Abstract:The present study aimed to understand the molecular mechanism underlying the light-induced depression of photosynthetic capacity in a typically shade-tolerant species Panax notoginseng grown under different light intensities. Photosynthetic performance was declined in the low-light- and high-light-grown plants. The physiological plasticity was low, while the plasticity of morpho-anatomy traits, biomass allocation and biochemical traits was great in response to the light environment. High light enhanced the expression of genes related to reactive oxygen scavenging, pentose phosphate pathway and glycolysis. The expression of genes involved in the light reaction was suppressed in low light, along with a reduction in Rubisco activity. The expression of genes encoding Rubisco was repressed in high light. Photosynthetic capacity in low light might be inhibited by the deceleration on light reaction and by the increase of inactivated Rubisco, high-light-induced decrease of photosynthetic performance might result from the deceleration on Calvin cycle. Photoprotection Contributes to Freezing Tolerance As Revealed by RNA-seq Profiling Ofrhododendronleaves During Cold Acclimation and Deacclimation over Time.

Abstract:Cold acclimation (CA) and deacclimation (DA), which are often accompanied by changes in freezing tolerance (FT), carbohydrates and hormones, are crucial for winter survival, especially under global warming. Plants with weak CA and premature DA caused by warm winters and/or unseasonal warm spells can be easily injured by adverse reactions to cold. Thus, understanding the molecular mechanisms of FT is imperative. In this study, we used high-throughput RNA-seq to profile the CA and DA of leaves of overwintering Rhododendron "Miyo-no-Sakae" over time; these leaves do not undergo dormancy but do undergo photoprotection during CA, and they do not grow during DA. Using Mfuzz and weighted gene coexpression network analysis, we identified specific transcriptional characteristics in each phase of CA and DA and proposed networks involving coexpressed genes and physiological traits. In particular, we discovered that the circadian rhythm is critical for obtaining the strongest FT, and high expression of circadian rhythm-related genes might be linked to sugar accumulation during winter. Furthermore, evergreen leaves exhibited robust photoprotection during winter, as revealed by high values of nonphotochemical quenching, high expression of transcripts annotated as "early light-induced proteins", loss of granum stacks and destacking of thylakoids, all of which were alleviated during DA. The strong requirement of photoprotection could be the reason for decreased abscisic acid (ABA) and jasmonic acid (JA) contents during CA, and decreases in ABA and JA contents may contribute to decreases in lignin content. Our data suggest that the molecular mechanisms of FT in overwintering leaves are unique, which may be due to the high requirements for photoprotection during winter.
Effects of irradiance on photosynthetic characteristics and growth of Mosla chinensis and M. scabra

Abstract:Photosynthetic and growth characteristics of Mosla chinensis and M. scabra were compared at three irradiances similar to shaded forest understory, forest edge, and open land. At 25 % full ambient irradiance, M. chinensis and M. scabra had similar photosynthetic characteristics, but saturation irradiance, compensation irradiance, and apparent quantum yield of M. chinensis were higher than those of M. scabra at full ambient irradiance and 70 % full ambient irradiance. At the same irradiance treatment, specific leaf area and leaf area ratio of M. chinensis were lower than those of M. scabra . Photon-saturated photosynthetic rate and water use efficiency of M. chinensis , however, were not significantly higher than those of M. scabra , and the leaf area and total biomass were lower than those of M. scabra . As a sun-acclimated plant, the not enough high photosynthetic capacity and lower biomass accumulation may cause that M. chinensis has weak capability to extend its population and hence be concomitant in the community.

Abstract:Light is the driving force of plant growth, providing the energy required for photosynthesis. However, photosynthesis is also vulnerable to light-induced damage caused by the production of reactive oxygen species (ROS). Plants have therefore evolved various protective mechanisms such as non-photochemical quenching (NPQ) to dissipate excessively absorbed solar energy as heat; however, photoinhibition and NPQ represent a significant loss in solar energy and photosynthetic efficiency, which lowers the yield potential in crops. To estimate light capture and light energy conversion in rice, a genotype with pale green leaves (pgl) and a normally pigmented control (Z802) were subjected to high (HL) and low light (LL). Chlorophyll content, light absorption, chloroplast micrographs, abundance of light-harvesting complex (LHC) binding proteins, electron transport rates (ETR), photochemical and non-photochemical quenching, and generation of ROS were subsequently examined. Pgl had a smaller size of light-harvesting chlorophyll antenna and absorbed less photons than Z802. NPQ and the generation of ROS were also low, while photosystem II efficiency and ETR were high, resulting in improved photosynthesis and less photoinhibition in pgl than Z802. Chlorophyll synthesis and solar conversion efficiency were higher in pgl under HL compared to LL treatment, while Z802 showed an opposite trend due to the high level of photoinhibition under HL. In Z802, excessive absorption of solar energy not only increased the generation of ROS and NPQ, but also exacerbated the effects of increases in temperature, causing midday depression in photosynthesis. These results suggest that photosynthesis and yield potential in rice could be enhanced by truncated light-harvesting chlorophyll antenna size.

