Abstract:BACKGROUND:The existing chemo/radiotherapy fail to eliminate cancer cells due to the restriction of either drug resistance or radio tolerance. The predicament urges researchers to continuously explore alternative strategy for achieving a potent curative effect.METHODS:Functional chlorin gold nanorods (Ce6-AuNR@SiO2-d-CPP) were fabricated aiming at treating breast cancer by photothermal/photodynamic therapy (PTT/PDT). The nanostructure was developed by synthesizing Au nanorods as the photothermal conversion material, and by coating the pegylated mesoporous SiO2 as the shell for entrapping photosensitizer Ce6 and for linking the D-type cell penetrating peptide (d-CPP). The function of Ce6-AuNR@SiO2-d-CPP was verified on human breast cancer MCF-7 cells and MCF-7 cells xenografts in nude mice.RESULTS:Under combinational treatment of PTT and PDT, Ce6-AuNR@SiO2-d-CPP demonstrated a strong cytotoxicity and apoptosis inducing effects in breast cancer cells in vitro, and a robust treatment efficacy in breast cancer-bearing nude mice. The uptake mechanism involved the energy-consuming caveolin-mediated endocytosis, and Ce6-AuNR@SiO2-d-CPP in PTT/PDT mode could induce apoptosis by multiple pathways in breast cancer cells.CONCLUSION:Ce6-AuNR@SiO2-d-CPP demonstrated a robust efficacy in the treatment of breast cancer by photothermal/photodynamic therapy. Therefore, the present study could offer a new promising strategy to treat the refractory breast cancer.
New-generation photosensitizer-anchored gold nanorods for a single near-infrared light-triggered targeted photodynamic–photothermal therapy

Abstract:Traditional combined photodynamic and photothermal therapy (PDT/PTT) was limited in clinical treatment of cancer due to the exceptionally low drug delivery efficiency to tumor sites and the activation by laser excitation with different wavelengths. We have accidentally discovered that our synthesized chlorin e6-C-15-ethyl ester (HB, a new type of photosensitizer) be activated by a laser with an excitation wavelength of 660 nm. Herein, we utilized Au nanorods (AuNRs) as 660 nm-activated PTT carriers to be successively surface-functionalized with HB and tumor-targeting peptide cyclic RGD (cRGD) to develop HB-AuNRs@cRGD for single NIR laser-induced targeted PDT/PTT. The HB-AuNRs@cRGD could be preferentially accumulated within tumor sites and rapidly internalized by cancer cells. Thereby, the HB-AuNRs@cRGD could exhibit amplified therapeutic effects by producing both significant reactive oxygen species (ROS) and hyperthermia simultaneously under the guidance of fluorescence imaging. The tumor inhibition rate on ECA109 esophageal cancer model was approximately 77.04%, and the negligible systematic toxicity was observed. This study proposed that HB-AuNRs@cRGD might be a promising strategy for single NIR laser-induced and imaging-guided targeted bimodal phototherapy.

Abstract:Photothermal therapy (PTT) has attracted increasing attention as an effective anticancer strategy, which has minimal side effects and could suppress the tumor growth effectively. Herein, poly (ethylene glycol)-stabilized polydopamine-modified gold nanostars (AuNSs@PDA-PEG) nanoparticles (NPs) were fabricated for PTT of cancer. The AuNSs@PDA-PEG NPs exhibited strong near-infrared (NIR) absorbance and excellent photostability for highly efficient PTT. Cytotoxicity assay indicated that the NPs possess high biocompatibility. Moreover, the AuNSs@PDA-PEG NPs exhibited excellent photothermal killing performance in vitro, in which the cell viability decreased to 10.02% under the 808 nm laser irradiation. The apoptosis assay indicated that HeLa cells incubated with AuNSs@PDA-PEG NPs can be seriously destroyed by the NIR laser irradiation. The mechanisms of NIR PTT-induced cancer cell death involved mitochondrial dysfunction, increase in lysosomal membrane permeability, and autophagy. Western blot analysis confirmed AuNSs@PDA-PEG NPs-induced decrease in Bcl-2 expression and elevation of Bax expression. Thus, the biocompatible AuNSs@PDA-PEG NPs exhibit great potential in PTT of cancer.

