Abstract:The climate has drastically changed over the past two decades. Rising temperatures and climate change may lead to increased evapotranspiration, specifically soil evaporation, causing water to evaporate and salt to accumulate in the soil, resulting in increased soil salinity. As a result, there is a need to evaluate methods for predicting and monitoring the effects of salinity on crop growth and production through rapid screening. Our study was conducted on 20 wheat genotypes, 10 sensitive and 10 tolerant, exposed to two salinity levels (90 and 120 mM NaCl) with the control under greenhouse conditions. Our results revealed significant differences in the genotypes’ response to salinity. Salt stress decreased chlorophyll index in sensitive genotypes but increased chlorophyll a and carotenoids in tolerant genotypes at 90 mM. Salt stress also increased protein, proline, lipoxygenase, and reactive thiobarbituric acid levels in all wheat genotypes. The study suggests that plant photosynthetic efficiency is a reliable, non-destructive biomarker for determining the salt tolerance of wheat genotypes, while other biochemical traits are destructive and time-consuming and therefore not suitable for rapid screening.

Abstract:Wheat is affected by various biotic and abiotic stresses, especially salinity, which reduces the growth and yield drastically. High salinity is a major constraint for wheat productivity in many countries, including Bangladesh. With this view, here, an experiment was conducted to observe genotypic differences in physiological, ion accumulation, agro-morphological and yield performance of wheat against different levels of salinity. Experimental variables consisted of five salt tolerant genotypes (G 20–2–2, G 20–1–2, G 13–2, G 22–2, G 9–2), one susceptible genotype (G 24–2) and one standard check variety (BARI ghom 25), which assigned to four levels of salinity with electrical conductivities control (0.3), 4, 8 and 12 dS m −1 . Irrespective of genotypes, salinity stress significantly decreased the yield and yield attributes. Results of analyses based on salt tolerance indices of plant growth related and yield contributing parameters, ionic balance (Na + , K + and Na + / K + ratio), and stress indicators such as chlorophyll content, photosynthetic rate (Pn), stomatal conductance (gs) and transpiration rate (Tr) revealed genotypes G 20–2–2, G 13–2 and G 20–1–2 as salt tolerant, genotype G 9–2 as moderately salt-tolerant and G 24–2 and G 22–2 as salt-sensitive genotypes. Additionally, lower accumulations of hydrogen peroxide and malondialdehyde, and higher activities of antioxidant enzymes in the salt-tolerant genotypes G 20–2–2 and G 13–2 than in the salt-sensitive genotype G 24–2 indicated reduced oxidative damage in genotypes G 20–2–2 and G 13–2 relative to that in genotype G 24–2. Collectively, our findings suggest that the optimum growth and yield of salt tolerant genotypes are associated with decreased Na + / K + ratio, increased antioxidant enzymes activity and reduced oxidative stress.

