Abstract:Relying on the electrochemiluminescence (ECL) and microfluidic technology, an immunosensor chip with high bioactivity was designed for sensitive determination of cytokeratin 19 fragment 21–1 (CYFRA 21–1). The mesoporous nanomaterial Fe3O4@Cu@Cu2O as the co-reaction accelerator was used to catalyze the S2O82− to produce more SO4•− to achieve the amplification of the ECL signal. In fact, the generating of SO4•− could not only be done with the aid of the reversible cycles of Fe2+ and Fe3+ and Cu+ and Cu2+, but could be achieved also through the catalase-like function of Fe3O4. What is more, it has also been proved that Fe3O4@Cu@Cu2O exhibited better catalytic performance than single Fe3O4, Cu2O, and Cu@Cu2O, which supported its application in this system. In addition, a portable microfluidic immunosensor chip for CYFRA 21–1-sensitive determination was assembled, which showed high selectivity, sensitivity, and strong universality in clinical cancer screening and diagnosis. It should be noted that HWRGWVC (HWR) was introduced as the antibody fixator to improve the incubation and binding efficiency of the antibody, which increased the ECL intensity and improved the sensitivity of the immunosensor. This strategy provided a new idea for cancer identification and diagnosis in clinical medicine.
Electrostatic Nanocage-Confined Probe for Electrochemical Detection of CA19-9 in Human Serum.

Abstract:Prompt and accurate detection of CA19-9 in human serum has great clinical significance for the early diagnosis and disease monitoring of cancer. Herein, we develop a convenient and antifouling electrochemical sensor for CA19-9 determination by immobilization of both an electrochemical redox probe [methylene blue (MB)] and immunorecognition element (CA19-9 antibody) on an electrostatic nanocage consisting of bipolar silica nanochannel array (bp-SNA). bp-SNA is composed of a negatively charged inner layer (n-SNA) and positively charged outer layer (p-SNA), which could be stably prepared on indium tin oxide (ITO) in several seconds using a two-step electrochemically assisted self-assembly approach and display asymmetric surface charges for confinement and enrichment of cationic MB into the inner n-SNA layer through electrostatic interaction. Modification of the CA19-9 antibody on the top surface of bp-SNA confers the sensing interface with specific recognition capacity. An antibody-antigen complex formed at the as-prepared immunosensor causes the decreased electrochemical signals of MB, achieving sensitive determination of CA19-9 with a wider linear dynamic range from 10 μU/mL to 50 U/mL and a low detection limit (3 μU/mL). Furthermore, accurate and feasible analysis of the CA19-9 amount in human serum samples by our proposed probe-integrated electrochemical immunosensor is realized.
An eco-friendly MIP-solid surface fluorescence immunosensor for detection of CA 19-9 tumor marker using Ni nanocluster as an emitter labels

Abstract:In the present study, an eco-friendly solid surface fluorescence immunosensor was fabricated for the ultra-selective and sensitive determination of CA 19-9 antigen based on polydopamine (PDA) molecular imprinted polymer (MIP) as an antibody substitution and Ni nanoclusters (Ni NCs) as the fluorescent signal probe. Here, the uniform MIP film for CA 19-9 antigen was established by electropolymerization of DA and target antigen on the surface of ITO electrode. When the imprinted cavities were occupied by CA 19-9 antigen and Ni NCs-Ab, respectively, the fluorescence emission of Ni NCs on the MIP-modified electrode surface was increased. Actually, MIP has been used in immunoassays as antibody substitute. Under optimized experimental conditions, taking advantage of both MIP-FL assays, the proposed immunosensor demonstrated a good analytical performance for the CA 19-9 antigen detection in an excellent linear range from 1 pg mL−1 to 48 ng mL−1 with detection limit of 0.13 pg mL−1 (S/N = 3). The application of the proposed MIP-FL immunosensor for detection of CA 19-9 antigen in human serum samples was evaluated, and the obtained results were found to be in acceptable agreement with those obtained with an ELISA assay as standard method. The developed fluorescence immunosensor demonstrated high specificity and can provide a simple, sensitive and reliable approach for the ultra-trace detection of other tumor markers in clinical monitoring.

