Abstract:Metagenomic studies on prokaryotic diversity of hypersaline soils from the Odiel saltmarshes, South-west Spain, revealed a high proportion of genomic sequences not related to previously cultivated taxa, that might be related to haloarchaea with a high environmental and nutritional flexibility. In this study, we used a culturomics approach in order to isolate new haloarchaeal microorganisms from these hypersaline soils. Four haloarchaeal strains, designated strains F24AT, F28, F27T, and F13T, phylogenetically related to the genus Halomicroarcula, were isolated and characterized in detail. The phylogenomic tree based on the 100 orthologous single-copy genes present in the genomes of these four strains as well as those of the type strains of the species Halomicroarcula pellucida CECT 7537T, Halomicroarcula salina JCM 18369T and Halomicroarcula limicola JCM 18640T, that were determined in this study, revealed that these four new isolates clustered on three groups, with strains F24AT and F28 within a single cluster, and altogether with the species of Halomicroarcula. Additionally, Orthologous Average Nucleotide Identity (OrthoANI), digital DNA-DNA hybridization (dDDH) and Average Amino-acid Identity (AAI) values, likewise phenotypic characteristics, including their polar lipids profiles, permitted to determine that they represent three new species, for which we propose the names Halomicroarcula rubra sp. nov. (type strain F13T), Halomicroarcula nitratireducens sp. nov. (type strain F27T) and Halomicroarcula salinisoli sp. nov. (type strain F24AT). An in deep comparative genomic analysis of species of the genus Halomicroarcula, including their metabolism, their capability to biosynthesize secondary metabolites and their osmoregulatory adaptation mechanisms was carried out. Although they use a salt-in strategy, the identification of the complete pathways for the biosynthesis of the compatible solutes trehalose and glycine betaine, not identified before in any other haloarchaea, might suggest alternative osmoadaptation strategies for this group. This alternative osmoregulatory mechanism would allow this group of haloarchaea to be versatile and eco-physiologically successful in hypersaline environments and would justify the capability of the species of this genus to grow not only on environments with high salt concentrations [up to 30% (w/v) salts], but also under intermediate to low salinities.

Abstract:The KTK 4A-related Thermoplasmata thrives in the sediment of saline lakes; however, systematic research on its taxonomy, environmental adaptation and metabolism is lacking. Here, we detected this abundant lineage in the sediment of five artificially separated ponds (salinity 7.0%-33.0%) within a Chinese soda-saline lake using culture-independent metagenomics and archaeal 16S rRNA gene amplicons. The phylogenies based on the 16S rRNA gene, and 122 archaeal ubiquitous single-copy proteins and genome-level identity analyses among the metagenome-assembled genomes demonstrate this lineage forming a novel order, Candidatus Haloplasmatales, comprising four genera affiliated with the identical family. Isoelectric point profiles of predicted proteomes suggest that most members adopt the energetically favourable 'salt-in' strategy. Functional prediction indicates the lithoheterotrophic nature with the versatile metabolic potentials for carbohydrate and organic acids as well as carbon monoxide and hydrogen utilization. Additionally, hydrogenase genes hdrABC-mvhADG are linked with incomplete reductive citrate cycle genes in the genomes, suggesting their functional connection. Comparison with the coupling of HdrABC-MvhADG and methanogenesis pathway provides new insights into the compatibility of laterally acquired methanogenesis with energy metabolism in the related order Methanomassiliicoccales. Globally, our research sheds light on the taxonomy, environmental adaptative mechanisms, metabolic potentials and evolutional significance of Ca. Haloplasmatales.

Decoding the Genomic Profile of the Halomicroarcula Genus: Comparative Analysis and Characterization of Two Novel Species

