Abstract:ABSTRACTBrucellosis is one of the most common zoonotic diseases, and current methods of detecting this pathogen are quite difficult. This work combines the benefits of a proximity ligation assay with those of a loop‐mediated isothermal amplification method to develop a novel proximity ligation‐based loop‐mediated isothermal amplification method useful forBrucelladetection. The genomic DNA extraction procedure is not needed. Sensitivity of this assay for detectingBrucella abortusis 1 × 104cells/mL in buffer and 1 × 105cells/mL in milk. The time to detection is within 2 h of initiating the procedure, and no special equipment is needed. This new method is also suitable for the detection of other pathogens, and as such will be useful in the food safety industry.PRACTICAL APPLICATIONSPolymerase chain reaction (PCR) is a sensitivity method for microbe detection, but the complicated genomic DNA extraction procedure and costly equipment needed for this method makes the PCR method unpopular in developing countries. In this study, we present the novel proximity ligation‐based loop‐mediated isothermal amplification (P‐LAMP) method forBrucelladetection; this is the first time to combine the monoclonal antibody for identify microbe and LAMP method for high performance amplification DNA. The genomic DNA extraction procedure is not needed and a water‐bath boiler is the only equipment required to complete the detection process. The P‐LAMP method is useful for food safety pathogen detection in developing countries.

Abstract:Trypanosoma hydrae from the broad-banded watersnake, Nerodia fasciata confluens, underwent development in the freshwater leech, Placobdella parasitica. Epimastigotes and transitional stages were present only in the crop and gastric caecum. Only one metacyclic form was observed. The potential of leeches as vectors is discussed. Two broad-banded watersnakes were infected by inoculation with culture forms of T. hydrae maintained on NNN medium. Parasitemias varied from 10(5)/ml to 10(6)/ml with dividing forms seen only rarely. A broad and a slender form of T. hydrae are described from the peripheral blood of a watersnake 7 months after an experimental infection.

Abstract:A man aged 65 years presented with symptomatic anaemia without overt gastrointestinal bleeding. An oesophagogastroduodenoscopy (EGD) was performed and he was found to have a large ulcerated pedunculated Brunner's gland hamartoma in the duodenal bulb. The polyp was resected using snare cautery in forward and retroflexed positions. Colonoscopy was also performed and a few diminutive polyps were resected. A year later, the patient returned for a surveillance EGD, and no residual polyp was noted. Haemoglobin and iron studies normalised within a few months after polypectomy, with resolution of symptoms.

Abstract:Real-time PCR-based assays specific for Brucella abortus, Brucella melitensis and Brucella suis were developed. The assays utilize an upstream primer that is derived from 3' end of the genetic element IS 711, whereas the downstream primers and probes are designed from signature sequences specific to a species or a biovar. The PCR reactions were monitored for fluorescence resonance energy transfer by including two adjacent labeled probes that hybridize to the amplicons as they are formed. The upstream probes were labeled with fluorescein at 3' end while Cy5 was attached to the 5' end of the downstream probes. An increase in the ratio of fluorescein to Cy5 fluorescence during the cycling was indicative of positive amplification event. The assays were accomplished in less than 30 min using a LightCycler in real-time mode. The assays were tested on known strains as well as field isolates and were found to be specific for all known biovars of B. abortus, B. melitensis and biovar 1 of B. suis. Therefore, specificity, sensitivity, speed and real-time detection make these assays attractive for use in epidemiological and ecological studies.

Abstract:Western media represents Th ird World destinations and communities as stagnant and primitive. Scholars have argued that these representations are fundamentally related to the practices of colonization. Except for a few valuable exceptions research on tourism representations of the Th ird World is scarce. In tourism research, previous investigations have been mainly limited to analyses of visual images and texts of tourist brochures, newspaper articles, postcards and travel magazines. However, researchers have paid little attention to conduct a historical study of Western tourism representations of the Th ird World taking a larger sample of data. Th us, this study explored if there is any diff erence between the way India and Indians are represented in the nineteenth century Western travel writing, and contemporary Western travel writing. Th e results revealed that much like in the nineteenth century, contemporary Western travel writing also emphasizes the romantic image of India and Indians – timeless, poverty-stricken, exotic women and eff eminate men. Th erefore, it can be argued that colonialism is still pervasive in contemporary Western tourism representations.

