Abstract:Three strains of Gram-negative, aerobic, neutrophilic, extremely halophilic archaea, designated ZJ206(T), ZJ203 and ZJ204, were isolated from a solar saltern in Zhe-Jiang Province, China. Phenotypically and on the basis of 16S rRNA gene sequences, the strains were very similar. Comparative 16S rRNA gene analysis revealed 96.4-97.4 % sequence similarity to members of the genus Haloferax. The major polar lipids were C(20)C(20) derivatives of phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester, diglycosyl glycerol diether and sulfated diglycosyl diether. The DNA G+C content of strain ZJ206(T) was 62.2 mol%. The results of DNA-DNA hybridizations and physiological and biochemical tests allowed genotypic and phenotypic differentiation of the isolates from closely related species. Therefore the isolates should be classified as members of a novel species, for which the name Haloferax larsenii sp. nov. is proposed. The type strain is ZJ206(T) (=CGMCC 1.5347(T)=JCM 13917(T)).

Abstract:Two extremely halophilic archaea, strains RO5-2(T) and RO5-14, were isolated from Rudong marine solar saltern in Jiangsu, China. Cells of the two strains were pleomorphic, motile and stained Gram-negative. Colonies were red-pigmented. Strains RO5-2(T) and RO5-14 were able to grow at 20-50 degrees C (optimum 37 degrees C), at 2.6-4.8 M NaCl (optimum 3.4-3.9 M NaCl), at 0.03-0.7 M MgCl2 (optimum 0.5 M MgCl2) and at pH 5.5-8.0 (optimum pH 6.5-7.0). Cells lyse in distilled water and the minimal NaCl concentration to prevent cell lysis was 12 % (w/v). The major polar lipids of the two strains were phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester and two major glycolipids chromatographically identical to sulfated mannosyl glucosyl diether (S-DGD-1) and mannosyl glucosyl diether (DGD-1). The 16S rRNA gene sequences of strains RO5-2T and RO5-14 showed 93.4-93.8% similarity to the closest cultivated relative, Halosarcina pallida. The DNA G + C content of strains RO5-2(T) and RO5-14 was 61.0 mol% and 59.9 mol%, respectively. The DNA DNA relatedness between strains RO5-2(T) and RO5-14 was 86.0%. The phenotypic, chemotaxonomic and phylogenetic properties suggest that strains RO5-2(T) and RO5-14 represent a novel species in a new genus within the family Halobacteriaceae, for which the name Halopelagius inordinatus gen. nov., sp. nov. is proposed. The type strain is RO5-2(T) (=CGMCC 1.7739(T) =JCM 15773(T)).

Abstract:Two extremely halophilic archaea, strains RO2-11(T) and HO2-1, were isolated from two Chinese marine solar salterns, Rudong solar saltern and Haimen solar saltern, respectively. Cells of the two strains were polymorphic and Gram-stain-negative; colonies were red-pigmented. The two strains grew at NaCl concentrations of 2.6-4.3 M (optimum 3.9 M) and required at least 0.1 M Mg2+ for growth. They were able to grow over a pH range of 6.0-8.0 and a temperature range of 20-50 degrees C, with optimal pH of 7.5 and optimal temperature of 37 degrees C. The major polar lipids of strain RO2-11(T) and strain HO2-1 were phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester and three glycolipids, two of them chromatographically identical to S-DGD-1 and DGD-1, the third unidentified. The 16S rRNA gene sequence similarity of strain RO2-11(T) and strain HO2-1 was 99.3 % and highest sequence similarity with the closest relative (Haloferax larsenii) was 91.4 %. Based on the data obtained, the two isolates could not be classified in any recognized genus of the family Halobacteriaceae. Strain RO2-11(T) and strain HO2-1 are thus considered to represent a novel species of a new genus within the family Halobacteriaceae, for which the name Halogranum rubrum gen. nov., sp. nov. is proposed. The type strain is RO2-11(T) (=CGMCC 1.7738(T) =JCM 15772(T)).

