Abstract:Attempt of developing bio-safety and functional layered double hydroxides (LDHs) modified plasma electrolytic oxidation (PEO) coatings, has become a hotspot in the protection of magnesium (Mg) based biomedical implants. In the present work, Mg-Fe LDH films with different Fe contents were fabricated on PEO coating via a novel two-step method: firstly, the FeOOH films were prepared by immersing PEO sample in Fe2+-containing solution, and then Mg-Fe LDH films were formed by transforming the FeOOH films via a hydrothermal treatment in water. The highly-oriented LDH nano-sheets could enhance the anti-corrosion performance of PEO coating, which was proved by the results of electrochemical test, hydrogen evolution and corroded morphology. PEO/Mg-Fe LDH coatings showed a low hemolysis rate (less 5%) than PEO coating. In addition, PEO/Mg-Fe LDH coatings were more favorable for cell adhesion and proliferation than PEO coating. Moreover, PEO/Mg-Fe LDH coatings showed good photothermal conversion property, which demonstrated the excellent rapid antibacterial effect under NIR light. In vitro culture of rat bone marrow stem cell (rBMSC) suggested that cells cultured in the extract of PEO/Mg-Fe LDH coatings had a better osteogenic activity. In vivo subcutaneous implantation test revealed that PEO/Mg-Fe LDH coatings exhibited good anti-corrosion and histocompatibility.

Abstract:In recent years, magnesium (Mg) alloys have emerged as possible biodegradable implant materials; however the degradation rate of Mg occurs at a higher rate than tolerable for the human body. Plasma electrolytic oxidation (PEO) has been used in the past as a useful surface treatment technique to improve the anti-corrosion properties of Mg alloys by forming protective coatings. This present work focuses on the effect of electrolyte solution on the corrosion, microstructural, and nanomechanical behavior of PEO coatings for possible use in biodegradable implants. The experimental parameters applied during PEO process did influence the structure, thickness, and morphology of the coating. Microstructural characterization of the coating was carried out by X-ray diffraction (XRD), scanning electron microscopy (SEM) followed by image analysis and energy dispersive spectroscopy (EDX). Further, nanoindentation was employed to evaluate nanohardness and Young’s modulus of the PEO coating. The results show beneficial effects of the PEO coating to enhance the corrosion resistance of the uncoated AZ31 magnesium alloy. The XRD pattern shows that the components of the film vary based on electrolyte solution. The film composition does affect the nanomechanical behavior.
Bioactivity Performance of Pure Mg after Plasma Electrolytic Oxidation in Silicate-Based Solutions

Abstract:The biodegradable metals, including magnesium (Mg), are a convenient alternative to permanent metals but fast uncontrolled corrosion limited wide clinical application. Formation of a barrier coating on Mg alloys could be a successful strategy for the production of a stable external layer that prevents fast corrosion. Our research was aimed to develop an Mg stable oxide coating using plasma electrolytic oxidation (PEO) in silicate-based solutions. 99.9% pure Mg alloy was anodized in electrolytes contained mixtures of sodium silicate and sodium fluoride, calcium hydroxide and sodium hydroxide. Scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), contact angle (CA), Photoluminescence analysis and immersion tests were performed to assess structural and long-term corrosion properties of the new coating. Biocompatibility and antibacterial potential of the new coating were evaluated using U2OS cell culture and the gram-positive Staphylococcus aureus (S. aureus, strain B 918). PEO provided the formation of a porous oxide layer with relatively high roughness. It was shown that Ca(OH)2 was a crucial compound for oxidation and surface modification of Mg implants, treated with the PEO method. The addition of Ca2+ ions resulted in more intense oxidation of the Mg surface and growth of the oxide layer with a higher active surface area. Cell culture experiments demonstrated appropriate cell adhesion to all investigated coatings with a significantly better proliferation rate for the samples treated in Ca(OH)2-containing electrolyte. In contrast, NaOH-based electrolyte provided more relevant antibacterial effects but did not support cell proliferation. In conclusion, it should be noted that PEO of Mg alloy in silicate baths containing Ca(OH)2 provided the formation of stable biocompatible oxide coatings that could be used in the development of commercial degradable implants.