Abstract:Studying the physiological mechanisms underlying the traits associated with salt tolerance in genotypes could lead to the discovery of new genetic resources for salt tolerance. In this study, the mechanisms of salt tolerance were evaluated, based on ionic, physiological, and agronomic traits in four varieties that differ in their salt tolerance and in 18 F8 recombinant inbred lines (RILs) grown in real field conditions. The salt tolerance of plant materials was assessed under both normal (3.5 mM NaCl) and high salinity stress (150 mM NaCl) conditions for two consecutive years. Different growth and physiological traits were assessed 75 days after sowing, while ion contents in the shoots, grain yield, and its components were determined at the maturity stage. Multivariate analysis was used to conduct a comprehensive evaluation of salt tolerance across various genotypes and traits. The ANOVA results showed significant differences (p ≤ 0.05 and 0.001) among salinity, genotypes, and their interactions for all ionic and agro-physiological traits, with a few exceptions. Salinity stress resulted in a considerable increase in Na+ content and canopy temperature (CT), with a simultaneous decrease of 11.3% to 94.5% in other ionic and agro-physiological traits compared to the control treatment. However, the salt-tolerant genotypes showed minimal increases in Na+ content and CT, as well as decreases in other ionic and agro-physiological traits when compared to salt-sensitive genotypes under salinity stress. All ionic and agro-physiological traits exhibited strong correlations with each other under salinity stress, but these correlations were weak or insignificant under control conditions. The principal component analysis identified Na+ and CT as negative indicators and other ionic and agro-physiological traits as positive indicators for salt tolerance under salinity stress. The negative indicators were strongly linked to salt-sensitive genotypes, while the positive indicators were closely associated with salt-tolerant genotypes. Heatmap clustering, using multiple traits, successfully differentiated the salt-tolerant genotypes from the salt-sensitive ones. The salt-tolerant group showed a significant reduction in Na+ content by 36.9%, in CT by 10.0%, and in HI by 16.7%, along with an increase of 6.3–51.4% in other ionic and agro-physiological traits compared to the salt-sensitive group. In conclusion, the mechanisms associated with Na+ exclusion and high K+/Na+ and Ca2+/Na+ ratios, as well as chlorophyll and relative water content, along with low CT, resulted in significant improvements in growth and yield under salinity stress conditions. Given that the effectiveness of various ionic and agro-physiological traits in evaluating salt tolerance in wheat has been proven in real field conditions, these traits will play a key role in the development of salt-tolerant wheat genotypes.

Abstract:Bottlenecks, including limited genetic variation and the ongoing loss of genetic diversity, have hindered the development of modern wheat cultivars., making it crucial to use genetic diversity from wild relatives to improve wheat's adaptation to abiotic stress, such as salinity. This study assessed the phenotypic and epigenetic variation of introgressed wheat lines (BC4F2) derived from hybridizing two wheat cultivars with Aegilops cylindrica (AC). This study assessed the phenotypic and epigenetic variation of 156 introgressed wheat lines (BC4F2) derived from hybridization between wheat cultivars "Chinese Spring" (CS) and "Roshan" (R) and Aegilops cylindrica (AC). These lines and their recurrent parents (total of 158) were evaluated under normal and saline field conditions for the agronomic traits and stress tolerance indices. The data were used to select the most tolerant and most sensitive lines. Then, the selected BC4F2 lines and their parents (AC, CS, and R) were subjected to physiological, DNA cytosine methylation, and expression analysis of HKT1;5, NHX1, and SOS1 genes under control and salt stress conditions. Agro-physiological, epigenetic, and gene expression analyses showed the significant effects of salt stress and genetic background, as well as the differential response of the BC4F2 lines to salt stress. The variations in leaf and root K, Na, and K/Na ratios, and leaf Chla, Chlb, Car, and MDA levels, unlike DPPH radical scavenging levels, between salt-tolerant and salt-sensitive BC4F2 lines under saline conditions indicated a substantial distinction in salinity tolerance responses. RT-qPCR indicated higher expression levels of NHX1 and SOS1 genes in the leaf and root tissues of tolerant lines than those of sensitive lines. Global leaf and root DNA methylation analysis revealed the significant effects of salinity on the methylation modifications and confirmed the successful introgression of the salt-tolerance epigenome from Ae. cylindrica into wheat. Exploiting the genetic diversity of wild wheat relatives is a crucial goal for increasing genetic and epigenetic variation to enhance plant adaptation to salt stress.

