Abstract:After investigating two anion-exchange resins, the purification factor and activity yields of P450 BM-3 were higher with Resource Q than with DEAE-Sepharose FF. Screening of HIC media showed that Source 15ISO was the most suitable for purification of P450 BM-3. An effective isolation and purification procedure of P450 BM-3 was developed and included three steps: 35%-70% saturation (NH(4))(2)SO(4) precipitation, Source 15ISO hydrophobic interaction chromatograph and Sephacryl S-200 gel filtration chromatography. Using this protocol, the purification factor and P450 BM-3 activity recovery was 13.5 and 13.7%, respectively.

Abstract:AIM: To separate propafenone enantiomers in rat liver microsomal incubates and investigate probable stereoselectivity in their N-dealkylation. METHODS: The concentration of each enantiomer in rat liver microsomal incubates was determined through precolumn derivatization with 2,3,4,6-tetra-O-acetyl-β-glucopyranosyl isothiocyanate (GITC), followed by RP-HPLC assay. RESULTS: A baseline separation of propafenone enantiomers was achieved on C 18-ODS column, with methanol - water - glacial acetic acid (67: 33:0.05) as mobile phase. The assay was linear from 0.5 to 320 μg·mL -1 for each enantiomer, and the limit of detection was 100 ng·mL -1. The method affords the average recoveries of 77.1% for R-propafenone and 76.0% for S-propafenone. Stereoselectivity was observed for the phase I metabolism of racemic propafenone in dexamethasone, β-naphthoflavone induced rat liver microsomal incubates,but not in controls. CONCLUSION: The method is simple, cheap and can be applied to study the metabolism of R- and S-propafenone in vitro. Stereoselectivity exists in their N-dealkylation.
Two-step Separation Strategy with Mixed-Mode Chromatography to Purify Recombinant Human Serum Albumin from Pichia Pastoris Culture Broth

Abstract:Mixed-mode chromatography (MMC) with the specially-designed ligands shows good adsorption selectivity for protein separation. In this study MMC was explored to purify recombinant human serum albumin (rHSA) from Pichia pastoris culture broth, which combined bind-elute and flow-through modes. Four mixed-mode resins were evaluated for their adsorption capacities of HSA. It was found that two home-made MMC resins showed special performances. IC-B-6FF resin with indole-3-acetic acid-cysteine ligand had high adsorption and selectivity towards HSA, and SFA-B-6FF resin with sulfapyridine ligand showed minimal binding to HSA. Therefore, a twostep MMC separation strategy was designed that IC-B-6FF was used to capture rHSA directly from yeast culture broth and then SFA-B-6FF resin was used to polish the remaining impurities by flow-through mode. The results revealed that the purity of rHSA reached 95.1% with a yield of 81.2% after the two-step separation. The removal rate of host cell proteins (HCPs) was 99.8% with final HCP level as low as 50 ppm. The results demonstrate that MMC combining bind-elute and flow-through modes has a promising potential to separate protein with high selectivity and process efficiency.

Abstract:The efficient separation of palladium, one of the precious metals, is one of the most challenging works that have not been solved. Based on soft-ligand multi-nitrogen organic agents, a new triazine-containing hybrid functional silica-adsorbent, BisDiOTP@SiO2, was prepared and characterized. The uptake of more than ten representative metals towards BisDiOTP@SiO2 was investigated in the wide range of 0.4-5.0 M HNO3. The high uptake ability and excellent selectivity of Pd(II) towards BisDiOTP@SiO2 were indicated. The enrichment and removal of Pd(II) by batch experiments through six cycles of uptake and desorption were conducted. The separation efficiency was 99.3% for Pd(II), 5.9% for Co(II) and less than 0.2% for all others. Pd(II) was separated effectively from others. A new technical process entitled SPMP (Simultaneous Partitioning of MA(III)/Pd(H) by Extraction Chromatography) for Pall) separation along with minor actinides has been developed.
Path Planning for Altruistically Negotiating Systems: The Near-Sighted Tarzan Algorithm

