Abstract:Botryosphaeria dothidea causes apple ring rot, which is among the most prevalent postharvest diseases of apples and causes significant economic loss during storage. In this study, we investigated the biocontrol activity and possible mechanism of Bacillus velezensis strain P2-1 isolated from apple branches against B. dothidea in postharvest apple fruit. The results showed strain P2-1, one of the 80 different endophytic bacterial strains from apple branches, exhibited strong inhibitory effects against B. dothidea growth and resulted in hyphal deformity. B. velezensis P2-1 treatment significantly reduced the ring rot caused by B. dothidea. Additionally, the supernatant of strain P2-1 exhibited antifungal activity against B. dothidea. Re-isolation assay indicated the capability of strain P2-1 to colonize and survive in apple fruit. PCR and qRT-PCR assays revealed that strain P2-1 harbored the gene clusters required for biosynthesis of antifungal lipopeptides and polyketides. Strain P2-1 treatment significantly enhanced the expression levels of pathogenesis-related genes (MdPR1 and MdPR5) but did not significantly affect apple fruit qualities (measured in fruit firmness, titratable acid, ascorbic acid, and soluble sugar). Thus, our results suggest that B. velezensis strain P2-1 is a biocontrol agent against B. dothidea-induced apple postharvest decay. It acts partially by inhibiting mycelial growth of B. dothidea, secreting antifungal substances, and inducing apple defense responses.

Abstract:Human exposure to heavy metals is a global public health problem. Heavy metals which cause neurological toxicity, such as lead (Pb2+) and manganese (Mn), are of particular concern due to the long-lasting and possibly irreversible nature of their effects. Pb2+ exposure in childhood can result in cognitive and behavioral deficits in children. These effects are long-lasting and persist into adulthood even after Pb2+ exposure has been reduced or eliminated. While Mn is an essential element of the human diet and serves many cellular functions in the human body, elevated Mn levels can result in a Parkinson’s disease (PD)-like syndrome and developmental Mn exposure can adversely affect childhood neurological development. Due to the ubiquitous presence of both metals, reducing human exposure to toxic levels of Mn and Pb2+ remains a world-wide public health challenge. In this review we summarize the toxicokinetics of Pb2+ and Mn, describe their neurotoxic mechanisms, and discuss common themes in heavy metal toxicology.
Biocontrol potential of Trichoderma harzianum CGMCC20739 (Tha739) against postharvest bitter rot of apples

Abstract:This study investigated the biocontrol ability of Trichoderma harzianum CGMCC20739 (Tha739) against apple bitter rot caused by Colletotrichum gloeosporioides. In vitro tests, Tha739 inhibited the mycelial growth of C. gloeosporioides. Microscopic observation showed that Tha739 grew in parallel with, coiled around, and deformed the hyphae of C. gloeosporioides. Tha739-derived metabolites decreased the conidia production of C. gloeosporioides. In vivo tests, the lesion diameters of wounded apples treated with Tha739 1 h before C. gloeosporioides were lower than those of wounded apples treated with Tha739 after pathogen inoculation. In addition, compared with the apples inoculated with C. gloeosporioides only, the disease index of unwounded apples inoculated with Tha739 and C. gloeosporioides decreased by 2.17-fold. Furthermore, compared with the control, the total soluble solid contents of apples treated with Tha739 were 9.02 % and 1.54 % higher at 1 and 3 d, respectively. The titratable acidity contents of apples treated with Tha739 were 10.02 % and 14.58 % higher than those in the control at 1 and 3 d after treatment, respectively. The soluble sugar content and weight loss in Tha739 treatment group and control were not significantly different. The results showed that Tha739 could control apple bitter rot and maintain the nutritional quality of the fruit. Thus, T. harzianum Tha739 is a potentially biocontrol agent for harvested apples.

