Abstract:Methamphetamine (METH) is the second abused drug which affects abusers' health and induces social crimes, developing novel methods with high sensitivity and selectivity for METH detecting is still challenging. In this paper, a colorimetric and fluorescent dual-channel sensor for METH has been constructed. We combine the enzyme-mimic catalytic activity of gold nanoparticles (GNPs) with high target specificity of METH aptamer to create a nanosensor (Apt-GNP), in the presence of METH, the absorption of 3,3',5,5'-tetramethylbenzidine (OxTMB) at 650 nm enhanced with METH concentration increasing, while the absorption characteristic peak of GNPs at 530 nm remained almost unchanged. The ratio of A650nm/A530nm and METH concentration had a good linear relationship when METH concentration was in the range of 5-50 μM, and the corresponding linear equation is A650nm/A530nm = 0.00727CMETH (μM) + 0.783 with R2 = 0.997 and LOD = 0.40 μM (LOD = 3σ/s, n = 11). Interestingly, the fluorescence emission of Rhodamine B (RB) overlaps with the absorption spectrum of OxTMB which represents the content of METH and the fluorescence signal of RB can be quenched through internal filtering effect (IEF). Hence, when RB was doped to the detection system, the decay of RB fluorescence can reflect the concentrations change of METH. Accordingly, the linear equation is F/FR = -0.00751CMETH (μM) + 0.895 with R2 = 0.993 and LOD = 0.40 μM, where F was the fluorescence of the analytical solution at 580 nm with METH and FR was fluorescence of RB control solution. The dual-channel sensor can measure METH in serum and artificial urine successfully which is potential to be applied in drug-using crime sites and provide direct evidence to law enforcement officials.
G-quadruplex–hemin DNAzyme molecular beacon probe for the detection of methamphetamine

Abstract:In this work, a simple, cost-effective, and label-free biosensor was constructed for methamphetamine (METH) detection. The biosensor consists of a G-quadruplex–hemin DNAzyme molecular beacon (DNAzyme MB), a METH aptamer, and a colorimetric substrate. The DNAzyme MB loses peroxidase activity when it hybridizes with the METH aptamer. In the presence of METH, DNAzyme MB dissociates from the inactive hybrid due to preferable hybridization of METH with the aptamer. This process recovers the activity of DNAzyme MB, which catalyzes a reaction with the colorimetric substrate to yield measurable signals. Under optimized conditions, a detection limit as low as 0.5 nM (74.6 ng L−1) was achieved. Common illicit drugs were found to have little interference on detection of METH. Recoveries of METH spiked in urines of addicts were greater than 85%. Good agreement was observed between METH concentrations in urines determined by the sensor and by liquid chromatography-tandem mass spectrometer. These results indicate that the G-quadruplex–hemin DNAzyme MB probe holds promise to detect METH not only in biological samples, but also in environmental matrices.

Abstract:A simple and rapid electrochemical sensing method with high sensitivity and specificity of aptamers was developed for the detection of methylamphetamine (MAMP). A short anti-MAMP thiolated aptamer (Apt) with a methylene blue (MB) probe at 3ʹ-end was immobilized on the surface of a gold electrode (MB-Apt-S/GE). The electrochemical signal appeared when MAMP presenting in the sample solution competed with cDNA for binding with MB-Apt-S. Under optimized conditions, the liner range of this signal-on electrochemical aptasensor for the detection of MAMP achieved from 1.0 to 10.0 nmol/L and 10.0–400 nmol/L. LOD 0.88 nmol/L were obtained. Satisfactory spiked recoveries of saliva and urine were also obtained. In this method, only 5 min were needed to incubate before the square wave voltammetry (SWV) analysis, which was much more rapid than other electrochemical sensors, leading to a bright and broad prospect for the detection of MAMP in biological sample. This method can be used for on-site rapid detection on special occasions, such as drug driving scenes, entertainment venues suspected of drug use, etc.

