Abstract:Waterborne pathogens have been found in biofilms grown in drinking water distribution system (DWDS). However, there is a lack of quantitative study on the culturability of pathogens in biofilms from metropolitan DWDS. In this study, we quantified culturable and viable but non-culturable (VBNC) Escherichia coli, Salmonella enterica, Pseudomonas aeruginosa and Vibrio cholerae in biofilms collected from five kinds of pipes (galvanized steel pipe, steel pipe, stainless steel clad pipe, ductile cast iron pipe and polyethylene pipe) and associated drinking water at an actual chlorinated DWDS in use from China. The results of these comprehensive analyses revealed that pipe material is a significant factor influencing the culturability of pathogen and microbial communities. Network analysis of the culturable pathogens and 16S rRNA gene inferred potential interactions between microbiome and culturability of pathogens. Although the water quality met the Chinese national standard of drinking water, however, VBNC pathogens were detected in both biofilms and water from the DWDS. This investigation suggests that stainless steel clad pipe (SSCP) was a better choice for pathogen control compared with other metal pipes. To our knowledge, this is the first study on culturable and VBNC pathogens in biofilms of different pipe materials in metropolitan DWDS.
Radial distribution shifts of microbial communities in biofilms developed under stagnation condition in water supply pipes

Abstract:Subject depends on the pilot platform in actual water supply system to conduct research in order to analyze radial distribution shifts of microbial communities and numbers of total bacteria in biofilms developed under stagnation condition in water supply pipes.Scanning electron microscope (SEM) was used to observe biofilms morphology.Biofilm community in different spatial pipe locations was collected and systematically investigated using the high-throughput sequencing of 16S rRNA gene by real-time fluorescence quantitative (RT-PCR) to count total number of bacteria in the biofilm.Results showed that the increase of total bacteria in the upper of pipe was little, but in the middle and bottom was significant.Radial distribution of microbial communities in the high density polyethylene pipe (HDPE) inner wall was uniform and the content changed little under stagnation condition, but as for ductile iron pipe(DCIP), the microbial communities increased sequentially from top to bottom and the change was distinct.The content of microbial community in the middle of pipe was dramatically changing, and the effect of microbial community in the region on pipe corrosion should be concerned.
Characteristics of Biostability of Drinking Water in Aged Pipes after Water Source Switching: ATP Evaluation, Biofilms Niches and Microbial Community Transition

Abstract:Delivering quality-changed water often contributes to the biological instability of drinking water distribution systems (DWDS). However, the potential effects of quality-changed water on the biostability within DWDS are not well understood, especially after water switching to quality-improved water. The objective of this study was to investigate the effects of quality-improved water on DWDS, focusing on the stability of biofilm. The practical aged-pipe was assembled into pipe reactors to simulate the effect of switching to quality-improve water. The adenosine triphosphate (ATP) concentration of bulk water in the pipe reactors increased from ∼1.2 ng/L to almost above 5 ng/L when fed water switching to TP 2. Biomass quantified by measuring ATP concentration confirmed that the risk of biofilm release through aged cast-iron (CI) pipe surfaces after water source switching. The changes in water characteristics due to quality-improved water source could cause bacteria release in DWDS at the initial period (at the first 7 days). However, the DWDS can establish the new stable phase after 42 days. Over time, biomass in the bulk water of the distribution system decreased significantly (The ATP concentration in the bulk maintains around 3 ng/L) after 42 days, indicating the improvement of water quality. The biofilm was dominated by bacteria related to iron-cycling process, and at the genus level, Desulfovibrio had the highest relative abundance, however, it decreased significantly (from 48% to 9.3%) after water source switching. And there was a slightly increase in the fraction of iron-oxidizing bacteria (IOB) and siderophore-producing bacteria (SPB), but a relatively higher increase in nitrate-reducing bacteria (NRB), nitrobacteria (NOB), and iron-reducing bacteria (IRB) was observed. Taken together, these results and the corrosion morphology, indicate that pipe biofilm and corrosion were chemically and microbially stable after re-stability under water source switching. In addition, the bulk water environment showed a marked decrease in selected bacteria at genus level, including pathogenic species, indicating the improvement of quality in drinking water.

