Abstract:Wheat breeding programs are currently focusing on using non-destructive and cost-effective hyperspectral sensing tools to expeditiously and accurately phenotype large collections of genotypes. This approach is expected to accelerate the development of the abiotic stress tolerance of genotypes in breeding programs. This study aimed to assess salt tolerance in wheat genotypes using non-destructive canopy spectral reflectance measurements as an alternative to direct laborious and time-consuming phenological selection criteria. Eight wheat genotypes and sixteen F8 RILs were tested under 150 mM NaCl in real field conditions for two years. Fourteen spectral reflectance indices (SRIs) were calculated from the spectral data, including vegetation SRIs and water SRIs. The effectiveness of these indices in assessing salt tolerance was compared with four morpho-physiological traits using genetic parameters, SSR markers, the Mantel test, hierarchical clustering heatmaps, stepwise multiple linear regression, and principal component analysis (PCA). The results showed significant differences (p ≤ 0.001) among RILs/cultivars for both traits and SRIs. The heritability, genetic gain, and genotypic and phenotypic coefficients of variability for most SRIs were comparable to those of measured traits. The SRIs effectively differentiated between salt-tolerant and sensitive genotypes and exhibited strong correlations with SSR markers (R2 = 0.56–0.89), similar to the measured traits and allelic data of 34 SSRs. A strong correlation (r = 0.27, p < 0.0001) was found between the similarity coefficients of SRIs and SSR data, which was higher than that between measured traits and SSR data (r = 0.20, p < 0.0003) based on the Mantel test. The PCA indicated that all vegetation SRIs and most water SRIs were grouped with measured traits in a positive direction and effectively identified the salt-tolerant RILs/cultivars. The PLSR models, which were based on all SRIs, accurately and robustly estimated the various morpho-physiological traits compared to using individual SRIs. The study suggests that various SRIs can be integrated with PLSR in wheat breeding programs as a cost-effective and non-destructive tool for phenotyping and screening large wheat populations for salt tolerance in a short time frame. This approach can replace the need for traditional morpho-physiological traits and accelerate the development of salt-tolerant wheat genotypes.

Abstract:Studying the physiological mechanisms underlying the traits associated with salt tolerance in genotypes could lead to the discovery of new genetic resources for salt tolerance. In this study, the mechanisms of salt tolerance were evaluated, based on ionic, physiological, and agronomic traits in four varieties that differ in their salt tolerance and in 18 F8 recombinant inbred lines (RILs) grown in real field conditions. The salt tolerance of plant materials was assessed under both normal (3.5 mM NaCl) and high salinity stress (150 mM NaCl) conditions for two consecutive years. Different growth and physiological traits were assessed 75 days after sowing, while ion contents in the shoots, grain yield, and its components were determined at the maturity stage. Multivariate analysis was used to conduct a comprehensive evaluation of salt tolerance across various genotypes and traits. The ANOVA results showed significant differences (p ≤ 0.05 and 0.001) among salinity, genotypes, and their interactions for all ionic and agro-physiological traits, with a few exceptions. Salinity stress resulted in a considerable increase in Na+ content and canopy temperature (CT), with a simultaneous decrease of 11.3% to 94.5% in other ionic and agro-physiological traits compared to the control treatment. However, the salt-tolerant genotypes showed minimal increases in Na+ content and CT, as well as decreases in other ionic and agro-physiological traits when compared to salt-sensitive genotypes under salinity stress. All ionic and agro-physiological traits exhibited strong correlations with each other under salinity stress, but these correlations were weak or insignificant under control conditions. The principal component analysis identified Na+ and CT as negative indicators and other ionic and agro-physiological traits as positive indicators for salt tolerance under salinity stress. The negative indicators were strongly linked to salt-sensitive genotypes, while the positive indicators were closely associated with salt-tolerant genotypes. Heatmap clustering, using multiple traits, successfully differentiated the salt-tolerant genotypes from the salt-sensitive ones. The salt-tolerant group showed a significant reduction in Na+ content by 36.9%, in CT by 10.0%, and in HI by 16.7%, along with an increase of 6.3–51.4% in other ionic and agro-physiological traits compared to the salt-sensitive group. In conclusion, the mechanisms associated with Na+ exclusion and high K+/Na+ and Ca2+/Na+ ratios, as well as chlorophyll and relative water content, along with low CT, resulted in significant improvements in growth and yield under salinity stress conditions. Given that the effectiveness of various ionic and agro-physiological traits in evaluating salt tolerance in wheat has been proven in real field conditions, these traits will play a key role in the development of salt-tolerant wheat genotypes.