Abstract:Apple white rot, caused by Botryosphaeria dothidea , is one of the important diseases in Korea. B. dothidea can cause pre- and postharvest decay on apple fruit as well as canker and dieback of apple trees. In this study, we isolated bacteria from the trunk of apple trees and tested their antagonistic activity against B. dothidea . Five bacterial isolates (23-168, 23-169, 23-170, 23-172, and 23-173) were selected that were most effective at inhibiting the mycelial growth of the pathogens. The isolate 23-172 was identified as Bacillus amyloliquefaciens and four isolates 23-168, 23-169, 23-170, and 23-173 were identified as Bacillus velezensis by RNA polymerase beta subunit ( rpoB ) and DNA gyraseA subunit ( gyrA ) gene sequencing. All isolates showed strong antagonistic activity against B. dothidea as well as Colletotrichum fructicola and Diaporthe eres . All isolates exhibited cellulolytic, proteolytic and phosphate solubilizing activities. In particular, two isolates 23-168, 23-169 were shown to significantly reduce the size of white rot lesions in pretreated apple fruits. These results will provide the basis for the development of a fungicide alternative for the control of white rot of apple.
Biocontrol efficiency of Meyerozyma guilliermondii Y-1 against apple postharvest decay caused by Botryosphaeria dothidea and the possible mechanisms of action

Abstract:Apple ring rot, caused by Botryosphaeria dothidea, is one of the important diseases in China. This pathogen infects branches and fruit and also results in fruit decay during storage. Biocontrol agents have been proposed to reduce apple decays during storage and are considered as a promising alternative strategy to traditional chemical treatment. In this study, Meyerozyma guilliermondii Y-1, isolated from healthy grape fruit, was firstly evaluated for its biocontrol efficiency against B. dothidea in postharvest apple fruit, and the possible mechanisms were investigated. The results revealed that M. guilliermondii Y-1 treatment effectively reduced apple ring rot caused by B. dothidea in vivo. The disease incidence and lesion diameter were reduced by 32.22% and 57.51% compared with those of control fruit. Furthermore, the use of filtrate and autoclaved culture of M. guilliermondii Y-1 also showed a certain degree of control efficiency against fruit ring rot. M. guilliermondii Y-1 significantly inhibited the mycelial growth and spore generation of B. dothidea in vitro and exhibited an obvious ability to colonize in apple fruit wounds and surface at 25 °C or 4 °C. In addition, M. guilliermondii Y-1 treatment significantly enhanced the activities of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), phenylalanine ammonialyase (PAL), and polyphenoloxidase (PPO), promoted the total phenolics content, and alleviated lipid peroxidation in apple fruit. As expected, we found that the expression of four pathogenesis-related proteins genes (MdPR1, MdPR5, MdGLU, and MdCHI) was remarkably increased by M. guilliermondii Y-1 treatment. Our data together suggest that M. guilliermondii Y-1 is a potential biocontrol agent against B. dothidea postharvest infection in apple fruit, partially through inhibiting mycelial growth and spore germination of B. dothidea, competing for space and nutrient with pathogen, and inducing resistance in apple fruit by stimulating a series of defense responses.
Antifungal Activity and Possible Mechanism of Bacillus amyloliquefaciens FX2 Against the Postharvest Apple Ring Rot Pathogen

Abstract:Botryosphaeria dothidea-induced apple ring rot is one of the most serious postharvest diseases in apple production. In our preliminary work, we isolated a bacterial strain (FX2) from an infested apple orchard. Here, we confirmed the strong antifungal activity of FX2 on B. dothidea. Through phylogenetic analysis and morphological observations, we identified FX2 as a Bacillus amyloliquefaciens strain. We also found that 10% cell-free supernatant (CFS) of FX2 significantly affected mycelial growth and morphology and almost completely inhibited spore germination and germ tube elongation in B. dothidea. Furthermore, 10% CFS damaged the cell ultrastructure, resulting in a remarkable increase in cellular leakage in B. dothidea mycelia. Thus, CFS has the potential to effectively reduce in vivo B. dothidea infection, reduced lesion diameters to 64.7% compared with the control group, and reduced disease incidence by 15%. Finally, ultrafiltration, desalting chromatography, and anion exchange chromatography showed that the antifungal constituents in CFS are composed mainly of antifungal proteins. We further characterized these potential antifungal proteins via liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis. Herein, we provide novel insights into the antifungal mechanisms of B. amyloliquefaciens FX2, and we highlight its potential as a novel biocontrol agent for controlling postharvest apple ring rot.
Identification and characterization of the Bacillus atrophaeus strain J-1 as biological agent of apple ring rot disease

