Abstract:This study aimed to explore the differences in the proteome and molecular pathways between two sampling locations (external, internal) of bovine Longissimus thoracis et lumborum (LTL) muscles at 0, 21, and 28 days of dry-aging (i.e. 3, 24, and 31 days post-mortem). It further assessed the impact of post-mortem aging on the meat proteome changes and the biological processes at interplay. Proteins related to defence response to bacterium and regulation of viral entry into host cell were identified to be more abundant on the external location before dry-aging, which may be associated to the oxidative conditions and microbial activity to which post-mortem muscle is exposed during dressing, chilling, and/or quartering of the carcasses. This highlights the relevance of sampling from interior tissues when searching for meat quality biomarkers. As dry-aging progressed, the meat proteome and related biological processes changed differently between sampling locations; proteins related to cell-cell adhesion and ATP metabolic processes pathways were revealed in the external location at 21 and 28 days, respectively. On the other hand, the impact of aging on the proteome of the interior meat samples, evidenced that muscle contraction and structure together with energy metabolism were the major pathways driving dry-aging. Additionally, aging impacted other pathways in the interior tissues, such as regulation of calcium import, neutrophil activation, and regeneration. Overall, the differences in the proteome allowed discriminating the three dry-aging times, regardless of the sampling location. Several proteins were proposed for validation as robust biomarkers to monitor the aging process (tenderization) of dry-aged beef: TTN, GRM4, EEF1A1, LDB3, CILP2, TNNT3, GAPDH, SERPINI1, and OMD.

Abstract:We describe the first case of sirolimus-induced drug fever in a female liver transplant recipient, with a history of hepatitis C-induced end-stage liver cirrhosis in 1999. In 2005, six years after transplantation, she developed calcineurin inhibitor-induced renal function impairment. Immunosuppression was switched from tacrolimus to sirolimus. Two days after the intake of sirolimus, she developed daily fever spikes, but no infectious focus was found. Antibiotic therapy had no influence on the fever. After fourteen days, sirolimus was switched back to tacrolimus and the fever disappeared. In history, the patient developed ciclosporin-induced generalized seizures eleven days after liver transplantation, followed by the development of a motoric speech disorder. Magnetic resonance imaging (MRI) findings were consistent with leucoencephalopathy, therefore immunosuppressive therapy was changed from ciclosporin to tacrolimus and the neurologic symptoms improved significantly. Our case is the first reported case of sirolimus-induced drug fever. In addition, the patient showed the rare occurrence of ciclosporin-induced leukencephalopathy with seizures.
Effects of relative humidity on dry-aged beef quality

Abstract:This study was conducted to evaluate the effects of relative humidity (RH) on moisture loss and flavor in dry-aged beef. Sixteen strip loins were assigned to one of the four aging treatments: vacuum (WET), dry-aging at 50% RH, dry-aging at 70% RH, or dry-aging at 85% RH and aged for 42 days at 2 °C. Loins were evaluated for evaporation loss, trim loss, tenderness, sensory, and microbiological characteristics. Results show that lower RH results in accelerated moisture loss during the first 3 days of the aging process without significantly affecting the total amount of moisture loss. Pseudomonadales dominated the aerobically dry-aged loins while Enterobacteriales was the most abundant in the wet-aged samples. Dry-aged samples had increased content of free amino acids in the cooked meat juice compared to the wet-aged counterpart. Dry aging at 50% RH tended to associate with more desirable flavor notes.
Effect of Dry Aging on Beef Muscle Proteins