Abstract:The biodegradability of magnesium (Mg) metal implants necessitates surface modification to mitigate the high corrosion rate in the body's physiological environment. This study focuses on synthesizing and applying a nanocomposite coating of polydimethylsiloxane-nanosized bioactive glass (PDMS-nBG) on the surface of a plasma electrolytic oxidation (PEO)-modified AZ91 Mg alloy. The PDMS-1 wt% nBG nanocomposite coating was deposited on the PEO-treated substrate using the dip-coating method. Surface and cross-sectional morphologies of the nanocomposite coatings were examined using field emission scanning electron microscopy (FESEM). Corrosion behavior of the AZ91 substrate, PEO coating, and double-layer PEO/PDMS-nBG coating was investigated using electrochemical impedance spectroscopy (EIS) and potentiodynamic polarization (PDP) tests. Results showed that the corrosion resistance and corrosion current density of Mg substrates increased and decreased from 4815 Ω.cm 2 and 8.96 μA.cm − 2 to 18 MΩ.cm 2 and 0.00016 μA.cm − 2 for AZ91 and PEO/PDMS-nBG coating, respectively. Additionally, the morphology and adhesion of cultured human mesenchymal cells on the PEO-modified AZ91 substrates with a single PDMS polymer coating and a PDMS-nBG nanocomposite coating were investigated using fluorescent light microscopy and FESEM. The PEO/PDMS-nBG bilayer coating effectively addressed the limitations of magnesium alloys and holds potential for biomedical applications, particularly in orthopedic implants, due to the enhanced biocompatibility and bioactivity of Mg alloys.
Biocompatibility and bone regeneration of PEO/Mg-Al LDH-coated pure Mg: an in vitro and in vivo study

Abstract:Forming a stable anti-corrosion surface layer on magnesium (Mg) and its alloys has become a major challenge in developing a desirable degradable medical implant in bone. In this study, a porous MgO layer was first formed on Mg by plasma electrolytic oxidation (PEO), and then a Mg-Al layered double hydroxide (LDH) layer was prepared to seal the porous structure of the PEO layer (LDH-2h and LDH-12h) via hydrothermal treatment. The bilayer structure composite coating, which can effectively resist the penetration of surrounding media, is similar to plain Chinese tiles. The in vitro results revealed that compared with other coatings, the LDH-12h composite coating can reduce the release of Mg ions and induce a milder change in pH when immersed in phosphate-buffered saline (PBS). In vitro rat bone marrow stem cell (rBMSC) culture suggested that the LDH-12h composite coating is favorable for cell activity, proliferation and could improve the osteogenic activity of rBMSCs. A subcutaneous implantation test revealed that the as-prepared sample showed enhanced corrosion resistance and histocompatibility in vivo, especially in the LDH-12h group. Moreover, LDH-12h had the lowest rate of degradation and the closest combination with the new bone after being inserted into a rat femur for 12 weeks with no major organ dysfunction. In summary, the asprepared PEO/Mg-Al LDH composite coating is able to improve the corrosion resistance and biocompatibility of Mg and to enhance osteogenic activity in vivo, suggesting its promising prospects for orthopedic applications.

Abstract:Magnesium and its alloys are considered excellent materials for biodegradable implants because of their good biocompatibility and biodegradability as well as their mechanical properties. However, the rapid degradation rate severely limits their clinical applications. Plasma electrolytic oxidation (PEO), also known as micro-arc oxidation (MAO), is an effective surface modification technique. However, there are many pores and cracks on the coating surface under conventional PEO process. The corrosive products tend to penetrate deeply into the substrate, reducing its corrosion resistance and the biocompatibility, which makes PEO-coated Mg difficult to meet the long-term needs of in vivo implants. Hence, it is necessary to modify the PEO coating. This review discusses the formation mechanism and the influential parameters of PEO coatings on Mg. This is followed by a review of the latest research of the pretreatment and typical amelioration of PEO coating on biodegradable Mg alloys in the past 5 years, including calcium phosphate (Ca-P) coating, layered double hydroxide (LDH)-PEO coating, ZrO2 incorporated-PEO coating, antibacterial ingredients-PEO coating, drug-PEO coating, polymer-PEO composite coating, Plasma electrolytic fluorination (PEF) coating and self-healing coating. Meanwhile, the improvements of morphology, corrosion resistance, wear resistance, biocompatibility, antibacterial abilities, and drug loading abilities and the preparation methods of the modified PEO coatings are deeply discussed as well. Finally, the challenges and prospects of PEO coatings are discussed in detail for the purpose of promoting the clinical application of biodegradable Mg alloys.

