Abstract:The current work aims to optimize and develop a scale-up process design for higher yields of phenolic compounds, as well as propose and validate their antidiabetic and cytotoxic effects when extracted from Cajanus cajan leaves. The optimization procedure included 17 experimental runs using microwave-assisted extraction parameters such as temperature (50–70 °C), power (400–1000 W), and time (15–45 min), and was optimized using response surface methodology for total phenolic content (TPC), total flavonoid content (TFC), total tannin content (TTC), and antioxidant activities like 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) was found to be 52.15 mg GAE/g dw, 25.12 mg QE/g dw, 43.02 mg TAE/g dw, 35.16%, and 43.25 mg TE/g dw, respectively. The optimized values of response variables were achieved at 700 W of power, 60 °C of temperature, and 40 min of processing time. Purification, thin-layer chromatography, fourier transform infrared (FTIR), and liquid chromatography-mass spectrometry (LC-MS) spectral analysis of the plant extract revealed the existence of 18 bioactive phenolic compounds. The in silico investigation found that fifteen out of eighteen bioactive phenolic compounds had strong binding affinities in the range of −4.0 to −8.8 kcal/mol, along with enhanced pharmacokinetic characteristics and a low to moderate level of toxicity. The IC 50 values of in vitro α-amylase and α-glucosidase inhibitory activity were 46.49 ± 2.2 and 59.51 ± 3.57 μg/mL, respectively, indicating a high level of enzymatic inhibition activity. In addition, the 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) and chorioallantoic membrane (CAM) tests revealed that cytotoxicity was minimal up to a concentration of 62.5 μg/mL of the extract. The results of this study will open new doors for researchers, scientists, and industrialists in the food and pharmaceutical industries who intend to develop antidiabetic food formulations and antidiabetic drugs as a way for diabetic people to manage their disease and improve their overall health.

Abstract:Unhealthy diets and lifestyles cause a resistance to and/or relative deficiency of insulin production. Therefore, it is thought worthwhile to develop a natural remedy that may effectively manage the disease symptoms to a certain extent without causing adverse consequences. The objective was to develop the active biological constituent(s) for the use of Coccinia indica and its relationship to treating type II diabetic rats. The soxhlet extraction method was used to get the cocktail of phytochemicals of C. indica by using methanol. The composition of a high-sugar diet, followed by fructose (66%), was used to induce T2DM in rats. The preliminary predictive markers were body weight (pre- and post-treatment), blood glucose level (pre- and post-treatment), serum insulin, and pancreatic insulin. And the secondary outcomes were the pro-inflammatory mediators interleukin 6 (IL-6), transforming growth factor-β (TGF-β), and tumour necrosis factor alpha (TNF alpha). Additionally, pancreatic tissue was used to estimate beta cell mass, size, and necrosis, and the cell supernatant was used to observe superoxide dismutase (SOD), lipid peroxidation (LPO), and catalase (CAT). High sugar diet showed significant increase in body weight (p < 0.01), fasting blood glucose level (p < 0.001), and decrease in serum and pancreatic insulin levels (p < 0.001), whereas rats treated with methanolic extract of C. indica showed significant reduction in post-treatment body weight (p < 0.01), blood glucose levels (p < 0.01), and increase in serum and pancreatic insulin (p < 0.001), especially in higher doses, i.e., 400 mg/kg. Pro-inflammatory cytokines (IL-6, TGF-beta, and TNF-α) can increase insulin resistance, which results in poor glucose homeostasis, which has been reduced by treatment with C. indica (p<0.001). Superoxide radicals and a deficiency in catalase, both of which are linked to diabetes, but the extract of the plant has been shown to enhance the secretion of enzymes SOD and CAT (p<0.001). It has been proven to have a crucial role in the regulation of apoptosis because it lowers oxidative stress and similarly reduces the level of LPO (p<0.01). Additionally, the treated rat pancreas shows islets of Langerhans that are normal in number and size. No necrosis or reduction in size was seen. The current study conclusively shows that the phytoconstituents of C. indica have the potential to tackle long-term health complications and manage symptoms.

