Abstract:Magnesium (Mg) alloys have been promised for biomedical implants in orthopedic field, however, the fast corrosion rate and mode challenge their clinical application. To push Mg alloys materials into practice, a composite coating with biodegradable and high compatible components to improve anticorrosion property of an Mg alloy (i.e., AZ31) is designed and fabricated. The inner layer is micro-nano structured Mg(OH)2 through hydrothermal treatment. Then stearic acid (SA) is introduced to modify Mg(OH)2 for better reducing the gap below a surface-degradation polymer layer of poly(1,3-trimethylene carbonate). Benefited by the SA modification effect, this sandwiched coating avoids corrosive medium penetration via enhancing the adhesion strength at the interface between outer and inner layers. Both in vitro and in vivo tests indicate that the composite coating modified AZ31 perform a better anticorrosion behavior and biocompatibility compared to bare AZ31. Strikingly, a 1.7-fold improvement in volume of newly formed bone is observed surrounding the composite coating modified implant after 12 week implantation. The sandwiched biocompatible coating strategy paves a hopeful way for future translational application of Mg alloys orthopedic materials in clinics.

Abstract:Biocompatible and biodegradable magnesium (Mg) and its alloys possess excellent potential as orthopedic implant biomaterials, but they corrode too quickly inside the human body to maintain such functions as supporting and fixation. In this study, we have successfully prepared a composite biomedical material with Mg as the substrate and a multilayered coating on the metal, which contains a chemical conversion layer of fluoride or phosphate, an adhesion layer, and a layer of biodegradable polylactic acid (PLA). The recorded weight loss and the release of ionic magnesium (Mg2+) in the simulated body fluid (SBF) demonstrate that the corrosion rate of the protected Mg has been reduced to 1/10 that of the unprotected bare Mg. The corrosion rate of the protected Mg can be adjusted with either the chemical conversion type or the thickness of the PLA layer. The bare and the protected Mg has been manufactured as an implant for bone defect repair of Sprague-Dawley rats. The X-ray and micro-CT images show an improved anticorrosion of the protected Mg in vivo, and the histological analysis indicates that the Mg2+ ions released at an appropriate rate are beneficial for bone growth. The preliminary tests prove that the biodegradable Mg has been effectively protected by the innovative multilayered coating, and the implant has met the required biological functions such as biocompatibility and promotion of bone growth.

Abstract:Abstract Nowadays, the need for bio‐implants, which can gradually degrade after fulfilling the therapeutic tasks is continuously increasing. Under such situation, magnesium (Mg) and its alloys have been proposed and intensively studied as the new‐generation medical implants due to their favorable biodegradability and biocompatibility. However, their swift corrosion in physiological environments can always cause an early fracture and further the surgical failure, greatly hindering their broad applications. Therefore, great efforts have been made to alter the degradation behaviors of Mg‐based implants. Biodegradable polymeric surface coatings have been revealed to be a straightforward and effective strategy for retarding the fast degradation and improving the bioactivity of Mg and its alloys. This article reviews the recent progress of polymer‐based coatings on Mg substrates, regarding the coating strategies, coating properties, and their performance in corrosive protection and biocompatibility promotion via in vitro as well as some in vivo models. The specific pros and cons of different polymeric coatings are also discussed. Finally, we put forward some perspectives on the future direction of polymeric coatings on biomedical Mg‐based implants to better adapt to clinical trials.