Abstract:A Gram-negative, aerobic, neutrophilic and extremely halophilic archaeon (strain AJ201(T)), isolated from Ayakekum salt lake on the Qinghai-Tibet Plateau, was investigated by a polyphasic approach. The DNA G+C content of strain AJ201(T) was 65.7 mol%. The major polar lipid profile and phylogenetic analysis based on 16S rRNA gene sequences supported the allocation of the strain to the genus Halorubrum. The results of DNA-DNA hybridizations and physiological and biochemical tests allowed genotypic and phenotypic differentiation of strain AJ201(T) from closely related species. Therefore, strain AJ201(T) represents a novel species of the genus Halorubrum, for which the name Halorubrum arcis sp. nov. is proposed. The type strain is strain AJ201(T) (=CGMCC 1.5343(T)=JCM 13916(T)).

Abstract:Three strains of Gram-negative, aerobic, neutrophilic and halophilic bacteria were isolated from samples of a salt lake on the Qinghai-Tibet Plateau and a subterranean saline well in the Si-Chuan Basin of China. These isolates, designated AJ275(T), AJ282(T) and ZG16(T), were investigated using a polyphasic approach. Based on 16S rRNA gene sequence analysis, the isolates could be affiliated to the genus Halomonas. Genomic DNA G+C contents were 65.9 mol% for AJ275(T), 56.7 mol% for AJ282(T) and 57.6 mol% for ZG16(T). The results of DNA-DNA hybridizations, fatty acid analysis and physiological and biochemical tests allowed the isolates to be differentiated genotypically and phenotypically from closely related species. It is proposed that strains AJ275(T) (=CGMCC 1.6493(T)=JCM 14606(T)=LMG 23976(T)), AJ282(T) (=CGMCC 1.6494(T)=JCM 14607(T)=LMG 23978(T)) and ZG16(T) (=CGMCC 1.6495(T)=JCM 14608(T)=LMG 23977(T)) represent the type strains of three novel species in the genus Halomonas: Halomonas saccharevitans sp. nov., Halomonas arcis sp. nov. and Halomonas subterranea sp. nov., respectively.

Abstract:Two novel halophilic archaeal strains (XZGYJ-43 T and ZJ1 T ) were isolated from Mangkang ancient solar saltern (Tibet, PR China) and Zhujiang river inlet (Guangdong, PR China), respectively. The comparison of the 16S rRNA gene sequences revealed that strain XZGYJ-43 T is related to the current species of the family Halobacteriaceae (89.2–91.7% similarity) and strain ZJ1 T showed 94.7–98.3% similarity to the current species of the genus Haladaptatus . Phylogenetic analyses based on 16S rRNA genes, rpoB ′ genes and genomes indicated that strain XZGYJ-43 T is separate from the related genera, Halocalculus , Salarchaeum and Halarchaeum of the family Halobacteriaceae , and strain ZJ1 T tightly clusters with the current species of the genus Haladaptatus . The average nucleotide identity, digital DNA–DNA hybridization and average amino acid identity values between strain XZGYJ-43 T and the current species of the family Halobacteriaceae were 71–75, 20–25 and 59–68 %, and these values between strain ZJ1 T and the current species of the genus Haladaptatus were 77–81, 27–32 and 76–82 %, respectively, clearly below the thresholds for prokaryotic species demarcation. These two strains could be distinguished from their relatives according to differential phenotypic characteristics. The major polar lipids of strain XZGYJ-43 T were phosphatidic acid (PA), phosphatidylglycerol (PG), phosphatidylglycerol phosphate methyl ester (PGP-Me), mannosyl glucosyl diether (DGD-1; DGD-PA) and sulphated mannosyl glucosyl diether (S-DGD-1; S-DGD-PA), and those of strain ZJ1 T were PA, PG, PGP-Me, DGD-PA, S-DGD-1 (S-DGD-PA) and sulphated galactosyl mannosyl glucosyl diether. Based on phenotypic, phylogenetic and genomic data, strain XZGYJ-43 T (=CGMCC 1.13890 T =JCM 33735 T ) represents a novel species of a new genus within the family Halobacteriaceae , and strain ZJ1 T (=CGMCC 1.18785 T =JCM 34917 T ) represents a novel species of the genus Haladaptatus , for which the names Halospeciosus flavus gen. nov., sp. nov. and Haladaptatus caseinilyticus sp. nov. are proposed, respectively.

