Abstract:Bone tissue engineering has emerged as a promising strategy to overcome the limitations of current treatments for bone-related disorders, but the trade-off between mechanical properties and bioactivity remains a concern for many polymeric materials. To address this need, novel polymeric blends of poly-L-lactic acid (PLLA), polycaprolactone (PCL) and poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) have been explored. Blend filaments comprising PLLA/PCL/PHBV at a ratio of 90/5/5 wt% have been prepared using twin-screw extrusion. The PLLA/PCL/PHBV blends were enriched with nano-hydroxyapatite (nano-HA) and strontium-substituted nano-HA (Sr-nano-HA) to produce composite filaments. Three-dimensional scaffolds were printed by fused deposition modelling from PLLA/PCL/PHBV blend and composite filaments and evaluated mechanically and biologically for their capacity to support bone formation in vitro. The composite scaffolds had a mean porosity of 40%, mean pores of 800 μm, and an average compressive modulus of 32 MPa. Polymer blend and enriched scaffolds supported cell attachment and proliferation. The alkaline phosphatase activity and calcium production were significantly higher in composite scaffolds compared to the blends. These findings demonstrate that thermoplastic polyesters (PLLA and PCL) can be combined with polymers produced via a bacterial route (PHBV) to produce polymer blends with excellent biocompatibility, providing additional options for polymer blend optimization. The enrichment of the blend with nano-HA and Sr-nano-HA powders enhanced the osteogenic potential in vitro.

Abstract:The clinical application of osteofixation materials is crucial for maxillofacial reconstruction and orthognathic surgeries. To overcome the limitations of traditional metallic implants, bioabsorbable materials are gaining popularity due to their ability to avoid secondary removal surgeries and reduce stress shielding. This study investigates third-generation biomaterials, focusing on polylactic acid (PLA) for its biocompatibility and biodegradability, and hydroxyapatite (HAP) for its bioactive osteoconductive and bioresorbable properties. Eggshell nanoparticles (ES-NP), HAP, and bioinert alumina particles coated with titanium dioxide (TiO 2 @Al 2 O 3 ) were prepared using ball milling, co-precipitation, and sol-gel methods, respectively. PLA-based nanocomposites PLA/ESNP/Al 2 O 3 (PEA), PLA/HAP/Al 2 O 3 (PHA), PLA/ESNP/TiO 2 @Al 2 O 3 (PEAT), and PLA/HAP/TiO 2 @Al 2 O 3 (PHAT) were fabricated via solvent casting. Characterization techniques including X-ray Diffraction (XRD), Fourier Transform Infrared Spectroscopy (FT-IR), and Field-Emission Scanning Electron Microscopy (FE-SEM) were used to analyze the developed nanoparticles and composites. Results indicated PEAT and PHAT composites exhibited tensile strengths of 33.59 ± 0.38 MPa and 32.46 ± 0.46 MPa, tensile moduli of 1756.17 ± 95.43 MPa and 2367.21 ± 158.84 MPa, and shore d hardness values of 84.10 ± 1.45 SHN and 78.00 ± 2.25 SHN, respectively. Both composites achieved a wettability angle of ∼65° and surface roughness below 2.19 μm, enhancing osteoblast adhesion. Additionally, MG63 cell viability was approximately 80 %, and hemolysis rates were below 2.17 %, demonstrating their potential for maxillofacial implant applications.

