Abstract:Isolated chloroplasts which have lost their envelopes and, in consequence, the soluble components which constitute the stroma, will nevertheless evolve O(2) when supplied with an artificial oxidant (the Hill reaction). They will also evolve O(2) with NADP as the Hill oxidant if supplemented with ferredoxin. With catalytic NADP, continuing O(2) evolution can be maintained by the inclusion of a suitable reaction or reaction sequence which reoxidizes NADPH.In the Benson-Calvin cycle the terminal oxidant is glycerate 1,3-bisphosphate which is generated by phosphorylation of 3-phosphoglycerate, its immediate precursor. Experiments with a reconstituted chloroplast system are described in which this reaction sequence is catalyzed by stromal protein and supported by photophosphorylation of catalytic ADP. In the presence of CO(2), 3-phosphoglycerate can be progressively replaced by ribulose 1,5-bisphosphate, ribose 5-phosphate, fructose 6-phosphate, fructose 1,6-bisphosphate, and finally by glyceraldehyde 3-phosphate. In this last instance the natural oxidant is regenerated from its own reduction product (via the carboxylation step) and the reaction sequence therefore involves the entire photosynthetic carbon cycle.
Influence of two different feeding strategies in the dry period on dry matter intake and plasma protein peroxidative and antioxidative profile during dry period and early lactation

Abstract:Abstract Background Dairy cows undergo dramatic changes in endocrine and metabolic status around parturition and in early lactation. Meeting the nutritional requirements of transition dairy cows is important for animal health, production and animal wellbeing. Dry cow feeding and managing play an essential role in this. The changes in metabolism of periparturient cows also lead to a rise in the production of oxidising agents, leading to oxidative stress. The relationship between dry cow diet composition and oxidative stress has received little research attention so far. In the present study, the influence of two different dry cow feedings (single diet with medium energy content over the whole dry period versus traditional two-phase diet with a low-energy “far-off” ration and a high energy “close-up” ration) on dry matter intake, energy intake and plasma protein peroxidative and antioxidative profile was investigated. Results The examined parameters revealed a dynamic profile within the experimental period. Dry matter intake (DMI) did not differ between groups. However, there was a time and a group x time interaction effect: Group 1 (“one-phase”) had a very constant DMI with a slow and even decrease until calving. In Group 2 (“two-phase”), an initial increase in DMI two weeks antepartum (a.p.) was followed by a sharp drop at week 1 a.p.. The highest total antioxidant capacity and sulfhydryl residue concentration was noted at partus. In contrast, concentration of formylokinurenine and bityrosine bridges as representatives of protein peroxidation were lowest at parturition. The time course of formylokinurenine and bityrosine bridges showed parallels to the DMI. The contents of sulfhydryl groups, formylokinurenine and total antixoxidant capacity did not differ between groups. In contrast, concentration of bityrosine bridges was always higher in Group 2 compared with Group 1 and these differences were statistically significant at week 3 a.p., week 2 a.p., week 1 a.p. and at parturition. Conclusion The results of our study suggest time-related changes of pro- and antioxidative plasma parameters. Different dry cow feeding affected antepartal DMI. Furthermore, DMI and diet compositions seemed to have an influence on plasma protein peroxidative profile and activity of antioxidative defence.

Association of dry matter intake, milk yield, and days to first ovulation with cytological endometritis in Holstein cows