Abstract:Diabetes Mellitus (DM) can appear due to the absence of insulin (DM1- type 1) or poor response of cells to insulin (DM2-type 2). Even though DM1 cannot be controlled using general treatments, DM2 can be easily controlled or prevented using pharmaceuticals, nutraceuticals, or dietary practices. Ceylon cinnamon (Cinnamomum zeylanicum) is one such natural remedy that has been consumed against elevated blood glucose levels in the past. Cinnamon and different types of cinnamon extracts have been scientifically tested for their activities on the inhibition of α-amylase and α-glucosidase enzymes that are responsible for carbohydrate metabolism and are effective in blood glucose regulation. However, the combined effect of aqueous and ethanol extracts of cinnamon bark on blood glucose regulation is still lacking. In this study, Water Extract of Cinnamon (CWE), Ethanol Extracts of Cinnamon (hot ethanol extract of cinnamon-CHEE, cold ethanol extract of cinnamon-CEE, and 50% ethanol extract of cinnamon- CEE-50) were studied for their sugar-controlling properties. This study was performed to identify the efficacy of different cinnamon extracts on the inhibition of α-amylase and α-glucosidase enzymes, followed by animal studies to confirm the use of the extracts in nutraceutical formulations. To study alpha-amylase and alpha-glucosidase inhibition properties, cinnamon extracts. To study post-prandial glucose regulation using the properties of cinnamon extracts. To evaluate the possibility of incorporating cinnamon extracts in nutraceutical applications. Water and ethanol-based extraction method was used to prepare cinnamon extracts. These extracts have been scientifically tested for their activities on the inhibition of α-amylase and α-glucosidase enzymes. Molecular docking studies were used to identify the binding of the active molecules to the substrate binding sites of α-amylase and α-glucosidase. In-vivo time dependence postprandial blood glucose regulation studies have been performed with healthy Wistar male rats. Yields of the CHEE, CEE, and CWE were 14±2%, 12±2%, and 8±1% respectively. According to the LCMS data, the major component in the CEE was cinnamaldehyde. Both CWE and CEE were subjected to the Total Polyphenol assay (TPC) and Total Flavonoids (TFC) assays. The TPC of CWE and CEE were 117±1 mg (Gal)/g and 170±10 mg (Gal)/g, while the TFC of CWE and CEE were 359±1 mg (Qc)/g and 254±4 mg (Qc)/g, respectively. In the α-amylase inhibition assay, Acarbose; a known α-amylase inhibitor, and CEE showed IC50 values of 65.4 ppm and 2.6 ppm, while CWE failed to show inhibition against α-amylase. In the α-glucosidase inhibition assay, Acarbose; a known α-amylase inhibitor, CEE, and CWE showed IC50 values of 312 ppm, 4.5 ppm, and 1.3 ppm, respectively. In-vivo time dependence postprandial blood glucose regulation studies that have been performed with healthy Wistar male rats showed a lowering of blood glucose concentrations by 22%, 11%, and 10% of glucose at 30 min, 60 min, and 90 min compared to the control group. The CEE contains polyphenols and flavonoids and is effective in inhibiting both α- amylase and α-glucosidase. The CWE also contains polyphenols and a comparatively higher level of flavonoids and is effective in inhibiting α-glucosidase while not affecting α-amylase inhibition. Overall, the IC50 data, TPC data, and TFC data proposed that the inhibition of carbohydrate hydrolyzing enzymes by polyphenols may depend on the polarity of particular polyphenols. Based on the rat trials, it can be concluded that the 1:1 combination of CWE and CEE may be useful in formulating postprandial blood glucose level-regulating nutraceuticals.