Abstract:In this study, a novel split-type electrochemical immunosensor based on controlled release strategy was proposed for sensitive analysis and detection of tumor marker carbohydrate antigen 199 (CA19-9). Specifically, glucose (Glu) was encapsulated in carrier mesoporous silica (MSN) with encapsulation technology, and surface functionalized Zinc sulfide (ZnS) caps were used as "gatekeepers". The complex is formed by encapsulating Glu within MSN with ZnS (ZnS@MSN-Glu) as a signal amplifier labeled on the signal antibody (Ab2). And the Ab2 can detect the presence of antibodies. To reduce the interference of biological analysis, the immune recognition process of ZnS@MSN-Glu-Ab2 bioconjugate and antigen was carried out in 96-well microplate, which did not interfere with the electrochemical analysis process. Therefore, the low sensitivity detection caused by biofouling of nanomaterials and immunoreaction on the testing platform is eliminated. Subsequently, the opening and timed release of mesopores were controlled by external stimuli, the disulfide bond cleavage by dithiothreitol (DTT), and glucose was effectively released. Then nickel cobalt layered double hydroxide (NiCo-LDH) were directly hydrothermally grown on carbon cloth (CC) electrodeposited with copper selenide (CuSe) nanosheets to construct three-dimensional (3D) cactus-like NiCo-LDH/CuSe/CC sensing platform. It can realize the catalytic oxidation of released glucose, triggering glucose-mediated signal amplification. The synergistic effect of the 3D cactus structure and active nanomaterials promotes electron conduction. Taking the detection of carbohydrate antigen CA19-9 as an example, the immunosensor shows a wide linear concentration range (0.001-100 U/mL) with the limit of detection of 0.0005 U/mL, realizing highly sensitive detection of CA19-9. This biosensing technique has considerable advantages and provides an innovative approach for trace detection of other biomarkers.

Abstract:Evaluating the levels of the biomarker carbohydrate antigen 19-9 (CA19-9) is crucial in early cancer diagnosis and prognosis assessment. In this study, an antifouling electrochemical immunosensor was developed for the label-free detection of CA19-9, in which bovine serum albumin (BSA) and graphene were cross-linked with the aid of glutaraldehyde to form a 3D conductive porous network on the surface of an electrode. The electrochemical immunosensor was characterized through the use of transmission electron microscopy (TEM), scanning electron microscopy (SEM), atomic force microscope (AFM), UV spectroscopy, and electrochemical methods. The level of CA19-9 was determined through the use of label-free electrochemical impedance spectroscopy (EIS) measurements. The electron transfer at the interface of the electrode was well preserved in human serum samples, demonstrating that this electrochemical immunosensor has excellent antifouling performance. CA19-9 could be detected in a wide range from 13.5 U/mL to 1000 U/mL, with a detection limit of 13.5 U/mL in human serum samples. This immunosensor also exhibited good selectivity and stability. The detection results of this immunosensor were further validated and compared using an enzyme-linked immunosorbent assay (ELISA). All the results confirmed that this immunosensor has a good sensing performance in terms of CA19-9, suggesting its promising application prospects in clinical applications.

Abstract:A CA19-9 electrochemical immunosensor was constructed using a hybrid self-assembled membrane modified with a gold electrode and applied to detect real samples. Hybrid self-assembled membranes were selected for electrode modification and used to detect antigens. First, the pretreated working electrodes were placed in a 3-mercaptopropionic acid (MPA)/β-mercaptoethanol (ME) mixture for 24 h for self-assembly. The electrodes were then placed in an EDC/NHS mixture for 1 h. Layer modification was performed by stepwise dropwise addition of CA19-9 antibody, BSA, and antigen. Differential pulse voltammetry was used to characterize this immunosensor preparation process. The assembled electrochemical immunosensor enables linear detection in the concentration range of 0.05–500 U/mL of CA19-9, and the detection limit was calculated as 0.01 U/mL. The results of the specificity measurement test showed that the signal change of the interfering substance was much lower than the response value of the detected antigen, indicating that the sensor has good specificity and strong anti-interference ability. The repeatability test results showed that the relative standard deviations were less than 5%, showing good accuracy and precision. The CA19-9 electrochemical immunosensor was used for the actual sample detection, and the experimental results of the standard serum addition method showed that the RSD values of the test concentrations were all less than 10%. The recoveries were 102.4–115.0%, indicating that the assay has high precision, good accuracy, and high potential application value.