Abstract:Manipulating the degradation rate of biomedical magnesium alloys poses a challenge. The characteristics of a microarc oxidation (MAO), prepared in phytic acid, and poly(L-lactic acid) (PLLA) composite coating, fabricated on a novel Mg-1Li-1Ca alloy, were studied through field emission scanning electron microscopy (FE-SEM), electron probe X-ray microanalysis (EPMA), energy dispersive X-ray spectroscopy (EDS) and X-ray diffraction (XRD). The corrosion behaviors of the samples were evaluated via hydrogen evolution, potentiodynamic polarization and electrochemical impedance spectroscopy in Hanks' solution. The results indicated that the MAO/PLLA composite coatings significantly enhanced the corrosion resistance of the Mg-1Li-1Ca alloy. MTT and ALP assays using MC3T3 osteoblasts indicated that the MAO/PLLA coatings greatly improved the cytocompatibility, and the morphology of the cells cultured on different samples exhibited good adhesion. Hemolysis tests showed that the composite coatings endowed the Mg-1Li-1Ca alloys with a low hemolysis ratio. The increased solution pH resulting from the corrosion of magnesium could be tailored by the degradation of PLLA. The degradation mechanism of the composite coatings was discussed. The MAO/PLLA composite coating may be appropriate for applications on degradable Mg-based orthopedic implants.

Abstract:Magnesium alloy is becoming one of the main candidates for biodegradable orthopedic implants due to its excellent biocompatibility and mechanical properties matching with natural bone. However, its rapid degradation rate hinders its clinical application. In this study, the organic (polydopamine) and inorganic (calcium phosphates) composite coating was prepared by chemical conversion and one step impregnation method to improve corrosion resistance and osteogenic differentiation for orthopedic implant materials . The formation mechanism of the coating was discussed by phase analysis and surface morphology observation. The results of anti-corrosion performance showed that the Ca-Mg-P/PDA coating greatly reduced the hydrogen evolution amount compared with WE43 alloy substrate and Ca-Mg-P coating. The biocompatibility and osteogenic differentiation ability of the coated alloys was studied, the alloy with Ca-Mg-P/PDA composite coating exhibited the best osteogenic differentiation ability in both short and long term of osteogenic differentiation.

Abstract:Magnesium alloys have great application prospects as ideal bone implant materials. However, their poor corrosion resistance limits their clinical orthopedic application. Surface modification promotes the corrosion resistance of magnesium. Conversion coatings, such as calcium phosphate (Ca-P) coating, microarc oxidation (MAO) treatment, and fluoride (FLU) treatment, have been extensively investigated in in vivo studies. This systematic review and network meta-analysis compared the influence of different conversion coatings on bone repair, material properties, and systemic host response in orthopedic applications. Using the PICOS model, the inclusion criteria for biodegradable magnesium and its alloys were determined for in vivo studies. Four databases were used. The standard and weight mean differences with 95% confidence intervals were used to analyze new bone formation and degradation rate. Network structure and forest plots were created, and ranking probabilities were estimated. The risk of bias and quality of evidence were assessed using SYRCLE, CERQual, and GRADE tools. In the qualitative analysis, 43 studies were selected, and the evaluation of each outcome indicator was not entirely consistent from article to article. In the quantitative analysis, 21 articles were subjected to network meta-analysis, with 16 articles on implant degradation and 8 articles for new bone formation. Additionally, SUCRA indicated that Ca-P coating exhibited the highest corrosion resistance, followed by FLU treatment. MAO demonstrated the best capability for new bone formation, followed by Ca-P coating. Ca-P coating exhibited the highest overall performance. To conclude, coated Mg can promote better new bone formation than bare Mg and has considerable biocompatibility. Ca-P-coated Mg and MAO-coated Mg have the greatest potential to significantly promote corrosion resistance and bone regeneration, respectively. The findings of this study will provide a theoretical basis for the investigation of composite coatings and guidance for the orthopedic application of Mg bone implants.

