Abstract:Antibiotics in soil environments are a growing concern. Identifying transformation products is key to elucidating degradation pathways and mechanisms of antibiotics and other organic micropollutants. The primary challenge of transformation product identification is the interference of matrices. In this study, stable-isotope assisted nontarget screening was used to identify biodegradation products of florfenicol in soil. A total of 74 candidates were prioritized from thousands of mass features observed by a tiered peak filtering approach. Moreover, with the support of in silico prediction tools, the structures of 12 transformation products were elucidated, and 9 of them were reported for the first time. A biodegradation map of florfenicol consisting of amide hydrolysis, dechlorination, dehydration, defluorination, and sulfone reduction was established based on these identified products. A total of 8 products were also found in 6 field soil samples with manure application. Because of the structural similarity to florfenicol, some transformation products might still keep antimicrobial activity toward a variety of bacterial species. The strategies demonstrated in this study provide a basis for efficient identification of transformation products of other organic micropollutants in a variety of environmental matrices. The results also shed light on the degradation mechanisms, risk assessments, and regulations of these compounds.
Aerobic Microbial Transformation of Fluorinated Liquid Crystal Monomer: New Pathways and Mechanism

Abstract:Fluorinated liquid crystal monomers (FLCMs) have been suggested as emerging contaminants, raising global concern due to their frequent occurrence, potential toxic effects, and endurance capacity in the environment. However, the environmental fate of the FLCMs remains unknown. To fill this knowledge gap, we investigated the aerobic microbial transformation mechanisms of an important FLCM, 4-[difluoro(3,4,5-trifluorophenoxy)methyl]-3, 5-difluoro-4'-propylbiphenyl (DTMDPB), using an enrichment culture termed as BG1. Our findings revealed that 67.5 ± 2.1% of the initially added DTMDPB was transformed in 10 days under optimal conditions. A total of 14 microbial transformation products obtained due to a series of reactions (e.g., reductive defluorination, ether bond cleavage, demethylation, oxidative hydroxylation and aromatic ring opening, sulfonation, glucuronidation, O-methylation, and thiolation) were identified. Consortium BG1 harbored essential genes that could transform DTMDPB, such as dehalogenation-related genes [e.g., glutathione S-transferase gene (GST), 2-haloacid dehalogenase gene (2-HAD), nrdB, nuoC, and nuoD]; hydroxylating-related genes hcaC, ubiH, and COQ7; aromatic ring opening-related genes ligB and catE; and methyltransferase genes ubiE and ubiG. Two DTMDPB-degrading strains were isolated, which are affiliated with the genus Sphingopyxis and Agromyces. This study provides a novel insight into the microbial transformation of FLCMs. The findings of this study have important implications for the development of bioremediation strategies aimed at addressing sites contaminated with FLCMs.

Abstract:The fate of per and polyfluoroalkyl substances (PFASs) in aqueous filmforming foams (AFFFs) under anaerobic conditions has not been well characterized, leaving major gaps in our understanding of PFAS fate and transformation at contaminated sites. In this study, the biotransformation of 6:2 fluorotelomer thioether amido sulfonate (6:2 FtTAoS), a component of several AFFF formulations, was investigated under sulfate-reducing conditions in microcosms inoculated with either pristine or AFFF-impacted solids. To identify the transformation products, we used high-resolution mass spectrometry and employed suspect-screening and nontargeted compound identification methods. These analyses demonstrated that 6:2 FtTAoS was transformed primarily to a stable polyfluoroalkyl compound, 6:2 fluorotelomer thioether propionate (6:2 FtTP). It did not undergo further reactions to produce the perfluoroalkyl carboxylates and fluorotelomer sulfonates and carboxylates that were observed during aerobic transformations. Here, the 6:2 FtTP was recalcitrant to biotransformation, indicating the stability of the thioether group under sulfate reducing conditions. The total oxidizable precursor (TOP) assay was used to assess the presence of other PFASs. Although nearly all of the PFAS mass initially present was recovered from the pristine microcosms, only 67% of the initial PFAS mass was recovered from the contaminated microcosms, suggesting the formation of volatile biotransformation products or those that could not be detected by the TOP assay.