Abstract:Gold nanorods (AuNRs), because of their strong absorption of near-infrared (NIR) light, are very suitable for in vivo photothermal therapy of cancer. However, appropriate surface modification must be performed on AuNRs before their in vivo application because of the high toxicity of their original stabilizer cetyltrimethylammonium bromide. Multidentate ligands have attracted a lot of attention for modification of inorganic nanoparticles (NPs) because of their high ligand affinity and multifunctionality, while the therapeutic effect of multidentate ligands modified NPs in vivo remains unexplored. Here, we modified AuNRs with a polythiol PEG-based copolymer. The multidentate PEG coated AuNRs (AuNR-PTPEGm950) showed good stabilities in high saline condition and wide pH range. And they had much stronger resistance to ligand competition of dithiothreitol (DTT) than AuNRs coated by monothiol-anchored PEG. The AuNR-PTPEGm950 had very low cytotoxicity and showed high efficacy for the ablation of cancer cells in vitro. Moreover, the AuNR-PTPEGm950 showed good stability in serum, and they had a long circulation time in blood that led to a high accumulation in tumors after intravenous injection. In vivo photothermal therapy showed that tumors were completely cured without reoccurrence by one-time irradiation of NIR laser after a single injection of these multidentate PEG modified AuNRs.

Abstract:The combination therapy of photothermal therapy and chemotherapy as a promising strategy has drawn extensive attention by overcoming the limitations of conventional treatments in tumor therapy. Gold nanorods-based nanoplatforms were herein designed by integrating doxorubicin (DOX)- and polydopamine-coated gold nanorods (GNRs@PDA) for tumor metastasis inhibition and multifunctional drug delivery. The GNRs@PDA poly(ethylene glycol) (PEG) DOX nanocomplex showed robust stability and excellent near-infrared (NIR) photothermal conversion efficiency under laser irradiation. The release of loaded DOX from GNRs@PDA PEG DOX nanocomposites was improved in tumor microenvironments. Furthermore, the PDA-functionalized GNR nanocomposites were expected to be potential photoacoustic imaging agents for imaging-guided tumor therapy. Upon NIR laser irradiation, the efficiency of tumor inhibition of GNRs@PDA PEG DOX is greater than that of the other group in vitro and in vivo, which was confirmed by immunohistochemistry staining, demonstrating a promising strategy for suppression of tumor metastasis and low long-term systemic toxicity. These results illustrated a promising strategy of tailor-made GNRs@PDA PEG DOX nanoplatforms for ablation of tumor and suppression of tumor metastasis in clinical application.

Abstract:Photothermal/photodynamic therapies (PTT/PDT) have been widely accepted as non-invasive therapeutic modalities to erase tumours. However, both therapies face the problem of precisely locating tumours and reducing their side effects. Herein, chlorin e6 conjugated gold nanorod, (Ce6-PEG-AuNR), a type of gold nanorod-photosensitizer conjugate, is designed as a kind of nano-therapeutic agent to simultaneously realize combined PTT/PDT. Compared to free Ce6, the fluorescence of Ce6 adhered to the conjugate is effectively quenched by the longitudinal surface plasmon resonance (LSPR) of in the Ce6-PEG-AuNR. However, the specific fluorescence of Ce6 can be recovered in tumour tissue when Ce6 is separated from the conjugate owing to the cleavage of hydrazone bond between Ce6 and PEG caused by intracellular acidic conditions in tumour tissue. Based on this effect, we can precisely locate tumours and further kill cancer cells by combined PTT/PDT. In addition, the combined therapy (PTT/PDT) function is more efficient in cancer treatment than that of PTT or PDT alone. Therefore, Ce6-PEG-AuNR can serve as a promising dual-modal phototherapeutic agent as well as a tumour-sensitive fluorescent probe to diagnose and treat cancer.

