Abstract:We describe the design and performance of a scanning tunneling microscope (STM) which operates at a base temperature of 30 mK in a vector magnetic field. The cryogenics is based on an ultra-high vacuum (UHV) top-loading wet dilution refrigerator that contains a vector magnet allowing for fields up to 9 T perpendicular and 4 T parallel to the sample. The STM is placed in a multi-chamber UHV system, which allows in-situ preparation and exchange of samples and tips. The entire system rests on a 150-ton concrete block suspended by pneumatic isolators, which is housed in an acoustically isolated and electromagnetically shielded laboratory optimized for extremely low noise scanning probe measurements. We demonstrate the overall performance by illustrating atomic resolution and quasiparticle interference imaging and detail the vibrational noise of both the laboratory and microscope. We also determine the electron temperature via measurement of the superconducting gap of Re(0001) and illustrate magnetic field-dependent measurements of the spin excitations of individual Fe atoms on Pt(111). Finally, we demonstrate spin resolution by imaging the magnetic structure of the Fe double layer on W(110).

Abstract:One of the daunting challenges in modern low temperature scanning tunneling microscopy (STM) is the difficulty of combining atomic resolution with cryogen free cooling. Further functionality needs, such as ultra-high vacuum (UHV), high magnetic field, and compatibility with {\mu}m-sized samples, pose additional challenges to an already ambitious build. We present the design, construction, and performance of a cryogen free, UHV, low temperature, and high magnetic field system for modular STM operation. An internal vibration isolator reduces vibrations in this system allowing atomic resolution STM imaging while maintaining a low base temperature of ~4K and magnetic fields up to 9T. Samples and tips can be conditioned in-situ utilizing a heating stage, an ion sputtering gun, an e-beam evaporator, a tip treater, and sample exfoliation. In-situ sample and tip exchange and alignment are performed in a connected UHV room temperature stage with optical access. Multisite operation without breaking vacuum is enabled by a unique quick-connect STM head design. A novel low-profile vertical transfer mechanism permits transferring the STM between room temperature and the low temperature cryostat.

Abstract:We present the design and performance of a cryogenic scanning tunneling microscope (STM) which operates inside a water-cooled Bitter magnet, which can attain a magnetic field of up to 38 T. Due to the high vibration environment generated by the magnet cooling water, a uniquely designed STM and vibration damping system are required. The STM scan head is designed to be as compact and rigid as possible, to minimize the effect of vibrational noise as well as fit the size constraints of the Bitter magnet. The STM uses a differential screw mechanism for coarse tip - sample approach, and operates in helium exchange gas at cryogenic temperatures. The reliability and performance of the STM are demonstrated through topographic imaging and scanning tunneling spectroscopy (STS) on highly oriented pyrolytic graphite (HOPG) at T = 4.2 K and in magnetic fields up to 34 T.

Abstract:We present a novel homebuilt scanning tunneling microscope (STM) with atomic resolution integrated into a cryogen-free superconducting magnet system with a variable temperature insert. The STM head is designed as a nested structure of double piezoelectric tubes (PTs), which are connected coaxially through a sapphire frame whose top has a sample stage. A single shaft made of tantalum, with the STM tip on top, is held firmly by a spring strip inside the internal PT. The external PT drives the shaft to the tip-sample junction based on the SpiderDrive principle, and the internal PT completes the subsequent scanning and imaging work. The STM head is simple, compact, and easy to assemble. The excellent performance of the device was demonstrated by obtaining atomic-resolution images of graphite and low drift rates of 30.2 pm/min and 41.4 pm/min in the X-Y plane and Z direction, respectively, at 300K. In addition, we cooled the sample to 1.6 K and took atomic-resolution images of graphite and NbSe2. Finally, we performed a magnetic field sweep test from 0 T to 9 T at 70 K, obtaining distinct graphite images with atomic resolution under varying magnetic fields. These experiments show our newly developed STM's high stability, vibration resistance, and immunity to high magnetic fields.

Abstract:We describe the design and development of a scanning tunneling micoscope (STM) working at very low temperatures in ultra-high vacuum (UHV) and at high magnetic fields. The STM is mounted to the He3 pot of an entirely UHV compatible He3 refrigerator inside a tube which can be baked out to achieve UHV conditions even at room temperature. A base temperature of 315 mK with a hold time of 30 h without any recondensing or refilling of cryogenics is achieved. The STM can be moved from the cryostat into a lower UHV-chamber system where STM-tips and -samples can be exchanged without breaking UHV. The chambers contain standard surface science tools for preparation and characterization of tips and samples in particular for spin-resolved scanning tunneling spectroscopy (STS). Test measurements using either superconducting tips or samples show that the system is adequate for performing STS with both high spatial and high energy resolution. The vertical stability of the tunnel junction is shown to be 5 pmpp and the energy resolution is about 100 μeV.