Abstract:Although the use of Sulfluramid (N-ethyl perfluorooctane sulfonamide (N-EtFOSA)) has been restricted by the Stockholm Convention, it is still frequently detected in the environmental matrices and in use in some countries. Employing constructed wetlands as treatment systems requires understanding of the biodegradation process in the rhizosphere and the effect of contaminants on the microbes of wetlands. This study aimed to investigate the interactions between the microbial community and N-EtFOSA under aerobic and anaerobic conditions. Aerobic biotransformation of N-EtFOSA occurred with a half-life of 0.51 day and yielded 85.1 mol% PFOS of after 91 days. Kinetic modelling revealed that cleavage of the SN was the rate-limiting degradation step. Biotransformation was not observed under anaerobic and anoxic conditions, suggesting that N-EtFOSA is recalcitrant to biodegradation without dissolved oxygen. Under aerobic condition, the presence of N-EtFOSA and its biotransformation products decreased the microbial richness and diversity and exerted selective pressure on the microbial community. Enrichment of Methylocaldum was significant (49%) in the presence of N-EtFOSA compared to unexposed conditions (11%), suggesting that Methylocaldum is relatively tolerant to N-EtFOSA and potentially degrading N-EtFOSA. Under anaerobic conditions, the microbial richness and diversity were not significantly altered by the presence of N-EtFOSA. Only Methanomethylovorans increased significantly in the spiked microcosm (30% vs. 20%). These findings provide knowledge for comprehending the contribution of N-EtFOSA to other PFASs in various environmental conditions, information about microbial community changes in response to PFASs and robust microbial species which can degrade N-EtFOSA in the environment.
Generation of Perfluoroalkyl Acids from Aerobic Biotransformation of Quaternary Ammonium Polyfluoroalkyl Surfactants

Abstract:The aerobic biotransformation over 180 days of two cationic quaternary ammonium compounds (QACs) with perfluoroalkyl chains was determined in soil microcosms, and biotransformation pathways were proposed. This is the first time that polyfluoroalkyl cationic surfactants used in aqueous film-forming foam (AFFF) formulations were studied for their environmental fate. The biotransformation of perfluorooctaneamido quaternary ammonium salt (PFOAAmS) was characterized by a DT50 value (time necessary to consume half of the initial mass) of 142 days and significant generation of perfluoroalkyl carboxylic acid (PFOA) at a yield of 30 mol % by day 180. The biotransformation of perfluorooctane sulfonamide quaternary ammonium salt (PFOSAmS) was very slow with unobservable change of the spiked mass; yet the generation of perfluorooctanesulfonate (PFOS) at a yield of 0.3 mol % confirmed the biotransformation of PFOSAmS. Three novel biotransformation intermediates were identified for PFOAAmS and three products including perfluorooctane sulfonamide (FOSA) for PFOSAmS through high-resolution mass spectrometry (MS) analysis and t-MS(2) fragmentation. The significantly slower PFOSAmS biotransformation is hypothesized to be due to its stronger sorption to soil owing to a longer perfluoroalkyl chain and a bulkier sulfonyl group, when compared to PFOAAmS. This study has demonstrated that despite overall high stability of QACs and their biocide nature, the ones with perfluoroalkyl chains can be substantially biotransformed into perfluoroalkyl acids in aerobic soil.