Abstract:Au nanorods (AuNRs) have attracted considerable interest as drug delivery systems because of their enhanced cell internalization and stronger drug‐loading ability. In addition, the incorporation of photodynamic therapy (PDT) and photothermal therapy (PTT) into one nanosystem presents great promise to defect multiple drawbacks in cancer therapy. Herein, we fabricated a multifunctional and dual‐targeting nanoplatform based on hyaluronic acid‐grafted‐(mPEG/triethylenetetramine‐conjugated‐lipoic acid/tetra(4‐carboxyphenyl)porphyrin/folic acid) polymer ligand capped AuNRs (AuNRs@HA‐g‐(mPEG/Teta‐co‐(LA/TCPP/FA)) for combined photodynamic–photothermal therapy of cancer. The prepared nanoparticles displayed high TCPP loading capacity and excellent stability in different biological media. Furthermore, AuNRs@HA‐g‐(mPEG/Teta‐co‐(LA/TCPP/FA)) not only could produce a localized hyperthermia to conduct PTT, but also generate cytotoxic singlet oxygen (1O2) to perform PDT under laser irradiation. Confocal imaging results disclosed that this nanoparticle endowing the specific function of polymeric ligand could enhance cellular uptake, accelerate endo/lysosomal escape, as well as produce higher reactive oxygen species. Importantly, this combination therapy strategy could also induce higher anticancer potential than PDT or PTT only against MCF‐7 tumor cells in vitro. Therefore, this work presented an AuNRs‐based therapeutic nanoplatform with great potential in dual‐targeting and photo‐induced combination therapy of cancer.

Abstract:In terms of synergistic cancer therapy, biological nanomaterials with a second near-infrared (NIR-II) window response can greatly increase photothermal effects and photoacoustic imaging performance. Herein, we report a novel stimuli-responsive multifunctional drug-loading system which was constructed by integrating miniature gold nanorods (GNR) as the NIR-II photothermal nanorods and cyclic ternary aptamer (CTA) composition as a carrier for chemotherapy drugs. In this system, doxorubicin hydrochloride (DOX, a chemotherapy drug) binds to the G-C base pairs of the CTA, which exhibited a controlled release behavior based on the instability of G-C base pairs in the slightly acidic tumor microenvironment. Upon the 1064 nm (NIR-II biowindow) laser irradiation, the strong photothermal and promoted cargo release properties endow gold nanorods@CTA (GNR@CTA) nanoparticles displaying excellent synergistic anti-cancer effect. Moreover, the GNR@CTA of NIR also possesses thermal imaging and photoacoustic (PA) imaging properties due to the strong NIR region absorbance. This work enables to obtaining a stimuli-responsive "all-in-one" nanocarrier, which are promising candidate for bimodal imaging diagnosis and chemo-photothermal synergistic therapy.

Abstract:Gold nanorods (GNRs) with longitudinal surface plasmon resonance (LSPR) peaks in second near-infrared (NIR-II) window have attracted a great amount of attention as photothermal transducer because of their inherently excellent photothermal transition efficiency, high biocompatibility and versatile surface functionalization. One key question for the application of these GNRs against tumors in vivo is which size/shape and surface ligand conjugation are promising for circulation and tumor targeting. In this study, we prepared a series of gold nanorods (GNRs) of similar aspect ratio and LSPR peaks, and thus similar photothermal transfer efficiency under irradiation of 980 nm laser, but with tunable size in width and length. The obtained GNRs were subjected to surface modification with PEG and tumor targeting ligand lactoferrin. With these tailor-designed GNRs in hand, we have the chance to study the impact of dimension and surface property of the GNRs on their internalization via tumor cells, photothermal cytotoxicity in vitro, blood circulation and tissue distribution pattern in vivo. As a result, the GNRs with medium size (70 nm in length and 11.5 nm in width) and surface PEG/LF modification (GNR70@PEG-LF) exhibit the fastest cell internalization via HepG2 cells and best photothermal outcome in vitro. The GNR70@PEG-LF also display long circulation time and the highest tumor accumulation in vivo, due to the synergetic effect of surface coating and dimension. Finally, tumor ablation ability of the GNRs under irradiation of 980 nm light were validated on mice xenograft model, suggesting their potential photothermal therapy against cancer in NIR-II window.