Abstract:We present the design and construction of a nonmetallic tip-sample mechanical loop featured Scanning Tunneling Microscope (STM) that operates in a 22 T water-cooled magnet at a low temperature of l.8 K. The STM head mainly consists of a spider-drive motor, stand-alone scanner, moveable sapphire sample holder, and sapphire frame. All parts exist in the tip-sample mechanical loop are made of sapphire to reduce the interference from high magnetic fields. Except for the necessary movement of the tip and scanner, all STM parts are stationary. More importantly, the tip-sample mechanical loop is separate from the motor after detecting the tunneling current, which helps prevent the high voltage signal interference from entering the tip-sample junction, leading to a high stable imaging. A Janis liquid helium cryostat is used to obtain a variable temperature range from 1.8 K to 300 K, and the STM head is cooled down via helium exchange gas. The STM head hangs at the bottom of a probe with a two-stage spring suspension to prevent the huge vibration generated by the water-cooled magnet from entering the tip-sample junction. The performance is demonstrated by atomically resolved STM images of graphite surface at 0 T and 22.8 T under room temperature. Furthermore, the obtained atomic-resolution images of NbSe2 at 1.8 K and 22 T, as well as high-resolution dI/dV spectrums at temperatures from 1.8 K to 8.5 K and magnetic fields from 0 T to 22 T are displayed. This is the first STM capable of atomic-resolution imaging and dI/dV measurement at 1.8 K in a 22 T water-cooled magnet. The high immunity to the magnetic field makes the nonmetallic tip-sample mechanical loop widely useable for atomic-resolution STM imaging in ultra-high magnetic field conditions.

Abstract:We present a dual scanning tunneling microscope (DSTM) system operating between 2.2K and room temperature, in a split-coil superconducting magnetic field up to 12T and in ultrahigh vacuum. The DSTM consists of two compact STMs, each having x, y, and z coarse positioning piezoelectric steppers with embedded capacitive positioning sensor for navigation. Each STM can be operated independently and can achieve atomic resolution. The DSTM and the sample is configured in a way that allows the magnetic field orientation to be varied continuously from normal to parallel to the sample surface. Together with the sample, the DSTM can form a nanometer scale three terminal setup for transport measurement.

Abstract:We present the first nonmetallic scanning tunneling microscope (STM) featuring an ultra-stable tip-sample mechanical loop and capable of atomic-resolution imaging within a 12 T magnetic field that could be either perpendicular or parallel to the sample surface. This is also the first STM with an ultra-stable tip-sample mechanical loop but without a standalone scanner. The STM head is constructed only with two parts: an improved spider-drive motor and a zirconia tip holder. The motor performs both the coarse approach and atomic imaging. A supporting spring is set at the fixed end of the motor tube to decrease the tip-sample mechanical loop. The zirconia tip holder performs as the frame of the whole STM head. With the novel design, the STM head in three dimensions can be as small as 7.9 mm × 7.9 mm × 26.5 mm. The device's excellent performance is demonstrated by atomic-resolution images of graphite and NbSe2 obtained at 300 K and 2 K, as well as the high-resolution dI/dV spectrums of NbSe2 at variable temperatures. Low drift rates in the X-Y plane and Z direction further prove the imaging stability of our new STM. High-quality imaging of the Charge Density Wave (CDW) structure on a TaS2 surface shows the STM's good application capability. Continuous atomic images obtained in magnetic fields rangs from 0 T to 12 T with the direction of the magnetic field perpendicular or parallel to the sample surface show the STM's good immunity to high magnetic fields. Our results illustrate the new STM's broad application ability in extreme conditions of low temperature and high magnetic field.

Abstract:We describe the design and development of a scanning tunneling microscope (STM) which can operate at temperatures down to 240 mK and in magnetic fields up to 7 T with high spatial and energy resolution. The compact and rigid STM head is mounted directly on a low vibration, single shot, He3 refrigerator. This refrigerator can be operated at its base temperature continuously for several days before the He3 needs to be recondensed. The system is equipped with a sample transport manipulator from room temperature, and a cleavage device at low temperature, so that the cryogenic ultrahigh vacuum condition inside the cryostat can be utilized. A superconducting magnet provides a magnetic field of up to 7 T at the sample along the STM tip direction. Test results have shown that, at the base temperature, this instrument has better than 0.5 pm z-direction resolution in imaging mode, and better than 20 μV energy resolution in spectroscopy mode.