Abstract:Developing new bread wheat varieties that can be successfully grown in saline conditions has become a pressing task for plant breeders. High-throughput phenotyping tools are crucial for this task. Proximal remote sensing is gaining popularity in breeding programs as a quick, cost-effective, and non-invasive tool to assess canopy structure and physiological traits in large genetic pools. Limited research has been conducted on the effectiveness of combining RGB and thermal imaging to assess the salt tolerance of different wheat genotypes. This study aimed to evaluate the effectiveness of combining several indices derived from thermal infrared and RGB images with artificial neural networks (ANNs) for assessing relative water content (RWC), chlorophyll a (Chla), chlorophyll b (Chlb), total chlorophyll (Chlt), and plant dry weight (PDW) of 18 recombinant inbred lines (RILs) and their 3 parents irrigated with saline water (150 mM NaCl). The results showed significant differences in various traits and indices among the tested genotypes. The normalized relative canopy temperature (NRCT) index exhibited strong correlations with RWC, Chla, Chlb, Chlt, and PDW, with R2 values ranging from 0.50 to 0.73, 0.53 to 0.76, 0.68 to 0.84, 0.68 to 0.84, and 0.52 to 0.76, respectively. Additionally, there was a strong relationship between several RGB indices and measured traits, with the highest R2 values reaching up to 0.70. The visible atmospherically resistant index (VARI), a popular index derived from RGB imaging, showed significant correlations with NRCT, RWC, Chla, Chlb, Chlt, and PDW, with R2 values ranging from 0.49 to 0.62 across two seasons. The different ANNs models demonstrated high predictive accuracy for NRCT and other measured traits, with R2 values ranging from 0.62 to 0.90 in the training dataset and from 0.46 to 0.68 in the cross-validation dataset. Thus, our study shows that integrating high-throughput digital image tools with ANN models can efficiently and non-invasively assess the salt tolerance of a large number of wheat genotypes in breeding programs.

The characterization of wheat genotypes for salinity tolerance using morpho-physiological indices under hydroponic conditions

Abstract:Salinity affects plant growth, development, yield, and is a big challenge for wheat growth across the globe. Possible feasible solution is creation of salt-tolerant material, genetic variation is a criterion to developing genetically superior individuals. To assess the genetic variation for salt tolerance, nationally and internationally-derived 81 wheat genotypes were selected and evaluated in 0- and 150-mM salt in nutritional culture at seedling stage. Results indicate that salinity levels reveal significant (p ≤ 0.01) differences for fresh root weight (RW), shoot length (SL), fresh shoot weight (SW), total plant length (TL), total fresh weight (TW), root/shoot weight ratio (RSWR), root/shoot length ratio (RSLR), and relative growth rate for weight (RGR-Wt). While, there was no difference for root length (RL). Hierarchical Clustering and Pairwise correlation analysis showed TW, RGR-Wt, SL, SW, and RW were positively correlated among themselves, whereas RL had poor correlations with all the traits except TL and RSLR. Hence, selection of SL can improve the performance of other parameters. Based on PCA analysis, SW and RGR-Wt were the major discriminative components for wheat genotypes. Present study explained that shoot related parameters could be used as a selection criterion to categorize salt-tolerant genotypes. Outperforming genotypes 1104 and 1106 in saline conditions could be used as parents in creation of salt-tolerant wheat genotypes, and parameters such as SL, SW, TW, and RGR-Wt for early screening will be important for creating salt-tolerant and high yielding wheat genotypes.

