Abstract:A highly diastereoselective [3 + 2] cycloaddition of aziridines with difluorinated silyl enol ethers has been developed. This approach provides a facile methodology for highly functionalized gem-difluorinated pyrrolidines in good to excellent yields with good functional group tolerance. A one-pot, two-step approach for synthesis of structurally interesting fluorinated pyrroles has also been developed through a cycloaddition/aromatization/desulfonation sequence. Moreover, readily available substrates, mild reaction conditions, and easy scale-up synthesis show practical advantages.

Abstract:Benzofuran-derived azadienes (BDAs) have been widely used as four-atom synthons in transition-metal-mediated cycloaddition reactions, while the exploitation of their reactivity as a two-atom unit to construct spirocycles is still underdeveloped. Herein, we reported the first palladium(0)-catalyzed diastereo- and enantioselective [3 + 2] annulation of vinylcyclopropanes (VCPs) and BDAs. This transformation is featured with a broad substrate scope (31 examples), allowing for facile access to a variety of enantioenriched spirocycles bearing a quaternary stereogenic center in good yields with excellent regio-, diastereo-, and enantioselectivities (up to 93% yield, >20:1 dr, and mostly >99% ee) under mild reaction conditions. Abstract:A palladium(ii)-catalyzed intramolecular oxidative aza-Wacker-type reaction of vinyl cyclopropanecarboxamides to access a series of conformationally restricted highly substituted aza[3.1.0]bicycles is reported. The transformation proceeded through a typical aza-Wacker reaction mechanism to forge a new C-N bond with oxygen as the terminal oxidant. The desired fused heterocycles were obtained in moderate yields. The process is tolerant of a range of functional aryl groups under mild conditions.

Abstract:A TfOH-promoted synthesis of fluorinated poly-fused heterocycles via the cascade cyclization of azadienes and difluoroenoxysilanes has been developed, leading to the facile construction of fluorinated benzofuro[3,2-b]pyridines, 5H-indeno-[1,2-b]pyridines, and 5,6-dihydrobenzo[h]quinolines. This one-pot formal [4 + 2] approach involves 1,4-difluoroalkylation, desulfo-nylation, cyclization, and dehydrated and dehydrofluorinated aromatization and represents the first application of difluoroenoxysilane in cascade transformations. Furthermore, this methodology is highlighted by the synthesis of three fluoro analogues of bioactive molecules with potent topoisomerase inhibitory activities.

Abstract:A facile and highly efficient three-component reaction of thiazole or benzothiazole carbenes, disubstituted ketenes, and activated alkynes is disclosed. With this methodology, a polysubstituted ring system containing furo[2,3-c]thiazepine core can be constructed from simple and readily accessible starting materials in good yields. The scope and limitation of this transformation were investigated in detail by using various thiazole carbene, ketene, and alkyne components. Furthermore, the synthetic utilities of these unique polyheterocyclic compounds were demonstrated via their Diels-Alder reactions with benzynes to furnish thiazepine-fused 7-oxanorbornadiene derivatives in excellent yields.

Abstract:In this paper, we reported the palladium-catalyzed formal [5 + 4] cycloaddition reactions between 4-vinyl-4-butyrolactones (VBLs) and azadienes. Under mild reaction conditions, a wide range of benzofuran-fused 9-membered heterocyclic compounds had been provided in moderate to excellent yields with exclusive regioselectivities and excellent diastereoselectivities. The practical applicability of the synthesis was demonstrated through scale-up reaction and further transformation.

Abstract:The development of electron-demand disfavored [4 + 2] cycloaddition of two electron-deficient reacting partners poses a considerable challenge. An enantioselective aza-[4 + 2] cycloaddition of electron-deficient N-sulfonyl-1-aza-1,3-dienes is possible with vinyl ketones via phosphine catalysis, which provides facile access to a wide range of enantioenriched trifluoromethylated tetrahydropyridines in up to 97% yield with 97% ee and >20:1 dr. The mechanistic study indicated that this cycloaddition proceeds via a tandem intermolecular aza-Rauhut–Currier/intramolecular aza-Michael addition reaction.

Abstract:Although cycloadditions of bicyclobutanes (BCBs) have emerged as a reliable approach for producing bicyclo[n.1.1]alkanes such as azabicyclo[3.1.1]heptanes (aza-BCHeps), serving as saturated bioisosteres of arenes, the catalytic asymmetric variant remains underdeveloped and presents challenges. Herein, we developed several Lewis acid-catalyzed systems for the challenging dearomatizing (3+3) cycloaddition of BCBs and aromatic azomethine imines. This resulted in fused 2,3-diazabicyclo[3.1.1]heptanes, introducing a novel chemical space for the caged hydrocarbons. Moreover, an asymmetric Lewis acid catalysis strategy was devised for the (3+3) cycloadditions of BCBs and N-iminoisoquinolinium ylides, forming chiral diaza-BCHeps with up to 99% yield and 97% ee. This work represents the first successful demonstration of asymmetric (3+3) cycloaddition by introducing a chiral environment through the activation of BCBs.

Abstract:A new synthetic application of vinyl aziridines as N-containing three-atom components in a rhodium-catalyzed [4+3] cycloaddition reaction is described. The reaction proceeds well with various silyl dienol ethers and vinyl aziridines, and enables the efficient synthesis of highly functionalized azepines in an enantioselective manner with net inversion of absolute configuration. The salient features of the transformation include the use of readily available substrates, high selectivity, and mild reaction conditions, as well as the versatile functionalization of the products.

Abstract:The chemoselective [4+2] annulation/aromatization reactions between benzofuran‐derived azadienes and N‐Ts cyanamides are developed, affording a convenient method for synthesizing benzofuro[3,2‐d]pyrimidin‐2‐amines under mild conditions. Herein, N‐Ts cyanamides selectively participated in reactions absolutely via carbodiimide anion intermediates and the corresponding cyanamide anion intermediates derived products were not observed. The proposed chemoselective stepwise reaction mechanism was well supported by DFT calculations.

Abstract:Herein, we have developed a method for the construction of spiro[benzofuran-2,2'-bicyclo[2.1.1]hexanes] via BF3·Et2O-catalyzed formal [2π + 2σ] cycloaddition of bicyclo[1.1.0]butanes with benzofuran-derived oxa(aza)dienes. This transformation allowed for facile access to a variety of functionalized spiro-bicyclo[2.1.1]hexanes in good yields (up to 99% yield) with excellent regioselectivities and a broad substrate scope (34 examples) under mild reaction conditions. Moreover, the synthetic utility of the cycloadducts were further emphasized through a scale-up experiment and subsequent synthetic transformations.

Abstract:The construction of eight-membered N-heterocycles with good diastereo-and enantioselective control remains a formidable challenge in asymmetric catalysis. Herein, we report a palladium-catalyzed asymmetric [4+4] dipolar cycloaddition of aliphatic 1,4-dipoles with azadienes, furnishing chiral benzofuran- and indene-fused eight-membered N-heterocycles in good yields (up to 86%) and excellent stereoselectivities (up to >20:1 dr, 95% ee). This reaction not only represents the first catalytic asymmetric [4+4] cyclization of this aliphatic 1,4-dipoles, but also provides a new and efficient way to construct a variety of novel chiral fused eight-membered N-heterocycles.
