Abstract:Signal-amplification sensing strategies for rapid and accurate detection of tumor marker miRNA is of great practical significance for cancer prevention and treatment. Catalytic hairpin assembly (CHA) nucleic acid amplification reaction and CRISPR/Cas12a strategies have exhibited huge prospect due to their flexible design and efficient signal amplification. We constructed an electrochemiluminescence (ECL) biosensor based on horseradish peroxidase/gold nanoparticles (HRP/Au NPs) nanocomposite probe and CRISPR/Cas12a (LbCpf1), and completed a dual-mode detection of miRNA-21. When the target RNA is present in the system, the CHA) cycle was initiated, and the double-stranded DNA containing a specific protospacer adjacent motif (PAM) base sequence was generated to successfully activate the trans-cut activity of CRISPR/Cas12a. After the prepared HRP/Au NPs nanocomposite probe was cleaved by CRISPR/Cas12a, the supernatant containing HRP was collected through magnetic separation and subsequently introduced to the cathode of the bipolar electrode facilitating anodic ECL luminescence response. In addition, the supernatant was added to the TMB chromogenic solution for chromogenic reaction. Thus, ECL detection and visual detection of miRNA-21 were realized. Dual-mode detection improves the accuracy of the sensor, and the utilization of bipolar electrodes on indium tin oxide (ITO) coated glass could also provide some useful references for further development of point-of-care detecting devices for early cancer prevention and diagnosis.

Abstract:The latest in a series of speculations concerning the origins of HIV a Rolling Stone magazine article claiming that AIDS may have been introduced through the polio vaccines has touched off angry responses from vaccine experts including several who claim to have been misrepresented by the article. In an article entitled "The Origin of AIDS" found in the current issue of Rolling Stone free-lance writer Tom Curtis hypothesizes that AIDS first entered humans in Africa as a contaminant of an oral polio vaccine widely administered in the late 1950s a vaccine formulated at Philadelphias Wistar Institute by Hilary Koprowski. Curtis speculates that a still-undiscovered simian form of HIV may have contaminated Koprowskis polio vaccine developed from monkey kidney cell cultures. Some researchers say that the articles claim is not so far-fetched considering that simian viruses were found to contaminate some early polio vaccines. Most researchers however dismiss the theory as far too speculative arguing that no supporting evidence exists. Both the Wistar and the Center for Disease Control (CDC) have come out against the theory. Koprowski maintains that the existing data refutes Curtiss hypothesis since in preparing the vaccine he used wild macaques from the Philippines and India and no wild macaques have ever tested positive for a lentivirus. In response to Koprowskis claim Curtis quoted the chief of CDCs retrovirus branch Thomas Folks who offered a scenario in which the monkey kidney cells might have become infected. Folks however has angrily accuses Curtis of selective editing. Others interviewed by Curtis have also claimed to have been misrepresented. Despite the rejections the Wistar Institute has announced that it will form a committee to evaluate the theory.

Abstract:MicroRNAs (miRNAs) are a category of short non-coding RNA molecules which play essential roles in regulating gene expression. Correspondingly, the aberrant expression of miRNAs is commonly related to the development of various diseases including cancers and cardiovascular diseases. Hence, miRNA is considered as an ideal biomarker for disease diagnosis and its quantification is highly demanded. Herein, an isothermal and enzyme free miRNA assay based on catalytic hairpin assembly (CHA) amplification coupled with inductively coupled plasma mass spectrometry (ICP-MS) detection is proposed. Two hairpin probes are employed in CHA reaction with rare earth element tag on H1 for ICP-MS detection and biotin on H2 for immobilization on magnetic beads. In the presence of target miRNA, the two independent probes open up their stem-loop structure and hybridize. The target miRNA will be released after the hybridization to trigger another CHA reaction. Ultimately, one target miRNA molecule promotes a number of CHA cycles and produces several copies of hybridized complex leading to the enhanced ICP-MS signal. Based on this approach, miRNA-141 was successfully analyzed with high sensitivity and specificity. The limit of detection (LOD) was 88 fmol with a linear range 0.1-2 pmol. Compared with fluorescence mediated CHA method and ICP-MS mediated basic sandwich method, the current strategy displayed the lowest LOD. The developed method was also applied to analyze miRNA-141 and miRNA-21 simultaneously to demonstrate its multiple detecting capability and practical applicability. Furthermore, this miRNA assay has the potential to become a high-throughput method by employing more distinct rare earth elements.