Abstract:The genus Halomicroarcula, classified within the family Haloarculaceae, presently comprises eight haloarchaeal species isolated from diverse saline habitats, such as solar salterns, hypersaline soils, marine salt, and marine algae. Here, a detailed taxogenomic study and comparative genomic analysis of the genus Halomicroarcula was carried out. In addition, two strains, designated S1CR25-12T and S3CR25-11T, that were isolated from hypersaline soils located in the Odiel Saltmarshes in Huelva (Spain) were included in this study. The 16S rRNA and rpoB’ gene sequence analyses affiliated the two strains to the genus Halomicroarcula. Typically, the species of the genus Halomicroarcula possess multiple heterogeneous copies of the 16S rRNA gene, which can lead to misclassification of the taxa and overestimation of the prokaryotic diversity. In contrast, the application of overall genome relatedness indexes (OGRIs) augments the capacity for the precise taxonomic classification and categorization of prokaryotic organisms. The relatedness indexes of the two new isolates, particularly digital DNA–DNA hybridization (dDDH), orthologous average nucleotide identity (OrthoANI), and average amino acid identity (AAI), confirmed that strains S1CR25-12T (= CECT 30620T = CCM 9252T) and S3CR25-11T (= CECT 30621T = CCM 9254T) constitute two novel species of the genus Halomicroarcula. The names Halomicroarcula saliterrae sp. nov. and Halomicroarcula onubensis sp. nov. are proposed for S1CR25-12T and S3CR25-11T, respectively. Metagenomic fragment recruitment analysis, conducted using seven shotgun metagenomic datasets, revealed that the species belonging to the genus Halomicroarcula were predominantly recruited from hypersaline soils found in the Odiel Saltmarshes and the ponds of salterns with high salt concentrations. This reinforces the understanding of the extreme halophilic characteristics associated with the genus Halomicroarcula. Finally, comparing pan-genomes across the twenty Halomicroarcula and Haloarcula species allowed for the identification of commonalities and differences between the species of these two related genera.

Abstract:Recent advances in molecular and metagenomic analyses have enhanced the ability to precisely determine the microbiota of hypersaline environments of marine origin, such as solar salterns, saline lakes, and hypersaline soils, uncovering numerous yet-to-be-isolated prokaryotic groups. Our research focused on the hypersaline ecosystems within the Odiel Saltmarshes, a natural tidal wetland situated at the confluence of the Odiel and Tinto rivers in Huelva province, Southwestern Spain. Employing culture-dependent techniques, we aimed to isolate and characterize novel halophilic prokaryotes from this area. Two haloarchaeal strains, designated S1BR25-6 T and S3BR25-2 T were classified within the genus Halogeometricum based on Overall Genome Related Indexes (OGRIs) such as Orthologous Average Nucleotide Identity, digital DNA-DNA hybridization, and Average Amino Acid Identity as standard criteria for species delineation. Moreover, this study embarks on an exhaustive genome-based comparative analysis of the haloarchaeal genus Halogeometricum , delineating the metabolic capacities, osmoregulatory adaptations, and resistance to certain heavy metals of its species. The dual osmoregulatory mechanism observed by in-silico analysis of the Halogeometricum species combines "salt-in" and "salt-out" strategies which highlights the adaptive flexibility of these haloarchaea. In addition, capability for de novo thiamine biosynthesis of strain S1BR25-6 T along with other Halogeometricum species underscores their metabolic complexity and resilience, offering insights into their role in ecosystem dynamics and potential biotechnological applications. Wet lab experimental analysis of strains S1BR25-6 T and S3BR25-2 T confirmed their resistance to heavy metals, particularly to arsenic, zinc, and cadmium, emphasizing their potential for bioremediation applications. Furthermore, conducting fragment recruitment analysis across different metagenomic datasets revealed a predominant recruitment of species from the genus Halogeometricum in hypersaline soils of Odiel Saltmarshes (especially the two novel strains), and in the brines of marine saltern ponds with high salt concentrations. These results contribute to a reinforced understanding of the extremely halophilic characteristics inherent to the genus Halogeometricum . Finally, taxogenomic analysis has substantiated that strains S1BR25-6 T (= CCM 9250 T = CECT 30624 T ), and S3BR25-2 T (= CCM 9253 T = CECT 30622 T ) denote two previously unidentified species within the genus Halogeometricum , for which we propose the names Halogeometricum salsisoli sp. nov., and Halogeometricum luteum sp. nov., respectively.

