Abstract:Adenosine has received great attentions acting as a potential biomarker for monitoring lung cancer. Most of the reported studies for adenosine detection require large instruments and complicated procedures. Herein, a sensitive, rapid and in-situ colorimetric aptasensor was developed for adenosine detection. Moreover, a homemade biomimetic electronic-eye (E-eye) was established and utilized as a portable in-time detection equipment. The entire measurement can be completed within 20 min, including the combination of aptamer with adenosine or AuNPs and the detection of adenosine. Four different kinds of aptamer were compared and the results showed that the AuNPs-aptamer-biotin system was the most stable and with the widest detection range of 5.0 mu M-60.0 mu M and the lowest LOD of 0.17 mu M. Moreover, the artificial urine samples were also tested with a linear range from 5.0 to 50.0 mu M and a LOD of 0.48 mu M. The results validated that the aptasensor together with the E-eye can be a promising platform for adenosine detection. (C) 2020 Elsevier B.V. All rights reserved.
COLORIMETRIC DETERMINATION OF ADENOSINE IN URINE USING BIOTIN MODIFIED APTAMER AND GOLD NANOPARTICLES

Abstract:Adenosine can be a possible biomarker for monitoring the progress of lung cancer. Here, a sensitive and easily operated colorimetric methods based on aptasensor for the detection of adenosine has been developed. Gold nanoparticles (AuNPs) will aggregate in high-salt solutions, which causes color change, and single-stranded DNA will prevent this from happening. Based on changes in solution color, adenosine can be quantitatively determined. Besides a home-made bionic electronic-eye (E-eye) system was developed as a portable in-situ detection platform. Two different aptamers were used for comparison for adenosine detection. The linear correlation existed between 5.0-60.0μM with a detection limit of 0.17μM in water with modified aptamer. Moreover, the performance of the method was validated by testing the artificial urine samples with the linear range from 5.0 to 50.0μM and a detection limit of 0.48μM in artificial urine. This simple and cost-effective approach indicating its great potential for point-of-care applications.
A Novel Colorimetric Biosensor Based on Non-Aggregated Au@Ag Core–shell Nanoparticles for Methamphetamine and Cocaine Detection

Abstract:We report a novel colorimetric biosensor based on non-aggregation Au@Ag core-shell nanoparticles to detect methamphetamine and cocaine. The biosensor consisted of a reporter probe (RP) that is a specific single-stranded DNA (ssDNA) sequence coated on Au@Ag nanoparticles, a capture probe (CP) conjugated with magnetic beads, and an illicit drug-binding DNA aptamer (Apt). Au@Ag nanoparticles were synthesized by seed growth and characterized by scanning electron microscope (SEM), high-resolution transmission electron microscopy (HR-TEM), and UV-vis spectra. Methamphetamine (METH) was used as an example to evaluate the feasibility of the biosensor and to optimize the detection conditions. We demonstrated that this sensing platform was able to detect as low as 0.1nM (14.9ngL-1) METH with a negligible interference from other common illicit drugs. Various concentrations of METH were spiked into urines, and the biosensor yielded recoveries more than 83.1%. In addition, the biosensor also showed a high sensitivity to detect cocaine. These results demonstrated that our colorimetric sensor holds promise to be implemented as a visual sensing platform to detect multiple illicit drugs in biological samples and environmental matrices.
Rapid Duplexed Detection of Illicit Drugs in Wastewater Using Gold Nanoparticle Conjugated Aptamer Sensors

Abstract:The abuse of illicit drug addiction is a worldwide public health and social problem. In this paper, we reported on a simple and rapid colorimetric biosensor for duplexed detection of methamphetamine (METH) and cocaine in a single assay. Gold nanoparticles (AuNPs) and Au@Ag NPs were synthesized and functionalized with DNA reporter probes (RPs) for METH and cocaine, respectively. The magnetic beads (MBs) were conjugated with two capture probes (CPs) respective to METH and cocaine. The respective RPs and CPs were designed to hybridize with each illicit drug-binding DNA aptamers through DNA-DNA hybridization, forming a sandwich structure. This MBs-based sandwich structure could be removed with an external magnetic field. However, due to the higher affinity of DNA aptamers with illicit drugs, the sandwich structure was disassembled when illicit drugs are introduced into the solution, leading to the colour changes of the supernatant. Utilizing a non-negative matrix factorization (NMF) algorithm to process the data, we demonstrated the ability of our biosensor for the simultaneous quantification of two illicit drugs. Under the optimal condition, our sensors were able to detect both METH and cocaine at the nM level with a wide dynamic range. This sensing platform provides a huge potential on drug consumption evaluation at the community level for wastewater-based epidemiology.
Aptamer-controlled gold nanozyme sensor for fluorescent and colorimetric dual-channel detection of methamphetamine