Abstract:Diabetes is a prevalent metabolic disorder associated with various complications. Inhibition of α‐glucosidase and α‐amylase enzymes is an effective strategy for managing non‐insulin‐dependent diabetes mellitus. This study aimed to investigate the antioxidant and antidiabetic potential of Ormocarpum cochinchinense leaf through in vitro and in silico approaches. The methanol extract exhibited the highest phenolic and flavonoid content over solvent extracts aqueous, acetone, hexane and chloroform, same has been correlating with strong antioxidant activity. Furthermore, the methanol extract demonstrated significant inhibitory effects on α‐amylase and α‐glucosidase enzymes, indicating its potential as an antidiabetic agent. Molecular docking analysis identified compounds, including myo‐inositol, with favorable binding energies comparable to the standard drug metformin. The selected compounds displayed strong binding affinity towards α‐amylase and α‐glucosidase enzymes. Structural dynamics analysis revealed that myo‐inositol formed a more stable complex with the enzymes. These findings suggest that O. cochinchinense leaf possesses antioxidant and antidiabetic properties, making it a potential source for developing therapeutic agents.

Abstract:Curcuma angustifolia Roxb. is a plant with medicinal potential, traditionally used to treat different diseases. The present study was aimed to determine the antidiabetic activity of C. angustifolia rhizome in vitro and in silico. The methanolic extract of C. angustifolia rhizome was analysed by FTIR and GC‐MS to determine the phytochemicals present. The antidiabetic potential of the extract was evaluated by different assays in vitro. The extract inhibited both α‐amylase and α‐glucosidase enzymes and the glucose diffusion through the dialysis membrane in a concentration‐dependent manner with IC50 values of 530.39±0.09, 293.75±0.11, and 551.74±0.3 μg/ml respectively. The methanolic extract also improved yeast cell's ability to take up glucose across plasma membranes and the adsorption of glucose. The findings were supported by molecular docking studies. The results showed that the methanol extract of C. angustifolia rhizome has significant antidiabetic activity and thus can be also studied to isolate the potential compound with antidiabetic activities.

Abstract:A -Amylase and α -glucosidase inhibition is a hopeful curative target against type II diabetes as it can downgrade fierce digestion and absorption of carbohydrates into absorbable monosaccharides. Medicinal plants have been used as blood sugar reducers; on the other hand, the inhibitory action of phytocompounds found in these plants on α -amylase is yet limited. Palmarosa essential oil (PRO) from Cymbopogon martinii is complementary and conventional medicine worldwide. The aim of the study presented is compositional profiling of PRO and in silico docking of main bioactive compounds: geraniol, geranial, linalool, fenchyl alcohol, 6-methyl-hept-5-en-2-one, borneol, elemol, δ -cadinol against alpha-amylase and alpha-glucosidase. GC-FID was used to find out bioactive components in PRO. For docking, Cb-dock2 tool was utilized. Ligand–protein 2-D interactions were also studied. In silico ADMET pharmacoinformatics viewpoint along with PASS prediction of all the ligands has prospected from the point of view of human health. Wet laboratory validation was performed by using Aspergillus oryzae -derived α -amylase and Saccharomyces cerevisiae -derived α -glucosidase. GC-FID exposed presence of 13 bioactive components. Docking investigation demonstrated the effective binding of all the ligands with the enzymes. The interaction results imply that the enzyme-ligand complexes form hydrogen, hydrophobic, and other interactions. Furthermore, molecular dynamics (MD) simulations were also carried out for the best-docked complexes from docking studies ( α -amylase–elemol, α -glucosidase–elemol, and α -glucosidase– δ -cadinol) to clarify some information on their thermodynamic and dynamic properties and confirm the docking results as well. In silico ADMET examination disclosed that all the ligand molecules have no toxic effect and acceptable absorption as well. PRO retains inhibition in dose-dependent manner against α -amylase (IC50 = 194.78 μg/ml) and α -glucosidase (IC50 = 162.36 μg/ml). A kinetic study showed that PRO inhibited the α -amylase and α -glucosidase competitively. It could be inferred that the phytochemicals present in PRO might be responsible in α -amylase and α -glucosidase inhibition and thus have potential to be produced as an antidiabetic drug.