Abstract:Meat is an inherent part of human diet. Its quality develops at different stages of production, storage, and processing. In this respect, the stage of aging is especially important. This technology makes it possible to regulate biochemical processes in meat raw materials. Long-term dry aging is a promising method that presupposes conditions that limit the growth of microorganisms. The transformations in the protein component are an important but understudied aspect of meat quality formation during dry aging. The research featured Hereford beef carcasses of Siberian breeding. The samples were isolated from the inner part of bone spinal-lumbar cuts after 21, 35, and 42 days of dry aging under the following conditions: 0–1°C, 74–75% relative humidity, 0.5 m/s air velocity. The samples were subjected to dry aging after 24 h at 4°C. The fractional composition of proteins was controlled by vertical electrophoresis in a Mini-Protean Tetra System chamber. The amino acid composition was defined by high-performance liquid chromatography in a Shimadzu LC-20 Prominence liquid chromatograph with a Shimadzu SPD20MA diode-matrix detector and a Kromasil C-18 separation column. The protein digestibility was measured by sequential exposure to pepsin-trypsin proteinase system under simulated gastric digestion. Long-time dry aging triggered proteolysis under the action of endogenous enzymes. The electropherogram analysis showed that the proteolytic changes in high-molecular myofibrillar proteins of high-quality beef became more pronounced after a longer maturation period. The distribution of protein fractions by dry aging stages indicated a different rate of degradation of contractile, regulatory, and cytoskeletal proteins. As a result, the structural integrity of muscle fibers degraded, the meat grew tender, and the proteins became more available to digestive enzymes. The amino acid and protein digestibility analyses in vitro demonstrated an increase in the nutritional value of beef and the availability of proteins to the action of proteinases after 42 days of dry aging. Long-term dry aging of high-quality beef increased the digestibility of muscle proteins as a result of proteolysis that accompanied the accumulation of low-molecular fractions. According to the amino acid analysis, the optimal result was most pronounced on day 42 as proven by the moderate oxidative changes in proteins.

Abstract:Beef dry-aging consists of a selection of unpackaged prime cuts placed in a controlled environment cold room for several weeks. The goals are to concentrate flavors like nutty and beefy and to improve tenderness. The aim of this study was to verify the microbiological and physicochemical behavior and sensory perception of meat during a sample process example of meat dry-aging. Twelve beef loins were selected for 90 days of dry aging and placed in a cold room with average temperature, relative humidity and forced air 3.2 ± 0.7 °C, 60.7 ± 4.2% and 0.5–2 m/s, respectively. Samples of crust and lean meat were collected on days 1, 14, 21, 35, 60 and 90 of the dry aging process for microbiological, physicochemical (pH, aw, color L*a*b*) and sensory analysis. During drying, no pathogenic bacteria were detected, and the average counts were higher for the crust. The average values for water activity (aw) and pH were 0.98 ± 0.02 and 5.77 ± 0.015, respectively. A slight decrease in aw and an increase in pH were observed over the aging period (p < 0.05). The lower microbial counts on the lean meat and the overall assessment of freshness confirm the importance of good manufacturing and storage practices during dry aging.

Abstract:Big Data Era in Sky and Earth Observation (BIG-SKY-EARTH, http://www.bigskyearth.eu) is a recently started COST Action that aims at setting the ground for a long-term networking between astronomy and remote sensing research communities in the area of Big Data utilization. COST Action is a networking funding scheme that allows researchers to jointly develop their ideas and initiatives in a given scientific field. Even though COST does not fund research itself, COST Actions are active through a range of research supporting networking tools, such as meetings, workshops, conferences, training schools, short-term scientific missions (short travels of researchers to foster their collaborations) and dissemination activities. Abstract:The retinofugal axons of Tachyglossus aculeatus were found to project to two diencephalic nuclei, disparate in size and location and designated provisionally as LGNa and b, and to the pretectum and superior colliculus. The primary optic pathways of the echidna are crossed except for a few fibers which pass to the ipsilateral large diencephalic nucleus (LGNa). Further work is required before the homologies of the two diencephalic nuclei can be determined. In addition, entirely crossed projections were also found to a large number of optic nuclei including a medial terminal nucleus, four cell groups interstitial to the optic tract, and a nucleus located within the brachium of the superior colliculus which may represent a dorsal terminal nucleus. The latter nucleus may, however, actually be the nucleus of the optic tract which is displaced from its usual close association with the pretectal area. Only an inferior fasciculus of the accessory optic tract was found.
Studying an effect of thermal treatment methods on physico-chemical properties of beef depending on aging technology

