Abstract:Endothelial injury and damage as well as accumulated reactive oxygen species (ROS) in aging play a significant role in the development of cardiovascular disease (CVD). Recent studies show an association of high citrus fruit intake with a lower risk of CVD and stroke but the mechanisms involved are not fully understood. This study investigated the effects of pummelo (Citrus maxima Merr. var. Tubtim Siam, CM) fruit extract on human umbilical vein endothelial cell (HUVECs) migration and aging. The freeze-dried powder of fruit extract was characterized for antioxidant capacity (FRAP assay) and certain natural antioxidants, including ascorbic acid, gallic acid, hesperidin, and naringin (HPLC). Short-term (48 h) co-cultivation of HUVECs with CM enhanced cell migration as evaluated by a scratch wound assay and Boyden chamber assay. A long-term treatment with CM for 35 days significantly increased HUVEC proliferation capability as indicated by population doubling level (PDL). CM also delayed the onset of aging phenotype shown by senescence-associated β-galactosidase (SA-β-gal) staining. Furthermore, CM was able to attenuate increased ROS levels in aged cells when determined by 2',7'-dichlorodihydrofluorescein diacetate (DCDHF) while eNOS mRNA expression was increased but the eNOS protein level was not changed. Thus, further in vivo and clinical studies are warranted to support the use of pummelo as a functional fruit for endothelial health and CVD risk reduction.

Abstract:Diabetes is marked by postprandial hyperglycemia (PHG), an abnormal rise in blood glucose after meals. A key therapeutic goal to reduce PHG is the inhibition of α-amylase (αAM) and α-glucosidase (αGL), enzymes that break down carbohydrates into sugars. Cucurbita moschata has been shown to inhibit both enzymes. However, its inhibition mechanism has not been explored. This study investigated the in vitro inhibition mechanisms of αAM and αGL and conducted a metabolomic analysis of C. moschata (edible part) water-extract (CME), aiming to preliminarily identify its bioactive compounds (BCs). The inhibitory mechanisms were determined using Lineweaver-Burk plots. The BCs were identified and quantified using HPLC-QTOF-MS, employing both targeted and untargeted metabolomic approaches. CME had a significant higher effect (p < 0.05) on αAM activity than against αGL with IC50 of 28.99 and 698.42 mg/mL, respectively. The extract showed mixed and uncompetitive type inhibitions on αAM and αGL, respectively. The lowest inhibition constant (Ki) was 47.68 mg/mL on αAM activity at 20 mg/mL. Untargeted metabolic profiling by UPLC-MS-ESI-QTOF putatively identified 30 compounds in CME, such as amino acids, vitamins, phytohormones, fatty acids, cucurbitacins and phenolic acids, and flavonoids. Functional analysis of CME identified significant pathways, including pantothenate and CoA biosynthesis and phenylpropanoids, among others. The targeted analysis by UPLC-MS-ESI-QqQ allowed us to identify 12 compounds, with l-phenylalanine, p-hydroxybenzoic, and p-coumaric acid as majors. This study demonstrated the inhibitory potential of CME on αAM and αGL activities, which may be attributed to its metabolites. Thus, this plant represents a valuable source of BC against PHG. Practical Application: The research highlights that Cucurbita moschata has significant potential in managing postprandial hyperglycemia in diabetic patients by inhibiting enzymes like α-amylase and α-glucosidase. In addition, the identification of its compounds emphasizes its importance as a source of bioactive compounds. Therefore, C. moschata could be effectively utilized in the development of nutraceuticals or as an ingredient in functional foods specifically designed for postprandial hyperglycemia management. Thus, integrating C. moschata as part of the daily diet could offer patients with diabetes a natural alternative to control their blood glucose levels after eating.