Identification of antidiabetic inhibitors from Allophylus villosus and Mycetia sinensis by targeting α-glucosidase and PPAR-γ: In-vitro, in-vivo, and computational evidence

Abstract:Diabetes mellitus (DM) is a metabolic disorder arising from insulin deficiency and defectiveness of the insulin receptor functioning on transcription factor where the body loses control to regulate glucose metabolism in β-cells, pancreatic and liver tissues to homeostat glucose level. Mainstream medicines used for DM are incapable of restoring normal glucose homeostasis and have side effects where medicinal plant-derived medicine administrations have been claimed to cure diabetes or at least alleviate the significant symptoms and progression of the disease by the traditional practitioners. This study focused on screening phytocompounds and their pharmacological effects on anti-hyperglycemia on Swiss Albino mice of n-hexane, ethyl acetate, and ethanol extract of both plants Mycetia sinensis and Allophylus villosus as well as the in-silico investigations. Qualitative screening of phytochemicals and total phenolic and flavonoid content estimation were performed significantly in vitro analysis. FTIR and GC-MS analysis précised the functional groups and phytochemical investigations where FTIR scanned 14, 23 & 17 peaks in n-hexane, ethyl acetate, and ethanol extracts of Mycetia sinensis whereas the n-hexane, ethyl acetate, and ethanol extracts of Allophylus villosus scanned 11 peaks, 18 peaks, and 29 peaks, respectively. In GC-MS, 24 chemicals were identified in Mycetia sinensis extracts, whereas 19 were identified in Allophylus villosus extracts. Moreover, both plants' ethyl acetate and ethanol fractioned extracts were reported significantly (p < 0.05) with concentrations of 250 mg and 500 mg on mice for oral glucose tolerance test, serum creatinine test and serum alkaline phosphatase test. In In silico study, a molecular docking study was done on these 43 phytocompounds identified from Mycetia sinensis and Allophylus villosus to identify their binding affinity to the target Alpha Glucosidase (AG) and Peroxisome proliferator-activated receptor gamma protein (PPARG). Therefore, ADMET (absorption, distribution, metabolism, excretion, and toxicity) analysis, quantum mechanics-based DFT (density-functional theory), and molecular dynamics simulation were done to assess the effectiveness of the selected phytocompounds. According to the results, phytocompounds such as 2,4-Dit-butyl phenyl 5-hydroxypentanoate and Diazo acetic acid (1S,2S,5R)-2-isopropyl-5-methylcyclohexyl obtained from Mycetia sinensis and Allophylus villosus extract possess excellent antidiabetic activities.
Interleukin 2 immunotherapy in children with neuroblastoma after high-dose chemotherapy and autologous bone marrow transplantation.

Abstract:Four children with persistent neuroblastoma after marrow ablative chemoradiotherapy and autologous bone marrow transplantation received continuous infusion of recombinant interleukin 2, 75 to 120 days after the graft. Recombinant interleukin 2 therapy did not induce any major or nonreversible toxicity, hematological toxicity in particular. One patient entered complete remission for 9 months and a second patient had a long-lasting normalization of urinary catecholamine metabolites with more than 50% regression of bone marrow metastases (8 months). In three children, recombinant interleukin 2 and a second patient entered complete remission for 9 months therapy was followed by major increase and activation of circulating natural killer cells which amounted to 80% of the circulating mononuclear cells.
Computational study and in vitro alpha-glucosidase inhibitory effects of medicinal plants from a Thai folk remedy