Abstract:The gradual updating of the water supply network is one of the crucial ways to ensure the safety of drinking water all over the world. The phenomenon and regularity of the biological risk and chemical risk of biofilms of the new pipes in drinking water distribution system (DWDS) is inadequate researched by now. In order to explore the biochemical risks of biofilms after new pipes are used, this paper studied the growth of biofilms, the content of disinfection by-products (DBPs) and the potential for disinfection by-products (DBPsFP) after 2-year use by establishing a pilot test platform at both ends of the DWDS in City S. The results showed that the total bacterial count in new pipelines was between 1.38 x 10(8)-9.97 x 10(8)/cm(2); the DBPsFP at the front end and at the back end was subtly different. The overall DBPsFP of biofilms was the highest, followed by the ductile cast iron pipe and the galvanized pipe whereas the stainless steel pipe was the lowest. The HPC content of the 2-yearold pipe (1.68 x 10(5)-7.09 x 10(6) CFU/cm(2)) was remarkably higher than that of the 1-year-old pipe (1.04 x 10(5)-8.76 x 10(5) CFU/cm(2)), and the generation DBPsFP was about 50% higher. When a new pipeline was put into use in the urban drinking water distribution system, biofilms with certain biological hazards and risks of DBPs disinfection by-products would form in a short period of time.

Abstract:Water quality deterioration of drinking water distribution systems (DWDSs) caused by water source switching has been reported previously. However, systematic investigation of the biostability of DWDS under water source switching is limited. Aged pipes, including three commonly used pipe materials dug out from a practical DWDS, were used to systematically investigate the biofilm stability mechanism of DWDS under water source switching to quality-improved water. An increase in adenosine triphosphate (ATP) concentration in the bulk water during the initial stage of the switching period was observed, indicating the risk of biofilm release through aged pipe surfaces after water source switching. Sloughing of biofilms might contribute to temporary instability. From day 35, the ATP concentration in the polyethylene (PE) and plastic stainless steel composite (PS) pipes were maintained at approximately 2.40 and 2.56 ng/L, respectively. In contrast, the ATP concentration in the ductile iron (DI) pipes was higher, at approximately 3.43 ng/L from day 42. The water quality variation could cause areas of the biofilm to slough and reduce the biomass of biofilm, causing partial alteration of the microbial community. 16S rRNA gene amplicon sequencing–based functional prediction revealed that the biofilm could increase the abundance of chlorine-resistant bacteria attributed to the increase in Pseudomonas and Methylobacterium after switching to quality-improved water. Moreover, the profiles of specific pathways linked to human diseases, antibiotic resistance, and antibiotic biosynthesis revealed that the safety of the biofilm could improve after switching to quality-improved water. • The PE and PS biofilm showed improved resistance to water quality perturbation. • Greater number of Methylobacterium was found in the biofilm after water source switching. • 3.16S gene–based metagenomics prediction revealed that the safety of the biofilm under water source switching.
Niche Differentiation of Biofilm Microorganisms in a Full-scale Municipal Drinking Water Distribution System in China and Their Implication for Biofilm Control