Abstract:Occult hydrocephalus has received increasing attention during the last few years due to the reversibility of the serious, associated, symptoms, which include dementia and apraxia of gait. Abstract:This study aimed to extract polysaccharides from pumpkin, characterize the structures of four of them, and evaluate their in vitro antioxidant and hypoglycemic activities. Additionally, an animal model of type 2 diabetes mellitus (T2DM) was established and used to determine their hypoglycemic and hypolipidemic effects in vivo , and the underlying mechanisms related to the regulation of gut microbiota. Water-extracted crude pumpkin polysaccharides (W-CPPs), water extraction and alcohol precipitation crude pumpkin polysaccharides (WA-CPPs), deproteinized pumpkin polysaccharides (DPPs), and refined pumpkin polysaccharides (RPPs) were sequentially extracted and purified from pumpkin powder by hot water extraction, water extraction, and alcohol precipitation, deproteinization and DEAE-52 cellulose gel column, respectively. The extraction and purification methods had significant influence on the extraction yield, physicochemical properties, and in vitro antioxidant and hypoglycemic activities. W-CCP and RPPs had a significant positive free radical-scavenging capacities and inhibitory activities on α-glucosidase and α-amylase. RPP-3 not only inhibited the uptake of glucose in Caco-2 monolayer but also promoted the excretion of glucose, while RPP-2 had no inhibitory effect. Animal experiment results showed that W-CPP treatment significantly improved the T2DM symptoms in mice, which included lowering of fasting blood glucose (FBG), reducing insulin resistance (IR), and lowering of blood lipid levels. It increased the diversity of intestinal flora and reduced the harmful flora of model mice, which included Clostridium, Thermoanaerobe, Symbiotic bacteria, Deinococcus, Vibrio haematococcus, Proteus gamma , and Corio . At the family level, W-CPP (1,200 mg/kg) treatment significantly reduced the abundance of Erysipelotrichaceae , and the Akkermanaceae of Verrucobacterium became a biomarker. Pumpkin polysaccharides reshaped the intestinal flora by reducing Erysipelotrichaceae and increasing Akkermansia abundance, thereby improving blood glucose and lipid metabolism in the T2DM mice. Our results suggest that W-CCP and RPP-3 possess strong antioxidant and hypoglycemic activities, and are potential candidates for food additives or natural medicines.

Abstract:Biologically active phytochemicals from pumpkin reveal versatile medical applications, though little is known about their antiviral activity. The fucose-rich polysaccharide extraction conditions were optimized through Box-Behnken design and purified by column chromatography. The purified fucose-rich polysaccharide was characterized through SEM, FT-IR, 1H NMR, XRD, TGA, and GS-MS. The analysis results revealed an irregular and porous surface of the purified polysaccharide with high fucose, rhamnose, galactose, and glucose contents. The tested fucose-rich polysaccharides revealed significant antioxidant and anti-inflammatory activity at very low concentrations. The purified fucose-rich polysaccharides exerted a broad-spectrum antiviral activity against both DNA and RNA viruses, accompanied by high safety toward normal cells, where the maximum safe doses (EC100) were estimated to be about 3-3.9 mg/mL for both Vero and PBMC cell lines. Treatment of HCV, ADV7, HSV1, and HIV viruses with the purified polysaccharides showed a potent dose-dependent inhibitory activity with IC50 values of 95.475, 20.96, 5.213, and 461.75 μg/mL, respectively. This activity was hypothesized to be through inhibiting the viral entry in HCV infection and inhibiting the reverse transcriptase activity in HIV. The current study firstly reported the antioxidant, anti-inflammatory, and antiviral activities of Cucurbita maxima fucose-rich polysaccharide against several viral infections.

Abstract:Coprinus comatus (O.F. Mull.) Pers. is a medicinal and edible mushroom containing bioactive compounds known as antidiabetic and antioxidant agents. The pancreatic β-cells were sensitive to reactive oxygen species (ROS) attacks, which can cause necrosis and disrupt insulin production. Therefore, this research aimed to evaluate the C. comatus ethyl acetate extract for antidiabetic and antioxidant to decrease fasting blood glucose, dipeptidyl peptidase-4 enzyme (DPP-4) levels, and increase glutathione peroxidase (GPx). Subsequently, 24 male Wistar rats were divided into six groups, namely 2-5 streptozotocin (STZ)-induced 45 mg/kg, 1/HC (without any treatment), 2/NC (negative control), 3/PC (administered metformin 45 mg/kg Body weight (BW), 4-6 (T1: administered 250 mg, T2: administered 500 mg, and T3: administered 750 mg/kg BW extract). The parameters were analyzed using ANOVA, followed by Duncan’s multiple range test. According to the results, the extract significantly decreased DPP-4 levels, blood glucose and increased GPx (p ≤ 0.05). The 500 mg extract effectively reduced blood glucose levels and DPP-4 enzyme as well as increased levels of GPx.
Phenolic content of Thai Bao mango peel and its in-vitro antioxidant, anti-cholinesterase, and antidiabetic activities

