Abstract:Gold nanocages with a relatively small size (e.g., approximately 45 nm in edge length) have been developed, and the structure of these nanocages was tailored to achieve strong absorption in the near-infrared (NIR) region for photothermal cancer treatment. Numerical calculations show that the nanocage has a large absorption cross section of 3.48 x 10(-14) m(2), facilitating conversion of NIR irradiation into heat. The gold nanocages were conjugated with monoclonal antibodies (anti-HER2) to target epidermal growth factor receptors (EGFR) that are overexpressed on the surface of breast cancer cells (SK-BR-3). Our preliminary photothermal results show that the nanocages strongly absorb light in the NIR region with an intensity threshold of 1.5 W/cm(2) to induce thermal destruction to the cancer cells. In the intensity range of 1.5-4.7 W/cm(2), the circular area of damaged cells increased linearly with the irradiation power density. These results suggest that this new class of bioconjugated gold nanostructures, immuno gold nanocages, can potentially serve as an effective photothermal therapeutic agent for cancer treatment.

Abstract:In terms of synergistic cancer therapy, biological nanomaterials with a second near-infrared (NIR-II) window response can greatly increase photothermal effects and photoacoustic imaging performance. Herein, we report a novel stimuli-responsive multifunctional drug-loading system which was constructed by integrating miniature gold nanorods (GNR) as the NIR-II photothermal nanorods and cyclic ternary aptamer (CTA) composition as a carrier for chemotherapy drugs. In this system, doxorubicin hydrochloride (DOX, a chemotherapy drug) binds to the G-C base pairs of the CTA, which exhibited a controlled release behavior based on the instability of G-C base pairs in the slightly acidic tumor microenvironment. Upon the 1064 nm (NIR-II biowindow) laser irradiation, the strong photothermal and promoted cargo release properties endow gold nanorods@CTA (GNR@CTA) nanoparticles displaying excellent synergistic anti-cancer effect. Moreover, the GNR@CTA of NIR also possesses thermal imaging and photoacoustic (PA) imaging properties due to the strong NIR region absorbance. This work enables to obtaining a stimuli-responsive "all-in-one" nanocarrier, which are promising candidate for bimodal imaging diagnosis and chemo-photothermal synergistic therapy.

Abstract:Local drug delivery offers a means of achieving a high concentration of therapeutic agents directly at the tumor site, whilst minimizing systemic toxicity. For heterogenous cancers such as glioblastoma, multimodal therapeutic approaches hold promise for better efficacy. Herein, we aimed to create a well-defined and reproducible drug delivery system that also incorporates gold nanorods for photothermal therapy. Solvent-assisted micromolding was used to create uniform sacrificial templates in which microscale hydrogels were formed with and without gold nanorods throughout their structure. The microscale hydrogels could be loaded with doxorubicin, releasing it over a period of one week, causing toxicity to glioma cells. Since these microscale hydrogels were designed for direct intratumoral injection, therefore bypassing the blood–brain barrier, the highly potent breast cancer therapeutic doxorubicin was repurposed for use in this study. By contrast, the unloaded hydrogels were well tolerated, without decreasing cell viability. Irradiation with near-infrared light caused heating of the hydrogels, showing that if concentrated at an injection site, these hydrogels maybe able to cause anticancer activity through two separate mechanisms.
Gold Nanorods-Based Smart Nanoplatforms for Synergic Thermotherapy and Chemotherapy of Tumor Metastasis