Abstract:The genome of Bacillus cereus contains 26 Nudix hydrolase genes, second only to its closest relative, Bacillus anthracis which has 30. All 26 genes have been cloned, 25 have been expressed, and 21 produced soluble proteins suitable for analysis. Substrates for 16 of the enzymes were identified; these included ADP-ribose, diadenosine polyphosphates, sugar nucleotides, and deoxynucleoside triphosphates. One of the enzymes was a CDP-choline pyrophosphatase, the first Nudix hydrolase active on this substrate. Furthermore, as a result of this and previous work we have identified a new sub-family of the Nudix hydrolase superfamily recognizable by a specific amino acid motif outside of the Nudix box.

Abstract:A novel, enhanced photoelectrochemical immunoassay was established for sensitive and specific detection of carbohydrate antigen 19-9 (CA19-9, Ag). In this protocol, TiO2 nanowires (TiO2NWs) were first decorated with Au nanoparticles to form TiO2NWs/Au hybrid structure, and then coated with CdSe@ZnS quantum dots (QDs) via the layer-by-layer method, producing TiO2NWs/Au/CdSe@ZnS sensitized structure, which was employed as the photoelectrochemical matrix to immobilize capture CA19-9 antibodies (Ab1); whereas, bipyridinium (V2+) molecules were labeled on signal CA19-9 antibodies (Ab2) to form Ab2@V2+ conjugates, which were used as signal amplification elements. The TiO2NWs/Au/CdSe@ZnS sensitized structure could adequately absorb light energy and dramatically depress electron–hole recombination, resulting in evidently enhanced photocurrent intensity of the immunosensing electrode. While target Ag were detected, the Ab2@V2+ conjugates could significantly decrease the photocurrent detection signal because of strong electron-withdrawing property of V2+ coupled with evident steric hindrance of Ab2. Thanks to synergy effect of TiO2NWs/Au/CdSe@ZnS sensitized structure and quenching effect of Ab2@V2+ conjugates, the well-established photoelectrochemical immunoassay exhibited a low detection limit of 0.0039U/mL with a wide linear range from 0.01U/mL to 200U/mL for target Ag detection. This proposed photoelectrochemical protocol also showed good reproducibility, specificity and stability, and might be applied to detect other important biomarkers.

Abstract:Carbohydrate antigen 199 (CA199) is the most sensitive marker reported for pancreatic cancer, and it is a difficult task to develop a highly sensitive assay for CA199. During the experiment, a ratiometric electrochemical immunosensor for quantitative analysis of CA199 was prepared for NiCo@Fc-MWCNTs-LDH as the electrode sensing surface and 3D-rGOF@Ag/Au as the label of Ab 2 . NiCo@Fc-MWCNTs-LDH not only provide the required signal for the immunosensor, but also have a layered structure to obtain a large specific surface area, which can provide more sites for the placement of biological molecules. rGOF has the advantages of large specific surface area and high porosity, which can adsorb Ag electrochemical probe through redox reaction. The modification of gold nanoparticles can not only enhance the electrical conductivity of nano-composites, but also immobilize more biomolecules to improve the sensitivity of electrochemical sensors. With the beefing up of CA199 concentration, the oxidation peak current of Ag increases and the oxidation peak current of Fc-COOH decreases. The ratio (y = I Ag /I Fc-COOH ) of two different signals was linear with the logarithm of CA199 concentration in a certain value range. Under optimal conditions, the immunosensor showed excellent performance in the concentration range of 0.0001 U/mL to 10 U/mL, and the detection limit was 5.55 × 10 −4 U/mL. The strategy could clearly discriminate between matched and mismatched targets, demonstrating high specifificity. This approach further detects CA199 in human plasma to differentiate pancreatic cancer patients from healthy individuals with high accuracy. This method also provided a new idea for the ultrasensitive quantitative detection of other biomarkers.

