Abstract:To understand the behavior of particles within optical traps, dynamic analysis of laser trapping is necessary. A mathematical model has been established, which examines all the forces acting on a particle such as trapping force, gravity, buoyancy and Stokes drag. A simulator has been developed based on our mathematical model to define the motion of arbitrary shaped particles exerted primarily by trapping force and optical torque. The movement behavior of a designedtool made of silica trapped in air was predicted and the results showed that high rotation speed can be achieved when the torque produced by the laser beam is balanced by the drag torque exerted by the surrounding air. This gives a good verification that designed-tool made of silica or diamond trapped by laser beam can be used for nano-meter machining.

Abstract:Optical tweezers are widely used in materials assembly<a href="/articles/s41565-021-00852-0#ref-CR1">1</a>, characterization<a href="/articles/s41565-021-00852-0#ref-CR2">2</a>, biomechanical force sensing<a href="/articles/s41565-021-00852-0#ref-CR3">3</a>,<a href="/articles/s41565-021-00852-0#ref-CR4">4</a> and the in vivo manipulation of cells<a href="/articles/s41565-021-00852-0#ref-CR5">5</a> and organs<a href="/articles/s41565-021-00852-0#ref-CR6">6</a>. The trapping force has primarily been generated through the refractive index mismatch between a trapped object and its surrounding medium. This poses a fundamental challenge for the optical trapping of low-refractive-index nanoscale objects, including nanoparticles and intracellular organelles. Here, we report a technology that employs a resonance effect to enhance the permittivity and polarizability of nanocrystals, leading to enhanced optical trapping forces by orders of magnitude. This effectively bypasses the requirement of refractive index mismatch at the nanoscale. We show that under resonance conditions, highly doping lanthanide ions in NaYF4 nanocrystals makes the real part of the Clausius–Mossotti factor approach its asymptotic limit, thereby achieving a maximum optical trap stiffness of 0.086 pN μm–1 mW–1 for 23.3-nm-radius low-refractive-index (1.46) nanoparticles, that is, more than 30 times stronger than the reported value for gold nanoparticles of the same size. Our results suggest a new potential of lanthanide doping for the optical control of the refractive index of nanomaterials, developing the optical force tag for the intracellular manipulation of organelles and integrating optical tweezers with temperature sensing and laser cooling<a href="/articles/s41565-021-00852-0#ref-CR7">7</a> capabilities.

Quantitative measurements of force and displacement using an optical trap

Abstract:We combined a single-beam gradient optical trap with a high-resolution photodiode position detector to show that an optical trap can be used to make quantitative measurements of nanometer displacements and piconewton forces with millisecond resolution. When an external force is applied to a micron-sized bead held by an optical trap, the bead is displaced from the center of the trap by an amount proportional to the applied force. When the applied force is changed rapidly, the rise time of the displacement is on the millisecond time scale, and thus a trapped bead can be used as a force transducer. The performance can be enhanced by a feedback circuit so that the position of the trap moves by means of acousto-optic modulators to exert a force equal and opposite to the external force applied to the bead. In this case the position of the trap can be used to measure the applied force. We consider parameters of the trapped bead such as stiffness and response time as a function of bead diameter and laser beam power and compare the results with recent ray-optic calculations.

Comparing Femtosecond Optical Tweezers with Conventional CW Optical Tweezers

Abstract:In this work, we present a comparative study between continuous-wave (CW) and pulsed optical tweezers for 250 nm, 500 nm and 1-micron radius polystyrene beads at 5 different laser powers. We have used a Ti:Sapphire (MIRA 900F) laser that can be easily switched from CW to pulsed mode of operation, so there is no change in the experimental conditions in the two cases. We have measured the difference in the trap strength in both cases by fitting the power spectrum curve with Lorentzian. As it turns out, trapping with pulsed tweezers seems to be more effective for the smaller particles and as the particle size is increased both CW and pulsed tweezers appear to be equally effective at lower average laser powers but as the power is increased pulsed tweezers do a better job at stable trapping.