Abstract:Production of 8:2 fluorotelomer alcohol (8:2 FTOH) for industrial and consumer products, including aqueous film-forming foams (AFFFs) used for firefighting, has resulted in its widespread occurrence in the environment. However, the fate of 8:2 FTOH at AFFF-impacted sites remains largely unknown. Using AFFF-impacted soils from two United States Air Force Bases, microcosm experiments evaluated the aerobic biotransformation of 8:2 FTOH (extent and byproduct formation) and the dose-response on microbial communities due to 8:2 FTOH exposure. Despite different microbial communities, rapid transformation of 8:2 FTOH was observed during a 90-day incubation in the two soils, and 7:2 secondary fluorotelomer alcohol (7:2 sFTOH) and perfluorooctanoic acid (PFOA) were detected as major transformation products. Novel transformation products, including perfluoroalkane-like compounds (1H-perfluoroheptane, 1H-perfluorohexane, and perfluoroheptanal) were identified by liquid chromatography-high resolution mass spectrometry (LC-HRMS) and used to develop aerobic 8:2 FTOH biotransformation pathways. Microbial community analysis suggests that species from genus Sphingomonas are potential 8:2 FTOH degraders based on increased abundance in both soils after exposure, and the genus Afipia may be more tolerant to and/or involved in the transformation of 8:2 FTOH at elevated concentrations. These findings demonstrate the potential role of biological processes on PFAS fate at AFFF-impacted sites through fluorotelomer biotransformation.
Interspecies comparison of metabolism of two novel prototype PFAS

Abstract:As a result of proposed global restrictions and regulations on current-use per-and polyfluoroalkyl substances (PFAS), research on possible alternatives is highly required. In this study, phase I in vitro metabolism of two novel prototype PFAS in human and rat was investigated. These prototype chemicals are intended to be safer-by-design and expected to mineralize completely, and thus be less persistent in the environment compared to the PFAS available on the market. Following incubation with rat liver S9 (RL-S9) fractions, two main metabolites per initial substance were produced, namely an alcohol and a short-chain carboxylic acid. While with human liver S9 (HL-S9) fractions, only the short-chain carboxylic acid was detected. Beyond these major metabolites, two and five additional metabolites were identified at very low levels by non-targeted screening for the ether- and thioether-linked prototype chemicals, respectively. Overall, complete mineralization during the in vitro hepatic metabolism of these novel PFAS by HL-S9 and RL-S9 fractions was not observed. The reaction kinetics of the surfactants was determined by using the metabolite formation, rather than the substrate depletion approach. With rat liver enzymes, the formation rates of primary metabolite alcohols were at least two orders of magnitude higher than those of secondary metabolite carboxylic acids. When incubating with human liver enzymes, the formation rates of single metabolite carboxylic acids, were similar or smaller than those experienced in rat. It also indicates that the overall metabolic rate and clearance of surfactants are significantly higher in rat liver than in human liver. The maximum formation rate of the thioether congener exceeded 10-fold that of the ether in humans but were similar in rats. Overall, the results suggest that metabolism of the prototype chemicals followed a similar trend to those reported in studies of fluorotelomer alcohols.

Abstract:Per- and polyfluoroalkyl substances (PFAS) are a class of thousands of highly fluorinated, anthropogenic compounds that are used in a wide variety of consumer applications. Due to their widespread use and high persistence, PFAS are ubiquitous in drinking water, which is of concern due to the threats these compounds pose to human health. Reduction via the hydrated electron is a promising technology for PFAS remediation and has been well-studied. However, since previous work rarely reports fluorine atom balances and often relies on suspect screening, some transformation products are likely unaccounted for. Therefore, we performed non-target analysis using high-resolution mass spectrometry on solutions of perfluorooctanesulfonate (PFOS), perfluorobutanesulfonate (PFBS), perfluorooctanoate (PFOA), and 2,3,3,3-tetrafluoro-2-(heptafluoropropoxy)propanoate (GenX) that had been treated with UV/sulfite to produce hydrated electrons. We determined fluorine atom balances for all compounds studied, finding high fluorine atom balances for PFOS and PFBS. PFOA and GenX had lower overall fluorine atom balances, likely due to the production of volatile or very polar transformation products that were not measured by our methods. Transformation products identified by our analysis were consistent with literature, with a few exceptions. Namely, shorter-chain perfluorosulfonates (PFSA) and their H/F substituted counterparts were also detected from PFOS. This is an unexpected result based on literature, as no documented pathway exists for the formation of shorter-chain PFSA during UV/sulfite treatment. Furthermore, the nontarget approach we employed allowed for identification of novel, unsaturated products from the hydrated electron treatment of perfluorooctanesulfonate (PFOS) that warrant further investigation.
Biotransformation of 6:2/4:2 fluorotelomer alcohols by Dietzia aurantiaca J3: Enzymes and proteomics