Abstract:Robust and sensitive methods for the detection of microRNAs (miRNAs) are crucial in the clinical diagnosis of cancers. In this study, a novel electrochemical biosensor with high sensitivity for miRNA-21 detection is developed, which relies on the formation of a peptide nucleic acid (PNA)-DNA hetero-three-way junction (H3WJ) and target-recycling catalytic hairpin assembly (CHA) amplification. The electroneutral PNA probes are initially immobilized onto a gold electrode to construct the sensor. Upon introduction of miRNA-21, target-recycling CHA is initiated, resulting in abundant double-stranded CHA products. Subsequently, association between the PNA probes and these products leads to the formation of PNA-DNA H3WJs. Consequently, the electrode surface is densely populated with numerous electroactive Ferrocene (Fc) groups, resulting in a significantly amplified current response for highly sensitive detection of miRNA-21 at concentrations as low as 0.15 fM. This approach demonstrates remarkable specificity towards target miRNAs and can be utilized for quantitative monitoring of miRNA-21 expression in human cancer cells. More importantly, the sensor exhibits exceptional stability and shows a significant reduction in background noise during miRNA detection, making this method a highly promising sensing platform for monitoring various miRNA biomarkers to facilitate the diagnosis of diverse cancers.

Abstract:The cytosines in cluster-nucleation sequences play a vital role in the formation of silver nanoclusters (Ag NCs). Here, an innovative electrochemiluminescence (ECL) resonance energy transfer (RET) sensing system was developed using CdS quantum dots (QDs) as ECL donor and Ag NCs as ECL acceptor. Modulation of the number of cytosines in the cluster-nucleation sequences allowed tuning of Ag NCs absorption bands to match with the ECL emission spectrum of CdS QDs, yielding effective ECL-RET. The sensitivity of detection was improved by dual-target recycling amplification based on duplex-specific nuclease (DSN) and catalytic hairpin assembly. In the presence of target microRNA-21 (miRNA-21), DSN selectively cleaved the complementary DNA section (S1), resulting in the release of the transduction section (S2) and the reuse of miRNA-21 in the next recycling amplification. Interaction of the stem-loop structure of the DNA1 segment (H1) on CdS QDs-modified electrode with S2 led to the opening of the hairpin structure of H1 and the formation of H1:S2 duplex. Then, hairpin DNA2 encapsulated Ag NCs hybridized with the remaining single-stranded DNA segment of H1, and the S2 strand was replaced. Finally, the dissociated S2 participated in subsequent reaction cycles, introducing Ag NCs to the electrode surface and leading to ECL signal quenching of the CdS QDs. The proposed sensor showed excellent performance in detecting miRNA-21 at a wide linear range from 1 fM to 100 pM. The practical application ability of the strategy was tested in HeLa cells with acceptable results, suggesting that the detection platform is a promising approach for disease diagnosis and molecular biology research.

Abstract:<p>Multi-target detection has been widely applied for the sensitive measurement of cancer-related biomarkers; however, the design and application of single platforms for diverse target detection are still challenging. Herein, a robust and sensitive electrochemiluminescence (ECL) biosensing platform was constructed for the measurement of microRNA-21 (miRNA-21) and mucin 1 (MUC1) based on dual catalytic hairpin assembly (DCHA). The catalytic hairpin assembly (CHA) process (Cycle I) was initiated by the target miRNA-21 to introduce abundant CdS:Mn quantum dots (CdS:Mn QDs) on the electrode surface, leading to a considerable ECL response and the sensitive detection of miRNA-21 with a limit of detection as low as 11 aM. Subsequently, the second CHA process (Cycle II) was triggered by the MUC1-aptamer complex, which allowed copious amounts of Au nanoparticles (AuNPs) to approach the CdS:Mn QDs. A decreased ECL signal was obtained due to the ECL resonance energy transfer (ECL-RET) effect between the CdS:Mn QDs and AuNPs; meanwhile, MUC1 was sensitively detected with a limit of detection of 0.40 fg mL<sup>-1</sup>. This single sensing platform achieved dual cancer-related biomarker detection, which could provide a rational approach for future clinical analyse.</p>