Optical Torque Calculations and Measurements for DNA Torsional Studies

Abstract:The angular optical trap (AOT) is a powerful instrument for measuring the torsional and rotational properties of a biological molecule. Thus far, AOT studies of DNA torsional mechanics have been carried out using a high numerical aperture oil-immersion objective, which permits strong trapping, but inevitably introduces spherical aberrations due to the glass-aqueous interface. However, the impact of these aberrations on torque measurements is not fully understood experimentally, partly due to a lack of theoretical guidance. Here, we present a numerical platform based on the finite element method to calculate forces and torques on a trapped quartz cylinder. We have also developed a new experimental method to accurately determine the shift in the trapping position due to the spherical aberrations by using a DNA molecule as a distance ruler. We found that the calculated and measured focal shift ratios are in good agreement. We further determined how the angular trap stiffness depends on the trap height and the cylinder displacement from the trap center and found full agreement between predictions and measurements. As further verification of the methodology, we showed that DNA torsional properties, which are intrinsic to DNA, could be determined robustly under different trap heights and cylinder displacements. Thus, this work has laid both a theoretical and experimental framework that can be readily extended to investigate the trapping forces and torques exerted on particles with arbitrary shapes and optical properties.

Abstract:Optical trapping has proven to be a valuable experimental technique for precisely controlling small dielectric objects. However, due to their very nature, conventional optical traps are diffraction limited and require high intensities to confine the dielectric objects. In this work, we propose a novel optical trap based on dielectric photonic crystal nanobeam cavities, which overcomes the limitations of conventional optical traps by significant factors. This is achieved by exploiting an optomechanically induced backaction mechanism between a dielectric nanoparticle and the cavities. We perform numerical simulations to show that our trap can fully levitate a submicron-scale dielectric particle with a trap width as narrow as 56 nm. It allows for achieving a high trap stiffness, therefore, a high Q-frequency product for the particle's motion while reducing the optical absorption by a factor of 43 compared to the cases for conventional optical tweezers. Moreover, we show that multiple laser tones can be used further to create a complex, dynamic potential landscape with feature sizes well below the diffraction limit. The presented optical trapping system offers new opportunities for precision sensing and fundamental quantum experiments based on levitated particles.

Abstract:Multiphoton absorbing dye-coated trapped spherical bead at the focal plane of femtosecond optical tweezers shows nonlinear optical (NLO) phenomena. One such NLO process of two-photon fluorescence (TPF) has been used for the background-free imaging of a femtosecond laser-trapping event. Due to the high peak powers of femtosecond laser pulses with low average powers, it is possible to not only trap single nanospheres, but encourage optically directed self-assembly. The TPF signatures of trapped particles show evidence of such a directed self-assembly process which, in turn, can provide information about the structural dynamics during the process of cluster formation. We are able to trap and characterize structure and dynamics in 3D until pentamer formation from the decay characteristics of trapping at the focal plane.

Abstract:The angular optical trap (AOT) is a powerful technique for measuring the DNA topology and rotational mechanics of fundamental biological processes. Realizing the full potential of the AOT requires rapid torsional control of these processes. However, existing AOT quartz cylinders are limited in their ability to meet the high rotation rate requirement while minimizing laser-induced photodamage. In this work, we present a novel trapping particle design to meet this challenge by creating small metamaterial elliptical cylinders with tunable trapping force and torque properties. The optical torque of these cylinders arises from their shape anisotropy, with their optical properties tuned via multilayered SiO and Si N deposition. We demonstrate that these cylinders can be rotated at about 3 times the rate of quartz cylinders without slippage while enhancing the torque measurement resolution during DNA torsional elasticity studies. This approach opens new opportunities for previously inaccessible rotational studies of DNA processing.