Abstract:Two extremely halophilic archaeal strains, GSLN9 T and XZYJT29 T , were isolated from the saline soil in different regions of western China. Both strains GSLN9 T and XZYJT29 T have two 16S rRNA genes with similarities of 95.1 and 94.8 %, respectively. Strain GSLN9 T was mostly related to the genus Halomicrococcus based on 16S rRNA (showing 91.0–96.0 % identities) and rpoB′ genes (showing 92.0 % identity). Strain XZYJT29 T showed 92.1–97.6 % (16S rRNA gene) and 91.4–93.1 % ( rpoB′ gene) sequence similarities to its relatives in the genus Halosimplex , respectively. The polar lipid profile of strain GSLN9 T included phosphatidic acid (PA), phosphatidylglycerol (PG), phosphatidylglycerol phosphate methyl ester (PGP-Me), phosphatidylglycerol sulphate (PGS), sulphated mannosyl glucosyl diether (S-DGD-1) and sulphated galactosyl mannosyl glucosyl diether (S-TGD-1), mostly similar to that of Halomicrococcus hydrotolerans H22 T . PA, PG, PGP-Me, S-DGD-1 (S-DGD-PA), S 2 -DGD, S-TGD-1 and an unidentified glycolipid were detected in strain XZYJT29 T ; this polar lipid composition is similar to those of members of the genus Halosimplex . The average nucleotide identity, digital DNA–DNA hybridization and average amino acid identity values between these two strains and their relatives of the genera Halomicrococcus and Halosimplex were no more than 82, 27 and 80 %, respectively, much lower than the thresholds for species demarcation. Other phenotypic characterization results indicated that strains GSLN9 T and XZYJT29 T can be differentiated from the current species of the genera Halomicrococcus and Halosimplex , respectively. These results revealed that strains GSLN9 T (=CGMCC 1.15215 T =JCM 30842 T ) and XZYJT29 T (=CGMCC 1.15828 T =JCM 31853 T ) represent novel species of Halomicrococcus and Halosimplex , for which the names Halomicrococcus gelatinilyticus sp. nov. and Halosimplex aquaticum sp. nov. are proposed.

Abstract:A Gram-negative, aerobic, motile, halophilic bacterium, designated strain BC7(T), was isolated from a salt mine in north-western China and was subjected to a polyphasic taxonomic characterization. The isolate was able to grow in the presence of 0.5-20 % (w/v) NaCl and at pH 6.0-9.0 and 4-48 degrees C; optimum growth was observed with 3 % (w/v) NaCl and at pH 7.0 and 30 degrees C. Cells were long rods, 0.8-1.2 microm wide and 4.0-6.0 microm long. The major fatty acids were C(18 : 1)omega7c, C(16 : 0) and C(16 : 0)omega7c. The DNA G+C content was 65.5 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain BC7(T) belonged to the genus Halomonas and showed highest sequence similarity to the type strain of Halomonas pacifica (99.2 %). Levels of DNA-DNA relatedness between strain BC7(T) and H. pacifica CGMCC 1.2314(T) and Halomonas taeanensis DSM 16463(T) were 51 and 38 %, respectively. On the basis of phenotypic and genotypic data, strain BC7(T) is considered to represent a novel species of the genus Halomonas, for which the name Halomonas salifodinae sp. nov. is proposed. The type strain is BC7(T) (=CGMCC 1.6774(T) =JCM 14803(T)).

Abstract:An extremely halophilic archaeon, strain RO5-8(T), was isolated from a disused marine solar saltern in China. The cells were pleomorphic and flat. In static liquid medium, cells floated to the surface. Strain RO5-8(T) stained Gram-negative and colonies were pink-pigmented. It was able to grow at 30-50 degrees C (optimum 40 degrees C), at 2.6-4.3 M NaCl (optimum 3.1 M NaCl), at 0.03-0.5 M MgCl(2) (optimum 0.03 M MgCl(2)) and at pH 5.5-7.5 (optimum pH 6.0-6.5). Cells lysed in distilled water and the minimal NaCl concentration to prevent cell lysis was 12 % (w/v). The major polar lipids of strain RO5-8(T) were phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester, phosphatidylglycerol sulfate and one major glycolipid chromatographically identical to the sulfated mannosyl glucosyl diether S-DGD-1. On the basis of 16S rRNA gene sequence analysis, strain RO5-8(T) was closely related to three strains of Haloplanus natans with similarities of 97.3-97.6%. The DNA G+C content of strain RO5-8(T) was 62.1 mol%. The DNA DNA hybridization value between strain RO5-8(T) and Haloplanus natans JCM 14081(T) was 51.6%. It was concluded that strain RO5-8(T) represents a novel species of the genus Haloplanus, for which the name Haloplanus vescus sp. nov. is proposed. The type strain is RO5-8(T) (=CGMCC 1.8712(T) =JCM 16055(T)).
Genomic, Physiological, Biochemical, and Phenotypic Evidences Reveal a New Species, Halomicroarcula salaria sp. nov

