Plant Species Diversity Impacts Nitrogen Removal and Nitrous Oxide Emissions As Much As Carbon Addition in Constructed Wetland Microcosms

Abstract:With the aim to develop an approach for treating wastewater with low carbon (C) to nitrogen (N) ratio in constructed wetlands (CWs), we compared the effects of C addition and species diversity on N removal and nitrous oxide (N2O) emissions. Compared with the monocultures without C addition, C addition significantly increased N removal efficiency in monocultures (reached 71% on average); increasing species richness impacted N removal as much as C addition, and the N removal efficiency in the four-species mixture reached 75%. Both C addition and increasing species richness level significantly increased N2O emission rates (reached 1.26 and 1.40 mg m(-2) d(-1), respectively), but had no significant effect on N2O emissions per unit N removal. The effects of species richness on N removal and N2O emissions were stronger than species identity. Compared with C addition, assembling plant communities with high diversity in actual applications could be an effective and more economical approach for treating wastewater with low C/N ratio in CWs. Abstract:Rhizosphere microorganism is an important bio-component for wastewater treatment in constructed wetlands (CWs). Microbial abundance and enzyme activities in the rhizospheres of nine plant species were investigated in an integrated vertical-flow CW. The abundance of denitrifiers, as well as urease, acid, and alkaline phosphatase activities were positively correlated to plant root biomass. The abundance of bacteria, fungi, actinomycetes, ammonifiers, denitrifiers, and phosphorus decomposers, related to nutrient removal efficiencies in CWs, greatly varied among rhizospheres of different plant species (p < 0.05). Significant differences in rhizosphere enzyme activity among plant species were also observed (p < 0.05), with the exception of catalase activity. The principal component analysis using the data of microbial abundance and enzyme activity showed that Miscanthus floridulus, Acorus calamus, and Reineckia carnea were candidates to be used in CWs to effectively remove nitrogen and phosphorus from wastewater.

Abstract:In this study, an integrated treatment system was proposed and applied in situ, including detention tank, multistage constructed wetlands (CWs) and wastewater treatment plants (WWTPs), preventing nutrients flowing into Dianchi Lake, in which the treatment performance of multistage CWs were evaluated principally. Results skillfully realized the bypass purification of upstream river at dry reasons, as well as the effective management and treatment of the collected diffuse pollution at rainy reasons. The purified water flowing into water bodies could satisfy the Grade III of environmental quality standards for surface water in China with the average effluent concentrations of COD, NH4+-N, TN and TP decreased to 10 (51.2-72.7%), 0.5 (67.2-83.0%), 1.0 (71.2-79.6%) and 0.15 (72.3-89.4%) mg L-1, respectively. High-throughput sequencing results indicated that the application of poly-3-hydroxybutyrate-cohyroxyvelate-sawdust (PS) blends could enrich norank_f_Anaerolineaceae (7.95%) and Bradyrhizobium (10.2%), which were distinct from the dominant genera of Pleurocapsa (13.0%) in gravel-based CWs. Functional genes and metabolism analysis uncovered that the heterotrophic denitrification was the main pathway of nitrogen removal with the abundance of genes encoding TCA cycle, glycolysis and denitrification process up-regulated. In addition, molecular ecological network (MEN) analysis suggested the denitrification genes were positively correlated with the predominant microbes in PS-based CWs, favorable for denitrifiers to transfer and utilize electron donors during denitrification process. This study proved that the developed PS blends as carbon supplies in CWs and the proposed integrated treatment system are effective methods for watershed management, providing valuable reference to low-pollution wastewater treatment in practical engineering projects.
Learning as change of coordination dynamics: theory and experiment.