Abstract:A greenhouse hydroponic experiment was performed using salt-tolerant (cv. Suntop) and -sensitive (Sunmate) wheat cultivars and a salt-tolerant barley cv. CM72 to evaluate how cultivar and species differ in response to salinity stress. Results showed that wheat cv. Suntop performed high tolerance to salinity, being similar tolerance to salinity with CM72, compared with cv. Sunmate. Similar to CM72, Suntop recorded less salinity induced increase in malondialdehyde (MDA) accumulation and less reduction in plant height, net photosynthetic rate (Pn), chlorophyll content, and biomass than in sensitive wheat cv. Sunmate. Significant time-course and cultivar-dependent changes were observed in the activities of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR) in roots and leaves after salinity treatment. Higher activities were found in CM72 and Suntop compared to Sunmate. Furthermore, a clear modification was observed in leaf and root ultrastructure after NaCl treatment with more obvious changes in the sensitive wheat cv. Sunmate, rather than in CM72 and Suntop. Although differences were observed between CM72 and Suntop in the growth and biochemical traits assessed and modified by salt stress, the differences were negligible in comparison with the general response to the salt stress of sensitive wheat cv. Sunmate. In addition, salinity stress induced an increase in the Na+ and Na+/K+ ratio but a reduction in K+ concentrations, most prominently in Sunmate and followed by Suntop and CM72.

Abstract:Plant with a great diversity shows several responses towards the biotic and abiotic stresses. Among these abiotic stresses, salinity is the main damaging factor as it reduces the yield of wheat plant with moderate salt tolerance. For its survival, plant undergoes through some genetic, biochemical and physiological changes to tackle the stress. This review mainly describes the conditions where various ions present in the soil, especially sodium and chlorine, enter into the plant and the genes or proteins involved with survival mechanism against the damage in plants. Salt stress causes alteration in enzymatic activity and Photosynthesis, oxidative stress, damage of cellular structure and components and ionic imbalance. Ion toxicity stress occur due to accumulation of excessive sodium ion and chloride ion. Transcriptional factors TaPIMP, TaSRG and TaMYBsdu 1 play key role in gene expression mechanism to overcome the stress. High affinity potassium transporter gene family is responsible for salt tolerance in wheat plant. HKT1;4 and HKT1;5 genes are responsible for Na exclusion in Triticum monococcum . Forty QTLs were found with the marker assisted selection in bread wheat for salinity tolerance and some morphological traits, 5 QTLs were related to sodium ion exclusion. In bread wheat, salt stress tolerance mechanism is mainly an exclusion of Na + ions but also include K + ion concentration. The salinity tolerant germplasm MW#293 provides an opportunity for the development of future salinity tolerant bread wheat.

Abstract:Salinity stress is a barrier to crop production, quality yield, and sustainable agriculture. The current study investigated the plant growth promotion, biochemical and molecular characterization of bacterial strain Enterobacter cloacae PM23 under salinity stress (i.e., 0, 300, 600, and 900 mM). E. cloacae PM23 showed tolerance of up to 3 M NaCl when subjected to salinity stress. Antibiotic-resistant Iturin C (ItuC) and bio-surfactant-producing genes (sfp and srfAA) were amplified in E. cloacae PM23, indicating its multi-stress resistance potential under biotic and abiotic stresses. Moreover, the upregulation of stress-related genes (APX and SOD) helped to mitigate salinity stress and improved plant growth. Inoculation of E. cloacae PM23 enhanced plant growth, biomass, and photosynthetic pigments under salinity stress. Bacterial strain E. cloacae PM23 showed distinctive salinity tolerance and plant growth-promoting traits such as indole-3-acetic acid (IAA), siderophore, ACC deaminase, and exopolysaccharides production under salinity stress. To alleviate salinity stress, E. cloacae PM23 inoculation enhanced radical scavenging capacity, relative water content, soluble sugars, proteins, total phenolic, and flavonoid content in maize compared to uninoculated (control) plants. Moreover, elevated levels of antioxidant enzymes and osmoprotectants (Free amino acids, glycine betaine, and proline) were noticed in E. cloacae PM23 inoculated plants compared to control plants. The inoculation of E. cloacae PM23 significantly reduced oxidative stress markers under salinity stress. These findings suggest that multi-stress tolerant E. cloacae PM23 could enhance plant growth by mitigating salt stress and provide a baseline and ecofriendly approach to address salinity stress for sustainable agriculture.
Differences in Physiological and Biochemical Characteristics in Response to Single and Combined Drought and Salinity Stresses Between Wheat Genotypes Differing in Salt Tolerance