Abstract:Magnesium (Mg) alloys are being investigated as a biodegradable metallic biomaterial because of their mechanical property profile, which is similar to the human bone. However, implants based on Mg alloys are corroded quickly in the body before the bone fracture is fully healed. Therefore, we aimed to reduce the corrosion rate of Mg using a double protective layer. We used a magnesium-aluminum-zinc alloy (AZ91) and treated its surface with micro-arc oxidation (MAO) technique to first form an intermediate layer. Next, a bioceramic nanocomposite composed of diopside, bredigite, and fluoridated hydroxyapatite (FHA) was coated on the surface of MAO treated AZ91 using the electrophoretic deposition (EPD) technique. Our in vivo results showed a significant enhancement in the bioactivity of the nanocomposite coated AZ91 implant compared to the uncoated control implant. Implantation of the uncoated AZ91 caused a significant release of hydrogen bubbles around the implant, which was reduced when the nanocomposite coated implants were used. Using histology, this reduction in the corrosion rate of the coated implants resulted in an improved new bone formation and reduced inflammation in the interface of the implants and the surrounding tissue. Hence, our strategy using a MAO/EPD of a bioceramic nanocomposite coating (i.e., diopside-bredigite-FHA) can significantly reduce the corrosion rate and improve the bioactivity of the biodegradable AZ91 Mg implant.

A Study on the Structure and Biomedical Application Characteristics of Phosphate Coatings on ZKX500 Magnesium Alloys

Abstract:Magnesium-matrix implants can be detected by X-ray, making post-operative monitoring easier. Since the density and mechanical properties of Mg alloys are similar to those of human bones, the stress-shielding effect can be avoided, accelerating the recovery and regeneration of bone tissues. Additionally, Mg biodegradability shields patients from the infection risk and medical financial burden of needing another surgery. However, the major challenge for magnesium-matrix implants is the rapid degradation rate, which necessitates surface treatment. In this study, the ZKX500 Mg alloy was used, and a non-toxic and eco-friendly anodic oxidation method was adopted to improve corrosion resistance. The results indicate that the anodic coating mainly consisted of magnesium phosphate. After anodic oxidation, the specimen surface developed a coating and an ion-exchanged layer that could slow down the degradation and help maintain the mechanical properties. The results of the tensile and impact tests reveal that after being immersed in SBF for 28 days, the anodic oxidation-treated specimens maintained good strength, ductility, and toughness. Anodic coating provides an excellent surface for cell attachment and growth. In the animal experiment, the anodic oxidation-treated magnesium bone screw used had no adverse effect and could support the injured part for at least 3 months.

Abstract:Magnesium (Mg) and its alloys have proved promising as biodegradable candidates for the repair of bone tissue. Despite the encouraging bio-related properties of Mg, its high corrosion rate in contact with body fluids still presents a major challenge. An efficient approach to address this issue is to provide a protective coating on Mg. The present research evaluates, for the first time, in vitro bioactivity and biocompatibility of a novel multifunctional composite coating based on poly(methyl methacrylate) (PMMA) biopolymer and bioactive glass (BG) particles on Mg-based implant. Electrophoretic deposition (EPD) was utilized to obtain this coating from a bi-component suspension. Coatings' morphological analyses by scanning electron microscopy established that EPD provides a facile route for simultaneous deposition of the biopolymeric matrix and BG in the form of a dense and homogeneous coating on the surface of Mg. Chemical characterizations of the coatings performed by energy-dispersive spectroscopy and FT-IR spectroscopy confirmed the presence of both PMMA and BG components in the composite film. In vitro acellular bioactivity test in simulated body fluid (SBF) verified bone-like apatite formation on coatings due to the presence of BG particles. Additionally, in vitro cellular cytotoxicity using MTT assay demonstrated cellular viability in contact with the composite coatings. Moreover, the corrosion behavior of the pure Mg vs coated Mg samples in SBF was assessed electrochemically using linear polarization and impedance spectroscopy techniques. A significant increase in the polarization resistance from 65.89 Ω.cm 2 for uncoated Mg to 1365.1 Ω.cm 2 for Mg coated with a composite film (obtained from 45 g/L PMMA and 3.5 g/L BG suspension) was observed. This indicated a reduction of 95.17% in the corrosion rate of Mg substrate by application of a composite coating. The dramatic reduction in Mg substrate corrosion rate accompanied by in vitro bioactivity and cytocompatibility of the developed composite, reflect the high potential of the present strategy in the modification of Mg-based implants for successful bone tissue engineering applications.

