Abstract:The ability to detect multiple microRNAs (miRNAs) as cancer biomarkers with high sensitivity and good compatibility is still challenging. Here, we showed an ultrasensitive sensing system based on the fluorescence resonance energy transfer (FRET) effect between green-emitting and blue-emitting core@shell upconversion nanoparticles (UCNPs) and MoS2 nanosheets for simultaneously detecting two miRNAs. The core@shell UCNPs and MoS2 nanosheets served as energy donors and quenchers, respectively. The green-emitting and blue-emitting core@shell UCNPs were modified with single-stranded DNAs complementary to two breast cancer miRNA biomarkers, miR-593 and miR-155, respectively. When the two target miRNAs were absent in the samples, the ssDNA-modified nanoparticles bound to the MoS2 nanosheets directly through van der Waals forces, leading to the close proximity between the nano particles and nanosheets and thus the occurrence of FRET between them. When the two target miRNAs were present in the samples, each of the miRNAs hybridized with the complementary ssDNA on the nanoparticles, preventing the direct contact between the nanoparticles and the nanosheets and thereby causing miRNA concentration-dependent fluorescence recovery of the nanoparticles. Thus, monitoring the fluorescence recovery from the nanoparticles established a unique linear relationship between the fluorescence recovery and the target miRNA concentrations, allowing us to determine the concentration of the miRNAs by simply detecting the fluorescent signals. Our FRET sensor could reach an ultralow limit of detection (-0.17 nM and-0.25 nM for miR-593 and miR-155, respectively). It could selectively detect the target miRNAs but not the non-target molecules. Our sensor can be extended to the ultrasensitive detection of other biomolecules and holds promise for the early diagnosis of diseases such as breast cancer. (C) 2021 The Author(s). Abstract:MicroRNA-21 (miRNA-21) is currently the only known oncogenic miRNA that is upregulated in almost all malignant tumors and exhibits a broad spectrum of tumor recognition characteristics. It holds significant value in the early diagnosis, malignant degree assessment, and prognostic evaluation of tumors. In this study, a novel dual-mode self-powered sensing platform is developed using Au nanoparticles/graphdiyne as the electrode substrate and combined with DNA nanoring for highly sensitive and specific detection of miRNA-21. The DNA nanoring structure, which is easy to prepare and contains multiple recognition sites, induces significant electrochemical/colorimetric signal responses of the signaling molecule methylene blue. Under optimal conditions, the linear ranges of the electrochemical and colorimetric detection modes of this self-powered sensor are 0.1 fM-100 pM and 0.1 fM-10 nM, respectively, with the detection limits of 35.1 aM and 61.6 aM (S/N=3). This strategy provides a new reference for the sensitive detection of microRNA and has immense potential for application in the screening and detection of clinical nucleic acid diseases.
Sensitive detection of cancer biomarker with enzyme-free mediated cascade signal amplification empowered undisturbed dual-mode assay

Abstract:Cancer is a serious threat to human health, and early identification of cancer biomarkers is crucial for timely treatment. A dual-mode biosensing platform has been developed to identify colon carcinoma suppressor microRNA-199a (miRNA-199a). A biofuel cell is firstly constructed using hybridization chain reaction and catalytic hairpin assembly self-assembly. The anode is a flexible carbon cloth (CP) loaded with glucose oxidase-functionalized molybdenum disulfide nanorods (MoS 2 nanorods), and the bio-cathode is made up of double-stranded deoxyribonucleotide chains generated by nucleic acid amplification technology. The linear range of the electrochemical method is 0.1 fM-100 pM, and the limit of detection is 24.1 aM ( S/N = 3 ). The colorimetric method has a linear range of 0.1 fM-10000 pM with detection limit of 34.5 aM ( S/N = 3 ). The combination of MoS 2 nanorods, enzyme-free cascade signal amplification technology, and dual-mode detection strategy provides high sensitivity and accuracy, offering a promising method for accurate determination of cancer biomarkers.

Abstract:MicroRNAs (miRNAs) are closely bound up with the occurrence and deterioration of some cancers, and their early diagnoses have aroused great interest. However, stable and ultra-sensitive real-time detection remains challenge. In this work, we report a strategy for real-time ultrasensitive detection of miRNA-21 by capacitor/enzymatic biofuel cell based self-powered biosensor (EBFC-SPB) with hybridization chain reaction (HCR) for signal amplification. The nitrogen doped dodecahedral hollow carbon (N-DHC) is used for high load, exonuclease III (EXO III)-aided target cells for accurate recognition, and HCR for signal amplification. In addition, the detected signals are further enhanced by a good match with the commercial capacitor, and the real-time portable detection of tumor markers can be achieved by directly reading the data with an Android smartphone. The detection limit of the method is 0.04 fM (S/N = 3) in the linear range of 0.01–5.0 × 103 fM. This method provides reference and theoretical support for the early diagnosis of related tumor markers.

