Abstract:Objective: To explore the effects and the possible mechanism of electroacupuncture (EA) on diabetic bladder dysfunction (DBD) in streptozotocin-high fat diet (STZ-HFD) induced type 2 diabetes mellitus (T2DM) rats.Methods: The experiment was divided into Control, diabetic bladder dysfunction, electroacupuncture, and Sham electroacupuncture group. After 8 weeks of electroacupuncture intervention, the body mass, 24 h urine volume, intraperitoneal glucose tolerance test (IPGTT), and urodynamics were detected. After the wet weight of the bladder was detected, the hematoxylin-eosin (HE), Masson’s trichrome, and TUNEL were used to analyze histological changes. The PACAP38 expressions in the bladder were detected by Real-time PCR and Western blot.Results: Compared to the Control group, the bladder wet weight, 24 h urine volume, blood glucose, maximum bladder capacity, bladder compliance, bladder wall thickness, the smooth muscle/collagen ratio, and apoptosis rate of the diabetic bladder dysfunction group were significantly increased. Moreover, the body mass and leak point pressure were significantly reduced. Compared with the Sham electroacupuncture group, the bladder wet weight, maximum bladder capacity, bladder compliance, bladder wall thickness, and apoptosis rate of the electroacupuncture group were significantly reduced. In contrast, the leak point pressure was increased. The PACAP38 mRNA and PACAP38 protein expression of the diabetic bladder dysfunction group were significantly lower than the Control group, while electroacupuncture treatment could upregulate PACAP38 mRNA levels and PACAP38 protein expression of diabetic bladder dysfunction model rats.Conclusion: electroacupuncture could ameliorate bladder dysfunction in the diabetic bladder dysfunction model rats by reversing bladder remodeling, which might be mainly mediated by regulating the PACAP38 level.
Effects of electroacupuncture stimulation at different spinal segmental levels in a rat model of diabetes mellitus

Abstract:Background: Diabetes mellitus (DM) is associated with high morbidity, mortality and economic cost. Studies have shown that acupuncture can improve many symptoms of DM. Objectives: To examine for differences in effects of electroacupuncture (EA) stimulation at Weiwanxiashu, BL15 and BL23 in the streptozotocin (STZ)-induced DM rat model, to help guide clinical selection of acupuncture points. Methods: 90 male rats weighing 160Â±5 g were used. 12 rats were control fed (Normal group) and 78 were fed a high-fat high-sugar diet for 8 weeks and underwent intraperitoneal STZ injection to model DM. 60 animals that met modelling criteria were randomly divided into an untreated DM group and four groups receiving EA at Weiwanxiashu (DM+WWX group), BL15 (DM+BL15 group), BL23 (DM+BL23 group) or a non-traditional acupuncture point on the tail (DM+Tail group). Fasting blood glucose, total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) and insulin levels were determined and an oral glucose tolerance test (OGTT) performed. Results: EA at Weiwanxiashu had a glucose-lowering effect on the 21st and 28th days, decreased TC, TG and LDL-C levels, increase insulin levels and improved glucose tolerance. EA at BL15 had a glucose-lowering effect on the7th, 14th and 21st days of intervention but did not impact lipids, insulin or OGTT parameters. EA at BL23 or on the tail had no significant effects. Conclusion: EA at Weiwanxiashu and BL15 had differential effects on metabolic markers in the STZ-induced rat model of DM. These effects may be explained neuroanatomically by variations in the segmental innervation of the tissues at these locations.