Abstract:Methamphetamine (METH) is the second abused drug which affects abusers' health and induces social crimes, developing novel methods with high sensitivity and selectivity for METH detecting is still challenging. In this paper, a colorimetric and fluorescent dual-channel sensor for METH has been constructed. We combine the enzyme-mimic catalytic activity of gold nanoparticles (GNPs) with high target specificity of METH aptamer to create a nanosensor (Apt-GNP), in the presence of METH, the absorption of 3,3',5,5'-tetramethylbenzidine (OxTMB) at 650 nm enhanced with METH concentration increasing, while the absorption characteristic peak of GNPs at 530 nm remained almost unchanged. The ratio of A650nm/A530nm and METH concentration had a good linear relationship when METH concentration was in the range of 5-50 μM, and the corresponding linear equation is A650nm/A530nm = 0.00727CMETH (μM) + 0.783 with R2 = 0.997 and LOD = 0.40 μM (LOD = 3σ/s, n = 11). Interestingly, the fluorescence emission of Rhodamine B (RB) overlaps with the absorption spectrum of OxTMB which represents the content of METH and the fluorescence signal of RB can be quenched through internal filtering effect (IEF). Hence, when RB was doped to the detection system, the decay of RB fluorescence can reflect the concentrations change of METH. Accordingly, the linear equation is F/FR = -0.00751CMETH (μM) + 0.895 with R2 = 0.993 and LOD = 0.40 μM, where F was the fluorescence of the analytical solution at 580 nm with METH and FR was fluorescence of RB control solution. The dual-channel sensor can measure METH in serum and artificial urine successfully which is potential to be applied in drug-using crime sites and provide direct evidence to law enforcement officials.
Colorimetric and bare eye determination of urinary methylamphetamine based on the use of aptamers and the salt-induced aggregation of unmodified gold nanoparticles

Abstract:Methamphetamine (METH) is second only to marijuana as a widely used illicit drug. We are presenting a simple colorimetric assay for sensitive and visual detection of METH in human urine using a METH-specific aptamer as the recognition element and unmodified gold nanoparticles as indicators. The method is based on the finding that the presence of METH results in AuNPs solution’s color change from red to blue. Normally, aptamers attach to the surface of AuNPs and thereby increasing their resistance to NaCl-induced aggregation. If, however, the aptamer bind to METH via G-quartets, rapid salt induced aggregation occurs associated with the formation of a blue colored solution. Urinary METH can be quantified via this effect either visually or by measurement of the absorbance ratios at 660 and 525 nm, respectively. It works in the 2 μM to 10 μM concentration range with a detection limit at 0.82 μM. The method is fast and also works well in human urine. Filtered Saliva for Rapid and Accurate Analyte Detection for POC Diagnostics

Abstract:Saliva has shown considerable promise as a diagnostic medium for point-of-care (POC) and over-the-counter (OTC) diagnostic devices due to the non-invasive nature of its collection. However, a significant limitation of saliva-based detection is undesirable interference in a sensor's readout caused by interfering components in saliva. In this study, we develop standardized sample treatment procedures to eliminate bubbles and interfering molecules while preserving the sample's target molecules such as spike (S) protein and glucose. We then test the compatibility of the pretreatment system with our previously designed SARS-CoV-2 and glucose diagnostic biosensing systems for detecting S protein and glucose in subject saliva. Ultimately, the effectiveness of each filter in enhancing biomarker sensitivity is assessed. The results show that a 20 mg nylon wool (NW) filter shows an 80% change in viscosity reduction with only a 6% reduction in protein content, making it an appropriate filter for the salivary S protein diagnostic system. Meanwhile, a 30 mg cotton wool (CW) filter is identified as the optimal choice for salivary glucose detection, achieving a 90% change in viscosity reduction and a 60.7% reduction in protein content with a minimal 4.3% reduction in glucose content. The NW pretreatment filtration significantly improves the limit of detection (LOD) for salivary S protein detection by five times (from 0.5 nM to 0.1 nM) and it reduces the relative standard deviation (RSD) two times compared to unfiltered saliva. Conversely, the CW filter used for salivary glucose detection demonstrated improved linearity with an R2 of 0.99 and a sensitivity of 36.6 μA/mM·cm2, over twice as high as unfiltered saliva. This unique filtration process can be extended to any POC diagnostic system and optimized for any biomarker detection, making electrochemical POC diagnostics more viable in the current market.