Abstract:OBJECTIVES: Brucellosis is a major zoonotic disease that poses a significant public health threat worldwide. The classical bacteriological detection process used to identify Brucella spp. is difficult and time-consuming. This study aimed to develop a novel molecular assay for detecting brucellosis.METHODS: All complete sequences of chromosome 1 with 2.1-Mbp lengths were compared among all available Brucella sequences. A unique repeat sequence (URS) locus on chromosome 1 could differentiate Brucella abortus from Brucella melitensis. A primer set was designed to flank the unique locus. A total of 136 lymph nodes and blood samples were evaluated and classified by the URS-polymerase chain reaction (PCR) method in 2013-2014.RESULTS: Biochemical tests and bacteriophage typing as the golden standard indicated that all Brucella spp. isolates were B. melitensis biovar 1 and B. abortus biovar 3. The PCR results were the same as the bacteriological method for detecting Brucella spp. The sensitivity and specificity of the URS-PCR method make it suitable for detecting B. abortus and B. melitensis.CONCLUSION: Quick detection of B. abortus and B. melitensis can provide the most effective strategies for control of these bacteria. The advantage of this method over other presented methods is that both B. abortus and B. melitensis are detectable in a single test tube. Furthermore, this method covered 100% of all B. melitensis and B. abortus biotypes. The development of this URS-PCR method is the first step toward the development of a novel kit for the molecular identification of B. abortus and B. melitensis.
A Label-Based Polymer Nanoparticles Biosensor Combined with Loop-Mediated Isothermal Amplification for Rapid, Sensitive, and Highly Specific Identification of Brucella abortus

Abstract:Brucella abortus (B. abortus), an important zoonotic pathogen in Brucella spp., is the major causative agent of abortion in cattle (namely, bovine brucellosis). Currently, although the isolation and identification of the Brucella abortus were commonly accepted as the gold standard method, it cannot meet the requirements for early diagnostic strategies. Conventional PCR techniques and immunological tests can realize rapid detection of B. abortus, but the demands for PCR thermal cyclers and/or specific antibodies hinder their application in basic laboratories. Thus, rapid, sensitive, and specific diagnostic strategies are essential to prevent and control the spread of the bovine brucellosis. In this work, a novel detection method for the rapid identification of B. abortus, which uses loop-mediated isothermal amplification (LAMP) combined with a label-based polymer nanoparticles lateral flow immunoassay biosensor (LFIA), was established. One set of specific B. abortus-LAMP primers targeting the BruAb2_0168 gene was designed by the online LAMP primer design tool. The B. abortus-LAMP-LFIA assay was optimized and evaluated using various pathogens and whole blood samples. The optimal amplification temperature and time for B. abortus-LAMP-LFIA were determined to be 65°C and 50 min, respectively. The B. abortus-LAMP-LFIA method limit of detection (LoD) was 100 fg per reaction for pure genomic DNA of B. abortus. Meanwhile, the detection specificity was 100%, and there was no cross-reactivity for other Brucella members and non-Brucella strains. Furthermore, the entire procedure, including the DNA preparation for whole blood samples (30 min), isothermal incubation (50 min), and LFIA detection (2-5 min), can be completed in approximately 85 min. Thus, the B. abortus-LAMP-LFIA assay developed was a simple, rapid, sensitive, and reliable detection technique, which can be used as a screening and/or diagnostic tool for B. abortus in the field and basic laboratories.
Rapid, ultrasensitive, and highly specific identification of Brucella abortus utilizing multiple cross displacement amplification combined with a gold nanoparticles-based lateral flow biosensor

