Impact of nutrient restriction at dry-off on performance and metabolism

Abstract:Thanks to improvements in genetics, nutrition, and management, modern dairy cows can still produce large amounts of milk at the end of lactation, with possible negative effects on health and welfare, particularly when milking is stopped abruptly. To limit yield at dry-off, strategies involving different types of dietary restriction have been used worldwide. Thus, we aimed to investigate the effects of a reduced nutrient density at dry-off on milk production, metabolism, the pattern of rumen fermentation, and milk fatty acid profile around dry-off and in the ensuing periparturient period. During the last week before dry-off, 26 Holstein cows were enrolled in pairs according to the expected calving date and either fed ad libitum ryegrass hay (nutrient restricted, NR; 13 cows) or continued to receive lactation diet (control group, CTR; 13 cows). After dry-off, both groups received only grass hay for 7 d, and free access to water was always provided. Blood, milk, and rumen fluid samples were collected from 7 d before dry-off to 28 DIM. Milk production, DMI (during the periparturient period), and rumination times were recorded daily. At dry-off, NR cows had decreased milk yield (-62%) and milk lactose compared with CTR but had higher fat and protein contents. In the subsequent lactation, no significant differences were observed in milk yield and composition. The BCS did not differ between groups during the transition period, but it decreased in NR after dry-off. Before dry-off, NR had decreased glucose, urea, and insulin, but higher creatinine, BHB, and nonesterified fatty acids (NEFA). The day after dry-off, NEFA were lower in NR, but they were higher 7 d after calving. At dry-off, NR had higher rumen pH, lower lactate, urea, and total volatile fatty acid concentrations. Considering volatile fatty acid molar proportions, NR had increased acetate but decreased propionate and butyrate at dry-off. Rumination time dropped 6 d before dry-off in NR and after dry-off in CTR, but no differences were observed in the periparturient period. Milk fatty acid profile revealed a remarkably lower proportion of short-chain fatty acids in NR at dry-off and a higher proportion of medium- and long-chain ones. These results confirmed that decreasing nutrient density reduce milk yield before dry-off. However, metabolism around dry-off was significantly affected, as suggested by plasma, rumen fluid, and milk analyses. Further research is required to investigate the impact of the metabolic effects on the inflammatory response, liver function, and immune system, particularly concerning the mammary gland.
Effect of group size and health status on behavior and feed intake of multiparous dairy cows in early lactation

Abstract:Dairy cows in early lactation are often housed in a large group, where they may have to compete for access to feed and space. However, a cow's ability to compete may be impaired due to production disease, and housing in a small group with minimal competition may be beneficial for cow welfare. The aim of this study was to investigate the effect of group size and health on social and feeding behavior of cows during the first 3 d after introduction to a new group. Data included 54 multiparous Holstein-Friesian cows that were moved from an individual maternity pen and individually joined an existing group pen for 6 (N6) or 24 cows (N24) on d 4 after calving. Cows were considered sick if they were diagnosed with and treated for milk fever, mastitis, or retained placenta, diagnosed with subclinical ketosis or metritis within 3 d of calving, or were diagnosed and treated for any other infection (n = 22; balanced across treatments). Stocking density of both pens was 100% at the feeding and lying areas. Behavioral data were collected from video recordings during the 1 d after introduction to the group pen, as well as via electronic feed bins and leg-attached accelerometers during the first 3 d after introduction to the group pen. No interactions between health status and group size were discovered. During the 1 d after introduction, N6 cows displaced other cows from feed less frequently than N24 cows (1.22 vs. 5.76 times/24 h), were less likely to access feed after a displacement (replacement; 0.29 vs. 1.67 times/24 h), and were less frequently being butted by another cow (0.42 vs. 1.69 times/24 h). Second-parity cows received more head butting than later-parity cows. Data obtained from feed bins showed that the number of replacements peaked on d 2 after introduction to the group pen. During the first 3 d we observed no effect of group size on DMI, but sick cows ate less than cows that were not sick (15.2 vs. 16.6 kg of DM/d). However, cows in N6 visited the feeder less often (42.4 vs. 55.6 times/d). Over the 3 d after introduction DMI and feeding time increased, whereas feeding rate decreased. Lying time and the number of lying bouts increased from d 1 to 2. The number of steps decreased over days, but the number of steps was higher among N24 than N6 cows on d 1 and d 2. Results suggest that cows experience less competition when moved to a smaller group after calving regardless of health status. Thus, minimizing competition by housing dairy cows in a small group for the first days after calving may improve cow welfare under commercial conditions.
Behavioral and physiological effects of a short-term feed restriction in lactating dairy cattle with different body condition scores at calving

