Abstract:As a simple but beautiful model, the Drosophila larva has been extensively studied to investigate the sensorimotor mechanism by analyzing quantitatively measured behaviors. The current behavior descriptive methods are usually based on the movements of three oversimplified landmarks of the larval body, including the head, tail, and centroid, failing to reveal rich information of multisegment motion. Due to the highly elastic body shapes and sophisticated locomotion, it is challenging to extract body segment movements of a larva to develop a segment-based behavior model. Here, we propose a landmark regression method to automatically detect all the segment joint points of the larva, which can save massive manual efforts. A cascaded regression process is used to precisely estimate segment joint points in each video frame and obtain the detailed segment movement parameters. To train our model, we propose a novel larval body segment dataset, which consists of 3772 images, each image is annotated with 22 segment points. Our method could achieve 99.51% accuracy on the test dataset, which is accurate enough to process typical movement patterns in behavioral experiments for neural coding studies.

Abstract:This paper presented a novel tinny motion capture system for measuring bird posture based on inertial and magnetic measurement units that are made up of micromachined gyroscopes, accelerometers, and magnetometers. Multiple quaternion-based extended Kalman filters were implemented to estimate the absolute orientations to achieve high accuracy.Under the guidance of ornithology experts, the extending/contracting motions and flapping cycles were recorded using the developed motion capture system, and the orientation of each bone was also analyzed. The captured flapping gesture of the Falco peregrinus is crucial to the motion database of raptors as well as the bionic design.
Active vision shapes and coordinates flight motor responses in flies

Abstract:Animals use active sensing to respond to sensory inputs and guide future motor decisions. In flight, flies generate a pattern of head and body movements to stabilize gaze. How the brain relays visual information to control head and body movements and how active head movements influence downstream motor control remains elusive. Using a control theoretic framework, we studied the optomotor gaze stabilization reflex in tethered flight and quantified how head movements stabilize visual motion and shape wing steering efforts in fruit flies (Drosophila). By shaping visual inputs, head movements increased the gain of wing steering responses and coordination between stimulus and wings, pointing to a tight coupling between head and wing movements. Head movements followed the visual stimulus in as little as 10 ms-a delay similar to the human vestibulo-ocular reflex-whereas wing steering responses lagged by more than 40 ms. This timing difference suggests a temporal order in the flow of visual information such that the head filters visual information eliciting downstream wing steering responses. Head fixation significantly decreased the mechanical power generated by the flight motor by reducing wingbeat frequency and overall thrust. By simulating an elementary motion detector array, we show that head movements shift the effective visual input dynamic range onto the sensitivity optimum of the motion vision pathway. Taken together, our results reveal a transformative influence of active vision on flight motor responses in flies. Our work provides a framework for understanding how to coordinate moving sensors on a moving body.
Phosphorylation-Independent Inhibition of Cdc28p by the Tyrosine Kinase Swe1p in the Morphogenesis Checkpoint

Abstract:ABSTRACT The morphogenesis checkpoint in budding yeast delays cell cycle progression in G2 when the actin cytoskeleton is perturbed, providing time for cells to complete bud formation prior to mitosis. Checkpoint-induced G2 arrest involves the inhibition of the master cell cycle regulatory cyclin-dependent kinase, Cdc28p, by the Wee1 family kinase Swe1p. Results of experiments using a nonphosphorylatable CDC28Y19F allele suggested that the checkpoint stimulated two inhibitory pathways, one that promoted phosphorylation at tyrosine 19 (Y19) and a poorly characterized second pathway that did not require Cdc28p Y19 phosphorylation. We present the results from a genetic screen for checkpoint-defective mutants that led to the repeated isolation of the dominant CDC28E12K allele that is resistant to Swe1p-mediated inhibition. Comparison of this allele with the nonphosphorylatable CDC28Y19F allele suggested that Swe1p is still able to inhibit CDC28Y19F in a phosphorylation-independent manner and that both the Y19 phosphorylation-dependent and -independent checkpoint pathways in fact reflect Swe1p inhibition of Cdc28p. Remarkably, we found that a Swe1p mutant lacking catalytic activity could significantly delay the cell cycle in vivo during a physiological checkpoint response, even when expressed at single copy. The finding that a Wee1 family kinase expressed at physiological levels can inhibit a nonphosphorylatable cyclin-dependent kinase has broad implications for many checkpoint studies using such mutants in other organisms.