Abstract:Plant phenolics have been known for various biological activities. This study aims to extract and examine the presence of phenolics in Bao mango (Mangifera indica L. var.) peel ethanolic extract (MPE). Further, antioxidant, anti-diabetic (α-amylase, and α-glucosidase inhibitory activity), and anti- Alzheimer's disease (AD) (acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and β-secretase (BACE-1) inhibitory activity) efficacy of MPE were determined. The results indicated that mangiferin (8755.89 mg/ 100 g extract) was the major phenolic compound in MPE. An antioxidant mechanism revealed that MPE had a higher radical scavenging ability (4266.70 μmol TE/g extract) compared to reducing power (FRAP) or oxygen radical absorption capacity (ORAC). Further in-vitro enzyme inhibitory assay against diabetic and AD involved enzymes showed that MPE had stronger inhibitory action against an enzyme involved in diabetes compared to their standard drug (Acarbose) (P < 0.05). While a lower IC 50 value was observed against AD-involved enzymes compared to their standard drug (donepezil) (P < 0.05). The results show that Thai Bao mango peel byproduct can be a potential source of nutraceuticals to lower diabetes and improve cognitive health.

Abstract:This study evaluates the proximate and phytochemical composition and antioxidant, anti-diabetic, and anti-inflammatory properties of Capsicum annum L. seed powder (CSP) and pericarp powder (CPP). The CSP and CPP were analyzed for moisture, crude protein, ash, crude fat, crude fiber, nitrogen-free extract, flavonoids, phenol, tannins, alkaloids, saponins, 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonic) acid (ABTS), ferric ion reducing antioxidant power (FRAP), alpha-amylase, alpha-glucosidase, lipase and albumin inhibitory properties and anti-proteinase properties. The moisture, crude protein, ash and crude fat concentrations were significantly higher in CPP compared to CSP; while the concentrations of crude fiber and nitrogen-free extracts were significantly lower in CSP than CPP. The phenol, alkaloids and saponins concentrations of the CPP were significantly higher than CSP; while the flavonoids and steroid concentrations were higher in CPP compared to CSP. The vitamin C, DPPH, ABTS, FRAP and lipid peroxidation inhibition of CPP was significantly higher than CSP. The alpha-amylase inhibition and alpha-glucosidase inhibition of CPP were significantly higher than in CSP. The lipase inhibitory and anti-proteinase properties of CSP were higher than CPP; while the albumin inhibitory property was higher in CPP than in CSP. In conclusion, CPP offers better protein, minerals, and fat content compared to CSP with stronger antioxidant, anti-cholesterol, and anti-diabetic properties.

Abstract:Cortex Moutan (CM, root bark of Paeonia suffruticosa Andr.) is one of the common herbs found in anti-diabetic traditional Chinese medicine formulae. To study the potential anti-diabetic mechanisms of CM, four in vitro models (intestinal brush border membrane vesicles (BBMV), rat hepatoma cell line H4IIE, human skin fibroblasts cell line Hs68 and mouse adipocytes 3T3-L1) were used. CM showed significant in vitro anti-diabetic effects by inhibiting glucose uptake of BBMV and enhancing glucose uptake into Hs68 and 3T3-L1 cells. Using bioassay-guided fractionation, paeonol was confirmed to be one of the active constituents for inhibiting BBMV glucose uptake. With neonatal-streptozotocin diabetic rats, paeonol (200 and 400mg/kgbody wt.) was found to improve oral glucose tolerance in vivo. To the best of our knowledge, this is the first report on the anti-diabetic effect of paeonol.

