Abstract:[Objective] In this study,two L-aspartate α-decarboxylase genes (panD) from Listeria monocytogenes (panDL.m) and Corynebacterium jeikeium (panDC.j) were expressed in Escherichia coli,respectively,and the recombinant enzymes were purified and characterized.[Methods] The panD genes from L.monocytogenes and C.jeikeium were synthesized and inserted into pET28a(+) to obtain expression plasmids,which was then transformed into E.coli BL21(DE3).PanDL.m and PanDC.j were functionally expressed and the recombinant enzymes were purified by HisTrapTM affinity chromatography.Their catalytic properties were characterized and the effects of substrate concentration on enzymatic conversion were studied.[Results] Those α and β subunits were detected by Tricine-SDS-PAGE,indicating that both of the PanDs were processed by self-cleavage.The specific activities were 8.9 U/mg for PanDL.m and 11.8 U/mg for PanDC.j.They exhibited the same optimal temperature at 60 ℃,while they possessed different optimal pH at 7.0 and 6.0,respectively.Both the enzymes were stable at the condition of 30-50 ℃,and pH 4.0-7.0.Compared with the most studied PanD from C.glutamicum,the substrate inhibition effect of the PanDL.m was significantly less.[Conclusion] Under the high temperature and acidic conditions,PanDL.m as well as PanDC.j could specifically transform L-aspartic to β-alanine.PanDL.m could be more appropriate for industrial application because of less substrate inhibition.
Purification and characterization of oxaloacetate decarboxylase from Salmonella typhimurium heterologous expressed in Escherichia coli

Abstract:In order to know the biochemical characterization and function of oxaloacetate decarboxylase from Salmonella typhimurium,the gene of oxaloacetate decarboxylase was cloned and expressed in Escherichia coli.The product was purified by affinity chromatography on monomeric avidin-Sepharose column,and shown 95% pure on SDS-PAGE.It was shown to be composed of three subunits with relevant molecular weight around 65 × 103,46 × 103,8.8 × 103 corresponding to the predicated molecular weight of alpha,beta and gamma subunits respectively by SDS-PAGE.The holo-enzyme was unstable to heat,the activity of oxaloacetate decarboxylase was abolished at 50 ℃ for 1 h;the optimum pH was about 6.0.The activity of oxaloacetate decarboxylase was almost inhibited completely by 1 mmol·L-1 Zn2 + and 0.5 mmol·L-1 oxalate and 20 mmol·L-1 pyruvate,10-20 mmol·L-1 NaCl had obvious activation,but 200-500 mmol·L-1 NaCl had obvious inhibition for the catalytic activity of oxaloacetate decarboxylase.
Purification and characterization of human erythrocyte pyridoxine kinase

Abstract:A 6000-fold purification of pyridoxine kinase from human erythrocytes has been achieved by a combination of DEAE-cellulose chromatography, ammonium sulfate fractionation, gel filtration and preparative disc polyacrylamide gel electrophoresis. Analytic disc polyacrylamide gel electrophoresis at pH 8.7 reveals two protein bands with similar mobility in the purified enzyme, but only one of these has catalytic activity. The enzyme is found to have a pI of 5.5 and a molecular weight of 65000 by gel filtration, and a broad pH optimum of 8.5. The enzyme is labile at acidic pH. The order of activation of divalent metal ions on the enzyme is Co-2+ greater than Mn-2+ greater than Mg-2+ greater than Zn-2+ greater than Cu-2+ greater than Ni-2+ greater than Fe-2+. Of the monovalent cations studied, K+ is the most effective activator, NH+4 is slightly less effective while Na+ is inhibitory. ATP is the specific phosphate donor for the enzyme. Sulfhydryl reagents did not significantly inhibit the enzyme. The Km for pyridoxine is 5.7 with 10-minus 6 M. Pyridoxal, pyridoxamine and 4-deoxypyridoxine inhibit pyridoxine kinase. Inhibition by pyridoxal is competitive and pyridoxal is observed to be phosphorylated effectively by the enzyme. Young human red cells have a higher activity of pyridoxine kinase than old red cells.

Abstract:D-phenylalanine (D-Phe) is an important side-chain building block for semi-synthetic penicillins and cephalosporins. To synthesize D-Phe, D-amino acid aminotransferases (Dat) from Bacillus subtilis WB600, Bacillus licheniformis ATCC 14580, and Bacillus amyloliquefaciens were firstly compared. The theoretical 3 dimensional models of Dat were constructed, which showed appropriate configuration for synthesis of D-Phe in virtual screening. The comparison 3 different Dat on D-Phe formation was tested. All the heterogenous Dat showed high thermostability and pH stability, while Dat from B. subtilis demonstrated a better potential for Phe production than those from B. licheniformis and B. amyloliquefaciens. Then the dat from B. subtilis was expressed in an L-Phe producing chassis of Escherichia coli W14 (pR15ABK) to obtain D-Phe and different factors affecting D-Phe accumulation were tested. The engineered strain E. coli BCEA (pR15ABKApRdatBS) accumulated 1.72 g/L D-Phe in a 15-L jar fermenter which is the highest fermentation concentration that had been reported.