Abstract:The number of patients with type 2 diabetes mellitus (T2DM) has increased worldwide. Although an instant cure was achieved with the standard treatment acabose, unsatisfactory symptoms associated with cardiovascular disease after acabose administration have been reported. Therefore, it is important to explore new treatments. A Thai folk recipe has long been used for T2DM treatment, and it effectively decreases blood glucose. However, the mechanism of this recipe has never been proven. Therefore, the potential anti-T2DM effect of this recipe, which is used in Thai hospitals, was determined to involve alpha-glucosidase (AG) inhibition with a half maximal inhibitory concentration (IC50). In vitro experiments showed that crude Cinnamomum verum extract (IC50 = 0.35 ± 0.12 mg/mL) offered excellent inhibitory activity, followed by extracts from Tinospora crispa (IC50 = 0.69 ± 0.39 mg/mL), Stephania suberosa (IC50 = 1.50 ± 0.17 mg/mL), Andrographis paniculate (IC50 = 1.78 ± 0.35 mg/mL), and Thunbergia laurifolia (IC50 = 4.66 ± 0.27 mg/mL). However, the potencies of these extracts were lower than that of acabose (IC50 = 0.55 ± 0.11 mg/mL). Therefore, this study investigated and developed a formulation of this recipe using computational docking. Among 61 compounds, 7 effectively inhibited AG, including chlorogenic acid (IC50 = 819.07 pM) through 5 hydrogen bonds (HBs) and 2 hydrophobic interactions (HIs); β-sitosterol (IC50 = 4.46 nM, 6 HIs); ergosterol peroxide (IC50 = 4.18 nM, 6 HIs); borapetoside D (IC50 = 508.63 pM, 7 HBs and 2 HIs); borapetoside A (IC50 = 1.09 nM, 2 HBs and 2 His), stephasubimine (IC50 = 285.37 pM, 6 HIs); and stephasubine (IC50 = 1.09 nM, 3 HBs and 4 HIs). These compounds bind with high affinity to different binding pockets, leading to additive effects. Moreover, the pharmacokinetics of six of these seven compounds (except ergosterol peroxide) showed poor absorption in the gastrointestinal tract, which would allow for competitive binding to AG in the small intestine. These results indicate that the development of these 6 compounds into oral antidiabetic agents is promising.
An Integrated Metabolomics Study on Antidiabetic Activity of Christia vespertilionis Leaves Extract Using Chemometric and Molecular Docking Analysis

Abstract:In disease treatment, the utilisation of medicinal plants has witnessed a discernible rise, driven by concerns over the adverse effects associated with synthetic drugs available in the market. Analyses of the plant Christia vespertilionis (L.f.) Bakh. F., indigenous to Malaysia, has suggested its antidiabetic property linked to α-glucosidase inhibition, but metabolites responsible for antidiabetic are unexplored. The metabolomics approaches and molecular docking simulations were integrated to identify the putative α-glucosidase inhibitors and their enzyme interaction. In this study, the crude leaves extracted from this plant were extracted using solvents of varying polarity, followed by gas and liquid chromatography coupled with mass spectrometry metabolomics. The highest inhibition activity in a mixture of n-hexane and ethyl acetate (1:1, v/v)) was observed. Six putative metabolites corresponding to antidiabetic activity were identified: palmitic acid (2), linolenic acid (4), 7-tetradecenal (5), aloeemodin-8-monoglucoside (14), bruceine I (15), and sanjidin B (16). The mechanism of action of all the identified compounds is competitive, mainly involving hydrophobic and hydrogen bonding interactions with the protein residues. Compounds 14, 15, and 16 exhibited strong binding capabilities with both enzyme crystal structures compared to the positive control, quercetin. The metabolites extracted from C. vespertilionis leaves have demonstrated promising antidiabetic effects. These antidiabetic compounds can potentially commercialise new drug candidates in managing diabetes conditions.
Novel Phytochemical Constituents and their Potential to Manage Diabetes

Abstract:Background: Diabetes is a chronic disease affecting a large population worldwide and stands as one of the major global health challenges to be tackled. According to World Health Organization, about 400 million are having diabetes worldwide and it is the seventh leading cause of deaths in 2016. Plant-based natural products have been in use from ancient times as ethnomedicine for the treatment of several diseases, including diabetes. As a result of that, there are several reports on plant-based natural products displaying antidiabetic activity. In the current review, such antidiabetic potential compounds reported from all plant sources along with their chemical structures are collected, presented and discussed. These kinds of reports are essential to pool the available information to one source, followed by statistical analysis and screening to check the efficacy of all known compounds in a comparative sense. This kind of analysis can give rise to a few potential compounds from hundreds, which can further be screened through in vitro and in vivo studies, and human trails leading to the drug development. Methods: Phytochemicals, along with their potential antidiabetic property, were classified according to their basic chemical skeleton. The chemical structures of all the compounds with antidiabetic activities were elucidated in the present review. In addition to this, the distribution and their other remarkable pharmacological activities of each species are also included. Results: The scrutiny of literature led to the identification of 44 plants with antidiabetic compounds (70) and other pharmacological activities. For the sake of information, the distribution of each species in the world is given. Many plant derivatives may exert anti-diabetic properties by improving or mimicking insulin production or action. Different classes of compounds including sulfur compounds (1-4), alkaloids (5-11), phenolic compounds (12-17), tannins (18-23), phenylpropanoids (24-27), xanthanoids (28-31), amino acid (32), stilbenoid (33), benzofuran (34), coumarin (35), flavonoids (36-49) and terpenoids (50-70) were found to be potential active compounds for antidiabetic activity. Of the 70 listed compounds, majorly 17 compounds are obtained from triterpenoids, 13 from flavonoids and 7 from alkaloids. Among all the 44 plant species, the maximum number (7) of compounds were isolated from Lagerstroemia speciosa followed by Momordica charantia (6) and S. oblonga with 5 compounds. Conclusion: This is the first paper to summarize the established chemical structures of phytochemicals that have been successfully screened for antidiabetic potential and their mechanisms of inhibition. The reported compounds could be considered as potential lead molecules for the treatment of type-2 diabetes. Further, molecular and clinical trials are required to select and establish therapeutic drug candidates.