Abstract:An extremely halophilic archaeon strain named FL173 T was isolated from a salt mine (Anhui Province, China). Colonies on agar plate are orange-red, moist, and opaque. Cells are motile, Gram-stain-negative, polymorphic, and lyse in distilled water. Cells are able to grow at temperatures, NaCl concentrations, and pH ranging from 20 to 50 °C (optimum 42 °C), 2.6 to 5.1 M NaCl concentration (optimum 3.4 M), and 5.5 to 9.5 pH (optimum 7.0), respectively. Mg 2+ is not necessary for growth. The major polar lipids of strain FL173 T were phosphatidylglycerol (PG), phosphatidylglycerol phosphate methyl ester (PGP-Me), phosphatidylglycerol sulfonate (PGS), sulfonated mannosyl glycolipid (S-DGD-1). It has two copies of the 16S rRNA gene, which share the highest sequence similarity (93.04–99.02% sequence similarity) to the 16S rRNA genes of Halomicroarcula salinisoli F24A T , respectively. The rpoB' gene of strain FL173 T showed the highest sequence similarity (93.76%) to that of H. salinisoli F24A T . The genome-based analysis showed that the average amino-acid identity (AAI), orthologous average nucleotide identity (ANI) and in silico DNA–DNA hybridization values between strains FL173 T and H. salinisoli F24A T were 84.80%, 85.29%, and 29.70%, respectively, which are far below the threshold for the delineation of a prokaryotic new species. The DNA G+C content of strain FL173 T is 64.9%. Genomic, physiological, biochemical, and phenotypic evidences showed that strain FL173 T (CGMCC 1.18851=NBRC 114260) represents a new species of the genus Halomicroarcula , for which the name Halomicroarcula salaria sp. nov. is proposed.

Abstract:Two halophilic archaeal strains TS33T and KZCA124 were isolated from two distant salt lakes on the Qinghai-Xizang Plateau, respectively. Culture-independent analysis indicated that these two strains were original inhabitants but low abundant taxa in respective salt lakes. Strains TS33T and KZCA124 were able to grow at 20-60 °C (optimum were 42 and 35 °C, respectively), with 0.9-4.8 M NaCl (optimum were 3.0 and 2.6 M, respectively), with 0-0.7 M MgCl2 (optimum, 0.3 M) and at pH 5.0-9.5 (optimum were pH 7.5 and pH 7, respectively). The 16S rRNA and rpoB' gene similarities between these two strains were 99.7% and 99.4%, and these two similarities among strains TS33T, KZCA124, and existing species of the family Natrialbaceae were 90.6-95.5% and 84.4-89.3%, respectively. Phylogenetic and phylogenomic analyses indicated that strains TS33T and KZCA124 formed an independent branch separated from neighboring genera, Saliphagus, Natronosalvus, and Natronobiforma. The averagenucleotideidentity (ANI), digital DNA-DNAhybridization (dDDH), and average amino acid identity (AAI) values between strains TS33T and KZCA124 were 96.4%, 73.1%, and 96.7%, respectively, higher than the thresholds for species demarcation. The overall genome-related indexes between these two strains and existing species of family Natrialbaceae were 73-77%, 21-25%, and 63-70%, respectively, significantly lower than the species boundary thresholds. Strains TS33T and KZCA124 may represent a novel species of a new genus within the family Natrialbaceae judged by the cutoff value of AAI (≤76%) proposed to differentiate genera within the family Natrialbaceae. The major polar lipids of strains TS33T and KZCA124 were phosphatidic acid, phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester, sulfated mannosyl glucosyl diether, and sulfated galactosyl mannosyl glucosyl diether. These two strains could be distinguished from the related genera according to differential phenotypic characteristics. These phenotypic, phylogenetic, and genomic analyses revealed that strains TS33T (=KCTC 4310T = MCCC 4K00132T) and KZCA124 (=CGMCC 1.17432 = JCM 34316) represent a novel species of a new genus of the family Natrialbaceae and were named Halomontanus rarus gen. nov., sp. nov.