Abstract:A fourth ventricle lesion containing both areas of typical ependymoma and subependymoma has been studied with the electron microscope. Both foci show ependymal and astrocytic elements. In the subependymoma, a larger number of astrocytes, more frequent cytoplasmic degeneration, and increased glial filaments in the ependymal cells are found. These cellular changes in the subependymoma yield a distinctive light microscopic appearance. However, at the ultrastructural level, the similarity of cellular components, i.e. ependymal and astrocytic cells, makes us believe that subependymoma is a variant of ependymoma and not pure astrocytoma. Tissue culture study and further electron microscopic observation of these in vitro cells have also confirmed the presence of both ependymal and astrocytic cells. Like in vivo ependymal cells, these in vitro ependymal cells form not only glial filaments but also rosettes with specialized cell junctions, microvilli, and cilia. Perivascular pseudorosette formation is also described in detail.
Effect of Electroacupuncture on Spermatogenesis in Rats with Oligozoospermia of Insufficiency of Shen (Kidney) Essence Syndrome

Abstract:Objective: To assess the effect of electroacupuncture (EA) on expression of cytoskeletal proteins from Sertoli cells (SCs) and spermatogenesis in rats with oligozoospermia of insufficiency of Shen (Kidney) essence syndrome (OIKES). Methods: Twenty healthy male Sprague-Dawley rats were randomly assigned to four groups using a random number table: control, tripterygium glycosides (TG) treatment, sham and EA groups (n=5 in each group). A rat model of OIKES was established by oral gavage with TG. The EA group was treated with TG and received EA at Shenshu (BL 23) and Zusanli (ST 36) acupoints for 20 min, once daily for 30 days, while the sham group received EA at identical acupoints with skin penetration without stimulation. After 30 days, the final body weight and coefficients for the testis and epididymis were calculated and sperm parameters were measured. Immunohistochemical analyses were performed to detect expression of vimentin and α-tubulin in SCs and proliferating cell nuclear antigen (PCNA) immunoreactivity in germ cells. Apoptosis in germ cells was quantified by the transferase biotin-dUTP nick end labeling assay. Results: Compared with the control group, the final body weight and testis/epididymis coefficients of rats in the TG-treated group were not significantly different, but the sperm count and motility were lower (P<0.05). Expressions of vimentin and α-tubulin were also significantly weaker (P<0.01). The PCNA immunoreactivity of germ cells was decreased (P=0.059), whereas the apoptotic index of germ cells was increased significantly (P<0.01). In contrast, EA at BL 23 and ST 36 acupoints significantly improved the final body weight as well as the sperm count, concentration and motility (P<0.01 or P<0.05). EA increased expression of vimentin and α-tubulin in SCs markedly, and significantly enhanced PCNA immunoreactivity with decreased apoptosis in germ cells (P<0.01 or P<0.05). Conclusions: EA at BL 23 and ST 36 acupoints has protective effects on spermatogenesis in rats with OIKES. This effect seems to be achieved by attenuating TG-induced disruption of cytoskeletal protein in SCs.
A stab-and-roll biopsy technique to maintain gingival epithelium for desquamative gingivitis.

Abstract:BACKGROUND Desquamative gingivitis (DG) is a clinical manifestation common to several diseases. It is known that most cases of DG are caused by mucous membrane pemphigoid (MMP), oral lichen planus (OLP), or pemphigus vulgaris (PV). Early recognition and treatment of these diseases can improve the prognosis, but diagnostic delays are common in patients with DG because obtaining a diagnostic biopsy is technically challenging. A biopsy technique designed to maintain the gingival epithelium for patients with DG was developed. The usefulness of this technique is discussed. METHODS This study is based on a retrospective review of 27 DG cases. A stab-and-roll technique was used to obtain gingival tissue. This technique is designed to reduce lateral forces on the epithelium during the procedure and to thereby prevent the inadvertent removal of the epithelium from the biopsy specimen. A total of 52 biopsies comprising 27 for hematoxylin and eosin (H&E)-stained samples and 25 for direct immunofluorescence (DIF) testing were reviewed. RESULTS Fifty-one of the 52 biopsies (98.1%) maintained the epithelium. Only one biopsy (1.9%) showed that the epithelium was totally absent. Therefore, H&E and DIF features of 51 biopsies were analyzed. Definitive diagnoses of the diseases causing DG included MMP (13 cases), PV (eight cases), and OLP (six cases). CONCLUSIONS A diagnostic biopsy was obtained from the gingiva of patients with DG using the stab-and-roll technique. The gingival epithelium was well maintained, and the relationship with the underlying connective tissue was diagnostic. In the future, this stab-and-roll biopsy technique may facilitate early diagnosis and treatment of diseases causing DG.