Abstract:Soil salinity is one of the major constraints to crop production worldwide. The multifaceted nature of salinity tolerance traits complicates plant screening and the identification of salt‐tolerant germplasm to be used for the genetic advancement of corps. Many screening criteria have been suggested to distinguish between genotypes. Most of these were applied under controlled environmental conditions and limited to one developmental stage of plants. As a result, most of the reported tolerance could not be validated under field conditions. This study employed a membership function value (MFV) to assess NaCl tolerance of eight wheat (Triticum aestivum) genotypes measured at germination, vegetative and reproductive stages, as an integrative tool for the overall plant performance. Salt stresses had an adverse effect on plant physiological (residual transpiration, stomatal density, chlorophyll fluorescence characteristics; leaf N, Na+ and K+ content) and agronomical (plant height; biomass; root and tiller number; grain) characteristics. Based on this assessment, plants were divided into three contrasting groups: salt tolerant, moderately salt tolerant and salt sensitive. Although genotypes did not show the same degree of tolerance in germination, glasshouse and field experiments, the variety Yu‐07 showed consistently better performance in all trials, whilst Aus19720 was most sensitive in glasshouse and field experiments. These contrasting genotypes could be of a potential value for further studies to uncover the genetic mechanisms governing salt stress response in wheat.

Abstract:The climate has drastically changed over the past two decades. Rising temperatures and climate change may lead to increased evapotranspiration, specifically soil evaporation, causing water to evaporate and salt to accumulate in the soil, resulting in increased soil salinity. As a result, there is a need to evaluate methods for predicting and monitoring the effects of salinity on crop growth and production through rapid screening. Our study was conducted on 20 wheat genotypes, 10 sensitive and 10 tolerant, exposed to two salinity levels (90 and 120 mM NaCl) with the control under greenhouse conditions. Our results revealed significant differences in the genotypes’ response to salinity. Salt stress decreased chlorophyll index in sensitive genotypes but increased chlorophyll a and carotenoids in tolerant genotypes at 90 mM. Salt stress also increased protein, proline, lipoxygenase, and reactive thiobarbituric acid levels in all wheat genotypes. The study suggests that plant photosynthetic efficiency is a reliable, non-destructive biomarker for determining the salt tolerance of wheat genotypes, while other biochemical traits are destructive and time-consuming and therefore not suitable for rapid screening.

Abstract:Objectives: Soil salinity affects the growth of crop plants, leading to reduced productivity, and is a major challenge for wheat production worldwide. Various adaptations and mitigation approaches in combination with tolerant wheat genotypes can be useful for the sustainability of crop production in saline environments. However, the development of salt-tolerant wheat genotypes is one of the best and most efficient solutions for obtaining desirable yields. Considering these issues, an investigation was carried out under hydroponic nutrient culture conditions to assess the genetic variability and selection of salt-tolerant wheat genotypes by categorizing inequitable morphophysiological and genetic variability as well as multivariate analysis. Methods: To meet the objectives of this study, 100 wheat genotypes were tested hydroponically in 0 (control) and 15 dS m-1 salt solutions. Conclusion: For all the wheat genotypes grown under saline conditions, the shoot length (SL), root length (RL), shoot fresh weight (SFW), root fresh weight (RFW), total fresh weight (TFW), shoot dry weight (SDW), root dry weight (RDW), and total dry weight (TDW) decreased significantly. Furthermore, significant variation was observed among the genotypes in terms of their characteristics only under saline conditions. In the case of genetic diversity analysis, a high genotypic coefficient of variation (GCV), phenotypic coefficient of variation (PCV), genetic advance in the percentage of the mean (GAM) and high heritability (h2b) were recorded for all tested wheat genotypes based on the SDW, RDW and TDW. Correlation analysis for both genotypic and phenotypic relationships revealed strong positive correlations for TDW, SDW, TFW and SFW. Principal component analysis (PCA) revealed that TDW, TFW, SDW, and SFW were the most discriminative variables for the wheat genotypes, which was confirmed by discriminant function analysis (DFA). PCA-biplot analysis also revealed significant positive correlations between SDW and SFW and between TDW and TFW. Hierarchical cluster analysis was performed for ten clusters based on the relative performance of the genotypes, where the genotypes were characterized into salt-tolerant, medium-salt-tolerant, medium-salt-susceptible and salt-susceptible groups. Among the genotypes, G11, G25 and G29 under cluster VII were categorized as salt tolerant based on their outstanding performance in terms of characteristics only under saline conditions. D2 analysis proved that the wheat genotypes of this cluster were highly divergent from the other cluster genotypes; as a result, these genotypes might be utilized as parents in the development of salt-tolerant wheat genotypes. The current study concluded that SDW and TDW could be employed as criteria for selecting and defining salt-tolerant genotypes during the early growth stage of wheat.
Ability of Different Growth Indicators to Detect Salt Tolerance of Advanced Spring Wheat Lines Grown in Real Field Conditions