Abstract:Dry matter intake (DMI, kg/d) is closely related to the magnitude of negative energy and protein balance during the transition period, and the metabolic adaptations to support lactation in dairy cows. Thus, DMI might affect the development of cytological endometritis in the early postpartum period. Difficulty to adapt to these metabolic changes is related to impaired immune function and increased occurrence of reproductive disorders. We aimed to examine the association of pre- and postpartum DMI, body weight (BW), body condition score, milk yield and milk composition, and days to first ovulation with cytological endometritis at 15 (CYT15) and 30 DIM (CYT30). A second objective was to understand the association of vaginal discharge with CYT15 and CYT30 and performance. We conducted a pooled statistical analysis of 5 studies, including data from 280 multiparous Holstein cows. Based on the cutoffs for the percentage of uterine polymorphonuclear cells (PMN), determined by taking the median value of the data set for 15 and 30 DIM, cows were categorized as follows: LOW15 (PMN % at 15 DIM ≤24%; n = 125), HIGH15 (PMN % at 15 DIM >24%; n = 125), LOW30 (PMN % at 30DIM ≤7%; n = 141); and HIGH30 (PMN % at 30DIM >7%; n = 139). Cows in HIGH15 consumed an average of 1.97 ± 0.5 kg/d less DM than cows in LOW15 during prepartum, and 3.01 ± 0.5 kg/d less DM during postpartum. Dry matter intake (as a percentage of BW) was higher for cows in LOW15 during pre- and postpartum than for cows in HIGH15. Moreover, cows in HIGH15 tended to have lower milk yield than cows in LOW15 from the third until the fifth week postpartum. Although DMI was not associated with CYT30, DMI (as a percentage of BW) was lower for cows in LOW30 pre- and postpartum than for cows in HIGH30. There was no association between CYT30 and milk yield. Cows in LOW15 had greater days to first ovulation than cows in HIGH15, while cows in LOW30 also had greater days to first ovulation than cows in HIGH30. Simple regression analyses demonstrated linear associations of increased DMI, particularly postpartum, with decreased uterine PMN percentage and lower vaginal discharge score. Additionally, increased units of vaginal discharge score and increased percentage units of uterine PMN were linearly associated with decreased milk yield. Corroborating with the notion of the ovarian function being associated with uterine inflammatory status, cows in HIGH15 and HIGH30 ovulated on average 3 d before than cows in LOW15 and LOW30, respectively. Cytological endometritis at 15 DIM was associated with lower DMI from 4 wk before calving until 4 wk postpartum and was associated with lower milk yield. The association of vaginal discharge with cytological endometritis was variable and dependent on the day of evaluation.
Systemic inflammation in early lactation and its relation to the cows' oxidative and metabolic status, productive and reproductive performance, and activity

Abstract:A dysregulated inflammatory response contributes to the occurrence of disorders in cows during the transition period from pregnancy to lactation. However, a detailed characterization of clinically healthy cows that exhibit an enhanced inflammatory response during this critical period remains incomplete. In this experiment, a total of 99 individual transition dairy cows and 109 observations (18 cows monitored in 2 consecutive lactations), submitted to similar transition management were involved to evaluate the relationship between elevated an inflammatory response and metabolic and oxidative status, as well as transition outcomes. Blood was taken at -7, 3, 6, 9, and 21 DIM, and concentrations of metabolic parameters (glucose, β-hydroxybutyric acid, nonesterified fatty acids [NEFA], insulin, IGF-1, and fructosamine) were analyzed. Additionally, oxidative parameters (proportion of oxidized glutathione to total glutathione in red blood cells, the activity of glutathione peroxidase [GPx] and superoxide dismutase, concentrations of malondialdehyde, and oxygen radical absorbance capacity) and acute phase proteins (APP) including haptoglobin (Hp), serum amyloid A (SAA) and albumin-to-globulin ratio (A:G) were determined in the blood at 21 DIM. The 3 APP parameters were used to group clinically healthy cows into 2 categories through k-medoids clustering (i.e., a group showing an acute phase response, APR; n = 39) and a group not showing such a response (i.e., non-APR; n = 50). Diseased cases (n = 20) were handled in a separate group. Lower SAA and Hp concentrations as well as higher A:G were observed in the non-APR group, although for Hp, differences were observed from the APR group and not from the diseased group. Only 1 of the 5 oxidative parameters differed between the groups, with the non-APR group exhibiting lower GPx activity compared with the diseased group. The non-APR group showed the highest IGF-1 levels among the 3 groups and and lower NEFA concentrations compared with the diseased groups. Cows in the diseased group also showed reduced dry matter intake and milk yield compared with clinically healthy cows, regardless of their inflammatory status. Moreover, the APR group exhibited temporarily lower activity levels compared with the non-APR group. These findings highlight that cows with a lower inflammatory status after 21 DIM exhibited better metabolic health characteristics and productive performance, as well as activity levels. Nevertheless, the detrimental effects of a higher inflammatory status in the absence of clinical symptoms are still relatively limited.