Abstract:Biofilms on the inner surface of a drinking water distribution system (DWDS) affect water quality and stability. Understanding the niche differentiation of biofilm microbial communities is necessary for the efficient control of DWDS biofilms. However, biofilm studies are difficult to conduct in the actual DWDS because of inaccessibility to the pipes buried underground. Taking the opportunity of infrastructure construction and relevant pipeline replacement in China, biofilms in a DWDS (a water main and its branch pipes) were collected in situ, followed by analysis on the abundances and community structures of bacterial and archaeal using quantitative PCR and high-throughput sequencing, respectively. Results showed that archaea were detected only in the biofilms of the water main, with a range of 9.4×103~1.1×105 copies/cm2. By contrast, bacteria were detected in the biofilms of branch pipes and the distal part of the water main, with a range of 8.8×103~9.6×106 copies/cm2. Among the biofilm samples, the archaeal community in the central part of the water main showed the highest richness and diversity. Nitrosopumilus was found to be predominant (86.22%) in the biofilms of the proximal part of the water main. However, Methanobrevibacter (87.15%) predominated in the distal part of the water main. The bacterial community of the water main and branch pipes was primarily composed of Firmicutes and Proteobacteria at the phylum level, respectively. Regardless of archaea or bacteria, only few operational taxonomic units (OTUs) (<0.5% of total OTUs) were shared by all the biofilms, indicating the niche differentiation of biofilm microorganisms. Moreover, the high Mn content in the biofilms of the distal sampling location (D3) in the water main was linked to the predominance of Bacillus. Functional gene prediction revealed that the proportion of infectious disease-related genes was 0.44-0.67% in the tested biofilms. Furthermore, functional genes related to the resistance of the bacterial community to disinfections and antibiotics were detected in all the samples, that is, glutathione metabolism-relating genes (0.14-0.65%) and beta-lactam resistance gene (0.01-0.05%). The results of this study indicate the ubiquity of archaea and bacteria in the biofilms of water main and branch pipes, respectively, and pipe diameters could be a major influencing factor on bacterial community structure. In the water main, the key finding was the predominant existence of archaea, particularly Nitrosopumilus and methanogen. Hence, their routine monitoring and probable influences on water quality in pipelines with large diameter should be given more attention. Besides, since Mn-related Bacillus and suspected pathogenic Enterococcus were detected in the biofilm, supplementation of disinfectant may be a feasible strategy for inhibiting their growth and ensuring water quality. In addition, the monitoring on their abundance variation could help to determine the frequency and methods of pipeline maintenance.
Effects of drinking water distribution system on Vibrio amounts in biofilm

Abstract:Summary With the development of modern industries,drinking water in residential area and public building were supplied by water distribution systems.The drinking water distribution system is the final step from water plants to the users,and therefore is essential to assure drinking water safety for customers.Previous studies confirmed that many bacteria in the drinking water distribution systems existed as biofilm,which could induce corrosion and scaling of the pipe walls,reduce the water quality and water-carrying capacity of the pipelines. Moreover,as the pathogens growing in the biofilms transferred in the pipelines,the risks caused by these microorganisms would increase.Vibrio species is an estuarine bacterium widely distributed in the natural aquatic environment around the world.Among the discovered 76 kinds ofVibrio species,at least 12 kinds have pathogenic effect on human,and often result in severe diarrhea and dehydration.We found the Vibrio species in our previous survey of pathogenic bacteria in biofilm,indicating that the residents were at potential risk from Vibrio species. Therefore,the effect of pipe materials,pipe ages,and pipe diameters on amount of Vibrio species in biofilm of drinking water distribution system was necessary to investigate. In this study,1 2 biofilm samples were collected from the drinking water distribution system in east China. Vibrio was isolated using thiosulfate-citrate-bile salts-sucrose(TCBS)agar medium and was identified according to their biochemical reaction characteristics using biochemical identification kit for Vibrionaceae GYZ-9V.The amounts of total bacteria were determined by a spread-plating method according to the Ministry of Health StandardExaminationMethodsforDrinkingWater(2006).Samples and their dilutions were spread on Petri dishes with nutrient agar medium.Analysis of variance (ANOVA) by a least significant difference test was used to compare Vibrio counts from different pipe materials,ages and diameters. The results showed that Vibrio species could be detected in nodular cast iron,galvanized steel and stainless steel clad pipes,but not in plastic pipe.TheVibrio species amount on the biofilm attached to the nodular cast iron pipe(212±39.40 CFU/cm2)was significantly higher than those attached to the galvanized steel pipe(4.85±1.03 CFU/cm2)and stainless steel clad pipe(0.66±0.21 CFU/cm2).However,the rank of total bacteria amount was galvanized steel pipe > nodular cast iron pipe ≈ plastic pipe > stainless steel clad pipe.Pipe age had little influence on Vibrio amounts from galvanized steel and nodular cast iron pipes.Nodular cast iron pipe with a nominal diameter of 200 mm had the highest amount ofVibrio species(2.65×103 ±204 CFU/cm2),followed by 150 mm(212±39.40 CFU/cm2)and 300 mm(44.20±6.88 CFU/cm2),and the tendency of total bacteria amount was similar with Vibrio species. In conclusion, contamination from Vibrio species with varying levels is observed in nodular cast iron, galvanized steel and stainless steel clad pipes,but not in plastic pipe.The different pipe materials,especially nodular cart iron pipe,have strong influence on Vibrio species amount in biofilm from drinking water distribution system;therefore, the potential risks of Vibrio species from nodular cast iron pipe should be taken into consideration while laying the pipelines.
Bacterial communities of planktonic bacteria and mature biofilm in service lines and premise plumbing of a Megacity: Composition, Diversity, and influencing factors