Abstract:Forming a stable anti-corrosion surface layer on magnesium (Mg) and its alloys has become a major challenge in developing a desirable degradable medical implant in bone. In this study, a porous MgO layer was first formed on Mg by plasma electrolytic oxidation (PEO), and then a Mg-Al layered double hydroxide (LDH) layer was prepared to seal the porous structure of the PEO layer (LDH-2h and LDH-12h) via hydrothermal treatment. The bilayer structure composite coating, which can effectively resist the penetration of surrounding media, is similar to plain Chinese tiles. The in vitro results revealed that compared with other coatings, the LDH-12h composite coating can reduce the release of Mg ions and induce a milder change in pH when immersed in phosphate-buffered saline (PBS). In vitro rat bone marrow stem cell (rBMSC) culture suggested that the LDH-12h composite coating is favorable for cell activity, proliferation and could improve the osteogenic activity of rBMSCs. A subcutaneous implantation test revealed that the as-prepared sample showed enhanced corrosion resistance and histocompatibility in vivo, especially in the LDH-12h group. Moreover, LDH-12h had the lowest rate of degradation and the closest combination with the new bone after being inserted into a rat femur for 12 weeks with no major organ dysfunction. In summary, the asprepared PEO/Mg-Al LDH composite coating is able to improve the corrosion resistance and biocompatibility of Mg and to enhance osteogenic activity in vivo, suggesting its promising prospects for orthopedic applications.

Abstract:Magnesium alloy is currently regarded as the most favourable biodegradable metal; however, obstacles remain to be overcome in terms of managing its corrosion and ensuring its biocompatibility. In this study, a metal-organic complex comprising Ca ions incorporated in tannic acid (TA) was prepared and used to coat magnesium alloy by chemical conversion and dipping processes, followed by modification with stearic acid (SA). This metal-organic complex coating was demonstrated to be homogeneous and compact, and it significantly improved the electrochemical corrosion resistance and long-term degradation behaviour of the coated samples. Consequently, the well-controlled release of Mg and Ca ions, as well as the osteo-compatible TA and SA molecules, promoted the proliferation of osteoblast cells. This metal-organic complex coating offers a promising modifying strategy for magnesium-based orthopaedic implants.
Corrosion and biocompatibility improvement of magnesium-based alloys as bone implant materials: a review

Abstract:Magnesium (Mg) or its alloys are widely tested as potential orthopedic implants, particularly as biodegradable alloys for fracture fixation due to their mechanical properties are close to those of bone. Currently, available Mg or its alloys are confronted with challenges in passing regulatory biosafety tests prior to clinical trials due to its fast degradation and associated degradation products. The degradation of Mg is accompanied by the release of Mg ions, the rise of pH and osmolality in surrounding environments. According to the standard of ISO 10993 Part 13, the pH value shall be appropriate to the site of intended use maintaining in an appropriate range. Approaches to overcome these challenges include the selection of adequate alloying elements, proper surface treatment techniques and control of the degradation rate of Mg or its alloys developed as orthopedic implants. To date, Mg or its alloy-based bone implants have not yet been widely used in clinical applications as medical implants. This review critically summarized published methods to improve the corrosion resistance of Mg and its alloys. The current progress on in vitro cytotoxicity and in vivo biocompatibility properties of these metals was also reviewed. This review aimed to provide a reference for further research and development (R&D) of biodegradable Mg and its alloys with regard to the evaluation of their corrosion process and biocompatibility and facilitation of their translation to clinical applications.