Abstract:The combination therapy of photothermal therapy and chemotherapy as a promising strategy has drawn extensive attention by overcoming the limitations of conventional treatments in tumor therapy. Gold nanorods-based nanoplatforms were herein designed by integrating doxorubicin (DOX)- and polydopamine-coated gold nanorods (GNRs@PDA) for tumor metastasis inhibition and multifunctional drug delivery. The GNRs@PDA poly(ethylene glycol) (PEG) DOX nanocomplex showed robust stability and excellent near-infrared (NIR) photothermal conversion efficiency under laser irradiation. The release of loaded DOX from GNRs@PDA PEG DOX nanocomposites was improved in tumor microenvironments. Furthermore, the PDA-functionalized GNR nanocomposites were expected to be potential photoacoustic imaging agents for imaging-guided tumor therapy. Upon NIR laser irradiation, the efficiency of tumor inhibition of GNRs@PDA PEG DOX is greater than that of the other group in vitro and in vivo, which was confirmed by immunohistochemistry staining, demonstrating a promising strategy for suppression of tumor metastasis and low long-term systemic toxicity. These results illustrated a promising strategy of tailor-made GNRs@PDA PEG DOX nanoplatforms for ablation of tumor and suppression of tumor metastasis in clinical application.
Near Infrared Laser-Induced Targeted Cancer Therapy Using Thermoresponsive Polymer Encapsulated Gold Nanorods

Abstract:External stimuli, such as ultrasound, magnetic field, and light, can be applied to activate in vivo tumor targeting. Herein, we fabricated polymer encapsulated gold nanorods to couple the photothermal properties of gold nanorods and the thermo- and pH-responsive properties of polymers in a single nanocomposite. The activation mechamism was thus transformed from heat to near-infrared (NIR) laser, which can be more easily controlled. Doxorubicin, a clinical anticancer drug, can be loaded into the nanocomposite through electrostatic interactions with high loading content up to 24%. The nanocomposite's accumulation in tumor post systematic administration can be significantly enhanced by NIR laser irradiation, providing a prerequisite for their therapeutic application which almost completely inhibited tumor growth and lung metastasis. Since laser can be manipulated very precisely and flexibly, the nanocomposite provides an ideally versatile platform to simultaneously deliver heat and anticancer drugs in a laser-activation mechanism with facile control of the area, time, and dosage. The NIR laser-induced targeted cancer thermo-chemotherapy without using targeting ligands represents a novel targeted anticancer strategy with facile control and practical efficacy.

Abstract:Rattle-type nanostructures with movable cores, porous shells, and hollow interiors have become attractive nanoplatforms in the field of nanomedicine, especially for targeted and stimuli-responsive drug delivery. In this work, rattle-type gold nanorods@void@porous-SiO2 (GVPS) nanocomposites were fabricated facilely using the surface-protecting etching method and exhibited high photothermal conversion efficiency. Taking advantage of the porous shell and hollow interior, the nanocomposites have abundant space for drug loading and successfully improved the drug loading capacity up to ∼19.6%. To construct a multifunctional drug delivery system, GVPS was further functionalized with polyethylene glycol (PEG) and cyclic RGD peptides to improve biocompatibility as well as selectivity toward the targeted cancer cells. Besides, to achieve precise regulation and near-infrared laser activation of the drug release, a phase-changing material, 1-tetradecanol (1-TD, Tm: 39 °C), was employed as gatekeepers in this system. After incubation with an αVβ3 integrin receptor-overexpressed cell line, the as-prepared GVPSPR-DOX/TD nanocomposites exhibited great performances in combined photothermal therapy and chemotherapy. It is worth noting that the combined therapy showed superior efficiency in cancer cell killing to chemotherapy or photothermal therapy alone.