Abstract:Plant growth indicators (GIs) are important for evaluating how different genotypes respond to normal and stress conditions separately. They consider both the morphological and physiological components of plants between two successive growth stages. Despite their significance, GIs are not commonly used as screening criteria for detecting salt tolerance of genotypes. In this study, 36 recombinant inbred lines (RILs) along with four genotypes differing in their salt tolerance were grown under normal and 150 mM NaCl in a two-year field trial. The performance and salt tolerance of these germplasms were assessed through various GIs. The analysis of variance showed highly significant variation between salinity levels, genotypes, and their interaction for all GIs and other traits in each year and combined data for two years, with a few exceptions. All traits and GIs were significantly reduced by salinity stress, except for relative growth rate (RGR), net assimilation rate (NAR), and specific leaf weight (SLW), which increased under salinity conditions. Traits and GIs were more correlated with each other under salinity than under normal conditions. Principal component analysis organized traits and GIs into three main groups under both conditions, with RGR, NAR, and specific leaf area (SLA) closely associated with grain yield (GY) and harvest index, while leaf area duration (LAD) was closely associated with green leaf area (GLA), plant dry weight (PDW), and leaf area index (LAI). A hierarchical clustering heatmap based on GIs and traits organized germplasms into three and four groups under normal and salinity conditions, respectively. Based on the values of traits and GIs for each group, the germplasms varied from high- to low-performing groups under normal conditions and from salt-tolerant to salt-sensitive groups under salinity conditions. RGR, NAR, and LAD were important factors determining genotypic variation in GY of high- and low-performing groups, while all GIs, except leaf area duration (LAR), were major factors describing genotypic variation in GY of salt-tolerant and salt-sensitive groups. In conclusion, different GIs that reveal the relationship between the morphological and physiological components of genotypes could serve as valuable selection criteria for evaluating the performance of genotypes under normal conditions and their salt tolerance under salinity stress conditions.

Abstract:Soil salinity is a worldwide issue that affects wheat production. A comprehensive understanding of salt-tolerance mechanisms and the selection of reliable screening indices are crucial for breeding salt-tolerant wheat cultivars. In this study, 30 wheat genotypes (obtained from a rapid selection of 96 original varieties) were chosen to investigate the existing screening methods and clarify the salinity tolerance mechanisms in wheat. Ten-day-old seedlings were treated with 150 mM NaCl. Eighteen agronomic and physiological parameters were measured. The results indicated that the effects of salinity on the agronomic and physiological traits were significant. Salinity stress significantly decreased K + content and K + /Na + ratio in the whole plant, while the leaf K + /Na + ratio was the strongest determinant of salinity tolerance and had a significantly positive correlation with salt tolerance. In contrast, salinity stress significantly increased Na + concentration and relative gene expression ( TaHKT1;5 , TaSOS1 , and TaAKT1-like ). The Na + transporter gene ( TaHKT1;5 ) showed a significantly greater increase in expression than the K + transporter gene ( TaAKT1-like ). We concluded that Na + exclusion rather than K + retention contributed to an optimal leaf K + /Na + ratio. Furthermore, the present exploration revealed that, under salt stress, tolerant accessions had higher shoot water content, shoot dry weight and lower stomatal density, leaf sap osmolality, and a significantly negative correlation was observed between salt tolerance and stomatal density. This indicated that changes in stomata density may represent a fundamental mechanism by which a plant may optimize water productivity and maintain growth under saline conditions. Taken together, the leaf K + /Na + ratio and stomatal density can be used as reliable screening indices for salt tolerance in wheat at the seedling stage.