Abstract:Chlorophytum comosum is a plant with medicinal potential traditionally used to treat different diseases. The present study aimed to determine the bioactive compounds, hypoglycemic and antioxidant potential of C. comosum root and leaf. The ethyl acetate extracts of C. comosum root and leaf were analyzed by GC-MS to determine the bioactive compounds. The hypoglycemic potential of the extracts was evaluated by α-amylase, α-glucosidase, glucose diffusion inhibitory assays, and glucose adsorption assay. The ethyl acetate extract of C. comosum root inhibited α-amylase, α-glucosidase, and glucose diffusion in a concentration-dependent manner with IC50 values of 205.39 ± 0.15, 179.34 ± 0.3 and 535.248 μg/mL, respectively, and the leaf extract inhibited α-amylase and α-glucosidase enzymes with IC50 values of 547.99 ± 0.09, and 198.18 ± 0.25 μg/mL respectively. C. comosum root and leaf extracts also improved glucose adsorption. Heptadecanoic acid and dodecanoic acid were identified as potential compounds with hypoglycemic properties through molecular docking. The extracts were also assessed for their antioxidant activity using DPPH, ABTS, and FRAP assays. C. comosum root and leaf extracts were also able to scavenge DPPH radicals with IC50 values of 108.37 ± 0.06 and 181.79 ± 0.09 µM and ABTS radicals with IC50 values of 126.24 ± 0.13 and 264.409 ± 0.08 µM, respectively. The root and leaf extracts also reduced the ferricyanide complex to ferrocyanide with higher reducing powers of 2.24 ± 0.02 and 1.65 ± 0.03, respectively. The results showed that the ethyl acetate extract of C. comosum root has significant antioxidant and hypoglycemic potential compared to the leaf extract. Thus, it can also be studied to isolate the potential compounds with antihyperglycemic activities.

Abstract:Diabetes is marked by postprandial hyperglycemia (PHG), an abnormal rise in blood glucose after meals. A key therapeutic goal to reduce PHG is the inhibition of α-amylase (αAM) and α-glucosidase (αGL), enzymes that break down carbohydrates into sugars. Cucurbita moschata has been shown to inhibit both enzymes. However, its inhibition mechanism has not been explored. This study investigated the in vitro inhibition mechanisms of αAM and αGL and conducted a metabolomic analysis of C. moschata (edible part) water-extract (CME), aiming to preliminarily identify its bioactive compounds (BCs). The inhibitory mechanisms were determined using Lineweaver-Burk plots. The BCs were identified and quantified using HPLC-QTOF-MS, employing both targeted and untargeted metabolomic approaches. CME had a significant higher effect (p < 0.05) on αAM activity than against αGL with IC50 of 28.99 and 698.42 mg/mL, respectively. The extract showed mixed and uncompetitive type inhibitions on αAM and αGL, respectively. The lowest inhibition constant (Ki) was 47.68 mg/mL on αAM activity at 20 mg/mL. Untargeted metabolic profiling by UPLC-MS-ESI-QTOF putatively identified 30 compounds in CME, such as amino acids, vitamins, phytohormones, fatty acids, cucurbitacins and phenolic acids, and flavonoids. Functional analysis of CME identified significant pathways, including pantothenate and CoA biosynthesis and phenylpropanoids, among others. The targeted analysis by UPLC-MS-ESI-QqQ allowed us to identify 12 compounds, with l-phenylalanine, p-hydroxybenzoic, and p-coumaric acid as majors. This study demonstrated the inhibitory potential of CME on αAM and αGL activities, which may be attributed to its metabolites. Thus, this plant represents a valuable source of BC against PHG. Practical Application: The research highlights that Cucurbita moschata has significant potential in managing postprandial hyperglycemia in diabetic patients by inhibiting enzymes like α-amylase and α-glucosidase. In addition, the identification of its compounds emphasizes its importance as a source of bioactive compounds. Therefore, C. moschata could be effectively utilized in the development of nutraceuticals or as an ingredient in functional foods specifically designed for postprandial hyperglycemia management. Thus, integrating C. moschata as part of the daily diet could offer patients with diabetes a natural alternative to control their blood glucose levels after eating.