Abstract:Bacillus species are promising agents for the biological control of postharvest diseases. Based on the morphological, physiological and biochemical characteristics and 16S rRNA gene sequence analysis, the bacterial strain J-1 was identified as Bacillus atrophaeus. The B. atrophaeus strain J-1 showed broad spectrum antifungal activity against various plant pathogenic fungi. In assays in vitro the metabolites contained in the B. atrophaeus strain J-1 cell-free filtrate had a strong inhibitory effect on Botryosphaeria dothidea. The cell-free filtrate inhibited the growth of B. dothidea in vivo and reduced the disease incidence. Furthermore, PCR detection showed that the strain J-1 had lipopeptide antibiotic biosynthesis genes: fengycin, iturin A and surfactin. The lipopeptide crude extractions from the strain J-1 cell-free filtrate significantly suppressed the growth of B. dothidea. C14 iturin A was isolated by bioactivity-guided fractionation and identified by liquid chromatography–electronic spray ionization–tandem mass spectrometry analysis. This paper is the first to report that B. atrophaeus J-1 efficiently controls apple ring rot disease caused by B. dothidea. Using the B. atrophaeus strain J-1 may provide an alternative control method for the prevention apple ring rot disease.

Abstract:The biocontrol activity of Rhodotorula glutinis on gray mold decay and blue mold decay of apple caused by Botrytis cinerea and Penicillium expansum, respectively, was investigated, as well as its effects on postharvest quality of apple fruits. The results show there was a significant negative correlation between concentrations of the yeast cells and the disease incidence of the pathogens. The higher concentration of the R. glutinis, the better effect of the biocontrol capacity. At concentrations of R. glutinis 1×108CFUml−1, the amount of gray mold decay was completely inhibited after 5 days incubation at 20°C, after challenge with B. cinerea spores suspension of 1×105sporesml−1; While the blue mold decay was completely inhibited at concentrations of 5×108CFUml−1, at challenged with P. expansum spores suspension of 5×104sporesml−1. These results demonstrated that the efficacy of R. glutinis in controlling of gray mold decay of apples was better than the efficacy of controlling blue mold. R. glutinis within inoculated wounds on apples increased in numbers at 20°C from an initial level of 9.5×105CFU per wound to 2.24×107CFU at 20°C after 1 day. The highest population of the yeast was recovered 4 days after inoculation, the yeast population in wounds increased by 56.9 times. After that, the population of the yeast began to decline very slowly. R. glutinis significantly reduced the incidence of natural infections on intact fruit from 75% in the control fruit to 28.3% after 5 days at 20°C, and from 58.3 to 6.7% after 30 days at 4°C followed by 4 days at 20°C. R. glutinis treatment had no deleterious effect on quality parameters after 5 days at 20°C or after 30 days at 4°C followed by 4 days at 20°C.