High-Sensitivity Homogeneous Detection of miRNA-155 Governed by DNA Walker-Regulated Surface DNA Density of Magnetic Electrochemiluminescence Nanoparticles

Abstract:Homogeneous electrochemiluminescence (ECL) has gained attention for its simplicity and stability. However, false positives due to solution background interference pose a challenge. To address this, magnetic ECL nanoparticles (Fe3O4@Ru@SiO2 NPs) were synthesized, offering easy modification, magnetic separation, and stable luminescence. These were utilized in an ECL sensor for miRNA-155 (miR-155) detection, with locked DNAzyme and substrate chain (mDNA) modified on their surface. The poor conductivity of long-chain DNA significantly impacts the conductivity and electron transfer capability of Fe3O4@Ru@SiO2 NPs, resulting in weaker ECL signals. Upon target presence, unlocked DNAzyme catalyzes mDNA cleavage, leading to shortened DNA chains and reduced density. In contrast, the presence of short-chain DNA has minimal impact on the conductivity and electron transfer capability of Fe3O4@Ru@SiO2 NPs. Simultaneously, the material surface's electronegativity decreases, weakening the electrostatic repulsion with the negatively charged electrode, resulting in the system detecting stronger ECL signals. This sensor enables homogeneous ECL detection while mitigating solution background interference through magnetic separation. Within a range of 100 fM to 10 nM, the sensor exhibits a linear relationship between ECL intensity and target concentration, with a 26.91 fM detection limit. It demonstrates high accuracy in clinical sample detection, holding significant potential for clinical diagnostics. Future integration with innovative detection strategies may further enhance sensitivity and specificity in biosensing applications.
A Ratiometric Electrochemiluminescent/Electrochemical Strategy for Sensitive Detection of MicroRNA Based on Duplex-Specific Nuclease and Multilayer Circuit of Catalytic Hairpin Assembly.

Abstract:In this study, we proposed a ratiometric electrochemiluminescent (ECL) /electrochemical (EC) biosensor based on duplex-specific nuclease (DSN)-assisted target recycling and multilayer catalytic hairpin assembly (CHA) amplification cascades for the detection of microRNA (miRNA). The DSN-assisted target recycling transformed miRNAs into a large number of ssDNA, which then catalyzed multilayer CHA amplification cascade to produce numerous long dsDNA duplexes Hn/Hn+1 (n=2, 4, 6…). Then the Hn/Hn+1 displaced the ferrocene (Fc)-labeled ssDNA (Sx+1, x=1, 3, 5…) to hybridize with the Sx sequence on the gold electrode surface. Consequently, a great number of long Sx/Hn/Hn+1 duplexes were immobilized for binding Ru(phen)32+ to obtain an amplified ECL signal. Meanwhile, the EC signal of Fc reduced, and the quenching effect of Fc to ECL signal also decreased. By measuring the ratio of ECL signal of Ru(phen)32+ to EC signal of Fc, quantitative analysis of miRNA-499 with high accuracy and reproducibility was obtained. The ratiometric biosensor shows high sensitivity and a wide linear range of six orders of magnitude. With the help of DSN-assisted target recycling, this strategy can be easily extended to detect other miRNAs without redesigning the CHA cascade system. The proposed "hybrid" ratiometric ECL/EC strategy enriches the ratiometric sensors, and can find extensive applications in bioanalysis, especially for multiplex detection.