Abstract:Rapid and efficient detection of microRNA (miRNA) of breast cancer 1 gene mutation (BRCA1) at their earliest stages is one of the crucial challenges in cancer diagnostics. In this study, a highly-sensitive electrochemical DNA biosensor was fabricated by double signal amplification (DSA) strategy for the detection of ultra-trace miRNA of BRCA1. In the presence of target miRNA of BRCA1, the well-matched RNA-DNA duplexes were specifically recognized by double-strand specific nuclease (DSN), and the DNA part of the duplexes were then cleaved and miRNAs were released to trigger another following cycle, which produced a primarily amplified signal by such a cyclic enzymatic signal amplification (CESA). Then triple-CdTe quantum dot labelled DNA nanocomposites (3-QD@DNA NC) was selectively hybridized with the cleaved DNA probe on the electrode and produced multiply amplified signals. The biosensor exhibited a high sensitivity for the detection of miRNA of BRCA1 in concentrations ranging from 5 aM to 5 fM, and its detection limit of 1.2 aM was obtained, which is two or three orders of magnitude lower than those by single signal amplification strategy such as CESA or QD-labeled DNA probes. The as-prepared biosensor was successfully used to detect the miRNA of BRCA1 in human serum samples with acceptable stability, good reproducibility, and good recovery. The proposed DNA biosensor based on double signal amplification strategy provided a feasible, rapid, and sensitive platform for early clinical diagnosis and practical applications.

Abstract:Specific and sensitive microRNAs (miRNAs) detection is essential to early cancer diagnosis. The development of these technologies including functional nuclease-mediated target amplification and DNA nanotechnology possesses tremendous potential for the high-performance detection of miRNAs in the accurate diagnosis of disease. In this study, we have established an ultrasensitive electrochemical biosensor by combining nicking endonuclease-assisted primer exchange reaction (PER) cascade amplification with a DNA nanosphere (DNS)-mediated electrochemical signal-enhanced system for the detection of miRNA-21 (miR-21). The cascade amplification is initiated by a nicking endonuclease that can cleave specific DNA substrates and highly amplify translation of the target to single-stranded DNA fragments (sDNA). Then, the PER cascade is powered by strand-displacing polymerase and generates a large amount of nascent single stranded connector DNA (cDNA) via sDNA triggering of the dumbbell probe (DP), thus achieving the cascade amplification of the target. Finally, the DNS loaded with plenty of electroactive substances can be captured on the electrode via cDNA for further enhancing the electrochemical signal and highly sensitive detection of miR-21. The proposed electrochemical biosensor exhibits a wide detection range of 1 aM to 0.1 nM and a low detection limit of 0.58 aM. The excellent selectivity allows the biosensor to discriminate miR-21 from other miRNAs, even the one base-mismatched sequence. Moreover, the practicability of the biosensor is investigated by analyzing miR-21 in human serum and cancer cell lysate. Therefore, our proposed nicking endonuclease-assisted PER cascade amplification strategy provides a powerful platform for the early detection of miRNA-related disease and molecular diagnosis.

Abstract:Ultrasensitive detection for low concentration and abnormal expression of microRNA-155 (miR-155) in serum (fM level) at the early stage of cancer still exist great challenges. Therefore, a dual-model photoelectrochemical biosensor was developed based on label-free trimetallic nanoprobe and Mg 2+ dependent DNAzyme driven DNA walker to generate ratiometric dual-signal output for sensitive biosensing of miR-155 from breast cancer cells (MCF-7) and cervical cancer cells (HeLa). In signal amplification strategy, Blue nanozyme was selected as a signal nanoprobe with high catalytic activity and large specific surface area, while two dimensional MXene/AuNPs as surface modification material was applied to improve the active sites of the electrode. With the presence of the target, the DNAzyme assisted DNA walker process was initiated and abundant signal nanoprobe was introduced to the electrode surface, producing high current response and weak ultraviolet-visible spectrophotometry (UV–vis) response. Hence, the biosensor for miR-155 shows a low limit of detection of 1.2 fM from the linear range of 5 fM to 5 nM, exhibiting excellent stability and reproducibility. Moreover, the proposed biosensor demonstrates superior performance for miR-155 in complex human serum samples and tumor cell lysates, presenting great application potential for early-stage clinical diagnosis.