Abstract:Dried blood spot analysis is an innovative novel blood sampling technique gaining interest in drug discovery and development processes owing to its inherent advantages over the conventional whole blood, plasma or serum sample collection. The present manuscript describes the development and validation of a highly sensitive and precise method of evaluation of pharmacokinetics of (+) and (-) darunavir enantiomers on rat dried blood spots. The enantiomers on rat dried blood spots were extracted into methanol and separated by LC on a Chiralpak IA column using hexane and ethanol containing 0.1% DEA (75:25, v/v) as a mobile phase at 20°C; both the enantiomers were detected at 266 nm using a photodiode array detector. The method was validated in terms of selectivity, linearity, accuracy, precision and stability as per the US Food and Drug and Administration guidelines. The hematocrit effect on extraction recovery was evaluated and the mean recoveries of (-) and (+) enantiomers of darunavir from dried blood spots were found to be 85.76 and 88.91% respectively. The intra- and inter-day precision and accuracy were 3.1-8.4 and 0.8-4.8% respectively. The developed method was successfully applied to a pharmacokinetic study of (+) and (-) enantiomers of darunavir on rat dried blood spots.
Effect of dry period length on plasma minerals and oxidative stress around parturition and milk yield in high-producing Holstein dairy cows

Abstract:The aim of present study was to evaluate the effect of dry period length (56 vs. 28 days) on plasma minerals, albumin, total protein and globulin, oxidative status, and milk production in high-producing dairy cows (13,500 ± 1800 kg milk in previous 305-day lactation). A total of 120 multiparous Holstein cows were used in a repeated measurement mixed model (animal was considered random effect) with two experimental groups and 60 replicates each: 1-long dry period (LDP, 56-day dry period) and 2-short dry period (SDP, 28-day dry period). While LDP cows received two different rations in far-off and close-up periods (DCAD = 189 and - 130 Meq/Kg, respectively), the SDP cows received only the close-up ration (- 130 Meq/Kg) without passing the far-off stage. Blood samples for minerals, oxidative stress, and metabolites (albumin, total protein) were collected on calving day, 24 and 48 h after parturition. Milk samples of all cows in each group were collected monthly (four months from parturition to 120 DIM) and analyzed for milk fat, protein, and somatic cell count (SCC). Results showed that SDP had no effect on plasma calcium (Ca), ionized calcium (iCa), magnesium (Mg), and phosphorus (iP). Total antioxidant capacity (TAC) in the SDP cows was higher (P < 0.05) than the LDP cows. The treatment had no significant effect on plasma concentration of total protein, albumin, globulin, and superoxide dismutase (SOD) activity. A positive significant (P < 0.05) correlation was observed between plasma Mg and TAC in SDP group. Short dry period reduced (P < 0.05) milk production (48.39 vs. 51.95 kg/day; about 7%) during the first 120 days in milk. However, SDP cows had higher (P < 0.05) milk protein content compared to LDP group. The log-transformed of somatic cell count (LnSCC) was not affected by the treatment. It was concluded that a short dry period (28 days) had no negative effect on plasma Ca, iCa, Mg, and iP and even could improve TAC in high-producing Holstein cows.
Time profiles of energy balance in dairy cows in association with metabolic status, inflammatory status, and disease

Abstract:The early lactation period in dairy cows is characterized by complex interactions among energy balance (EB), disease, and alterations in metabolic and inflammatory status. The objective of this study was to cluster cows based on EB time profiles in early lactation and investigate the association between EB clusters and inflammatory status, metabolic status, oxidative stress, and disease. Holstein-Friesian dairy cows (n = 153) were selected and monitored for disease treatments during wk 1 to 6 in lactation. Weekly EB was calculated based on energy intake and energy requirements for maintenance and milk yield in wk 1 to 6 in lactation. Weekly plasma samples were analyzed for metabolic variables in wk 1 to 6, and inflammatory and oxidative stress variables in wk 1, 2, and 4 in lactation. Liver activity index (LAI) was computed from plasma albumin, cholesterol, and retino-binding protein concentration. First, cows were clustered based on time profiles of EB, resulting in 4 clusters (SP: stable positive; MN: mild negative; IN: intermediate negative; SN: severe negative). Cows in the SN cluster had higher plasma nonesterified fatty acids and β-hydroxybutyrate concentrations, compared with cows in the SP cluster, with the MN and IN cluster being intermediate. Cows in the SN cluster had a higher milk yield, lower dry matter intake in wk 1, lower insulin concentration compared with cows in the SP cluster, and lower glucose and IGF-1 concentration compared with cows in the SP and MN clusters. Energy balance clusters were not related with plasma haptoglobin, cholesterol, albumin, paraoxonase, and liver activity index (LAI). Second, cows were grouped based on health status [IHP: cows with treatment for inflammatory health problem (endometritis, fever, clinical mastitis, vaginal discharge or retained placenta); OHP: cows with no IHP but treatment for other health problem (milk fever, cystic ovaries, claw, and leg problems, rumen and intestine problems or other diseases); NHP: cows with no treatments, in the first 6 weeks after calving]. Energy balance was not different among health status groups. The IHP cows had lower nonesterified fatty acids and greater insulin concentration in plasma compared with OHP. The IHP cows had lower plasma albumin concentration, lower LAI and higher haptoglobin concentration compared with OHP and NHP. Overall, EB time profiles were associated with the metabolic status of dairy cows in early lactation, but were only limitedly related with markers of inflammation and oxidative stress status. Inflammatory and metabolic status were related to disease events in early lactation and caused prolonged effects on liver metabolism.
Association between uterine disease and indicators of neutrophil and systemic energy status in lactating Holstein cows