Abstract:This paper introduces a variant of the Rapidlyexploring Random Leafy Tree, called the Near-sighted Tarzan Algorithm, that performs path planning (for message routing) in a discrete graph simulating the communication network of an Altruistically Negotiating System. The Near-sighted Tarzan Algorithm is designed to operate in environments with similar characterstics as an ad hoc network. These characteristics include an unordered topology, large quantities of agents, network connectivity, and limited knowledge of the network by any given agent. Comparison tests with Dijkstra’s algorithm as a benchmark show the Tarzan algorithm to be superior when searching for paths between large distances. The Tarzan algorithm can be easily run in parallel making it suitable for use in an Altruistically Negotiating System. Abstract:S-adenosylmethionine synthetase (MAT) catalyzes the synthesis of S-adenosylmethionine (SAM) from ATP and L-methionine. SAM is the major methyl donor for more than 100 transmethylation reactions. It is also a common cosubstrate involved in transsulfuration and aminopropylation. However, product inhibition largely restrains the activity of MAT and limits the enzymatic synthesis of SAM. In this research, the product inhibition of MAT from Escherichia coli was reduced via semi-rational modification. A triple variant (Variant III, I303 V/I65 V/L186 V) showed a 42-fold increase in K-i,K-ATP and a 2.08-fold increase in specific activity when compared to wild-type MAT. Its K-i,K-ATP was 0.42 mM and specific acitivity was 3.78 +/- 0.19 U/mg. Increased K-i,K-ATP means reduced product inhibition which enhances SAM accumulation. The SAM produced by Variant III could reach to 3.27 mM while SAM produced by wild-type MAT was 1.62 mM in the presence of 10 mM substrates. When the residue in 104th of Variant III was further optimized by site-saturated mutagenesis, the specific activity of Variant IV (I303 V/I65 V/L186 V/N104 K) reached to 6.02 +/- 0.22 U/mg at 37 degrees C, though the SAM concentration decreased to 2.68 mM with 10 mM substrates. Analysis of protein 3D structure suggests that changes in hydrogen bonds or other ligand interactions around active site may account for the variety of product inhibition and enzyme activity. The Variant III and Variant IV with reduced inhibition and improved enzyme activity in the study would be more suitable candidates for SAM production in the future.
Purification and Activity Evaluation of Methionine Synthase

Abstract:Methionine synthase (MS, EC2.1.1.13), a key enzyme in the folate metabolism area catalyzing methyl transfer from N5-methyltetrahydrofolate to homocysteine to give tetrahydrofolate and methionine, takes a core position in folate cycle, one-carbon-unit transfer and sculpture amino acid pathways. Cobalamin-dependent methionine synthase was purified from rat liver. The enzyme was purified 609-fold to near homogeneity by batch chromatography on DE-52, anion-exchange chromatography on Q Sepharose Fast Flow and CHT-I hydroxyapatite column and was identified by SDS-PAGE and Western blotting. The enzyme activity was determined by spectrophotometric assay. In addition, the influencing factor and optimal reaction condition were performed. The steady state kinetic of rat liver methionine synthase was similar to that of other mammalian cobalamin-dependent methionine synthase which employed a Ping-Pong mechanism. The result indicated that cobalamin-dependent methionine synthase purified from rat liver is suitable for screening and studying methionine synthase specific inhibitors.
Isolation and purification of murine hepatic non-parenchymal cells by liver circulatory perfusion in situ

Abstract:Aim To establish a new method of liver perfusion in mice for isolation and purification of hepatic non-parenchymal cells.Methods The entire mouse abdominal cavity was exposed,portal vein and inferior vena cava were cannulated with intravenous catheters,then they were connected with three-way pipe and flexible tube to form a closed loop.Immediately after flushing liver,the digestion began with 60 ml collagenase Ⅳ in the circulatory loop at a flow rate of 20 ml.min-1 for up to 30 min.The liver fragments were pressed and cells were collected after incubation in collagenase solution for 30 min.Cell pellets were resuspended in 50% /35% isotonic Percoll solution to purify nonparenchymal cells.Results The yield of hepatic non-parenchymal cells per mouse was 0.5 × 107 ～ 1 × 107 and the percentage of CD45+ cells was(27.21 ± 1.47) %.Conclusion Liver collagenase perfusion in situ and Percoll density gradient centrifugation is an effective method in isolation and purification of hepatic nonparenchymal cells in mice.

Abstract:Efficient and reproducible sample preparation methods are key to successful separation in proteome. Meanwhile, efficient sample preparation also enriches target sample to some extent and plays a pre-separation role. In this study, for the first time we put forward using acidic and basic extraction buffer as complementary methods of standard ways to extract complex proteins in rat liver tissue. We also compared the effect of three extraction methods on sample pre-separation and enrichment. The acidic or basic condition was obtained by adding 0.1% trifluoroacetic acid or 40mM Tris, respectively. The rat liver samples were divided into three equal groups and different extraction buffers were used for each group. The concentration of protein from different methods was measured and the corresponding sample was separated using two-dimensional gel electrophoresis, followed by detection of MALDI-TOF-TOF mass spectrometry. Three extraction lysates were also separated by reversed-phase high performance liquid chromatography. The images in gel and mass spectrum indicated that the different methods can pre-separate and enrich some special proteins. Experimental results also showed that the three methods had good reproducibility and could be applied in proteomic analysis.