Abstract:How muscle actions are coordinated to realize animal movement is a fundamental question in behavioral study. To obtain the overall muscular activity patterns accompanying behaviors at high spatiotemporal resolution is technically difficult. In this work, we used light sheet microscopy to simultaneously image and analyze the activity, length and orientation of Drosophila larval muscles across body segments at single muscle resolution in nearly free behaviors. For typical behavioral modes such as peristalsis, head cast and turning, larval muscles showed behavioral mode specific activity patterns. Unexpectedly, reorientation of larval head involves muscle tone in the apparently motionless posterior segments. With a STGCN(spatial temporal graph convolution neural network)-Generator model, sequence of larval behavioral poses outlined by morphological patterns of muscles could be accurately predicted based on the time series of ventral but not dorsal muscle activities, and vice versa. Laser ablation of ventral but not dorsal muscles interrupted peristaltic wave and undermined head cast in both frequency and amplitude. Our results provide a simplified muscle activity representation of soft body motion that can be used for probing the key components of animal motor control. ### Competing Interest Statement The authors have declared no competing interest.
A visuomotor circuit for evasive flight turns in Drosophila

Abstract:Visual systems extract multiple features from a scene using parallel neural circuits. Ultimately, the separate neural signals must come together to coherently influence action. Here, we characterize a circuit in Drosophila that integrates multiple visual features related to imminent threats to drive evasive locomotor turns. We identified, using genetic perturbation methods, a pair of visual projection neurons (LPLC2) and descending neurons (DNp06) that underlie evasive flight turns in response to laterally moving or approaching visual objects. Using two-photon calcium imaging or whole-cell patch clamping, we show that these cells indeed respond to both translating and approaching visual patterns. Furthermore, by measuring visual responses of LPLC2 neurons after genetically silencing presynaptic motion-sensing neurons, we show that their visual properties emerge by integrating multiple visual features across two early visual structures: the lobula and the lobula plate. This study highlights a clear example of how distinct visual signals converge on a single class of visual neurons and then activate premotor neurons to drive action, revealing a concise visuomotor pathway for evasive flight maneuvers in Drosophila.

Abstract:Discovering principles underlying the control of animal behavior requires a tight dialogue between experiments and neuromechanical models. Until now, such models, including NeuroMechFly for the adult fly, Drosophila melanogaster, have primarily been used to investigate motor control. Far less studied with realistic body models is how the brain and motor systems work together to perform hierarchical sensorimotor control. Here we present NeuroMechFly v2, a framework that expands Drosophila neuromechanical modeling by enabling visual and olfactory sensing, ascending motor feedback, and complex terrains that can be navigated using leg adhesion. We illustrate its capabilities by first constructing biologically inspired locomotor controllers that use ascending motor feedback to perform path integration and head stabilization. Then, we add visual and olfactory sensing to this controller and train it using reinforcement learning to perform a multi-modal navigation task in closed loop. Finally, we illustrate more biorealistic modeling in two ways: our model navigates a complex odor plume using a Drosophila odor taxis strategy, and it uses a connectome-constrained visual system network to follow another simulated fly. With this framework, NeuroMechFly can be used to accelerate the discovery of explanatory models of the nervous system and to develop machine learning-based controllers for autonomous artificial agents and robots.

Abstract:Vision influences behavior, but ongoing behavior also modulates vision in animals ranging from insects to primates. The function and biophysical mechanisms of most such modulations remain unresolved. Here, we combine behavioral genetics, electrophysiology, and high-speed videography to advance a function for behavioral modulations of visual processing in Drosophila. We argue that a set of motion-sensitive visual neurons regulate gaze-stabilizing head movements. We describe how, during flight turns, Drosophila perform a set of head movements that require silencing their gaze-stability reflexes along the primary rotation axis of the turn. Consistent with this behavioral requirement, we find pervasive motor-related inputs to the visual neurons, which quantitatively silence their predicted visual responses to rotations around the relevant axis while preserving sensitivity around other axes. This work proposes a function for a behavioral modulation of visual processing and illustrates how the brain can remove one sensory signal from a circuit carrying multiple related signals.
High-resolution, high-throughput analysis of Drosophila geotactic behavior