Abstract:The pivotal role of microRNA (miRNA) in cancer diagnosis and monitoring is well recognized, yet current detection methods remain complex and costly, given the rarity of tumor miRNA in the bloodstream. This study proposes a simple and elegant miRNA sensing platform utilizing duplex-specific nuclease (DSN) and ultrasensitive graphene field-effect transistor (GFET) biosensors. The DSN-assisted GFET enables preamplification-free detection of miR-21, a promising cancer biomarker, at dilutions down to 25 aM. Along with the successful validation of its excellent specificity using clinical plasma samples without RNA extraction process, this innovative approach holds great promise for improving cancer diagnostics and monitoring.
Reliable Breast Cancer Mirnas Detection with Enhanced Silicon Nanowire Biosensor by PNA Probe and Optical Calibration

Abstract:MicroRNAs (miRNAs) have attracted significant attention in the field of cancer research as a promising class of biomarkers. However, precise, sensitive, and specific detection of miRNAs still confronts challenges due to their dynamic expression, low abundance, and high sequence similarity among families. The highly sensitive silicon nanowires (SiNWs) biosensors are limited by little interface discrepancy within the SiNWs, which has the potential to affect the final output results. In this study, the calibration of the SiNWs biosensor was initially suggested to be conducted through photo response. This approach successfully mitigated the impact of preparation and modification procedures, resulting in an enhanced correlation between the biosensor and q-PCR for the identification of breast cancer miRNA. Specifically, the correlation coefficient was raised from below 0.5 to above 0.8. Furthermore, the uncharged peptide nucleic acid (PNA) was used for device modification in order to address the issue of detecting miRNAs with a total length in below 0.01×PBS buffer. It was shown that the PNA probe exhibited greater sensitivity compared to the DNA probe in 0.001×PBS buffer. Significantly, the improved biosensor exhibited favorable selectivity and was capable of identifying single base mismatch. The sensor exhibited a high level of sensitivity in detecting miRNA within a concentration range of 1 fM to 10 pM when applied to actual human blood samples. The biosensor exhibited an excellent level of reproducibility in the analysis of spiked samples, achieving a recovery rate of 91 %, without requiring RNA extraction or amplification procedures. The biosensor has the capability to directly detect miRNA in authentic clinical samples, hence demonstrating significant promise for the timely diagnosis of cancer via the use of miRNA as biomarkers.

Abstract:The detection of trace tumor-related serum miRNA biomarkers is in great demand for the early diagnosis of cancer. Herein, for the first time, an electrochemical sensing platform based on atom transfer radical polymerization (ATRP) signal amplification strategy for ultrasensitive determination of the breast and prostate cancer marker miRNA-141 has been developed. The hairpin DNAs were immobilized on the benzoic acid modified electrode to capture the target miRNA-141, the recognition of miRNA-141 released thiol groups on the end of probes, followed by the association of ATRP initiators modified gold nanoparticles with thiol groups, and then triggered the polymerization on electrode surface, causing a great number of ferrocene (Fc) signal molecules grafted on the sensor interface. As a result, the electrochemical signal intensity of signal molecule has been greatly increased. The proposed biosensor has a linear range from 10 pM to 10 aM with a detection limit of 3.23 aM for miRNA-141, opening a new and promising path for ultrasensitive analysis of tumor-related miRNAs.

Abstract:MicroRNAs (miRNAs) are important cancer-associated biomarkers for early detection. However, accurately measuring miRNAs is challenging due to their low abundance and high homogeneity in human serum. CRISPR technology can greatly improve the specificity and sensitivity of detection in biosensor construction. In this study, we develop a remarkably responsive self-powered biosensing platform for detecting microRNA-21 (miRNA-21) using hybridization chain reaction (HCR) and CRISPR/Cas12a amplification cascade mediated by 3D DNA walkers. Carbon-coated molybdenum disulfide nanotubes with a large surface area and excellent conductivity are fabricated on flexible carbon paper as the anode and cathode for biological fuel cell construction. Glucose oxidase is loaded onto the anode surface, while serving as the biological cathode, it initiated HCR, generating abundant double-stranded DNA. The negatively charged DNA backbone captures electron acceptors [Ru(NH 3 ) 6 ] 3+ through electrostatic adsorption, resulting in a significant open circuit voltage ( E OCV ) signal. Through CRISPR/Cas12a amplification cascade mediated by 3D DNA walkers, a smaller E OCV is produced, enabling sensitive target detection. The method exhibits a detection range spanning from 0.0001 to 10,000 pM and capable of detecting as low as 19.0 aM ( S/N = 3 ). This highly selective and sensitive method provides an effective approach for accurately detecting tumor markers in clinical applications.