Abstract:This work took advantage of 3D biomimetic hydrangea-like bismuth oxychloride (BiOCl) and PtNi nanocubes to fabricate a facile electrochemical immunosensor for the detection of CA19-9. Herein, APTES functionalized BiOCl (APTES-BiOCl) with a large specific surface areas can provide an eco-friendly microenvironment for biomolecules. The PtNi nanocubes prepared by the classic hydrothermal method have good conductivity, which can facilitate electron transfer highly efficient. Meanwhile, PtNi nanocubes can immobilize the anti-CA19-9 via covalent Pt-N bonds to anchor a great number of biomolecules more firmly. In addition, reduced graphene oxide (rGO) with excellent conductivity was implemented as a matrix, which greatly promoted electron transfer at the sensing interface. Under optimal conditions, the proposed immunosensor can detect CA19-9 sensitively, from which a linear range of 0.01–150 U·mL–1 was derived accompanied with a low detection limit of 0.0034 U·mL–1. The results illustrate that the designed immunosensor exhibited excellent specificity, good reproducibility, and satisfactory stability. Moreover, it can be applied in the detection of real samples, which indicates that the immunosensor has great potential for early screening and diagnosis of diseases.

Abstract:We describe a novel electrochemical immunosensor for carbohydrate antigen 19-9 (CA19-9) based on the immobilization of CA19-9 with titania sol-gel on a graphite electrode (GE) by vapor deposition. The CA19-9 membrane was characterized using scanning electron microscopy and proved to be chemically clean, porous and homogeneous. The incubation of the immunosensor in a solution containing horseradish peroxidase (HRP)-labeled CA19-9 antibody led to the binding of HRP-labeled antibody with the immobilized antigen. The immobilized HRP catalyzed the oxidation of catechol by H2O2 and this provided a competitive method for the measurement of serum CA 19-9. The response current decreased with increasing CA 19-9 concentration in the incubation solution. The effects of pH, amount of HRP-labeled antibody, incubation time and temperature were explored to provide optimum analytical performance. Under optimal conditions, the current decrease of the immunosensor was proportional to CA19-9 concentrations in the range of 3-20 U/ml with a detection limit of 2.68 U/ml at a current decrease of 10%. The detection of CA19-9 in two serum samples obtained from clinically diagnosed patients with pancreatic carcinoma showed acceptable accuracy. Bimetallic cerium and ferric oxides nanoparticles embedded within mesoporous carbon matrix: Electrochemical immunosensor for sensitive detection of carbohydrate antigen 19-9

Abstract:A label-free electrochemical immunosensor was successfully developed for sensitively detecting carbohydrate antigen 19-9 (CA19-9) as a cancer marker. To achieve this, a series of bimetallic cerium and ferric oxide nanoparticles embedded within the mesoporous carbon matrix (represented by CeO2/FeOx@mC) was obtained from the bimetallic CeFe-based metal organic framework (CeFe-MOF) by calcination at different high temperatures. The formed CeO2 or FeOx nanoparticles were uniformly distributed within the highly graphitized mesoporous carbon matrix at the calcination temperature of 500 °C (represented by CeO2/FeOx@mC500). However, the obtained nanoparticles were aggregated into large size when calcined at the temperatures of 700 and 900 °C. The CA 19-9 antibody can be anchored to the CeO2/FeOx@mC network through chemical absorption between carboxylic groups of antibody and CeO2 or FeOx by ester-like bridging. The CeO2/FeOx@mC500-based immunosensor displayed superior sensing performance to the pristine CeFe-MOF, CeO2/FeOx@mC700- and CeO2/FeOx@mC900-based ones. Electrochemical impedance spectroscopy results showed that the developed immunosensor exhibited an extremely low detection limit of 10 μU·mL-1 (S/N = 3) within a wide range from 0.1 mU·mL-1 to 10 U·mL-1 toward CA 19-9. It also illustrated excellent specificity, good reproducibility and stability, and acceptable application analysis in the human serum solution which was diluted 100-fold with 0.01 M PBS solution (pH 7.4) and spiked with different amounts of CA19-9. Consequently, the proposed electrochemical immunosensor is capable enough of determining CA 19-9 in clinical diagnostics.
Ketone body utilization drives tumor growth and metastasis