Abstract:Rhizosphere respiration (Rroot) and rhizosphere priming effect (RPE) are crucial for regulating soil carbon dynamics. However, their responses to light intensity are not fully resolved. We investigated Rroot and RPE of soybean and sunflower using a continuous isotope-labeling technique. The two species were subjected to five levels of light intensity in a growth chamber. Plants were maintained at full light from seeding for 45 or 59 days, and switched to shading treatments for four days. Soil respiration was partitioned into root- and soil-derived CO2 during the last five days (day0, full light; day1–4, shading). Both soybean and sunflower Rroot showed significant positive relationships with light intensity, particularly after one day of shading. Moreover, both plants showed positive RPE (44–98%) at full light, but with increasing duration and intensity of shading, the RPE declined and even became negative (up to –20%). Indeed, a positive relationship between light intensity and RPE was observed in both species. Overall, our results showed that shading, by influencing light intensity and presumably photosynthesis rate and belowground carbon allocation, exerts a significant control of rhizosphere respiration rate and priming effect of the two species.
Light Energy Partitioning and Photoprotection from Excess Light Energy in Shade-Tolerant Plant Amorphophallus Xiei under Steady-State and Fluctuating High Light

Abstract:Light is the most essential environmental factor that plants require for the growth and development. However, light is highly heterogeneous in natural condition, and plants have to evolve a series of strategies to acclimate the dynamic light since it is sessile. The photosynthetic performance and regulatory mechanisms involved in acclimation were elucidated in shade-tolerant plant Amorphophallus xiei under steady state and fluctuating high light. The dissipation and allocation of light energy, photosynthetic capacity, and antioxidant defense were examined in A. xiei cultivated under 4%, 17%, and 100% of full sunlight. High-light-grown plants exhibited reduced photosynthesis and the slowest response to simulated sunflecks than the other two treatments. Maximum and actual efficiency of PSII photochemistry (Fv′/Fm′ and ΦPSII) and electron transport rate (ETR) were lowest, and non-photochemical quenching (NPQ) were highest in high-light-grown plants than in low- and intermediate-light-grown plants subjected to different photon flux densities (PFD) and intercellular CO2 concentrations (Ci). Fv′/Fm′, ΦPSII, ETR and NPQ were lowest in high-light-grown A. xiei than others, when plants were exposed to simulated sunflecks. In fully light-induced leaves, high-light-grown plants showed a maximum value in quantum efficiency of light-dependent thermal dissipation (ΦNPQ), and a minimum value in ΦPSII was recorded. Low-light-grown plants subjected to simulated sunflecks, a maximum value in ΦNPQ and ΦPSII were observed. Mass-based nitrogen content (Narea), specific leaf area (SLA), superoxide dismutase (SOD) and catalase (CAT) activities significantly declined with the increase of growth irradiance. The high-light-grown plants showed higher xanthophyll de-epoxidation and ascorbate–glutathione cycle activity, while low-light-grown plants showed higher neoxanthin and β-carotene contents. In general, high-light-grown A. xiei generally show a depressed photosynthetic capacity, and are efficient in dissipating excess light energy by NPQ under a steady-state light but not under a highly fluctuating light. Low-light-grown plants show a rapid photosynthetic assimilation response to sunflecks and then a rapid activation of the energy-dependent quenching (qE) component of NPQ under the sunflecks. In addition, high-light-grown plants, although using various strategies to reduce light absorption and to scavenge reactive oxygen species, have less efficient protection against photodamage.