Abstract:We describe the construction and performance of a scanning tunneling microscope (STM) capable of taking maps of the tunneling density of states with sub-atomic spatial resolution at dilution refrigerator temperatures and high (14 T) magnetic fields. The fully ultra-high vacuum system features visual access to a two-sample microscope stage at the end of a bottom-loading dilution refrigerator, which facilitates the transfer of in situ prepared tips and samples. The two-sample stage enables location of the best area of the sample under study and extends the experiment lifetime. The successful thermal anchoring of the microscope, described in detail, is confirmed through a base temperature reading of 20 mK, along with a measured electron temperature of 250 mK. Atomically-resolved images, along with complementary vibration measurements, are presented to confirm the effectiveness of the vibration isolation scheme in this instrument. Finally, we demonstrate that the microscope is capable of the same level of performance as typical machines with more modest refrigeration by measuring spectroscopic maps at base temperature both at zero field and in an applied magnetic field.

Abstract:We report the construction and performance of a low temperature, high field scanning tunneling microscope (STM) operating down to 350mK and in magnetic fields up to 9T, with thin film deposition and in-situ cleaving capabilities. The main focus lies on the simple design of STM head allowing top loading of the sample and innovative sample holder design that allows us to get spectroscopic data on superconducting thin films grown in-situ on insulating substrates. Other design details on sample transport, chamber and vibration isolation scheme are also described. We demonstrate the capability of our instrument through the atomic resolution imaging and spectroscopy on NbSe2 single crystal and spectroscopic maps obtained on homogeneously disordered NbN thin film.

Abstract:A variable-temperature ultrahigh vacuum (UHV) scanning tunneling microscope (STM) was designed and tested. Design details and initial results are presented. The STM is directly attached to the cold face of a continuous flow cryostat which is mounted into a two-chamber UHV system. A significant advantage of this system in comparison to many others is, that samples can be cooled down to base temperature of 6.5 K within very short times of below 2 h. This feature not only increases the potential sample throughput, it also allows to cycle the sample temperature within the regime below 20 K without losing track of given sample locations. The instrument was tested by imaging Au layers on graphite. The vertical stability at low temperature was found to be below 3 pm. Images recorded at 6.5 K show crystalline Au islands and the Au(111)22×√3 reconstruction with atomic resolution. Using a resistive heater, the sample temperature was adjusted between 6.5 and 20 K. After an equilibration time of 15 min, the displacement due to the temperature change remained below 150 nm. Scanning tunneling spectroscopy on Au(111) grains resolves the Au(111) surface state.

Abstract:We have designed and built a scanning tunneling microscope (STM) setup for operation at millikelvin temperatures in ultra high vacuum. A compact cryostat with an integrated dilution refrigerator has been built, that allows measurements at a base temperature of 25 mK in magnetic field up to 7.5 T with low mechanical and electronic noise. The cryostat is not larger than conventional helium bath cryostats (23 and 13 liters of nitrogen and helium respectively), so that the setup does not require a large experimental hall and fits easily into a standard lab space. Mechanical vibrations with running dilution circulation were kept below 300 fm/$\sqrt{\mathrm{Hz}}$ by mechanically decoupling the STM from the cryostat and the pumping system. All electronic input lines were low-pass filtered, reducing the electronic temperature to below 100 mK, as deduced from the quasiparticle peaks of superconducting aluminium. The microscope is optically accessible in the parked position, making sample and tip exchange fast and user-friendly. For measurement the STM is lowered 60 mm down so that the sample ends in the middle of a wet superconducting magnetic coil.

Abstract:We present the design and performance of a novel scanning tunnelling microscope (STM) operating in a cryogen‐free superconducting magnet. Our home‐built STM head is compact (51.5 mm long and 20 mm in diameter) and has a single arm that provides complete openness in the scanning area between the tip and sample. The STM head consists of two piezoelectric tubes (PTs), a piezoelectric scanning tube (PST) mounted on a well‐polished zirconia shaft, and a large PT housed in a sapphire tube called the motor tube. The main body of the STM head is made of tantalum. In this design, we fixed the sapphire tube to the frame with screws so that the tube's position can be changed quickly. To analyse the stiffness of the STM head unit, we identified the lowest eigenfrequencies with 3 and 4 kHz in the bending modes, 8 kHz in a torsional mode, and 9 kHz in a longitudinal mode by finite element analysis, and also measured the low drift rates in the X–Y plane and in the Z direction. The high performance of the home‐built STM was demonstrated by images of the hexagonal graphite lattice at 300 K and in a sweeping magnetic field from 0 T to 9 T. Our results confirm the high stability, vibration resistance, insensitivity to high magnetic fields and the application potential of our newly developed STM for the investigation of low‐frequency systems with high static support stiffness in physics, chemistry, material and biological sciences.