Abstract:Although simulated studies have provided valuable knowledge regarding the communities of planktonic bacteria and biofilms, the lack of systematic field studies have hampered the understanding of microbiology in real-world service lines and premise plumbing. In this study, the bacterial communities of water and biofilm were explored, with a special focus on the lifetime development of biofilm communities and their key influencing factors. The 16S rRNA gene sequencing results showed that both the planktonic bacteria and biofilm were dominated by Proteobacteria. Among the 15,084 observed amplicon sequence variants (ASVs), the 33 core ASVs covered 72.8 %, while the 12 shared core ASVs accounted for 62.2 % of the total sequences. Remarkably, it was found that the species richness and diversity of biofilm communities correlated with pipe age. The relative abundance of ASV2 (f_Sphingomonadaceae) was lower for pipe ages 40-50 years (7.9 %) than for pipe ages 10-20 years (59.3 %), while the relative abundance of ASV10 (f_Hyphomonadaceae) was higher for pipe ages 40-50 years (19.5 %) than its presence at pipe ages 20-30 years (1.9 %). The community of the premise plumbing biofilm had significantly higher species richness and diversity than that of the service line, while the steel-plastics composite pipe interior lined with polyethylene (S-PE) harbored significantly more diverse biofilm than the galvanized steel pipes (S-Zn). Interestingly, S-PE was enriched with ASV27 (g_Mycobacterium), while S-Zn pipes were enriched with ASV13 (g_Pseudomonas). Moreover, the network analysis showed that five rare ASVs, not core ASVs, were keystone members in biofilm communities, indicating the importance of rare members in the function and stability of biofilm communities. This manuscript provides novel insights into real-world service lines and premise plumbing microbiology, regarding lifetime dynamics (pipe age 10-50 years), and the influences of pipe types (premise plumbing vs. service line) and pipe materials (S-Zn vs. S-PE).
Survey of Pathogenic Bacteria of Biofilms in a Metropolitan Drinking Water Distribution System

Abstract:ABSTRACT Bacteria, especially pathogenic bacteria, were detected in order to estimate the safety of drinking water distribution systems (DWDSs). Sixteen biofilms and 12 water samples (six retained and six flowing) were collected from a city DWDS in eastern China. Biofilms were observed using scanning electron microscopy. Cultivable bacteria of biofilms were counted by heterotrophic plate counts, ranging from 3.61 × 101 to 1.67 × 106 CFU·cm−2. Coliforms, Salmonella, Shigella, Vibrio and Legionella were separated by Eosin-Methylene Blue (EMB) agar, Salmonella chromogenic medium, Shigella chromogenic medium, Thiosulfate Citrate Bile Salts Sucrose (TCBS) agar and Buffered Charcoal Yeast Extract (BCYE) agar and 13/16, 8/16, 7/16, 6/16, 0/16 biofilm samples were found to be positive, respectively. Retained and flowing water samples were collected to estimate the influence of hydrodynamic conditions on biofilm detachment. All six retained water samples were positive for bacteria, the count ranged from 1.2 × 103 to 2.8 × 104 CFU·mL−1 and 2/6, 3/6, 2/6, 0/6, 0/6 samples were positive for coliforms, Salmonella, Shigella, Legionella and Vibrio, respectively. While only three of six flowing water samples were bacteria positive, the counts ranged from 102 to 103 CFU·mL−1, 2/6 were coliform positive and no pathogens were detected under testing. The results show that there are pathogens in DWDS biofilms, which can cause health-related problems if detached from their surfaces.
Assessing the Contribution of Biofilm to Bacterial Growth During Stagnation in Shower Hoses