Abstract:Drosophila innate response to gravity, geotaxis, has been previously used to assess the impact of aging and disease on motor performance. Despite its rich history, fly geotaxis continues to be largely measured manually and assessed through simplistic metrics. The manual nature of this assay introduces substantial experimental variability while simplistic metrics provide limited analytic insights into the behavior. To address these shortcomings, we have constructed a fully automated, programable apparatus, and developed a multi-object tracking software capable of following sub-second movements of individual flies, thus allowing reproducible, detailed, and quantitative analysis of geotactic behavior. The apparatus triggers and monitors geotaxis of 10 fly cohorts simultaneously, with each cohort consisting of up to 7 flies. The tracking program isolates cohorts and records individual fly coordinate outputs allowing for simultaneous multi-group, multi-fly tracks per experiment, greatly improving throughput and resolution. The algorithm tracks individual flies during the entire run with ~97% accuracy, yielding detailed climbing curve, speed, and movement direction with 1/30 second resolution. Our tracking also allows the construction of multi-variable metrics and the detection of transitory movement phenotypes, such as slips and falls, which have thus far been neglected in geotaxis studies due to limited spatio-temporal resolution. Through a combination of automation and robust tracking, the platform is therefore poised to advance Drosophila geotaxis assay into a comprehensive assessment of locomotor behavior

Abstract:Biological sensors have evolved to act as matched filters that respond preferentially to the stimuli they expect to receive during ecologically relevant tasks [ – ]. For instance, insect visual systems are tuned to detect stimuli ranging from the small-target motion of mates [ , ] or prey [ , ] to the polarization pattern of the sky [ , ]. In flies, individually identified neurons called lobula plate tangential cells respond to optic flow fields matched to specific self-motions [ , ], forming the output layer of what is presently nature’s best-understood deep convolutional neural network [ – ]. But what functional principle does their tuning embed, and how does this aid motor control? Here we test the hypothesis that evolution co-tunes physics and physiology by aligning the preferred directions of an animal’s sensors to the most dynamically-significant directions of its motor system [ ]. We build a state-space model of blowfly flight by combining visual electrophysiology, synchrotron-based X-ray microtomography, high-speed videogrammetry, and computational fluid dynamics. We then apply control-theoretic tools to show that the tuning of the fly’s widefield motion vision system maximizes the flow of energy from control inputs and disturbances to sensor outputs, rather than optimizing state estimation as is the conventional approach to sensor placement in engineering [ , ]. We expect the same functional principle to apply across sensorimotor systems in other organisms, with implications for the design of novel control architectures for robotic systems combining high performance with low computational load and low power consumption.

Behavioral signatures of structured feature detection during courtship in Drosophila

Abstract:Many animals detect other individuals effortlessly. In Drosophila, previous studies have examined sensory processing during social interactions using simple blobs as visual stimulation; however, whether and how flies extract higher-order features from conspecifics to guide behavior remains elusive. Arguing that this should be reflected in sensorimotor relations, we developed unbiased machine learning tools for natural behavior quantification and applied these tools, which may prove broadly useful, to study interacting pairs. By transforming motor patterns with female-centered reference frames, we established circling, where heading and traveling directions intersect, as a unique pattern of social interaction during courtship. We found circling to be highly visual, with males exhibiting view-tuned motor patterns. Interestingly, males select specific wing and leg actions based on the positions and motions of the females' heads and tails. Using system identification, we derived visuomotor transformation functions indicating history-dependent action selection, with distance predicting action initiation and angular position predicting wing choices and locomotion directions. Integration of vision with somatosensation further boosts these sensorimotor relations. Essentially comprised of orchestrated wing and leg maneuvers that are more variable in the light, circling induces mutually synchronized conspecific responses stronger than wing extension alone. Finally, we found that actions depend on integrating spatiotemporally structured features with goals. Altogether, we identified a series of sensorimotor relations during circling, implying that during courtship, flies detect complex spatiotemporally structured features of conspecifics, laying the foundation for a mechanistic understanding of conspecific recognition in Drosophila.