Abstract:One of the key characteristics of cellular Machine-to-Machine (M2M) communications is the generation of small data packets by a large number of devices. The excessive control signaling required for dynamic scheduling of such uplink data in the Long Term Evolution (LTE) standard impedes the deployment of LTE networks for M2M communications. Traditional Semi-persistent scheduling (SPS) reduces the load on control channel but it is currently only developed for constant data rate voice packets. For realistic M2M environments, non-deterministic data traffic needs to be served in a resource efficient way. In this paper, we propose a buffer based adaptive SPS scheme for the LTE uplink which does not require the control signaling overhead of dynamic scheduling, but offers the same flexibility of increasing or decreasing the number of allocated radio resource blocks according to the latest buffer status of the device. We examine the influence of SPS periodicity on quality of Service (QoS) satisfaction of delay constrained stochastic M2M traffic and compare the performance of the proposed adaptive SPS scheme with rigid fixed allocation SPS schemes for such traffic. We also demonstrate how the adaptive SPS scheme can be implemented in a resource efficient way to meet the QoS requirements of a large number of machine type nodes through data aggregation.
Modelling of the inventory replenishment problem with a two-segment piecewise linear demand

Abstract:The classical inventory replenishment problem with a linear function in demand uses a ‘single-segment’ linear function as its demand and can be modelled by a simple algorithm. Moreover, this article extends the algorithm to provide a heuristic solution for the inventory replenishment model with a two-segment linear function in demand called the ‘two-segment piecewise linear demand model’. In addition, this article proposes a general procedure for solving both models. Meanwhile, several examples taken from the literature illustrate our algorithm for these two models with convincing results. Furthermore, this study shows that when the demand is a two-segment piecewise linear function over time, it is better to use the proposed algorithm rather than devising a decoupled solution approach by treating segments separately. Finally, a sensitivity analysis of two factors, demand and cost, is performed. The model is highly extensible and applicable, so it can serve as an inventory planning tool to solve the replenishment problem.

Abstract:Hall et al (Headache. 2004;44:204-208) recorded cortical activity during migraine visual aura using magnetoencephalography. These authors conclude that their results are consistent with the theory of cortical spreading depression (CSD). If CSD is the neuro-physiological equivalent—and basis—of the migraine aura, it cannot be the attack-initiating event because the migraine prodrome—that precedes both the headache as well as the aura—can last several hours or a few days.1 About 20 years ago, Pearce underscored that CSD is not the inaugural event at the level of the brain.2 The advent of sophisticated neuroimaging does not diminish the importance of such fundamental pathophysiological considerations. Any theoretical model must correspond with clinical reality, by which benchmark the theory of CSD has serious drawbacks.3,4 Pharmacological agents that do not cross the blood-brain barrier (BBB) instantaneously abort migraine aura.4 In cats, CSD is not associated with release of calcitonin gene-related peptide.5 Dihydroergotamine, acetylsalicylic acid, metoprolol, or valproate do not affect CSD in the cat.6 Spreading depression cannot be elicited in the brain stem of the adult rat, even after conditioning.7 The brain stem is the focus of several pathogenetic concepts for migraine. Premature philosophic commitment encourages scientists to ignore contradictory evidences. A Medline review reveals a number of studies that show the neuroprotective effect of CSD following brain injury or ischemia.8,9 CSD is a powerful yet largely benign stimulus that acutely is capable of providing long-lasting ischemic tolerance probably through ischemic preconditioning; prolonged increase in atrial natriuretic peptide after an acute episode of CSD possibly contributes to CSD-induced neuroprotection.10 The neuroprotective effect of CSD has been known since about two decades; needle puncture of the brain improves survival following forebrain ischemia in mice.11 There is little evidence to support the popular belief that CSD is an important brain pathophysiological mechanism. Insofar as migraine pathophysiology is concerned, the ability of atenolol—that does not cross BBB—to prevent migraine with or without aura heralds the demise of a popular neurological theoretical concept.12 The scientific imperative to explain occurrence of migraine aura must not dull our ability to think logically.