Abstract:This paper describes a bidirectional electrochemiluminescence (ECL) biosensor for the detection of microRNA-141 (miRNA-141) with aM level detection limit and two-segment 8 orders of magnitude linear range. Hemin/G-quadruplex DNAzyme was assembled on carbon nitride nanosheets and Au nanoparticles modified electrode. The ECL of carbon nitride nanosheets could be enhanced by the right amount of H2O2 (no more than 10 mM) and then inhibited by excessive H2O2 when 0.1 M K2S2O8 acted as coreactant. In the presence of excessive H2O2 (20 mM), a recovery of ECL intensity was obtained due to the catalytic reduction of H2O2 caused by hemin/G-quadruplex DNAzyme at lower target concentrations, and then an ECL decrease occurred mainly by biocatalytic precipitation (BCP)-induced charge transfer resistance on the electrode surface at higher target concentrations. Therefore, based on the change of the ECL intensity caused by the catalytic reduction of H2O2 and BCP, a highly sensitive bidirectional miRNA sensor with ultralow detection limit of 7.9 aM and wide linear range from 10-17 to 10-9 M was obtained. This work could attract more attention on the study of multiple mechanisms and also provides a more sensitive and precise method for the analysis of nucleic acids.

Abstract:Since microRNAs (miRNAs) are predictors of tumorigenesis, accurate identification and quantification of miRNAs with highly similar sequences are expected to reflect tumor diagnosis and treatment. In this study, a highly selective and sensitive electrochemiluminescence (ECL) biosensor was constructed for miRNAs determination based on Y-shaped junction structure equipped with locked nucleic acids (LNA), graphene oxide-based nanocomposite to enrich luminophores, and conductive matrix. Specifically, two LNA-modified probes were designed for specific miRNA recognition, that is, a dual-amine functionalized hairpin capture probe and a signal probe. A Y-shaped DNA junction structure was generated on the electrode surface upon miRNA hybridizing across the two branches, so as to enhance the selectivity. Carbon quantum dots-polyethylene imine-graphene oxide (CQDs-PEI-GO) nanocomposites were developed to enrich luminophores CQDs, and thus enhancing the ECL intensity. For indirect signal amplification, an electrochemically activated poly(2-aminoterephthalic acid) (ATA) film decorated with gold nanoparticles was prepared on electrode as an effective matrix to accelerate the electron transfer. The fabricated ECL biosensor achieved sensitive determination of miRNA-222 with a limit-of-detection (LOD) as low as 1.95 fM (S/N = 3). Notably, Y-shaped junction structures equipped with LNA probes endowed ECL biosensor with salient single-base discrimination ability and anti-interference capacity. Overall, the proposed Y-shaped ECL biosensor has considerable promise for clinical biomarker determination.

High Fidelity Detection of miRNAs from Complex Physiological Samples Through Electrochemical Nanosensors Empowered by Proximity Catalysis and Magnetic Separation

Abstract:Electrochemical detection of miRNA biomarkers in complex physiological samples holds great promise for accurate evaluation of tumor burden in the perioperative period, yet limited by reproducibility and bias issues. Here, nanosensors installed with hybrid probes that responsively release catalytic DNAzymes (G-quadruplexes/hemin) were developed to solve the fidelity challenge in an immobilization-free detection. miRNA targets triggered toehold-mediated strand displacement reactions on the sensor surface and resulted in amplified shedding of DNAzymes. Subsequently, the interference background was removed by Fe 3 O 4 core-facilitated magnetic separation. Binding aptamers of the electrochemical reporter (dopamine) were tethered closely to the catalytic units for boosting H 2 O 2 -mediated oxidation through proximity catalysis. The one-to-many conversion by dual amplification from biological-chemical catalysis facilitated sufficient homogeneous sensing signals on electrodes. Thereby, the nanosensor exhibited a low detection limit (2.08 fM), and high reproducibility (relative standard deviation of 1.99%). Most importantly, smaller variations (RSD of 0.51-1.04%) of quantified miRNAs were observed for detection from cell lysates, multiplexed detection from unprocessed serum, and successful discrimination of small upregulations in lysates of tumor tissue samples. The nanosensor showed superior diagnostic performance with an area under curve (AUC) of 0.97 and 94% accuracy in classifying breast cancer patients and healthy donors. These findings demonstrated the synergy of signal amplification and interference removal in achieving high-fidelity miRNA detection for practical clinical applications.

