. By enhancing the positive correlation between microorganisms and metabolites, biochar alleviated the inhibitory effect of salt ions on microorganisms. In conclusion, the incorporation of biochar significantly improves the soil microenvironment, reduces soil salinity, and shows promise in ameliorating saline-alkaline soil conditions.

Continuously applying compost for three years alleviated soil acidity and heavy metal bioavailability in a soil-asparagus lettuce system

Abstract:Soil acidification and heavy metal pollution are two common barrier factors threatening plant growth and agro-product quality. Applying manure compost is promising to alleviate soil acidity, while it may increase heavy metal accumulation in soil. In a 3-year field experiment, compost was applied for 12 consecutive harvest seasons at 15, 30, and 45 t ha −1 in a slightly acidic soil. Samples were taken at the twelfth season to examine the changes of soil properties, vegetable productivity, heavy metal accumulation and bioavailability in the soil-asparagus lettuce system. The results showed that the pH values of the topsoil were increased by 0.49–0.75 units in compost added soils compared with no compost control, soil organic matter (SOM) contents and cation exchange capacity (CEC) were increased by 34–101% and 43–44%, respectively. The soil nutrient contents were also increased in compost treatments. Continuously applying compost increased Cd, Cu, and Zn concentrations in topsoil by up to 32, 20, and 22% and decreased Pb by 10%, while soil available Cd and Zn concentrations were reduced by up to 54 and 86%, and available Cu was increased by 19–63%. The biomass of asparagus lettuce was increased by 30–59% in compost treatments, with Cd and Zn concentrations in the plant tissues reduced by 28–50% and 14–67%. Cu concentrations in the lettuce shoots were increased by 20–39%. The concentration factor and total uptake of Cd and Zn in lettuce were effectively reduced in compost treatments. Cd was more prone to be taken up, translocated and accumulated from soil to the lettuce plant than the other heavy metals. Continuously applying compost over 3 years increased soil pH, SOM, CEC, nutrient contents, and lettuce productivity, decreased Cd and Zn bioavailability in the soil-lettuce system, while posing a risk of increasing heavy metal accumulation in topsoil.

Abstract:To mitigate the issues of severe farmland soil salinization, the environmental degradation stemming from the overuse of chemical fertilizers, and suboptimal soil composition, a study was conducted to investigate the influence of different types and ratios of organic fertilizers on the physical and chemical attributes of saline–alkali soil. This study aimed to investigate the relationship between different types and proportions of organic fertilizers, soil moisture, organic fertilizer application rates, organic carbon molecular structure, and the soil environment in saline–alkali soils. Reducing the application of chemical fertilizers and substituting them with organic fertilizers can improve the soil quality of saline–alkali lands. The results indicated that replacing a part of the urea with organic fertilizer in saline–alkali farmland reduced the soil salinity by 11.1 to 22.8% in the 0–60 cm soil layer, decreased the soil pH by 0.11 to 1.52%, and increased the soil redox potential (Eh) values by 2.5 to 4.3% in the 0–20 cm layer of the mild and moderate saline–alkali soils. It also decreased the accumulation of the soil organic matter (OM) during the growing season. Compared to commercial organic fertilizers, natural organic fertilizers increased the accumulation of the soil soluble carbon (DOC) and nitrogen (DON), resulting in less soil salinity accumulation. When commercial organic fertilizer was applied in a 1:1 ratio with inorganic fertilizer, the salt accumulation was minimized. Compared to conventional fertilization, organic fertilizer reduced the accumulation of the NH4+-N (ammonium nitrogen) and NO3−-N (nitrate nitrogen) in the soil by 3.1 to 22.6%. In comparison to conventional chemical fertilizers, the application of organic fertilizer in the mild and moderate saline–alkali soils increased the accumulation of the DOC, DON, microbial biomass carbon (MBC), microbial biomass nitrogen (MBN), and microbial quotient during the grain-filling stage. Specifically, it increased the DOC, DON, and DOC/DON by 12.7 to 26.7%, 12 to 59.3%, and 15.2 to 35.5%, respectively. The application of commercial organic fertilizer in the mild saline–alkali soils increased the MBC, MBN, MBC/SOC, and MBN/TN by 37.1, 65.6, 36.7, and 4.7%, respectively. Through analyzing the relative proportions of soil surface organic carbon functional groups during the grain filling period, we observed that, after the application of organic fertilizer, the OM in the mildly salinized soils primarily originated from terrestrial plant litter, whereas, in moderately salinized soils, the OM was mainly derived from microbial sources.

Response of Soil Fungal Community in Coastal Saline Soil to Short-Term Water Management Combined with Bio-Organic Fertilizer

Abstract:This study aimed to elucidate the response of soil microbial communities to saline soil amelioration via biological organic fertilizer. A year-long experiment was conducted on coastal saline soil, employing water and fertilizer strategies. Three treatments were compared: dry field (control, CK), paddy field (W), and combined dry and irrigated fields with biological organic fertilizer (BW). Soil DNA was extracted and sequenced using high-throughput methods, revealing significant reductions in soil electrical conductivity (EC) and pH with W and BW treatments. Moreover, the BW treatment notably increased soil organic carbon content by 17.2%, as well as soil urease and alkaline phosphatase activity. Fungal community richness increased, with the BW treatment showing a 36% rise in the ACE index and a 24% increase in the Shannon index, while the Simpson index decreased by 59%. Dominant fungal phyla were Ascomycota, Mortierellomycota, and Basidiomycota, with Basidiomycota prevailing at the genus level. Redundancy analysis (RDA) indicated that soil pH, EC, and organic carbon were key determinants of fungal community distribution, with the BW treatment correlating negatively with pH and salt and positively with soil organic carbon (SOC). Fungal functional groups varied among treatments, with saprophytic fungi predominating, but the BW treatment showed a higher relative abundance of animal pathogenic fungi. In summary, the integration of biological organic fertilizer with flooding ameliorates soil properties and influences the changes in soil fungal community structure and function in the short term. These results could enhance the scientific basis for the efficient utilization and development of saline soil resources in coastal areas.

Abstract:Soil organic carbon (SOC) plays a vital role in improving soil quality and alleviating global warming. Understanding the dynamic changes in SOC is crucial for its accumulation induced by compost application in agroecosystem. In this study, soil samples were collected from three treatments: high-rate bio-compost (BioM h ), low-rate bio-compost (BioM l ), and control (CK, no fertilization) during 2002–2020 in a wheat-maize double cropping system in North China. The soils were separated into three functional fractions, i.e., coarse particle organic matter (cPOM, >250 μm), microaggregates (μAgg, 53–250 μm) and mineral-associated organic matter (MAOM, < 53 μm), and the associated SOC contents were determined. During 1993–2002, SOC contents in bulk soil significantly increased with the duration in the BioM h and BioM l plots. However, there was no significant correlation between SOC content and duration during 2002–2020. These results suggested that compost application positively improved SOC sequestration, while the duration of SOC sequestration (i.e., the longevity of increased SOC with time) under compost inputs maintained only 9 years. Moreover, there was a significant increase in mean annual SOC contents in bulk soil with compost application rate during 2002–2020, indicating that carbon saturation did not occur. Additionally, the SOC contents in the cPOM fraction increased with time ( p < 0.01), but the corresponding μAgg and MAOM associated SOC was insignificant ( p > 0.05). The MAOM fraction exhibited no additional carbon accumulation with expanding compost application, confirming a hierarchical carbon saturation in these fractions. We concluded that soils under wheat-maize double cropping system in North China have greater potential to sequester C through additional compost inputs, despite showing hierarchical saturation behavior in the non-protected coarse particulate fraction.