Abstract:S-adenosylmethionine decarboxylase (SAMDC) is an essential enzyme for the synthesis of spermidine and spermine in the biosynthetic pathway of polyamines. The total RNA was extracted from colon cancer tissue and amplified by reverse-transcription PCR with two primers, which span the coding region of SAMDC alpha subunit. Clone vector pMD18-T-SAMDC-alpha was successfully constructed by using T-A clone technique. pMD18-T-SAMDC-alpha and pTriEx-4 were digested by NcoI and XhoI double enzymes. The purified SAMDC-alpha fragment was subcloned into the expression vector pTriEx-4 to construct the prokaryotic expression plasmid pTriEx-4-SAMDC-alpha. The recombinant plasmid pTriEx-4-SAMDC-alpha was transformed into competence E. coli JM109 (DE3). The bacterium was induced by IPTG and its lysates were loaded directly onto SDS-PAGE. An approximately 32 kDa exogenous protein was observed on the SDS-PAGE. The protein was verified by Western blot with anti His.Tag monoclonal antibody. The fusion protein including 6 x His.Tag was purified by Ni-NTA chromatographic column. Then, the purified protein can be applied for further research of the immunity of SAMDC.
Characteristics of socially withdrawn youth in France: A retrospective study

Abstract:Background: Poor social interactions have been recognized as a symptom since the beginnings of psychiatry. As far as socially withdrawn youth (SWY) are concerned, studies were mostly conducted on patients seeking care. Our psychiatric outreach team called Psymobile was able to reach SWY patients who were not seeking mental health care. Aims: To identify the clinical and socio-demographic characteristics of SWY patients referred to our Psymobile unit. Method: We carried out a retrospective study on the records of patients aged 18–34 years, who were referred to Psymobile for ‘withdrawal’, between April 2012 and December 2015. Results: In total, 66 patients were included in the study. SWY are predominantly male (80%) from large families or single-parent ones. About 42% had no prior contact with a mental health professional before being referred to Psymobile. The mean duration of withdrawal is 29 months. In all, 42% of SWY use cannabis and 73% present disorders of the sleep–wake schedule. About 71% maintain relations with their families and 73% go out occasionally. They are mostly diagnosed with schizophrenia (37%) or mood disorders (23%). Conclusion: Over one-third of Psymobile patients aged 18–34 years were referred on grounds of social withdrawal. Our data may illustrate more accurately the situation of youth social withdrawal amid the general population than data from help-seeking patients or online questionnaires.

Abstract:A pCW vector harboring rat liver serine dehydratase cDNA was expressed in Escherichia coli. The expressed level was about 5-fold higher in E. coli BL21 than in JM109 cell extract; the former lacked two kinds of proteases. Immunoblot analysis revealed the occurrence of a derivative other than serine dehydratase in the JM109 cell extract. The recombinant enzyme was purified to homogeneity. Staphylococcus aureus V8 protease and trypsin cleaved the enzyme at Glu-206 and Lys-220, respectively, with a concomitant loss of enzyme activity. Spectrophotometrically, the nicked enzyme showed a approximately 50% reduced capacity for binding of the coenzyme pyridoxal phosphate and no spectral change of circular dichroism in the region at 300-480 nm, whereas circular dichroism spectra of both enzymes in the far-UV region were similar, suggesting that proteolysis impairs the coenzyme binding without an accompanying gross change of the secondary structure. Whereas the nicked enzyme behaved like the intact enzyme on Sephadex G-75 column chromatography, it was dissociated into two fragments on the column containing 6 M urea. Upon the removal of urea, both fragments spontaneously refolded. These results suggest that serine dehydratase consists of two folding domains connected by a region that is very susceptible to proteases.

Stereoselective Metabolism of Tetrahydropalmatine Enantiomers in Rat Liver Microsomes

Abstract:Tetrahydropalmatine (THP), with one chiral center, is an active alkaloid ingredient in Rhizoma Corydalis. The aim of the present paper is to study whether THP enantiomers are metabolized stereoselectively in rat, mouse, dog, and monkey liver microsomes, and then, to elucidate which Cytochrome P450 (CYP) isoforms are predominately responsible for the stereoselective metabolism of THP enantiomers in rat liver microsomes (RLM). The results demonstrated that (+)-THP was preferentially metabolized by liver microsomes from rats, mice, dogs, and monkeys, and the intrinsic clearance (Cl(int)) ratios of (+)-THP to (-)-THP were 2.66, 2.85, 4.24, and 1.67, respectively. Compared with the metabolism in untreated RLM, the metabolism of (-)-THP and (+)-THP was significantly increased in dexamethasone (Dex)-induced and β-naphthoflavone (β-NF)-induced RLM; meanwhile, the Cl(int) ratios of (+)-THP to (-)-THP in Dex-induced and β-NF-induced RLM were 5.74 and 0.81, respectively. Ketoconazole had stronger inhibitory effect on (+)-THP than (-)-THP, whereas fluvoxamine had stronger effect on (-)-THP in untreated and Dex-induced or β-NF-induced RLM. The results suggested that THP enantiomers were predominately metabolized by CYP3A1/2 and CYP1A2 in RLM, and CYP3A1/2 preferred to metabolize (+)-THP, whereas CYP1A2 preferred (-)-THP.