Abstract:The combined drought and salinity stresses pose a serious challenge for crop production, but the physiological mechanisms behind the stresses responses in wheat remains poorly understood. Greenhouse pot experiment was performed to study differences in genotype response to the single and combined (D + S) stresses of drought (4% soil moisture, D) and salinity (100 mM NaCl, S) using two wheat genotypes: Jimai22 (salt tolerant) and Yangmai20 (salt-sensitive). Results showed that salinity, drought and/or D + S severely reduces plant growth, biomass and net photosynthetic rate, with a greater effect observed in Yangmai20 than Jimai22. A notable improvement in water use efficiency (WUE) by 239, 77 and 103% under drought, salinity and D + S, respectively, was observed in Jimai22. Moreover, Jimai22 recorded higher root K+ concentration in drought and salinity stressed condition and shoot K+ under salinity alone than that of Yangmai20. Jimai22 showed lower increase in malondialdehyde (MDA) accumulation, but higher activities of superoxide dismutase (SOD, EC 1.15.1.1) and guaicol peroxidase (POD, EC 1.11.1.7), under single and combined stresses, and catalase (CAT, EC 1.11.1.6) and ascorbate peroxidase (APX, EC 1.11.1.11) under single stress. Our results suggest that high tolerance of Jimai22 in both drought and D + S stresses is closely associated with larger root length, higher Fv/Fm and less MDA contents and improved capacity of SOD and POD. Moreover, under drought Jimai22 tolerance is firmly related to higher root K+ concentration level and low level of Na+ , high-net photosynthetic rate and WUE as well as increased CAT and APX activities to scavenge reactive oxygen species.
The characterization of wheat genotypes for salinity tolerance using morpho-physiological indices under hydroponic conditions

Abstract:Salinity affects plant growth, development, yield, and is a big challenge for wheat growth across the globe. Possible feasible solution is creation of salt-tolerant material, genetic variation is a criterion to developing genetically superior individuals. To assess the genetic variation for salt tolerance, nationally and internationally-derived 81 wheat genotypes were selected and evaluated in 0- and 150-mM salt in nutritional culture at seedling stage. Results indicate that salinity levels reveal significant (p ≤ 0.01) differences for fresh root weight (RW), shoot length (SL), fresh shoot weight (SW), total plant length (TL), total fresh weight (TW), root/shoot weight ratio (RSWR), root/shoot length ratio (RSLR), and relative growth rate for weight (RGR-Wt). While, there was no difference for root length (RL). Hierarchical Clustering and Pairwise correlation analysis showed TW, RGR-Wt, SL, SW, and RW were positively correlated among themselves, whereas RL had poor correlations with all the traits except TL and RSLR. Hence, selection of SL can improve the performance of other parameters. Based on PCA analysis, SW and RGR-Wt were the major discriminative components for wheat genotypes. Present study explained that shoot related parameters could be used as a selection criterion to categorize salt-tolerant genotypes. Outperforming genotypes 1104 and 1106 in saline conditions could be used as parents in creation of salt-tolerant wheat genotypes, and parameters such as SL, SW, TW, and RGR-Wt for early screening will be important for creating salt-tolerant and high yielding wheat genotypes.
Multidimensional screening and evaluation of morpho‐physiological indices for salinity stress tolerance in wheat