Abstract:The development of biodegradable implants is certainly intriguing, and magnesium and its alloys are considered significant among the various biodegradable materials. Nevertheless, the fast degradation, the generation of a significant amount of hydrogen gas, and the escalation in the pH value of the body solution are significant barriers to their use as an implant material. The appropriate approach is able to solve this issue, resulting in a decrease the rate of Mg degradation, which can be accomplished by alloying, surface adjustment, and mechanical treatment. Surface modification is a practical option because it not only improves corrosion resistance but also prepares a treated surface to improve bone regeneration and cell attachment. Metal coatings, ceramic coatings, and permanent polymers were shown to minimize degradation rates, but inflammation and foreign body responses were also suggested. In contrast to permanent materials, the bioabsorbable polymers normally show the desired biocompatibility. In order to improve the performance of drugs, they are generally encapsulated in biodegradable polymers. This study summarized the most recent advancements in manufacturing polymeric coatings on Mg alloys. The related corrosion resistance enhancement strategies and future potentials are discussed. Ultimately, the major challenges and difficulties are presented with aim of the development of polymer-coated Mg-based implant materials.

Abstract:The biodegradability of magnesium (Mg) metal implants necessitates surface modification to mitigate the high corrosion rate in the body's physiological environment. This study focuses on synthesizing and applying a nanocomposite coating of polydimethylsiloxane-nanosized bioactive glass (PDMS-nBG) on the surface of a plasma electrolytic oxidation (PEO)-modified AZ91 Mg alloy. The PDMS-1 wt% nBG nanocomposite coating was deposited on the PEO-treated substrate using the dip-coating method. Surface and cross-sectional morphologies of the nanocomposite coatings were examined using field emission scanning electron microscopy (FESEM). Corrosion behavior of the AZ91 substrate, PEO coating, and double-layer PEO/PDMS-nBG coating was investigated using electrochemical impedance spectroscopy (EIS) and potentiodynamic polarization (PDP) tests. Results showed that the corrosion resistance and corrosion current density of Mg substrates increased and decreased from 4815 Ω.cm 2 and 8.96 μA.cm − 2 to 18 MΩ.cm 2 and 0.00016 μA.cm − 2 for AZ91 and PEO/PDMS-nBG coating, respectively. Additionally, the morphology and adhesion of cultured human mesenchymal cells on the PEO-modified AZ91 substrates with a single PDMS polymer coating and a PDMS-nBG nanocomposite coating were investigated using fluorescent light microscopy and FESEM. The PEO/PDMS-nBG bilayer coating effectively addressed the limitations of magnesium alloys and holds potential for biomedical applications, particularly in orthopedic implants, due to the enhanced biocompatibility and bioactivity of Mg alloys.

In this study, we developed a novel functional and biocompatible coating strategy through polydopamine mediated assembly of hydroxyapatite nanoparticles and growth factor, bone morphogenetic protein-2 (BMP-2), onto the surface of AZ31 Mg alloys. Such functional coating has strong bonding with the substrate and can increase surface hydrophilicity of magnesium alloys. In vitro electrochemical corrosion and hydrogen evolution tests demonstrate that the coating can significantly enhance the corrosion resistance and therefore slow down the degradation of AZ31 Mg alloys. In vitro cell culture reveals that immobilization of HA nanoparticles and BMP-2 can obviously promote cell adhesion and proliferation. Furthermore, in vivo implantation tests indicate that with the synergistic effects of HA nanoparticles and BMP-2, the coating does not cause obvious inflammatory response and can significantly reduce the biodegradation rate of the magnesium alloys and induce the new bone formation adjacent to the implants. © 2017 Wiley Periodicals, Inc. A surface-engineered multifunctional TiO2 based nano-layer simultaneously elevates the corrosion resistance, osteoconductivity and antimicrobial property of a magnesium alloy