Abstract:Objective To establish an effective method to isolate hepatic non-parenchymal cells including hepatic stellate cells (HSCs),sinusoidal endothelial cells (SECs) and macrphages respectivly.Methods The whole livers were removed from rats under sterile condition,then the livers were perfused with digestive enzyme in vitro and cut into species.The species were digested to generate single cells suspension at 37 ℃.By using the methords of Percoll gradient centrifugation and adherent culture,the single cells were isolated into HSCs,SECs,and macrophages.The cell activity was observed by the trypan blue excluding test,and the cell purity was identified using spontaneous fluorescence,immunofluorescence and cellular morphology.Results The purity of hepatic non-parenchymal cells was more than 90％,and the survival rate of HSCs and SECs was more than 85％.The isolated HSCs and SECs could be cultured with 7-10 passages.Conclusion This isolation method is easy and reliable to separate HSCs,SECs and macrophages from the liver of rats with a high purity.
On Molecular Multiple-Access, Broadcast, and Relay Channels in Nanonetworks

Abstract:Molecular communication is a novel paradigm that uses molecules as an information carrier to enable nanomachines to communicate with each other. Interconnections of the nanomachines with molecular communication is envisioned as a nanonetwork. Nanonetworks are expected to enable nanomechines to cooperatively share information such as odor, flavour, light, or any chemical state. In this paper, we develop and present models for the molecular multiple-access, broadcast, and relay channels in a nanonetwork and derive their capacity expressions. Numerical results reveal that the molecular multiple-access of nanomachines to a single nanomachine can be possible with the high molecular communication capacity by selecting the appropriate molecular communication parameters. Similarly, the molecular broadcast can also allow a single nanomachine to communicate with a number of nanomachines with high molecular communication capacity. As a combination of the molecular multiple-access and broadcast channel, we show that the molecular relay channel can improve the molecular communication capacity between two nanomachines using a relay nanomachine.

Abstract:Currently, many prostate cancer patients, detected through the prostate specific antigen test, harbor organ-confined indolent disease that cannot be differentiated from aggressive cancer according to clinically and pathologically known measures. Spermine has been considered as an endogenous inhibitor for prostate-confined cancer growth and its expression has shown correlation with prostate cancer growth rates. If established clinically, measurements of spermine bio-synthesis rates in prostates may predict prostate cancer growth and patient outcomes. Using rat models, we tested the feasibility of quantifying spermine bio-synthesis rates with C-13 NMR. Male Copenhagen rats (10 weeks, n = 6) were injected with uniformly C-13-labeled L-ornithine HCl, and were sacrificed in pairs at 10, 30, and 60 min after injection. Another two rats were injected with saline and sacrificed at 30 min as controls. Prostates were harvested and extracted with perchloric acid and the neutralized solutions were examined by C-13 NMR at 600 MHz. C-13 NMR revealed measurable ornithine, as well as putrescine-spermidine-spermine syntheses in rat prostates, allowing polyamine bio-synthetic and ornithine bio-catabolic rates to be calculated. Our study demonstrated the feasibility of C-13 NMR for measuring bio-synthesis rates of ornithine to spermine enzymatic reactions in rat prostates. The current study established a foundation upon which future investigations of protocols that differentiate prostate cancer growth rates according to the measure of ornithine to spermine bio-synthetic rates may be developed.
[Nano Flow Multidimensional Liquid Chromatography for Proteome Analysis of Rat Liver].

Abstract:A nano flow comprehensive multidimensional liquid chromatography was developed. By using this separation platform, complex samples can be injected, desalted, separated and analyzed in complete automatization. Coupling of the strong cation-exchange (SCX) with capillary reversed-phase liquid chromatography (cRPLC) is described for proteomics of rat liver. A step gradient was applied for SCX column, and a linear gradient was applied for RPLC. The flow rate through the nano scale analytical column during the separation process was maintained at 200 nL/min. The peak capacity for the nano-2D-LC was up to 620 under these conditions.