Abstract:Body condition score (BCS) around calving, and the typical BCS loss for up to 100 d after parturition, is associated with both production and reproductive performance of dairy cattle. In addition, there is public concern that thin cows may have impaired welfare, particularly in early lactation where feed demand exceeds pasture growth, and a lag exists between peak milk energy requirements and intake. The aim of this experiment was to determine how BCS at calving influences behavioral and physiological responses to a short-term feed restriction at 47 DIM. Body condition score (on a 10-point scale) at calving was manipulated by modifying the diets in the previous lactation of healthy dairy cattle to generate 3 treatment groups: low BCS (3.4; n=17), medium BCS (4.6; n=18), or high BCS (5.4; n=20). Cows were tested in 4 groups for 8 consecutive days; testing consisted of different levels of feed allocation (d 1 and 2: 100%; d 3 and 4: 75%; d 5: 50%; d 6 to 8: 125%), where 100% was 15kg of DM/cow per day. All BCS groups had similar and marked behavioral and physiological responses to feed restriction. For example, they increased vocalization, time spent eating silage and grazing, aggressive behavior, and fat metabolism (as measured by concentrations of β-hydroxybutyrate and nonesterified fatty acids), and reduced milk production. Body condition affected some of these responses. Fewer cows with low BCS engaged in aggressive interactions in a feed competition test (trough filled with silage that could be consumed in 15 min) on the first day of feed restriction (low: 32%; medium: 74%; high: 64%; standard error of difference=15.4%). High BCS cows had greater concentrations of β-hydroxybutyrate and nonesterified fatty acids throughout the experimental period, which suggests more fat mobilization; however, plasma leptin and fecal glucocorticosteroid metabolite concentrations were unaffected by BCS. Whereas cows demonstrated marked responses to feed restriction, the results suggest that a BCS of 3.4, 4.6, or 5.4 in healthy cows at calving does not overwhelmingly influence this response at 47 DIM.

Abstract:In the commercial dairy industry worldwide, it is common practice to periodically regroup cows as part of their management strategy within housed systems. While this animal husbandry practice is intended to improve management efficiency, cows may experience social stress as a result of the social environment changes, which may have an impact on their behavioural patterns, performance, and welfare. We investigated whether regrouping altered dairy cows' behaviour and impacted their cortisol concentration (a physiological marker of stress), oxytocin, milk yield, and quality in a robotic milking system. Fifty-two lactating cows (17 primiparous; 35 multiparous) were moved in groups of 3-5 individuals into established pens of approximately 100 cows. Behaviour of the regrouped cows was directly observed continuously for 4 h/day across 4-time blocks (day-prior (d-1), day-of regrouping (d0), day-after (d + 1), and 6-days after (d + 6) regrouping). Cows were categorised as being with others, alone, or feeding every 2.5 min prior to the assessment of behavioural dynamics. Milk yield (MY) and composition, total daily activity, and rumination time (RUM) data were extracted from the Lely T4C management program (Lely Industries, Maassluis, the Netherlands), and milk samples were collected for cortisol and oxytocin concentration analyses; data were analysed using linear mixed-effect modelling. Primiparous cows were less likely to be interacting with others on d + 1 than d-1 compared with multiparous. However, average bout duration (minutes) between being alone and feeding activity states were similar on d-1, d + 1, and d + 6, for both primiparous and multiparous cows. A reduction in the average alone and feeding bout duration was observed on d0. Multiparous cows spent significantly more total time being alone on d0 compared to d-1. Neither regrouping nor parity statistically influenced milk DM content, energy, or cortisol concentration. Primiparous cows produced 3.80 ± 2.42 kg (12.2%) less MY on d + 1 compared to their d-1, whereas multiparous cows did not change MY. A significant decrease of 0.2% fat was found in both parity groups following regrouping and remained low up to d + 6. Daily activity in both parity groups increased significantly and RUM reduced after regrouping. A significant decrease in oxytocin concentration was observed in all cows on d + 1. The results, specifically for primiparous cows, indicated a negative impact of regrouping on social interactions, due to changes in the social environment which may lead to short-term social instability. Multiparous cows may benefit from previous regrouping experiences.

Abstract:Isolated chloroplasts which have lost their envelopes and, in consequence, the soluble components which constitute the stroma, will nevertheless evolve O(2) when supplied with an artificial oxidant (the Hill reaction). They will also evolve O(2) with NADP as the Hill oxidant if supplemented with ferredoxin. With catalytic NADP, continuing O(2) evolution can be maintained by the inclusion of a suitable reaction or reaction sequence which reoxidizes NADPH.In the Benson-Calvin cycle the terminal oxidant is glycerate 1,3-bisphosphate which is generated by phosphorylation of 3-phosphoglycerate, its immediate precursor. Experiments with a reconstituted chloroplast system are described in which this reaction sequence is catalyzed by stromal protein and supported by photophosphorylation of catalytic ADP. In the presence of CO(2), 3-phosphoglycerate can be progressively replaced by ribulose 1,5-bisphosphate, ribose 5-phosphate, fructose 6-phosphate, fructose 1,6-bisphosphate, and finally by glyceraldehyde 3-phosphate. In this last instance the natural oxidant is regenerated from its own reduction product (via the carboxylation step) and the reaction sequence therefore involves the entire photosynthetic carbon cycle.
Dietary fiber source and direct-fed microbial supplementation effects on lactation performance and feeding behavior of high-producing dairy cows