Abstract:Photosynthetic adaptive strategies vary with the growth irradiance. The potential photosynthetic adaptive strategies of shade-tolerant species Panax notoginseng (Burkill) F. H. Chen to long-term high light and low light remains unclear. Photosynthetic performance, photosynthesis-related pigments, leaves anatomical characteristics and antioxidant enzyme activities were comparatively determined in P. notoginseng grown under different light regimes. The thickness of the upper epidermis, palisade tissue, and lower epidermis were declined with increasing growth irradiance. Low-light-grown leaves were declined in transpiration rate (Tr) and stomatal conductance (Cond), but intercellular CO 2 concentration ( C i ) and net photosynthesis rate ( P n ) had opposite trends. The maximum photo-oxidation P 700 + ( P m ) was greatly reduced in 29.8% full sunlight (FL) plants; The maximum quantum yield of photosystem II ( F v / F m ) in 0.2% FL plants was significantly lowest. Electron transport, thermal dissipation, and the effective quantum yield of PSI [Y(I)] and PSII [Y(II)] were declined in low-light-grown plants compared with high-light-grown P. notoginseng . The minimum value of non-regulated energy dissipation of PSII [Y(NO)] was recorded in 0.2% FL P. notoginseng . OJIP kinetic curve showed that relative variable fluorescence at J-phase ( V J ) and the ratio of variable fluorescent F K occupying the F J - F O amplitude ( W k ) were significantly increased in 0.2% FL plants. However, the increase in W k was lower than the increase in V J . In conclusion, PSI photoinhibition is the underlying sensitivity of the typically shade-tolerant species P. notoginseng to high light, and the photodamage to PSII acceptor side might cause the typically shade-tolerant plants to be unsuitable for long-term low light stress.

Abstract:The allocation of recently assimilated carbon (C) by plants depends on developmental stage and on environmental factors, but the underlying mechanisms are still a matter of debate. In the present study, we investigated the regulation of C uptake and allocation and their adjustments during plant growth. We induced different allocation strategies in the Mediterranean shrub Halimium halimifolium L. by a reduction of light (Low L treatment) and nutrient availability (Low N treatment) and analyzed allocation parameters as well as morphological and physiological traits for 15 months. Further, we conducted a (13)CO2 pulse-labeling and followed the way of recently assimilated carbon to eight different tissue classes and respiration for 13 days. The plant responses were remarkably distinct in our study, with mainly morphological/physiological adaptions in case of light reduction and adjustment of C allocation in case of nutrient reduction. The transport of recently assimilated C to the root system was enhanced in amount (c. 200%) and velocity under nutrient limited conditions compared to control plants. Despite the 57% light reduction the total biomass production was not affected in the Low L treatment. The plants probably compensated light reduction by an improvement of their ability to fix C. Thus, our results support the concept that photosynthesis is, at least in a medium term perspective, influenced by the C demand of the plant and not exclusively by environmental factors. Finally, our results indicate that growing heterotrophic tissues strongly reduce the C reflux from storage and structural C pools and therefore enhance the fraction of recent assimilates allocated to respiration. We propose that this interruption of the C reflux from storage and structural C pools could be a regulation mechanism for C translocation in plants.

Abstract:Photoprotection mechanisms of rice plants were studied when its seedlings were subjected to the combined stress of water and high light. The imposition of water stress, induced by PEG 6000 which was applied to roots, resulted in substantial inhibition of stomatal conductance and net photosynthesis under all irradiance treatments. Under high light stress, the rapid decline of photosynthesis with the development of water stress was accompanied by decreases in the maximum velocity of RuBP carboxylation by Rubisco (V(cmax)), the capacity for ribulose-1,5-bisphosphate regeneration (J(max)), Rubisco and stromal FBPase activities, and the quantum efficiency of photosystem II, in the absence of any stomatal limitation of CO(2) supply. Water stress significantly reduced the energy flux via linear electron transport (J(PSII)), but increased light-dependent and DeltapH- and xanthophyll-mediated thermal dissipation (J(NPQ)). It is concluded that the drought-induced inhibition of photosynthesis under different irradiances in the rice was due to both diffusive and metabolic limitations. Metabolic limitation of photosynthesis may be related to the adverse effects of some metabolic processes and the oxidative damage to the chloroplast. Meanwhile, an enhanced thermal dissipation is an important process to minimize the adverse effects of drought and high irradiance when CO(2) assimilation is suppressed.
High-resolution Imaging of Rhizosphere Oxygen (O2) Dynamics in Potamogeton Crispus: Effects of Light, Temperature and O2 Content in Overlying Water