Abstract:Synthetic bone graft substitutes have attracted increasing attention in tissue engineering. This study aimed to fabricate a novel, bioactive, porous scaffold that can be used as a bone substitute. Strontium and zinc doped nano-hydroxyapatite (Sr/Zn n-HAp) were synthesized by a water-based sol-gel technique. Sr/Zn n-HAp and poly (lactide-co-glycolide) (PLGA) were used to fabricate composite scaffolds by supercritical carbon dioxide technique. FTIR, XRD, TEM, SEM, and TGA were used to characterize Sr/Zn n-HAp and the composite scaffolds. The synthesized scaffolds were adequately porous with an average pore size range between 189 to 406 µm. The scaffolds demonstrated bioactive behavior by forming crystals when immersed in the simulated body fluid. The scaffolds after immersing in Tris/HCl buffer increased the pH value of the medium, establishing their favorable biodegradable behavior. ICP-MS study for the scaffolds detected the presence of Sr, Ca, and Zn ions in the SBF within the first week, which would augment osseointegration if implanted in the body. nHAp and their composites (PLGA-nHAp) showed ultimate compressive strength ranging between 0.4-19.8 MPa. A 2.5% Sr/Zn substituted nHAp-PLGA composite showed a compressive behavior resembling that of cancellous bone indicating it as a good candidate for cancellous bone substitute.
Strontium-Substituted Nanohydroxyapatite Containing Biodegradable 3D Printed Composite Scaffolds for Bone Regeneration

Abstract:Treatment of large-size bone defects is difficult, and acquiring autografts may be challenging due to limited availability. A synthetic patient-specific bone substitute can be developed by using 3D printing technologies in such cases. In the present study, we have developed photocurable composite resins with poly(trimethylene carbonate) (PTMC) containing a high percentage of biodegradable bioactive strontium-substituted nanohydroxyapatite (SrHA, size 30-70 nm). These photocurable resins have then been employed to develop high-surface-area 3D-printed bone substitutes using the digital light processing (DLP) technique. To enhance the surface area of the 3D-printed substitute, cryogels alone and functionalized with bioactive components of bone morphogenetic protein (BMP) and zoledronic acid (ZA) were filled within the 3D-printed scaffold/substitute. The scaffolds were tested in vitro for biocompatibility and functionality in vivo in two therapeutically relevant rat models with large bone defects (4 mm). The porosities of 3D printed scaffolds were found to be 60.1 ± 0.9%, 72.9 ± 0.5%, and 74.3 ± 1.6% for PTMC, PTMC-HA, and PTMC-SrHA, respectively, which is in the range of cancellous bone (50-95%). The thermogravimetric analysis demonstrated the fabrication of 3D printed composites with HA and SrHA concentrations of 51.5 and 57.4 wt %, respectively, in the PTMC matrix. The tensile Young's modulus (E), compressive moduli, and wettability increased post incorporation of SrHA and HA in the PTMC matrix. In vitro and in vivo results revealed that SrHA integrated into the PTMC matrix exhibited good physicochemical and biological properties. Furthermore, the osteoactive molecule-functionalized 3D printed composite scaffolds were found to have an adequate osteoconductive and osteoinductive surface that has shown increased bone regeneration and defect repair in both tibial and cranial bone defects. Our findings thus support the use of PTMC-SrHA composites as next-generation patient-specific synthetic bioactive biodegradable bone substitutes.

Abstract:This article is intended to present an optimized Biocomposites material to prepare customized implants by using additive manufacturing technology. The most preferred implant material, polylactic acid (PLA) and hydroxyapatite (HAp), is chosen as matrix and reinforcements with different weight percentages of reinforcements. The 3D-printed composites underwent structural, mechanical, thermal stability, and in-vitro characterization. In the composites, the functional groups "OH, CH, CO, CO, and PO 4 " identified by Fourier transform infrared spectroscopy (FTIR). The thermal stability of the composites was identified by Thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC). For the composition of PLA-HAp, a maximum tensile strength of 62 ± 0.5 MPa (30 wt%), a maximum compressive strength of 61 ± 0.5 MPa at 50 wt%), a maximum flexural strength of 46 ± 0.5 MPa (20 wt%), and a Shore-D hardness of 82 ± 0.5 SHN (50 wt%) were obtained. Modeled maximal principal stress in PLA and HAp. The 30 wt% of HAp-PLA composite carried out the in-vitro direct cytotoxicity, and it revealed 97% cell viability and 3% cytotoxicity through the L929 cell line. The results indicate that the addition of reinforcement increased the mechanical strength and cell visibility; therefore, the HAp-PLA composite would be used in biomedical applications.