Abstract:Photothermal therapy (PTT) has become effective method for the treatment of malignant cancer. The development of PTT system with high anti-tumour effect is still the feasible research direction. Here, a new type of gold nanorods (AuNRs)-doxorubicin (DOX)/mPEG10K-peptide/P(AAm-co-AN) (APP-DOX) nano drug delivery system was proposed. Among them, AuNRs was used as high-efficiency photothermal agent. APP-DOX had a suitable size and can be targeted to accumulate in tumour tissues through circulation in the body. The abundant matrix metalloproteinase 2 (MMP-2) in the tumour environment intercepted and cut off the short peptide chain structure grafted on APP-DOX. At the same time, the removal of the PEG segment leaded to an increase in the hydrophobic properties of the system. Nanoparticles aggregated into large particles, causing them to stay and aggregate further at the tumour site. When irradiated by 808 nm near-infrared laser, APP-DOX achieved a gradual heating process. High temperature can effectively ablate tumours and enable UCST polymer to achieve phase transition, resulting in more anti-cancer drugs loaded in the polymer layer DOX was released, effectively killing cancer cells. Animal experiments had verified the possibility of the nano drug-carrying system and good tumour treatment effect. What’s more worth mentioning is that compared with free DOX, the nano drug delivery system had lower biological toxicity and not cause obvious harmful effects on normal organs and tissues.
Targeting Mesoporous Silica-Encapsulated Gold Nanorods for Chemo-Photothermal Therapy with Near-Infrared Radiation

Abstract:Mesoporous silica-encapsulated gold nanorods (GNRs@mSiO(2)) have great potential both in photothermal therapy and drug delivery. In this paper, we firstly developed GNR5@mSiO(2) as a synergistic therapy tool for delivery heat and drug to the tumorigenic region. We studied the ablation of tumor both in vitro and in vivo by the combination of photothermal therapy and chemotherapy using doxorubicin (DOX)-loaded GNR5@rnSiO(2). Significantly greater cell killing was observed when A549 cells incubated with DOX-loaded GNR5@mSiO(2) were irradiated with near-infrared (NIR) illumination, attributable to both GNRs@mSiO(2)-mediated photothermal ablation and cytotoxicity of light-triggered DOX release. We then performed in vivo therapy studies and observed a promising tumor treatment. Compared with chemotherapy or photothermal treatment alone, the combined treatment showed a synergistic effect, resulting in higher therapeutic efficacy. Abstract:Gold nanorods (GNRs) are well known in photothermal therapy based on near-infrared (NIR) laser absorption of the longitudinal plasmon band. Herein, we developed an effective stimulus system -- GNRs and doxorubicin co-loaded polymersomes (P-GNRs-DOX) -- to facilitate co-therapy of photothermal and chemotherapy. DOX can be triggered to release once the polymersomes are corrupted under local hyperthermic condition of GNRs induced by NIR laser irradiation. Also, the cytotoxicity of GNRs caused by the residual cetyltrimethylacmmonium bromide (CTAB) was reduced by shielding the polymersomes. The GNRs-loaded polymersomes (P-GNRs) can be efficiently taken up by the tumor cells. The distribution of the nanomaterial was imaged by IR-820 and quantitatively analyzed by ICP-AES. We studied the ablation of tumor cells in vitro and in vivo, and found that co-therapy offers significantly improved therapeutic efficacy (tumors were eliminated without regrowth.) compared with chemotherapy or photothermal therapy alone. By TUNEL immunofluorescent staining of tumors after NIR laser irradiation, we found that the co-therapy showed more apoptotic tumor cells than the other groups. Furthermore, the toxicity study by pathologic examination of the heart tissues demonstrated a lower systematic toxicity of P-GNRs-DOX than free DOX. Thus, the chemo-photothermal treatment based on polymersomes loaded with DOX and GNRs is a useful strategy for maximizing the therapeutic efficacy and minimizing the dosage-related side effects in the treatment of solid tumors.
Multidentate Polyethylene Glycol Modified Gold Nanorods for in Vivo Near-Infrared Photothermal Cancer Therapy.