Abstract:The objective of this study was to evaluate the effects of dietary fiber source and direct-fed microbial supplementation on lactation performance and feeding behavior of high-producing dairy cows. Sixty-four multiparous Holstein cows (3.5 ± 1.6 [SD] lactations; 76 ± 22 DIM and 735 ± 67 kg of BW at covariate period initiation) and 32 gate feeders were enrolled in a study with a completely randomized design and a 2 × 2 factorial arrangement. Cows and gate feeders were randomly assigned to treatments (16 cows and 8 gate feeds per treatment). Cows were allowed 1 wk to acclimate to feeding gates followed by a 2-wk covariate period. During the acclimation and covariate periods, all cows were fed the same diet to meet or exceed the nutrient requirements. Following the covariate period, cows were enrolled in a 8-wk treatment period during which cows were randomly assigned to 1 of 4 treatments consisting of forage inclusion in the diet, either 45.8% (LF) or 56.7% (HF) of DM, and the supplementation of 75 mL/head per day of a direct-fed microbial (DFM) containing herbal extracts (mallow, mint, and sage), Lactiplantibacillus plantarum, Lentilactobacillus buchneri, Saccharomyces cerevisiae, and sugar cane molasses (Valibiom Mix, Valibiotics AG, Emmen, Switzerland) or without supplementation (CON). The average covariate period value of each variable was used as a covariate. Three-way interactions were observed for DMI and feed efficiency. Dry matter intake was 2 to 3 kg greater for LF-DFM than HF-CON and HF-DFM during wk 2, 3, 5, and 8 of the treatment period. Milk production was 2.1 kg/d greater for LF than HF diets. Both milk fat and MUN concentrations were greater for HF than LF diets. Conversely, milk protein concentration was lower for HF than LF diets. The respiration rate measured in the morning was lower with DFM supplementation than CON. Rectal temperature measured in the morning and averaged for the day were greater for LF than HF diets. Under the conditions of the present study, feeding high-forage diets may be an alternative for producers to reduce feeding costs depending on the price of purchased feeds. However, nonforage fiber sources (i.e., soy hulls) must be considered when producers are challenged by either forage shortages or forage with a lower nutritive value. Additionally, DFM supplementation reduced respiration rate in the morning and affected meal behavior of lactating cows.

Abstract:This study aimed to use a range of non-invasive monitoring technologies to investigate the behavioural and physiological responses of individually housed dairy calves to age at change in milk replacer (MR) feeding frequency. Forty-eight Holstein Friesian calves were individually penned and fed MR (625 g/d) as solids in one of three feeding regimes: (i) once-a-day feeding commencing at age 14 d (OAD14), (ii) once-a-day feeding commencing at age 28 d (OAD28) and (iii) twice-a-day feeding (TAD). Several behavioural (automatic activity sensors), physiological (infrared [IR] thermography and heart rate variability [HRV]) and haematological indicators were used to examine calf responses. Reduction in milk feeding frequency at 14 or 28 d of age increased daily concentrate intakes and drinking water consumption throughout the pre-wean period. Calf lying behaviour was unaffected by reduction in milk feeding frequency; however, TAD calves recorded a significant decrease in total daily lying time during the post-wean period compared with OAD28s. There was no effect of treatment on IR eye or rectal temperature throughout the experiment; however, there was an effect of age, with IR temperature decreasing as calf age increased. OAD14 calves tended to have decreased HRV at days 14 and 16, which is suggestive of an increased stress load. The findings suggest that under high levels of animal husbandry and whilst maintaining the same amount of milk powder/d (625 g/d), reduction in milk feeding frequency from twice to once daily at 28 d can occur without significant impact to behavioural, performance and physiological parameters assessed here.