Abstract:Earlier we showed that in serum-starved (27 h), washed mouse fibroblasts and other cell lines 40-80 mM concentrations of ethanol (EtOH) potentiate, in a zinc (Zn2+)-dependent manner, the combined stimulatory effects of calcium (Ca2+) and insulin (Ins) on DNA synthesis. We now report that the promitogenic EtOH effects require removal of the used medium at least 6 h prior to treatments with EtOH, Zn2+, and Ins. If serum-starved (27 h) cells were continuously incubated for another 18-h period without replacing the medium, a secreted cellular factor moderately enhanced the mitogenic effect of Ins and simultaneously blocked the potentiating effect of EtOH on DNA synthesis measured during the last hour of treatments. However, the presence of Ca2+ (2.8 mM) plus Zn2+ (25 microM) or 25-300 nM phorbol 12-myristate 13-acetate (PMA) during the serum starvation period partially restored the promitogenic effect of EtOH. The PMA effect was blocked by the protein kinase C (PKC) inhibitor GF 109203X added for the second (18 h) period. Even at 300 nM, PMA failed to fully downregulate PKC-alpha, the major PKC isoform, over a 28-h period, suggesting that an activated PKC enzyme was involved in the restoration of EtOH effect. When EtOH (40-80 mM) was added for the entire serum starvation period and the incubations were continued for 18 h without removing the medium, EtOH inhibited both the combined actions of Ins and cellular factor as well as the promoting effect of newly added EtOH on Ins-dependent DNA synthesis. Coaddition of Zn2+ and PMA with EtOH prevented these inhibitory effects of EtOH. The results indicate that in mouse fibroblasts EtOH can both enhance and inhibit Ins-dependent DNA synthesis depending on the timing of EtOH treatment as well as the presence of Zn2+, cellular factors, and activators of the PKC system.

Abstract:Three strains of Gram-negative, aerobic, neutrophilic, extremely halophilic archaea, designated ZJ206(T), ZJ203 and ZJ204, were isolated from a solar saltern in Zhe-Jiang Province, China. Phenotypically and on the basis of 16S rRNA gene sequences, the strains were very similar. Comparative 16S rRNA gene analysis revealed 96.4-97.4 % sequence similarity to members of the genus Haloferax. The major polar lipids were C(20)C(20) derivatives of phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester, diglycosyl glycerol diether and sulfated diglycosyl diether. The DNA G+C content of strain ZJ206(T) was 62.2 mol%. The results of DNA-DNA hybridizations and physiological and biochemical tests allowed genotypic and phenotypic differentiation of the isolates from closely related species. Therefore the isolates should be classified as members of a novel species, for which the name Haloferax larsenii sp. nov. is proposed. The type strain is ZJ206(T) (=CGMCC 1.5347(T)=JCM 13917(T)).

Abstract:A Gram-negative, aerobic, neutrophilic and extremely halophilic archaeon (strain AJ201(T)), isolated from Ayakekum salt lake on the Qinghai-Tibet Plateau, was investigated by a polyphasic approach. The DNA G+C content of strain AJ201(T) was 65.7 mol%. The major polar lipid profile and phylogenetic analysis based on 16S rRNA gene sequences supported the allocation of the strain to the genus Halorubrum. The results of DNA-DNA hybridizations and physiological and biochemical tests allowed genotypic and phenotypic differentiation of strain AJ201(T) from closely related species. Therefore, strain AJ201(T) represents a novel species of the genus Halorubrum, for which the name Halorubrum arcis sp. nov. is proposed. The type strain is strain AJ201(T) (=CGMCC 1.5343(T)=JCM 13916(T)).

Abstract:Four halophilic archaeal strains, BCD28 T , BND7 T , PSR21 T , and PSRA2 T , were isolated from coastal and inland saline soil, respectively. The 16S rRNA and rpoB′ gene sequence similarities among these four strains and current species of Halomarina were 95.9–96.6% and 86.9–90.3%, respectively. Phylogenetic and phylogenomic analyses revealed that these four strains tightly cluster with the current species of the genus Halomarina . The AAI, ANI, and dDDH values among these four strains and current species of Halomarina were 65.3–68.4%, 75.8–77.7%, and 20.3–22.0%, respectively, clearly below the threshold values for species demarcation Strains BCD28 T , BND7 T , PSR21 T , and PSRA2 T could be differentiated from the current species of Halomarina based on the comparison of diverse phenotypic characteristics The major polar lipids of these four strains were phosphatidylglycerol (PG), phosphatidylglycerol phosphate methyl ester (PGP-Me), and four to five glycolipids. Phosphatidylglycerol sulfate (PGS) was only detected in strain BND7 T . The phenotypic, phylogenetic, and genome-based analyses suggested that strains BCD28 T (= CGMCC 1.18776 T = JCM 34908 T ), BND7 T (= CGMCC 1.18778 T = JCM 34910 T ), PSR21 T (= CGMCC 1.17027 T = JCM 34147 T ), and PSRA2 T (= CGMCC 1.17214 T = JCM 34148 T ) represent four novel species of the genus Halomarina , for which the names Halomarina litorea sp. nov., Halomarina pelagica sp. nov., Halomarina halobia sp. nov., and Halomarina ordinaria sp. nov. are proposed.