Abstract:Aims Radial oxygen loss (ROL) for macrophytes is intimately involved in their survival and growth, thus detailed characterizations of ROL and its implication for geochemical processes are of particular interest. We experimentally investigated ROL patterns from the submerged macrophyte species Potamogeton crispus and determined how light, temperature and O 2 content in overlying water regulate O 2 micro-distributions in the rhizosphere. Method Planar optodes were firstly used for non-destructive imaging of the O 2 micro-distributions around single roots of P. crispus planted in rhizotrons containing sediment. The dynamic changes in below-ground O 2 concentrations and oxygenation expansion at different light intensities (0–216 μmol photons m −2 ), temperatures (14 and 25 °C) and O 2 content in overlying waters (0–256 μmoL −1 ) were quantified. Results P. crispus -mediated ROL is predominantly localized to the root apices with an average rate of 168.1 ± 21.4 nmol m −2 s −1 under nearly natural conditions (light, saturated overlying water at 14 °C), maintaining a visible oxygenated rhizosphere zone with a radius of 1.33 ± 0.21 mm. ROL is closely correlated with light intensity, suggesting photosynthetic O 2 evolution. In darkness, the rhizosphere O 2 availability is significantly reduced and strongly affected by the O 2 content in overlying water, which passively diffuses into the plant leaves. Elevated temperatures lead to diminished ROL as a result of increased O 2 demand of the surrounding sediment. Conclusion A high O 2 microheterogeneity around P. crispus root apices is firstly demonstrated, and we provide direct empirical evidences that ROL are regulated dynamically by the shifting light, temperature and O 2 content in overlying water.
High nitrogen-driven photosynthesis limitation in shade-demanding species mediated by stomatal conductance through plasma membrane intrinsic proteins

Abstract:Photosynthesis is susceptible to nitrogen (N) surplus. However, the mechanism of reduced photosynthesis by N surplus need to be further verified in the shade-demanding and high-N-sensitive species. Photosynthetic capacity, N allocation, stomatal parameters, the correlation between PIP (encoding plasma membrane intrinsic proteins) expression and total stomatal conductance (g t ) were analyzed in Panax notoginseng grown under the levels of low N (LN), moderate N (MN) and high N (HN). N content per unit leaf area (N area ) was increased with increasing N application. HN resulted in an increase in N allocation to the light capture system ( P L ) and a decrease in N allocation to the carboxylation system ( P C ). Net photosynthetic rate ( P n ), stomatal conductance (g s ), the maximum electron transport rate ( J max ) and the maximum carboxylation rate ( V cmax ) were lower in the LN and HN plants. Under HN condition, the limitation to stomatal conductance (S L ) and limitation to biochemical factors (B L ) were the main factors for reducing photosynthetic efficiency. The stomatal area index (SAI) was more closely correlated with stomatal density (SD) than with stomatal length (SS). g s was partially correlated with SS or SD. The correlation analysis showed that the relative expression of PIP1.3 , PIP1.4 and PIP2.7 is positively correlated with g t /N area . The results obtained suggest that high-N-driven limitation to photosynthesis in the shade-demanding and HN-sensitive medicinal crop, such as P. notoginseng , is mainly derived from the decrease in g s , and partly from the decrease in the carboxylation capacity of RuBP.

Source-to-sink relationship between green leaves and green pseudobulbs of C 3 orchid in regulation of photosynthesis

Abstract:In this paper, photosynthetic characteristics of green leaves (GL) and green pseudobulbs (GPSB) of C 3 orchid Oncidium Golden Wish were first studied. Light saturation for photosynthesis and maximum photosynthetic rates ( P max ) were significantly higher in GL than in GPSB. The results of the optimal PSII quantum yield (F v /F m ratio), electron transport rate (ETR), the effective photochemical quantum yield (ΔF/F m ′) and nonphotochemical quenching (NPQ) of Chl fluorescence revealed that GPSB had lower light utilization than that of GL. Significantly higher photosynthetic pigments were found in GL than in GPSB. Alteration of source/sink ratio had no impact on all photosynthetic parameters for both GL and GPSB after a short term of 3 days or even a long term of 2 weeks of treatments although there were significant decreases in GL carbohydrate concentration of GL-darkened plants by the end of the day. However, decreases of all photosynthetic parameters of GL were observed in GL-darkened plants after 4 weeks of treatment compared to those of fully illuminated (FI) and GPSB-darkened plants. These results indicate that the level of carbohydrates in GL plays an important role in regulating their photosynthesis. Due to their lower photosynthetic capacities, GPSB function mainly as sinks. Darkening GPSB up to 2 weeks did not affect their own P max and the P max of GL and thus, did not result in significant decreases of total carbohydrate concentration of GPSB. As GPSB store a large amount of carbohydrates, it could also act as a source when the level of carbohydrates decreased. Thus, GL could depend on GPSB carbohydrates to regulate their photosynthesis when their source capacity was removed. However, 4 weeks after treatments, photosynthetic capacities of GL were significantly lower in GL- and GPSB-darkened plants than in FI plants, which could be due to the lower total soluble and insoluble sugar concentrations of both GL and GPSB in these plants.
Light Signaling-dependent Regulation of Photoinhibition and Photoprotection in Tomato.