Abstract:We describe a novel biosensing strategy for sensitive detection of methylamphetamine (MAMP) based on surface enhanced Raman scattering (SERS) by the mediation of spacing between 4-mercaptobenzoic acid (4-MBA) labeled Au@Ag core-shell nanoparticles (Au@Ag). To achieve a favorable SERS substrate, Au@Ag shell-core nanoparticle was synthesized with seeds growth method and well characterized by SEM, TEM and UV-vis spectrometer. The uniform Au@Ag shows an excellent dispersion ability for SERS detection. Under the optimized conditions, the novel biosensor shows a good logarithm linear correlation with the concentration of MAMP ranging from 0.5 ppb to 40 ppb (R-2 = 0.986), with a limit of detection at 0.16 ppb of MAMP (3 sigma). Furthermore, our biosensors hold an excellent selectivity, demonstrated by the negligible interference from the detection of other illicit drugs and metabolites. The concentrations determined with our biosensor from spiked MAMP in human urine sample fell within the same range with the results from mass spectrometry. This indicates that our sensor has a clear potential for the rapid detection of illicit drug in real samples.

Abstract:Largely due to the high extinction coefficients and distance-dependent optical properties, metallic nanoparticles and quantum dots have been shown to be very attractive in DNA-related colorimetric and fluorescent assays, respectively. We have used DNAzymes (DNA molecules with catalytic activities) for directed assembly of gold nanoparticles and its application as colorimetric sensors for metal ions such as lead. This methodology has been expanded to an even broader range of molecules by using aptamers (nucleic acid-based binding molecules obtained through systematic evolution of ligands by exponential enrichment (SELEX)). We have developed a general sensor design method that is simple to design, easy to operate and gives fast color change with minimal materials consumption. To demonstrate the generality, sensors for adenosine and cocaine have been designed, both of which can produce color changes in seconds and at room temperature. Since no special feature on the aptamers is required, the design should be able to be used for any molecule of choice. Finally, to make the operation even easier to use and less venerable to errors, we have demonstrated dipsticks tests for a broad range of contaminants in water. These and other recent advances in this area will be summarized.
Social Comparison and Affect: A Study Among Elderly Women

Abstract:The author investigated how elderly women who recently moved to senior centers reacted in comparison to same-age persons who were either better off or worse off than were the former. The author expected 2 individual difference variables related to uncertainty about oneself—sense of control and social comparison orientation—to interact with social comparison direction to produce affect. Contrary to this expectation, the author observed only main effects of social comparison direction on affect. The results indicated that downward comparison produced more negative affect (and less positive affect) than did upward comparison, independently of participants' individual differences. The results provide some useful information on the social psychology of aging.

Abstract:Aptamers are single-stranded nucleic acids or peptides that can bind target molecules with high affinity and specificity. The conformation of an aptamer usually changes upon binding to its target analyte, and this property has been used in a wide variety of sensing applications, including detections based on fluorescence, electrochemistry, mass, or color change. Because native nucleic acids do not possess signaling moieties required for most detection methods, aptamer sensors usually involve labeling of external signaling groups. Among the many kinds of labels, inorganic nanoparticles are emerging as highly attractive candidates because some of their unique properties. Here, we describe protocols for the preparation of aptamer-linked gold nanoparticles (AuNPs) that undergo fast disassembly into red dispersed nanoparticles upon binding of target analytes. This method has been proven to be generally applicable for colorimetric sensing of a broad range of analytes. The sample protocols have also been successfully applied to quantum dots and magnetic nanoparticles. Finally, to increase the user friendliness of the method, the sensors have been converted into simple dipstick tests using lateral flow devices.