Abstract:Soil salinity is one of the major constraints to crop production worldwide. The multifaceted nature of salinity tolerance traits complicates plant screening and the identification of salt‐tolerant germplasm to be used for the genetic advancement of corps. Many screening criteria have been suggested to distinguish between genotypes. Most of these were applied under controlled environmental conditions and limited to one developmental stage of plants. As a result, most of the reported tolerance could not be validated under field conditions. This study employed a membership function value (MFV) to assess NaCl tolerance of eight wheat (Triticum aestivum) genotypes measured at germination, vegetative and reproductive stages, as an integrative tool for the overall plant performance. Salt stresses had an adverse effect on plant physiological (residual transpiration, stomatal density, chlorophyll fluorescence characteristics; leaf N, Na+ and K+ content) and agronomical (plant height; biomass; root and tiller number; grain) characteristics. Based on this assessment, plants were divided into three contrasting groups: salt tolerant, moderately salt tolerant and salt sensitive. Although genotypes did not show the same degree of tolerance in germination, glasshouse and field experiments, the variety Yu‐07 showed consistently better performance in all trials, whilst Aus19720 was most sensitive in glasshouse and field experiments. These contrasting genotypes could be of a potential value for further studies to uncover the genetic mechanisms governing salt stress response in wheat.

Abstract:The tolerance to salinity stress is an intricate phenomenon at cellular and whole plant level that requires the knowledge of contributing physiological and biochemical processes and the genetic control of participating traits. In this context, present study was conducted with objective to evaluate the physiological, biochemical, and genetic responses of different wheat genotypes including bread wheat (BW) and synthetic hexaploids (SHs) under saline and control environment. The experiment was conducted in two factorial arrangement in randomized complete block design (RCBD), with genotypes as one factor and treatments as another factor. A significant decline in physiological traits (chlorophyll, photosynthesis, stomatal conductance, transpiration, and cell membrane stability) was observed in all genotypes due to salt stress; however, this decline was higher in BW genotypes as compared to four SH genotypes. In addition, the biochemical traits including enzymes [superoxide dismutase, catalase, and peroxidase (POD)] activity, proline, and glycine betaine (GB) illustrated significant increase along with increase in the expression of corresponding genes ( TaCAT1 , TaSOD , TaPRX2A , TaP5CS , and TaBADH-A1 ) due to salt stress in SHs as compared to BW. Correspondingly, highly overexpressed genes, TaHKT1;4 , TaNHX1 , and TaAKT1 caused a significant decline in Na + /K + in SH as compared to BW genotypes under salt stress. Moreover, correlation analysis, principal component analysis (PCA), and heatmap analysis have further confirmed that the association and expression of physiological and biochemical traits varied significantly with salinity stress and type of genotype. Overall, the physiological, biochemical, and genetic evaluation proved SHs as the most useful stock for transferring salinity tolerance to other superior BW cultivars via the right breeding program.

Abstract:This study investigates the differential responses of two maize genotypes, SC180 and SC168, to salt stress, aiming to elucidate the mechanisms underlying salinity tolerance and identify traits associated with improved stress resilience. Salinity stress, imposed by 150 mM NaCl, adversely affected various growth parameters in both genotypes. SC180 exhibited a more pronounced reduction in shoot length (13.6%) and root length (13.6%) compared to SC168, which showed minimal reductions (3.0% and 2.3%, respectively). Additionally, dry weight losses in SC180's leaves, stems, and roots were significantly greater than those in SC168. Under salinity stress, both genotypes accumulated Na + in all organs, with SC168 showing higher Na + concentrations. However, K + levels decreased more significantly in SC180's leaves than in SC168's. The study also assessed physiological responses, noting that SC180 experienced a substantial reduction in relative water content (RWC) in leaves (22.7%), while SC168's RWC remained relatively stable (5.15%). Proline accumulation, a marker for osmotic adjustment, increased 2.3-fold in SC168 compared onefold in SC180. Oxidative stress indicators, such as electrolyte leakage and hydrogen peroxide levels, were elevated in both genotypes under salt stress, with SC180 showing higher increases (48.5% and 48.7%, respectively) than SC168 (35.25% and 22.0%). Moreover, antioxidant enzymes (APX, CAT, POD, SOD, GR) activities were significantly enhanced in SC168 under salinity stress, whereas SC180 showed no significant changes in these activities. Stress indices, used to quantify and compare salinity tolerance, consistently ranked SC168 as more tolerant (average rank = 1.08) compared to SC180 (average rank = 1.92). Correlation analyses further confirmed that SC168's superior tolerance was associated with better Na + regulation, maintenance of K + levels, and a robust antioxidant defense system. In conclusion, SC168 demonstrated greater resilience to salinity stress, attributed to its efficient ion regulation, stable water status, enhanced osmotic adjustment, and strong antioxidant response. These findings provide valuable insights for breeding and developing salinity-tolerant maize varieties.