Abstract:Embryonic stem cells (ESCs) are established from the inner cell mass of preimplantation embryos, are capable of self‐renewal, and exhibit pluripotency. Given these unique properties, ESCs are expected to have therapeutic potential in regenerative medicine and as a powerful tool for in vitro differentiation studies of stem cells. Various growth factors and extracellular matrix components regulate the pluripotency and differentiation of ESC progenies. Thus, the cell surface receptors that bind these regulatory factors are crucial for the precise regulation of stem cells. To identify membrane proteins that are involved in the regulation of pluripotent stem cells, the membrane proteins of murine ESCs cultured with or without leukemia inhibitory factor (LIF) were purified and analyzed by quantitative proteomics. 2‐D PAGE‐based analysis using fluorescently labeled proteins and shotgun‐based analysis with isotope‐labeled peptides identified 338 proteins, including transmembrane, membrane‐binding, and extracellular proteins, which were expressed specifically in pluripotent or differentiated murine ESCs. Functions of the identified proteins revealed cell adhesion molecules, channels, and receptors, which are expected to play important roles in the maintenance of murine ESC pluripotency. Membrane proteins that are expressed in pluripotent ESCs but not in differentiated cells such as Slc16a1 and Bsg could be useful for the selection of the stem cells in vitro.
State-dependent decoupling of sensory and motor circuits underlies behavioral flexibility in Drosophila

Abstract:An approaching predator and self-motion toward an object can generate similar looming patterns on the retina, but these situations demand different rapid responses. How central circuits flexibly process visual cues to activate appropriate, fast motor pathways remains unclear. Here we identify two descending neuron (DN) types that control landing and contribute to visuomotor flexibility in Drosophila. For each, silencing impairs visually evoked landing, activation drives landing, and spike rate determines leg extension amplitude. Critically, visual responses of both DNs are severely attenuated during non-flight periods, effectively decoupling visual stimuli from the landing motor pathway when landing is inappropriate. The flight-dependence mechanism differs between DN types. Octopamine exposure mimics flight effects in one, whereas the other probably receives neuronal feedback from flight motor circuits. Thus, this sensorimotor flexibility arises from distinct mechanisms for gating action-specific descending pathways, such that sensory and motor networks are coupled or decoupled according to the behavioral state.

Abstract:Tracking visual objects while maintaining stable gaze is complicated by the different computational requirements for figure-ground discrimination, and the distinct behaviors that these computations coordinate. Drosophila melanogaster uses smooth optomotor head and body movements to stabilize gaze, and impulsive saccades to pursue elongated vertical bars. Directionally selective motion detectors T4 and T5 cells provide inputs to large-field neurons in the lobula plate, which control optomotor gaze stabilization behavior. Here, we hypothesized that an anatomically parallel pathway represented by T3 cells, which provide inputs to the lobula, drives bar tracking body saccades. We combined physiological and behavioral experiments to show that T3 neurons respond omnidirectionally to the same visual stimuli that elicit bar tracking saccades, silencing T3 reduced the frequency of tracking saccades, and optogenetic manipulation of T3 acted on the saccade rate in a push-pull manner. Manipulating T3 did not affect smooth optomotor responses to large-field motion. Our results show that parallel neural pathways coordinate smooth gaze stabilization and saccadic bar tracking behavior during flight.
Inhibitory control of locomotor statistics in walking Drosophila

Abstract:In order to forage for food, many animals regulate not only specific limb movements but the statistics of locomotor behavior over time, switching between long-range dispersal and localized search depending on resource availability. How pre-motor circuits regulate such locomotor statistics is not clear. Here we analyze and model locomotor statistics in walking Drosophila, and their modulation by attractive food odor. Odor evokes three motor regimes in flies: baseline walking, upwind running during odor, and search behavior following odor loss. During search behavior, we find that flies adopt higher angular velocities and slower ground speeds, and tend to turn for longer periods of time in one direction. We further find that flies spontaneously adopt periods of different mean ground speed, and that these changes in state influence the length of odor-evoked runs. We next developed a simple model of neural locomotor control that suggests that contralateral inhibition plays a key role in regulating the statistical features of locomotion. As the fly connectome predicts decussating inhibitory neurons in the lateral accessory lobe (LAL), a pre-motor structure, we gained genetic access to a subset of these neurons and tested their effects on behavior. We identified one population of neurons whose activation induces all three signature of search and that bi-directionally regulates angular velocity at odor offset. We identified a second group of neurons, including a single LAL neuron pair, that bi-directionally regulate ground speed. Together, our work develops a biologically plausible computational architecture that captures the statistical features of fly locomotion across behavioral states and identifies potential neural substrates of these computations.