Abstract:This article reviews the recent developments of electrochemical sensors for the detection of disease‐signaling protein biomarkers in saliva bio‐samples. The role of nanomaterials in synergy with bio‐recognition probes and electrochemical sensing techniques for boosting the analytical performance of resultant sensors is described and discussed. Perspectives and views in this field are also given. The focus on precise medicine enhances the need for timely diagnosis and frequent monitoring of chronic diseases. Moreover, the recent pandemic of severe acute respiratory syndrome coronavirus 2 poses a great demand for rapid detection and surveillance of viral infections. The detection of protein biomarkers and antigens in the saliva allows rapid identification of diseases or disease changes in scenarios where and when the test response at the point of care is mandated. While traditional methods of protein testing fail to provide the desired fast results, electrochemical biosensors based on nanomaterials hold perfect characteristics for the detection of biomarkers in point‐of‐care settings. The recent advances in electrochemical sensors for salivary protein detection are critically reviewed in this work, with emphasis on the role of nanomaterials to boost the biosensor analytical performance and increase the reliability of the test in human saliva samples. Furthermore, this work identifies the critical factors for further modernization of the nanomaterial‐based electrochemical sensors, envisaging the development and implementation of next‐generation sample‐in‐answer‐out systems.

Abstract:Gold nanoparticles (AuNPs) with localized surface plasmon resonance effect have been widely used for colorimetric detection based on the interparticle plasmon coupling during AuNPs aggregation. However, it is still challenging to develop portable and quantitative methods with good sensitivity and excellent selectivity. In this study, a smartphone-based colorimetric assay is developed on the principle of surfactant-mediated AuNPs aggregation assisted with rolling circle amplification (RCA) on magnetic beads (MBs). The detection of adenosine is demonstrated as an example. The cetyl trimethyl ammonium bromide (CTAB) causes the negatively charged AuNPs to aggregate, which results in the color change from red to blue. When adenosine is in solution, the RCA process is triggered on the MBs because of specific adenosine-aptamer recognition, resulting in prolongation of single-stranded nucleic acid (ssDNA). The solution color remains red due to the electrostatic interaction between CTAB and ssDNA. Using this method, the limit of detection (LOD) for adenosine can be as low as 16 pM. Besides, it also works well in human serum. In addition, a portable device integrated with in-situ RGB analysis software is developed for the detection with a smartphone. This study offers a new strategy to improve the sensitivity and selectivity for the AuNPs-based colorimetric assay, taking advantages of specific aptamer recognition, in-situ RCA on MBs, magnetic separation, and smartphone-based portable device.

Abstract:The abuse of illicit drugs has been prevalent in recent years and is associated with crime and public health issues. To strengthen public security and fortify public health services with respect to the increasing severity of drug abuse, academic and government institutes have been devoted to constructing relative analytical methods for illicit drugs. To date, the development of sensors has been greatly emphasized due to their features of high sensitivity, prompt detection and flexible manipulation; thus, sensors can serve as alternatives to conventional sophisticated instruments. Recently, the use of nanomaterials has inspired the development of a series of innovative sample pretreatment and detection strategies in the field of analytical chemistry. Herein, this review elaborated the application of nanomaterials in analytical methods, including sample pretreatments, colorimetric sensors and fluorescent sensors. The utilization of nanomaterials in the analytical field provides novel perspectives for the development of detection platforms and facilitates the monitoring of illicit drugs in diverse complex matrices.

Aptamer-based Au Nanoparticles-Enhanced Surface Plasmon Resonance Detection of Small Molecules.

Abstract:Small molecules are difficult to detect by conventional SPR technique directly because the changes in the refractive index resulting from the binding processes of small biomolecules are often small. In order to extend the application of SPR biosensor in detecting a small molecule, we combine the advantage of aptamer technique with the amplifying effect of Au nanoparticles to design a sensitive SPR sensor for detecting small molecules. The principle of this sensor is based on surface inhibition detection. The aptamer is first immobilized on SPR gold film with its ss-DNA structure. The aptamer possessing this structure can be hybridized with Au nanoparticles-tagged complementary ss-DNA and result in a large change of SPR signal. However, the aptamer will change its structure from ss-DNA to tertiary structure after adenosine is added to the SPR cell. The aptamer possessing tertiary structure could not hybridize with Au nanoparticles-tagged complementary ss-DNA. Thus, the change of SPR signal resulted in the hybridization reaction between aptamer and Au nanoparticles-tagged complementary ss-DNA will decrease with the increase of the number of aptamers possessing tertiary structure, which is proportional to the concentration of the small molecule. Based on this principle, we choose a simple system (antiadenosine aptamer/adenosine) to detect the sensing ability of this SPR biosensor for a small molecule. The experimental results confirm that the SPR sensor we developed possesses a good sensitivity and a high selectivity for adenosine. The detection range for adenosine is from 1 x 10 (-9) to 1 x 10 (-6) M. More significantly, it is fairly easy to generalize this strategy to detect a spectrum of small molecules by SPR spectroscopy using different aptamers. Therefore, it is expected that this method may offer a new direction in designing high-performance SPR biosensors for sensitive and selective detection of a wide spectrum of small molecules.