Abstract:This study was designed to check the potential of secondary metabolites of the selected plants; Citrullus colocynthis, Solanum nigrum, Solanum surattense, Calotropis procera, Agave americana, and Anagallis arvensis for antioxidant, antibacterial, antifungal, and antidiabetic agents. Plant material was soaked in ethanol/methanol to get the crude extract, which was further partitioned via solvent extraction technique. GCMS and FTIR analytical techniques were applied to check the compounds responsible for causing antioxidant, antimicrobial, and antidiabetic activities. It was concluded that about 80% of studied extracts/fractions were active against α-amylase, ranging from 43 to 96%. The highest activity (96.63%) was exhibited by butanol fractions of A. arvensis while the least response (43.65%) was shown by the aqueous fraction of C. colocynthis and the methanol fraction of fruit of S. surattense. The highest antioxidant activity was shown by the ethyl acetate fraction of Anagallis arvensis (78.1%), while aqueous as well as n-hexane fractions are the least active throughout the assay. Results showed that all tested plants can be an excellent source of natural products with potential antimicrobial, antioxidant, and antidiabetic potential. The biological response of these species is depicted as a good therapeutic agent, and, in the future, it can be encapsulated for drug discovery.

Abstract:BackgroundChlorogenic acid is amongst the well-known polyphenolic compounds being used in human food and beverages. Its presence has been reported in tea leaves, roasted green beans, coffee, cocoa, berry fruits, apples, citrus fruits, and pears.ObjectiveThe present study aims to elucidate the effectiveness of chlorogenic acid on in silico and in vitro inhibition of glucose metabolising enzymes (α-amylase and α-glucosidase) and on blood-based markers associated with diabetic complications in vivo.MethodsDocking and molecular dynamics studies were performed using GLIDE (Schrodinger, LLC, NY, 2019-2) and Maestro-Desmond Interoperability Tools, version 4.1 (Schrödinger, NY, 2015), respectively. α-Amylase and α-glucosidase inhibitory activities of chlorogenic acid were measured in vitro. Diabetes was induced in adult Wistar rats by injecting streptozotocin (50 mg/kg). Biochemical assays were performed using standard kits.ResultThe in silico studies for α-amylase and α-glucosidase with chlorogenic acid suggested that the ligand was stable and strongly bound with the above-mentioned proteins. During in vitro studies, chlorogenic acid inhibited both the enzymes in a dose-dependent manner (5–30 μg/mL). In addition, chlorogenic acid treatment for 28 days significantly suppressed the increase in blood glucose, total cholesterol, triglyceride, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, γ-glutamyl transferase, alkaline phosphatase, total bilirubin, creatinine, urea, uric acid, and feed intake levels in diabetic rats. Chlorogenic acid also caused significant improvement in body weight, serum HDL-cholesterol, total protein, and albumin levels leading to betterment in atherogenic indices related to diabetes-associated cardiovascular risks.ConclusionThe findings indicated that chlorogenic acid inhibited α-amylase and α-glucosidase and significantly decreased diabetes associated hyperglycemia, hyperlipidemia, and hepatorenal damage, making it a possible functional food ingredient and drug candidate for the management of diabetes and related complications.