Abstract:Methamphetamine (METH) abuse has become a global problem, and new strategies for rapid and sensitive detection of METH are in urgent need. Herein, a novel technique is developed for highly sensitive detection of METH in solution with a broad concentration range by the assembly of AIE luminogen of tetrakis(4-(pyridin-4-yl)phenyl)ethene (TPPE) and cucurbit[7]uril (CB[7]). The H-1 NMR and isothermal titration calorimetry (ITC) characterizations confirm that TPPE and CB[7] are self-assembled in a ratio of 1:4. While, the interaction of CB[7] with METH exhibits a binding ratio of 1:1 and stronger binding ability than with TPPE. Based on the competitive binding affinity, CB[7]superset of TPPE could be used as a chemosensor to detect METH in solution, which exhibits remarkable advantages such as rapid response, high sensitivity and low limit of detection (LOD). When the concentration of METH solution is larger than 40 mu M, the enhancement of fluorescence intensity and change of fluorescent colour could be directly observed upon UV irradiation. When the METH solution concentration is higher than 10 mu M, we can use our fabricated test strips to rapidly and visibly detect. If above detections do not work, we can measure the photoluminescence (PL) spectra of the sensing system, which shows a LOD as low as 0.43 mu M. This unique CB[7]superset of TPPE based chemosensor is potentially applicable in real-time and visual detection of METH in solution and provides a powerful platform for preventing chemical solvent transport.

Abstract:Methamphetamine (METH) is second only to marijuana as a widely used illicit drug. We are presenting a simple colorimetric assay for sensitive and visual detection of METH in human urine using a METH-specific aptamer as the recognition element and unmodified gold nanoparticles as indicators. The method is based on the finding that the presence of METH results in AuNPs solution’s color change from red to blue. Normally, aptamers attach to the surface of AuNPs and thereby increasing their resistance to NaCl-induced aggregation. If, however, the aptamer bind to METH via G-quartets, rapid salt induced aggregation occurs associated with the formation of a blue colored solution. Urinary METH can be quantified via this effect either visually or by measurement of the absorbance ratios at 660 and 525 nm, respectively. It works in the 2 μM to 10 μM concentration range with a detection limit at 0.82 μM. The method is fast and also works well in human urine. Abstract:We describe a novel biosensing strategy for sensitive detection of methylamphetamine (MAMP) based on surface enhanced Raman scattering (SERS) by the mediation of spacing between 4-mercaptobenzoic acid (4-MBA) labeled Au@Ag core-shell nanoparticles (Au@Ag). To achieve a favorable SERS substrate, Au@Ag shell-core nanoparticle was synthesized with seeds growth method and well characterized by SEM, TEM and UV-vis spectrometer. The uniform Au@Ag shows an excellent dispersion ability for SERS detection. Under the optimized conditions, the novel biosensor shows a good logarithm linear correlation with the concentration of MAMP ranging from 0.5 ppb to 40 ppb (R-2 = 0.986), with a limit of detection at 0.16 ppb of MAMP (3 sigma). Furthermore, our biosensors hold an excellent selectivity, demonstrated by the negligible interference from the detection of other illicit drugs and metabolites. The concentrations determined with our biosensor from spiked MAMP in human urine sample fell within the same range with the results from mass spectrometry. This indicates that our sensor has a clear potential for the rapid detection of illicit drug in real samples.