Abstract:We have previously proposed that catabolic fibroblasts generate mitochondrial fuels (such as ketone bodies) to promote the anabolic growth of human cancer cells and their metastasic dissemination. We have termed this new paradigm “two-compartment tumor metabolism.” Here, we further tested this hypothesis by using a genetic approach. For this purpose, we generated hTERT-immortalized fibroblasts overexpressing the rate-limiting enzymes that promote ketone body production, namely BDH1 and HMGCS2. Similarly, we generated MDA-MB-231 human breast cancer cells overexpressing the key enzyme(s) that allow ketone body re-utilization, OXCT1/2 and ACAT1/2. Interestingly, our results directly show that ketogenic fibroblasts are catabolic and undergo autophagy, with a loss of caveolin-1 (Cav-1) protein expression. Moreover, ketogenic fibroblasts increase the mitochondrial mass and growth of adjacent breast cancer cells. However, most importantly, ketogenic fibroblasts also effectively promote tumor growth, without a significant increase in tumor angiogenesis. Finally, MDA-MB-231 cells overexpressing the enzyme(s) required for ketone re-utilization show dramatic increases in tumor growth and metastatic capacity. Our data provide the necessary genetic evidence that ketone body production and re-utilization drive tumor progression and metastasis. As such, ketone inhibitors should be designed as novel therapeutics to effectively treat advanced cancer patients, with tumor recurrence and metastatic disease. In summary, ketone bodies behave as onco-metabolites, and we directly show that the enzymes HMGCS2, ACAT1/2 and OXCT1/2 are bona fide metabolic oncogenes.
A Nanotube/Polymer Composite Biosensing Thin-Film Transistor Platform for C-Reactive Protein Detection

Abstract:In the present study, a back-gate thin-film transistor (TFT) based on a single-walled carbon nanotubes (SWCNTs)/poly(9,9-dioctylfluorene-co-bithiophene) (F8T2) composite semiconductor is proposed as electronic label-free immunosensor for the detection of the C-reactive protein (CRP). The nano-composite semiconducting ultra-thin film is deposited on the channel area of the TFT and is functionalized with a monoclonal anti-CRP antibody coating acting as biorecognition layer. The immunesensor was then used for the selective detection of the CRP both in phosphate buffered saline (PBS) solution and in human serum samples. The proposed immunosensor exhibits high current (10 - 20 μA) and good field-effect carriers mobility. Furthermore, the CRP could be revealed at a detection limit as low as 1 mg/L and in a wide dynamic range (from 0.4 nM to 2.2 μM).

Abstract:Tumor-derived exosomal miRNAs may have important functions in the onset and progression of cancers and are potential biomarkers for early diagnosis and prognosis monitoring. Yet, simple, sensitive, and label-free detection of exosomal miRNAs remains challenging. Herein, an ultrasensitive, label-free, and stable field-effect transistor (FET) biosensor based on a polymer-sorted high-purity semiconducting carbon nanotube (CNT) film is reported to detect exosomal miRNA. Different from conventional CNT FETs, the CNT FET biosensors employed a floating gate structure using an ultrathin Y2O3 as an insulating layer, and assembled Au nanoparticles (AuNPs) on Y2O3 as linkers to anchor probe molecules. A thiolated oligonucleotide probe was immobilized on the AuNP surface of the sensing area, after which miRNA21 was detectable by monitoring the current change before and after hybridization between the immobilized DNA probe and target miRNA. This method achieved both high sensitivity (LOD: 0.87 aM) and high specificity. Furthermore, the FET biosensor was employed to test clinical plasma samples, showing significant differences between healthy people and breast cancer patients. The CNT FET biosensor shows the potential applications in the clinical diagnosis of breast cancer.