Abstract:Learning of coordination patterns was investigated theoretically from the point of view of a dynamic theory of biological coordination and with reference to recent experiments on the learning of relative timing patterns. The theory is based on theoretical and experimental work showing that coordinated movement is characterized not only by the actually performed pattern of coordination but by an entire dynamics of coordination. Theoretically, such dynamics are captured as equations of motion of relevant collective variables. Experimentally, signatures of these underlying dynamics can be found in the temporal stability of coordination patterns, which can be assessed through various stability measures as well as through processes of pattern change. We argue that not only intrinsic coordination tendencies, but also specific behavioral requirements, be they perceived, memorized, or intended, must be expressed in terms of such dynamics. The concept of behavioral information captures such requirements as part of the coordination dynamics. We expound two hypotheses on the nature of learning in this framework. First, we assume that at each point during the learning process the system is governed by a well-defined coordination dynamics. This equation of motion evolves with learning so as to acquire an attractor solution near the to-be-learned pattern. Second, we hypothesize that this change of the coordination dynamics, captured by the time course of memorized behavioral information, can itself be ascribed to an additional layer of dynamics, the slower learning dynamics. Testable consequences of these views are discussed in the light of recent experimental findings on the learning of a relative phase in rhythmic movement: (a) Learning affects dynamic properties of performed coordination patterns, in particular, their stability; (b) the change of the coordination dynamics due to learning leads to specific changes of behavior also under conditions other than the learned condition, namely, to systematic deviation toward the learned patterns; (c) learning may lead to instabilities in the coordination behavior if initial and learned performance differ sufficiently; and (d) the dynamic properties of the performed coordination patterns are distinct on the two time scales of learning and of performance.

Abstract:The effluents from conventional wastewater treatment plants (WWTP), even if accomplishing quality regulations, substantially differ in their characteristics with those of waters in natural environments. Constructed wetlands (CWs) serve as transitional ecosystems within WWTPs, mitigating these differences and restoring natural features before water is poured into the natural environment. Our study focused on an experimental surface-flow CW naturalizing the WWTP effluent in a semiarid area in Eastern Spain. Despite relatively low pollutant concentrations entering the CW, it effectively further reduced settled organic matter and nitrogen. Dissolved organic matter (DOM) reaching the CW was mainly protein-like, yet optical property changes in the DOM indicated increased humification, aromaticity, and stabilization as it flowed through the CW. Flow cytometry analysis revealed that the CW released less abundant but more active bacterial populations than those received. MiSeq Illumina sequencing highlighted changes in the prokaryotic community composition, with phyla Proteobacteria, Bacteroidetes, Firmicutes, and Actinobacteria dominating the CW outflow. Functional prediction tools (FaproTax and PICRUSt2) demonstrated a shift towards microbial guilds aligned with those of the natural aquatic environments, increased aerobic chemoheterotrophs, photoautotrophs, and metabolic reactions at higher redox potentials. Enhanced capabilities for degrading plant material correlated well with changes in the DOM pool. Our findings emphasize the role of CWs in releasing biochemically stable DOM and functionally suited microbial populations for natural receiving environments. Consequently, we propose CWs as a naturalization nature-based solution (NBS) in water-scarce regions like the Mediterranean, where reclaimed discharged water can significantly contribute to ecosystem's water resources compared to natural flows.

Abstract:Plant development greatly influence the composition structure and functions of microbial community in constructed wetlands (CWs) via plant root activities. However, our knowledge of the effect of plant development on microbial nitrogen (N) cycle is poorly understood. Here, we investigated the N removal performance and microbial structure in subsurface flow CWs at three time points corresponding to distinct stages of plant development: seedling, mature and wilting. Overall, the water parameters were profoundly affected by plant development with the increased root activities including radial oxygen loss (ROL) and root exudates (REs). The removal efficiency of NH 4 + -N was significantly highest at the mature stage ( p < 0.01), while the removal performance of NO 3 − -N at the seedling stage. The highest relative abundances of nitrification- and anammox-related microbes ( Nitrospira, Nitrosomonas , and Candidatus Brocadia, etc.) and functional genes (Amo, Hdh, and Hzs) were observed in CWs at the mature stage, which can be attributed to the enhanced intensity of ROL, creating micro-habitat with high DO concentration. On the other hand, the highest relative abundances of denitrification- and DNRA-related microbes ( Petrimonas, Geobacter , and Pseudomonas , etc.) and functional genes (Nxr, Nir, and Nar, etc.) were observed in CWs at the seedling and wilting stages, which can be explained by the absence of ROL and biological denitrification inhibitor derived from REs. Results give insights into microbial N cycle in CWs with different stages of plant development. More importantly, a potential solution for intensified N removal via the combination of practical operation and natural regulation is proposed.