Abstract:Glycosaminoglycans (GAGs) have been implicated in many functions of human blood cells. Normal human monocytes grown on plastic culture dishes for five days display macrophage-like morphology and functional characteristics concurrent with the appearance of a 4,6-disulfated chondroitin. This process has been regarded as in vitro differentiation. HL-60 human promyelocytic leukemia cells produce chondroitin 4-sulfate, which is the major GAG constituent of normal granulocytes and monocytes. These cells can be induced by phorbol myristate acetate (PMA) to develop many macrophage-like characteristics. This study investigates GAG production by HL-60 cells after prolonged exposure to PMA. Disulfated chondroitin molecules were not detected. This biochemical abnormality may be an important factor in the functional deficiencies of these induced cells.

Abstract:Abstract Background Diabetes mellitus is one of the major current public health problems. Electro-acupuncture at ST-36 showed a significant reduction in plasma glucose levels in diabetic rats. There are lacks of studies reporting its hypoglycemic effect in humans and thus, the present study was conducted to evaluate the effect of needling at ST-36 (Zusanli) on blood glucose levels in patients with type 2 diabetes mellitus (T2DM). Methods Sixty T2DM participants were recruited and randomized into acupuncture group (n=30) and placebo control group (n=30). The participants of the acupuncture group received needling at ST-36 (one finger breadth lateral to the inferior border of the tibial tuberosity), and the participants those in the placebo control group received needling at placebo point (midpoint between the apex of the patella and tibial tuberosity). For both the groups, needles were retained for 30 min. Baseline and post-test assessments were performed prior to and after each intervention. Statistical analysis was performed using SPSS version 16. Results The present study showed a significant reduction in random blood glucose levels in the acupuncture group compared to the placebo control group. Conclusion This study suggests that 30 min of bilateral acupuncture needling at ST-36 with manual stimulation is effective in reducing blood glucose levels in T2DM patients.
Autonomically mediated anti-inflammatory effects of electrical stimulation at acupoints in a rodent model of colonic inflammation

Abstract:Background: Acupuncture has been widely accepted for treatments of many diseases. This study was performed to determine effects and mechanisms of electroacupuncture (EA) by chronically implanted electrodes at acupoint ST36 on colonic inflammation induced by TNBS in rats. Methods: After intrarectal administration of TNBS, the rats were treated with sham-EA, EA1/EA2 (two sets of parameters) for 3 weeks. Disease activity index (DAI), macroscopic and microscopic lesions, plasma levels of TNF-α, IL-1β and IL-6 were observed as evaluation of inflammatory responses. The autonomic function was assessed by analysis of the heart rate variability. Results: (a) Vagal activity was significantly increased with both acute and chronic EA1/EA2; (b) DAI was significantly decreased with both chronic EA1 and EA2, and EA2 was more potent than EA1 (P < 0.05); (c) The macroscopic score was 6.4 ± 0.6 with sham-EA and reduced to 4.9 ± 0.1 with EA1 (P < 0.05) and 4.0 ± 0.2 with EA2 (all P < 0.05). The histological score was 4.05 ± 0.58 with sham-EA and remained unchanged (3.71 ± 0.28) with EA1 (P > 0.05) but reduced to 3.0 ± 0.3 with EA2 (P < 0.01); (d) The plasma levels of TNF-α, IL-1β and IL-6 were significantly decreased with EA2. Conclusions: Electrical stimulation at ST36 improves colonic inflammation in TNBS-treated rats by inhibiting pro-inflammatory cytokines via the autonomic mechanism.