Abstract:Objective. To evaluate accuracy of transvaginal sonography (TVS)and hysteroscopy in detection of intrauterine pathology in infertilewomen. Subjects and methods. This retrospective study was conductedin 56 infertile women with abnormal transvaginal ultrasoundfindings of the uterine cavity which was performed during the midfollicularphase as a part of routine infertility workup. Hysteroscopy wasperformed between 6th and 10th day of cycle. Results. The mean age ofthe subjects was 31.9±4.0. The most frequent ultrasound finding wasendometrial polyp in 34 (60.7%) patients, septate uterus in 8 (14.3%)patients, submucosal myoma in 7 (12.5%) patients, endometrial hyperplasiain 5 (8.9%) patients and Syndroma Ascherman in 2 (3.6%)patients. Hysteroscopy confirmed 20 (35.7%) polyps, the same numberof myomas, septate uterus and Syndroma Ascherman as detectedby ultrasound, (7 (12.5%), 8 (14.3%) and 2 (3.6%), respectively) and19 (33.9%) endometrial hyperplasia. In 46 women with histologicalexcamination, the sensitivity of TVS and hysteroscopy in the diagnosisof endometrial polyps were identical - 100%, while the specificity washigher in hysteroscopy than in TVS (92.3% versus 56.4%, p<0.001).The sensitivity of TVS in the diagnosis of endometrial hyperplasia washigher than that of hysteroscopy (86.4% versus 22.7%, p<0.001), whilespecificity was identical, of 100%. Accordance between hysteroscopyand histology was good (k=0.79), between ultrasound and hystologywas moderete (k=0.59). Conclusion. Hysteroscopy appeared tobe more reliable in diagnosis than TVS. The use of a high frequencyultrasound probe leads us to a lack of diagnostic clarity between endometrialpolyps and hyperplasia.
Prepartum anti-inflammatory therapies in Holstein dairy cows blocked by parity and body condition score group: Effects on metabolic stress, systemic inflammation, performance, and health

Abstract:The objective of this study was to assess the effects of prepartum administration of anti-inflammatory therapies on body condition score (BCS), β-hydroxybutyrate (BHB) concentration, haptoglobin (HP) concentration, milk yield, milk components, rumination time, clinical health events and reproductive performance in Holstein dairy cows. At 14 d before the expected calving date, cows (PAR; n = 170) and heifers (nulliparous [NUL]; n = 63) were blocked by BCS group (optimal = 3-3.5 [OPT]; over-conditioned cows [OVERC; BCS ≥ 3.75 pts.]) and parity (NUL; PAR) and randomly allocated to one of 3 treatment groups: 1) ASA (n = 78): receive one oral administration of acetylsalicylic acid (4 boluses; 480 grain/bolus); 2) MEL (n = 76): receive one oral administration with meloxicam (1mg/kg of BW), or 3) PLC (n = 77): receive one oral treatment with gelatin capsules filled with water. Body condition score was assessed, and blood samples were collected, weekly starting one week before treatment until 3 weeks after calving. Daily milk yields and daily rumination times were collected from on-farm computer records. Dairy Herd Improvement Association (DHIA) monthly test data were collected to assess milk yield, somatic cell counts, and milk components. Furthermore, health events, culling rate, and reproductive performance data were collected from on-farm computer records. The data were analyzed using MIXED, GLIMMIX, and LIFETEST procedures of SAS as a randomized complete block design. On average, MEL-NUL cows produced 4.77 ± 0.93 kg/d and 4.81 ± 0.92 kg/d more milk from wk 6 to wk 21 of lactation compared with ASA-NUL and PLC-NUL cows, respectively. Similarly, there was a week by treatment by body condition group interaction (P = 0.01), where OVERC cows treated with MEL produced more milk from wk 10 to wk 15 of lactation compared with ASA- OVERC and PLC-OVERC cows. Parous cows treated with ASA had lower BCS compared with PAR cows treated with MEL or PLC. A lower percentage of OVERC cows treated with ASA became sick in the first 60 DIM compared with MEL- OVERC and PLC- OVERC cows (ASA = 23.88 ± 7.26%, MEL = 46.36 ± 8.57%; PLC = 46.74 ± 8.53%; P = 0.04). Parous cows treated with ASA had (P = 0.03) a higher hazard ratio to become pregnant by 300 DIM compared with PAR MEL cows. Although the study was not sized for finding treatment differences in blocking criteria groups, these results suggest that treatment with prepartum anti-inflammatory therapies may have positive effects on milk yield and postpartum health in specific groups of cows, such as NUL and OVERC cows, while it may not be recommended for other animal categories, such as parous cows and cows with optimal BCS. Larger studies are needed to strengthen the associations observed in this study.