Abstract:Stagnation occurs in building water supplies when there is little or no water usage. As a result, the number of bacteria increase, and this often leads to the deterioration of water quality. Still, the role of biofilm in stagnation remains unclear. This study used shower hoses as the model system and investigated the contribution of biofilm and microbes in fresh water to the bacterial growth in water under different stagnation times from 6 to 24 h. Bacterial counts in water were observed to increase significantly after 12 h stagnation but longer stagnation did not lead to further increase, indicating different mechanisms contributing to bacterial growth during stagnation. 16S rRNA gene sequencing and Sourcetracker2 further confirmed that the contribution of fresh water to the microbial core community did not increase significantly with stagnation time, whereas the contribution of biofilm increased significantly after 24 h stagnation (53.5%) compared with 6 h stagnation (11.2%) (p< 0.05). The present results differentiated the contribution between planktonic and biofilm phase to the bacterial growth during stagnation, and provided insights into its mechanism. These findings serve as a framework for future development of strategies to manage biological water quality at the distal end of the building water supplies.
Biofilm formation dynamics in long-distance water conveyance pipelines: Impacts of nutrient levels and metal stress

Abstract:Biofilm formation in long-distance water conveyance pipelines poses significant risks to water quality, particularly under varying nutrient levels and heavy metal stress. However, the impacts of pipeline material on biofilm formation dynamics under different raw water conditions remain elusive. This study investigated the effects of nutrient availability and Fe-Mn stress on biofilm development, structural stability, bacterial community composition, and the occurrence of viable but non-culturable (VBNC) bacteria. Using reactors with different nutrient conditions, we observed that increased nutrient levels promote biofilm growth but lead to greater instability, heightening the risk of secondary contamination. Notably, nutrient escalation beyond a critical threshold had a diminishing impact on biofilm community composition. Additionally, Fe-Mn stress, while initially enhancing microbial adhesion and metabolic activity, ultimately inhibited biofilm formation over time and increases the prevalence of VBNC bacteria, particularly on stainless steel (SS) surfaces. Our findings also highlighted the importance of material selection for pipelines, with polyvinyl chloride (PVC) showing reduced biofilm formation compared to SS, making it a more suitable option for transporting raw water in environments with high metal content. Dispersal limitation determined the bacterial community assembly during the biofilm formation, accounting for 64.53-90.67 % of the variability in different scenarios. These insights offer valuable guidance for managing biofilm-related issues in water distribution systems, emphasizing the need for careful control of nutrient levels and material choice to ensure water safety over long distances.
Bacterial Community Radial-Spatial Distribution in Biofilms along Pipe Wall in Chlorinated Drinking Water Distribution System of East China

Abstract:Biofilms in the pipe wall may lead to water quality deterioration and biological instability in drinking water distribution systems (DWDSs). In this study, bacterial community radial-spatial distribution in biofilms along the pipe wall in a chlorinated DWDS of East China was investigated. Three pipes of large diameter (300, 600, and 600 mm) were sampled in this DWDS, including a ductile cast iron pipe (DCIP) with pipe age of 11 years and two gray cast iron pipes (GCIP) with pipe ages of 17 and 19 years, and biofilms in the upper, middle, and lower parts of each pipe wall were collected. Real-time quantitative polymerase chain reaction (qPCR) and culture-based method were used to quantify bacteria. 454 pyrosequencing was used for bacterial community analysis. The results showed that the biofilm density and total solid (TS) and volatile solid (VS) contents increased gradually from the top to the bottom along the pipe wall. Microorganisms were concentrated in the upper and lower parts of the pipe wall, together accounting for more than 80 % of the total biomass in the biofilms. The bacterial communities in biofilms were significantly different in different areas of the pipe wall and had no strong interaction. Compared with the upper and lower parts of the pipe wall, the bacterial community in the middle of the pipe wall was distributed evenly and had the highest diversity. The 16S rRNA genes of various possible pathogens, including Escherichia coli, Staphylococcus epidermidis, Pseudomonas aeruginosa, and Salmonella enterica, were detected in the biofilms, and the abundances of these possible pathogens were highest in the middle of the pipe wall among three areas. The detachment of the biofilms is the main reason for the deterioration of the water quality in DWDSs. The results of this study suggest that the biofilms in the middle of the pipe wall have highly potential risk for drinking water safety, which provides new ideas for the study of the microbial ecology in DWDS.
Influence of microbial community on iron release under stagnation condition in drinking water systems