Abstract:The tolerance to salinity stress is an intricate phenomenon at cellular and whole plant level that requires the knowledge of contributing physiological and biochemical processes and the genetic control of participating traits. In this context, present study was conducted with objective to evaluate the physiological, biochemical, and genetic responses of different wheat genotypes including bread wheat (BW) and synthetic hexaploids (SHs) under saline and control environment. The experiment was conducted in two factorial arrangement in randomized complete block design (RCBD), with genotypes as one factor and treatments as another factor. A significant decline in physiological traits (chlorophyll, photosynthesis, stomatal conductance, transpiration, and cell membrane stability) was observed in all genotypes due to salt stress; however, this decline was higher in BW genotypes as compared to four SH genotypes. In addition, the biochemical traits including enzymes [superoxide dismutase, catalase, and peroxidase (POD)] activity, proline, and glycine betaine (GB) illustrated significant increase along with increase in the expression of corresponding genes ( TaCAT1 , TaSOD , TaPRX2A , TaP5CS , and TaBADH-A1 ) due to salt stress in SHs as compared to BW. Correspondingly, highly overexpressed genes, TaHKT1;4 , TaNHX1 , and TaAKT1 caused a significant decline in Na + /K + in SH as compared to BW genotypes under salt stress. Moreover, correlation analysis, principal component analysis (PCA), and heatmap analysis have further confirmed that the association and expression of physiological and biochemical traits varied significantly with salinity stress and type of genotype. Overall, the physiological, biochemical, and genetic evaluation proved SHs as the most useful stock for transferring salinity tolerance to other superior BW cultivars via the right breeding program.

Identification and Characterization of Wheat Germplasm for Salt Tolerance

Abstract:Salinity is one of the limiting factors of wheat production worldwide. A total of 334 internationally derived wheat genotypes were employed to identify new germplasm resources for salt tolerance breeding. Salt stress caused 39, 49, 58, 55, 21 and 39% reductions in shoot dry weight (SDW), root dry weight (RDW), shoot fresh weight (SFW), root fresh weight (RFW), shoot height (SH) and root length (RL) of wheat, respectively, compared with the control condition at the seedling stage. The wheat genotypes showed a wide genetic and tissue diversity for the determined characteristics in response to salt stress. Finally, 12 wheat genotypes were identified as salt-tolerant through a combination of one-factor (more emphasis on the biomass yield) and multifactor analysis. In general, greater accumulation of osmotic substances, efficient use of soluble sugars, lower Na+/K+ and a higher-efficiency antioxidative system contribute to better growth in the tolerant genotypes under salt stress. In other words, the tolerant genotypes are capable of maintaining stable osmotic potential and ion and redox homeostasis and providing more energy and materials for root growth. The identified genotypes with higher salt tolerance could be useful for developing new salt-tolerant wheat cultivars as well as in further studies to underline the genetic mechanisms of salt tolerance in wheat.

Abstract:Successfully promoting drought tolerance in wheat genotypes will require several procedures, such as field experimentations, measuring relevant traits, using analysis tools of high precision and efficiency, and taking a complementary approach that combines analyses of phenotyping and genotyping at once. The aim of this study is to assess the genetic diversity of 60 genotypes using SSR (simple sequence repeat) markers collected from several regions of the world and select 13 of them as more genetically diverse to be re-evaluated under field conditions to study drought stress by estimating 30 agro-physio-biochemical traits. Genetic parameters and multivariate analysis were used to compare genotype traits and identify which traits are increasingly efficient at detecting wheat genotypes of drought tolerance. Hierarchical cluster (HC) analysis of SSR markers divided the genotypes into five main categories of drought tolerance: four high tolerant (HT), eight tolerant (T), nine moderate tolerant (MT), six sensitive (S), and 33 high sensitive (HS). Six traits exhibit a combination of high heritability (>60%) and genetic gain (>20%). Analyses of principal components and stepwise multiple linear regression together identified nine traits (grain yield, flag leaf area, stomatal conductance, plant height, relative turgidity, glycine betaine, polyphenol oxidase, chlorophyll content, and grain-filling duration) as a screening tool that effectively detects the variation among the 13 genotypes used. HC analysis of the nine traits divided genotypes into three main categories: T, MT, and S, representing three, five, and five genotypes, respectively, and were completely identical in linear discriminant analysis. But in the case of SSR markers, they were classified into three main categories: T, MT, and S, representing five, three, and five genotypes, respectively, which are both significantly correlated as per the Mantel test. The SSR markers were associated with nine traits, which are considered an assistance tool in the selection process for drought tolerance. So, this study is useful and has successfully detected several agro-physio-biochemical traits, associated SSR markers, and some drought-tolerant genotypes, coupled with our knowledge of the phenotypic and genotypic basis of wheat genotypes.