A label-free electrochemical biosensor based on a bimetallic organic framework for the detection of carbohydrate antigen 19-9

Abstract:Carbohydrate antigen 19-9 (CA19-9) is an important marker for pancreatic cancer, ovarian cancer and other tumors, and its rapid and stable detection is the basis for early diagnosis and treatment. In this paper, a label-free electrochemical immunosensor for the sensitive detection of CA19-9 has been developed. First, the synthesis of two novel core-shell bimetallic nanomaterials, namely Ce-MOF-on-Fe-MOF and Fe-MOF-on-Ce-MOF, was accomplished using the MOF-on-MOF approach. The poor electrical conductivity of MOF materials was addressed by incorporating polyethylenimide (PEI) functionalized rGO with Ce-MOF-on-Fe-MOF and Fe-MOF-on-Ce-MOF nanomaterials. Simultaneously, toluidine blue (Tb) was employed as a redox probe and physically adsorbed onto the synthesized materials, resulting in the formation of two nanomaterials: rGO@Ce-MOF-on-Fe-MOF@Tb and rGO@Fe-MOF-on-Ce-MOF@Tb. The fundamental characterization reveals that the sensing performance of the rGO@Ce-MOF-on-Fe-MOF@TB-based immune sensor surpasses that of the rGO@Fe-MOF-on-Ce-MOF@TB-based immune sensor, which is attributed to the fact that, unlike the interlayer-constrained structure of Fe-MOF-on-Ce-MOF, in Ce-MOF-on-Fe-MOF, Ce-MOF penetrates into Fe-MOF to form a heterogeneous structure due to the relatively large pore size of Fe-MOF, which better combines the excellent biocompatibility and strong anchoring effect of Fe MOFs on antibodies, as well as the high electrochemical activity and conductivity of Ce-MOF, to enhance sensing performance. The proposed label-free immunosensor based on rGO@Ce-MOF-on-Fe-MOF@Tb has a wide linear range (1-100 000 mU mL-1), a low detection limit (0.34 mU mL-1), good stability, reproducibility, and repeatability, and satisfactory applicability, which provides a potential platform for clinical applications.

Abstract:Accurate and sensitive detection of cancer antigen 125 (CA125) is important in early ovarian cancer diagnosis and monitoring. In the present work, a novel electrochemical ratiometric immunosensor for the determination of CA125 based on three-dimensional reduced graphene oxide/multi-walled carbon nanotube carboxylic acid- thionine (3DrGO/MWCNTs-Thi) and UIO-66-NH 2 /ferrocenecarboxylic acid (UiO-66-Fc) was constructed. 3DrGO/MWCNTs-Thi of immobilized primary antibody (Ab 1 ) and UiO-66-Fc of labeled second antibody (Ab 2 ) were used as capture probes and signaling probes for CA125, respectively. As the concentration of CA125 increases, the oxidation peak currents (I Thi and I Fc ) generated by the two materials were changed in opposite directions, and therefore the ratio of the currents (I Thi /I Fc ) was negatively correlated with the concentration of CA125. Thanks to the amplification of high loading of signaling molecules by the framework nanocomposites and the ratiometric strategy, the constructed electrochemical immunosensor exhibits outstanding accuracy, stability, and sensitivity. Under optimal conditions, the detection range spanned from 0.01 U mL −1 to 80 U mL −1 , with a calculated limit of detection (LOD) of 0.0089 U mL −1 . The immunosensor was successfully applied to real serum sample tests, indicating its potential for clinical detection applications.