Abstract:Magnesium (Mg) is a promising biodegradable metal for orthopedic applications, and plasma electrolytic oxidation (PEO) has been widely studied as a corrosion protection coating on Mg-based implants. However, the porous structures and easily formed cracks in fluid are disadvantageous for long-term corrosion protection. In this study, a SiO 2 layer was deposited on PEO-treated Mg to inhibit the formation of cracks on the PEO layer and prevent the permeation of corrosive fluid. The SiO 2 layer did not alter the surface morphology of the PEO layer but considerably enhanced its corrosion resistance. The in vitro culture of MC3T3-E1 cells demonstrated the good cytocompatibility and osteogenic induction ability of SiO 2 -coated PEO-treated Mg, which could be attributed to Mg and Si ions released from the coating. The coating also favored the angiogenesis behaviors of HUVEC. Furthermore, with the continuous release of Mg and Si ions, the as-prepared implant showed a superior osseointegration ability in a rat bone implantation model. In summary, this newly designed Mg-based implant shows promising potential for orthopedic applications.

Abstract:Biocompatible and biodegradable magnesium (Mg) and its alloys possess excellent potential as orthopedic implant biomaterials, but they corrode too quickly inside the human body to maintain such functions as supporting and fixation. In this study, we have successfully prepared a composite biomedical material with Mg as the substrate and a multilayered coating on the metal, which contains a chemical conversion layer of fluoride or phosphate, an adhesion layer, and a layer of biodegradable polylactic acid (PLA). The recorded weight loss and the release of ionic magnesium (Mg2+) in the simulated body fluid (SBF) demonstrate that the corrosion rate of the protected Mg has been reduced to 1/10 that of the unprotected bare Mg. The corrosion rate of the protected Mg can be adjusted with either the chemical conversion type or the thickness of the PLA layer. The bare and the protected Mg has been manufactured as an implant for bone defect repair of Sprague-Dawley rats. The X-ray and micro-CT images show an improved anticorrosion of the protected Mg in vivo, and the histological analysis indicates that the Mg2+ ions released at an appropriate rate are beneficial for bone growth. The preliminary tests prove that the biodegradable Mg has been effectively protected by the innovative multilayered coating, and the implant has met the required biological functions such as biocompatibility and promotion of bone growth.

Abstract:Magnesium (Mg) alloys have been promised for biomedical implants in orthopedic field, however, the fast corrosion rate and mode challenge their clinical application. To push Mg alloys materials into practice, a composite coating with biodegradable and high compatible components to improve anticorrosion property of an Mg alloy (i.e., AZ31) is designed and fabricated. The inner layer is micro-nano structured Mg(OH)2 through hydrothermal treatment. Then stearic acid (SA) is introduced to modify Mg(OH)2 for better reducing the gap below a surface-degradation polymer layer of poly(1,3-trimethylene carbonate). Benefited by the SA modification effect, this sandwiched coating avoids corrosive medium penetration via enhancing the adhesion strength at the interface between outer and inner layers. Both in vitro and in vivo tests indicate that the composite coating modified AZ31 perform a better anticorrosion behavior and biocompatibility compared to bare AZ31. Strikingly, a 1.7-fold improvement in volume of newly formed bone is observed surrounding the composite coating modified implant after 12 week implantation. The sandwiched biocompatible coating strategy paves a hopeful way for future translational application of Mg alloys orthopedic materials in clinics.