Abstract:The efficacy of Rhodotorula mucilaginosa against postharvest gray mold, blue mold and natural decay development of apples and the possible mechanisms involved were investigated. The decay incidence and lesion diameter of gray mold and blue mold of apples treated by R. mucilaginosa were significantly reduced compared with the control fruits, and the higher concentration of R. mucilaginosa, the better the efficacy of the biocontrol. R. mucilaginosa also significantly reduced the natural decay development of apples following storage at 20°C for 35 days or at 4°C for 45 days followed by 20°C for 15 days. Germination and survival of spores of Penicillium expansum and Botrytis cinerea were markedly inhibited by R. mucilaginosa in an in vitro test. Rapid colonization of the yeast in apple wounds was observed whether stored at 20°C or 4°C. In apples, the activities of peroxidase (POD) and polyphenoloxidase (PPO) were significantly induced and lipid peroxidation (malondialdehyde (MDA) content) was highly inhibited by R. mucilaginosa treatment compared with those of the control fruits. All these results indicated that R. mucilaginosa has great potential for development of commercial formulations to control postharvest pathogens on fruits. Its modes of action were based on competition for space and nutrients with pathogens, inducement of activities of defense-related enzymes such as POD, PPO and inhibition of lipid peroxidation (MDA content) of apples, so as to enhance the resistance and delay the ripening and senescence of apples.

Highlighting the complexity of pathogenesis: the host microbiota impacts disease development on apple fruit and is a cornerstone for its biocontrol

Abstract:Apple fruit is the most produced temperate fruit with a trade value estimated at 7.5 billion $ and is usually stored up to one year after harvest. Postharvest pathogens often compromise storage, responsible for up to 55% of fruit losses, depending on the country and fruit. They are also a source of mycotoxin contamination. A sustainable way to control that pathogen is using beneficial microorganisms called biocontrol agents (BCA). Despite promising efficacy in laboratory conditions, BCA's efficacy is variable and often reduced once applied at a large scale through either orchard or postharvest treatment. We hypothesized that the epiphytic microbiota plays a role in the variability of BCA efficiency (Pichia anomala, strain K) and postharvest disease development due to Botrytis cinerea on apples. A diverse set of 18 epiphytic microbial communities were harvested from apple carposphere and bio-banked. The analysis of their bacterial and fungal taxonomic composition and carbon metabolic footprint confirmed that contrasted microbiotas were harvested. Their impact on B. cinerea disease development was evaluated through a standardized in vivo bioassay. The reduction of B. cinerea rot development ranged from 20% to 80% when the microbiotas were applied alone. In addition, three microbiotas enhanced the biological control efficiency of strain K (up to +100%, whatever the tested microbiota concentrations) while others limited its action (down to -27%). A co-clustering analysis of biocontrol efficacy with carbon profiles or taxonomic composition was carried out. It identified promising molecules whose high metabolization was associated with high biocontrol by the microbiota and taxa with higher abundance in microbiota limiting B. cinerea rot development. Putative beneficial taxa were isolated from the most efficient microbiota. In vivo bioassays confirmed the efficacy of two molecules and two strains belonging to species never mentioned for their biocontrol properties against plant disease. This study, the first of its kind, demonstrates that natural epiphytic microbiota significantly influences postharvest disease development in apples and the variability in biocontrol efficacy. Our approach identified promising molecules and taxa that enhance biocontrol, offering new insights for sustainable postharvest pathogen management.

Effectiveness of a Novel Endophyte Bacillus velezensis Strain B1 in the Biocontrol of Pear Postharvest Ring Rot

Abstract:Pear ring rot caused by Botryosphaeria dothidea severely affects the quality and storage life of pear fruit. Plant endophytes are potential new sources of biocontrol agents due to their abilities to produce novel bioactive products. In this work, we focused on an endophytic strain B1, which was isolated from roots of Dendrobium huoshanense. Notably, strain B1 exhibited strongly inhibitory effects against the growth of Botryosphaeria dothidea with an inhibition rate of 73.2% after 1 week of co-cultivation. Furthermore, it displayed a broad-spectrum inhibitory activity. Strain B1 also effectively delayed the onset of ring rot caused by B. dothidea in pear fruit and reduced the lesion diameter by 73.4%. Strain B1 was identified as Bacillus velezensis based on core-genome phylogeny. Genome mining with antiSMASH revealed 13 potential gene clusters involved in antimicrobial metabolites. Three main groups of lipopeptides (surfactin, iturin and fengycin) were identified with MALDI-TOF-MS, and the lipopeptides in the inhibition zone were greatly upregulated by B. dothidea; especially, fengycin isoforms were detected with higher abundance. These results proved that lipopeptides produced by strain B1 had significant antagonistic effect on B. dothidea. In conclusion, our results suggest that strain B1 has potential applications in the biocontrol of B. dothidea in postharvest fruit.