Abstract:Composite materials composed of polylactide (PLA) and nano-hydroxyapatite (n-HA) have been recognized as excellent candidate material in bone repai The difference in hydrophilicity/hydrophobicity and poor interfacial compatibility between n-HA filler and PLA matrix leads to non-uniform dispersion of n-HA in PLA matrix and consequent poor reinforcement effect. In this study, an HA/PLA nanocomposite was designed based on the surface modification of n-HA with poly(D-lactide) (PDLA), which not only can improve the dispersion of n-HA in the poly(L-lactide) (PLLA) matrix but also could form a stereocomplex crystal with the matrix PLLA at the interface and ultimately lead to greatly enhanced mechanical performance The n-HA/PLA composites were characterized by means of scanning electron microscopy, Fourier transform infrared spectroscopy, X-Ray diffraction, thermal gravity analysis, differential scanning calorimetry, and a mechanical test; in vitro cytotoxicity of the composite material as well as its efficacy in inducing osteogenic differentiation of rat bone marrow stromal cells (rMSCs) were also evaluated. Compared with those of neat PLLA, the tensile strength, Young’s modulus, interfacial shear strength, elongation at break and crystallinity of the composites increased by 34%, 53%, 26%, 70%, and 17%, respectively. The adhesion and proliferation as well as the osteogenic differentiation of rMSCs on HA/PLA composites were clearly evidenced. Therefore, the HA/PLA composites have great potential for bone repai.

Abstract:Poly (L-lactic acid) (PLLA) has emerged as a promising orthopedic implant material due to its favorable strength and biodegradability. However, challenges such as low toughness and limited osteoinductivity hinder its widespread use in bone fixation. This study focuses on enhancing the toughness and osteogenic activity of PLLA-based orthopedic implants. Inspired by reinforcement techniques in the construction industry, we designed a structure comprising flexible fibers enveloped by PLLA/hydroxyapatite (HA) crystalline phases. Initially, PLLA/poly (butylene succinate- co -adipate) (PBSA)/HA composites with "sea-island" morphology were prepared through melt-compounding. Subsequently, the highly oriented PBSA fibers were in situ formed during microinjection molding for bone screw fabrication. Comprehensive investigation into the structural-mechanical property relationship revealed a significant increase in elongation at break (from 5.4 % to 59.4 % with an optimal PBSA/HA ratio), while maintaining a high stiffness and a slight decrease in tensile strength (from 62.0 MPa to 56.0 MPa). The flexural tests of the resulting composite bone screws demonstrated a significant increase in toughness. Additionally, the in vivo studies corroborated the osteogenic potential of the microinjection molded bone screws by using hematoxylin and eosin (HE) and Masson staining. The methodology presented in this study offers a promising approach for advancing PLLA-based fixation devices in bone repair applications.