Abstract:Specific and sensitive microRNAs (miRNAs) detection is essential to early cancer diagnosis. The development of these technologies including functional nuclease-mediated target amplification and DNA nanotechnology possesses tremendous potential for the high-performance detection of miRNAs in the accurate diagnosis of disease. In this study, we have established an ultrasensitive electrochemical biosensor by combining nicking endonuclease-assisted primer exchange reaction (PER) cascade amplification with a DNA nanosphere (DNS)-mediated electrochemical signal-enhanced system for the detection of miRNA-21 (miR-21). The cascade amplification is initiated by a nicking endonuclease that can cleave specific DNA substrates and highly amplify translation of the target to single-stranded DNA fragments (sDNA). Then, the PER cascade is powered by strand-displacing polymerase and generates a large amount of nascent single stranded connector DNA (cDNA) via sDNA triggering of the dumbbell probe (DP), thus achieving the cascade amplification of the target. Finally, the DNS loaded with plenty of electroactive substances can be captured on the electrode via cDNA for further enhancing the electrochemical signal and highly sensitive detection of miR-21. The proposed electrochemical biosensor exhibits a wide detection range of 1 aM to 0.1 nM and a low detection limit of 0.58 aM. The excellent selectivity allows the biosensor to discriminate miR-21 from other miRNAs, even the one base-mismatched sequence. Moreover, the practicability of the biosensor is investigated by analyzing miR-21 in human serum and cancer cell lysate. Therefore, our proposed nicking endonuclease-assisted PER cascade amplification strategy provides a powerful platform for the early detection of miRNA-related disease and molecular diagnosis.
Nano-matrixes Propped Self-Enhanced Electrochemiluminescence Biosensor for Microrna Detection.

Abstract:MicroRNAs (miRNA) are the potential biomarker for breast cancer, a biosensor for detecting miRNA-21 was successfully prepared by covalently linking carbohydrazide (CON4H6) and tris (4,4 '- dicarboxylic acid-2,2' -bipyridyl) ruthenium dichloride (Ru (dcbpy)32+) as a self-enhanced emitter (Ru-CON4H6). The biosensor was prepared by coating the electrode with mesoporous silica encapsulated Ru-CON4H6 as luminophores (RMSNs) to covalently link a couple of DNA strands (Q1-H2). The RMSNs coated electrode exhibited strong ECL emission due to the intramolecular electron transfer between the electrochemically oxidized Ru (dcbpy)32+ and co-reactant CON4H6. In the presence of target miRNA-21 and an assistant hairpin H1, H2 could be released from the sur-face through a strand displacement reaction (SDR), and the reserved Q1 could form G-quadruplex upon the addition of K+. The formed G-quadruplex then interacted with Q2-Fc in the presence of Mg2+ to form a DNA complex on the biosensor surface, which quenched the nano-matrixes propped self-enhanced ECL emission through the electron exchange between Fc and electrode or oxidized ECL intermediates. Under optimal conditions, the ECL decrease showed a correlation with target concentration, leading to a biosensing method for sensitive detection of miRNA-21. The proposed ECL method demonstrated a detectable concentration range from 0.1 fM to 1 nM along with a detection limit of 0.03 fM, good accuracy, and acceptable reproducibility, showing that the self-enhanced ECL biosensing strategy supported by nano-matrix provided a new way for the ultra -sensitive detection of miRNA, and promoted the development of breast cancer diagnosis.