Abstract:A halophilic archaeon, strain RO1-6(T), was isolated from a marine solar saltern in eastern China. Cells of strain RO1-6(T) were pleomorphic and motile and stained Gram-negative. Strain RO1-6(T) grew well on complex medium and colonies were red-pigmented. It was able to grow at 20-50°C (optimum 37°C), in 2.1-5.1M NaCl (optimum 3.9 M NaCl), in 0.05-0.70 M MgCl₂ (optimum 0.30 M MgCl₂) and at pH 6.5-8.0 (optimum pH 7.0). Cells lysed in distilled water and the minimal NaCl concentration to prevent cell lysis was 12 % (w/v). The major polar lipids of strain RO1-6(T) were phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester and two glycolipids that were chromatographically identical to S-DGD-1 and S₂-DGD. The 16S rRNA gene sequence of strain RO1-6(T) showed similarities of 96.9 and 96.4 % to those of the type strains of Halosarcina pallida and Halogeometricum borinquense, respectively, members of the most closely related recognized genera within the family Halobacteriaceae. The DNA G+C content of strain RO1-6(T) was 61.2 mol%. Phenotypic characterization and phylogenetic analysis revealed that strain RO1-6(T) is related to Halosarcina pallida and represents a novel species of the genus Halosarcina, for which the name Halosarcina limi sp. nov. is proposed; the type strain is RO1-6(T) (=CGMCC 1.8711(T) =JCM 16054(T)).

Abstract:An extremely halophilic archaeon strain named FL173 T was isolated from a salt mine (Anhui Province, China). Colonies on agar plate are orange-red, moist, and opaque. Cells are motile, Gram-stain-negative, polymorphic, and lyse in distilled water. Cells are able to grow at temperatures, NaCl concentrations, and pH ranging from 20 to 50 °C (optimum 42 °C), 2.6 to 5.1 M NaCl concentration (optimum 3.4 M), and 5.5 to 9.5 pH (optimum 7.0), respectively. Mg 2+ is not necessary for growth. The major polar lipids of strain FL173 T were phosphatidylglycerol (PG), phosphatidylglycerol phosphate methyl ester (PGP-Me), phosphatidylglycerol sulfonate (PGS), sulfonated mannosyl glycolipid (S-DGD-1). It has two copies of the 16S rRNA gene, which share the highest sequence similarity (93.04–99.02% sequence similarity) to the 16S rRNA genes of Halomicroarcula salinisoli F24A T , respectively. The rpoB' gene of strain FL173 T showed the highest sequence similarity (93.76%) to that of H. salinisoli F24A T . The genome-based analysis showed that the average amino-acid identity (AAI), orthologous average nucleotide identity (ANI) and in silico DNA–DNA hybridization values between strains FL173 T and H. salinisoli F24A T were 84.80%, 85.29%, and 29.70%, respectively, which are far below the threshold for the delineation of a prokaryotic new species. The DNA G+C content of strain FL173 T is 64.9%. Genomic, physiological, biochemical, and phenotypic evidences showed that strain FL173 T (CGMCC 1.18851=NBRC 114260) represents a new species of the genus Halomicroarcula , for which the name Halomicroarcula salaria sp. nov. is proposed.

Thiohalospira halophila gen. nov., sp. nov. and Thiohalospira alkaliphila sp. nov., novel obligately chemolithoautotrophic, halophilic, sulfur-oxidizing gammaproteobacteria from hypersaline habitats