Abstract:Magnesium biometals exhibit great potentials for orthopeadic applications owing to their biodegradability, bioactive effects and satisfactory mechanical properties. However, rapid corrosion of Mg implants in vivo combined with large amount of hydrogen gas evolution is harmful to bone healing process which seriously confines their clinical applications. Enlightened by the superior biocompatibility and corrosion resistance of passive titanium oxide layer automatically formed on titanium alloy, we employ the Ti and O dual plasma ion immersion implantation (PIII) technique to construct a multifunctional TiO2 based nano-layer on ZK60 magnesium substrates for enhanced corrosion resistance, osteoconductivity and antimicrobial activity. The constructed nano-layer (TiO2/MgO) can effectively suppress degradation rate of ZK60 substrates in vitro and still maintain 94% implant volume after post-surgery eight weeks. In animal study, a large amount of bony tissue with increased bone mineral density and trabecular thickness is formed around the PIII treated group in post-operation eight weeks. Moreover, the newly formed bone in the PIII treated group is well mineralized and its mechanical property almost restores to the level of that of surrounding mature bone. Surprisingly, a remarkable killing ratio of 99.31% against S. aureus can be found on the PIII treated sample under ultra-violet (UV) irradiation which mainly attributes to the oxidative stress induced by the reactive oxygen species (ROS). We believe that this multifunctional TiO2 based nano-layer not only controls the degradation of magnesium implant, but also regulates its implant-to-bone integration effectively. STATEMENT OF SIGNIFICANCE: Rapid corrosion of magnesium implants is the major issue for orthopaedic applications. Inspired by the biocompatibility and corrosion resistance of passive titanium oxide layer automatically formed on titanium alloy, we construct a multifunctional TiO2/MgO nanolayer on magnesium substrates to simultaneously achieve superior corrosion resistance, satisfactory osteoconductivity in rat intramedullary bone defect model and excellent antimicrobial activity against S. aureus under UV irradiation. The current findings suggest that the specific TiO2/MgO nano-layer on magnesium surface can achieve the three objectives aforementioned and we believe this study can demonstrate the potential of biodegradable metals for future clinical applications.
Biodegradable Magnesium Bone Implants Coated with a Novel Bioceramic Nanocomposite

Abstract:Magnesium (Mg) alloys are being investigated as a biodegradable metallic biomaterial because of their mechanical property profile, which is similar to the human bone. However, implants based on Mg alloys are corroded quickly in the body before the bone fracture is fully healed. Therefore, we aimed to reduce the corrosion rate of Mg using a double protective layer. We used a magnesium-aluminum-zinc alloy (AZ91) and treated its surface with micro-arc oxidation (MAO) technique to first form an intermediate layer. Next, a bioceramic nanocomposite composed of diopside, bredigite, and fluoridated hydroxyapatite (FHA) was coated on the surface of MAO treated AZ91 using the electrophoretic deposition (EPD) technique. Our in vivo results showed a significant enhancement in the bioactivity of the nanocomposite coated AZ91 implant compared to the uncoated control implant. Implantation of the uncoated AZ91 caused a significant release of hydrogen bubbles around the implant, which was reduced when the nanocomposite coated implants were used. Using histology, this reduction in the corrosion rate of the coated implants resulted in an improved new bone formation and reduced inflammation in the interface of the implants and the surrounding tissue. Hence, our strategy using a MAO/EPD of a bioceramic nanocomposite coating (i.e., diopside-bredigite-FHA) can significantly reduce the corrosion rate and improve the bioactivity of the biodegradable AZ91 Mg implant.

Abstract:The market for orthopedic implant alloys has seen significant growth in recent years, and efforts to reduce the carbon footprint of medical treatment (i.e., green medicine) have prompted extensive research on biodegradable magnesium-based alloys. Magnesium alloys provide the mechanical strength and biocompatibility required of medical implants; however, they are highly prone to corrosion. In this study, Mg-9Li alloy was immersed in cell culture medium to simulate degradation in the human body, while monitoring the corresponding effects of the reaction products on cells. Variations in pH revealed the generation of hydroxyl groups, which led to cell death. At day-5 of the reaction, a coating of MgCO 3 (H 2 O) 3 , HA, and α -TCP appeared on sample surfaces. The coating presented three-dimensional surface structures (at nanometer to submicron scales), anti-corrosion effects, and an altered surface micro-environment conducive to the adhesion of osteoblasts. This analysis based on bio-simulation immersion has important implications for the clinical use of Mg alloys to secure regenerated periodontal tissue.