Abstract:Modern methods for aging and thermal treatment facilitate an improvement in beef sensory properties. The aim of the research was to study an effect of different types of thermal treatment on quality of steaks from dry-aged beef. Steaks were obtained from the rib cut from carcasses of grain-fed young bulls; aging duration was 35 days (temperature 0–1 oС, air relative humidity 74–75%). Aging duration in the control sample was five days (temperature 0–4 oС, air relative humidity 80–85%). The methods for thermal treatment were as follows: sous vide cooking at 58 oС, grill heating at 250 oС, sous vide cooking in combination with grill treatment. During processing, the following parameters were controlled: weight loss by the direct method, hydrophobicity of myofibrillar proteins by the reaction with bromophenol blue, soluble collagen by the method of acid hydrolysis with determination of oxyproline, mass fraction of moisture and fat by the method from one analytical unit, sensory properties by the reference method. According to the data obtained, minimal losses (6.8%) were noticed upon sous vide cooking of dry-aged beef steaks. In combined heating and dry heating on grill, they increased by 6.1% and 12%. More significant weight losses were observed in beef steaks with aging time of five days upon any tested treatment types. For dry-aged beef steaks treated by sous vide and combined heating, a higher content of soluble collagen compared to other samples was established. The results of measuring mass fraction of moisture and fat indicate an increase in the nutritional value of dry-aged beef steaks. According to the total data obtained with consideration for the results of sensory evaluation, dry-aged beef steaks subjected to sous vide cooking with the following short-term dry heating on grill have higher quality.
Effects of the Aging Period and Method on the Physicochemical, Microbiological and Rheological Characteristics of Two Cuts of Charolais Beef

Abstract:Wet-aging (WA) and dry-aging (DA) methods are usually used in the beef industry to satisfy the consumers' tastes; however, these methods are not suitable for all anatomical cuts. In this study, WA and DA were applied to improve the quality of two cuts of Charolais beef (Longissimus dorsi and Semitendinosus). For 60 days (i.e., 2 days, 15 days, 30 days and 60 days of sampling), a physicochemical, rheological, and microbiological analysis were performed at WA (vacuum packed; temperature of 4 ± 1 °C) and at DA (air velocity of 0.5 m/s; temperature of 1 ± 1 °C; relative humidity of 78 ± 10%) conditions. The results showed that the aging method influenced the aging loss (higher in the DA), cooking loss (higher in the WA), malondialdehyde concentration (higher in the DA) and fatty acid profile (few changes). No differences in the drip loss and color were observed, which decreased after 30 days of aging. The WBSF and TPA test values changed with increasing an aging time showing that the aging improved the tenderness of meat regardless of the aging method. Moreover, the aging method does not influence the microbiological profile. In conclusion, both WA and DA enhanced the quality of the different beef cuts, suggesting that an optimal method-time and aging combination could be pursued to reach the consumers' preferences.

Assessment of water relaxometry of meat under different ageing processes using time domain nuclear magnetic resonance relaxometry

Abstract:This study assessed water relaxometry of beef exposed to different ageing techniques by examining the inner and surface regions using time-domain nuclear magnetic resonance (TD-NMR) relaxometry. Beef strip loins were aged under vacuum (Wet), under vacuum using moisture absorbers (Abs), under vacuum using moisture absorbers and with mechanical tenderisation (AbsTend), or without any packaging (Dry). The ageing technique significantly influenced various meat parameters, including dehydration, total loss, and the moisture content of the meat surface. The transverse (T2) relaxation times provided a more sensitive indicator of the changes in meat water relaxometry than the longitudinal (T1) relaxation times. The Dry samples exhibited distinct differences in the T2 signals between the surface and inner regions of the meat. In particular, for the inner region, there were significant differences in signal areas between the Wet and Dry samples, and the Abs and AbsTend samples were positioned closely together between the Dry and Wet samples. The principal component analysis supported these findings: it indicated some differentiation among the ageing techniques in the score plot, but the differentiation was more pronounced when analysing the surface region. Additionally, there was a strong correlation between dehydration and the T2 values, leading to a clustering of the samples based on the ageing technique. The overlap between the Abs and AbsTend samples, situated between the Dry and Wet samples, suggests the potential of these treatments to produce meat with properties that are intermediate to Wet and Dry meat. Furthermore, tenderisation did not lead to greater dehydration.
[Family contact examination for tuberculosis--the current situations and perspective].