Abstract:The objective of this study was to evaluate the association between uterine disease and indicators of neutrophil (PMN) and systemic energy status in dairy cows. Peripheral blood (120 mL) was collected weekly from 84 Holstein cows for PMN isolation and plasma collection from calving until 42 d in milk (DIM). The final analysis included 80 cows. Of those, 20 cows were classified as having metritis (fetid uterine discharge and fever), 15 as having subclinical endometritis (SCE; >or=10% PMN on uterine cytology), and 45 as healthy controls. Plasma haptoglobin concentration was increased only in cows that developed metritis. Neutrophil glycogen content was reduced in cows developing metritis compared with healthy cows on the day of calving and at 7 and 42 DIM. Cows with SCE had lower PMN glycogen content than healthy cows at 7, 28, and 42 DIM. Blood glucose was affected by disease status within parity. Primiparous metritis cows had greater blood glucose concentrations than healthy primiparous cows. Multiparous metritis cows tended to have lower blood glucose concentration than multiparous SCE cows. Cows that developed metritis and SCE had or tended to have greater NEFA and BHBA than healthy cows, mainly around calving. At calving, cows that developed metritis had higher plasma estradiol concentration than healthy cows and greater plasma cortisol than cows that had SCE. Plasma insulin was not affected. Plasma glucagon was increased for SCE cows. Cows that developed uterine disease experienced a greater degree of negative energy balance and had decreased lower intracellular PMN glycogen levels, which could be a major predisposing factor for disease because of decreased availability of oxidative fuels.

Abstract:This study aimed to analyze the possible relationship between days to conception and different oxidative stress (OS) biomarkers and liver functional parameters in multiparous dairy cows. Besides, a fast reliable method for the accurate measurement of malondialdehyde (MDA) by liquid chromatography–tandem mass spectrometry was developed in several matrices. During lactation, the days to conception of 28 cows were determined for a retrospective study. According to this parameter, cows were divided into two groups: high and low days to conception (HDC and LDC, respectively). Blood, urine and liver biopsies were sampled 21 days before the expected calving date, and 7 and 21 days after calving. The method developed for MDA was validated according to international requirements. The lower limit of quantification was 0.25 μmol/L for plasma and urine and 10.00 μmol/L for liver tissue. No differences between groups were observed in the systemic concentration of non-esterified fatty acids, β-hydroxybutyric acid and liver triacylglycerol content (P > 0.05). Cholesterol concentration was higher in the LDC than in the HDC group (P < 0.05). Plasma 3-nitrotyrosine (3-NT) concentration was lower in the LDC than in the HDC group on day 21 post-calving (P < 0.05). Superoxide dismutase activity was higher in the LDC than in the HDC group (P < 0.05). Particularly, in the liver, 3-NT and MDA concentrations were lower in the LDC than in the HDC group (P < 0.05). These results allow inferring that the amelioration of OS biomarkers in plasma and liver could be related to a better reproductive performance of dairy cows.