Abstract:Salt stress is a prominent abiotic stressor that imposes constraints on grain yield and quality across various crops, including wheat ( Triticum aestivum ). This study focused on assessing the genetic diversity of 20 wheat genotypes categorized as tolerant, moderately tolerant, and sensitive with three genotypes of unknown tolerance. To address salinity stress-related problems, different morpho-physiological, osmoprotectant, biochemical, yield, and grain quality-related parameters were analyzed under control (pH 8.0, EC 3.9) and saline-sodic (pH 9.4, EC 4.02) conditions in field.

Response of winter wheat genotypes to salinity stress under controlled environments

Abstract:This study was conducted in controlled environmental conditions to systematically evaluate multi-traits responses of winter wheat ( Triticum aestivum L.) genotypes to different salinity levels. Responses were assessed at the germination to early seedling stage (Experiment 1). Seeds of different genotypes (n=292) were subjected to three salinity levels (0 [control], 60, and 120 mM NaCl). Principal Component Analysis (PCA) revealed that among studied traits seedling vigor index (SVI) contributed more towards the diverse response of genotypes to salinity stress. Based on SVI, eight contrasting genotypes assumed to be tolerant (Gage, Guymon, MTS0531, and Tascosa) and susceptible (CO04W320, Carson, TX04M410211) were selected for further physio-biochemical evaluation at the booting stage (Experiment 2) and to monitor grain yield. Higher level of salinity (120 mM NaCl) exposure at the booting stage increased thylakoid membrane damage, lipid peroxidation, sugars, proline, and protein while decreasing photosynthesis, chlorophyll index, starch, and grain yield. Based on grain yield, the assumed magnitude of the genotypic response shown in Experiment 1 was not analogous in Experiment 2. This indicates the necessity of individual screening of genotypes at different sensitive growth stages for identifying true salinity-tolerant and susceptible genotypes at a particular growth stage. However, based on higher grain yield and its least percentage reduction under higher salinity, Guymon and TX04M410211 were identified as tolerant, and Gage and CO04W320 as susceptible at the booting stage, and their biparental population can be used to identify genomic regions for booting stage-specific salinity response.