Abstract:The aim of this study was to evaluate the impact of salt stress on morphological, yield, biochemical, and molecular attributes of different barley genotypes. Ten genotypes were cultivated at Fayoum Research Station, El-Fayoum Governorate, Egypt, during two seasons (2020-2021 and 2021-2022), and they were exposed to two different salt concentrations (tap water as a control and 8000 ppm). The results showed that genotypes and salt stress had a significant impact on all morphological and physiological parameters. The morphological parameters (plant height) and yield attributes (spike length, number of grains per spike, and grain yield per plant) of all barley genotypes were significantly decreased under salt stress as compared to control plants. Under salt stress, the total soluble sugars, proline, total phenol, total flavonoid, ascorbic acid, malondialdehyde, hydrogen peroxide, and sodium contents of the shoots of all barley genotypes significantly increased while the potassium content decreased. L1, which is considered a sensitive genotype was more affected by salinity stress than the tolerance genotypes L4, L6, L9, and Giza 138. SDS-PAGE of seed proteins demonstrated high levels of genetic variety with a polymorphism rate of 42.11%. All genotypes evaluated revealed significant variations in the seed protein biochemical markers, with new protein bands appearing and other protein bands disappearing in the protein patterns of genotypes cultivated under various conditions. Two molecular marker techniques (SCoT and ISSR primers) were used in this study. Ten Start Codon Targeted (SCoT) primers exhibited a total of 94 fragments with sizes ranging from 1800 base pairs to 100 base pairs; 29 of them were monomorphic, and 65 bands, with a polymorphism of 62.18%, were polymorphic. These bands contained 21 unique bands (9 positive specific markers and 12 negative specific markers). A total of 54 amplified bands with molecular sizes ranging from 2200 to 200 bp were produced using seven Inter Simple Sequence Repeat (ISSR) primers; 31 of them were monomorphic bands and 23 polymorphic bands had a 40.9% polymorphism. The techniques identified molecular genetic markers associated with salt tolerance in barley crop and successfully marked each genotype with distinct bands. The ten genotypes were sorted into two main groups by the unweighted pair group method of arithmetic averages (UPGMA) cluster analysis based on molecular markers and data at a genetic similarity coefficient level of 0.71.
Physiological responses, ion accumulation and yield performance of wheat ( Triticum aestivum L.) to salt stress

Abstract:Wheat is affected by various biotic and abiotic stresses, especially salinity, which reduces the growth and yield drastically. High salinity is a major constraint for wheat productivity in many countries, including Bangladesh. With this view, here, an experiment was conducted to observe genotypic differences in physiological, ion accumulation, agro-morphological and yield performance of wheat against different levels of salinity. Experimental variables consisted of five salt tolerant genotypes (G 20–2–2, G 20–1–2, G 13–2, G 22–2, G 9–2), one susceptible genotype (G 24–2) and one standard check variety (BARI ghom 25), which assigned to four levels of salinity with electrical conductivities control (0.3), 4, 8 and 12 dS m −1 . Irrespective of genotypes, salinity stress significantly decreased the yield and yield attributes. Results of analyses based on salt tolerance indices of plant growth related and yield contributing parameters, ionic balance (Na + , K + and Na + / K + ratio), and stress indicators such as chlorophyll content, photosynthetic rate (Pn), stomatal conductance (gs) and transpiration rate (Tr) revealed genotypes G 20–2–2, G 13–2 and G 20–1–2 as salt tolerant, genotype G 9–2 as moderately salt-tolerant and G 24–2 and G 22–2 as salt-sensitive genotypes. Additionally, lower accumulations of hydrogen peroxide and malondialdehyde, and higher activities of antioxidant enzymes in the salt-tolerant genotypes G 20–2–2 and G 13–2 than in the salt-sensitive genotype G 24–2 indicated reduced oxidative damage in genotypes G 20–2–2 and G 13–2 relative to that in genotype G 24–2. Collectively, our findings suggest that the optimum growth and yield of salt tolerant genotypes are associated with decreased Na + / K + ratio, increased antioxidant enzymes activity and reduced oxidative stress.