Abstract:The development of a convenient and efficient assay using miRNA-21 as a lung cancer marker is of great importance for the early prevention of cancer. Herein, an electrochemical biosensor for the detection of miRNA-21 was successfully fabricated under blue light excitation using click chemistry and photocatalytic atom transfer radical polymerization (photo-ATRP). By using hairpin DNA as a recognition probe, the electrochemical sensor deposits numerous electroactive monomers (ferrocenylmethyl methacrylate) on the electrode surface under the reaction of photocatalyst (fluorescein) and pentamethyldiethylenetriamine, thereby achieving signal amplification. This biosensor is sensitive, precise and selective for miRNA-21, and is highly specific for RNAs with different base mismatches. Under optimal conditions, the biosensor showed a linear relationship in the range of 10 fM ∼1 nM (R 2 = 0.995), with a detection limit of 1.35 fM. Furthermore, the biosensor exhibits anti-interference performance when analyzing RNAs in serum samples. The biosensor is based on green chemistry and has the advantages of low cost, specificity and anti-interference ability, providing economic benefits while achieving detection objectives, which makes it highly promising for the analysis of complex samples.

Abstract:AIM to explore Hong Kong Chinese women's experiences and perceptions of elective caesarean section performed under regional anaesthesia. DESIGN an exploratory study using a qualitative approach. Data were collected by tape-recorded in-depth interviews two to five days after caesarean section. PARTICIPANTS a purposive sample of 18 Hong Kong Chinese women having an uncomplicated, elective caesarean section under successful regional anaesthesia. SETTING a post-caesarean ward of a University affiliated District General Hospital in Hong Kong. KEY FINDINGS the three categories relating to the overall theme of Consciousness during Surgery were: Interacting with others, Experience of birthing, and Awareness of the environment. IMPLICATIONS FOR PRACTICE the experiences of women, related to the general theme and categories cited above, affected their feelings of security and fulfillment. On the whole, women were satisfied with the regional anaesthesia, preferring to remain conscious throughout the surgery. They appreciated being able to interact with others and listening to music, but needed information regarding what was happening. Many were distressed by the sight and noise of surgical instruments, the narrowness of the operating table, the operating theatre lamps and the coldness of the theatre. Recommendations for practice are made relating to these issues.
An “on-off” signal-switchable electrochemiluminescence biosensor for ultrasensitive detection of dual microRNAs based on DNAzyme-powered DNA walker

Abstract:Herein, an "on-off" signal-switchable electrochemiluminescence (ECL) biosensor for dual miRNAs (miRNA-21 and miRNA-155) detection was constructed based on carbon dots (CDs)-labeled DNA walker, which can effectively avoid problems of the signal overlap or limited available labels that general exist in multiplex assays. In the presence of miRNA-21, the CDs-DNA walker were captured on the electrode via nucleic acid hybridization, producing a strong ECL signal corresponding to miRNA-21 (signal on). For miRNA-155, the biosensor was pre-incubated with CDs-DNA walker and assistant DNA (DNA-21). Target miRNA-155 could trigger metal ion-dependent DNAzyme cleavage. With the assistance of Mn2+, the substrate strands were cleaved and CDs detached from the electrode, resulting in a reduced ECL signal (signal off). The interference of miRNA-21 was eliminated because no more CDs-DNA walker was added. As a result, benefiting from the signal amplification offered by the CDs-DNA walker and the inherent properties of ECL technique, this biosensor achieved low detection limits of 33 fM for miRNA-21 and 33 aM for miRNA-155. This paper also demonstrates the biosensor's bioanalytical application for multiplex detection of miRNA-21/155 in human serum samples and the total RNA extraction solution from the human cancer cell lines.

Abstract:Abstract Aberrant expression of microRNAs (miRNAs) has been shown to be linked to several crucial biological processes, including as carcinogenesis, metastasis, and progression. The advancement of innovative miRNA detection technologies can enhance the early detection of malignancies by merging with conventional diagnostic methods, such as ultrasound technology. Herein, we reported a simple, sensitive, and label-free miRNA detection method by integrating the proximity-catalytic hairpin assembly (proximity-CHA) and DNAzyme-assisted signal amplification. Compared with traditional CHA, in which the signal amplification efficiency is greatly limited by the concentration of hairpin probes, the proposed method possesses a greatly improved signal amplification efficiency. The target facilitated the non-enzymatic CHA-driven sequential formation of DNAzyme nanostructures, resulting in the effective DNAzyme-facilitated cleavage of a substrate modified with a fluorophore and quencher, leading to the production of an intensified fluorescence signal. The proximity-CHA-DNAzyme system possesses appealing analytical characteristics, making it highly promising for the analysis of many analytes in clinical research domains.