Abstract:The use of microbial biocontrol agents for control of postharvest disease has been the subject of intensive research over the past three decades resulting in commercialization of several biocontrol products. The objective of this research was to test endospore-forming bacteria collected from apple leaves for suppression of bitter rot and blue mold on apple. Bacteria were collected from abandoned, low-input, organic, and conventionally managed orchards in Pennsylvania and were screened for their ability to produce endospores, hydrolyze chitin, reduce pathogen growth in vitro, and suppress postharvest disease in vivo. Several isolates reduced bitter rot lesion size on ‘Rome Beauty’ from 40–89% compared to untreated controls. Bacillus megaterium isolates, A3-6 and Ae-1, resulted in the greatest suppression of bitter rot lesion size. One isolate, A3-2, suppressed blue mold lesion size. Scanning electron microscopy of inoculated apple wounds suggests parasitism as a mode of action explains the suppression of bitter rot lesion size by isolate A3-6. Of the top seventeen isolates exhibiting biocontrol potential, 70% were collected from abandoned or unmanaged locations. This research demonstrates abandoned apple orchards can be a source of new biocontrol agents for control of postharvest diseases of apple.

Coherence and content of conflict-based narratives: associations to family risk and maladjustment.

Abstract:This study examined the role of structural and content characteristics of children's conflict-based narratives (coherence, positive and aggressive themes) in the association between early childhood family risk and children's internalizing and externalizing problems in a sample of 193 children (97 girls, 96 boys) aged 3 to 5 years (M = 3.85, SD = .48). Parents participated in an interview on family related risk factors; teachers and parents completed the Strengths and Difficulties Questionnaire; children completed conflict-based narratives based on the MacArthur Story Stem Battery (MSSB). We specifically investigated the mediating and moderating role of narrative coherence and content themes in the association between family risk and children's internalizing and externalizing problems. Children's narrative coherence was associated with better adjustment, and had a buffering effect on the negative relation between family risk on children's internalizing problems. Positive themes were negatively associated with externalizing problems. Telling narratives with many positive and negative themes buffered the negative association of family risk and teacher-reported externalizing problems. In sum, the findings suggest that in children, being able to tell coherent and enriched narratives may buffer the impact of family risk on their symptoms, and being able to produce positive themes rather than aggressive themes is associated with lower externalizing problems.
Endophytic Bacterium Serratia plymuthica From Chinese Leek Suppressed Apple Ring Rot on Postharvest Apple Fruit

Abstract:Apple ring rot caused by Botryosphaeria dothidea is an economically significant plant disease that spreads across the apple production areas in China. The pathogen infects apple fruits during the growing season and results in postharvest fruits rot during storage, which brings about a huge loss to plant growers. The study demonstrated that an endophytic bacterium Serratia plymuthica isolated from Chinese leek ( Allium tuberosum ) significantly suppressed the mycelial growth, severely damaging the typical morphology of B. dothidea , and exerted a high inhibition of 84.64% against apple ring rot on postharvest apple fruit. Furthermore, S. plymuthica significantly reduced the titratable acidity (TA) content, enhanced the soluble sugar (SS) content, vitamin C content, and SS/TA ratio, and maintained the firmness of the fruits. Furthermore, comparing the transcriptomes of the control and the S. plymuthica treated mycelia revealed that S. plymuthica significantly altered the expressions of genes related to membrane (GO:0016020), catalytic activity (GO:0003824), oxidation-reduction process (GO:0055114), and metabolism pathways, including tyrosine metabolism (ko00280), glycolysis/gluconeogenesis (ko00010), and glycerolipid metabolism (ko00561). The present study provided a possible way to control apple ring rot on postharvest fruit and a solid foundation for further exploring the underlying molecular mechanism.