Abstract:Denitrification is strongly dependent on carbon quantity and quality in most constructed wetlands (CWs). In this study, four batch CWs were designed, and were fed with nitrate-dominated water to investigate nitrate removal affected by plant and external cattail litter with or without alkali pretreatment. The results showed that the unit with plant and alkali-pretreated litter was more efficient in the initial stage whereas unit with plant and unpretreated litter was superior to other units in the middle and terminal stages. Plant accounted for less than 37% of the nitrate removal in biomass-up added CWs. The different nitrate removal rates were found to be greatly affected by the composition of the plant biomass as well as the quantity and quality of the available organic matters. It was also observed that plant biomass degradation over the period of this study resulted in various N species and concentrations in effluent.

Abstract:A limited understanding of microbial interactions and community assembly mechanisms in constructed wetlands (CWs), particularly with different substrates, has hampered the establishment of ecological connections between micro-level interactions and macro-level wetland performance. In this study, CWs with distinct substrates (zeolite, CW_A; manganese ore, CW_B) were constructed to investigate the nutrient removal efficiency, microbial interactions, metabolic mechanisms, and ecological assembly for treating rural sewage with a low carbon-to-nitrogen ratio. CW_B showed higher removal of ammonia nitrogen and total nitrogen by about 1.75-6.75 % and 3.42-5.18 %, respectively, compared to CW_A. Candidatus_Competibacter (denitrifying glycogen-accumulating bacteria) was the dominant microbial genus in CW_A, whereas unclassified_f_Blastocatellaceae (involved in carbon and nitrogen transformation) dominated in CW_B. The null model revealed that stochastic processes (drift) dominated community assembly in both CWs; however, deterministic selection accounted for a higher proportion in CW_B. Compared to those in CW_A, the interactions between microbes in CW_B were more complex, with more key microbes involved in carbon, nitrogen, and phosphorus conversion; the synergistic cooperation of functional bacteria facilitated simultaneous nitrification-denitrification. Manganese ores favour biofilm formation, increase the activity of the electron transport system, and enhance ammonia oxidation and nitrate reduction. These results elucidated the ecological patterns exhibited by microbes under different substrate conditions thereby contributing to our understanding of how substrates shape distinct microcosms in CW systems. This study provides valuable insights for guiding the future construction and management of CWs.

Abstract:Constructed wetlands (CWs) are a cost-effective and environmentally friendly wastewater treatment technology. The influent chemical oxygen demand (COD)/nitrogen (N) ratio (CNR) plays a crucial role in microbial activity and purification performance. However, the effects of CNR changes on microbial diversity, interactions, and assembly processes in CWs are not well understood. In this study, we conducted comprehensive mechanistic experiments to investigate the response of CWs to changes in influent CNR, focusing on the effluent, rhizosphere, and substrate microbiota. Our goal is to provide new insights into CW management by integrating microbial ecology and environmental engineering perspectives. We constructed two groups of horizontal subsurface flow constructed wetlands (HFCWs) and set up three influent CNRs to analyse the microbial responses and nutrient removal. The results indicated that increasing influent CNR led to a decrease in microbial α-diversity and niche width. Genera involved in nitrogen removal and denitrification, such as Rhodobacter , Desulfovibrio , and Zoogloea , were enriched under medium/high CNR conditions, resulting in higher nitrate ( NO3--N ) removal (up to 99 %) than that under lower CNR conditions (<60 %). Environmental factors, including water temperature (WT), pH, and phosphorus (P), along with CNR-induced COD and NO3--N play important roles in microbial succession in HFCWs. The genus Nitrospira , which is involved in nitrification, exhibited a significant negative correlation ( p < 0.05) with WT, COD, and P. Co-occurrence network analysis revealed that increasing influent CNR reduced the complexity of the network structure and increased microbial competition. Analysis using null models demonstrated that the microbial community assembly in HFCWs was primarily driven by stochastic processes under increasing influent CNR conditions. Furthermore, HFCWs with more stochastic microbial communities exhibited better denitrification performance ( NO3--N removal). Overall, this study enhances our understanding of nutrient removal, microbial co-occurrence, and assembly mechanisms in CWs under varying influent CNRs.