Abstract:A previously unknown ecotype of obligately chemolithoautotrophic, sulfur-oxidizing bacteria was discovered in sediments of various inland hypersaline lakes and a solar saltern. The salt requirements for these bacteria were similar to those of haloarchaea, representing the first example of extreme halophiles occurring among the chemolithoautotrophs. They were enriched and isolated at 4 M NaCl under aerobic conditions with thiosulfate or tetrathionate as the electron donor or under micro-oxic conditions with sulfide. In total, 20 strains were obtained from hypersaline inland lakes in central Asia, central Russia and Crimea and a sea saltern of the Adriatic Sea. The isolates were thin, motile spirilla, some of which possessed a yellow, membrane-bound pigment. They were obligately aerobic, chemolithoautotrophic, sulfur-oxidizing bacteria that used thiosulfate, sulfide, sulfur and tetrathionate as electron donors. The characteristic feature of the group was the production of large amounts of tetrathionate as an intermediate during the oxidation of thiosulfate to sulfate. All but one of the strains grew within the pH range 6.5-8.2 (optimally at pH 7.3-7.8) and at NaCl concentrations from 2.0 to 5 M (optimally at 3.0 M). A single strain, designated ALgr 6sp(T), obtained (by enrichment) from the hypersaline alkaline lakes of the Wadi Natrun valley, was found to be moderately halophilic and facultatively alkaliphilic (capable of growth at pH 10). The predominant cellular fatty acids were quite unusual, with 10-methyl C(16 : 0) and C(16 : 0) predominating. Cells grown at 4 M NaCl accumulated extremely high concentrations of glycine betaine as a compatible solute. The 20 neutrophilic isolates contained three genospecies (on the basis of DNA-DNA relatedness data) but could not be discriminated phenotypically. On the basis of the phenotypic and genotypic analyses, the isolates constitute a novel genus and species, for which the name Thiohalospira halophila gen. nov., sp. nov. is proposed. The type strain of Thiohalospira halophila is HL 3(T) (=DSM 15071(T)=UNIQEM U219(T)). The haloalkaliphilic strain ALgr 6sp(T) represents a second species of the new genus, for which the name Thiohalospira alkaliphila sp. nov. is proposed. The type strain of Thiohalospira alkaliphila is ALgr 6sp(T) (=DSM 17116(T)=UNIQEM U372(T)).

Abstract:Moderately halophilic,strictly anaerobic eubacteria S191 was isolated and purified from the sediment in Northeast Pacific deep-sea nodule province by using Hungate roll-tube technique,the 16S rDNA encoding gene of S191 was proliferated by PCR,the sequence was determined,and homology analysis was completed by "Clustalx" and "Mega" software,a phylogenetic tree of Halanaerobium was constructed,S191 is considered to be a new member of Halanaerobium.It is a Gram-negative,rod-shaped,non-motile,non-spore-forming obligate anaerobe that owns a cell size of 0.3-0.6 μm×1.8-3.0 μm.Growth was observed in the presence of 5%-25% NaCl,the optimum salt concentration for growth is 7.5%-10%.It grows at pH 6.0-8.9 and the optimum pH for growth is 7.8-8.0.It grows well at temperature 15-42 ℃,and the optimum temperature for growth is 32-37 ℃.The doubling time is about 3.2 h when grown at optimal condition.

Abstract:Three strains of Gram-negative, aerobic, neutrophilic and halophilic bacteria were isolated from samples of a salt lake on the Qinghai-Tibet Plateau and a subterranean saline well in the Si-Chuan Basin of China. These isolates, designated AJ275(T), AJ282(T) and ZG16(T), were investigated using a polyphasic approach. Based on 16S rRNA gene sequence analysis, the isolates could be affiliated to the genus Halomonas. Genomic DNA G+C contents were 65.9 mol% for AJ275(T), 56.7 mol% for AJ282(T) and 57.6 mol% for ZG16(T). The results of DNA-DNA hybridizations, fatty acid analysis and physiological and biochemical tests allowed the isolates to be differentiated genotypically and phenotypically from closely related species. It is proposed that strains AJ275(T) (=CGMCC 1.6493(T)=JCM 14606(T)=LMG 23976(T)), AJ282(T) (=CGMCC 1.6494(T)=JCM 14607(T)=LMG 23978(T)) and ZG16(T) (=CGMCC 1.6495(T)=JCM 14608(T)=LMG 23977(T)) represent the type strains of three novel species in the genus Halomonas: Halomonas saccharevitans sp. nov., Halomonas arcis sp. nov. and Halomonas subterranea sp. nov., respectively.

Abstract:A marine bacterium,designated strain S3-22,was isolated from the ballast water sample from Zhoushan archipelago,China.16S rDNA sequences similarity of strain S3-22 to all recognized bacterial species was not greater than 97%.The DNA G+C content was 54.9 mol%.The dominant fatty acid was iso-C17:1ω 9c(24.99%).The major respiratory quinine was MK-5.Cells of strain S3-22 were gram-negative.Optimal growth of strain S3-22 was at pH 7.0,30–37 ℃ and 3%(w/v) NaCl.It was halophilic and tested positive for catalase,oxidase,amylase,and esterase.In addition,it could deoxidize the nitrate.Based on the evidence of 16S rDNA sequences similarities,phylogenetic analysis and cellular and molecular level identification,the halophile formed a phyletic lineage which was distinct from the known Kordiimonas species.Strain S3-22 represented a novel species in the Kordiimonas,its Genebank Accession number was FJ847942.