Abstract:Apple replant disease (ARD), caused by Fusarium pathogens, is a formidable threat to the renewal of apple varieties in China, necessitating the development of effective and sustainable control strategies. In this study, the bacterial strain BA-4 was isolated from the rhizosphere soil of healthy apple trees in a replanted orchard, demonstrating a broad-spectrum antifungal activity against five crucial apple fungal pathogens. Based on its morphology, physiological and biochemical traits, utilization of carbon sources, and Gram stain, strain BA-4 was tentatively identified as Bacillus amyloliquefaciens . Phylogenetic analysis using 16S rDNA and gyrB genes conclusively identified BA-4 as B. amyloliquefaciens . In-depth investigations into B. amyloliquefaciens BA-4 revealed that the strain possesses the capacity to could secrete cell wall degrading enzymes (protease and cellulase), produce molecules analogous to indole-3-acetic acid (IAA) and siderophores, and solubilize phosphorus and potassium. The diverse attributes observed in B. amyloliquefaciens BA-4 underscore its potential as a versatile microorganism with multifaceted benefits for both plant well-being and soil fertility. The extracellular metabolites produced by BA-4 displayed a robust inhibitory effect on Fusarium hyphal growth and spore germination, inducing irregular swelling, atrophy, and abnormal branching of fungal hyphae. In greenhouse experiments, BA-4 markedly reduced the disease index of Fusarium -related ARD, exhibiting protective and therapeutic efficiencies exceeding 80% and 50%, respectively. Moreover, BA-4 demonstrated plant-promoting abilities on both bean and Malus robusta Rehd. (MR) seedlings, leading to increased plant height and primary root length. Field experiments further validated the biocontrol effectiveness of BA-4, demonstrating its ability to mitigate ARD symptoms in MR seedlings with a notable 33.34% reduction in mortality rate and improved biomass. Additionally, BA-4 demonstrates robust and stable colonization capabilities in apple rhizosphere soil, particularly within the 10-20 cm soil layer, which indicates that it has long-term effectiveness potential in field conditions. Overall, B. amyloliquefaciens BA-4 emerges as a promising biocontrol agent with broad-spectrum antagonistic capabilities, positive effects on plant growth, and strong colonization abilities for the sustainable management of ARD in apple cultivation.

Abstract:Apple ring rot caused by Botryosphaeria dothidea is an essential and prevalent disease in the apple orchard in China. Our previous study demonstrated that dimethyl trisulfide (DT) from Chinese leek ( Allium tuberosum ) significantly suppressed the mycelial growth of B. dothidea and inhibited the incidence of apple ring rot postharvest. However, the mechanism underlying the inhibitory role of DT against B. dothidea is not fully understood. Comparing the control and the DT-treated B. dothidea mycelial transcriptomes revealed that heat shock protein 20 (Hsp20) strongly responded to DT treatment. This study identified four Hsp20 genes throughout the B. dothidea genome (BdHsp20_1-4). Each BdHsp20 gene had a conserved ACD with a variable N-terminal region and a short C-terminal extension. The segmental duplication event has contributed to the expansion of the BdHsp20 gene family. Compared to the wild-type strain, the CRISPR/Cas9 gene-edited BdHsp20 mutant (ΔBdHsp20) decreased the mycelial growth by 55.95% and reduced the disease symptom in postharvest apple fruit by 96.34%. However, the BdHsp20 complemented strain (ΔBdHsp20_C) significantly restored the growth and pathogenicity, which suggested that the BdHsp20 gene was closely involved in the growth and pathogenicity of B. dothidea . This study would accelerate the exploration of the molecular mechanism of the inhibitory effect of DT against B. dothidea and also provide new insights for the management of apple ring rot disease.