Abstract:Gold nanorods (AuNRs), because of their strong absorption of near-infrared (NIR) light, are very suitable for in vivo photothermal therapy of cancer. However, appropriate surface modification must be performed on AuNRs before their in vivo application because of the high toxicity of their original stabilizer cetyltrimethylammonium bromide. Multidentate ligands have attracted a lot of attention for modification of inorganic nanoparticles (NPs) because of their high ligand affinity and multifunctionality, while the therapeutic effect of multidentate ligands modified NPs in vivo remains unexplored. Here, we modified AuNRs with a polythiol PEG-based copolymer. The multidentate PEG coated AuNRs (AuNR-PTPEGm950) showed good stabilities in high saline condition and wide pH range. And they had much stronger resistance to ligand competition of dithiothreitol (DTT) than AuNRs coated by monothiol-anchored PEG. The AuNR-PTPEGm950 had very low cytotoxicity and showed high efficacy for the ablation of cancer cells in vitro. Moreover, the AuNR-PTPEGm950 showed good stability in serum, and they had a long circulation time in blood that led to a high accumulation in tumors after intravenous injection. In vivo photothermal therapy showed that tumors were completely cured without reoccurrence by one-time irradiation of NIR laser after a single injection of these multidentate PEG modified AuNRs.
A Redox-Responsive, In-Situ Polymerized Polyplatinum(IV)-Coated Gold Nanorod as An Amplifier of Tumor Accumulation for Enhanced Thermo-Chemotherapy

Abstract:It remains a major challenge to develop an effective therapeutic system based on gold nanorods (GNRs) for cancer therapy. Herein, we developed a redox-responsive, in-situ polymerized polyplatinum(IV)-coated gold nanorod ((IV)) with coupling of the near-infrared (NIR)-induced hyperthermal effect and redox-triggered drug release in one therapeutic platform as an amplifier of tumor accumulation through mild hyperthermia for enhanced synergistical thermo-chemotherapy. After in-situ polymerized with 2-methacryloyloxy ethyl phosphorylcholine (MPC) and Pt(IV) complex-based prodrug monomer (PPM) onto the surface of GNRs, the nanosized (IV) exhibited the advantages of high drug encapsulation efficiency, triggered drug release, and reduced side effect. As demonstrated by thermal imaging and photoacoustic imaging in vitro and in vivo, this (IV) exhibited an excellent NIR-associated hyperthermal effect and outstanding capacity of tumor accumulation. Importantly, under a mild hyperthermia process, the vascular endothelial growth factor (VEGF) and hypoxia-inducible factor-1α (HIF-1α) were upregulation, resulting in angiogenic vessel around the tumor. Combination with accelerated blood flow and angiogenesis by mild hyperthermia, a dramatic increase of drug accumulation in tumor could be realized after systematic administration. As a result, this amplification fashion of tumor accumulation would contribute the (IV) to inhibit tumor progression effectively. Such a facile and simple methodology for enhanced therapeutic effect based on GNRs holds great promises for cancer therapy with further development.

Abstract:Novel composite nanoparticles based on poly(N-isopropylacrylamide-co-N-hydroxymethyl acrylamide) (P(NIPAM-co-NHMA)) layer coated gold nanorod@mesoporous silica (GNR@mSiO(2)) has been successfully synthesized by precipitation polymerization. The composite nanoparticles exhibited a thermo/near-infrared (NIR) light sensitivity. The volume phase transition temperature (VPTT) could be precisely regulated by the content of NHMA in monomers and an excellent photothermal conversion effect was expressed due to the surface plasmon resonance of gold nanorods in the composite nanoparticles. Doxorubicin hydrochloride (DOX) was applied as a model drug and the drug storage/release behavior was investigated. The results demonstrated that DOX could be effectively loaded into the composite nanoparticles with 21% loading capacity. The cumulative release of DOX-loaded composite nanoparticles was temperature dependent and the release rate was significantly enhanced above the VPTT. Therefore, the composite nanoparticles applied as a drug delivery system could reduce the side effect of DOX to normal tissues as only a small fraction of DOX was released from the composite nanoparticles at 37 degrees C. In addition, the DOX-loaded composite nanoparticles demonstrated a synergistic effect, the therapeutic efficacy was improved significantly by the combination of photothermal therapy and traditional chemotherapy with low composite nanoparticle concentration and short laser irradiation time in an in vitro study.
Gold Nanorods with Silica Shell and PAMAM Dendrimers for Efficient Photothermal Therapy and Low Toxic Codelivery of Anticancer Drug and Sirna