Abstract:The Stroop Test is a quick and frequently used measure in screening for brain damage, dysfunction of selective attention, and cognitive flexibility. The purpose of the present study is to provide normative data for Trenerry's Stroop Neuropsychological Screening Test (SNST) in a sample of 605 healthy Greek participants (age range: 18-84 years, education range: 6-18 years). Results revealed that age and education significantly contributed to SNST scores, accounting for a significant proportion of variance in time needed to complete the color task and in the interference Color-Word score. Performance on most of the measures decreases with increasing age and lower levels of education. Normative data stratified by age and education for the Greek adult population are provided as a useful set of norms for clinical practice.
Influence of two different feeding strategies in the dry period on dry matter intake and plasma protein peroxidative and antioxidative profile during dry period and early lactation

Abstract:Abstract Background Dairy cows undergo dramatic changes in endocrine and metabolic status around parturition and in early lactation. Meeting the nutritional requirements of transition dairy cows is important for animal health, production and animal wellbeing. Dry cow feeding and managing play an essential role in this. The changes in metabolism of periparturient cows also lead to a rise in the production of oxidising agents, leading to oxidative stress. The relationship between dry cow diet composition and oxidative stress has received little research attention so far. In the present study, the influence of two different dry cow feedings (single diet with medium energy content over the whole dry period versus traditional two-phase diet with a low-energy “far-off” ration and a high energy “close-up” ration) on dry matter intake, energy intake and plasma protein peroxidative and antioxidative profile was investigated. Results The examined parameters revealed a dynamic profile within the experimental period. Dry matter intake (DMI) did not differ between groups. However, there was a time and a group x time interaction effect: Group 1 (“one-phase”) had a very constant DMI with a slow and even decrease until calving. In Group 2 (“two-phase”), an initial increase in DMI two weeks antepartum (a.p.) was followed by a sharp drop at week 1 a.p.. The highest total antioxidant capacity and sulfhydryl residue concentration was noted at partus. In contrast, concentration of formylokinurenine and bityrosine bridges as representatives of protein peroxidation were lowest at parturition. The time course of formylokinurenine and bityrosine bridges showed parallels to the DMI. The contents of sulfhydryl groups, formylokinurenine and total antixoxidant capacity did not differ between groups. In contrast, concentration of bityrosine bridges was always higher in Group 2 compared with Group 1 and these differences were statistically significant at week 3 a.p., week 2 a.p., week 1 a.p. and at parturition. Conclusion The results of our study suggest time-related changes of pro- and antioxidative plasma parameters. Different dry cow feeding affected antepartal DMI. Furthermore, DMI and diet compositions seemed to have an influence on plasma protein peroxidative profile and activity of antioxidative defence.

Abstract:The aim of the present study was to investigate the incidence and causes of ovarian inactvity and evaluate the efficacy of single administration of GnRH for treatment of inactive ovaries in dairy cows. Two experiments were carried out to fulfill the aims of the study. In experiment 1, 122 cows which had not shown estrous signs ³ 45 after calving were examined twice by transrectal ultrasonography at a 7-10 day interval to diagnose the causes of anestrous and assess the incidence of ovarian inactivity. In Experiment 2, blood samples were collected from 30 cows with ovarian inactivity and 13 cyclic cows to compare the blood concentration of insulin, glucose, calcium and phosphorus between the two types of cows. Cows (n=30) suffering from ovarian inactivity were treated with either 500 mg gonadorelin acetate intramuscular (G1; n=20) or saline intramuscular (G2; n=10). The ovaries of cows in G1 and G2 were examined by transrectal palpation twice before and twice after the treatment at a week interval. After transrectal palpation, blood samples were collected from cows for assessment of progesterone, insulin and glucose. Also the cows were observed for estrous signs and those cows showed estrus were inseminated. In Experiment 1, silent or unobserved estrus (47.5%), inactive ovaries (41%), cystic ovaries (9%) and pyometra (2.5%) were the reasons of anestrous in the studied dairy cows. In Experiment 2, there were significant differences in the blood concentrations of insulin (P< 0.0005) and glucose (P< 0.01) between cyclic and non-cyclic cows. On the other hand, there were non-significant differences in the blood concentrations of calcium and phosphorus between the cyclic and non-cyclic cows. Estrus induction rates within 14 days after treatment were significantly (P< 0.05) higher in G1 (60%) than in G2 (10%). Seven of 12 cows (58.3%) responded to administration of GnRH in G1 and the responded cow (1/1) in G2 were pregnant at Day 60 after AI. In conclusion, ovarian inactivity is the main reason of postpartum anestrous in dairy cows. Lower level of insulin and glucose might be among the reasons of ovarian inactivity. Single administration of GnRH is reasonably effective for treatment of ovarian inactivity in dairy cows.
Exploring vaginal discharge scoring to assess clinical metritis severity: Comparison between intrauterine dextrose and systemic antibiotics treatments