Strain RO1-6(T) grew well on complex medium and colonies were red-pigmented. It was able to grow at 20-50°C (optimum 37°C), in 2.1-5.1M NaCl (optimum 3.9 M NaCl), in 0.05-0.70 M MgCl₂ (optimum 0.30 M MgCl₂) and at pH 6.5-8.0 (optimum pH 7.0). Cells lysed in distilled water and the minimal NaCl concentration to prevent cell lysis was 12 % (w/v). The major polar lipids of strain RO1-6(T) were phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester and two glycolipids that were chromatographically identical to S-DGD-1 and S₂-DGD. The 16S rRNA gene sequence of strain RO1-6(T) showed similarities of 96.9 and 96.4 % to those of the type strains of Halosarcina pallida and Halogeometricum borinquense, respectively, members of the most closely related recognized genera within the family Halobacteriaceae. Abstract:Two halophilic archaea, strains RO1-4(T) and RO1-64, were isolated from a marine solar saltern in Jiangsu, China. Cells of the two strains were pleomorphic, motile, and stained Gram-negative. Colonies were red-pigmented. Strains RO1-4(T) and RO1-64 were able to grow at 25-55 °C (optimum 40-42 °C), at 2.1-5.1 M NaCl (optimum 3.9 M NaCl), at 0.05-0.7 M MgCl(2) (optimum 0.3 M MgCl(2)) and at pH 6.0-8.5 (optimum pH 7.0). Cells lyse in distilled water and the minimal NaCl concentration to prevent cell lysis is 12 % (w/v). On the basis of 16S rRNA gene sequence analysis, strains RO1-4(T) and RO1-64 were closely related to Halogeometricum borinquense PR3(T) (98.0 and 98.2 % similarity, respectively) and Halosarcina pallida BZ256(T) (97.8 and 97.9 %). The major polar lipids of the two strains were phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester and two glycolipids (S-DGD-1 and DGD-1). The DNA G+C contents of strains RO1-4(T) and RO1-64 are 64.9 and 62.4 mol%, respectively. The DNA-DNA hybridization value between strains RO1-4(T) and RO1-64 was 83.0 % and both strains showed low DNA-DNA relatedness with Halogeometricum borinquense PR3(T) (42.5 and 50.1 % relatedness, respectively) and Halosarcina pallida BZ256(T) (37.6 and 42.1 % relatedness). It was concluded that strains RO1-4(T) and RO1-64 represent a novel species of the genus Halogeometricum, for which the name Halogeometricum rufum sp. nov. is proposed. The type strain is RO1-4(T) (=CGMCC 1.7736(T) =JCM 15770(T)).

Abstract:Two novel extremely halophilic archaeal strains, designated H27T and FL145T, were isolated from a salt mine and a kelp salt sample, respectively. Cells of both strains were Gram-stain-negative, motile and pleomorphic. The 16S rRNA and rpoB' gene sequence similarities between strains H27T and FL145T were 96.60 and 88.77%. Strains H27T and FL145T were both closely related to Halorhabdus rudnickae WSM-64T, Halorhabdus tiamatea SARL4BT and Halorhabdus utahensis AX-2T, with a 16S rRNA gene sequence similarities of 98.14, 96.34 and 96.27% for strain H27T and 96.42, 95.82 and 96.17% for strain FL145T. The genome-based average nucleotide identity (ANI) values between strains H27T and FL145T, and these three species were 83.93, 79.79 and 79.09% (for strain H27T), and 78.32, 77.95 and 77.05% (for strain FL145T), respectively. The ANI value between strains H27T and FL145T was 78.65 %. The digital DNA-DNA hybridization values between strains H27T and FL145T, and these three species were less than 27.40%, which were below the recommended threshold for membership of the same species. The major polar lipids of both strains were found to consist of sulfated diglycosyl diether, triglycosyl diether, phosphatidylglycerol phosphate methyl ester and phosphatidylglycerol. The DNA G+C content was determined from genome to be 62.10 mol% for strain H27T and 61.51 mol% for strain FL145T. Based on phylogenetic, phenotypic, chemotaxonomic and genomic analyses, these two new isolates should be classified as representing two novel species in the genus Halorhabdus, with strain H27T (=CGMCC 1.16342T=NBRC 113589T) as the type strain of a new species for which we propose the name Halorhabdus amylolytica sp. nov., and strain FL145T (=CGMCC 1.13888T=NBRC 114260T) as the type strain of another new species for which we propose the name Halorhabdus salina sp. nov.