Abstract:Per- and polyfluoroalkyl substances (PFAS) are recalcitrant synthetic organohalides known to negatively impact human health. Short-chain fluorotelomer alcohols are considered the precursor of various perfluorocarboxylic acids (PFCAs) in the environment. Their ongoing production and widespread detection motivate investigations of their biological transformation. Dietzia aurantiaca strain J3 was isolated from PFAS-contaminated landfill leachate using 6:2 fluorotelomer sulphonate (6:2 FTS) as a sulphur source. Resting cell experiments were used to test if strain J3 could transform fluorotelomer alcohols (6:2 and 4:2 FTOH). Strain J3 transformed fluorotelomer alcohols into PFCAs, polyfluorocarboxylic acids and transient intermediates. Over 6 days, 80 % and 58 % of 6:2 FTOH (0.1 mM) and 4:2 FTOH (0.12 mM) were degraded with 6.4 % and 14 % fluoride recovery respectively. Fluorotelomer unsaturated carboxylic acid (6:2 FTUCA) was the most abundant metabolite, accounting for 21 to 30 mol% of 6:2 FTOH (0.015 mM) applied on day zero. Glutathione (GSH) conjugates of 6:2/4:2 FTOH and 5:3 FTCA adducts were also structurally identified. Proteomics studies conducted to identify enzymes in the biotransformation pathway have revealed the role of various enzymes involved in β oxidation. This is the first report of 6:2/4:2 FTOH glutathione conjugates and 5:3 FTCA adducts in prokaryotes, and the first study to explore the biotransformation of 4:2 FTOH by pure bacterial strain.
Microbial Defluorination of Unsaturated Per- and Polyfluorinated Carboxylic Acids under Anaerobic and Aerobic Conditions: A Structure Specificity Study

Abstract:The recently discovered microbial reductive defluorination of two C6 branched and unsaturated fluorinated carboxylic acids (FCAs) provided valuable insights into the environmental fate of per- and polyfluoroalkyl substances (PFASs) and potential bioremediation strategies. However, a systematic investigation is needed to further demonstrate the role of C═C double bonds in the biodegradability of unsaturated PFASs. Here, we examined the structure-biodegradability relationships of 13 FCAs, including nine commercially available unsaturated FCAs and four structurally similar saturated ones, in an anaerobic defluorinating enrichment and an activated sludge community. The anaerobic and aerobic transformation/defluorination pathways were elucidated. The results showed that under anaerobic conditions, the α,β-unsaturation is crucial for FCA biotransformation via reductive defluorination and/or hydrogenation pathways. With sp2 C–F bonds being substituted by C–H bonds, the reductive defluorination became less favorable than hydrogenation. Moreover, for the first time, we reported enhanced degradability and defluorination capability of specific unsaturated FCA structures with trifluoromethyl (−CF3) branches at the α/β-carbon. Such FCA structures can undergo anaerobic abiotic defluorination in the presence of reducing agents and significant aerobic microbial defluorination. Abstract:Nearly all per- and polyfluoroalkyl substances (PFAS) biotransformation studies reported to date have been limited to laboratory-scale batch reactors. The fate and transport of PFAS in systems that more closely represent field conditions, i.e., in saturated porous media under flowing conditions, remain largely unexplored. This study investigated the biotransformation of 6:2 fluorotelomer sulfonate (6:2 FTS), a representative PFAS of widespread environmental occurrence, in one-dimensional water-saturated flow-through columns packed with soil obtained from a PFAS-contaminated site. The 305-day column experiments demonstrated that 6:2 FTS biotransformation was rate-limited, where a decrease in pore-water velocity from 3.7 to 2.4 cm/day, resulted in a 21.7–26.1 % decrease in effluent concentrations of 6:2 FTS and higher yields (1.0–1.4 mol% vs. 0.3 mol%) of late-stage biotransformation products (C 4 C 7 perfluoroalkyl carboxylates). Flow interruptions (2 and 7 days) were found to enhance 6:2 FTS biotransformation during the 6–7 pore volumes following flow resumption. Model-fitted 6:2 FTS column biotransformation rates (0.039–0.041 cmw3/gs/d ) were ∼3.5 times smaller than those observed in microcosms (0.137 cmw3/gs/d ). Additionally, during column experiments, planktonic microbial communities remained relatively stable, whereas the composition of the attached microbial communities shifted along the flow path, which may have been attributed to oxygen availability and the toxicity of 6:2 FTS and associated biotransformation products. Genus Pseudomonas dominated in planktonic microbial communities, while in the attached microbial communities, Rhodococcus decreased and Pelotomaculum increased along the flow path, suggesting their potential involvement in early- and late-stage 6:2 FTS biotransformation, respectively. Overall, this study highlights the importance of incorporating realistic environmental conditions into experimental systems to obtain a more representative assessment of in-situ PFAS biotransformation.