Abstract:Based on characteristics of the tumor microenvironment (TME), including acidity, oxygen‐depleted, inflammation and hydrogen peroxide overload, combined with emerging nanotechnologies, designing nanoplatforms with TME specificity/ responsiveness for tumor treatment is a promising nanotherapeutic strategy. In this work, a multifunctional gold‐palladium bimetallic cascade nanozyme was constructed for effective photothermal‐enhanced cascade catalyzed synergistic therapy of tumors. The dumbbell‐like Au‐Pd bimetallic nanomaterials (Au NRs‐Pd@HA) were obtained by reducing palladium on gold nanorods with ascorbic acid (AA) and further modified with hyaluronic acid (HA). The introduction of HA brings biocompatibility and targeting properties. The zebrafish embryos model showed that Au NRs‐Pd@HA had good biocompatibility and low biotoxicity. Au NRs‐Pd@HA can induce catalytic conversion of glucose to generate H2O2 efficiently, and subsequently undergoes cascade reaction to produce abundant ·OH radicals, exhibiting peroxidase‐like (POD‐like) and glucose oxidase‐like (GOD‐like) capabilities. The generated ·OH was a key factor for tumor ablation. Meanwhile, Au NRs‐Pd@HA exhibits good photothermal performance under 808 nm irradiation, in favor of photothermal therapy (PTT). Especially, the POD‐like and GOD‐like activities were significantly enhanced due to the photothermal effect. Abstract:Cancer is still one of the major health issues faced by human beings today. Various nanomaterials have been designed to treat tumors and have made great progress. Herein, we used amino-functionalized metal organic framework (UiO-66-NH2) as superior templates and successfully synthesized the UiO-66-NH2@Aushell composite nanoparticles (UA) with high loading capacity and excellent photothermal properties through a simple and gentle method. In addition, due to the rich pore structure and excellent biocompatibility of the as-prepared composite nanoparticles, the hydrophobic NO donor BNN6 (N,N′-Di-sec-butyl-N,N′-dinitroso-1, 4-phenylenediamine) molecule was efficiently delivered. Based on the phenomenon where BNN6 molecules can decompose and release NO at high temperature, when UiO-66-NH2@Aushell-BNN6 composite nanoparticles (UA-BNN6) entered tumor cells and were irradiated by NIR, the porous gold nanoshells on the surface of composite nanoparticles induced an increase in temperature through the photothermal conversion process and promoted the decomposition of BNN6 molecules, releasing high concentration of NO, thus efficiently killing HeLa cells through the synergistic effect of NO-photothermal therapy. This effective, precise and safe treatment strategy controlled by NIR laser irradiation represents a promising alternative in the field of cancer treatment.

Abstract:Gold nanorods (GNRs) are well known in photothermal therapy based on near-infrared (NIR) laser absorption of the longitudinal plasmon band. Herein, we developed an effective stimulus system -- GNRs and doxorubicin co-loaded polymersomes (P-GNRs-DOX) -- to facilitate co-therapy of photothermal and chemotherapy. DOX can be triggered to release once the polymersomes are corrupted under local hyperthermic condition of GNRs induced by NIR laser irradiation. Also, the cytotoxicity of GNRs caused by the residual cetyltrimethylacmmonium bromide (CTAB) was reduced by shielding the polymersomes. The GNRs-loaded polymersomes (P-GNRs) can be efficiently taken up by the tumor cells. The distribution of the nanomaterial was imaged by IR-820 and quantitatively analyzed by ICP-AES. We studied the ablation of tumor cells in vitro and in vivo, and found that co-therapy offers significantly improved therapeutic efficacy (tumors were eliminated without regrowth.) compared with chemotherapy or photothermal therapy alone. By TUNEL immunofluorescent staining of tumors after NIR laser irradiation, we found that the co-therapy showed more apoptotic tumor cells than the other groups. Furthermore, the toxicity study by pathologic examination of the heart tissues demonstrated a lower systematic toxicity of P-GNRs-DOX than free DOX. Thus, the chemo-photothermal treatment based on polymersomes loaded with DOX and GNRs is a useful strategy for maximizing the therapeutic efficacy and minimizing the dosage-related side effects in the treatment of solid tumors.
Pt/Ag‐PEG‐Ce6 Nanosystem with Enhanced Near‐Infrared Absorption and Peroxidase‐Like Activity for Synergistic Photodynamic/Photothermal Therapy