Abstract:Photoreceptor-mediated light signaling plays a critical role in plant growth, development, and stress responses but its contribution to the spatial regulation of photoinhibition and photoprotection within the canopy remains unclear. Here, we show that low-red/far-red (L-R/FR) ratio light conditions significantly alleviate PSII and PSI photoinhibition in the shade leaves of tomato (Solanum lycopersicum) plants. This protection is accompanied by a phytochrome A-dependent induction of LONG HYPOCOTYL5 (HY5). HY5 binds to the promoter of ABA INSENSITIVE5 (ABI5), triggering RESPIRATORY BURST OXIDASE HOMOLOG1 (RBOH1)-dependent H2O2 production in the apoplast. Decreased levels of HY5, ABI5, and RBOH1 transcripts increased cold-induced photoinhibition and abolished L-R/FR-induced alleviation of photoinhibition. L-R/FR illumination induced nonphotochemical quenching (NPQ) of chlorophyll a fluorescence and increased the activities of Foyer-Halliwell-Asada cycle enzymes and cyclic electron flux (CEF) around PSI. In contrast, decreased HY5, ABI5, and RBOH1 transcript levels abolished the positive effect of L-R/FR on photoprotection. Loss of PROTON GRADIENT REGULATION5-dependent CEF led to increased photoinhibition and attenuated L-R/FR-dependent NPQ. These data demonstrate that HY5 is an important hub in the cross talk between light and cold response pathways, integrating ABA and reactive oxygen species signaling, leading to the attenuation of photoinhibition by enhanced induction of photoprotection in shade leaves.

Effects of Light and Soil Water Availability on Leaf Photosynthesis and Growth of Arisaema Heterophyllum , a Riparian Forest Understorey Plant

Abstract:The effects of soil-water availability on leaf light acclimation and whole-plant carbon gain were examined in Arisaema heterophyllum Blume, a riparian deciduous forest understorey plant. Photosynthesis, above-ground morphology and ramet biomass accumulation (relative growth rate: RGR of a corm for a full leaf life-span) were measured on plants raised under three light treatments combined with two soil water conditions. The two higher light treatments during growth (high: max. 550 µmol photons m–2 s–1; medium: 150 µmol photons m–2 s–1) resulted in a twofold increase in RGRs, 30% higher photosynthetic capacities and 20% less photosynthetic low-light use efficiency than those under a low light condition (50 µmol photons m–2 s–1). Leaf area was the smallest and leaf mass area ratio was the largest under the high light treatment. Water stress decreased both photosynthetic rate and leaf area and, hence, RGR in all the light regimes. However, water stress did not alter the general patterns of physiological and morphological responses to different light regimes. We estimated that higher photosynthetic low-light use efficiency and larger leaf area in the low light leaf would lead to a threefold carbon gain as compared with the high light leaf under simulated low light conditions. Both experimental and simulation results suggest that the physiological and morphological acclimations tend to be beneficial to carbon gain when light availability is low, whereas they favor increased water use efficiency when light availability is sufficiently high.
Effects of Nitrogen Deficiency on Gas Exchange, Chlorophyll Fluorescence, and Antioxidant Enzymes in Leaves of Rice Plants