Abstract:Black-odor water pollution in rural areas, especially swine wastewater, can lead to the deterioration of water quality and thus seriously affect the daily life of people in the area. However, there is a lack of effective treatment measures with simultaneous attention to carbon, nitrogen and sulfur pollution in rural black-odor water bodies. This study evaluated the feasibility of an in-situ pilot-scale constructed wetland (CW) for the synchronous removal of COD, ammonium, and sulfur compounds in the swine wastewater. In this study, the operation strategy of CW sediment pretreated with free nitrous acid (FNA) and Myriophyllum aquaticum plantation was established. Throughout the 114-day operation, the total removal efficiencies of COD and ammonium nitrogen in experimental groups were 81.2 ± 4.2 % and 72.8 ± 1.8 %, respectively, which were significantly higher than CW without any treatment. Removal efficiencies of Sulfur compounds, i.e. sulfide, sulfate, thiosulfate, and sulfite, were 92.3 ± 1.9 % (61.2 % higher than the no-treatment group), 42.1 ± 3.8 %, 97.9 ± 1.7 %, and 42.7 ± 4.5 % respectively. High-throughput sequencing and qPCR revealed that experimental group significantly increased denitrification genes (nirK, nosZ) and sulfur oxidation genes (soxB, fccAB) and enriched the corresponding microbial taxa (Bacillus, Conexibacter and Clostridium sensu stricto). Moreover, metabolic pathways related to nitrogen and sulfur and the degradation of organic matter were up-regulated. These results indicated that partial nitrification in CW planted with M. aquaticum promoted sulfur oxidation denitrification and heterotrophic denitrification. Overall, the in-situ pilot-scale study revealed that the cultivation of M. aquaticum in FNA-treated CW can be a sustainable approach to treat black-odor water bodies.

Abstract:Understanding of mechanisms in nitrous oxide (N2O) emission from constructed wetland (CW) is particularly important for the establishment of related strategies to reduce greenhouse gas (GHG) production during its wastewater treatment. However, plant biomass accumulation, microbial communities and nitrogen transformation genes distribution and their effects on N2O emission from CW as affected by different nitrogen forms in aquatic environment have not been reported. This study investigated the interactive effects of aquatic nitrogen and plant biomass on N2O emission from subsurface CW with NH4+-N (CW-A) or NO3−-N (CW–B) wastewater. The experimental results show that NH4+-N and NO3−-N removal efficiencies from CW mesocosms were 49.4% and 87.6%, which indirectly lead to N2O emission fluxes of CW-A and CW-B maintained at 213 ± 67 and 462 ± 71 μg-N/(m2·h), respectively. Correlation analysis of nitrogen conversion dynamic indicated that NO2−-N accumulation closely related to N2O emission from CW. Aquatic NH4+-N could up-regulate plant biomass accumulation by intensifying citric acid cycle, glycine-serine-threonine metabolism etc., resulting in more nitrogen uptake and lower N2O emission/total nitrogen (TN) removal ratio of CW-A compared to CW-B. Although the abundance of denitrifying bacteria and N2O reductase nosZ in CW-B were significantly higher than that of CW-A, after fed with mixed NH4+-N and NO3−-N influent, N2O fluxes and N2O emission/TN removal ratio in CW-A were extremely close to that of CW-B, suggesting that nitrogen form rather than nitrogen transformation microbial communities and N2O reductase nosZ determines N2O emission from CW. Hence, the selection of nitrate-loving plants will play an important role in inhibiting N2O emission from CW.

Abstract:Due to its low C/N ratio and high concentrations of nitrogen and phosphorus, the effluent of anaerobic cyanobacteria fermentation cannot be directly discharged without further treatment. To effectively reduce nutrient loads and utilize the nutrient resources of biogas slurry generated from the anaerobic digestion of stored algae, two different aerobic treatment units (AUs) were combined with an ecological treatment unit (EU) to create two different treatment systems. The two AUEU systems paired a constructed wetland (CW) with either a cascade biological contact reactor (CBCR) or a carrousel oxidation ditch reactor (CODR). In this paper, the water quality characteristics of biogas slurry were measured, and comprehensive experiments on the two trial-treatment systems were carried out to validate their performance in removing pollutants and utilizing resources. Furthermore, the pollutant removal efficiencies of the combined systems, along with the removal mechanisms and utilization of the nitrogen and phosphorus in the CWs, were also investigated. The results showed that the CWs, with aquatic vegetation, took up the majority of removed nitrogen and phosphorus by absorption, which effectively reduced the concentration of pollutants in the effluent and enabled the nitrogen and phosphorus to be reused in plants. Biomass assimilation by the absorption by vegetation took up 75.8%, 66.1%, 70.3%, and 86% of the removed NH4+-N, NOx−-N, TN, and TP, respectively.