Abstract:The multifunctional nanocomposites with grafting polyamidoamine dendrimers up to third generation (G(3)) are grown onto the surface of mesoporous silica-coated gold nanorods (AuNRs@SiO2) via a divergent technique. The resultant AuNRs@SiO2-G(3) nanocomposites with uniform size and excellent stability not only enable their utilization as targeted contrast agents for photothermal cancer therapy but also serve as scaffolds for the intracellular delivery of anticancer drug and small interfering RNA (siRNA) to enhance the efficiency of cancer therapy. The controlled doxorubicin (DOX) release from AuNRs@SiO2-G(3) nanocomposites is significantly improved under lower pH condition and near-infrared laser irradiation. The Bcl-2-targeted siRNA is transfected into tumor cells and induces knockdown of the protein expression, which is confirmed by western blot assays. Furthermore, the complementary effect of chemo- and gene therapy to MCF-7 cells for improved photothermal therapy is demonstrated by MTT assay. The DOX and siRNA coloaded AuNRs@SiO2-G(3) nanocomposites show much lower cytotoxicity compared to the DOX, resulting in low toxicity to normal tissues. The multifunctional nanocomposites have potential application as nanoplatforms to integrate the photothermal-chemo-gene tumor therapy with high efficiency.
Gold-nanobranched-shell based drug vehicles with ultrahigh photothermal efficiency for chemo-photothermal therapy

Abstract:Development of combined chemo-photothermal nanoplatform is of great interest for enhancing antitumor efficacy. Herein, a multifunctional drug delivery system was synthesized based on gold-nanobranched coated betulinic acid liposomes (GNBS-BA-Lips) for chemo-photothermal synergistic therapy. In this system, GNBS-BA-Lips exhibited broad near-infrared (NIR) absorption, preferable photothermal response and good photostability under NIR irradiation. Importantly, the gold-nanobranched nanostructure possessed high photothermal conversion efficiency (η = 55.7%), and the temperature change (ΔT) reached 43.2 °C after laser irradiation for 5 min. Upon NIR irradiation, the nanocarriers apparently endowed higher cell uptake, resulting in an enhanced intracellular drug accumulation. Furthermore, the tumor growth inhibition ratio achieved from chemo-photothermal therapy of GNBS-BA-Lips was 86.9 ± 1.1%, which was higher than that of the chemotherapy or photothermal therapy alone, showing an outstanding synergistic anticancer effect. Our data suggested that the nanoplatform should be considered as a critical platform in the development of cancer multi-mode therapies.

Abstract:Abstract Owing to strong and tunable surface plasmon resonance (SPR) effect and good biocompatibility, gold nanoparticles have been suggested to be a versatile platform for a broad range of biomedical applications. In this study, a new nanoplatform of thermo-responsive polymer encapsulated gold nanorods incorporating indocyanine green (ICG) was designed to couple the photothermal properties of gold nanorods (AuNRs) and the photodynamic properties of ICG to enhance the photodynamic/photothermal combination therapy (PDT/PTT). In addition to the significantly increased payload and enhancing photostability of ICG, the polymer shell in the nanoplatform also has thermo-responsive characteristics that can control the release of drugs at tumour sites upon the laser irradiation. On the basis of these improvements, the nanoplatform strongly increased drug aggregation at the tumour site and improved the photothermal/photodynamic therapeutic efficacy. These results suggest that this nanoplatform would be a great potential system for tumour imaging and antitumour therapy.