Abstract:The motions that make up animal behavior arise from the interplay between neural circuits and the mechanical parts of the body. Therefore, in order to comprehend the operational mechanisms governing behavior, it is essential to examine not only the underlying neural network but also the mechanical characteristics of the animal's body. The locomotor system of fly larvae serves as an ideal model for pursuing this integrative approach. By virtue of diverse investigation methods encompassing connectomics analysis and quantification of locomotion kinematics, research on larval locomotion has shed light on the underlying mechanisms of animal behavior. These studies have elucidated the roles of interneurons in coordinating muscle activities within and between segments, as well as the neural circuits responsible for exploration. This review aims to provide an overview of recent research on the neuromechanics of animal locomotion in fly larvae. We also briefly review interspecific diversity in fly larval locomotion and explore the latest advancements in soft robots inspired by larval locomotion. The integrative analysis of animal behavior using fly larvae could establish a practical framework for scrutinizing the behavior of other animal species.

Flies tune the sensitivity of their multifunctional gyroscope

Abstract:Locomotion requires navigating unpredictable and complex environments, demanding both stability and maneuverability within short timeframes. This is particularly important for flying insects, and the true flies (Diptera) stand out among this group for their impressive flight capabilities. Flies’ aerial abilities are partially attributed to halteres, tiny club-shaped structures that evolved from the hindwings and play a crucial role in flight control. Halteres oscillate during flight, in antiphase with the wings, providing rhythmic input to the wing steering system arrays of embedded mechanosensors called campaniform sensilla. These sensor arrays convey timing information to the wing steering muscles, but linking haltere sensor location to sensor activity and the functional organization of the wing steering system remains a central challenge. Here, we use calcium imaging during tethered flight to obtain population-level recordings of the haltere sensory afferents in specific fields of sensilla. We find that haltere feedback is continuously modulated by visual stimuli to stabilize flight. Additionally, this feedback is present during saccades and help flies actively maneuver. We also find that the haltere’s multifaceted role arises from the steering muscles of the haltere itself, regulating haltere stroke amplitude to modulate campaniform activity. Taken together, our results underscore the crucial role of biomechanics in regulating the dynamic range of sensors and provide insight into how the sensory and motor systems of flies coevolved.

Abstract:Flying insects exhibit remarkable navigational abilities controlled by their compact nervous systems. Optic flow, the pattern of changes in the visual scene induced by locomotion, is a crucial sensory cue for robust self-motion estimation, especially during rapid flight. Neurons that respond to specific, large-field optic flow patterns have been studied for decades, primarily in large flies, such as houseflies, blowflies, and hover flies. The best-known optic-flow sensitive neurons are the large tangential cells of the dipteran lobula plate, whose visual-motion responses, and to a lesser extent, their morphology, have been explored using single-neuron neurophysiology. Most of these studies have focused on the large, Horizontal and Vertical System neurons, yet the lobula plate houses a much larger set of 'optic-flow' sensitive neurons, many of which have been challenging to unambiguously identify or to reliably target for functional studies. Here we report the comprehensive reconstruction and identification of the Lobula Plate Tangential Neurons in an Electron Microscopy (EM) volume of a whole Drosophila brain. This catalog of 58 LPT neurons (per brain hemisphere) contains many neurons that are described here for the first time and provides a basis for systematic investigation of the circuitry linking self-motion to locomotion control. Leveraging computational anatomy methods, we estimated the visual motion receptive fields of these neurons and compared their tuning to the visual consequence of body rotations and translational movements. We also matched these neurons, in most cases on a one-for-one basis, to stochastically labeled cells in genetic driver lines, to the mirror-symmetric neurons in the same EM brain volume, and to neurons in an additional EM data set. Using cell matches across data sets, we analyzed the integration of optic flow patterns by neurons downstream of the LPTs and find that most central brain neurons establish sharper selectivity for global optic flow patterns than their input neurons. Furthermore, we found that self-motion information extracted from optic flow is processed in distinct regions of the central brain, pointing to diverse foci for the generation of visual behaviors.