Abstract:Noble metals and their alloy‐based nanomaterials are widely used in biomedicine, especially in the fields of photodynamic therapy (PDT) and photothermal therapy (PTT) on tumors, due to their remarkable physicochemical properties. Nonetheless, novel doping strategies with unexpected efficacies are still challenging and formidable. Herein, a photosensitizer‐alloy nanosystem is designed for highly efficient PDT and PTT. Specifically, the optimized Pt/Ag alloy nanoparticles are facilely synthesized in one step at room temperature and are simultaneously endowed with enhanced peroxidase‐like activity and photothermal conversion, and further activated cytotoxic singlet oxygen (1O2) productivity after covalent functionalization of chlorin e6 (Ce6). A small amount of Ag doping can kill two birds with one stone by simultaneously enhancing the synergistic PDT and PTT potential of Pt/Ag‐PEG‐Ce6 nanoparticles. Both in vitro and in vivo experimental results indicate that the significant temperature increase triggered by 808 nm laser and a large amount of toxic 1O2 excited by 671 nm laser synergistically and strongly kill tumor cells. Therefore, through proper doping and improved physicochemical properties, this nanosystem achieves effective PDT and PTT of tumors and also inspires the advanced design of noble metal‐based nanomedicine. A novel photosensitizer‐alloy nanotherapeutic reagent for efficient photodynamic therapy (PDT) and photothermal therapy (PTT), Pt/Ag‐PEG‐Ce6, has been successfully fabricated and is applied to effectively kill tumor cells in vivo and in vitro, not only remarkably improving the PDT and PTT therapeutic effects, but also broadening the biomedical applications of noble metal‐based nanosystems and their alloy.
Gold Nanostar@Polyaniline Theranostic Agent with High Photothermal Conversion Efficiency for Photoacoustic Imaging-Guided Anticancer Phototherapy at a Low Dosage.

Abstract:Due to the strong and tunable photothermal effect, metallic nanoparticles are of enormous interest in light-activated biomedical applications, such as photoacoustic imaging (PAI) and photothermal therapy (PTT). However, the photothermal conversion efficiency (PCE) of existing metallic photothermal agents is still unsatisfactory. Herein, we develop an efficient photothermal theranostic agent based on a gold nanostar@polyaniline core-shell nanocomposite with high PCE for PAI-guided PTT at a low dosage. After optimizing the relative composition of polyaniline (PANI) and gold nanostars (AuNSs), this nanocomposite eventually empowers an outstanding PCE of up to 78.6%, which is much better than AuNSs or PANI alone and most of the existing photothermal theranostic agents. Besides, the nanocomposite can act as a targeted probe for tumors by hyaluronic acid (HA) modification without compromising the photothermal performance. The obtained nanoprobes named AuNSPHs exhibit promising biocompatibility and great performance of PAI-guided PTT to treat triple-negative breast cancer both in vitro and in vivo. More importantly, a single injection of AuNSPHs significantly suppresses tumor growth with a low dosage of Au (0.095 mg/kg), which is attributed to the high PCE of AuNSPHs. Taking advantage of the exhilarating photothermal conversion ability, this theranostic agent can safely potentiate the antitumor therapeutic efficacy of laser-induced ablation and holds great potential for future medical applications.

Abstract:Abstract Owing to strong and tunable surface plasmon resonance (SPR) effect and good biocompatibility, gold nanoparticles have been suggested to be a versatile platform for a broad range of biomedical applications. In this study, a new nanoplatform of thermo-responsive polymer encapsulated gold nanorods incorporating indocyanine green (ICG) was designed to couple the photothermal properties of gold nanorods (AuNRs) and the photodynamic properties of ICG to enhance the photodynamic/photothermal combination therapy (PDT/PTT). In addition to the significantly increased payload and enhancing photostability of ICG, the polymer shell in the nanoplatform also has thermo-responsive characteristics that can control the release of drugs at tumour sites upon the laser irradiation. On the basis of these improvements, the nanoplatform strongly increased drug aggregation at the tumour site and improved the photothermal/photodynamic therapeutic efficacy. These results suggest that this nanoplatform would be a great potential system for tumour imaging and antitumour therapy.