Abstract:Gold nanomaterials with light-responsive properties can be exploited as light-triggered delivery vehicles to enhance the therapeutic efficacy of anticancer drugs. Additionally, different wavelengths of light can be utilized to achieve the combined effects of light-triggered release of therapeutics and light-induced localized heating, which results in improved anticancer efficacy. Herein, we describe methods to develop gold nanorod (AuNR) complexes that provide drug delivery or photothermal therapy when activated by ultraviolet (UV) or near-infrared (NIR) wavelengths of light, respectively. The surface functionalization of AuNRs with three key components is presented. The first component, cyclodextrin, serves to encapsulate drugs of interest. The second component, dextran-phenyl-azo-benzoic acid (DexAzo), serves as a capping agent that undergoes a conformational change upon UV light activation to expose the drugs for release. The third component is a folic acid-based targeting ligand that provides efficient delivery of the AuNR complexes to cancer cells. The dual wavelength activation of these drug-loaded AuNR complexes, which enables one to achieve highly efficient anticancer therapy through the combined effects of UV-triggered drug release and NIR-induced hyperthermia, is also described.

Abstract:New combination therapy strategy, which takes the advantages of co-delivery two or more therapeutic agents in one nanocarrier platform, has been widely used in the clinic and achieved immense popularity in cancer treatment. Here, we have rationally developed a multifunctional platform using a self-assembly strategy to incorporate materials with specific functions of chemotherapeutics, hyperthermia, and especially immunotherapy, which can collectively contribute to the effective cancer treatment. We design the immunomodulatory CpG ODNs based platform that is conjugated with NIR-responsive gold nanorods and doxorubicin for cancer therapy. The gold nanorods can be applied as the nanocarrier to simultaneously address the three kinds of treatments, which lead to a significant benefit relative to the use of each method alone. Both in vitro and in vivo assays reveal that this engineered vehicle exhibits significant antitumor efficacy. Our studies provide strong evidence that the AuNRs-CpG-Dox conjugates can be utilized as efficient antitumor agents.
Nir-Triggered Chemo-Photothermal Therapy By Thermosensitive Gold Nanostar@Mesoporous Silica@Liposome-Composited Drug Delivery Systems

Abstract:Multimodal therapeutic strategy that combines drug delivery with other treatments (e.g., photodynamic therapy) is highly desired for biomedical applications. Here, we engineer a promising versatile platform based on gold nanostar liposomeentrapped mesoporous silica core-shell nanostructure (GSMS@lip). This system not only served as a container for therapeutic drugs (hydrophilic doxorubicin and hydrophobic docetaxel), but also had high photothermal conversion efficiency. Specifically, the thermosensitive liposome wrapping around mesoporous silica acted as a gatekeeper to encapsulate the drug molecules within the GSMS/Dox/Doc@lip drug delivery system. After exposure to near-infrared (NIR) laser, gold nanostar significantly raised the ambient temperature via photothermal conversion, thereby rupturing the thermosensitive liposomes and subsequently triggering the drug release. Benefiting from the combined chemo-photothermal therapy, this composite system showed enhanced cytotoxicity to MDA-MB-231 and MCF-7 cells. Furthermore, effective drug release and synergistic inhibition of tumor growth were achieved after intratumoral injection of GSMS/Dox/Doc@lip followed by NIR laser irradiation. The proposed multifunctional drug delivery system is capable of codelivering hydrophilic-hydrophobic drugs, providing a potential strategy for multimodal tumor therapies.

Abstract:The rational design of the nanocarriers with active tumor targeting and specific chemotherapy for heat shock protein 90 (Hsp90) inhibition is attractive for combined chemo-photothermal therapy (PTT). Herein, a smart gold nanorod (GNR)-cored micelle (T-GNR AAG ) was developed to encapsulate 17-allylamino-17-demethoxygeldanamycin (17-AAG, Hsp90 inhibitor) through a facile preparation approach. The characteristics of T-GNR AAG were evaluated both in vitro and in vivo . The designed nanoplatform possessed sufficient 17-AAG loading content and lactobionic acid-mediated active targeting for hepatoma cells. More importantly, the 808 nm laser irradiation not only initiated PTT for cell killing but also remotely triggered 17-AAG liberation within cancer cells via inducing the phase transition of poly(ɛ-caprolactone). All these features forcefully supported the effectiveness of T-GNR AAG in tumor growth inhibition. This work represents a proof-of-concept combinatorial chemo-PTT treatment.