Abstract:Introduction Benincasa hispida (Thunb.) Cogn. (Cucurbitaceae) is an essential food plant in India possessing antihyperglycemic and antihyperlipidemic activities. Objective The objective included comparative estimation of α‐glucosidase and α‐amylase enzyme inhibition potential of B. hispida fractions prepared by microwave‐assisted extraction and prediction of metabolite interaction against non‐insulin‐dependent diabetes mellitus by metabolite profiling based network pharmacology analysis. Methods A validated microwave‐assisted extraction method was employed to obtain different fractions of B. hispida fruits. The in vitro enzyme assay was done with p‐nitrophenyl‐α‐D‐glucopyranoside and acarbose as standard to evaluate antidiabetic potential. The phytomolecules present in the active fraction were identified by UHPLC‐QToF‐MS/MS analysis. Network pharmacology analysis gave possible gene and disease association, combination synergy network, and predicted probable mechanism of action. Results The highest enzyme inhibition potential (IC50) was shown by the ethyl acetate fraction (0.546 ± 0.17 mg/mL and 1.134 ± 0.42 mg/mL) compared to acarbose (0.298 ± 0.08 mg/mL and 0.532 ± 0.38 mg/mL), respectively, for α‐glucosidase and α‐amylase addressing the potential role in ameliorating non‐insulin‐dependent diabetes mellitus. Metabolite profiling resulted in the identification of 17 metabolites, and a synergy between the identified molecules suggested multimolecule action in the amelioration of non‐insulin‐dependent diabetes mellitus through insulin resistance pathway, AMPK signaling pathway, PPAR signaling pathway, and PI3K–Akt signaling pathway. Combination synergy of identified molecules was observed through a multitarget approach to manage non‐insulin‐dependent diabetes mellitus. Conclusion Polyphenol‐enriched fraction of B. hispida fruits and identified phytocompounds ameliorate non‐insulin‐dependent diabetes mellitus. Thus, enriched extract of B. hispida can be further investigated in order to develop high‐quality, safe, and effective products for the management of non‐insulin‐dependent diabetes mellitus.

Abstract:Objective: Diabetic individuals have a higher probability of suffering from illness and death due to small blood vessel-related problems such as retinopathy, neuropathy, nephropathy, and stroke than other complications. There are many synthetic anti-diabetic agents available, but these can be expensive and have undesirable pathological effects. The enzyme α-amylase (hydrolase), catalyzes the hydrolysis of starch to maltose and glucose via the cleavage of α-1,4-glucosidic linkages. Diabetes mellitus patients may benefit from a therapeutic strategy that involves slowing the hydrolysis of starch by inhibiting the activity of α-amylase. Thus, looking for cost-effective, natural, and safe antidiabetic agents is essential. This study aims to screen phytoconstituents and evaluate the in-vitro and in-silico α-amylase inhibitory activity of the ethanolic extract of Adhatoda vasica leaves. Methods: The extraction of Adhatoda vasica leaves was performed using ethanol via the Soxhlet extraction process. Different concentrations (100 μg/mL to 1000 μg/mL) of ethanolic extract, Acarbose, and Sitagliptin, were prepared and evaluated for α-amylase inhibitory activity using the spectrophotometric method. Molecular docking (AutodockVina 1.2.0) and toxicity profiling (SToPToX web server) studies were performed. Results: The ethanolic extract of Adhatoda vasica leaves showed the highest percentage inhibition against α-amylase (56.763 ± 0.0035) at a concentration of 1000 μg/mL. The in-silico study supported this inhibitory activity. Vasicoline (C5) and Quercetin (C9), the active constitute of Adhatoda vasica, showed the best binding energies of -8.3 and -8.0 Kcal/mol, respectively against α-amylase enzyme (PDBID: 4W93). A toxicity study revealed the safety profile of the plant extract. Conclusion: It was concluded that Adhatoda vasica leaves possess some bioactive compounds that are responsible for controlling blood glucose levels, and their identification, purification, and isolation may lead to the development of new therapeutic agents with fewer side effects than the available drugs.