Abstract:Brucella abortus (B. abortus) as an important infectious agent of bovine brucellosis cannot be ignored, especially in countries/regions dominated by animal husbandry. Thus, the development of an ultrasensitive and highly specific identification technique is an ideal strategy to control the transmission of bovine brucellosis. In this report, a novel detection protocol, which utilizes multiple cross displacement amplification (MCDA) combined with a gold nanoparticles-based lateral flow biosensor (AuNPs-LFB) targeting the BruAb2_0168 gene was successfully devised and established for the identification of B. abortus (termed B. abortus-MCDA-LFB). Ten specific primers containing engineered C1-FAM (carboxyfluorescein) and D1-biotin primers were designed according to the MCDA reaction mechanism. These genomic DNA extracted from various bacterial strains and whole blood samples were used to optimize and evaluate the B. abortus-MCDA-LFB assay. As a result, the optimal reaction conditions for the B. abortus-MCDA-LFB assay were 66°C for 40 min. The limit of detection of the B. abortus-MCDA-LFB was 10 fg/μl (~3 copies/μl) for genomic DNA extracted from pure cultures of B. abortus isolate. Meanwhile, the B. abortus-MCDA-LFB assay accurately identified all tested B. abortus strains, and there was no cross-reaction with non-B. abortus pathogens. Moreover, the detection workflow of the B. abortus-MCDA-LFB assay for whole blood samples can be completed within 70 min, and the cost of a single test is approximately 5.0 USD. Taken together, the B. abortus-MCDA-LFB assay is a visual, fast, ultrasensitive, low-cost, easy-to-operate, and highly specific detection method, which can be used as a rapid identification tool for B. abortus infections.

Abstract:ABSTRACT Brucellosis is a zoonotic disease caused by Brucella spp. and is a major threat to human and livestock health. The accurate and rapid detection of Brucella DNA is essential for the diagnosis and treatment of this disease. However, traditional diagnostic methods, such as culture and serological tests, have limitations in terms of sensitivity, specificity, and time consumption. To address these challenges, we developed a highly sensitive one-tube nested quantitative real-time PCR (qPCR) method to detect Brucella DNA using a closed-tube approach with two primers and two probes that sequentially react with encoding a Brucella outer membrane protein. The analytical sensitivity of this one-tube nested qPCR approach was 100 fg/μL, which is 100-fold higher than that of a conventional qPCR. Intra-batch and inter-batch replicates showed low coefficients of variation, both less than 5%. The study included 250 clinical samples, showing that this one-tube nested qPCR method has a specificity of 100% and a sensitivity of 98.6%, exceeding the sensitivity of conventional qPCR (84.1%) at a cycle threshold (CT) cutoff of 38. More importantly, the one-tube nested qPCR reduced the CT values by an average of 6.4 compared to conventional qPCR, significantly improving the detection rate of low-load samples (CT > 35). In conclusion, the bcsp31 one-tube nested qPCR method is a promising tool for the detection of brucellosis, owing to its high sensitivity and operational simplicity. This study highlights the potential of qPCR-based methods to improve the accuracy and speed of brucellosis diagnoses. IMPORTANCE This study developed a highly sensitive and efficient method for the detection of brucellosis by introducing a one-tube nested quantitative real-time PCR (qPCR) approach, representing a remarkable advance in the field. The method demonstrated an impressive analytical sensitivity of 100 fg/μL, surpassing conventional qPCR and enabling the detection of even low levels of Brucella DNA. In addition, the study’s comprehensive evaluation of 250 clinical samples revealed a specificity of 100% and a sensitivity of 98.6%, underscoring its reliability and accuracy. Most importantly, the new method significantly improved the detection rate of low-burden samples, reducing cycle threshold values by an average of 6.4. These results underscore the immense potential of this approach to facilitate rapid and accurate brucellosis diagnosis, which is critical for effective disease management and control.