Abstract:Over the last years, orthopedic procedures for bone repairs have been developed due to an increase in trauma and diseases. The development of bioactive composites using biodegradable polymers like poly(lactic acid) (PLA) and bioactive fillers as hydroxyapatite (HA) originate biomaterials, which combine bioactivity of HA and PLA biocompatibility. Therefore, using additive manufacturing is possible for the production of customized products made from these materials; however, a thorough study of these materials is required. In this context, melt‐compounding has been used to manufacture bioactive composites of PLA/HA, and rheological, molecular, and thermomechanical behavior were assessed. The biocomposite of PLA with 10 wt% HA presented a strong shear thinning behavior, which makes it more suitable for fused filament fabrication since lower printing pressure is required. Furthermore, this composite presented an enhancement of 12% in thermomechanical properties in comparison to PLA and a slight increase in cell proliferation. PLA and PLA/HA were fabricated and used to produce 3D calibrations cube as a proof of concept. They presented good printability and high accuracy, and therefore, further investigation needs to be performed to unleash its use in bone tissue engineering applications. POLYM. ENG. Abstract:The medical sector needs medical devices that are biocompatible or bioabsorbable, and Poly Lactic Acid (PLA) satisfies that need. PLA is a thermoplastic polymer that degrades naturally and has good mechanical strength. In order to create anchors, screws, plates, pins, rods, and mesh for medical implants, polylactic acid, which is harmless, can be broken down into lactic acid. The primary objective of this work is to characterize the mechanical properties and material properties of polymer composite materials reinforced with hydroxyapatite (HAp), zeolite, magnesium, and zinc particulates for biomedical implant applications. Using an injection moulding process, the composite samples were created. Mechanical qualities such as tensile, compression, flexural strength, and shore D hardness were examined in accordance with American Society for Testing and Materials (ASTM) standards. The maximum tensile, compressive, and flexural strength were found to be 39.23 MPa, 36.80 MPa, and 68.25 MPa, respectively, for 65 wt.% PLA + 35 wt.% HAp + 5% wt.% Zeolite polymer composites were observed, and a maximum Shore D hardness of 79.8 SHN was found. Further, material characterizations were carried out for this composite material. The morphological characterization using Field Emission Scanning Electron Microscopy (FESEM) and Energy Dispersive X-ray (EDAX) analysis was used to analyze the surface morphology and confirm the various proportions of elements in the PLA composites. Fourier-transform infrared spectroscopy (FT IR) was used to confirm the different functional groups. Thermogravimetric analysis (TGA) and derivative thermogravimetric (DTG) analysis were used to study the thermal properties of polymer composites. The results show that the fabricated polymer composites could have better mechanical strength and be suitable for medical implant applications.

Abstract:<h3 class="a-plus-plus">Abstract</h3> Nowadays polylactic acid (PLA) is widely used in orthopedics surgeries as implants material due to well mechanical characterization and biomedical properties. But the PLA implants suffer from slow degradation rate when it is used in real-life scenario. In the present research work, the PLA specimens using additive manufacturing technique are fabricated and further assessed for mechanical characterization and its degradation behavior with different parameters. The change in weight of scaffolds was measured using digital weight measure, and pH value was measured using pH meter. Morphology and elemental composition of PLA scaffolds were characterized by SEM and EDS, respectively, while compressive strength is measured by the universal testing machine. Apatite formation and biocompatible nature of fabricated scaffolds were analyzed by in vitro simulated body fluid study. The outcomes of characterization exposed that scaffold with 60% infill percentage had maximum porosity, which is beneficial for the apatite formation and osseointegration. The average change in compressive strength was measured as 49.79 MPa after 14 days and 46.11 MPa after 28 days, whereas the average change in pH value was measured as 5.67 and 5.27 after 14 and 28 days of incubation period, respectively. The degradation rate of specimen 3 was 27.92% less than that of specimen 1, 35.69% less than that of specimen 5, and 87.98% more than that of specimen 9. This study concludes the positive effect of process parameters on degradation rate and biocompatible behavior of PLA implants.

Abstract:Controlled incorporation of bioactive hydroxyapatite (HA) into poly(lactic acid) (PLA) signifies a promising approach to design and development of biomedical-adaptive materials. Here we unravel a microwave-assisted biomineralization approach to synthesis of HA nanowhiskers (HANWs), which were characterized by well-controlled diameter (∼30 nm) and length (80 − 120 nm), combined with a desirable calcium − phosphorus ratio (Ca/P) of 1.67. A high-shear liquid dispersion (HSLD) method that provided a combination of high pressure (up to 50 kPa) and high shear rate approaching 10000 s −1 was established to fabricate homogeneous PLA/HANWs composites. In particular, upon incorporation of 30 wt % HANWs the tensile strength and elastic modulus of PLA-HA30 (76.7 MPa and 3.3 GPa) were elevated by 48% and 84% compared to those of pure PLA, respectively, as accompanied by a nearly 2-fold increase in the cell viability. This work paves a facile yet effective roadway to strong and osteoconductive PLA composites appealing for bone tissue repairing. Graphical Synopsis: Biomineralization concepts are abstracted to synthesize well-defined and bioactive HANWs, which could be uniformly dispersed in PLA biofilms by high-shear liquid exfoliation.