Abstract:The misuse of kanamycin in the breeding industry can pose a threat to human health through food exposure. Therefore, it is crucial to monitor kanamycin (Kana) levels in food. This study presents a novel colorimetric approach for detecting kanamycin based on the aggregation of gold nanoparticles (AuNPs) induced by kanamycin. To achieve this, a single-standard DNA (ssDNA) aptamer was employed to bind the surface of AuNPs and maintain their dispersion under high salt concentrations. Upon adding Kana, the aptamer selectively binds to it and separates from the gold surface, resulting in the aggregation of AuNPs. This leads to a color change in the solution (from red to purple to blue) which can be observed under salt conditions. The proposed sensor demonstrated a linear range of 0.5 - 3 nM and a limit of detection (LOD) of 0.11 nM under optimal conditions. Its practicability was tested by monitoring kanamycin in three food samples, namely milk, honey, and vitamin C effervescent tablets, which resulted in satisfactory spiked recoveries. The sensor's miniaturization, convenience, simplicity, and low cost make it a desirable choice for fast and highly sensitive detection of Kana.

However, the effect of the biological sample matrix has not been fully explored. In this work, we investigated the salt-induced aggregation of AuNPs as well as DNA adsorption in serum and milk. AuNPs of 13, 30, and 50 nm were used as probes. The detection was successful only in a clean buffer but failed in serum or milk. Serum and milk exhibited excellent protective properties, even 250 mM NaCl added did not induce the aggregation of AuNPs. After centrifugation of milk, the supernatant did not protect the AuNPs, whereas the redispersed pellet showed protection. The limit concentration of serum to prevent AuNPs from aggregating was 0.04% for 13 nm AuNPs and 0.01% serum for 50 nm AuNPs. In addition, serum reduced DNA adsorption, and the DNA was adsorbed to the protein corona instead of directly to the AuNP surface. These two factors can explain the difficulty of detection in protein-containing samples. This study articulates the adsorption of proteins by AuNPs in biological samples and offers useful insights into the biosensor design.
A colorimetric aptasensor based on gold nanoparticles for detection of microbial toxins: an alternative approach to conventional methods

Abstract:Frequent contamination of foods with microbial toxins produced by microorganisms such as bacteria, fungi, and algae represents an increasing public health problem that requires the development of quick and easy tools to detect them at trace levels. Recently, it has been found that colorimetric detection methods may replace traditional methods in the field because of their ease of use, quick response, ease of manufacture, low cost, and naked-eye visibility. Therefore, it is suitable for fieldwork, especially for work in remote areas of the world. However, the development of colorimetric detection methods with low detection limits is a challenge that limits their wide applicability in the detection of food contaminants. To address these challenges, nanomaterial-based transduction systems are used to construct colorimetric biosensors. For example, gold nanoparticles (AuNPs) provide an excellent platform for the development of colorimetric biosensors because they offer the advantages of easy synthesis, biocompatibility, advanced surface functionality, and adjustable physicochemical properties. The selectivity of the colorimetric biosensor can be achieved by the combination of aptamers and gold nanoparticles, which provides an unprecedented opportunity to detect microbial toxins. Compared to antibodies, aptamers have significant advantages in the analysis of microbial toxins due to their smaller size, higher binding affinity, reproducible chemical synthesis and modification, stability, and specificity. Two colorimetric mechanisms for the detection of microbial toxins based on AuNPs have been described. First, sensors that use the localized surface plasmon resonance (LSPR) phenomenon of gold nanoparticles can exhibit very strong colors in the visible range because of changes caused by aggregation or disaggregation. Second, the detection mechanism of AuNPs is based on their enzyme mimetic properties and it is possible to construct a colorimetric biosensor based on the 3,3',5,5'-tetramethylbenzidine/Hydrogen peroxide, TMB/H 2 O 2 reaction to detect microbial toxins. Therefore, this review summarizes the recent applications of AuNP-based colorimetric aptasensors for detecting microbial toxins, including bacterial toxins, fungal toxins, and algal toxins focusing on selectivity, sensitivity, and practicality. Finally, the most important current challenges in this field and future research opportunities are discussed.