Abstract:To clarify the recent problems of family contact examination for tuberculosis, studies were conducted from August 1989 through to Mach 1990. 1) The examinations were carried out in 405 (94%) out of 431 household contacts of 119 index cases with bacillary pulmonary tuberculosis who were newly registered in 1988 in Yamagata prefecture. The contacts of smear-positive cases had a significantly higher incidence rate of active tuberculosis (4.8%) and a markedly shorter interval between discovery of the index cases and their household examinations than did the contacts of smear-negative cases (i.e. bacilli positive only on culture). 2) The study on the disease history of 129 cases with bacillary pulmonary tuberculosis, newly registered in 1988 in Yamagata, revealed that one-third of them might be have been "preventable", if the existing prevention and control methods had been effectively used. Especially, the preventable cases under 20 years of age occurred among those with inadequate family contact examinations. 3) 21 families were specially investigated because they had more than one active tuberculosis. Half of them underwent insufficient family contact examinations. It was indicated that tuberculin skin tests were necessary even to those aged 16 and above as well as to those aged 15 and below, when they were family contacts of smear-positive cases. 4) The policy and methods of family contact examination were evaluated by a mail questionnaire to 74 Health Centers in Tohoku Districts. Changes in Physico-Chemical and Storage Properties of Dry-Aged Beef Loin Using Electric Field Refrigeration System

Abstract:The aim of this study is to establish the dry aging period of beef loin in an electric field refrigeration system. Beef loins (Korea quality grade 2) were dry aged at 0, −1, and −2 °C temperature in an electric field refrigeration system (air velocity, 5 ± 2 m/s) and aging stopped as the value of TPC reached 7 log CFU/g. Samples were examined by aging yield, trimming yield, pH, color, water holding capacity (WHC), cooking yield, shear force, total plate count (TPC), 2-thiobarbituric acid reactive substances (TBARS), and volatile basic nitrogen (VBN). The results for aging yield, trimming yield, redness, yellowness, and chroma decreased with increasing the dry aging period. Contrariwise, those for pH, lightness, hue angle, WHC, and cooking yield increased with the dry aging period. In shear force, the lowest value occurred at four weeks at all temperatures. The results for TPC, TBARS, and VBN increased with aging period, and VBN at 6 weeks at 0 °C and 9 weeks at −1 °C exceed the standard value (20 mg/100 g), while dry aging temperature had an effect on physico-chemical and storage properties by lower temperatures showed slower progress. Therefore, dry aging on an electric field refrigerate system can be used until 4 weeks at 0 °C, 8 weeks at −1 °C, and 10 weeks at −2 °C. However, considering physico-chemical properties, 4 weeks at every temperature is suitable for manufacturing soft dry-aged beef loin.

Abstract:This study was performed to compare the differences in pH, myofibril fragmentation index (MFI), total protein solubility (TPS), sarcoplasmic protein solubility (SPS), myofibrillar protein solubility (MPS), and the microstructure of seven beef muscles during aging. From the six beef carcasses of Xinjiang brown cattle, a total of 252 samples from semitendinosus (ST), longissimus thoracis (LT), rhomboideus (RH), gastrocnemius (GN), infraspinatus (IN), psoas major (PM), and biceps femoris (BF) muscles were collected, portioned, and assigned to six aging periods (1, 3, 7, 9, 11, and 14 day/s) and 42 samples were used per storage period. IN muscle showed the highest pH (p < 0.05) from 1 to 14 days and the lowest TPS (p 0.05) as the aging period increased. The lowest TPS was found in the RH muscle on day 1, 3, and 7 and in the IN muscle on day 9, 11, and 14. The pH showed negative correlations with the MFI, TPS, and MPS (p < 0.01). The results suggest that changes in protein solubility and muscle fiber structure are related to muscle location in the carcass during aging. These results provide new insights to optimize the processing and storage of different beef muscles and enhance our understanding of the biological characteristics of Xinjiang brown cattle muscles.