Abstract:The major problem in bone tissue engineering is the development of scaffolds which can simultaneously meet the requirements of porous structure, as well as have the ability to guide the regeneration of damaged tissue by biological fixation. Composites containing biodegradable matrix and bioactive filler are the new hope in this research field. Herein we employed a simple and facile solvent casting particulate-leaching method for producing polylactide acid/hydroxyapatite (PLA/HA) composites at room temperature. FT-IR analysis confirmed the existence of necessary functional groups associated with the PLA/HA composite, whereas energy-dispersive X-ray (EDX) spectra indicated the uniform distribution of hydroxyapatite particles in the polymer matrix. The beehive-like surface morphology of the composites revealed the presence of macropores, ranged from 300 to 400 μm, whereas the thickness of the pores was noticed to be 1–2 μm. The total porosity of the scaffolds, calculated by hydrostatic weighing, was found to be 79%. The water contact angle of pure PLA was decreased from 83.6 ± 1.91° to 62.4 ± 4.17° due to the addition of hydroxyapatite in the polymer matrix. Thus, the wettability of the polymeric biomaterial could be increased by preparing their composites with hydroxyapatite. The adhesion of multipotent mesenchymal stromal cells over the surface of PLA/HA scaffolds was 3.2 times (p = 0.03) higher than the pure PLA sample. Subcutaneous implantation in mice demonstrated a good tolerance of all tested porous scaffolds and widespread ingrowth of tissue into the implant pores. HA-containing scaffolds showed a less pronounced inflammatory response after two weeks of implantation compared to pure PLA. These observations suggest that PLA/HA composites have enormous potential for hard tissue engineering and restoring maxillofacial defects.

Abstract:Poly-lactic acid (PLA) is an attractive biopolymer where it is biocompatible with respect to biomedical applications. Hydroxyapatite (HA) ceramics have been broadly practiced as bone substitutes because of their chemical composition close to the mineral phase of bone. Indirectly, the combination of PLA and HA would be a suitable biomaterial candidate for implant and bone repair applications. In this study, the feedstock composition of PLA/HA has been investigated to obtain good rheological properties before undergoing an injection moulding process. The process begins with mixing the specific powder loadings of 60 wt.% PLA/ 40 wt.% HA at 30 rpm of rotating speed and 180 oC of mixing temperature. Then, the feedstock is evaluated using a capillary rheometer at three different temperatures, 140 oC, 150 oC, and 160 oC. Feedstock exhibiting pseudoplastic flow (n<1) is chosen for the injection moulding process. The result shows injected moulded part successfully produced at barrel and nozzle temperature of 160 oC, mould temperature of 37 oC and injection pressure of 0.7 MPa. The injected parts were characterized for physical and mechanical properties. The PLA/HA specimen density is 1.564 ± 0.02 g/cm3. The Young’s Modulus of the tensile is 1.82 ± 0.5 GPa within the range of human bone, 1.15 to 5.44 GPa.

Abstract:Different types of implants are used in orthopedic applications. Due to the stress shielding and a second surgery caused by applying metal implants as orthopedic joints, using biodegradable bone implants with antibacterial properties is advantageous. Therefore, this study aims at fabricating a multifunctional poly (lactic acid) (PLA)-polycaprolactone (PCL)-based scaffold reinforced with hydroxyapatite (HA) and zinc oxide (ZnO) nanoparticles. Solvent casting combined with die-casting techniques was employed for fabricating the scaffolds, and their physical, mechanical, and biological properties were characterized. The morphology of the samples and the growth of hydroxyapatite crystals on the surface of the samples were examined by field emission scanning electron microscope. In dry conditions, adding nanoparticles to the scaffolds has significantly ( p < .05) increased the tensile Young's modulus from 450.00 ± 87.18 MPa to 1050.66 ± 104.05 MPa. Also, adding ZnO has improved the antibacterial properties of the scaffolds against S.aureus and E.coli bacteria strains. Moreover, the MTT assay results (after 72 h cell culture) show that the viability of L929 cells has increased from 22.18 ± 0.62% for the PLA-PCL scaffold to 70.03 ± 0.25% for the optimal nanostructure ( p < .05). In conclusion, the PCL-PLA/HA/1% ZnO scaffold could potentially be used as absorbable spongy bone implants.