Abstract:Fluorinated organic chemicals have a wide variety of industrial and consumer applications. For long time perfluorooctane sulfonate and perfluorooctanoic acid have been used as precursors for manufacture of such chemicals. However, these C 8 chain compounds have been demonstrated to be toxic, persistent, and bioaccumulative, thus inducing their phase-out. Currently, C 6 telomer based fluorocarbon surfactants are considered better alternatives to C 8 products because of their low bioaccumulability. But, their high persistency suggests that in the near future their concentrations will increase in the environment and in industrial waste. Being a solid state non-thermal technology, mechanochemical treatment is a good candidate for the destruction of emerging C 6 fluorotelomers in solid waste. In the present study, 6:2 fluorotelomer sulfonate is effectively destroyed (~100%) in rapid manner (<1 h) by high energy ball milling with KOH. Stoichiometric fluoride formation confirms its entire mineralization, assuring that no toxic by-products are generated. Reaction mechanism and kinetics indicate that effective mineralization of the perfluorinated moiety is obtained thanks to a rapid CF 2 “flake-off” process through radical mechanism.

Abstract:Following the global regulation of legacy PFAS molecules, fluorotelomer molecules have been widely employed as replacements to PFOS in aqueous film-forming foam (AFFF) and PFOA in other products. Recent field studies indicate that fluorotelomer molecules are increasingly identified in environmental settings including groundwater, soil and sediments. Consequently, gaining a comprehensive understanding of the fate and transport of fluorotelomers in soils and sedimentary environments is vital. In this study, the behavior of two different fluorotelomers, 6:2 FTS and 6:2 FTC, in three common soil minerals (kaolinite, montmorillonite and illite) having quite different interfacial properties are reported using molecular dynamics simulations. The interfacial adsorption and dynamical characteristics of 6:2 FTS and 6:2 FTC vary substantially between the three minerals. Irrespective of the mineral composition, 6:2 FTS exhibits surface complexation while 6:2 FTC coordinates only with neutral and low charged clay minerals. In addition, the fundamental interactions that dictate the adsorption, interfacial structure of 6:2 FTS and 6:2 FTC are completely different for the three minerals. The large, aggregated clusters of 6:2 FTS at the surface experienced greater stability for longer periods of time and restricted mobility than 6:2 FTC for all three clay minerals. Importantly, the current study provides cluster size dependent diffusion behavior of surface adsorbed fluorotelomer molecules in each clay mineral. Such detailed mechanistic insights are necessary to understand the environmental footprint of fluorotelomers around contaminated sites.