Abstract:New drugs and illicit synthesized mixtures detection at crime scenes is a great challenge for detection method, which requires anti-interference and ultrasensitive methods to detect methamphetamine (METH) in seized street samples and biological fluids. Herein, we constructed a surface-enhanced Raman sensing method based on aligner mediated cleavage (AMC) of nucleic acid for quantitative detection of METH for the first time. This method we proposed relied on AMC to achieve programmable sequence-specific cleavage of METH aptamer linked by gold nanoparticles (METH aptamer-Au NPs), the cleavage product-Au NPs conjugates (cleavage aptamer-Au NPs) would hybridize with complementary DNA (cDNA)-Au NPs, resulting in the aggregation of the Au NPs and concomitant plasmonic coupling effect. Besides, due to the base number of METH aptamer-Au NPs was decreased, the interparticle distance of the Au NPs was shortened, which increased the electric field enhancement factor. Thus, under the irradiation of the laser, rhodamine 6G (R6G) adsorbed on Au NPs generated a strong Raman signal. The detection limit reached 7 pM, the linear range was from 10 pM to 10 nM, and this detection method also showed good anti-interference ability and reproducibility in serum. (C) 2020 Elsevier B.V. Abstract:The realization of drug detection in drug-using crime sites can provide law enforcement officials with direct evidence. This research has developed and demonstrated an easy-to-use handheld sensor that can quickly detect methamphetamine (MA) in the field. The core of the handheld UV spectrophotometer device (HUVSD) is the STM32F103 series of single-chip micro-controller, which has a 32-bit microcontroller and two embedded 12-bit high-precision analog-to-digital converter (ADC) modules. Through Bluetooth-wireless transmission protocol, the spectral information can be displayed in the cell phone's app, and it is possible to visually determine whether the test sample contains methamphetamine hydrochloride substances based on the characteristic peak at 410 nm. The readily available and inexpensive inducible compound 3A and the phosphate pillar[5]arene@silver nanoparticle (PP5@AgNPs) colloidal solution were used as the reactants. The PP5@AgNPs colloidal solution and 3A were mixed and reacted at room temperature, and the color changed to gray-black. The color change was caused by the aggregation of AgNPs induced by the molecular recognition between the induction compound 3A and PP5 on the AgNPs surface. After continuing to add the drug MA, the color of the colloidal solution turned yellow again. This is due to the occurrence of competitive molecular recognition, and the interaction between PP5 and 3A/MA was investigated by molecular docking simulations. The HUVSD has high sensitivity, simple equipment, time-saving, color change visualization and suitable for on-site deployment. It only needs a Pasteur pipette, which has great potential to realize rapid on-site detection.

Abstract:We report a novel colorimetric biosensor based on non-aggregation Au@Ag core-shell nanoparticles to detect methamphetamine and cocaine. The biosensor consisted of a reporter probe (RP) that is a specific single-stranded DNA (ssDNA) sequence coated on Au@Ag nanoparticles, a capture probe (CP) conjugated with magnetic beads, and an illicit drug-binding DNA aptamer (Apt). Au@Ag nanoparticles were synthesized by seed growth and characterized by scanning electron microscope (SEM), high-resolution transmission electron microscopy (HR-TEM), and UV-vis spectra. Methamphetamine (METH) was used as an example to evaluate the feasibility of the biosensor and to optimize the detection conditions. We demonstrated that this sensing platform was able to detect as low as 0.1nM (14.9ngL-1) METH with a negligible interference from other common illicit drugs. Various concentrations of METH were spiked into urines, and the biosensor yielded recoveries more than 83.1%. In addition, the biosensor also showed a high sensitivity to detect cocaine. These results demonstrated that our colorimetric sensor holds promise to be implemented as a visual sensing platform to detect multiple illicit drugs in biological samples and environmental matrices.

Abstract:Methamphetamine (MA), a psychoactive substance with many medicinal applications in different countries, has destructive impacts on the nervous system and brain and can lead to addiction. The optimal system for MA determination must be able to measure the tiny amount of MA in complex matrixes accurately. In the current work, a simple and biocompatible sensitive optical probe was developed based on molecularly imprinted polymers (MIPs) technique and by using green CQDs and mesoporous structured imprinting microspheres (SiO2@CQDs@ms-MIPs). CQDs (ФF = 33%) were synthesized via the hydrothermal method using natural chewing gum as carbon source. SiO2 nanoparticles were used as the backup substrate for the placement of CQDs. In spite of biocompatibility, porosity and having high specific area are the unique features of SiO2 nanoparticles. When MA is present, the fluorescence response of MIPs enhances. This is caused by the passivation and adjustment of active clusters that are present on the surface of CQDs. By this optical sensor, the favorable linear dynamic range (5.0-250 μM) and the detection limit (1.6 μM) were obtained. The applicability of the advanced sensor was studied in real samples such as human urine and human blood plasma. Acceptable results were obtained and recovery amounts were in the 92-110% interval.
Rapid Duplexed Detection of Illicit Drugs in Wastewater Using Gold Nanoparticle Conjugated Aptamer Sensors