Abstract:Increasing atmospheric temperature can significantly reduce global wheat productivity; despite a mounting demand for wheat grain supplies. Developing genotypes with superior performance under current and future hot climates is a key challenge for wheat breeders. Multidimensional tools have supported plant breeders in increasing the genetic stability rate of agro-physiological indices that influence wheat productivity. We used 25 agro-physiological indices to classify 20 bread wheat genotypes for their heat stress tolerance. Agro-physiological indices and multidimensional analyses to identify differences in genetic and phenotypic were used, combining these analyses to reach selection criteria of accurate and credible. The 25 studied indices reflected high genotypic and environmental variations. We used 16 indices, which have brought together high heritability and genetic gain as indicators for screening heat-tolerant genotypes. Based on the seven principal comprehensive indices of (D value), wheat genotypes were classified into three highly heat-tolerant, four heat-tolerant, six moderately heat-tolerant, five heat-sensitive, and two highly heat-sensitive wheat genotypes. Based on four critical indices [grain yield (GY), grain-filling duration (GFD), spike length (SL) and canopy temperature (CT)] obtained from stepwise multiple linear regression (SMLR), the genotypes were grouped as four genotypes highly heat-tolerant, six heat-tolerant, two moderately heat-tolerant, four heat-sensitive and four highly heat-sensitive. The classification D value and SMLR distances were significantly correlated based on the Mantel test, with a perfect match in nine genotypes. SMLR indicated that a mathematical equation for the evaluation of wheat heat tolerance was established: GY = 0.670 + 0.504 × GFD + 0.334 × SL − 0.466 × CT (R2 = 0.739; average prediction accuracy of 94.12%). SMLR-based classification of wheat genotypes for heat tolerance was further verified through discriminant analysis, which showed that prior and posterior classification was identical in eighteen genotypes. Cross-validation showed that prior and posterior classification was identical in thirteen genotypes. Based on this study, we can recommend tolerated new wheat lines (DHL25, DHL05, DHL23 and DHL08) and cultivar Pavone-76 as a promising genetic source for heat-tolerant breeding programs.

Abstract:A genetic framework underpinning salinity tolerance at reproductive stage was revealed by genome-wide SNP markers and major adaptability genes in synthetic-derived wheats, and trait-associated loci were used to predict phenotypes. Using wild relatives of crops to identify genes related to improved productivity and resilience to climate extremes is a prioritized area of crop genetic improvement. High salinity is a widespread crop production constraint, and development of salt-tolerant cultivars is a sustainable solution. We evaluated a panel of 294 wheat accessions comprising synthetic-derived wheat lines (SYN-DERs) and modern bread wheat advanced lines under control and high salinity conditions at two locations. The GWAS analysis revealed a quantitative genetic framework of more than 200 loci with minor effect underlying salinity tolerance at reproductive stage. The significant trait-associated SNPs were used to predict phenotypes using a GBLUP model, and the prediction accuracy (r2) ranged between 0.57 and 0.74. The r2 values for flag leaf weight, days to flowering, biomass, and number of spikes per plant were all above 0.70, validating the phenotypic effects of the loci discovered in this study. Furthermore, the germplasm sets were compared to identify selection sweeps associated with salt tolerance loci in SYN-DERs. Six loci associated with salinity tolerance were found to be differentially selected in the SYN-DERs (12.4 Mb on chromosome (chr)1B, 7.1 Mb on chr2A, 11.2 Mb on chr2D, 200 Mb on chr3D, 600 Mb on chr6B, and 700.9 Mb on chr7B). A total of 228 reported markers and genes, including 17 well-characterized genes, were uncovered using GWAS and EigenGWAS. A linkage disequilibrium (LD) block on chr5A, including the Vrn-A1 gene at 575 Mb and its homeologs on chr5D, were strongly associated with multiple yield-related traits and flowering time under salinity stress conditions. The diversity panel was screened with more than 68 kompetitive allele-specific PCR (KASP) markers of functional genes in wheat, and the pleiotropic effects of superior alleles of Rht-1, TaGASR-A1, and TaCwi-A1 were revealed under salinity stress. To effectively utilize the extensive genetic information obtained from the GWAS analysis, a genetic interaction network was constructed to reveal correlations among the investigated traits. The genetic network data combined with GWAS, selective sweeps, and the functional gene survey provided a quantitative genetic framework for identifying differentially retained loci associated with salinity tolerance in wheat.