Abstract:Plasmonic nano-optical tweezers enable the non-invasive manipulation of nano-objects under low illumination intensities, and have become a powerful tool for nanotechnology and biophysics. However, measuring the trap stiffness of nanotweezers remains a complicated task, which hinders the development of plasmonic trapping. Here, we describe an experimental method to measure the trap stiffness based on the temporal correlation of the fluorescence from the trapped object. The method is applied to characterize the trap stiffness in different double nanohole apertures and explore the influence of their design parameters in relationship with numerical simulations. Optimizing the double nanohole design achieves a trap stiffness 10x larger than the previous state-of-the-art. The experimental method and the design guidelines discussed here offer a simple and efficient way to improve the performance of nano-optical tweezers.

Abstract:This paper presents the most complete framework to date for understanding the nonlinear optical trapping of highly absorbing nanoparticles within the dipole regime. Highly absorbing and plasmonic particles garner considerable interest due to their enhanced optical interactions, yet traditional linear theory fails to predict their trapping behavior, particularly in the longitudinal direction. We introduce a more comprehensive theory that incorporates four-wave mixing and two-photon absorption, addressing significant gaps in the current literature. Our research shows TPA to be crucial for longitudinal trapping stability. Furthermore, our findings propose a novel phenomenon of multiple split traps in both saturable absorption and reverse saturable absorption regimes. Our framework extends beyond existing models by including factors such as self-induced back-action and scattering forces, aligning theoretical predictions with experimental observations, and laying a robust foundation for future nonlinear optical trapping and manipulation research.

Abstract:Optical trapping has proven to be an efficient method to control particles, including biological cells, single biological macromolecules, colloidal microparticles, and nanoparticles. Multiple types of particles have been successfully trapped, leading to various applications of optical tweezers ranging from biomedical through physics to material sciences. However, precise manipulation of particles with complex composition or of sizes down to nanometer-scales can be difficult with conventional optical tweezers, and an alternative manipulation tool is desirable. Optical nanofibers, that is, fibers with a waist diameter smaller than the propagating wavelength of light, are ideal candidates for optical manipulation due to their large evanescent field that extends beyond the fiber surface. They have the added advantages of being easily connected to a fibered experimental setup, being simple to fabricate, and providing strong electric field confinement and intense magnitude of evanescent fields at the nanofiber's surface. Many different particles have been trapped, rotated, transported, and assembled with such a system. This article reviews particle trapping using optical nanofibers and highlights some challenges and future potentials of this developing topic.

Abstract:We present experimental evidence of plasmonic-enhanced optical tweezers, of polystyrene beads in deionized water in the vicinity of metal-coated nanostructures. The optical tweezers operate with a continuous wave near-infrared laser. We employ a Cu/Au bilayer that significantly improves dissipation of heat generated by the trapping laser beam and avoid de-trapping from heat convection currents. We investigate the improvement of the optical trapping force and the effective trapping quality factor, and observe an exponential distance dependence of the trapping force from the nanostructures, indicative of evanescent plasmonic enhancement.

Abstract:Low-power trapping of nanoscale objects can be achieved by using the enhanced fields near plasmonic nanoantennas. Unfortunately, in this approach the trap site is limited to the position of the plasmonic hotspots and continuous dynamic manipulation is not feasible. Here, we report a low-frequency electrothermoplasmonic tweezer (LFET) that provides low-power, high-stability and continuous dynamic manipulation of a single nanodiamond. LFET harnesses the combined action of the laser illumination of a plasmonic nanopillar antenna array and low-frequency alternating current (ac) electric field to establish an electrohydrodynamic potential capable of the stable trapping and dynamic manipulation of single nanodiamonds. We experimentally demonstrate the fast transport, trapping, and dynamic manipulation of a single nanodiamond using a low-frequency ac field below 5 kHz and low-laser power of 1 mW. Abstract:Advances in optical tweezers, coupled with the proliferation of two-photon polymerization systems, mean that it is now becoming routine to fabricate and trap non-spherical particles. The shaping of both light beams and particles allows fine control over the flow of momentum from the optical to mechanical regimes. However, understanding and predicting the behaviour of such systems is highly complex in comparison with the traditional optically trapped microsphere. In this Article, we present a conceptually new and simple approach based on the nature of the optical force density. We illustrate the method through the design and fabrication of a shaped particle capable of acting as a passive force clamp, and we demonstrate its use as an optically trapped probe for imaging surface topography. Further applications of the design rules highlighted here may lead to new sensors for probing biomolecule mechanics, as well as to the development of optically actuated micromachines.