Abstract:Brucellosis is a bacterial infectious disease caused mainly by Brucella. Transmission is mainly by contact with infected domestic or wild animals or their excreta. Clinical diagnosis of brucellosis is usually based on qualitative total antibody tests, which make it difficult to differentiate between acute and pre-existing infections. In this study, for the simultaneous detection of anti-brucellosis IgG and IgM, we innovatively developed a surface-enhanced Raman scattering (SERS)-lateral flow immunoassay (LFIA) detection system, and evaluated its performance and diagnostic effect by using clinical serum samples. The key design of this biosensor involves the preparation of two immunoprobes using two Raman spheres (R-Sphere) with distinct signals conjugated to mouse anti-human IgG and IgM, respectively. In this design, brucellosis-specific antigens are embedded in the T-line, allowing simultaneous capture of anti-brucellosis IgG and IgM in a specific binding reaction. A portable Raman instrument is used to detect the characteristic signal intensities generated by IgM and IgG on the T-line, which can then be used to analyze the target IgM and IgG antibodies. Based on the analysis of 40 clinical samples, the method has a sensitivity and specificity of 100%, and the detection time is only 15 min. The advantages of this technology are fast speed, convenient use, high sensitivity, and it can distinguish the disease course to achieve precise treatment. These features make it an ideal solution for large-scale brucellosis testing in remote and nomadic areas, where it can play a crucial role in improving public health protection.

Abstract:A pair of primers were designed according to the sequence of specific Brucella gene of Outer membrane protein(OMP31),the OMP31 gene was amplified by PCR and cloned into pMD18-T vector,and then the gene sequence was detected.The results showed that the target gene band at a length 602 bp was amplified by PCR,with a homology of 99.7% to the gene sequence reported in GenBank.The experiments had proved that PCR assay possessed a high specificity,DNA bands couldn't amplified in other bacterial except Brucella.And the sensitivity test results indicated that PCR assay was more sensitive,which could detect OMP31 with only 0.9 pg/μL DNA.The successfully construction of the PCR assay provides strongly technical support for detection,identification and epidemiological investigations of bovine brucellosis.
Strain-level Identification of Brucella Melitensis Reference Strain 63/9 Using Multiplex PCR Method by Targeting BMEA_B0162 and BMEA_A1238

Abstract:Brucellosis is one of the most prevalent bacterial zoonosis worldwide. Brucella melitensis (B. melitensis), Brucella abortus (B. abortus), Brucella canis (B. canis), Brucella suis (B. suis) are common to cause disease in humans and B. melitensis is the most pathogenic causative agent of brucellosis in humans and animals. Fast, efficient and accurate identification of Brucella reference strains at the strain-level is indispensable for microbiological method quality assurance and downstream applications. B. melitensis 63/9 is recognized as an important reference strain for the microbiological culture collection organizations worldwide, and the identification of B. melitensis strain 63/9 is still lacking. The genomic sequences of B. melitensis 63/9 and nine other Brucella strains were compared. Two specific genes were selected for the multiplex PCR method. Gene BMEA_B0162 with unknown function is the key target to identify B. melitensis 63/9, and gene BMEA_A1238 annotated as TRAP transporter solute receptor is included as a control gene for the Brucella genus. A multiplex PCR was established in this study to differentiate B. melitensis reference strain 63/9 from 39 B. melitensis strains, 13 B. abortus strains, 5 B. suis strains, 6 B. canis strains, 3 E. coli, and 4 Salmonella strains by targeting the BMEA_B0162 and the BMEA_A1238 in the genome. This method allows at least 100 pg of B. melitensis 63/9 genomic DNA to be detected. We established a fast, and a cost-effective method to distinguish B. melitensis 63/9 from other Brucella strains and some non-Brucella bacteria strains with high sensitivity and specificity, making the first report about the identification of Brucella reference strain recognized by World Organization for Animal Health at the strain-level.
A rapid minor groove binder PCR method for distinguishing the vaccine strain Brucella abortus 104M