Abstract:Magnesium (Mg) alloys have recently attracted attention in biomedicine as biodegradable materials with non-toxic degradable products. Such compounds have become a frontier in the study of biodegradable materials because of their remarkable biomechanical compatibility and superior biocompatibility. The use of Mg-based implants reduces the negative consequences of permanent biological implants by eliminating the necessity for biomaterial surgery following the healing process. However, the quick deterioration, formation of considerable gas of hydrogen volumes and a rise in the body environment pH are obstacles in the application of Mg as an implant material. Hence, compelling advances for erosion resistance and biocompatibility of magnesium and its alloys are noteworthy. Surface modification may be a practical approach because it improves the erosion resistance compared with extensive preparation of a treated surface for progressed bone recovery and cell attachment. Coating produced by plasma electrolytic oxidation (PEO) seems a compelling method in order to enhance magnesium and the properties of its alloys. PEO-formed coatings cannot provide long-term protection in the physiological environment due to their porous nature. Thus, a polymer coating is applied on the porous PEO-formed coating, which is steadily applied on the surface. Polymer coatings improve the biocompatibility properties of Mg and its alloys and increase corrosion resistance. In this article, the most recent advancements in PEO/polymer composite coatings are reviewed, and the biocompatibility of such coatings is examined.

Abstract:Biodegradable materials, especially Magnesium (Mg) and its alloys, are preferred for bone implants due to their compatibility with host tissue and biomechanical support. However, their rapid corrosion and vulnerability to structural failure, compounded by infections from antibiotic-resistant bacteria, pose significant challenges. Addressing these issues, we developed silicate-based composite coatings with varying CuO concentrations (1, 5, and 7 wt%) on Mg using plasma electrolytic oxidation (PEO). This study evaluated the coatings' surface characteristics, degradation behavior, and optimum composition. Our analyses, including XPS and EDX, confirmed the presence and influence of CuO on the coatings. We observed enhanced hydroxyapatite crystallization in CuO-doped coatings, which is critical for implant biocompatibility. These coatings also showed improved corrosion resistance and controlled ion release, likely due to silicate composition and CuO's sealing effects. Antimicrobial assays revealed that CuO concentration influenced bacterial and fungal adhesion, varying with microorganism type. Our findings demonstrate that incorporating inert copper particles into PEO coatings alters their chemical and elemental properties, enhancing bioactivity, corrosion resistance, and antimicrobial efficacy. Abstract:Magnesium AZ31 alloy has been chosen as bio-resorbable temporary prosthetic implants to investigate the degradation processes in a simulating body fluid (SBF) of the bare metal and the ones coated with low and high-molecular-weight PEO hydrogels. Hydrogel coatings are proposed to control the bioresorption rate of AZ31 alloy. The alloy was preliminary hydrothermally treated to form a magnesium hydroxide layer. 2 mm discs were used in bioresorption tests. Scanning electron microscopy was used to characterize the surface morphology of the hydrothermally treated and PEO-coated magnesium alloy surfaces. The variation of pH and the mass of Mg2+ ions present in the SBF corroding medium have been monitored for 15 days. Corrosion current densities (Icorr) and corrosion potentials (Ecorr) were evaluated from potentiodynamic polarisation tests on the samples exposed to the SBF solution. Kinetics of cumulative Mg ions mass released in the corroding solution have been evaluated regarding cations diffusion and mass transport parameters. The initial corrosion rates for the H- and L-Mw PEO-coated specimens were similar (0.95 ± 0.12 and 1.82 ± 0.52 mg/cm2day, respectively) and almost 4 to 5 times slower than that of the uncoated system (6.08 mg/cm2day). Results showed that the highly swollen PEO hydrogel coatings may extend into the bulk solution, protecting the coated metal and efficiently controlling the degradation rate of magnesium alloys. These findings focus more research effort on investigating such systems as tunable bioresorbable prosthetic materials providing idoneous environments to support cells and bone tissue repair.