Effect of Dry Maturation Time and the Curing Composition on Proteins in High Quality Beef

Abstract:Meat quality largely depends on the maturation conditions and natural biochemical processes that affect its taste, aroma, tenderness, and technological properties. Dry maturation and technological processing attract a lot of scientific attention. The research objective was to study the effect of dry maturation time on the physicochemical parameters of high-quality beef, as well as the effect of a new curing mix on the properties of raw materials. The research featured dorsal-lumbar cuts of high-quality beef on maturation days 21 and 40. Matured meat was tested for the main components, the solubility of sarcoplasmic and myofibrillar proteins, the hydrophobicity of myofibrillar proteins, protein oxidation products, and the activity of catalase and peroxidase. The chemical composition was evaluated depending on the dry maturation time using the arbitration method. The solubility of proteins was determined by the calorimetric method with a biuret reagent. The hydrophobicity of myofibrillar proteins was determined by bromophenol blue reaction, and the activity of catalase and peroxidase was determined by standard methods. Proteins were extracted with potassium phosphate buffer (pH 7.2), while myofibrillar proteins were extracted with Tris-HCl and KCl buffers at pH 7.5 and 7.0. The meat samples were deboned and salted using sodium chloride and a combined mix of 70% magnesium chloride and 30% sodium chloride. The solubility of myofibrillar proteins on day 21 increased by 23.95% but decreased by 14.1% by day 40. The solubility of sarcoplasmic proteins decreased continuously (22.10 and 31.12%, respectively). The obtained data matched the hydrophobicity of proteins. Dry maturation initiated protein oxidation, as demonstrated by carbonyl and sulfhydryl groups of myofibrillar proteins on maturation day 40 (27.85 nmol/L and 27.3 μmol/g of protein, respectively). Sodium chloride and its mix increased the extractability of proteins by 5.2 and 6.9% on day 21 and by 6.8 and 10.6% on day 40 but triggered protein oxidation. Muscle proteins of high-quality beef proved functional after 21 days of dry maturation. The new mix with reduced sodium content can be recommended for high-quality dry-aged beef production.

Abstract:The objective of this study was to determine the effect of aging/freezing sequence on meat quality, oxidative stability and biochemical attributes of beef muscles. At 3 days postmortem, Longissimus lumborum and Semitendinosus muscles were obtained from 10 beef carcasses, cut into 3 sections and vacuum-packaged. The sections were randomly assigned to aging/freezing treatments (aging only, aging then freezing/thawing, and freezing then thaw/aging). Freezing first then-thaw/aging showed more enlarged gaps between muscle fibers and widely opened extracellular drip channels, resulting in more purge/exudate loss compared to other treatments (P < .05). No differences in other meat quality attributes (e.g. shear force, color and lipid oxidative stability) were found between the aging/freezing sequence treatments (P > .05). A greater desmin degradation was observed in both freezing treatments, while troponin-T degradation was not affected (P > .05). The results suggest that freezing (and aging) itself would be the critical factor affecting those quality attributes of frozen/thawed meat rather than the sequence of aging/freezing.