Abstract:Per‐ and polyfluoroalkyl substances (PFAS) are a class of chemicals present in a wide range of commercial and consumer products due to their water‐repellency, non‐stick, or surfactant properties, resulting from their chemical and thermal stability. This stability, however, often leads to persistence in the environment when they are inevitability released. Here we utilized microbial microcosms from WWTP sludge to determine how employing different functional groups such as heteroatom linkages, varying chain lengths, and hydrofluoroethers (HFEs) will impact the ultimate fate of these novel PFAS structures. A suite of five novel fluorosurfactant building blocks, (F7C3OCHFCF2SCH2CH2OH (FESOH), F3COCHFCF2SCH2CH2OH (MeFESOH), F7C3OCHFCF2OCH2CH2OH (ProFdiEOH), F7C3OCHFCF2CH2OH (ProFEOH), F3COCHFCF2OCH2CH2OH (MeFdiEOH)), and their select transformation products, were incubated in WWTP aerobic microcosms to determine structure‐activity relationships. HFE alcohol congeners with a thioether (FESOH and MeFESOH) were observed to transform faster than the ether congeners, while also producing second generation HFE acid products (F7C3OCHFC(O)OH (2H‐3:2 PFECA), and F3COCHFC(O)OH (2H‐1:2 PFECA). Subsequent biodegradation experiments with 2H‐1:2 PFESA and 2H‐1:2 PFECA, displayed no further transformation over 74 d. Surface water Photofate experiments compared 2H‐1:2 PFECA, and 2H‐1:2 PFESA to their fully fluorinated ether acid counterparts, and demonstrated the potential for both HFE acid species to completely mineralize over extended periods of time, a fate that highlights the value in studying novel PFAS functionalization.

Abstract:The aerobic biological transformation of octylphenoxyacetic acid (OP1EC) and its brominated analog (BrOP1EC) by groundwater enrichment cultures was studied, and persistent metabolites were identified by GC/MS. OP1EC is a representative of the class of alkylphenol ethoxycarboxylates (APEC), formed from alkylphenol polyethoxylate nonionic surfactants during sewage treatment. BrOP1EC is a byproduct formed during chlorine disinfection in the presence of bromide. The metabolite 2,4,4-trimethyl-2-pentanol was detected in stoichiometric quantities in OP1EC-metabolizing enrichment cultures, representing the intact alkyl side chain as a tertiary alcohol. BrOP1EC was transformed by the OP1EC-utilizing cultures only if OP1EC was simultaneously metabolized, suggesting a cometabolic mechanism of transformation. Brominated intermediates were also detected: brominated octylphenol and a compound tentatively identified as 2-aminomethoxy-3-bromo-5-(1,1,3,3-tetra- methylbutyl)phenol.

Abstract:The biodegradation of 2 common fluorotelomer surfactants used in aqueous film forming foams (AFFFs), 6:2 fluorotelomer sulfonamide alkylamine (FTAA) and 6:2 fluorotelomer sulfonamide alkylbetaine (FTAB), was investigated over 109 d with aerobic wastewater-treatment plant (WWTP) sludge. Results show that biodegradation of 6:2 FTAA and 6:2 FTAB produces 6:2 fluorotelomer alcohol (FTOH), 6:2 fluorotelomer carboxylic acid (FTCA), 6:2 fluorotelomer unsaturated carboxylic acid (FTUCA), 5:3 FTCA, and short-chain perfluoroalkyl carboxylates (PFCAs). Additional degradation products included 6:2 fluorotelomer sulfonamide (FTSAm), which was a major degradation product in the presence of either active or sterilized sludge, whereas 6:2 fluorotelomer sulfonate (FTSA) production was measured with sterilized sludge only. Six additional degradation products were tentatively identified by quadrupole time-of-flight mass spectrometry (qTOF-MS) and attributed to N-dealkylation and oxidation of 6:2 FTAA.