Abstract:To enhance the mechanical strength and bioactivity of poly(lactic acid) (PLA) to the level that can be used as a material for spinal implants, poly(glycolic acid) (PGA) fibers and hydroxyapatite (HA) were introduced as fillers to PLA composites. To improve the poor interface between HA and PLA, HA was grafted by PLA to form HA-g-PLA through coupling reactions, and mixed with PLA. The size of the HA particles in the PLA matrix was observed to be reduced from several micrometers to sub-micrometer by grafting PLA onto HA. The tensile and flexural strength of PLA/HA-g-PLA composites were increased compared with those of PLA/HA, apparently due to the better dispersion of HA and stronger interfacial adhesion between the HA and PLA matrix. We also examined the effects of the length and frequency of grafted PLA chains on the tensile strength of the composites. By the addition of unidirectionally aligned PGA fibers, the flexural strength of the composites was greatly improved to a level comparable with human compact bone. In the bioactivity tests, the growth of apatite on the surface was fastest and most uniform in the PLA/PGA fiber/HA-g-PLA composite.

Abstract:Polylactic acid (PLA), pure magnesium powder, and calcium phosphate powder were used to form a three-phase degradable biomedical composite. The effects of various powder proportions in polylactic acid-Mg-Ca3(PO4)2 composites were analyzed through mechanical and biological tests, which revealed that both the tensile and impact strength of the composite increased. Additionally, ductility presented only after a small proportion of powder was added. Hardness slightly increased because of dispersion strengthening. Furthermore, the addition of pure magnesium and calcium phosphate accelerated the degradation rate, and biocompatible salts were generated after degradation, which can improve healing and renewal in bone tissue. None of the composites exhibited cytotoxicity, meeting biological safety requirements. Overall, PLA10M10C (10 wt.% Mg, 10 wt.% Ca3(PO4)2) exhibited superior performance. Accordingly, PLA10M10C can serve as a reference for degradable biomedical material applications in orthopedic implants.

Abstract:INTRODUCTION: poly(l-lactic acid) (PLLA) has been approved for clinical use by the US Food and Drug Administration (FDA); however, their stronger hydrophobicity and relatively fast degradation rate restricted their widespread application. In consideration of the composition of bone, the inorganic-organic composite has a great application prospect in bone tissue engineering. Many inorganic-organic composite scaffolds were prepared by directly mixing the active ingredient, but this method is uncontrolled and will lead to lack of homogeneity in the polymer matrix. Strontium (Sr) is an admirable addition to improve the bioactivity and bone induction of hydroxyapatite (HA). To our knowledge, the application of biomimetic mineralized strontium-doped hydroxyapatite on porous poly(l-lactic acid) (Sr-HA/PLLA) scaffolds for bone defect repair has never been reported till date. Biomimetic mineralized Sr-HA/PLLA porous scaffold was developed in this study. The results indicated that the Sr-HA/PLLA porous scaffold could improve the surface hydrophobicity, reduce the acidic environment of the degradation, and enhance the osteoinductivity; moreover, the ability of protein adsorption and the modulus of compression were increased. The results also clearly showed the effectiveness of the Sr-HA/PLLA porous scaffold in promoting cell adhesion, proliferation, and alkaline phosphatase (ALP) activity. The micro computed tomography (micro-CT) results showed that more new bones were formed by Sr-HA/PLLA porous scaffold treatment. The histological results confirmed the osteoinductivity of the Sr-HA/PLLA porous scaffold. The results suggested that the Sr-HA/PLLA porous scaffold has a good application prospect in bone tissue engineering in the future.PURPOSE: The purpose of this study was to promote the bone repair.MATERIALS AND METHODS: Surgical operation of rabbits was carried out in this study.RESULTS: The results showed that formation of a large number of new bones by the Sr-HA/PLLA porous scaffold treatment is possible.CONCLUSION: Biomimetic mineralized Sr-HA/PLLA porous scaffold could effectively promote the restoration of bone defects in vivo.