Abstract:Objectives: Soil salinity affects the growth of crop plants, leading to reduced productivity, and is a major challenge for wheat production worldwide. Various adaptations and mitigation approaches in combination with tolerant wheat genotypes can be useful for the sustainability of crop production in saline environments. However, the development of salt-tolerant wheat genotypes is one of the best and most efficient solutions for obtaining desirable yields. Considering these issues, an investigation was carried out under hydroponic nutrient culture conditions to assess the genetic variability and selection of salt-tolerant wheat genotypes by categorizing inequitable morphophysiological and genetic variability as well as multivariate analysis. Methods: To meet the objectives of this study, 100 wheat genotypes were tested hydroponically in 0 (control) and 15 dS m-1 salt solutions. Conclusion: For all the wheat genotypes grown under saline conditions, the shoot length (SL), root length (RL), shoot fresh weight (SFW), root fresh weight (RFW), total fresh weight (TFW), shoot dry weight (SDW), root dry weight (RDW), and total dry weight (TDW) decreased significantly. Furthermore, significant variation was observed among the genotypes in terms of their characteristics only under saline conditions. In the case of genetic diversity analysis, a high genotypic coefficient of variation (GCV), phenotypic coefficient of variation (PCV), genetic advance in the percentage of the mean (GAM) and high heritability (h2b) were recorded for all tested wheat genotypes based on the SDW, RDW and TDW. Correlation analysis for both genotypic and phenotypic relationships revealed strong positive correlations for TDW, SDW, TFW and SFW. Principal component analysis (PCA) revealed that TDW, TFW, SDW, and SFW were the most discriminative variables for the wheat genotypes, which was confirmed by discriminant function analysis (DFA). PCA-biplot analysis also revealed significant positive correlations between SDW and SFW and between TDW and TFW. Hierarchical cluster analysis was performed for ten clusters based on the relative performance of the genotypes, where the genotypes were characterized into salt-tolerant, medium-salt-tolerant, medium-salt-susceptible and salt-susceptible groups. Among the genotypes, G11, G25 and G29 under cluster VII were categorized as salt tolerant based on their outstanding performance in terms of characteristics only under saline conditions. D2 analysis proved that the wheat genotypes of this cluster were highly divergent from the other cluster genotypes; as a result, these genotypes might be utilized as parents in the development of salt-tolerant wheat genotypes. The current study concluded that SDW and TDW could be employed as criteria for selecting and defining salt-tolerant genotypes during the early growth stage of wheat.

Abstract:Physiological and biochemical parameters of plants among five wheat genotypes: KH-65, KRL-210, KRL-99, PBW-343 and PBW-373 were studied. Wheat plantlets, at three-leaf stage, were supplemented with 0, 50, 100, 150, 200, 250 and 300 mM of NaCl for 48 hours. Principal component analysis revealed chlorophyll and carotenoid degradation as best salinity indicator for studied wheat genotypes. Salt tolerance levels of studied wheat genotypes were in the order: KH-65 greater than KRL-210 greater than KRL-99 greater than PBW-343 greater than PBW-373. The study has revealed that observed physiological and biochemical data may provide an insight into the existence of internal mechanism in salt tolerant genotypes to cope up with salinity stress.
Resemblance and Difference of Seedling Metabolic and Transporter Gene Expression in High Tolerance Wheat and Barley Cultivars in Response to Salinity Stress

Abstract:To elucidate inter-specific similarity and difference of tolerance mechanism against salinity stress between wheat and barley, high tolerant wheat cv. Suntop and sensitive cv. Sunmate and tolerant barley cv. CM72 were hydroponically grown in a greenhouse with 100 mM NaCl. Glutathione, secondary metabolites, and genes associated with Na+ transport, defense, and detoxification were examined to discriminate the species/cultivar difference in response to salinity stress. Suntop and CM72 displayed damage to a lesser extent than in Sunmate. Compared to Sunmate, both Suntop and CM72 recorded lower electrolyte leakage and reactive oxygen species (ROS) production, higher leaf relative water content, and higher activity of PAL (phenylalanine ammonia-lyase), CAD (cinnamyl alcohol dehydrogenase), PPO (polyphenol oxidase), SKDH (shikimate dehydrogenase), and more abundance of their mRNA under salinity stress. The expression of HKT1, HKT2, salt overly sensitive (SOS)1, AKT1, and NHX1 was upregulated in CM72 and Suntop, while downregulated in Sunmate. The transcription factor WRKY 10 was significantly induced in Suntop but suppressed in CM72 and Sunmate. Higher oxidized glutathione (GSSG) content was accumulated in cv. CM72 and Sunmate, but increased glutathione (GSH) content and the ratio of GSH/GSSG were observed in leaves and roots of Suntop under salinity stress. In conclusion, glutathione homeostasis and upregulation of the TaWRKY10 transcription factor played a more important role in wheat salt-tolerant cv. Suntop, which was different from barley cv. CM72 tolerance to salinity stress. This new finding could help in developing salinity tolerance in wheat and barley cultivars.