Abstract:To investigate the influence of total bacteria and microbial community structure on the iron release in water supply pipes under stagnation condition, the research was carried on the pilot platform constructed in actual water supply system. The number of culturable corrosive bacteria was counted by conventional R2A cultural method, and microbial community structure was analyzed using high-throughput sequencing (454 pyrosequencing). Results show that Proteobacteria, which account for 86.69%~91.36% in pipe biofilms under stagnation condition, are the dominant microbial community in phylum level. In class level, the content of Alphaproteobacteria, Betaproteobacteria and Actinobacteria excesses half of the total amount. The total iron concentration and iron oxidizing bacteria had a strong correlation with sulfate reducing bacteria. In ductile cast iron pipes, the influence of corrosive bacteria on total iron release is greater than that of HDPE pipes. By correlation coefficient and RDA analysis, it is observed that Proteobacteria, Actinobacteria and Acidobacteria in phylum level and Alphaproteobacteria, Betaproteobacteria and Bacilli in class level promote iron release.

Abstract:Biofilms in drinking water distribution systems (DWDSs) could cause several types of problems, such as the deterioration of water quality, corrosion of pipe walls, and potential proliferation of opportunistic pathogens. In this study, ten biofilm samples from different pipe materials, including ductile cast iron pipe (DCIP), gray cast iron pipe (GCIP), galvanized steel pipe (GSP), stainless steel clad pipe (SSCP), and polyvinyl chloride (PVC), were collected from an actual DWDS to investigate the effect of pipe material on bacterial community. Real-time quantitative polymerase chain reaction (qPCR) and culture-based method were used to quantify bacteria. 454 pyrosequencing was used for bacterial community analysis. The results showed that the numbers of total bacteria and culturable heterotrophic bacteria from iron pipes were higher than that in PVC, while the numbers of Shigella and vibrios were low in biofilms from iron pipes. Bacterial community analysis showed that Hyphomicrobium or Desulfovibrio were the predominant microorganism in iron pipes, whereas Sphingomonas or Pseudomonas were dominant in other types of pipe. This study revealed differences in bacterial communities in biofilms among different pipe materials, and the results were useful for pipeline material selection in DWDSs.
Spatiotemporal Change Rules of Water Quality in Cut-off Water Pipelines in District Metering Areas (DMA)

Abstract:The pilot platform was constructed with water pipelines of nine years old to study the spatiotemporal changes of essential physical, chemical and biological parameters of water in cut-off pipelines, in order to explore the potential impact of cut-off pipelines and stagnant areas which are created in the construction of district metering areas (DMAs) on water quality. Results show that, as the depth of cut-off pipeline increases, the biofilm and limescale fall off more quickly and the water quality deterioration rate gets faster; the temporal changes of iron release, turbidity and microorganism vary with time at different depths. After twelve hours, the water quality deterioration rate was significantly accelerated. The correlation analysis of the deterioration of water quality parameters shows that, the deeper the depth of cut-off pipeline is, the less the water disturbed by the main pipe is and the higher the correlation level of each parameter is.