Abstract:Diabetes is a metabolic disease characterised by decreased insulin signalling and persistent hyperglycaemia. The most frequent progression of diabetes is known as diabetes mellitus which is defined as persistent hyperglycaemia brought on by either peripheral insulin resistance or impaired pancreatic β cell production of insulin. This condition is caused by the disruptions in the digestion of carbohydrates, fats and proteins which are caused by defects in the production, release and regulation of insulin. The breakdown of carbohydrates is significantly aided by the intestinal digestive enzymes α-glucosidase and α-amylase. One type of antidiabetic treatment is to lower the blood glucose levels after a meal by blocking the enzymes α-glucosidase and α-amylase. This could be a key tactic in blood glucose control. When compared to other commercial pharmaceuticals used to treat diabetes, herbal treatments are thought to be more in harmony with the human body and to have less harmful side effects. This herbal remedy is reasonably priced as well. The aim of the current study is to compare the anti- diabetic activity of the methanolic extracts of Momordica charantia and Psidium guajava. The methods used in the determination of invitro antidiabetic activity are alpha amylase and alpha glucosidase inhibitory assays. In the current study, when compared the individual inhibition activity of Momordica charantia and Psidium guajava on the intestinal enzymes (alpha amylase and alpha glucosidase), Momordica charantia has the highest inhibition that is 79.83% at 0.8 mL concentration while the Psidium guajava has shown only 70.58% inhibition at 0.8 mL concentration. Which suggests that the presence of bio active compounds may be responsible for the plants many medicinal uses, including the treatment of diabetes.

Antioxidant Activities and Properties of Coprinus comatus Mushroom Both Mycelium and Fruiting Body Extracts In Streptozotocin-Induced Hyperglycemic Rats Model

Abstract:Coprinus comatus, well-known as Shaggy Ink Cap mushroom, is potential herbal medicine. Synthetic medicines sometimes cause side effects; therefore, it is necessary to innovate with herbal medications with minimal side effects. The study evaluated in vitro and in vivo treatments to evaluate the antioxidant effect and activity of C. comatus. The in vivo treatment was conducted using six groups of Wistar rats (n = 24). Group 1 healthy control (HC), groups 2–6 received 45 mg/Kg BW of streptozotocin once, group 2 just streptozotocin-induced (NC), group 3 was given 45 mg/kg BW of metformin (PC), groups 4–6 were given 250 (T1), 500 (T2), and 750 mg (T3) of C. comatus extract for 14 days, and the in vitro was conducted using an antioxidant oxidant assay. Data were analyzed using analysis of variance and Duncan's multiple range tests. Based on qualitative analysis, C. comatus mycelium extract contained polyphenol, flavonoids, terpenoids, and fruiting body extract had flavonoids, alkaloids, and saponins. The in vitro analysis showed that the mycelium extract had an antioxidant activity by inhibiting free radicals up to 58.51% with an IC50 value of 72.77 mg/L. The in vivo treatment using C. comatus fruiting body extract showed that it could increase the endogenous antioxidant levels of GPx, SOD, catalase and reduce MDA levels (p 0.05). The most effective dose of C. comatus extract is 500 mg. This research has shown the potential of mycelium and C. comatus fruitbody extract as an antioxidant supplement in a diabetic rat model.