Abstract:Five halophilic archaeal strains, XH8T, CK5-1T, GDY1T, HW8-1T, and XH21T, were isolated from commercial coarse salt produced in different regions of China. Their 16S rRNA and rpoB' gene sequences indicated that four of the strains (CK5-1T, GDY1T, HW8-1T, and XH21T) represent distinct species within the genus Haloplanus (family Haloferacaceae), while strain XH8T represents a novel genus within the same family. These assignments were supported by phylogenetic and phylogenomic analyses, which showed that strains CK5-1T, GDY1T, HW8-1T, and XH21T cluster with the current species of the genus Haloplanus, while strain XH8T forms a separate branch from the genus Haloplanus. The digital DNA-DNA hybridization and average amino acid identity (AAI) values among these four strains and the current members of the genus Haloplanus were 23.1%-35.2% and 75.9%-83.8%, respectively; and those values between strain XH8T and other genera in the family Haloferacaceae were 18.8%-33.6% and 59.8%-66.6%, respectively, much lower than the threshold values for species demarcation. Strain XH8T may represent a novel genus of the family Haloferacaceae according to the cut-off value of AAI (≤72.1%) proposed to differentiate genera within the family Haloferacaceae. These five strains could be distinguished from the related species according to differential phenotypic characteristics. Based on these results, it is proposed that strain XH8T represents a novel genus within the family Haloferacaceae, and strains CK5-1T, GDY1T, HW8-1T, and XH21T represent four novel species of the genus Haloplanus, respectively. Additionally, these five strains possess genes encoding enzymes critical for the fermentation process in salt-fermented foods, indicating their potential as starter cultures for these applications.

Abstract:A halophilic, Gram-negative bacterial strain, designated AJ261(T), which was isolated from a soil sample from a salt lake on the Qinghai-Tibet Plateau, was subjected to a polyphasic taxonomic study. The isolate grew optimally in the presence of 3-5% NaCl and used various carbohydrates as sole carbon and energy sources. The genomic DNA G + C content was 63.0 mol%. The predominant fatty acids were C(18:1)omega 7c, C(16:0) and C(12:0). A phylogenetic analysis based on 16S rRNA gene sequences indicated that the isolate had the highest sequence similarity with respect to type strains of Halomonas elongata (98.2%), Halomonas eurihalina (98.1%) and Halomonas halmophila (97.2%). The DNA-DNA relatedness of strain AJ261(T) with respect to H. elongata NBRC 15536(T), H. eurihalina CGMCC 1.2318(T) and H. halmophila DSM 5349(T) was 42, 25 and 26%, respectively. Overall, the phenotypic, genotypic and phylogenetic results demonstrate that strain AJ261(T) represents a novel species within the genus Halomonas, for which the name Halomonas caseinilytica is proposed, The type strain is AJ261(T) (=CGMCC 1.6773(T) =JCM 14802(T)).

Abstract:Since the first genome of a halophilic archaeon was sequenced in 2000, microbes inhabiting hypersaline environments have been investigated largely based on genomic characteristics. Salinigranum rubrwn GX10(T), the type species of the genus Salinigranum belonging to the euryarchaeal family Haloferacaceae, was isolated from the brine of Gangxi marine solar saltern near Weihai, China. Similar with most members of the class Halobacteria, S. rubrum GX10(T) is an extreme halophile requiring at least 1.5 M NaCl for growth and 3.1 M NaCl for optimum growth. We sequenced and annotated the complete genome of S. rubrum GX10(T), which was found to be 4,973,118 bp and comprise one chromosome and five plasmids. A total of 4966 protein coding genes, 47 tRNA genes and 6 rRNA genes were obtained. The isoelectric point distribution for the predict proteins was observed with an acidic peak, which reflected the adaption of S. rubrum GX10(T) to the halophilic environment. Genes related to potassium uptake, sodium efflux as well as compatible-solute biosynthesis and transport were identified, which were responsible for the resistance to osmotic stress. Genes related to heavy metal resistance, CRISPR-Cas system and light transform system were also detected. This study reports the first genome in the genus Salinigranum and provides a basis for understanding resistance strategies to harsh environment at the genomic level.