Abstract:The deterioration of drinking water quality due to corrosion of the water supply network has become inevitable and regular renewal of pipes has become a common means of doing so. Severely corroded pipes release certain nutrients (e.g., elemental phosphorus), however, little has been reported on the effect of old pipes on the young biofilm of new pipe sections and on ensuring water safety in the early stages of the water supply. The aim of our study was to model the effect of key phosphorus nutrients released from corroded old pipes on the morphological characteristics of young biofilms in new pipe sections, mediated disinfection byproducts (DBPs) production and their combined toxicity. Based on the experimental results, phosphorus showed significant differences in the morphological characteristics, spatial structure of extracellular polymers (EPS), functional abundance, disinfection byproduct formation potential (DBPsFP) and toxicity of young biofilms. Under residual chlorine (1.0 ± 0.2 mg/L) incubation, the functional abundance of young biofilm metabolism was dominant, particularly amino acid metabolism and carbohydrate metabolism. There is a dynamic balance between the trophic and shedding effects of phosphorus, where concentration changes affect young biofilm morphology and DBPFP. Relatively moderate phosphorus concentrations resulted in the highest density of PN/PS organic precursors in EPS and a clear advantage of DBPFP; relatively high phosphorus conditions had limited promotion of young biofilm, while membrane structure shedding was more pronounced, increasing young biofilm-mediated DBPs production. Nitrogen-containing disinfection byproducts (N-DBPs) in young biofilms had a clear toxicity advantage, with HANs and HNMs being key to controlling cytotoxicity and genotoxicity, respectively.

Abstract:Pipe wall biofilm plays a vital role in water quality purification in raw water transportation system (RWTS). This study investigated the pollutants removal, bacterial community (abundant, rare and functional taxa) variations, and their correlations during biofilm development process under different pipe materials and raw water quality. Polyethylene pipe had higher removal efficiency on ammonia nitrogen (NH4 +-N) and dissolved organic carbon (DOC), while stainless steel pipe achieved a better total nitrogen (TN) degradation performance. Increasing influent DOC promoted the removal of nitrogen pollutants. Compared with water quality, pipe material showed a greater influence on bacterial diversity and community structure. Pseudomonas and Planctopirus were the dominant genera in abundant and rare taxa, respectively. There were complex interactions within the abundant and rare taxa, with Haliscomenobacter and Reyranella as the hub genera, respectively, and many abundant genera co-occurred with rare genera. Serratia of abundant taxa and Filomicrobium of rare taxa had positive correlations with the influent NH4 +-N and DOC concentration, respectively. Complete ammonia oxidation and ammonia assimilatory contributed to the great NH4 +-N transformation, while limited denitrification resulted in the undesirable TN removal. Functional bacteria and genes displayed statistically strong correlations with water quality. Abundant and rare taxa were significantly affected by environmental conditions and organic matter, respectively, while the variations in functional taxa were mainly explained by organic matter, nitrogen and their interactions. These findings provide novel insights into the evolution of abundant, rare and functional taxa during biofilm development in RWTS, and highlight their correlations with water purification performance and environmental factors.

Abstract:In newly commissioned drinking-water polyethylene (PE) pipes, biofilm develops on the inner pipe surface. The microbial community composition from colonization to the establishment of mature biofilms is less known, including the effect on the distributed water quality. Biofilm development was followed through 1.5 years in PE-pipe side streams at two locations of a full-scale, non-chlorinated drinking-water distribution system (leaving a waterworks versus 5–6 km from a waterworks) along with inlet and outlet water quality. Mature biofilms were established after ∼8–9 months, dominated by Proteobacteria , Actinobacteria and Saccharibacteria (61–93% relative abundance), with a higher diversity (OTUs/Shannon Index/16S rRNA gene amplicon sequencing) in pipes in the far end of the distribution system. Comamonadaceae , and specifically Aquabacterium (>30% of reads), dominated young (∼1.5-month-old) biofilms. Young biofilms were linked to increased microbiological counts in drinking water (HPC/ATP/qPCR), while the establishment of mature biofilms led to a drop in HPC and benefited the water quality, highlighting the importance of optimizing commissioning procedures for rapidly achieving mature and stable biofilms.