Abstract:The objectives of this study were to assess: 1) differences in the metabolic status, systemic inflammation, daily milk yield, and daily rumination time between Holstein dairy cows with different vaginal discharge scores (VDS) in the first 7±3 DIM, and 2) effects of intrauterine dextrose infusion on metabolic status, systemic inflammation, daily milk yield and daily rumination time in dairy cows with VDS4 and VDS5. Cows (n=641) from a farm located in central Pennsylvania were screened at 7±3 DIM (study d 0) to assess vaginal discharge scores. Vaginal discharge was scored using a five-point scale (i.e., 1- clear fluid, 2- <50% white purulent fluid, 3- >50% white purulent fluid, 4- red-brownish fluid without fetid smell, and 5- fetid red-brownish watery fluid). Cows with VDS4 and VDS5 were blocked by parity and randomly assigned to one of two treatment groups: 1) CONV (VDS4 n=15; VDS5 n= 23): two injections of ceftiofur (per label; 6.6 mg/Kg) 72 h apart; and 2) DEX (VDS4 n=15; VDS5 n=22): three intrauterine infusions of a 50% dextrose solution (1 L/cow) every 24 h. Cows that presented a VDS 1, 2, and 3 were categorized as normal vaginal discharge animals (NOMVDS; n=35) and were randomly selected and matched by parity to CONV and DEX cows. Daily milk yield and rumination time for the first 150 DIM were collected from on-farm computer records. Blood samples were collected to assess haptoglobin (HP) and β-hydroxybutyrate (BHB) concentrations at study d 0, d 7, and d 14 relative to enrollment. Subclinical ketosis was defined as having a BHB concentration >1.2 mmol/dL at any of the sampling points. The data were analyzed using the MIXED and GLIMMIX procedures of SAS as a randomized complete block design. When comparing cows with different VDS (i.e., NOMVDS, VDS4, VDS5) separately, cows with VDS5 had the highest concentration of HP at enrollment compared to cows with VDS4 and NOMVDS; however, cows with VDS4 had higher concentrations of HP compared to cows with NOMVDS. Cows with VDS4 or VDS5 had a higher incidence of subclinical ketosis compared to cows with NOMVDS (p=0.005; VDS4= 62.08±9.16%; VDS5=74.44±6.74%; NOMVDS=34.36±8.53%). Similarly, daily milk yield (p<.0001; VDS4=30.17±1.32 kg/d; VDS5=27.40±1.27 kg/d; NOMVDS=35.14±1.35 kg/d) and daily rumination time (p=0.001; VDS4=490.77±19.44 min; VDS5=465±16.67 min; NOMVDS=558.29±18.80 min) was lower for cows with VDS4 and VDS5 compared to cows with NOMVDS at 7±3 days in milk. When analyzing HP concentration between treatment groups in cows with VDS4 (p=0.70), VDS5 (p=0.25), or VDS4 and VDS5 combined (p=0.31), there was no difference in HP concentration by study d 14 between treatment groups. Interestingly, when only cows with VDS4 were considered for treatment, both treatments, DEX and CONV, increased the daily milk yield to the levels of NOMVDS cows by 14 days in milk. On the other hand, when only cows with VDS5 were considered for treatment, cows treated with DEX produced, on average, 4.48 kg/d less milk in the first 150 days in milk compared to cows treated with CONV or cows that had NOMVDS. Similarly, when cows with either VDS4 or VDS5 were considered for treatment, DEX treatment also impaired milk yield. These results suggest that cows with either VDS 4 or 5 have an altered inflammatory status, and decreased milk yield and rumination compared to cows with NOMVDS. Furthermore, DEX treatment may have similar effects on daily milk yield and metabolic status compared to CONV in cows with VDS4, while DEX is not recommended for cows with VDS5.
Impact of postpartum milking frequency on the immune system and the blood metabolite concentration of dairy cows

Abstract:The transition from pregnancy to lactation is marked by metabolic, hormonal, and immunological changes that have an impact on the incidence of infectious and metabolic diseases. The aim of this study was to evaluate the effect on immune function and blood metabolite concentration of limiting milk production in early lactation to reduce negative energy balance. Twenty-two multiparous Holstein cows were milked either once a day (1x) or twice a day (2x) for the first week postpartum. All cows were milked twice daily for the rest of lactation. Blood concentrations of nonesterified fatty acids (NEFA), beta-hydroxybutyric acid (BHBA), calcium, bilirubin, urea, phosphorus, glucose, leptin, stanniocalcin-1, and 17beta-estradiol were determined in samples collected from 5 wk before scheduled calving to 5 wk after calving. Polymorphonuclear leukocytes (PMNL) were isolated from blood to conduct assays for chemotaxis, phagocytosis, and respiratory burst. Peripheral blood mononuclear cells (PBMC) were isolated to evaluate lymphocyte proliferation and cytokine production (tumor necrosis factor-alpha, IL-4, and interferon-gamma). Cows milked 1x produced 31% less milk than cows milked 2x during the first week of lactation. Over the following 13 wk of lactation, the milk production of cows milked 1x during the first week was 8.1% lower than for cows milked 2x. However, because the percentages of fat and protein were greater in the milk from 1x cows, the yields of milk components and energy-corrected milk were similar. Calving induced an increase in the concentrations of NEFA, BHBA, urea, and bilirubin. The increases in levels of NEFA and BHBA were greater in cows milked 2x than in cows milked 1x. During the same period, the serum glucose concentration decreased but remained greater in cows milked 1x. Serum calcium on d 4 and serum phosphorus on d 4 and 5 were greater in cows milked 1x. The differences between the 2 groups persisted beyond treatment until postpartum d 24 for NEFA and glucose and until postpartum d 14 for BHBA. After calving, the concentrations of leptin and stanniocalcin-1 decreased. During the first week postpartum, the decrease of leptin was less marked in cows milked 1x. The immune functions of PBMC and PMNL isolated from experimental cows and incubated using a standard medium did not show clear-cut peripartum immunosuppression. These variables were not significantly affected by the treatments, with the exception of interferon-gamma secretion, which was greater on d 5 and 14 in cows milked 1x. In conclusion, limiting milk production in early lactation had positive effects on metabolite concentration, but larger studies are necessary to establish if this could reduce disease incidence.
The effect of cultural and environmental factors on potato seed tuber morphology and subsequent sprout and stem development