Abstract:The mechanism by which xylazine raises plasma glucose but lowers insulin concentrations was examined in Holstein cows. Intravenous injection of xylazine (15-150 micrograms/kg) induced a dose-dependent hyperglycemia and hypoinsulinemia for 3-4 h. An alpha 2-adrenergic blocking agent, yohimbine, and an alpha 1- and alpha 2-adrenergic blocking agent, phentolamine, at 500 micrograms/kg each reduced or abolished xylazine-induced hyperglycemia and hypoinsulinemia. The alpha 1-adrenergic blocking agents, prazosin and phenoxybenzamine, at 500 micrograms/kg each did not exert such antagonism. Prazosin at the dose studied even prolonged xylazine-induced hypoinsulinemia. The alpha-adrenergic blocking agents alone at 500 micrograms/kg each did not change either plasma glucose or insulin concentrations. The present study suggests that xylazine-induced hyperglycemia and hypoinsulinemia are mediated by alpha 2-adrenergic receptors, possibly in beta-cells of pancreatic islets which inhibit the release of insulin. The present study further suggests the use of xylazine as a pharmacological tool in the study of adrenergic influence on in vivo insulin release.

Abstract:Achieving sensitive detection and accurate identification of cancer cells is vital for diagnosing and treating the disease. Here, we developed a logic signal amplification system using DNA tetrahedron-mediated three-dimensional (3D) DNA nanonetworks for sensitive electrochemiluminescence (ECL) detection and subtype identification of cancer cells. Specially designed hairpins were integrated into DNA tetrahedral nanostructures (DTNs) to perform a catalytic hairpin assembly (CHA) reaction in the presence of target microRNA, forming hyperbranched 3D nanonetworks. Benefiting from the "spatial confinement effect," the DNA tetrahedron-mediated catalytic hairpin assembly (DTCHA) reaction displayed significantly faster kinetics and greater cycle conversion efficiency than traditional CHA. The resulting 3D nanonetworks could load a large amount of Ru(phen)32+, significantly enhancing its ECL signal, and exhibit detection limits for both miR-21 and miR-141 at the femtomolar level. The biosensor based on modular logic gates facilitated the distinction and quantification of cancer cells and normal cells based on miR-21 levels, combined with miR-141 levels, to further identify different subtypes of breast cancer cells. Overall, this study provides potential applications in miRNA-related clinical diagnostics.

Abstract:The variations in microRNA (miRNA) expression levels can be useful biomarkers for the diagnosis of different cancers. In this work, on the basis of a new miRNA-triggered molecular machine for enzyme-free target recycling signal amplification, the development of a simple electronic sensor for highly sensitive detection of miRNA-21 from human breast cancer cells is described. The three-stand DNA duplex probes are self-assembled on the gold electrode surface to fabricate the sensor. The miRNA-21 target binds to the terminal toehold region of the probes, displaces one of the short strands through toehold-mediated strand displacement reactions, and exposes the secondary toehold region for subsequent hybridization with the methylene blue (MB)-modified DNA fuel strand, which further displaces both the miRNA-21 target and the other short strand to activate the operation of the molecular machine. As a result, the miRNA-21 target is cyclically reused, and many MB-DNA fuel strands are attached to the sensor surface, leading to a significantly amplified current response for sensitive detection of miRNA-21 down to 1.4 fM. The developed sensor also shows high sequence discrimination capability and can be used to monitor miRNA-21 expression levels in cancer cells. Moreover, this sensor avoids the involvement of any enzymes for target recycling amplification and features with highly minimized background noise for miRNA detection, which makes this method hold great potential for convenient monitoring of different miRNA biomarkers for early diagnosis of various cancers.