Abstract:Optical trapping using focused laser beams (laser tweezers) has been proven to be extremely useful for contactless manipulation of a variety of small objects, including biological cells, organelles within cells, and a wide range of other dielectric micro- and nano-objects. Colloidal metal nanoparticles have drawn increasing attention in the field of optical trapping because of their unique interactions with electromagnetic radiation, caused by surface plasmon resonance effects, enabling a large number of nano-optical applications of high current interest. Here we try to give a comprehensive overview of the field of laser trapping and manipulation of metal nanoparticles based on results reported in the recent literature. We also discuss and describe the fundamentals of optical forces in the context of plasmonic nanoparticles, including effects of polarization, optical angular momentum, and laser heating effects, as well as the various techniques that have been used to trap and manipulate metal nanoparticles. We conclude by suggesting possible directions for future research.

Abstract:Optical manipulation has attracted remarkable interest owing to its versatile and non-invasive nature. However, conventional optical trapping remains inefficient for the nanoscopic world. The emergence of plasmonics in recent years has brought a revolutionary change in overcoming limitations due to diffraction and the requirements for high trapping laser powers. Among the near-field optical trapping cavity-based systems, Fano resonant optical tweezers have a robust trapping capability. In this work, we experimentally demonstrate sequential trapping of 20 nm particles through the use of metamaterial plasmonic optical tweezers. We investigate the multiple trapping via trap stiffness measurements for various trapping positions at low and high incident laser intensities. Our configuration could be used as a light-driven nanoscale sorting device under low laser excitation. Our results provide an alternative approach to trap multiple nanoparticles at distinct hotspots, enabling new ways to control mass transport on the nanoscale.

Enhancing the Applied Force and Range of Axial Optical Tweezers

Abstract:Axial optical tweezers provide a natural geometry for performing biomechanical assays such as rupture force measurements of protein binding. Axial traps, however, are typically weaker than their lateral counterparts requiring high laser power to maintain a well calibrated, linear restoring force. Here we show how to extend the spatial range over which well calibrated forces can be applied by considering aberration effects and extend the range of applied forces by accounting for the nonlinear response that appears when an optically trapped bead is moved far from the trap center. These refinements to the force calibration can be used to apply higher axial forces at reduced laser powers deeper into a sample. To illustrate the method, we reproduce both the linear extension regime and overstretching transition observed in dsDNA at significantly reduced laser powers.

Resonant optical trapping of Janus nanoparticles in plasmonic nanoaperture

Abstract:Controlled trapping of light absorbing nanoparticles with low-power optical tweezers is crucial for remote manipulation of small objects. This study takes advantage of the synergetic effects of tightly confined local fields of plasmonic nanoaperture, self-induced back-action of nanoparticles, and resonant optical trapping method to demonstrate enhanced manipulation of Janus nanoparticles in metallic nanohole aperture. We theoretically demonstrate that displacement of Au-coated Janus nanoparticles toward plasmonic nanoaperture and proper orientation of the metal coating give rise to enhanced near-field intensity and pronounced optical force. We also explore the effect of resonant optical trapping by employing dual laser system, where an on-resonant green laser excites the metal-coated nanoparticle whereas an off-resonant near-infrared laser plays trapping role. It is found that, at optimum nanoparticle configuration, the resonant optical trapping technique can result in three-fold enhancement of optical force, which is attributed to excitation of surface plasmon resonance in Janus nanoparticles. The findings of this study might pave the way for low power optical manipulation of light-absorbing nanoparticles with possible applications in nanorobotics and drug delivery.