Abstract:Seed crops of the variety Estima were grown in each of 2 years using two planting dates, two harvest dates, two plant densities and two irrigation regimes to produce seed tubers which had experienced different cultural and environmental conditions. The effects of these treatments on tuber characteristics, sprout production and stem development in the ware crop were then determined in subsequent experiments using storage regimes of 3 and 10 °C. Time of planting the seed crop affected numbers of eyes, sprouts and above ground stems in the subsequent ware crop because environmental conditions around the time of tuber initiation appeared to alter tuber shape. Cooler, wetter conditions in the 7 days after tuber initiation were associated with tubers which were longer, heavier and had more eyes, sprouts and above ground stems. In contrast, the time of harvesting the seed crop did not affect tuber shape or numbers of above ground stems and there was no interaction with tuber size. The density of the seed crop had no effect on any character measured and irrigation well after tuber initiation did not affect tuber shape, numbers of sprouts or numbers of stems. Seed production treatments, which resulted in earlier dormancy break, were associated with tubers that produced more sprouts and above ground stems, in contrast to the conventional understanding of apical dominance. Storage at 3 °C gave fewer sprouts, a lower proportion of eyes with sprouts and fewer stems than storage at 10 °C. The major effects on stem production appear to result from environmental conditions at the time of tuber initiation of the seed crop and sprouting temperature.
Negatively controlled trial investigating the effects of dry cow therapy on clinical mastitis and culling in multiparous cows

Abstract:Blanket dry cow therapy (DCT) is a major contributor to overall antibiotic usage on dairy farms in the United States. With low prevalence of intramammary infections at dry-off in US herds today, alternative DCT approaches have been the focus of much research. We hypothesized that complete cessation of DCT [i.e., use of internal teat sealants (ITS) only at dry-off] could be a practical alternative to blanket DCT in well-managed herds. The objective of this negatively controlled clinical trial was to determine the effects of DCT on clinical mastitis (CM) and removal from the herd during the dry period and the first 200 d of the subsequent lactation in multiparous dairy cows treated with only ITS at dry-off. As a secondary objective, we conducted exploratory analysis to identify subpopulations in the herd (based on parity, previous CM history, and dry-period length) where DCT would not affect postcalving udder health, to generate hypotheses about potential alternative selective DCT programs. The study was conducted in a commercial dairy herd in South Dakota from June 2020 to January 2021. Dry-off sessions (n = 43) were scheduled such that all cows at a given session were dried off using ITS alone (ITS only, n = 20 sessions, n = 1,108 cows) or an intramammary DCT product containing 500 mg of cloxacillin (Dry-Clox, Boehringer Ingelheim) followed by ITS (ITS+ABX, n = 23 sessions, n = 1,331 cows). Culling and CM events were recorded by farm workers who were blinded to the treatment status of cows. Hazard ratios (HR) for the effects of the treatment group on CM and removal from the herd were estimated using multivariable Cox proportional hazards, adjusting for the clustered treatment allocation strategy. Risk of removal from the herd during the dry period was lower in ITS+ABX than ITS-only cows (1.1 vs. 2.7%; HR = 0.45; 95% CI: 0.25 to 0.81). Risk of removal from the herd during the first 200 d of lactation was similar in ITS+ABX and ITS-only cows (17.3 vs. 18.0%; HR = 0.98; 95% CI: 0.82 to 1.18). Risk of CM during the first 200 d of lactation was lower in ITS+ABX cows (6.9%; HR = 0.56; 95% CI: 0.41 to 0.76) compared with ITS-only cows (13.4%). The beneficial effects of DCT on CM and removal from the herd were consistently observed across strata of parity, previous CM history, and dry-period length, indicating that no subpopulations could be identified to withhold DCT. The findings from this study indicate that the omission of DCT from the dry-off procedure, when udder health is not taken into consideration, in multiparous cows can have a negative effect on cow health and welfare. Findings from previous research suggest that culture- or algorithm-guided selective dry cow therapy are likely to be safer approaches to improving antibiotic stewardship.
Reducing milking frequency from twice to once daily as an adjunct treatment for ketosis in lactating dairy cows-A randomized controlled trial