Abstract:Soil salinity is a worldwide issue that affects wheat production. A comprehensive understanding of salt-tolerance mechanisms and the selection of reliable screening indices are crucial for breeding salt-tolerant wheat cultivars. In this study, 30 wheat genotypes (obtained from a rapid selection of 96 original varieties) were chosen to investigate the existing screening methods and clarify the salinity tolerance mechanisms in wheat. Ten-day-old seedlings were treated with 150 mM NaCl. Eighteen agronomic and physiological parameters were measured. The results indicated that the effects of salinity on the agronomic and physiological traits were significant. Salinity stress significantly decreased K + content and K + /Na + ratio in the whole plant, while the leaf K + /Na + ratio was the strongest determinant of salinity tolerance and had a significantly positive correlation with salt tolerance. In contrast, salinity stress significantly increased Na + concentration and relative gene expression ( TaHKT1;5 , TaSOS1 , and TaAKT1-like ). The Na + transporter gene ( TaHKT1;5 ) showed a significantly greater increase in expression than the K + transporter gene ( TaAKT1-like ). We concluded that Na + exclusion rather than K + retention contributed to an optimal leaf K + /Na + ratio. Furthermore, the present exploration revealed that, under salt stress, tolerant accessions had higher shoot water content, shoot dry weight and lower stomatal density, leaf sap osmolality, and a significantly negative correlation was observed between salt tolerance and stomatal density. This indicated that changes in stomata density may represent a fundamental mechanism by which a plant may optimize water productivity and maintain growth under saline conditions. Taken together, the leaf K + /Na + ratio and stomatal density can be used as reliable screening indices for salt tolerance in wheat at the seedling stage.

Abstract:Agricultural production is exposed to different environmental challenges such as salinity, drought, and global warming; plants have used different physiological and biochemical responses to adapt and survive under abiotic stress conditions. Drought and salinity are the most important abiotic constraints to plant survival and to crop productivity; they furthermore have main harmful effects on the plant tissue such as the negative effects on the cellular energy supply and redox homeostasis that are stable by re-programming of plant primary metabolism and modification of cellular composition. The agricultural sector is considered the main user of freshwater resources in numerous regions of the world, with growing population and increasing water scarcity in various regions of the world, thus reducing the amount of crop production, which affects the availability of global food quantity in the near future. Drought can affect plants in various ways such as decrease in photosynthesis and growth inhibition; accumulation of abscisic acid (ABA), proline, mannitol, sorbitol; formation of radical scavenging compounds (ascorbate, glutathione, α-tocopherol etc.); and synthesis of new proteins and mRJNAs; also, water stress results in stomatal closure and reduced transpiration rates, a reduce in the water potential of plant tissues, and therefore, plants under drought stress use two processing to control the relation between photosynthetic potential and relative water content of leaves; first one through decreased stomatal conductance and reduced photosynthesis and decreased CO2 concentration inside the leaf, while the other one by elevated CO2 which decreases progressively as relative water content (RWC) declines, and reduced gas exchange; with both type there is reducing in metabolic processing in leaf tissue. Salinity stress causes changes in numerous physiological and metabolic processes in plant tissue, unfortunately, the majority of economic crop species are glycophytes, therefore, salinity inhibits crop productivity worldwide. Salinity stress causes changes in various physiological and metabolic processes, depending on severity and duration of the stress, and eventually hamper crop production; glycophytes plant have different physiological mechanisms such as ion homeostasis, compatible solute, antioxidant regulation, and polyamines production. Generally, plants could combine a range of response to avoiding drought and salinity stresses injuries by different mechanisms to be able to complete their life cycle. By using different strategies such as using maximum available resources, they store reserves in plant organs and use them for fruit production; also, plants can tolerate stress conditions by avoiding tissue dehydration.