Abstract:Gas exchange, chlorophyll (Chl) fluorescence, and contents of photosynthetic pigments, soluble proteins (ribulose-1,5-bisphosphate carboxylase/oxygenase, RuBPCO), and antioxidant enzymes were characterized in the fully expanded 6 th leaves in rice seedlings grown on either complete (CK) or on nitrogen-deficient nutrient (N-deficiency) solutions during a 20-chase period. Compared with the control plants, the lower photosynthetic capacity at saturation irradiance ( P max ) was accompanied by an increase in intercellular CO 2 concentration (C i ), indicating that in N-deficient plants the decline in P max was not due to stomatal limitation but due to the reduced carboxylation efficiency. The fluorescence parameters Φ PS2 , F v ′/F m ′, electron transport rate (ETR), and q P showed the same tendency as P max in N-deficient plants. Correspondingly, a higher q N paralleled the rise of the ratio of carotenoid (Car) to Chl contents. However, F v /F m was still diminished, suggesting that photoinhibition did occur in the photosystem 2 (PS2) reaction centres. In addition, the activities of antioxidant enzymes on a fresh mass basis were gradually lowered, leading to the aggravation of membrane lipid peroxidation with the proceeding N-deficiency. The accumulation of malonyldialdehyde resulted in the lessening of Chl and soluble protein content. Analyses of regression showed PS2 excitation pressure (1 - q P ) was linearly correlated with the content of Chl and inversely with soluble protein (particularly RuBPCO) content. There was a lag phase in the increase of PS2 excitation pressure compared to the decrease of RuBPCO content. Therefore, the increased excitation pressure under N-deficiency is probably the result of saturation of the electron transport chain due to the limitation of the use of reductants by the Calvin cycle. Rice plants responded to N-deficiency and high irradiance by decreasing light-harvesting capacity and by increasing thermal dissipation of absorbed energy.
Variable Light Condition Improves Root Distribution Shallowness and P Uptake of Soybean in Maize/Soybean Relay Strip Intercropping System

Abstract:In this study, soybean root distribution in an inter-cropping system was influenced by various environmental and biotic cues. However, it is still unknown how root development and distribution in inter-cropping responds to aboveground light conditions. Herein, soybeans were inter- and monocropped with P (phosphorus) treatments of 0 and 20 kg P ha yr−1 (P0 and P20, respectively) in field experiment over 4 years. In 2019, a pot experiment was conducted as the supplement to the field experiment. Shade from sowing to V5 (Five trifoliolates unroll) and light (SL) was used to imitate the light condition of soybeans in a relay trip inter-cropping system, while light then shade from V5 to maturity (LS) was used to imitate the light condition of soybeans when monocropped. Compared to monocropping, P uptake and root distribution in the upper 0–15 cm soil layer increased when inter-cropped. Inter-cropped soybeans suffered serious shade by maize during a common-growth period, which resulted in the inhibition of primary root growth and a modified auxin synthesis center and response. During the solo-existing period, plant photosynthetic capacity and sucrose accumulation increased under ameliorated light in SL (shade-light). Increased light during the reproductive stage significantly decreased leaf P concentration in SL under both P-sufficient and P-deficient conditions. Transcripts of a P starvation response gene (GmPHR25) in leaves and genes (GmEXPB2) involved in root growth were upregulated by ameliorated light during the reproductive stage. Furthermore, during the reproductive stage, more light interception increased the auxin concentration and expression of GmYUCCA14 (encoding the auxin synthesis) and GmTIR1C (auxin receptor) in roots. Across the field and pot experiments, increased lateral root growth and shallower root distribution were associated with inhibited primary root growth during the seedling stage and ameliorated light conditions in the reproductive stage. Consequently, this improved topsoil foraging and P uptake of inter-cropped soybeans. It is suggested that the various light conditions (shade-light) mediating leaf P status and sucrose transport can regulate auxin synthesis and respond to root formation and distribution.
Photoprotective energy dissipation is greater in the lower, not the upper, regions of a rice canopy: a 3D analysis

Abstract:High light intensities raise photosynthetic and plant growth rates but can cause damage to the photosynthetic machinery. The likelihood and severity of deleterious effects are minimised by a set of photoprotective mechanisms, one key process being the controlled dissipation of energy from chlorophyll within PSII known as non-photochemical quenching (NPQ). Although ubiquitous, the role of NPQ in plant productivity is important because it momentarily reduces the quantum efficiency of photosynthesis. Rice plants overexpressing and deficient in the gene encoding a central regulator of NPQ, the protein PsbS, were used to assess the effect of protective effectiveness of NPQ (pNPQ) at the canopy scale. Using a combination of three-dimensional reconstruction, modelling, chlorophyll fluorescence, and gas exchange, the influence of altered NPQ capacity on the distribution of pNPQ was explored. A higher phototolerance in the lower layers of a canopy was found, regardless of genotype, suggesting a mechanism for increased protection for leaves that experience relatively low light intensities interspersed with brief periods of high light. Relative to wild-type plants, psbS overexpressors have a reduced risk of photoinactivation and early growth advantage, demonstrating that manipulating photoprotective mechanisms can impact both subcellular mechanisms and whole-canopy function.
Nitric Oxide Induced by Ammonium/Nitrate Ratio Ameliorates Low-Light Stress in Brassica pekinesis: Regulation of Photosynthesis and Root Architecture