Abstract:In traditional medicine, Plagiorhegma dubia Maxim (PD) is used to treat diabetes, irritability, insomnia, high fever, and coma. In this study, to identify the components in PD, we first performed three preliminary phytochemical tests, which were all positive for the presence of alkaloids. Next, we evaluated the total alkaloid content and performed 13 different in vitro antioxidant experiments for three solvent extracts of PD. According to the results, the methanol extract was selected for further research. Ultra-performance liquid chromatography with electrospray ionization Orbitrap mass spectrometry was used to identify 41 compounds in the methanol extract. Thirty-eight of these compounds were alkaloids, and the main components were magnoflorine, berberrubine, and betaine. Addition of the methanol extract to cooking oil increased the stability against oxidation and greatly reduced the peroxide value. Cell viability and oral acute toxicity studies in mice indicated that the methanol extract was relatively non-toxic. Histopathological examination and biochemical analysis showed that the methanol extract was protective against liver damage in rats induced by d-galactosamine. The methanol extract also showed a good antidiabetic effect for streptozotocin-induced diabetes in rats, where it lowered blood sugar levels and protected vital organs against damage. Lipid profile parameters and oxidative stress parameters also improved. The therapeutic effects towards diabetes and liver damage were attributed to the ability of the three main components in the methanol extract to resist oxidative stress. Our results show that the alkaloid-rich methanol extract of PD is potential candidate for treatment of diseases associated with oxidative imbalances. This understanding of the hepatoprotective effect and antidiabetic activity of PD provides new insight for its use as a substitute for Coptis chinensis and establishes a foundation for development of PD as an economic crop.

Abstract:Cortex Mori Radicis (CMR) is enriched in various phenolics, this study aimed to estimate the antioxidant effect, enzyme (lipase, α-amylase, α-glucosidase and acetyl-cholinesterase) inhibition and the anti-proliferative effect of the phenolic compounds in CMR. However, the amount of these compounds obtained from CMR is highly dependent on the processing conditions. In this study, the processing parameters of extracting the phenolics from CMR using ultrasonic technique pooled with high-speed shearing extraction (UTPHSE) were optimized. Subsequently, the phenolics from Cortex Mori Radicis (PCMR) were purified using AB-8 macroporous resin, and their chemical analysis, antioxidant, enzyme inhibition, and antiproliferative activities were studied. Based on our findings, the optimal parameters of UTPHSE were: L/S ratio 25.8:1 (mL/g), voltage 81.0 V, ultrasonic temperature 51.8 ℃ and ultrasonic time 289 s. And under the optimal extraction conditions, the extraction rate of the PCMR was 0.531 ± 0.004%. Compared with PCMR, the contents of “total phenolics, flavonoids, flavonols, flavanols and phenolic acids” increased 2.30, 2.67, 2.59, 3.63 and 2.72 times in the purified phenolics from Cortex Mori Radicis (PPCMR), respectively. In addition, PPCMR depicted significant DPPH, ABTS+ and superoxide anion radicals’ scavenging capability, reducing power, ferric ion reducing antioxidant power (FRAP) and remarkable inhibitory activities on “lipase, alpha-amylase, alpha-glucosidase, and the proliferation of HeLa, HepG2 and NCI-H460”. At the same time, the morphological changes of HeLa, HepG2 and NCI-H460 cells suggested that PPCMR could effectively inhibit the proliferation of tumor cells in vitro. Therefore, PPCMR have good potential as natural antioxidants, antilipidemic, hypoglycemic, and antineoplastic agents in functional foods and pharmaceuticals.

Potential Anti-Diabetic Benefits of Momordica charantia Extracts on Experimental Diabetes and its Complications

Abstract:Background and Objective: The Momordica charantia L. (Cucurbitaceae) plant is a natural medicine that is commonly used in the treatment of a wide range of ailments, including diabetes. The current study validates two common types of extracts (methanolic and water) of Momordica charantia (MC) for antidiabetic effects and compares their efficacy in preventing experimental diabetic complications. Materials and Methods: The rats with a fasting serum glucose level above 200 mg dL–1 at 96 hrs after streptozotocin (STZ) injection were considered diabetic and included in the study. They were administered with any of the three doses (45, 90 or 180 mg kg–1) of methanolic or aqueous extracts of MC based on their respective experimental groups. After 14 and 28 days, parameters such as body weight, serum glucose, lipid profile, hemoglobin content and antioxidant status were recorded. The observations were statistically examined using a One-way ANOVA and p<0.05 indicated significance. Results: According to the research findings, rats with STZ-induced diabetes had a significant (p<0.05) variance in their body weight (177±5.64), serum glucose levels (216.4±3.96), lipid profiles [cholesterol (107.38±3.70), triglycerides (168.68±14.70)], hemoglobin levels (7.5±0.14) and antioxidant status. These effects were found to become more prominent after 14 days of exposure. By providing either methanolic or water extracts of MC at different doses, the STZ-induced alterations in body weight and hyperglycemia were significantly (p<0.05) reversed. Besides, the extracts attenuated the hyperlipidemic, depleted antioxidant stress and improved hemoglobin levels in diabetic rats up to 28 days. Comparing the two extracts, the methanolic extract has shown greater efficacy than the water extract and the effects matches with that of glibenclamide. Conclusion: The findings of the current investigation verify the anti-diabetic potential of the MC and show its ability to serve as an alternate therapy for managing hyperglycemia-induced complications. Besides, attenuation in oxidative stress, diminished hemoglobin level, weight loss and hyperlipidemia, both extracts might benefit diabetic conditions by minimizing the complications related to hyperglycemia.