In Vitro Studies and Evaluation of Antibacterial Properties of Biodegradable Bone Joints Based on PLA/PCL/HA

Abstract:Background: Due to the history of using permanent implants and the ability of adaptations of polymers to physiological environments such as the body environment, the need to design a polymer implant with a new formulation for orthopedic applications was felt. Methods: Polymer joints in this study were made by solvent casting method. The mechanical properties of the samples were investigated by bending tests, before and after immersion in simulated body fluid (SBF). Morphology of nanocomposites, bioactivity of samples and initiation of degradation process were performed by field emission scanning electron microscope (FESEM). Toxicity test was performed to evaluate the toxicity of nanocomposites. The antibacterial properties of the samples were investigated by examining the zone of inhibition and measuring the photometric concentration. Biodegradability test was performed to prove the biodegradability of polymer joints. Results: It was found that the mechanical properties of nanostructures increased with the addition of nanoparticles. Also, the presence of oxide and graphene nanoparticles affected the antibacterial properties of the composite nanostructure. Immersion in SBF solution showed that the nanostructures were biodegradable and bioactive. The results of this study indicate that the optimal nanocomposite PLA-PCL-HA-1% ZNO-1% GR has a Young’s modulus close to spongy bone and reduces the stress shielding phenomenon. The flexural Yang modulus of the PLA-PCL-HA nanocomposite was 2139.037 ± 381.312 MPa. The presence of zinc oxide and graphene nanoparticles increased the Young’s modulus to 4363.636 ± 127.498 MPa. The optimal sample has the necessary lethality against two strains of gram-positive and gram-negative bacteria and due to its bioactivity is a suitable option for use in spongy bone tissue. In this study, the viability of fibroblast cells in the vicinity of the polymer matrix versus the optimal matrix increased from 22.14 ± 0.623 to 82.96 ± 1.101% after 72 hours. Conclusions: Improving cell viability indicates a reduction in the optimal matrix toxicity compared to the polymer matrix.
Biomimetic, mussel-inspired surface modification of 3D-printed biodegradable polylactic acid scaffolds with nano-hydroxyapatite for bone tissue engineering

Abstract:Polylactic acid (PLA) has been widely used as filaments for material extrusion additive manufacturing (AM) to develop patient-specific scaffolds in bone tissue engineering. Hydroxyapatite (HA), a major component of natural bone, has been extensively recognized as an osteoconductive biomolecule. Here, inspired by the mussel-adhesive phenomenon, in this study, polydopamine (PDA) coating was applied to the surface of 3D printed PLA scaffolds (PLA@PDA), acting as a versatile adhesive platform for immobilizing HA nanoparticles (nHA). Comprehensive analyses were performed to understand the physicochemical properties of the 3D-printed PLA scaffold functionalized with nHA and PDA for their potent clinical application as a bone regenerative substitute. Scanning electron microscopy (SEM) and element dispersive X-ray (EDX) confirmed a successful loading of nHA particles on the surface of PLA@PDA after 3 and 7 days of coating (PLA@PDA-HA3 and PLA@PDA-HA7), while the surface micromorphology and porosity remain unchanged after surface modification. The thermogravimetric analysis (TGA) showed that 7.7 % and 12.3% mass ratio of nHA were loaded on the PLA scaffold surface, respectively. The wettability test indicated that the hydrophilicity of nHA-coated scaffolds was greatly enhanced, while the mechanical properties remained uncompromised. The 3D laser scanning confocal microscope (3DLS) images revealed that the surface roughness was significantly increased, reaching Sa (arithmetic mean height) of 0.402 μm in PLA@PDA-HA7. Twenty-eight days of in-vitro degradation results showed that the introduction of nHA to the PLA surface enhances its degradation properties, as evidenced by the SEM images and weight loss test. Furthermore, a sustainable release of Ca 2+ from PLA@PDA-HA3 and PLA@PDA-HA7 was recorded, during the degradation process. In contrast, the released hydroxyl group of nHA tends to neutralize the local acidic environments, which was more conducive to osteoblastic differentiation and extracellular mineralization. Taken together, this facile surface modification provides 3D printed PLA scaffolds with effective bone regenerative properties by depositing Ca 2+ contents, improving surface hydrophilicity, and enhancing the in-vitro degradation rate.