Abstract:Low-light intensity affects plant growth and development and, finally, causes a decrease in yield and quality. There is a need for improved cropping strategies to solve the problem. We previously demonstrated that moderate ammonium:nitrate ratio (NH4+:NO3−) mitigated the adverse effect caused by low-light stress, although the mechanism behind this alleviation is unclear. The hypothesis that the synthesis of nitric oxide (NO) induced by moderate NH4+:NO3− (10:90) involved in regulating photosynthesis and root architecture of Brassica pekinesis subjected to low-light intensity was proposed. To prove the hypothesis, a number of hydroponic experiments were conducted. The results showed that in plants exposed to low-light intensity, the exogenous donors NO (SNP) and NH4+:NO3− (N, 10:90) treatments significantly increased leaf area, growth range, and root fresh weight compared with nitrate treatment. However, the application of hemoglobin (Hb, NO scavenger), N-nitro-l-arginine methyl ester (L-NAME, NOS inhibitor), and sodium azide (NaN3, NR inhibitor) in N solution remarkably decreased the leaf area, canopy spread, the biomass of shoot and root, the surface area, and volume and tips of the root. The application of N solution and exogenous SNP significantly enhanced Pn (Net photosynthetic rate) and rETR (relative electron transport rates) compared with solo nitrate. While all these effects of N and SNP on photosynthesis, such as Pn, Fv/Fm (maximum quantum yield of PSII), Y(II) (actual photosynthetic efficiency), qP (photochemical quenching), and rETR were reversed when the application of Hb, L-NAME, and NaN3 in N solution. The results also showed that the N and SNP treatments were more conducive to maintaining cell morphology, chloroplast structure, and a higher degree of grana stacking of low-light treated plants. Moreover, the application of N significantly increased the NOS and NR activities, and the NO levels in the leaves and roots of mini Chinese cabbage seedlings treated with N were significantly higher than those in nitrate-treated plants. In conclusion, the results of this study showed that NO synthesis induced by the appropriate ammonia–nitrate ratio (NH4+: NO3− = 10:90) was involved in the regulation of photosynthesis and root structure of Brassica pekinesis under low-light stress, effectively alleviating low-light stress and contributing to the growth of mini Chinese cabbage under low-light stress.

Abstract:Cuttings of the southeast Asian tropical rain forest tree species, Pongamia pinnata (L.) Pierre were raised in growth chambers providing a photosynthetic photon flux density (PPFD) of 60 micromol m(-2) s(-1) with either a low or a high red:far-red light ratio (LR and HR, repectively). The chambers were supplied with air containing CO(2) at a concentration of either 400 (LR4 and HR4, respectively) or 800 micromol mol(-1) (HR8 and LR8, respectively). After 4 months, leaf morphology and photosynthetic characteristics were determined. Relative to HR4, the LR4 treatment increased leaf area and total chlorophyll concentration (Chl) by 24 and 25%, respectively, but reduced leaf mass per unit area (LMA) by 19%. Elevated [CO(2)] significantly increased leaf area and LMA but did not affect Chl of LR or HR plants. Leaf nitrogen concentration was unaffected by the red:far-red light ratio but decreased significantly in seedlings in the elevated [CO(2)] treatment. Photosynthesis measured in situ under the growth conditions of ambient light and [CO(2)] (A(amb)) was 30% lower on an area basis and 14% lower on a mass basis in LR4 plants than in HR4 plants. Elevated [CO(2)] reduced the activity of ribulose-1,5-bisphosphate carboxylase/oxygenase and thus decreased light-saturated photosynthetic rate in both HR and LR plants. Elevated [CO(2)] increased mean leaf area and decreased respiration rates in both LR and HR plants. The LR8 plants had significantly higher A(amb) than LR4 plants, but similar A(amb) to HR8 plants.