Abstract:Background: Brucellosis is a widespread zoonotic disease caused by Gram-negative Brucella bacteria. Immunisation with attenuated vaccine is an effective method of prevention, but it can interfere with diagnosis. Live, attenuated Brucella abortus strain 104M has been used for the prevention of human brucellosis in China since 1965. However, at present, no fast and reliable method exists that can distinguish this strain from field strains. Single nucleotide polymorphism (SNP)-based assays offer a new approach for such discrimination. SNP-based minor groove binder (MGB) and Cycleave assays have been used for rapid identification of four Brucella vaccine strains (B. abortus strains S19, A19 and RB51, and B. melitensis Rev1). The main objective of this study was to develop a PCR assay for rapid and specific detection of strain 104M. Results: We developed a SNP-based MGB PCR assay that could successfully distinguish strain 104M from 18 representative strains of Brucella (B. abortus biovars 1, 2, 3, 4, 5, 6, 7 and 9, B. melitensis biovars 1, 2 and 3, B. suis biovars 1, 2, 3 and 4, B. canis, B. neotomae, and B. ovis), four Brucella vaccine strains (A19, S19, S2, M5), and 55 Brucella clinical field strains. The assay gave a negative reaction with four non-Brucella species (Escherichia coli, Pasteurella multocida, Streptococcus suis and Pseudomonas aeruginosa). The minimum sensitivity of the assay, evaluated using 10-fold dilutions of chromosomal DNA, was 220 fg for the 104M strain and 76 fg for the single non-104M Brucella strain tested (B. abortus A19). The assay was also reproducible (intra- and inter-assay coefficients of variation = 0.006-0.022 and 0.012-0.044, respectively). Conclusions: A SNP-based MGB PCR assay was developed that could straightforwardly and unambiguously distinguish B. abortus vaccine strain 104M from non-104M Brucella strains. Compared to the classical isolation and identification approaches of bacteriology, this real-time PCR assay has substantial advantages in terms of simplicity and speed, and also reduces potential exposure to live Brucella. The assay developed is therefore a simple, rapid, sensitive, and specific tool for brucellosis diagnosis and control.

Abstract:We have developed a combinatorial polymerase chain reaction (PCR) procedure to identify four major species of the genus Brucella simultaneously. Four pairs of primers targeting the genes encoding a cell surface protein (BCSP31) and outer membrane proteins (omp2b, omp2a and omp31) were prepared. PCR using these primers gave rise to specific patterns of amplification for each Brucella spp. examined in this study. B. abortus could be identified when fragments of BCSP31 and omp2b/2a were amplified by B. abortus-specific primers. B. melitensis could be identified by the amplification of fragments of BCSP31, omp2b/2a and omp31 using pair of primers B4/B5, JRF/JPR-ab and omp31. Identification of B. canis could be achieved when the amplicons of omp2b/2a were detected by B. canis-specific primers, as could the identification of BCSP31 and omp31. If specific amplifications occurred using all pairs of primers, the strain was identified as B. suis. Combinatorial PCR reported here thus appeared to be an ideal method of identifying Brucella spp., the causative pathogen of human brucellosis.

Abstract:Laboratory detection of Brucella is based largely on bacterial isolation and phenotypic characterization. These methods are lengthy and labor-intensive and have been associated with a heightened risk of laboratory-acquired infection. Antibody based indirect detection methods also suffer from limitations in proper diagnosis of the organism. To overcome these problems, nucleic acid amplification has been explored for rapid detection and confirmation of the presence of Brucella spp. PCR-based diagnostics is useful for screening large populations of livestock to identify infected individuals and confirms the presence of the pathogen. Random Amplification of Polymorphic DNA (RAPD) was performed and identified a 1.3 kb PCR fragment specifically amplifiable from DNA isolated from Brucella. A BLAST search revealed no significant homology with the reported sequences from species other than the members of Brucella. The isolated fragment seems to be a part of d-alanine-d-alanine ligase gene in Brucella sp. Translational BLAST revealed certain degree of homology of this sequence with orthologs of this gene reported from other microbial species at the deduced amino acid level. The sequence information was used to develop PCR based assays to detect Brucella sp. from various samples. The minimum detection limit of Brucella from blood and milk samples spiked with Brucella DNA was found to be 1 ng/ml and 10 ng/ml, respectively. In conclusion, we demonstrated that the PCR based detection protocol was successfully used for the detection of Brucella from various organs and spiked samples of diseased sheep. Diagnosis of Brucellosis by PCR based method reported in this study is relatively rapid, specific and simple.