In this study, patients with resected stage III adenocarcinoma of the colon were assigned to treatment with fluorouracil (FU) plus leucovorin (LV; weekly 6, every 8 weeks for four cycles) or FU plus LV plus irinotecan (CPT-11; weekly 4, every 6 weeks for five cycles). This welldesigned and well-conducted clinical trial by the publicly funded National Cancer Institute (Bethesda, MD) cooperative group system clearly demonstrated that CPT-11 plus FU plus LV failed to improve disease-free, recurrence-free, or overall survival compared with FU plus LV. These results were unexpected. After all, CPT-11 had previously shown clear activity in patients with metastatic cancer—the accepted proving ground for subsequent adjuvant therapies. In 1996, CPT-11 was granted accelerated US Food and Drug Administration approval based on phase II single-agent activity in patients with metastatic colorectal cancer refractory to FU. Full approval was granted approximately 2 years later based on randomized phase III trials that demonstrated a modest (2 to 3 months) survival benefit from CPT-11 compared with best supportive care, or to infusional FU in patients who had previously received bolus FU. Subsequent phase III studies showed that CPT-11 improved survival when added to FU plus LV compared with FU plus LV in the initial management of patients with metastatic colorectal cancer. One of these CPT-11 regimens, CPT-11 plus FU plus LV, was selected as the experimental arm of CALGB 89803. We may consider several potential explanations for the negative results of CALGB 89803. One might initially question whether the weekly bolus CPT-11 plus FU plus LV regimen was an optimal choice for the experimental arm. At the time that CALGB 89803 was designed, the CPT-11 plus FU plus LV regimen was the standard in North America FU plus LV plus CPT-11 regimen in North America. During the study, toxicity concerns surfaced, including an excess of toxic deaths on the experimental arm. It would be useful to know whether there was a substantial imbalance in scheduled therapy delivered between the study conditions that might help explain the outcomes. In fact, Saltz et al note that nearly twice as many patients withdrew consent or discontinued treatment on the CPT-11 plus FU plus LV arm than in the control group. Subsequent to this trial, data emerged to suggest that infusional FU delivery has a superior therapeutic index in combination with CPT-11. However, preliminary results of two additional adjuvant studies comparing infusional FU plus LV to infusional FU plus LV plus CPT-11 also failed to demonstrate superiority of the CPT-11– containing arms. Therefore, a demonstrable lack of activity for CPT-11 in the adjuvant setting has been consistently observed. These data must be interpreted in the context of other studies that demonstrated improved recurrence-free survival when oxaliplatin is added to a FU plus LV backbone as adjuvant therapy. The benefit of CPT-11 in patients with metastatic disease in terms of response rate and progression is similar to that observed with oxaliplatin. In addition, two studies comparing infusional FU plus oxaliplatin to infusional FU plus CPT-11 as initial therapy in patients with metastatic colorectal cancer showed equivalent response rates, time to progression, and overall survival between the study arms. Thus, one may have reasonably expected similar results in the adjuvant setting. However, these advanced disease studies comparing oxaliplatin and CPT-11 were of moderate size and were not powered to demonstrate equivalency. The trial reported by Tournigand et al included 220 patients and was designed to have 80% power to detect a 20% difference in the proportion of patients without progression at 15 months. Similarly, Colucci et al enrolled 336 patients with response rate as the primary objective, with statistical power to demonstrate a 15% difference between the arms. Thus, it is plausible that there are subtle differences in the clinical activity of CPT-11 and oxaliplatin that were not observed in underpowered trials in patients with metastatic disease, but became manifest in much larger trials in the adjuvant setting. Furthermore, patient preferences and tolerance of specific toxicities that differ between regimens (such as alopecia or hospitalization rate) might become more apparent in the adjuvant setting where premature treatment discontinuation could affect long-term outcomes. Underlying successful application of adjuvant therapy is the hypothesis that microscopic metastatic disease exists and can be eradicated with systemic treatment. Another possible explanation for the findings of CALGB 89803, and other adjuvant studies of CPT-11 in combination with FU plus LV, is that mechanisms of CPT-11 resistance might be expressed in micrometastases to a greater extent than in macrometastatic colon cancer. Abstract:To speed up the migration of moisture during air drying under common condition (35°C, 60% relative humidity, 3 m/s wind speed), the relationship between crust formation and moisture migration was investigated. The results showed that shear force was inversely proportional to moisture content, meanwhile moisture content in surface decreased more quickly than that in the inner part of lamb cubes during air drying (p < .05). This indicated that the crust of meat surface was due to quicker decline of moisture content. Light microscopy showed that, the formation of crust and decrease of crust's porosity with drying was partly due to shrinkage of the connective tissue. This study concluded that changing the shrinkage characteristics of connective tissue might be a way to increase the meat porosity, influence the water migration, and shorten the drying time. Practical applicationsJerky processing, especially drying of it, is time and energy consuming. The aim of this article was to find the relationship between crust and water migration during air drying. We found that changing the shrinkage characteristics of connective tissue might be a way to increase the meat porosity, influence the water migration, and shorten the drying time. This study provides some ideas for shortening the drying time of jerky.