Abstract:Drought presents a major risk to wheat growth and productivity under changing climates. During the last few years, various morphological and physiological approaches were used to overcome drought stress-associated problems. Cultivation of tolerant wheat cultivars can serve as a sustainable choice to raise wheat yield under water stress. Herein, field trials were carried out at the experimental farm of Ismailia Agricultural Research Station, Egypt, in two successive growing seasons (2020/2021 and 2021/2022) to investigate the response of four Egyptian bread wheat cultivars (Misr 1, Misr 3, Giza 171, and Sakha 95) to drought stress according to morpho-physiological characteristics, yield, and stress indices. Irrigation treatments and cultivars were assigned to the main and sub-plots, consequently, in a split-plot design with three replicates. The findings revealed that in both the first and second seasons, drought drastically revoked growth vigor of shoot, growth vigor of flag leaf, relative water content (%), membrane stability index (%), photosynthetic pigments, heading (days), maturity (days), as well as yield and yield attributes: spike length, number of spikes/m 2 , spike weight, grain number/spike, 100-kernel weight, grain yield/m 2 , straw yield/m 2 , biological yield/m 2 , and harvest index of all four wheat cultivars. Conversely, drought caused a marked increase in saturation water deficit (%), carotenoids content, and NKP uptake of all four wheat cultivars in both study seasons. The current study found that all four of the wheat cultivars were drought-tolerant plants. These cultivars exhibited similar drought-tolerant behaviors, which included decreased loss in relative water content, membrane stability, and photosynthetic pigment levels, consequently reducing wheat grain yield loss under water stress. Additionally, the drought tolerance indices of Sakha 95 > Giza 171 > Misr 1 > Misr 3 were revealed by the stress sensitivity index (SSI), mean productivity (MP), stress tolerance index (STI), and yield stability index (YSI). In conclusion, Misr 3 was the least tolerant wheat cultivar and Sakha 95 was the most tolerant. These results can be applied to breeding programs by plant breeders.

Abstract:Abstract Background Salinity is one of the major limiting abiotic stresses that decrease crop production worldwide. To recommend genotypes for cultivation under saline stress conditions, a comprehensive understanding of the genetic basis and plant responses to this stress is needed. In the present study, a total of 20 barley genotypes were investigated to identify potential salt-tolerant genotypes, both at the early growth stage using a hydroponic system, and in adult plants under field conditions. For these purposes, the multi-trait genotype-ideotype distance index (MGIDI) was used to identify salt-tolerant barley genotypes at the seedling stage, and the weighted average of absolute scores (WAASB) index was used to identify the high-yielding and stable genotypes in adult plant stage. At the early growth stage, barley seedlings were treated with two salinity levels: 0 mM NaCl (as control conditions) and 200 mM NaCl (as stress conditions) for 30 days, and during this period different growth and physiological traits were measured. Besides, the yield performance and stability of the investigated barley genotypes were evaluated across five environments during the 2018–2020 cropping seasons. Results Salinity stress significantly decreased growth and physiological traits in all seedling plants; however, some salt-tolerant genotypes showed minimal reduction in the measured traits. Multivariate analysis grouped the measured traits and genotypes into different clusters. In the early growth stage, the G12, G14, G6, G7, and G16 were selected as the most salt-tolerant genotypes using MGIDI index. In the multi-environment trials experiment, AMMI analysis showed that grain yields of the tested barley genotypes were influenced by the environment (E), genotype (G), and GE interaction. Based on the weighted average of absolute scores of the genotype index (WAASB) and other stability statistics, G7, G8, G14, and G16 were selected as superior genotypes. Conclusion Together the MGIDI and WAASB indices revealed that three genotypes—G7, G14 and G16—can be recommended as new genetic resources for improving and stabilizing grain yield in barley programs for the moderate climate and saline regions of Iran. Our results suggest that using the MGIDI index in the early growth stage can accelerate screening nurseries in barley breeding programs. Besides, the WAASB index can be used as a useful stability measurement for identify high-yielding and stable genotypes in multi-environment trials.