Strength reliability and in vitro degradation of three-dimensional powder printed strontium-substituted magnesium phosphate scaffolds

Abstract:Strontium ions (Sr(2+)) are known to prevent osteoporosis and also encourage bone formation. Such twin requirements have motivated researchers to develop Sr-substituted biomaterials for orthopaedic applications. The present study demonstrates a new concept of developing Sr-substituted Mg3(PO4)2 - based biodegradable scaffolds. In particular, this work reports the fabrication, mechanical properties with an emphasis on strength reliability as well as in vitro degradation of highly biodegradable strontium-incorporated magnesium phosphate cements. These implantable scaffolds were fabricated using three-dimensional powder printing, followed by high temperature sintering and/or chemical conversion, a technique adaptable to develop patient-specific implants. A moderate combination of strength properties of 36.7MPa (compression), 24.2MPa (bending) and 10.7MPa (tension) were measured. A reasonably modest Weibull modulus of up to 8.8 was recorded after uniaxial compression or diametral tensile tests on 3D printed scaffolds. A comparison among scaffolds with varying compositions or among sintered or chemically hardened scaffolds reveals that the strength reliability is not compromised in Sr-substituted scaffolds compared to baseline Mg3(PO4)2. The micro-computed tomography analysis reveals the presence of highly interconnected porous architecture in three-dimension with lognormal pore size distribution having median in the range of 17.74-26.29μm for the investigated scaffolds. The results of extensive in vitro ion release study revealed passive degradation with a reduced Mg(2+) release and slow but sustained release of Sr(2+) from strontium-substituted magnesium phosphate scaffolds. Taken together, the present study unequivocally illustrates that the newly designed Sr-substituted magnesium phosphate scaffolds with good strength reliability could be used for biomedical applications requiring consistent Sr(2+)- release, while the scaffold degrades in physiological medium. Statement of significance: The study investigates the additive manufacturing of scaffolds based on different strontium-substituted magnesium phosphate bone cements by means of three-dimensional powder printing technique (3DPP). Magnesium phosphates were chosen due to their higher biodegradability compared to calcium phosphates, which is due to both a higher solubility as well as the absence of phase changes (to low soluble hydroxyapatite) in vivo. Since strontium ions are known to promote bone formation by stimulating osteoblast growth, we aimed to establish such a highly degradable magnesium phosphate ceramic with an enhanced bioactivity for new bone ingrowth. After post-processing, mechanical strengths of up to 36.7MPa (compression), 24.2MPa (bending) and 10.7MPa (tension) could be achieved. Simultaneously, the failure reliability of those bioceramic implant materials, measured by Weibull modulus calculations, were in the range of 4.3-8.8. Passive dissolution studies in vitro proved an ion release of Mg(2+) and PO4(3-) as well as Sr(2+), which is fundamental for in vivo degradation and a bone growth promoting effect. In our opinion, this work broadens the range of bioceramic bone replacement materials suitable for additive manufacturing processing. The high biodegradability of MPC ceramics together with the anticipated promoting effect on osseointegration opens up the way for a patient-specific treatment with the prospect of a fast and complete healing of bone fractures.