Abstract:Objective Renal microvascular injury, as the result of diabetic toxicity, plays a vital role in the pathogenesis of diabetic kidney disease (DKD) during diabetes progression. Here, we investigated whether electroacupuncture (EA) could ameliorate renal microvascular impairment to prevent DKD and its underlying mechanism. Methods The male db/db mice with Leprdb mutation were used as the model of type 2 diabetes mellitus-induced DKD and treated with EA at “Zusanli (ST36)” and “Weiwanxiashu (EX-B3)” acupoints for 4 weeks. Age-matched wild-type mice were used as control group. Renal protection of EA was evaluated by mouse urine production, water consumption, renal index and tubules dilation. Two-photon microscope imaging was applied to visualize renal microvascular blood flow in vivo. Immunostaining and western blot analysis were used to detect the glomerular alternations and inflammatory signaling. Results EA significantly attenuated renal dysfunction in db/db mice. The protective effect of EA on renal microvascular recovery was observed both in function and structure analysis. Firstly, EA restored the renal microvascular blood flow in db/db mice. Then, glomerular hypertrophy and glomerular barrier destruction were suppressed after EA, as respectively demonstrated by the reduction of glomerular dilation, Collagen Ⅳ and Claudin-1 deposits. In mechanism, EA suppressed the diabetes-induced inflammatory response in renal microvessels, presenting as the downregulation of inflammatory cytokines interleukin-1β (IL-1β) and tumor necrosis factor (TNF-α), intercellular cell adhesion molecule-1 (ICAM-1) activation, and macrophage infiltration after EA treatment. Conclusion These findings indicated the benefits of EA against renal microvascular impairment and DKD progression, which was associated with the action of anti-inflammation, and supported EA as a promising modality for DKD management.

[Effect of Electroacupuncture Intervention on Expression of Testicular P 450 Scc/17 Β-Hsd3 in Rats with Partial Androgen Deficiency].

Abstract:OBJECTIVE:To observe the effect of electroacupuncture (EA) intervention on expression of cytochrome P 450 side chain cleavage (P 450 scc) and 17 β-hydroxysteroid dehydrogenase 3 (17 β-HSD3) in the testis in partial androgen deficiency of aging male (PADAM) rats so as to reveal its mechanism underlying improving PADAM.METHODS:Thirty male SD rats were randomly and equally divided into control, model, and EA groups. The PADAM model was established by intraperitoneal injection of cyclophosphamide (20 mg · kg(-1) · d(-1)), once daily for 5 days. EA (20-30 Hz, 1-3 mA) was applied to bilateral "Shenshu" (BL 23) and "Guanyuan" (CV 4) for 15 min, once daily for 8 weeks. Serum total testosterone (TT) and free testosterone (FT) levels were measured using enzyme-linked immunosorbent assay (ELISA). The expression levels of P 450 scc/17 β-HSD3 proteins and mRNA in the testis tissue were assayed by immunohistochemistry, Western bolt (WB) and RT-polymerase chain reaction (RT-PCR), separately.RESULTS:Compared with the control group, both serum TT, FT levels and expression levels of P450 scc/17 β-HSD3 proteins and mRNA in the testis tissue in the model group were significantly down-regulated (P<0.01). After EA intervention, compared with the model group, the cyclophosphamide-induced decrease of serum TT, FT levels and the expression levels of P 450 scc/17 β-HSD3 proteins and mRNA in the testis was reversed in the EA group (P<0.01).CONCLUSION:EA intervention is effective in up-regulating serum TT and FT, testicular P 450 scc and 17 β-HSD3 proteins and mRNA levels in PADAM rats, which may be one of its mechanisms underlying improvement of PADAM.