Abstract:Abstract Nowadays, the need for bio‐implants, which can gradually degrade after fulfilling the therapeutic tasks is continuously increasing. Under such situation, magnesium (Mg) and its alloys have been proposed and intensively studied as the new‐generation medical implants due to their favorable biodegradability and biocompatibility. However, their swift corrosion in physiological environments can always cause an early fracture and further the surgical failure, greatly hindering their broad applications. Therefore, great efforts have been made to alter the degradation behaviors of Mg‐based implants. Biodegradable polymeric surface coatings have been revealed to be a straightforward and effective strategy for retarding the fast degradation and improving the bioactivity of Mg and its alloys. This article reviews the recent progress of polymer‐based coatings on Mg substrates, regarding the coating strategies, coating properties, and their performance in corrosive protection and biocompatibility promotion via in vitro as well as some in vivo models. The specific pros and cons of different polymeric coatings are also discussed. Finally, we put forward some perspectives on the future direction of polymeric coatings on biomedical Mg‐based implants to better adapt to clinical trials.

Abstract:Magnesium (Mg) and its alloys have been intensively studied to develop the next generation of bone implants recently, but their clinical application is restricted by rapid degradation and unsatisfied osteogenic effect in vivo . A bioactive chemical conversion Mg-phenolic networks complex coating (eEGCG) was stepwise incorporated by epigallocatechin-3-gallate (EGCG) and exogenous Mg 2+ on Mg-2 Zn magnesium alloy. Simplex EGCG induced chemical conversion coating (cEGCG) was set as compare group. The in vitro corrosion behavior of Mg-2 Zn alloy, cEGCG and eEGCG was evaluated in SBF using electrochemical (PDP, EIS) and immersion test. The cytocompatibility was investigated with rat bone marrow mesenchymal stem cells (rBMSCs). Furthermore, the in vivo tests using a rabbit model involved micro computed tomography (Micro-CT) analysis, histological observation, and interface analysis. The results showed that the eEGCG is Mg-phenolic multilayer coating incorporated Mg-phenolic networks, which is rougher, more compact and much thicker than cEGCG. The eEGCG highly improved the corrosion resistance of Mg-2 Zn alloy, combined with its lower average hemolytic ratios, continuous high scavenging effect ability and relatively moderate contact angle features, resulting in a stable and suitable biological environment, obviously promoted rBMSCs adhesion and proliferation. More importantly, Micro-CT, histological and interface elements distribution evaluations all revealed that the eEGCG effectively inhibited degradation and enhanced bone tissue formation of Mg alloy implants. This study puts forward a promising bioactive chemical conversion coating with Mg-phenolic networks for the application of biodegradable orthopedic implants.

Abstract:The use of Mg‐based biomaterials with a number of their advantageous properties are overshadowed by uncontrollable metal corrosion. Moreover, the use of implants goes alongside with the threat of pathogens‐associated complications. In this study, PEO coated Mg biomaterial loaded with antibacterial Ag(I) and Cu(II) complexes is produced and tested to meet both appropriate protective characteristics as well as sufficient level of antibacterial activity. To achieve a suitable level of anticorrosion protection phosphate and fluoride‐phosphate electrolytes are used in the PEO process. Investigation of the surface thickness and morphology done by means of cross‐section analysis and scanning electron microscopy (SEM), as well as electrochemical impedance spectroscopy (EIS) assay show precedence of the fluoride containing PEO coating and make it the material of choice for further modification with Ag(I) and Cu(II) complexes. The presence of the complexes on the PEO surface is confirmed by energy dispersive X‐ray spectroscopy (EDX). X‐ray photoelectron spectroscopy (XPS), transmission electron microscopy (TEM) and glow discharge optical emission spectroscopy (GDOES) are used to estimate the complexes' chemical state and depth of penetration in the coating surface. Based on the results of antibacterial assay, the modified coatings are found to be active against both Gram‐positive and Gram‐negative bacteria.