Identification and evaluation of an endophytic antagonistic yeast for the control of gray mold (Botrytis cinerea) in apple and mechanisms of action

Abstract:Gray mold, caused by Botrytis cinerea, is a prevalent postharvest disease of apple that limits their shelf life, resulting in significant economic losses. The use of antagonistic microorganisms has been shown to be an effective approach for managing postharvest diseases of fruit. In the present study, an endophytic yeast strain PGY-2 was isolated from apples and evaluated for its biocontrol efficacy against gray mold and its mechanisms of action. Results indicated that strain PGY-2, identified as Bullera alba, reduced the occurrence of gray mold on apples and significantly inhibited lesion development in pathogen-inoculated wounds. Gray mold control increased with the use of increasing concentrations of PGY-2, with the best disease control observed at 108 cells/mL. Notably, Bullera alba PGY-2 did not inhibit the growth of Botrytis cinerea in vitro indicating that the yeast antagonist did not produce antimicrobial compounds. The rapid colonization and stable population of PGY-2 in apple wounds at 4 °C and 25 °C confirmed its ability to compete with pathogens for nutrients and space. PGY-2 also had a strong ability to form a biofilm and enhanced the activity of multiple defense-related enzymes (POD, PPO, APX, SOD, PAL) in host tissues. Our study is the first time to report the use of Bullera alba PGY-2 as a biocontrol agent for postharvest diseases of apple and provide evidence that Bullera alba PGY-2 represents an endophytic antagonistic yeast with promising biocontrol potential and alternative to the use of synthetic, chemical fungicides for the control of postharvest gray mold in apples.
Characterization and Biocontrol Efficacy of Bacillus velezensis GYUN-1190 against Apple Bitter Rot

Abstract:The application of synthetic fungicides has resulted in environmental pollution and adverse effects on non-target species. To reduce the use of agrochemicals, crop disease management requires microbial biological control agents. Bacillus-related genera produce secondary metabolites to control fungal pathogens. Bacillus velezensis GYUN-1190, isolated from soil, showed antagonistic activity against Colletotrichum fructicola, the apple anthracnose pathogen. Volatile organic compounds and culture filtrate (CF) from GYUN-1190 inhibited C. fructicola growth in vitro, by 80.9% and 30.25%, respectively. The CF of GYUN-1190 inhibited pathogen spore germination more than cell suspensions at 10 8 cfu/ml. Furthermore, GYUN-1190 CF is effective in inhibiting C. fructicola mycelial growth in vitro, and it suppresses apple fruit bitter rot more effectively than GYUN-1190 cell suspensions and pyraclostrobin in planta. The mycelial growth of C. fructicola was completely inhibited 48 h after immersion into the CF, in compared with positive controls and GYUN-1190 cell suspensions. The genetic mechanism underlying the biocontrol features of GYUN-1190 was defined using its whole-genome sequence, which was closely compared to similar strains. It consisted of 4,240,653 bp with 45.9% GC content, with 4,142 coding sequences, 87 tRNA, and 28 rRNA genes. The genomic investigation found 14 putative secondary metabolite biosynthetic gene clusters. The investigation suggests that B. velezensis GYUN-1190 might be more effective than chemical fungicides and could address its potential as a biological control agent.
Study on Screening of Antagonistic Bacteria for Apple Postharvest Diseases and Biocontrol Efficacy

Abstract:3 strains of antagonistic bacteria were screened from healthy 'Red Fuji' apple peel by the flat panel separation,and the inhibition of antagonistic bacteria to blue mould was studied through the living test and in vitro test.The results showed that the bacteriostatic effect was obvious;the control effect of strain BF-2 reached 87.6%,and the inhibition rate of BF-3 was 33.3% in the confrontation experiment.The antagonistic bacteria were fermented with the beef extract peptone liquid medium,and there was bacteriostatic factor in fermentation medium,and the diameter of bacteriostatic ring caused by strain BF-3 zymotic fluid was 27.4 mm.The antibacterial ability was affected by the fermentation time and temperature.