Abstract:The taxonomic status of some species of Halobellus , Haloferax , Halogranum , and Haloplanus within the family Haloferacaceae was elucidated by phylogenetic, phylogenomic, and comparative genomic analyses. The relative species of each genus should constitute a single species based on the overall genome-related indexes proposed for species demarcation. The cutoff values of AAI (72.1%), ANI (82.2%), and rpoB′ gene similarity (90.7%) were proposed to differentiate genera within the family Haloferacaceae . According to these standards, a novel genus related to the genus Halobaculum was proposed to accommodate Halobaculum halophilum Gai3-2 T and Halobaculum salinum NJ-3-1 T . Five halophilic archaeal strains, DT31 T , DT55 T , DT92 T , SYNS20 T , and YSMS11 T , isolated from a tidal flat and a marine solar saltern in China, were subjected to polyphasic classification. The phenotypic, phylogenetic, phylogenomic, and comparative genomic analyses revealed that strains DT31 T (= CGMCC 1.18923 T = JCM 35417 T ), DT55 T (= CGMCC 1.19048 T = JCM 36147 T ), DT92 T (= CGMCC 1.19057 T = JCM 36148 T ), SYNS20 T (= CGMCC 1.62628 T = JCM 36154 T ), and YSMS11 T (= CGMCC 1.18927 T = JCM 34912 T ) represent five novel species of the genus Halobaculum , for which the names, Halobaculum lipolyticum sp. nov., Halobaculum marinum sp. nov., Halobaculum litoreum sp. nov., Halobaculum halobium sp. nov., and Halobaculum limi sp. nov., are proposed.

Abstract:Extremely halophilic archaea (Haloarchaea, Nanohaloarchaeota, Methanonatronarchaeia and Halarchaeoplasmatales) thrive in saturating salt concentrations where they must maintain osmotic equilibrium with their environment. The evolutionary history of adaptations enabling salt tolerance remains poorly understood, in particular because the phylogeny of several lineages is conflicting. Here we present a resolved phylogeny of extremely halophilic archaea obtained using improved taxon sampling and state-of-the-art phylogenetic approaches designed to cope with the strong compositional biases of their proteomes. We describe two uncultured lineages, Afararchaeaceae and Asbonarchaeaceae, which break the long branches at the base of Haloarchaea and Nanohaloarchaeota, respectively. We obtained 13 metagenome-assembled genomes (MAGs) of these archaea from metagenomes of hypersaline aquatic systems of the Danakil Depression (Ethiopia). Our phylogenomic analyses including these taxa show that at least four independent adaptations to extreme halophily occurred during archaeal evolution. Gene-tree/species-tree reconciliation suggests that gene duplication and horizontal gene transfer played an important role in this process, for example, by spreading key genes (such as those encoding potassium transporters) across extremely halophilic lineages.

Abstract:A Gram-negative, aerobic, motile, halophilic bacterium, designated strain BC7(T), was isolated from a salt mine in north-western China and was subjected to a polyphasic taxonomic characterization. The isolate was able to grow in the presence of 0.5-20 % (w/v) NaCl and at pH 6.0-9.0 and 4-48 degrees C; optimum growth was observed with 3 % (w/v) NaCl and at pH 7.0 and 30 degrees C. Cells were long rods, 0.8-1.2 microm wide and 4.0-6.0 microm long. The major fatty acids were C(18 : 1)omega7c, C(16 : 0) and C(16 : 0)omega7c. The DNA G+C content was 65.5 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain BC7(T) belonged to the genus Halomonas and showed highest sequence similarity to the type strain of Halomonas pacifica (99.2 %). Levels of DNA-DNA relatedness between strain BC7(T) and H. pacifica CGMCC 1.2314(T) and Halomonas taeanensis DSM 16463(T) were 51 and 38 %, respectively. On the basis of phenotypic and genotypic data, strain BC7(T) is considered to represent a novel species of the genus Halomonas, for which the name Halomonas salifodinae sp. nov. is proposed. The type strain is BC7(T) (=CGMCC 1.6774(T) =JCM 14803(T)).