Abstract:Treating wastewater with low carbon-to-nitrogen (C/N) ratios by constructed wetlands (CWs) is still problematic. Adding chemicals is costly and may cause secondary pollution. Configuring plant diversity in substrate-based CWs has been found to be a better way to treat low-C/N wastewater, but wastewater treatment in floating CWs needs to be studied. In this study, wastewater with C/N ratios of 5 and 10 were set in simulated floating CWs, and 9 combinations with plant species richness (SR) of 1, 3, and 4 were configured. The results showed that (1) increasing SR improved the total N mass removal (NMR) by 29% at a C/N ratio of 5 but not 10; (2) the presence of Oenanthe javanica in the microcosms increased the NMR by 13% and 20% with C/N ratios of 5 and 10, respectively; (3) increasing SR mitigated the net global warming potential (GWP) by 120% at a C/N ratio of 5 but not 10; and (4) a Hemerocallis fulva × O. javanica × Echinodorus parviflours × Iris hybrids mixture resulted in a high NMR and low net GWP. In summary, assembling plant diversity in floating CWs is an efficient and clean measure during the treatment of wastewater with a C/N ratio of 5.

Abstract:A pilot hybrid constructed wetland (CW) planted with reeds (Phragmite australis) and rice (Oryza sativa L.) was designed to treat liquid anaerobic digestate in the Yangtze River Delta, China. The hybrid CW system was composed of four stages: two reed vegetated vertical subsurface flow beds (VSSF: U1 and U2) in sequence, followed by a reed vegetated horizontal subsurface flow bed (HSSF: U3) and a rice vegetated surface flow bed (SF: U4). The average loading rate of digestate was 3.6 m3 per day during the experimental period.The average concentrations of TN and TP in the influent were 379 ± 58 mg L−1 and 29.6 ± 9.2 mg L−1, while the average removal efficiencies of TN and TP were 94.6% and 88.4%. Both TN and TP removal efficiencies in the second VSSF containing zeolite gave the highest removal performance, in which the mass removal rates were 21.3 ± 8.0 g-N m−2 d−1 and 0.99 ± 0.69 g-P m−2 d−1, respectively. Similarly, the highest removal performance for COD was also observed in the second VSSF with a mass removal rate of 79.9 ± 72.4 g-COD m−2 d−1.On the other hand, the average CH4 and N2O fluxes were highest in the first VSSF, at 31.8 ± 12.9 mg m−2 h−1 and 3.7 ± 2.8 mg m−2 h−1, respectively. There was a significant linear relationship between CH4 flux and DOC concentration in the pore water as well as a correlation between N2O flux and TN concentration. Total GWP of the hybrid CWs, derived from CH4 and N2O emissions, was 792.4 kg CO2-eq, of which CH4 and N2O emissions accounted for 66.0% and 34.0%, respectively. Consequently, the hybrid CWs emitted on average 0.93 kg CO2-eq to remove 1 kg COD while the average EF of TNin was 0.34%, suggesting that the use of multistage hybrid CWs could be efficiency-wise and environmentally a promising strategy for anaerobic digestate treatment.

Abstract:Constructed wetlands (CWs) have been proved to be an alternative to the treatment of various wastewater. However, there are few studies focused on the removal performance and mechanisms of pollutants in pilot-scale CWs packed with novel solid carbon. In this study, we investigated the effect of poly-3-hydroxybutyrate-co-3hydroxyvalerate/polyacetic acid (PHBV/PLA) blends as carbon source on pollutant's transformation, microbial communities and functional genes in pilot-scale aeration-anoxic two-stage CWs for polishing rural runoff in southern China. Results showed a striking improvement of TN removal in CWs with PHBV/PLA blends (64.5%) compared to that in CWs with ceramsite (52.9%). NH4+-N (61.3-64.6%), COD (40.4-53.8%) and TP (43.6-47.1%) were also removed effectively in both two CWs. In addition, the strains of Rhodocyclaceae and Bacteroidetes were the primary denitrifiers on the surface of PHBV/PLA blends. Further, the aerobic stage induced gathering of 16 S and amoA genes and the anoxic zone with PHBV/PLA blends increased the nirS genes, which fundamentally explained the better denitrification performance in CW based on PHBV/PLA blends. Consequently, this study will provide straightforward guidance for the operation of engineering CWs packed with polymers to govern the low-C/N rural wastewater.