Abstract:The abuse of illicit drug addiction is a worldwide public health and social problem. In this paper, we reported on a simple and rapid colorimetric biosensor for duplexed detection of methamphetamine (METH) and cocaine in a single assay. Gold nanoparticles (AuNPs) and Au@Ag NPs were synthesized and functionalized with DNA reporter probes (RPs) for METH and cocaine, respectively. The magnetic beads (MBs) were conjugated with two capture probes (CPs) respective to METH and cocaine. The respective RPs and CPs were designed to hybridize with each illicit drug-binding DNA aptamers through DNA-DNA hybridization, forming a sandwich structure. This MBs-based sandwich structure could be removed with an external magnetic field. However, due to the higher affinity of DNA aptamers with illicit drugs, the sandwich structure was disassembled when illicit drugs are introduced into the solution, leading to the colour changes of the supernatant. Utilizing a non-negative matrix factorization (NMF) algorithm to process the data, we demonstrated the ability of our biosensor for the simultaneous quantification of two illicit drugs. Under the optimal condition, our sensors were able to detect both METH and cocaine at the nM level with a wide dynamic range. This sensing platform provides a huge potential on drug consumption evaluation at the community level for wastewater-based epidemiology.
Integrated Sensing Platform Validated for the Efficient and On-Site Screening of Amine-Containing Illicit Drugs

Abstract:Efficient and reliable technologies for the on-site detection of illicit drugs are important in drug-facilitated crime investigations. However, the development of such technologies is challenging. Based on the synthetic optimization, introducing a boron ester functional group to the two furanic indicators endows the stimulus-responsive properties synergistically. The ring-opening reaction of the indicators in the presence of amine-containing illicit drugs generated well-known donor-acceptor Stenhouse adducts, accompanied by strong color changes. A small-size and lightweight laminated sensor was integrated based on the outstanding ratiometric variations of the two active furanic indicators. A prototype platform was fabricated equipped with a circuit control, a mini pump, and a signal processing system. A user-friendly detection and efficient screening of amine-containing illicit drugs, including phenethylamines, amphetamines, cathinones, and tryptamines in the liquid states were conducted. The ratiometric response of the sensor was linear in the concentration range of 2.1-10.6 μg·mL-1 for methamphetamine·HCl and methcathinone ·HCl. The detection limits for the two illicit drugs at the sublevel (ng·mL-1) were found to be 8.4 and 9.0 ng·mL-1, respectively. Double-blind field tests and different illicit drugs were evaluated with good screening capability. Successful trials showed the potential applications of the developed prototype platform for efficient and on-site analytical determination.
Two-stage tricuspid valve replacement for mixed bacterial endocarditis.

Abstract:Mixed Staphylococcus aureus and Enterobacter endocarditis of the tricuspid valve in a heroin addict failed to respond to large doses of antibiotics to which these organisms were sensitive. However, following tricuspid valvulectomy, the synergistic combination of cefanone and gentamicin sulfate (Garamycin) effectively eradicated the infection. Though the patient tolerated absence of the tricuspid valve for eight weeks, his exercise capacity was limited until a tricuspid valve prosthesis was inserted. The Enterobacter strain isolated from this patient inactivated penicillin and oxacillin in broth culture. Studies of a sonicate from this organism demonstrated considerable β-lactamase activity against penicillin and oxacillin and less against cefanone. Those findings suggest a mechanism by which the presence of Enterobacter perpetuated staphylococcal endocarditis despite prolonged therapy with antibiotics.

Abstract:Effective detection of methamphetamine(Met)requires a fast,sensitive,and cheap testing assay.However,commer-cially available methods require expensive instruments and highly trained operators,which are time-consuming and labor-in-tensive.Herein,an antibody-modified graphene transistor assay is developed for sensitive and minute-level detection of Met in complex environments.The anti-Met probe captured charged targets within 120 s,leading to a p-doping effect near the graphene channel.The limit of detection reaches 50 aM(5.0×10-17 M)Met in solution.The graphene transistor would be a valu-able tool for Met detection effective prevention of drug abuse.
Red blood cell storage age – what we know from clinical trials