Abstract:BACKGROUND: We investigated anti-diabetic, hypolipedimic and antioxidant activity of hydroalcoholic extract from leaves and fruit peel of Punica granatum.MATERIALS AND METHODS: Streptozotocin induced diabetic Wister rats were used in this study consisting of seven groups of six animals each. Groups (1) normal control, (2) diabetic control, (3) leaves extract 100 mg/kg b.w. of P. granatum, (4) leaves extract 200 mg/kg b.w. of P. granatum, (5) fruit peel extract 100 mg/kg b.w. of P. granatum, (6) peel extract 200 mg/kg b.w. of P. granatum and (7) glibenclamide respectively. Fasting blood sugar was recorded on 1(st), 7(th), 14(th), 21(st) and 28(th) day. At the end of the experiment Lipid profile and levels of antioxidants were determined. Safety profile of both extracts was evaluated using acute and chronic toxicity studies.RESULTS: Higher dose of fruit peel extract of P. granatum (PEPG) and glibenclamide significantly lowered blood glucose level from 7(th) day onwards however glibenclamide was found to be more effective. Leaves extract at higher dose and fruit extract at lower dose also significantly lowered blood glucose level from 14(th) day onwards. Leaves extract at lower dose also significantly lowered blood glucose level from 21(st) day onwards. Glibenclamide and higher dose of fruit PEPG extract significantly reduced the total cholesterol, triglyceride levels and significantly increased the high density lipoprotein cholesterol level. Glibenclamide followed by higher dose was found more effective in reducing plasma thiobarbituric acid reactive substances and increasing levels of antioxidant enzymes (superoxide dismutase and catalase). No toxicity was observed even when both extracts were administered at 10 times of higher dose used in this study and no significant changes were seen when it were used chronically.CONCLUSION: Leaves and fruit PEPG possesses significant anti-diabetic, hypolipedimic and antioxidant properties. This study supports the traditional use of P. granatum in diabetes. Fruit peel which is normally thrown by many while eating pomegranate fruit is having anti-diabetic, hypolipedimic and Antioxidant activity. Furthermore high therapeutic index is safe for chronic use.

Abstract:Background: Diabetes Mellitus (DM) is a major independent risk factor for cardiovascular disease (CVD) that oxidative stress plays key role for diabetescomplication vascular. Antioxidant agent is needed to attenuate the oxidative stress. Objective: This study aimed to evaluate polysaccharide peptide (PsP) of the extract of Ganoderma lucidum as an antioxidant that attenuate oxidative stress toinhibits the process of atherogenesis in rats model of diabetes. Methods: The study was experimental study design. Thirty Wistar rats (Rattus novergicus) were divided into 5 groups: group treated with High Fat Diet (HFD) and Streptozotocin injection subcutan given PsP dose 50, 150 and 300 mg/KgBW and group without PsP, normal rats as a control group. Parameters measured were the levels of MDA, SOD and H2O2. Result: After 5 weeks of treatment with PsP, level of MDA and H2O2in rats model of diabetes waslower significantly (p = 0.006), (p = 0.003) respectivelyand PsP can significantly enhance the level of SOD (p = 0.000). Conclusion: Based on this study, PsP can be usefull alternativeas antioxidant that attenuate oxidative stress to inhibit the process of atherogenesis in a mouse model of diabetes induced by HFD and streptozotocin.