Abstract:Limited information exists about the relationship of adipose tissue with inflammation, oxidative stress, and energy metabolism during the transition period in dairy cows. The objective of this study was to assess the changes and relation of some adipokines, cytokines, oxidative biomarkers, and serum biochemical parameters related to energy balance (EB) in cows during the transition period. Thirty multiparous Holstein cows were selected based on estimated parturition date, and blood samples were collected from jugular vein on one-week prepartum and one and three weeks postpartum and used to measure the parameters. The serum levels of beta-hydroxybutyric acid (BHB), non-esterified fatty acid, cholesterol, high-density lipoprotein (HDL), aspartate aminotransferase, and total antioxidant capacity increased significantly, and glucose, urea, triglyceride (TG), and low-density lipoprotein (LDL) decreased significantly after parturition. The serum values of adiponectin, resistin, leptin, and cytokines including interleukin 6 (IL-6) and tumor necrosis factor (TNF)-α were not changed significantly during the experiment. The results of the Pearson correlation revealed a significant negative correlation between BHB with glucose, albumin, cholesterol, HDL, LDL, and a positive correlation with TG and malondialdehyde. Also, there was a significant direct correlation between insulin and leptin, adiponectin, resistin, IL-6 and TNF-α in the whole experiment period. These emphasize the difficulty of dairy cows to manage the energy requirements during the transition period. It can be stated that adipokines and cytokines may have an essential role in the metabolic status in this period, and control of their production and, or secretion could be helpful in EB during the transition period.

Abstract:The prepartal dietary energy level is tightly correlated with the degree of tissue mobilization that the animal experiences around parturition (giving birth). To better understand the link between the dry period dietary energy management and the inflammatory status around parturition, 12 multiparous Holstein cows were fed for the entire dry period either a high-wheat straw/lower-energy diet to supply at least 100% of the calculated net energy for lactation (NEL) (control, CON) or a higher-energy diet to supply >140% of NEL (overfed, OVE). The blood was sampled throughout the transition period for biomarker analyses. Liver tissue samples were taken on days -14, 7, 14, and 30 relative to parturition for triacylglycerol (TAG) composition and gene expression analysis. Fifty genes involved in inflammation, endoplasmic reticulum (ER), and oxidative stress, and cell cycle and growth were evaluated. Although blood biomarkers did not reveal signs of a greater inflammatory status compared with OVE, CON cows had a greater activation of the intrahepatic unfolded protein response prepartum. However, postpartum mRNA profiling indicated that the OVE group experienced a mild but sustained level of ER stress, with higher oxidative stress and impairment of antioxidant mechanisms. After parturition, inflammation-related genes were upregulated in OVE cows compared with CON. However, CON cows experienced a gradual increase in expression of key inflammatory transcription regulators up to 30 days postpartum which agreed with the lower plasma albumin and cholesterol, suggesting an inflammatory state. Data underscored that ER stress is not necessarily linked with inflammation during the peripartal period. Gene expression data also suggest that prepartum overnutrition could have negative effects on normal cell cycle activity. Overall, allowing cows to overconsume energy prepartum increased the hepatic pro-inflammatory response prepartum and up to the point of parturition. Subsequently, cows fed the lower-energy diet experienced a gradual increase in the inflammatory response. The lack of differences between groups in voluntary feed intake and lactation capacity suggests that nutritional management prepartum triggers different mechanisms that affect ER and oxidative stress along with inflammation. Although no clinical disorders were detected, these alterations expose animals to the development of immuno-metabolic disorders.

Abstract:The dry-off of dairy cows represents an important phase of the lactation cycle, influencing the outcome of the next lactation. Among the physiological changes, the severity of the inflammatory response can vary after the dry-off, and this response might have consequences on cow adaptation in the transition period. The plasma protein profile is a diagnostic tool widely used in humans and animals to assess the inflammatory status and predict the outcome of severe diseases. The albumin-to-globulin ratio (AG) can represent a simple and useful proxy for the inflammatory condition. In this study, we investigated the relationship between AG before dry-off and inflammation, metabolic profile, and performance of 75 Holstein dairy cows. Blood samples were collected from -62 (7 d before dry-off) to 28 d relative to calving (DFC) to measure metabolic profile biomarkers, inflammatory variables, and liver function. Daily milk yield in the first month of lactation was recorded. Milk composition, body condition score, fertility, and health status were also assessed. The AG calculated 1 wk before dry-off (-62 DFC) was used to retrospectively group cows into tertiles (1.06 ± 0.09 for HI, 0.88 ± 0.04 for IN, and 0.72 ± 0.08 for LO). Data were subjected to ANOVA using the PROC MIXED program in SAS software. Differences among groups observed at -62 DFC were almost maintained throughout the period of interest, but AG peaked before calving. According to the level of acute-phase proteins (haptoglobin, ceruloplasmin, albumin, cholesterol, retinol-binding protein), bilirubin, and paraoxonase, a generally overall lower inflammatory condition was found in HI and IN than in the LO group immediately after the dry-off but also after calving. The HI cows had greater milk yield than LO cows, but no differences were observed in milk composition. The somatic cell count reflected the AG ratio trend, with higher values in LO than IN and HI either before dry-off or after calving. Fertility was better in HI cows, with fewer days open and services per pregnancy than IN and LO cows. Overall, cows with high AG before dry-off showed an improved adaptation to the new lactation, as demonstrated by a reduced systemic inflammatory response and increased milk yield than cows with low AG. In conclusion, the AG ratio before dry-off might represent a rapid and useful proxy to evaluate the innate immune status and likely the ability to adapt while switching from the late lactation to the nonlactating phase and during the transition period with emphasis on early lactation.