Abstract:Magnesium biometals exhibit great potentials for orthopeadic applications owing to their biodegradability, bioactive effects and satisfactory mechanical properties. However, rapid corrosion of Mg implants in vivo combined with large amount of hydrogen gas evolution is harmful to bone healing process which seriously confines their clinical applications. Enlightened by the superior biocompatibility and corrosion resistance of passive titanium oxide layer automatically formed on titanium alloy, we employ the Ti and O dual plasma ion immersion implantation (PIII) technique to construct a multifunctional TiO2 based nano-layer on ZK60 magnesium substrates for enhanced corrosion resistance, osteoconductivity and antimicrobial activity. The constructed nano-layer (TiO2/MgO) can effectively suppress degradation rate of ZK60 substrates in vitro and still maintain 94% implant volume after post-surgery eight weeks. In animal study, a large amount of bony tissue with increased bone mineral density and trabecular thickness is formed around the PIII treated group in post-operation eight weeks. Moreover, the newly formed bone in the PIII treated group is well mineralized and its mechanical property almost restores to the level of that of surrounding mature bone. Surprisingly, a remarkable killing ratio of 99.31% against S. aureus can be found on the PIII treated sample under ultra-violet (UV) irradiation which mainly attributes to the oxidative stress induced by the reactive oxygen species (ROS). We believe that this multifunctional TiO2 based nano-layer not only controls the degradation of magnesium implant, but also regulates its implant-to-bone integration effectively. STATEMENT OF SIGNIFICANCE: Rapid corrosion of magnesium implants is the major issue for orthopaedic applications. Inspired by the biocompatibility and corrosion resistance of passive titanium oxide layer automatically formed on titanium alloy, we construct a multifunctional TiO2/MgO nanolayer on magnesium substrates to simultaneously achieve superior corrosion resistance, satisfactory osteoconductivity in rat intramedullary bone defect model and excellent antimicrobial activity against S. aureus under UV irradiation. The current findings suggest that the specific TiO2/MgO nano-layer on magnesium surface can achieve the three objectives aforementioned and we believe this study can demonstrate the potential of biodegradable metals for future clinical applications.
Micro-arc oxidation-assisted sol-gel preparation of calcium metaphosphate coatings on magnesium alloys for bone repair

Abstract:Calcium phosphate coating is an attractive surface modification strategy for magnesium alloys, since it can increase their corrosion resistance and endow them with osteogenic function simultaneously. Herein, a calcium metaphosphate (CMP) coating was fabricated on magnesium alloy by using sol-gel approach assisted with micro-arc oxidation pre-treatment. Scanning electron microscopy showed that the micro-pores and cracks in micro-arc oxidation inner layer generated during the pre-treatment process were sealed by the grainy sol-gel outer layer. Energy dispersive spectrometry and X-ray diffraction results demonstrated the identity of the coating as CMP. The cross-cut test showed that the adhesion of CMP coating was strong. Applying bare magnesium alloy substrate as a control, the CMP coating surface was rougher and more hydrophilic. The potentiodynamic polarization test demonstrated that the corrosion resistance was significantly improved by using CMP coating. Hydrogen evolution in immersion test further confirmed that the degradation rate was decelerated within 14 days. Moreover, CMP coating facilitated the adhesion speed, spreading area, and focal adhesion formation of bone marrow stem cells. The number of cells in the active proliferating state and proliferated cells present on the CMP coating also increased. Additionally, CMP coating upregulated alkaline phosphatase activity and osteogenic gene expression in cells. In summary, the micro-arc oxidation assisted sol-gel CMP coatings increased the corrosion resistance and promoted the interfacial cell behavior for magnesium alloy implants, which might inform the further development of surface modifications on magnesium alloys for bone related applications.

Abstract:The rapid degradation rate of magnesium alloy affects its mechanical support as implant materials, which limits its application as medical implant materials. Therefore, it is of great significance to design a protective coating with low degradation rate and excellent biological activity. In this paper, a PLGA+MAO composite coating was prepared on a magnesium alloy. The long-period degradation behavior of PLGA+MAO coating in vivo and in vitro and the bone tissue growth were studied. The results have shown that the anti-corrosion performance of PLGA+MAO composite coating transcended that of MAO coating and uncoated Mg alloy both in vivo and in vitro . Obvious new bone growth was found around the PLGA+MAO coated implants in vivo . However, localized corrosion is still the main corrosion form once the PLGA+MAO coating is failed at some region, which could be an issue for the biocompatibility at the fast localized corrosion region and should be studied in the future.