Abstract:Agricultural biomass waste in rural areas has been identified as an economical solid carbon sources in constructed wetlands (CWs) for treating low C/N ratio domestic sewage. However, little information is available regarding its optimal utilization as a media amendment for enhancing nitrogen removal in CWs. In this study, vertical flow CWs with different walnut peel amendment proportions (0%, 25%, 50%, 75%) were developed to explore the effects of biomass dosage on the treatment performance, nitrous oxide (N2O) emission and microbial metabolites. Results showed that the addition of biomass significantly enhanced the denitrification performance in all CWs, and the higher total nitrogen (TN) removal efficiency (91.14-97.16%) was achieved in CWs with the optimal dosage of 25%. While the addition of biomass resulted in a slight increase in N2O emission (20.56-270.13 μg m-2 h-1) compared with control systems. Additionally, the biomass addition increased the accumulation of extracellular polymeric substances (EPS) by facilitating microbial processes. Higher total EPS production was observed in CW with 25% biomass, and the proportion of tightly bound EPS (48%) dominated in the total EPS in different CWs.

Abstract:To synchronously decrease the risk of secondary pollution from decomposing litter of the widely planted warm-season macrophytes (WSMs) and improve nitrogen (N) removal in the early cold season, a strategy of planting both cold-season macrophytes (CSMs) and WSMs (in the form of litter, WSML) in a single unit of constructed wetlands (CWs) was tested in pilot-scale floating CWs. The interaction between CSMs and WSML as well as the pathway of N removal was examined. CSMs stimulated carbon source release but inhibited N and phosphorus (P) release from WSML, while WSML inhibited CSM growth, resulting in a decrease in N and P absorption by CSMs. The average removal efficiencies (REs) of total nitrogen (TN), NH4+-N and NO3−-N were 2.0%, 8.8% and 14.9% for the WSML-only treatment and 25.8%, 90.3% and 25.7% for the CSM-only treatment, respectively. In contrast, the combined CSM and WSML treatment obtained average REs of 44.8%, 94.7% and 48.9% for TN, NH4+-N and NO3−-N, respectively. WSML was not only the source of NH4+-N but also the possible pathway of its removal by adsorption. CSM absorption is an important pathway for NH4+-N and NO3−-N removal, especially the former. Therefore, planting both CSMs and WSMs in a single unit is recommended to reduce the risk from the current overreliance on WSMs for most CWs.

Abstract:The inefficient nitrogen removal in constructed wetlands (CWs) can be attributed to insufficient carbon sources for low carbon-to-nitrogen (C/N) ratio wastewater. In this study, sugarcane bagasse fermentation liquid (SBFL) was used as a supplemental carbon source in intermittently aerated CWs to enhance nitrogen removal. The impact of different regulated influent C/N ratios on nitrogen removal and greenhouse gas (GHG) emissions was investigated. Results demonstrated that SBFL addition significantly enhanced the denitrification capacity, resulting in faster NO 3 - -N removal compared to sucrose. Moreover, intermittently aerated CWs significantly improved NH 4 + -N removal efficiency compared to non-aerated CWs. The highest total nitrogen removal efficiency (98.3 %) was achieved at an influent C/N ratio of 5 in intermittently aerated CWs with SBFL addition. The addition of SBFL resulted in a reduction of N 2 O emissions by 17.8 %–43.7 % compared to sucrose. All CWs exhibited low CH 4 emissions, with SBFL addition (0.035–0.066 mg·m -2 h -1 ) resulting in lower emissions compared to sucrose. Additionally, higher abundance of denitrification ( nirK, nirS and nosZ ) genes as well as more abundant denitrifying bacteria were shown in CWs of SBFL inputs. The results of this study provide a feasible strategy for applying SBFL as a carbon source to improve nitrogen removal efficiency and mitigate GHG emissions in CWs.