Abstract:A delayed recovery of the reproductive tract from natural inflammatory processes associated with postpartum involution will compromise further reproductive function. Following a literature review, we selected serum amyloid A (SAA) and α1-acid glycoprotein (α1-AGP) to assess as potential circulating markers of acute uterine inflammation, as concentrations of these 2 acute phase proteins were reported to be elevated early postpartum in dairy cows with active uterine infection. Convenience serum samples from an induced model of uterine infection were used to measure concentrations of these markers. Infection was induced by infusing either 107 or 109 cfu of Trueperella pyogenes (n = 9 cows each; bacteria group n = 18) or saline as a control (n = 18) into the uterus at 48 d postpartum. Although infection stimulated an increase in uterine polymorphonuclear neutrophils, SAA and α1-AGP concentrations in serum were not different between infusion groups. Cows were subsequently classified into uterine health groups based on the presence of endometritis, with or without the presence of T. pyogenes in uterine culture in response to uterine infusion. Mean SAA concentrations were greater in cows that were either endometritis negative – infection positive (n = 9), endometritis positive – infection negative (n = 5), or endometritis positive – infection positive, compared with the endometritis negative – infection negative (n = 11) cows. There was no difference between uterine health groups for α1-AGP concentrations. Mean SAA and α1-AGP concentrations changed over time, reaching maximum concentrations on d 3 and 7 post-infusion, respectively, before decreasing thereafter to d 17 post-infusion.
Prepartum anti-inflammatory therapies in Holstein dairy cows blocked by parity and body condition score group: Effects on metabolic stress, systemic inflammation, performance, and health

Abstract:The objective of this study was to assess the effects of prepartum administration of anti-inflammatory therapies on body condition score (BCS), β-hydroxybutyrate (BHB) concentration, haptoglobin (HP) concentration, milk yield, milk components, rumination time, clinical health events and reproductive performance in Holstein dairy cows. At 14 d before the expected calving date, cows (PAR; n = 170) and heifers (nulliparous [NUL]; n = 63) were blocked by BCS group (optimal = 3-3.5 [OPT]; over-conditioned cows [OVERC; BCS ≥ 3.75 pts.]) and parity (NUL; PAR) and randomly allocated to one of 3 treatment groups: 1) ASA (n = 78): receive one oral administration of acetylsalicylic acid (4 boluses; 480 grain/bolus); 2) MEL (n = 76): receive one oral administration with meloxicam (1mg/kg of BW), or 3) PLC (n = 77): receive one oral treatment with gelatin capsules filled with water. Body condition score was assessed, and blood samples were collected, weekly starting one week before treatment until 3 weeks after calving. Daily milk yields and daily rumination times were collected from on-farm computer records. Dairy Herd Improvement Association (DHIA) monthly test data were collected to assess milk yield, somatic cell counts, and milk components. Furthermore, health events, culling rate, and reproductive performance data were collected from on-farm computer records. The data were analyzed using MIXED, GLIMMIX, and LIFETEST procedures of SAS as a randomized complete block design. On average, MEL-NUL cows produced 4.77 ± 0.93 kg/d and 4.81 ± 0.92 kg/d more milk from wk 6 to wk 21 of lactation compared with ASA-NUL and PLC-NUL cows, respectively. Similarly, there was a week by treatment by body condition group interaction (P = 0.01), where OVERC cows treated with MEL produced more milk from wk 10 to wk 15 of lactation compared with ASA- OVERC and PLC-OVERC cows. Parous cows treated with ASA had lower BCS compared with PAR cows treated with MEL or PLC. A lower percentage of OVERC cows treated with ASA became sick in the first 60 DIM compared with MEL- OVERC and PLC- OVERC cows (ASA = 23.88 ± 7.26%, MEL = 46.36 ± 8.57%; PLC = 46.74 ± 8.53%; P = 0.04). Parous cows treated with ASA had (P = 0.03) a higher hazard ratio to become pregnant by 300 DIM compared with PAR MEL cows. Although the study was not sized for finding treatment differences in blocking criteria groups, these results suggest that treatment with prepartum anti-inflammatory therapies may have positive effects on milk yield and postpartum health in specific groups of cows, such as NUL and OVERC cows, while it may not be recommended for other animal categories, such as parous cows and cows with optimal BCS. Larger studies are needed to strengthen the associations observed in this study.
Comparison of behavioral and physiological changes in postpartum healthy and inflammatory diseased cows