Abstract:In this study, for the first time, the erbium-doped graphene quantum dots (Er-GQDs) were decorated onto a three-dimensional MoS 2 nanoflower (EGM) for electrochemically sensitive and selective detection of carbohydrate antigen, CA 19-9, in phosphate buffer saline (PBS) and biological fluids. The MoS 2 nanoflowers (MoS 2 NF) were hydrothermally prepared at 200 °C for 24 h, and then 2 – 8 nm of Er-GQDs were uniformly deposited onto 200 nm MoS 2 NF. The MoS 2 NF has a high electric conductivity for electrochemical performance, whereas Er-GQDs act as an effective anchor with anti-CA19-9 antibodies (CA19-9 Ab) for ultra-selectivity to CA19-9 antigen. The SPE||EGM/Ab immunosensor has substantial benefits by providing a long-lasting, ultra-sensitive, and highly selective platform for CA19-9 antigen detection. The dynamic ranges in PBS, human serum, and saliva are 5×10 −5 – 250, 5×10 −4 – 250, and 3×10 −4 – 250 U mL −1 , respectively, with limits of detection (LODs) of (0.18 − 2.95) ×10 −4 U mL −1 . Furthermore, the SPE||EGM/Ab immunosensor can detect CA19-9 in human serum with a recovery of 97 – 108%. The non-invasive detection of CA19-9 in saliva is stably sensitive before and after 1 h of breakfast. Moreover, the SPE||EGM/Ab immunosensor has excellent selectivity toward CA19-9 over the other 17 interferences. Results elaborate that the SPE||EGM/Ab immunosensor is a promising electrochemical biosensing probe for detecting CA19-9 in human serum and saliva for clinical diagnosis and prognosis of pancreatic cancer in the early stage.

Abstract:It is tremendously desirable for the timely and effective detection of cancer to facilitate the ultra-highly sensitive monitoring of tumor marker in clinical serum sample. In this study, an electromagnetic and chemical synergistically enabled recyclable immunoassay based on surface-enhanced Raman scattering (SERS) was proposed by realizing the anisotropic growth of sea-urchin-like gold nanoflowers (Au NFs) on two-dimensional red phosphorus (RP) nanoplates. Besides the achieved enhancement factor as high as 2.24 × 106, it was found that the photocatalytic and SERS activities were kept at a high level for the hybrid substrate of RP/Au NFs throughout 7 cycles of immunoassay. In combination with a non-metallic immunoprobe, the limit of detection was drove to 7.41 × 10−5 IU·mL−1 for cancer antigen 19–9. The comparative experiments of nonspecific monitoring verified the promising selectivity of this strategy. Considering the intriguing features of high sensitivity, recyclability, and specificity, the proposed multifunctional RP/Au NFs exhibited its superior role in the early detection of cancer and can be adapted for point-of-care diagnosis.

Abstract:A novel label-free electrochemical immunosensor for the ultrasensitive determination of cancer antigen (CA724) was developed using a novel composite of CoAl layered double hydroxides (CoAl-LDH) and carboxyl-functionalized multiwall carbon nanotubes (MWCNTs-COOH) as platform and ferrocenecarboxylic acid (Fc-COOH) as signal label. The MWCNTs-COOH/Fc-COOH@CoAl-LDH composite was prepared by a convenient and simple one-step ultrasonic method, and various characterization techniques consisting of scanning electron microscopy (SEM), transimission electronic microscopy (TEM), TEM-Mapping, fourier transform infrared (FT-IR), X-ray diffraction (XRD) and X-ray photoelectronic energy spectrum (XPS) were applied to study the size and morphological features. Due to its large specific surface area and multilayer structure, the CoAl-LDH can be post-doped to embed a large amount of signal probe to realize the amplification of the internal reference signal Fc. In addition, the higher conductivity of MWCNTs-COOH compensates for the deficiency of CoAl-LDH, which effectively strengthened the electron transfer efficiency of electrochemical signaling substances. The optimal experimental conditions were detected to be 2.5 mmol of Fc-COOH, 4.0 mg/mL of concentration, pH 6.0, incubation time of 40 min, and incubation temperature of 37 °C. Under optimal conditions, the fabricated sensor exhibits linearity in a wide dynamic range covering the physiological concentration, from 0.001 to 100 U/mL and the limit of detection (LOD) was 0.03962 mU/mL, the calibration equation is stated as △I = 7.76363 log 10 C CA724 + 40.50351 (R 2 = 0.99674). The sensor is successfully detects trace levels of CA724 in human serum with excellent recovery rates ranging from 100.52 %-102.30 %, proving the synergy of MWCNTs-COOH/Fc-COOH@CoAl-LDH as a promising platform for electrochemical sensing for clinical detection of other disease markers.