Abstract:Red blood cell (RBC) transfusion has become one of the most commonly utilized life-saving therapies in medicine with approximately 12–16 million units transfused in the US per year [1,2]. Since storage solutions were developed 100 years ago, they have been modified to allow for increased storage duration, primarily to increase availability, especially in remote areas, such as during combat operations [3]. Currently in the US and Europe, RBCs can be stored up to 42–56 days based upon at least 75% of radiochromium-labeled cells remaining in the circulation at 24 h after infusion and hemolysis less than 1% [4,5]. As RBCs are stored over time, the metabolic and biochemical changes that occur have been coined ‘the storage lesion.’ To further improve the availability of RBCs, it is standard for blood distributors to transfer RBCs that are close to expiration at community hospitals to medical centers where utilization rates are higher, and for hospital blood banks to issue the ‘first-in, first-out’ inventory to reduce RBC outdating and waste [2,6]. A consequence of these policies focused on inventory management is that patients with increased severity of illness are transfused RBCs of increased storage duration compared to less acutely ill patients. It is important to recognize that as storage duration for RBC units has increased for logistical purposes, there has not been a commensurate evaluation of clinical efficacy or safety. As a result of the preferential allocation of older RBCs to sicker patients, there has been concern regarding the quality of RBCs after prolonged storage related to both efficacy and safety. This has led to a significant amount of research into the clinical relevance of the storage lesion with laboratory, preclinical, and clinical studies [4,7–9]. The results of these studies suggested biologic plausibility that the RBC storage lesion was potentially able to affect outcomes, especially in critically ill populations. These studies though are limited due to potential bias and higher-quality studies were needed to more accurately determine if RBC storage age affects outcomes. Consequently, a few randomized controlled trials (RCTs) have been completed, and a few are still underway. Abstract:A novel polydimethylsiloxane (PDMS)-poly-L-lysine (PLL) microfluidic chip, comprised of 24 reaction channels with 2.5,L volume of each reaction channel only, was proposed for quantitative analysis of methamphetamine (MET) based on time-resolved immunoassay techniques. The chip utilized the adsorption characteristics of PDMS and PLL towards proteins to immobilize MET complete antigens (MET-BSA) on the surface of reaction channel. Then the competition reaction could happen between MET-Ag in the sample solution and MET-BSA on the inner surfaces of the reaction channels with MET-Ab in the reagent. The surface of latex microspheres was labeled by lanthanide, which could emit red fluorescence under the exposure of ultraviolet (UV). Based on the principle of competitive immunoassay, the more MET-Ag, the less the fluorescence intensity in the reaction channel. The detection results of this chip were acquired using UV irradiation fluorescence imaging method. With this method, 24 samples could be detected and analyzed simultaneously on a chip by just taking the fluorescence image of the chip. The method allows the detection of MET antigens ranging from 100 ng/mL to 3000 ng/mL, with less sample consumption and high-throughput. This chip is suitable for the police preliminary screening work and has a good application prospects.
Social Comparison and Affect: A Study Among Elderly Women

Abstract:The author investigated how elderly women who recently moved to senior centers reacted in comparison to same-age persons who were either better off or worse off than were the former. The author expected 2 individual difference variables related to uncertainty about oneself—sense of control and social comparison orientation—to interact with social comparison direction to produce affect. Contrary to this expectation, the author observed only main effects of social comparison direction on affect. The results indicated that downward comparison produced more negative affect (and less positive affect) than did upward comparison, independently of participants' individual differences. The results provide some useful information on the social psychology of aging.
Rationally Designed Ionically Conductive Metal‐Organic Frameworks for Ultrasensitive Methamphetamine Analogs Sensor