Abstract:We describe the design, construction, and performance of an ultra-high vacuum (UHV) scanning tunneling microscope (STM) capable of imaging at dilution-refrigerator temperatures and equipped with a vector magnet. The primary objective of our design is to achieve a high level of modularity by partitioning the STM system into a set of easily separable, interchangeable components. This naturally segregates the UHV needs of STM instrumentation from the typically non-UHV construction of a dilution refrigerator, facilitating the usage of non-UHV materials while maintaining a fully bakeable UHV chamber that houses the STM. The modular design also permits speedy removal of the microscope head from the rest of the system, allowing for repairs, modifications, and even replacement of the entire microscope head to be made at any time without warming the cryostat or compromising the vacuum. Without using cryogenic filters, we measured an electron temperature of 184 mK on a superconducting Al(100) single crystal.

Abstract:We present the design and performance of an ultra-high vacuum (UHV) scanning tunneling microscope (STM) that uses adiabatic demagnetization of electron magnetic moments for controlling its operating temperature in the range between 30 mK and 1 K with the accuracy of up to 7 $\mu$K. The time available for STM experiments at 50 mK is longer than 20 h, at 100 mK about 40 h. The single-shot adiabatic demagnetization refrigerator (ADR) can be regenerated automatically within 7 hours while keeping the STM temperature below 5 K. The whole setup is located in a vibrationally isolated, electromagnetically shielded laboratory with no mechanical pumping lines penetrating through its isolation walls. The 1K pot of the ADR cryostat can be operated silently for more than 20 days in a single-shot mode using a custom-built high-capacity cryopump. A high degree of vibrational decoupling together with the use of a specially-designed minimalistic STM head provides an outstanding mechanical stability, demonstrated by the tunneling current noise, STM imaging, and scanning tunneling spectroscopy measurements all performed on atomically clean Al(100) surface.

Abstract:The design and performance of a cryogen-free low temperature scanning tunneling microscope (STM) housed in ultrahigh vacuum (UHV) are reported. The cryogen-free design was done by directly integrating a Gifford-McMahon cycle cryocooler to a Besocke-type STM, and the vibration isolation was achieved by using a two-stage rubber bellow between the cryocooler and a UHV-STM interface with helium exchange gas cooling. A base temperature of 15 K at the STM was achieved, with a possibility to further decrease by using a cryocooler with higher cooling power and adding additional low temperature stage under the exchange gas interface. Atomically sharp STM images and high resolution dI/dV spectra on various samples were demonstrated. Furthermore, we reported the inelastic tunneling spectroscopy on a single carbon monoxide molecule adsorbed on Ag(110) surface with a cryogen-free STM for the first time. Being totally cryogen-free, the system not only saves the running cost significantly but also enables uninterrupted data acquisitions and variable temperature measurements with much ease. In addition, the system is capable of coupling light to the STM junction by a pair of lens inside the UHV chamber. We expect that these enhanced capabilities could further broaden our views to the atomic-scale world.
Quantitative detection of tumor M2-pyruvate kinase in plasma of patients with lung cancer in comparison to other lung diseases.

Abstract:Lung cancer is one of the predominant causes of cancer death. The aim of this project is the development of a screening method in persons with high risk for developing lung cancer, based on the measurement of Tumor M2-pyruvate kinase (Tumor M2-PK). Tumor M2-PK is quantitatively detectable in ethylenediaminetetraacetic acid-plasma with a sensitive enzyme-linked immunosorbent assay. So far, 60 patients with newly diagnosed lung cancer were included. These were compared to 24 patients with acute inflammatory lung diseases, 56 patients with pneumoconiosis, 22 patients with obstructive airway diseases, and 28 healthy persons. Tumor patients and some individuals suffering from severe inflammatory lung diseases had significantly higher Tumor M2-PK concentrations in ethylenediaminetetraacetic acid-plasma than all the other groups. The histologic tumor type had no influence on the plasma levels of Tumor M2-PK. Tumor M2-PK concentrations correlate strongly with the tumor stage, showing significantly increasing concentrations with progressive tumor stages. The present data indicate that Tumor M2-PK could be a valuable tumor marker for the detection of lung cancer.

Abstract:The design and performance of a fast-scanning, low- and variable-temperature, scanning tunneling microscope (STM) incorporated in an ultrahigh vacuum system is described. The sample temperature can be varied from 25 to 350 K by cooling the sample using a continuous flow He cryostat and counter heating by a W filament. The sample temperature can be changed tens of degrees on a time scale of minutes, and scanning is possible within minutes after a temperature change. By means of a software implemented active drift compensation the drift rate can be as low as 1 nm/day. The STM is rigid, very compact, and of low weight, and is attached firmly to the sample holder using a bayonet-type socket. Atomic resolution on clean metal surfaces can be achieved in the entire temperature range. The performance of the instrument is further demonstrated by images of adsorbed hexa-tert-butyl-decacyclene molecules on Cu(110), by STM movies, i.e., sequential STM images with a time resolution down to 1 s/image (100×100 Å2 with 256×256 pixels), of the mobility of these molecules, and finally by constant current images of standing waves in the electronic local density of states on Cu(110).