Abstract:The purpose of this study was to examine the effects of Electroacupuncture (EA) Zusanli (ST36) on release of nitric oxide (NO) in the gracile nucleus (GN) and functional neuropathic changes in Zucker Diabetic Fatty (ZDF) rats and Lean Control (LC) rats. The foot withdrawal responses to mechanical, hot and cold stimuli were measured before and after EA stimulation. A microdialysis probe was implanted in GN and dialysate samples were collected 20 min before, during and after EA stimulation. Total nitrate and nitrite (NOx‐) concentrations in the samples were quantified by using chemiluminescence. The baseline withdrawal latencies to mechanical, cold and hot stimuli were significantly lower in ZDF rats than that of LC rats. The latencies to these stimuli were increased 20 min after EA ST36 both in LC and ZDF rats (p<0.05), but not altered by non‐point stimulation. The baseline dialysate NOx‐ concentrations in GN were decreased in ZDF rats compared to LC rats (p<0.05). Dialysate NOx‐ releases in GN were significantly increased during and 20 min after EA ST36 in ZDF rats. These results show that EA ST36 increases NO release in GN, which matches the time interval of EA‐induced changes in latencies to mechanical, cold and hot nociception stimuli. Abstract:Objective: To investigate the effects of different electroacupuncture (EA) parameters for the treatment of asthenozoospermia in rats. Methods: One hundred and five male Sprague-Dawley rats were randomly divided into 2 Hz-EA treatment daily in 3 d group (n=9), sham-EA group (n=10), model group (n=10); 2 Hz-EA treatment every other day in 5 d group, sham-EA group, model group (8 rats in each group); 2 Hz-EA treatment every other day in 9 d group, sham-EA group, model group (10 rats in each group); 100 Hz-EA treatment every other day in 9 d group (n=7), sham-EA group (n=8), model group (n=7). Asthenozoospermia model was established by intragastric administration of ornidazole (ORN,400 mg·kg-1·d-1) once daily till the end of treatment. EA treatments (2 Hz or 100 Hz) were applied to "Shenshu" (BL 23,bilateral), "Zusanli" (ST 36, bilateral) for 30 min, intensity of 1-2-3 mA (increasing 1 mA per 10 min), once a day or once every other day for 3 times or 5 times. Sham-EA groups were treated with similar procedure except that the output leads of the stimulator were disconnected. The sperm density, viability, motility, the number of grade A sperm, and grade A+B sperm were examined by computer-assisted sperm analysis. Results: (1) 2 Hz-EA treatment daily in 3 d:compared with the model group and the sham-EA group, 2 Hz-EA treatment once daily had no significant effect on all of the sperm motility indexes in the asthenozoospermic rats (P>0.05). (2) 2 Hz-EA treatment every other day in 5 d:compared with the model group, EA treatment could increase the sperm motility (P<0.05), the number of grade A sperm (P<0.05), and the number of grade A+B sperm (P<0.05) in the asthenozoospermic rats. However, compared with the sham-EA group, EA treatment could only improve the number of grade A+B sperm (P<0.05). (3) 2 Hz-EA treatment every other day in 9 d:compared with both the model group and the sham-EA group, EA treatment could markedly improve the sperm viability (P<0.001), the sperm motility (P<0.001), the number of grade A sperm (P<0.001), and the number of grade A+B sperm (P<0.001) in the asthenozoospermic rats. (4) 100 Hz-EA treatment every other day in 9 days:compared with both the model group and the sham-EA group, all of the sperm indexes in the asthenozoospermic rats including the sperm viability (P<0.001 vs. the model group, P<0.05 vs. the sham-EA group), the sperm motility (P<0.001 vs. the model group, P<0.01 vs. the sham-EA group), the number of grade A sperm (P<0.01) and the number of grade A+B sperm (P<0.01) also could be improved after EA treatment. Unexpectedly,none of the EA treatment had significant influence on the sperm density in the asthenozoospermic rats. Conclusions: Both 2 Hz-EA and 100 Hz-EA treatment once every other day for 5 times in 9 d had a therapeutic effect on asthenozoospermia by improving the sperm viability and the sperm motility in the rats.