Abstract:Halophilic archaeal strain TBN37(T) was isolated from Taibei marine solar saltern near Lianyungang city of Jiangsu province, China. Cells were pleomorphic, flat and contained gas vesicles. Cells of strain TBN37(T) stained Gram-negative and the colonies were pink-pigmented. The strain was able to grow at 25-50 °C (optimum, 37-40 °C), with 1.4-5.1 M NaCl (optimum, 2.1 M NaCl), with 0-1.0 M MgCl(2) (optimum, 0.01 M MgCl(2)) and at pH 6.0-9.0 (optimum, pH 7.5). Cells lysed in distilled water and the minimal NaCl concentration to prevent cell lysis was 8 % (w/v). The major polar lipids of strain TBN37(T) were phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester, phosphatidylglycerol sulfate and one major glycolipid chromatographically identical to sulfated mannosyl glucosyl diether (S-DGD-1). On the basis of 16S rRNA gene sequence analysis, strain TBN37(T) was closely related to Haloplanus natans and Haloplanus vescus, with the same similarity of 97.4 %. The DNA G+C content of strain TBN37(T) is 64.1 mol%. DNA-DNA hybridization values between strain TBN37(T) and Haloplanus natans JCM 14081(T) and between strain TBN37(T) and Haloplanus vescus RO5-8(T) were 37.6 % and 42.1 %, respectively. It was concluded that strain TBN37(T) represents a novel species of the genus Haloplanus, for which the name Haloplanus aerogenes sp. nov. is proposed. The type strain is TBN37(T) ( = CGMCC 1.10124(T) = JCM 16430(T)).
Genomic Insights Into New Species of the Genus Halomicroarcula Reveals Potential for New Osmoadaptative Strategies in Halophilic Archaea

Abstract:Metagenomic studies on prokaryotic diversity of hypersaline soils from the Odiel saltmarshes, South-west Spain, revealed a high proportion of genomic sequences not related to previously cultivated taxa, that might be related to haloarchaea with a high environmental and nutritional flexibility. In this study, we used a culturomics approach in order to isolate new haloarchaeal microorganisms from these hypersaline soils. Four haloarchaeal strains, designated strains F24AT, F28, F27T, and F13T, phylogenetically related to the genus Halomicroarcula, were isolated and characterized in detail. The phylogenomic tree based on the 100 orthologous single-copy genes present in the genomes of these four strains as well as those of the type strains of the species Halomicroarcula pellucida CECT 7537T, Halomicroarcula salina JCM 18369T and Halomicroarcula limicola JCM 18640T, that were determined in this study, revealed that these four new isolates clustered on three groups, with strains F24AT and F28 within a single cluster, and altogether with the species of Halomicroarcula. Additionally, Orthologous Average Nucleotide Identity (OrthoANI), digital DNA-DNA hybridization (dDDH) and Average Amino-acid Identity (AAI) values, likewise phenotypic characteristics, including their polar lipids profiles, permitted to determine that they represent three new species, for which we propose the names Halomicroarcula rubra sp. nov. (type strain F13T), Halomicroarcula nitratireducens sp. nov. (type strain F27T) and Halomicroarcula salinisoli sp. nov. (type strain F24AT). An in deep comparative genomic analysis of species of the genus Halomicroarcula, including their metabolism, their capability to biosynthesize secondary metabolites and their osmoregulatory adaptation mechanisms was carried out. Although they use a salt-in strategy, the identification of the complete pathways for the biosynthesis of the compatible solutes trehalose and glycine betaine, not identified before in any other haloarchaea, might suggest alternative osmoadaptation strategies for this group. This alternative osmoregulatory mechanism would allow this group of haloarchaea to be versatile and eco-physiologically successful in hypersaline environments and would justify the capability of the species of this genus to grow not only on environments with high salt concentrations [up to 30% (w/v) salts], but also under intermediate to low salinities.