Abstract:As many cities around the world face the prospect of replacing aging drinking water distribution systems (DWDS), water utilities must make careful decisions on new pipe material (e.g., cement-lined or PVC) for these systems. These decisions are informed by cost, physical integrity, and impact on microbiological and physicochemical water quality. Indeed, pipe material can impact the development of biofilm in DWDS that can harbor pathogens and impact drinking water quality. Annular reactors (ARs) with cast iron and cement coupons fed with chloraminated water from a municipal DWDS were used to investigate the impact of pipe material on biofilm development and composition over 16 months. The ARs were plumbed as closely as possible to the water main in the basement of an academic building to simulate distribution system conditions. Biofilm communities on coupons were characterized using 16S rRNA sequencing. In the cast iron reactors, β-proteobacteria, Actinobacteria, and α-proteobacteria were similarly relatively abundant (24.1, 22.5, and 22.4%, respectively) while in the cement reactors, α-proteobacteria and Actinobacteria were more relatively abundant (36.3 and 35.2%, respectively) compared to β-proteobacteria (12.8%). Mean alpha diversity (estimated with Shannon H and Faith’s Phylogenetic Difference indices) was greater in cast iron reactors (Shannon: 5.00 ± 0.41; Faith’s PD: 15.40 ± 2.88) than in cement reactors (Shannon: 4.16 ± 0.78; Faith’s PD: 13.00 ± 2.01). PCoA of Bray-Curtis dissimilarities indicated that communities in cast iron ARs, cement ARs, bulk distribution system water, and distribution system pipe biofilm were distinct. The mean relative abundance of Mycobacterium spp. was greater in the cement reactors (34.8 ± 18.6%) than in the cast iron reactors (21.7 ± 11.9%). In contrast, the mean relative abundance of Legionella spp. trended higher in biofilm from cast iron reactors (0.5 ± 0.7%) than biofilm in cement reactors (0.01 ± 0.01%). These results suggest that pipe material is associated with differences in the diversity, bacterial composition, and opportunistic pathogen prevalence in biofilm of DWDS.

Abstract:Pipe material and residual chlorine are key factors for the drinking water distribution system, and understanding the biofilm ecosystem is vital for water quality safeguard. The aim of our study was to determine the influence of pipe materials (ductile iron, steel, polyethylene) and chlorination on the biofilm structure and microbial community, as shown by the physicochemical properties, extracellular polymeric substances (EPS) structural characteristics, bacterial community composition, and functional traits. EPS spatial properties were studied based on a semi-quantitative confocal laser scanning microscope (CLSM) description. Regarding the impact of chlorination, residule chlorine (1.0 ± 0.3 mg L<sup>-1</sup> free chlorine) could inhibit the bacteria colonization, and initiate a potential response to external disinfectants revealed by the EPS spatial distribution changes and communities variation compared to unchlorinated system. Regarding the impact of pipe material, polyethylene (PE) biofilms displayed lower biomass, loose zoogloea structure, lower proteins and polysaccharides content, and poor microbial diversity in contrast to ductile iron and steel biofilms. Pipe material was the more possible driving factor of the biofilm community composition compared to the chlorination based on principal coordinates analysis (PCoA) and permutational multivariate analysis of variance (PERMANOVA). Actinobacteria was dominant in the PE biofilms (45.57%-83.32%), while Alphaproteobacteria (34.30%-73.22%) and Gammaproteobacteria (6.46%-36.82%) were the major classes in the steel and ductile iron biofilms. The genus Rhodococcus was predominant in the PE biofilms. Rhodococcus, Pseudomonas, and Sphingomonas seemed to have a better growth advantage in the chlorinated system and display a stronger disinfectant resistance. Functional sketch prediction indicated the potential impact of pipe material and chlorination on functional pathway abundnce, possible functional pathways associated with infectious disease included. This study provides insights into the impact of pipe material and chlorination on biofilm structure and microbial community and might help to develop monitoring or maintenance strategies to protect the biosafety of the drinking water.