Abstract:MicroRNAs (miRNAs), useful biomarkers for cancer diagnosis, play an important role in tumorigenesis and progression, but many of the current analysis methods can suffer from excessive protease dependence, being time-consuming and unsatisfactory performance. Therefore, a reliable sensing strategy for the protein-free, ultrasensitive analysis of tumor-associated miRNAs is desired. The proposed dual-walker biosensing strategy based on an entropy-driven catalytic (EDC) walker coupled with a smart-responsive DNAzyme walker was demonstrated for the dual-amplification detection of miRNA-21. Namely, the target miRNA-21 initiates the three-stranded substrate complex of the traditional EDC circuit to release the input trigger of the Dz walker, which recognizes the circular binding domain to restore the cleavage activity of the DzS-AuNP walker. The fluorescence signal continuously released from the AuNPs was recorded by a fluorescence reader for miRNA-21 sensing. The optimized dual-walker exhibited appreciable sensitivity with a detection limit of 70 fM, satisfactory flexibility, fine specificity and ideal stability for clinical serum sample assays. The proposed strategy may open a new avenue for the development of powerful DNA molecular tools for cancer diagnosis.

Abstract:In this work, an ultrasensitive electrochemiluminescence (ECL) biosensor was constructed based on DNA-stabilized Au Ag nanoclusters (DNA-Au Ag NCs) as the efficient luminophore and Au NPs@Ti3C2 as a new coreaction accelerator for determining microRNA-221 (miRNA-221) related to liver cancer. Impressively, DNA-Au Ag NCs were stabilized by the high affinity of the periodic 3C sequence, exhibiting an excellent ECL efficiency of 27% compared with classical BSA-Au Ag NCs (16%). Moreover, the Au NPs@Ti3C2 nanocomposites, as a new coreaction accelerator, were first introduced to accelerate the production of abundant sulfate free radicals (SO4•-) for promoting the ECL efficiency of DNA-Au Ag NCs in the DNA-Au Ag NCs/Au NPs@Ti3C2/S2O82- ternary system due to the energy band of Au NPs@Ti3C2 being well-matched with the frontier orbital of S2O82-. Furthermore, the trace target (miRNA-221) could drive the rolling circle amplification to generate an amount of output DNA with periodic 3C and 10A sequences. Through covalent bonds on the surface of poly A and Au NPs, the distance between the luminophor and the coreaction accelerator could be narrowed to further enhance the detection sensitivity. As a result, the constructed sensor has been applied for the ultrasensitive detection of miRNA-221 with a low detection limit of 50 aM and successfully monitored miRNA-221 in MHCC-97L and HeLa cell lysates. This strategy could be utilized for guiding the synthesis of light-emitting DNA-metal NCs, which has great potential in the construction of ultrasensitive biosensors for the early diagnosis of diseases.

Abstract:MicroRNAs (miRNAs) play vital regulatory roles in cancer development and a variety of diseases, which make them become promising biomarkers. Here, a simple electrochemical biosensor was developed for highly sensitive and specific detection of target miRNA using mismatched catalytic hairpin assembly (CHA). The target miRNA triggered the toehold strand displacement assembly of two hairpin substrates, which led to the cyclic reuse of the target miRNA and the CHA products. Compared with the traditional CHA, mismatched CHA could decrease the nonspecific CHA products, which reduced the background signal significantly. Under the optimal experimental conditions and using differential pulse voltammetry, the established biosensor could detect target miRNA down to 0.6 pM (S/N=3) with a linear range from 1 pM to 25 nM, and discriminate target miRNA from mismatched miRNA with a high selectivity. It was also applied to the determination of miRNA spiked into human total RNA samples. Thus, this biosensing strategy might become a potential alternative tool for detection of miRNA in biomedical research and early clinical diagnosis.