Abstract:During parturition, cows often experience intense pain and stress, which increases the risk of inflammatory diseases. This study aimed to compare the postpartum health status between healthy cows and those diagnoses with inflammatory diseases by examining behavioral and heart rate (HR) variability (HRV) changes, to provide information before the onset of disease. Eight Holstein cows were used in this study. HR, parameters of HRV (low-frequency power: LF; high-frequency power: HF; LF/HF ratio, and total power) and time budget of individual maintenance behaviors (standing, recumbency, feeding, rumination while standing and lying, and sleep) were continuously recorded from 0 to 168 h postpartum. Milk and blood samples were collected daily. Cows were categorized as diseases based on the positive result of California mastitis test and/or serum haptoglobin concentration that exceeded 50 μg/ml after all blood samples have been collected. Compared to healthy individuals (n = 3), diseased cows (n = 5) exhibited higher HR, LF/HF, and lower total power (p < 0.05), suggesting the dominance of the sympathetic nervous system in cows with inflammatory diseases. Additionally, diseased cows showed an increased standing time budget and reduced recumbency (p < 0.05), which may be a behavioral strategy in response to discomfort from inflammation.

Abstract:Abstract Background: A standard chemotherapy for recurrent/metastatic salivary gland cancers has not been established. Combination chemotherapy of carboplatin and paclitaxel should be evaluated as a treatment option. Methods: This study retrospectively reviewed salivary gland cancer patients who received combination chemotherapy of carboplatin and paclitaxel. The differences in objective responses and in the prognoses according to the different pathological diagnoses were evaluated. Results: A total of 38 patients were enrolled in the study; of them, 18 had salivary duct carcinomas (SDCs), nine had adenoid cystic carcinomas (ACCs), and 11 had other pathological diagnoses. Objective responses were observed in 15 (39%) patients. The median progression-free survival (PFS) was 6.5 months, and the median overall survival (OS) was 26.5 months. ACC patients had relatively low response rates (9%), but there were no significant differences in PFS or OS compared to other sub-types. The treatment was well tolerated, with few adverse events. Conclusion: Salivary gland cancer patients showed a moderate clinical response to the combination chemotherapy of carboplatin and paclitaxel. The objective response rates differed according to the pathological diagnoses, but there were no significant differences in prognoses.

Abstract:The objective of this study was to determine the effect of feeding different combinations of dietary vitamin A supplementation (0 or 110 IU/kg body weight), protein (10.3% or 12.2%), and an ionophore (monensin at 0 or 400 mg/day) on retinoid metabolism and immune function of dairy cows. Eighty multiparous Holstein dairy cows were studied from d −35 to +21 relative to expected parturition in a complete randomized block design with a 2 × 2 × 2 factorial arrangement of treatments. The significance of treatments was declared at p ≤ 0.05. Dairy cows receiving high crude protein (CP) diets with monensin had a greater retinol-binding protein serum concentration than cows receiving high CP diets without monensin (p = 0.04). Animals supplemented with vitamin A showed lower SCC (p = 0.04) and a higher thiobarbituric acid reactive substances concentration (p = 0.06) than cows non-supplemented. Moreover, cows receiving low crude protein diets had a greater haptoglobin concentration (p = 0.01). In addition, cows fed a high crude protein diet had a greater TNF-α expression in peripheral blood mononuclear cells (p = 0.04). Animals fed diets without monensin had a greater serum haptoglobin on day 3 postpartum than those fed monensin (p = 0.01). Moreover, dietary vitamin A increased serum 13-cis retinoic acid postpartum. We conclude that vitamin A, crude protein levels, and monensin fed during the close-up period affect milk somatic cell count, some vitamin statuses, and inflammatory markers during early lactation.