Abstract:This randomized controlled trial investigated the effects of temporarily reducing milking frequency (MF) on the resolution of ketosis and milk production in dairy cows in early lactation. To detect ketosis [blood β-hydroxybutyrate (BHB) ≥1.2 mmol/L], Holstein cows were screened daily from 3 to 16 d in milk using a cow-side meter. Cows diagnosed with ketosis (n = 104) were randomly assigned to twice-daily milking (TDM) or reduced to once-daily milking (ODM) for 2 wk, then returned to twice-daily milking. Both treatment groups received a 5-d treatment of an oral propylene glycol drench (PG; 300 g) beginning on the afternoon of the diagnosis; cows received additional 5-d PG treatments if they had a ketotic test result (blood BHB ≥1.2 mmol/L) at least 4 d after finishing the first PG treatment. Blood BHB tests were conducted for the first 3 d after ketosis diagnosis, and then once every 3 d for 21 d of trial (DOT). Milk and milk component data were collected weekly for 15 wk following trial enrollment. The ODM group showed rapidly and markedly decreased blood BHB concentrations (primiparous cows: 1 DOT, 0.92 ODM vs. 1.22 TDM, 15 DOT, 0.55 vs. 0.81 mmol/L; multiparous cows: 1 DOT, 1.01 vs. 1.40, 15 DOT, 0.78 vs. 1.65 mmol/L). In addition, a logistic regression model indicated that ODM cows were less likely to have blood BHB concentrations ≥1.2 mmol/L [primiparous cows: 3 DOT: ODM 1% (95% confidence interval: 0-10%) vs. TDM 43% (30-58%), 15 DOT ODM 0% (0-0.2%) vs. TDM 22% (13-36%); multiparous cows: 3 DOT: ODM 33% (24-44%) vs. TDM 59% (48-69%), 15 DOT ODM 20.9% (13-31%) vs. TDM 64% (53-74%)]. The proportion of ODM cows that required additional treatments of PG were substantially lower than the TDM group (ODM: 39%; TDM: 64%) than the TDM cows during the initial 21-d period. However, during the 2-wk treatment period, cows in the ODM group produced 26% less milk and 25% less energy-corrected milk than the TDM cows. During wk 3 to 15, when all cows were milked twice daily, ODM cows produced less milk (-14%) and energy-corrected milk (-12%) compared with the TDM group. Milk protein percentage was greater, and milk fat percentage and linear score tended to be greater in the ODM group over 15 wk. In conclusion, a 2-wk reduction of MF in ketotic cows from twice to once daily with treatment with PG resolved ketosis and decreased blood BHB concentrations more effectively than treating TDM cows with PG alone. However, the 2-wk MF reduction had immediate and long-term (up to 13 wk after cessation of MF reduction) negative effects on milk production.
Effects of prenatal ethanol exposure on parvalbumin-expressing GABAergic neurons in the adult rat medial septum.

Abstract:Exposure of human fetuses to ethanol often results in the fetal alcohol syndrome. Animal models of fetal alcohol syndrome have been developed and used to examine the consequences of prenatal ethanol exposure on the central nervous system. The objective of this study was to determine the long-term effects of prenatal ethanol exposure on parvalbumin-expressing (PA+) GABAergic neurons of the rat medial septum. Pregnant Long-Evans rats were maintained on 1 of 3 diets from gestational day 0 to 21: an ethanol-containing liquid diet in which ethanol accounted for 35% of the total calories, a similar diet with the isocaloric substitution of sucrose for ethanol, or a lab chow control diet. Offspring were killed on postnatal day 60, and their brains were prepared for parvalbumin immunocytochemistry. Female rats exposed to the ethanol-containing diet during gestation had 42% fewer PA+ neurons in the medial septum and reduced PA+ cell density when compared with female rats exposed to the sucrose diet. Ethanol females also had fewer PA+ neurons per unit volume than sucrose females. Male rats exposed to ethanol did not display a similar reduction in PA+ neurons or density. No effect of prenatal diet was found on the area or volume of the medial septum, nor were cell diameters affected. As such, prenatal exposure to ethanol seems to reduce permanently the number of PA+ neurons in the female rat medical septum without affecting area, volume, or neuronal size. Functional implications and possible relations to the fetal alcohol syndrome are discussed.