Abstract:Synergetic photothermal/photodynamic/chemotherapy receives significant attention for precise in vivo cancer treatment. Despite plenty of encouraging photosensitizers explored, integrated nanoagents with multiple functions are still highly desired. In this study, novel nanocomposites coupling black phosphorus (BP) nanosheets, gold nanorods (AuNRs), carbon nanodots (CDs), and doxorubicin (Dox) are prepared. The nanoagents exhibit high antitumor activity on account of their broad light absorption, excellent catalytic ability, and significant photothermal and photodynamic effects. CDs not only emit bright fluorescence for accurate diagnosis and guiding of tumor treatment but also catalyze the generation of ROS for photodynamic therapy (PDT). The released Dox induces apoptosis of cells and increases the levels of H2O2 to promote PDT. AuNRs are the main photothermal therapy (PTT) material that converts light into heat. Moreover, BP can be used to enhance both PTT and PDT efficiencies, and the two therapy modes can be cooperatively reinforced. It is also found that the local immune microenvironment of the tumors is activated. The strategy makes good use of the features of each component. Satisfactory antitumor phenomena are well confirmed by in vitro and in vivo results. This study provides new insights into enhanced synergetic therapy, highlighting the great utility of BP-based nanoagents in the field of nanomedicine.

Abstract:Photothermal therapy (PTT) employs photosensitizing agents, which are taken up by cells and generate heat when irradiated with near-infrared (NIR) light, to enable the photoablation of cancer cells. High absorption in the NIR region is crucial for a photosensitizing agent to achieve efficient PTT. Different combinations between gold nanoparticles and fluorescent agents always influence their spectrum properties. Herein, we fabricated a novel combination of a fluorescent agent (doxorubicin, DOX, also a popular chemotherapeutic agent) with gold nanospheres by synthesizing hybridized DOX-Au nanospheres (DAuNS), where a part of the DOX molecules and Au co-formed a hybridized matrix as the shell and the remaining DOX molecules precipitated as the core. The unique structure of DAuNS induced interesting changes in the characteristics including spectrum properties, morphology, drug loading and antitumor activity. We observed that DAuNS exhibited a significantly enhanced surface plasmon absorption in the NIR region, inducing a more efficient photothermal conversion and stronger tumor-cell killing ability under NIR laser irradiation. In addition, our study presents a new and simple platform to load a drug into nanoparticles. DAuNS could be a promising nanoparticle with the "two punch" efficacy of PTT and chemotherapy and could be used in clinical applications due to its controllable synthesis, small size, and narrow size distribution.

Abstract:Real-time, high-quality, 3D scanning of large-scale scenes is key to mixed reality and robotic applications. However, scalability brings challenges of drift in pose estimation, introducing significant errors in the accumulated model. Approaches often require hours of offline processing to globally correct model errors. Recent online methods demonstrate compelling results but suffer from (1) needing minutes to perform online correction, preventing true real-time use; (2) brittle frame-to-frame (or frame-to-model) pose estimation, resulting in many tracking failures; or (3) supporting only unstructured point-based representations, which limit scan quality and applicability. We systematically address these issues with a novel, real-time, end-to-end reconstruction framework. At its core is a robust pose estimation strategy, optimizing per frame for a global set of camera poses by considering the complete history of RGB-D input with an efficient hierarchical approach. We remove the heavy reliance on temporal tracking and continually localize to the globally optimized frames instead. We contribute a parallelizable optimization framework, which employs correspondences based on sparse features and dense geometric and photometric matching. Our approach estimates globally optimized (i.e., bundle adjusted) poses in real time, supports robust tracking with recovery from gross tracking failures (i.e., relocalization), and re-estimates the 3D model in real time to ensure global consistency, all within a single framework. Our approach outperforms state-of-the-art online systems with quality on par to offline methods, but with unprecedented speed and scan completeness. Our framework leads to a comprehensive online scanning solution for large indoor environments, enabling ease of use and high-quality results.1