Abstract:Tailored ionically conductive metal‐organic framework (IC‐MOFs) sensor exhibited the excellent sensing performance toward the simulator of addictive drugs (N‐methylphenethylamine) at room temperature, including rapid response time, excellent selectivity, low theoretical detection limit and ultrahigh long‐term stability, owing to its strong binding interaction and fast ionic migration. It provides advanced insights on the exploitation of smart sensor for ice‐drug detection and various healthcare applications. Achieving convenient, sensitive, low‐cost, and non‐contact detection of trace addictive drugs is a challenging problem. Herein, a novel ionically conductive metal‐organic framework (IC‐MOFs) is designed through a controlled interface assembly strategy. The active metal anions are incorporated into the layered MOFs with a porous structure, forming charge carriers that served as effective adsorption/binding sites for N‐methylphenethylamine (MPEA), a crucial simulator of addictive drugs. The in situ integrated Zn3(HHTP)2‐MOF sensor device designed in this study demonstrated real‐time detection of sub‐ppb level MPEA at room temperature, with an exceptionally low theoretical detection limit of 20 ppt. The overall sensing performance of this sensor surpassed all previously reported chemical sensors for detecting methamphetamine. The Zn3(HHTP)2‐MOF sensor exhibited outstanding selectivity, rapid response time (ca. 5 s), excellent long‐term stability, amenable miniaturization, and device consistency. The device successfully passed dual 85 test (500 h at 85 °C and 85% of humidity), which is rarely report previously. Density functional theoretical calculations (DFT) and spectral characterization confirmed that the prominent selectivity of Zn3(HHTP)2‐MOF toward MPEA is attributed to the strong binding ability. The general and straightforward strategy provides brand‐new route to exploiting smart sensors for drug prevention and surveillance.

Abstract:The escalating issue of drug abuse poses a significant threat to public health and societal stability worldwide. An on-site drug detection platform is vital for combating drug abuse and trafficking, as it eliminates the need for additional tools, extensive processes, or specialized training. Therefore, it is imperative to develop a fast, sensitive, non-invasive, and reliable multiplex drug testing platform. In this study, we have presented a silica core@dual quantum dot-shell nanocomposite (SI/DQD)-based fluorescent lateral flow immunoassay (LFIA) platform for the highly sensitive and simultaneous point-of-care (POC) detection of methamphetamine (MET) and tramadol (TR). A 3D-printed attachment was designed to integrate optical and electrical components, facilitating the miniaturization of the instrument and reducing both cost and complexity. The device's advanced hardware and effective fluorescence extraction algorithm with waveform reconstruction enable swift, automatic noise reduction and data analysis. SI/DQD nanocomposites were utilized as fluorescent nanotags in the LFIA strips due to their outstanding luminous efficiency and robustness. This LFIA platform achieves impressive detection limits (LODs) of 0.11 ng mL −1 for MET and 0.017 ng mL −1 for TR. The method has also successfully detected MET and TR in complex biological samples, demonstrating its practical application capabilities. The proposed fluorescent LFIA platform, based on SI/DQD technology, holds significant promise for the swift and accurate POC detection of these substances. Its affordability, compact size, and excellent analytical performance make it suitable for on-site drug testing, including at borders and roadside checks, and open up new possibilities for the design and implementation of drug testing methods.

Abstract:<p>Aptamer electrochemical sensors using immobilized aptamers with redox tag rely on the target binding-induced changes of current signal on electrode, offering advantages in operation convenience, no separation, rapidity, and sensitivity. Usually, the redox tag is placed on aptamer terminal, however, sometimes the terminal label may be insensitive to target-binding and fail to generate sensitive responses. The redox tag methylene blue (MB) labeled on different sites of aptamer may experience distinct changes in local environment, distance to electrode, or interactions with aptamer bases during affinity binding, which affect the current signal. Thus, it is possible to construct aptamer electrochemical sensors with sensitive and significant responses to targets by screening a series of sites (e.g., internal thymine T) of the aptamer and placing MB tag on a specific site of the aptamer. With this strategy, we successfully fabricated an electrochemical sensor on gold electrode for rapid, reagentless, and sensitive detection of aflatoxin B1 (AFB1), an important mycotoxin causing great health risks, by using a 26-mer DNA aptamer with MB on an internal T site (e.g., 18th T) and a thiol moiety at 5′ terminal. This sensor generated remarkable signal-on responses to AFB1, allowed a detection limit of 6 pM, and enabled detection of AFB1 in wine, milk and corn flour samples. This sensor can be well regenerated by rinsing with deionized water and reused, and shows good stability. This sensor and the demonstrated strategy are promising in wide applications.</p>