Abstract:Malignant tumors are significant diseases that pose a threat to human life and health. The advancement of highly sensitive and specific methods is of excellent significance. This study proposes a novel detection platform that employs a smartphone-assisted self-powered sensing system with binary 3D DNA Walkers mediated CRISPR/Cas12a system and ultra-thin 2D graphdiyne for ultrasensitive dual-mode detection of breast cancer biomarker microRNA-let-7a. The binary 3D DNA Walkers possess high efficiency in movement and multifunctional characteristics. When combined with the CRISPR/Cas12a system, they can retain the advantages of 3D DNA Walker while releasing a large of trigger strands complementary to CRISPR RNA, thereby greatly enhancing the sensitivity of the sensor. In addition, capacitors are also integrated into the sensing system to further amplify the detection signal, resulting in a 20.7-fold increase in sensitivity of the method. Additionally, the adoption of an electrochemical/colorimetric dual-mode detection strategy significantly enhances the accuracy of the new approach. The newly developed detection method exhibits a linear range of 0.0001–10000 pM with a detection limit of 8.11 aM (S/N = 3 ). This work offers a reliable portable sensor for sensitive and accurate detection of tumor biomarkers, enabling remote disease sensing and on-site monitoring applications.

Multiple signal amplification strategy induced by biomarkers of lung cancer: A self-powered biosensing platform adapted for smartphones

Abstract:Lung cancer is a major malignant cancer with low survival rates, and early diagnosis is crucial for effective treatment. Herein, a biosensing platform that is self-powered derived from a capacitor-coupled EBFC has been developed for ultra-sensitive real-time identification of microRNA-21 (miRNA-21) with the assistance of a mobile phone. The flexible substrate of the platform is prepared on a carbon paper modified with graphdiyne and gold nanoparticles. The biosensor employs DNAzyme-mediated dual strand displacement amplification, which enhances the signal output intensity of the EBFC and improves selectivity. The coupling of the capacitor with the EBFC significantly amplifies the sensing signal, causing a 10.6-fold surge in current respond and further improving the sensitivity of the sensing platform. The established detection approach demonstrates a linear relationship varied from 0.0001 to 10,000 pM, with a sensitivity down to 32.3 aM as the minimum detectable limit, which has been effectively utilized for detecting miRNA-21 in practical samples. This sensing system provides strong support for the construction of portable detection devices, and the strategy of the platform construction provides an effective method for ultra-sensitive and accurate detection of miRNA, holding great potential in clinical diagnosis, prognosis evaluation, and drug screening for cancer.

Abstract:The latest research shows that the expression level of microRNA-141 can predict the number of prostate cancer cells in the human body and has become an important biomarker. In this paper, an all-carbon sandwich self-powered biosensor based on graphene and carbon cloth is constructed for the highly sensitive detection of the prostate tumor marker miRNA-141. First, gold nanoparticles modified carbon cloth is applied for substrate electrode, and bilirubin oxidase is then immobilized on it to prepare the biocathode of the biofuel cell. Then, aptamer 1 is immobilized on gold nanoparticles-modified carbon cloth as the electrode substrate. The bioconjugate is prepared by immobilizing the aptamer 2-glucose oxidase complex on gold nanoparticles/graphene. In the biofuel cell-based self-powered sensing system, when the target microRNA-141 is present, it undergoes complementary base pairing with aptamer 1 and aptamer 2, and the bioconjugates are immobilized on the anode to form the sandwich structure. The enzyme on the anode undergoes an oxidation reaction to catalyze the reduction of oxygen, and the electrochemical respond of the system increases significantly. The results show that the concentration of microRNA-141 is proportional to the open-circuit voltage value ranging from 0.0001 to 1000 pmol/L with a detection limit of 50 amol/L (S/N = 3). The method has high sensitivity and excellent selectivity and can be applied to sensitively detect tumor marker microRNA-141 in biological matrix.

Abstract:MicroRNAs (miRNAs) are crucial regulators in various pathological and physiological processes, and their misregulation is a hallmark of many diseases. In this study, we introduce an advanced DNA nanomachine using split-type molecular beacons (STMBs) for sensitive detection of miR-21, a key biomarker in cancer diagnostics. Utilizing an innovative STMB-mediated cascade strand displacement amplification (STMB-CSDA) technique, our approach offers a powerful means for the precise quantification of miRNAs, using miR-21 as a primary example. The system operates through target-induced linkage of STMBs, initiating a series of strand displacement amplifications resulting in exponential signal amplification. Coupled with the precision of T4 DNA ligase, this mechanism translates minimal miRNA presence into significant fluorescence signals, offering detection sensitivity as low as 5.96 pM and a dynamic range spanning five orders of magnitude. Characterized by its high specificity, which includes the ability to identify single-base mismatches, along with its user-friendly design, our method represents a significant leap forward in miRNA analysis and molecular diagnostics. Its successful application in examining total RNA from cancer cells and clinical serum samples demonstrates its immense potential as a groundbreaking tool for early cancer detection and gene expression studies, paving the way for the next generation of non-invasive diagnostics in personalized healthcare.