Abstract:The diagnosis of brucellosis is the corner stone in any control and eradication program. Therefore, the main objective of the present study was to apply more advanced techniques for rapid and accurate diagnosis of brucellosis that can overcome the draw backs of the traditional diagnostic techniques. Different polymerase chain reaction (PCR) assays were applied in the present study, either singly or in a multiplex format, that enable to detect and differentiate most of Brucella species. The PCR assay detection limit was evaluated in a preliminary study. The obtained results recommend the PCR assay as a valuable, rapid, very specific, highly sensitive and safe laboratory diagnostic test that can be used not only for detection of Brucella antigen either in culture or in clinical samples but also in differentiating most of the virulent and vaccinal strains. Abstract:Objective To establish a real-time quantitative PCR (RT-PCR) preliminary screening method for rapid detection of Brucella. Methods Based on the nucleotide sequence encoding Brucella's outer membrane proteins omp10 and omp31, four primers and probes of omp10, omp10-1, omp31 and omp31-1 were designed. The primers and probes were used to detect 19 strains of 6 categories of Brucella DNA of known organisms, 10 strains of Brucella DNA that were isolated from Yuxi of Yunnan. And 224 negative Brucella DNA, including 35 strains of Bartonella DNA, 103 strains of the Lord Komori enterocolitis DNA (including 4 parts of O∶3 and 9 parts of O∶9) and 86 samples of hybrids bacteria DNA. Then the specificity of primers and probes were evaluated based on the test results. Results The DNA of 19 standard Brucella strains could be amplified by omp10 and omp10-1, and the peak time and amplification curve of omp10 is better than omp10-1, and the DNA of negative control strains could not be amplified by omp10. The DNA of 16 standard Brucella strains could be amplified by omp31-1. The DNA of standard Brucella strains could not be amplified by omp31. The average Ct values of 10 strains of Brucella DNA which were isolated from Yuxi of Yunnan that were detected by omp10, omp10-1 and omp31-1 respectively were 19.87, 19.14 and 17.52. Conclusion Omp10 has strong specificity and only specific for Brucella, so it can be used for rapid detection of Brucella. Key words: Brucella; Real-time PCR; Rapid detection; Omp10; Specificity

Why are blue visual pigments blue? A resonance Raman microprobe study.

Abstract:A resonance Raman microscope has been developed to study the structure of the retinal prosthetic group in the visual pigments of individual photoreceptor cells. Raman vibrational spectra are obtained by focusing the probe laser on intact photoreceptors frozen on a 77 K cold stage. To elucidate the mechanism of wavelength regulation in blue visual pigments, we have used this apparatus to study the structure of the chromophore in the 440-nm absorbing pigment found in "green rods" of the toad (Bufo marinus). The 9-cis isorhodopsin form of the green rod pigment exhibits a 1662-cm-1 C = NH+ Schiff base stretching mode that shifts to 1636 cm-1 in deuterium-substituted H2O. This demonstrates that the Schiff base linkage to the protein is protonated. Protonation of the Schiff base is sufficient to explain the 440-nm absorption maximum of this pigment without invoking any additional protein-chromophore interactions. The absence of additional perturbations is supported by the observation that the ethylenic band and the perturbation-sensitive C-10-C-11 and C-14-C-15 stretching modes have the same frequency as those of the 9-cis protonated retinal Schiff base in solution. Our demonstration that a blue visual pigment contains an unperturbed protonated Schiff base provides experimental evidence that the protein charge perturbation responsible for the opsin shift in the 500-nm absorbing pigments is removed in the opsins of blue pigments, as suggested by the sequence data.