Abstract:Taking DL-phenylalanine as substrate,D-phenylalanine was obtained via using phenylalaine deaminase from Bacillus megaterium AS1.127,which has the specific oxidative deamination ability for L-phenylalanine.The factors affecting the enzymatic reaction were discussed and the optimum conditions were obtained.The results showed that the optimum temperature was 40℃ and pH was 5.8.The presence of 0.2 mol/L phosphate buffer solution resulted in an enhancement of the enzyme activity.The conversion rate was remarkably enhanced in the presence of 0.2% Tween-80 and 10~(-5) mol/L K~+.The conversion rate was related with the concentration of both the substrate and the mycelia.When the concentration of the substrate DL-phenylalanine was 0.09 mol/L,the mycelia was 0.03 g/mL and the reaction time was 18 h,the yield was more than 98%.D-phenylalanine was obtained after decoloring of the enzymic reaction solution,concentrating and crystallizing,and its optical purity was higher than 95%.

Abstract:Ultrasonic was the best method of disrupting cells. Using this method, DAAO activity per milliliter was two times more than that by other methods. The highest activity could reach 12000U/mL. Furthermore, 50% (NH4)2SO4 precipitation could achieve a recovery of 85%. Moreover, a Q-Sepharose column combined with a S-100 column could achieve the best purification. There was a single band on SDS-PAGE. The result of SDS-PAGE indicates that, using the methods mentioned above, we can achieve pure DAAO and the total recovery is 40%.

Abstract:The aproteinogenic amino acid, L-phenylglycine, is an important side chain building block for some drugs. It would be of great commercial and environmental value to biocatalyse L-phenylalanine to L-phenylglycine, and thus replace the organic synthesis method. To produce L-phenylglycine from L-phenylalanine, an L-phenylglycine aminotransferase was screened and characterized. HpgTAO showed high homology to α-aminoadipate aminotransferase. The L-phenylalanine binding site was near the residues S26, R401, N201, and G46 in HpgTAO, and L-phenylalanine formed a hydrogen bond with Asn20, which was similar to the substrate binding mechanism of α-aminoadipate aminotransferase. HpgTAO showed increased activity in alkalescent environment below 40°C. The kinetic analysis showed that L-phenylalanine had the highest affinity to HpgTAO, which ensured the recycle biosynthesis of Lphenylglycine from L-phenylalanine. To date, it was the only aminotransferase using L-phenylalanine as an optimal amino donor. The L-phenylglycine biocatalysis operon was also constructed by co-expressing the hmaS, hmo and hpgT by a single plasmid. The first in vitro conversion of L-phenylalanine to L-phenylglycine was achieved by directly using the L-phenylalanine fermentation broth as the raw material.

Abstract:Purification of a cis-epoxysuccinic acid hydrolase was achieved by ammonium sulfate precipitation, ionic exchange chromatography, hydrophobic interaction chromatography followed by size-exclusion chromatography. The enzyme was purified 177-fold with a yield of 14.4%. The apparent molecular mass of the enzyme was determined to be 33kDa under denaturing conditions. The optimum pH for enzyme activity was 7.0, and the enzyme exhibited maximum activity at about 45 degrees C in 50mM sodium phosphate buffer (pH 7.5). EDTA and o-phenanthrolin inhibited the enzyme activity remarkably, suggesting that the enzyme needs some metal cation to maintain its activity. Results of inductively coupled plasma mass spectrometry analysis indicated that the cis-epoxysuccinic acid hydrolase needs Zn(2+) as a cofactor. Eight amino acids sequenced from the N-terminal region of the cis-epoxysuccinic acid hydrolase showed the same sequence as the N-terminal region of the beta subunit of the cis-epoxysuccinic acid hydrolase obtained from Alcaligenes sp.

Abstract:ABSTRACT Four bacterial phenolic acid decarboxylases (PAD) from Lactobacillus plantarum , Pediococcus pentosaceus , Bacillus subtilis , and Bacillus pumilus were expressed in Escherichia coli , and their activities on p -coumaric, ferulic, and caffeic acids were compared. Although these four enzymes displayed 61% amino acid sequence identity, they exhibit different activities for ferulic and caffeic acid metabolism. To elucidate the domain(s) that determines these differences, chimeric PAD proteins were constructed and expressed in E. coli by exchanging their individual carboxy-terminal portions. Analysis of the chimeric enzyme activities suggests that the C-terminal region may be involved in determining PAD substrate specificity and catalytic capacity. In order to test phenolic acid toxicity, the levels of growth of recombinant E. coli displaying and not displaying PAD activity were compared on medium supplemented with different concentrations of phenolic acids and with differing pHs. Though these acids already have a slight inhibitory effect on E. coli , vinyl phenol derivatives, created during decarboxylation of phenolic acids, were much more inhibitory to the E. coli control strain. To take advantage of this property, a solid medium with the appropriate pH and phenolic acid concentration was developed; in this medium the recombinant E. coli strains expressing PAD activity form colonies approximately five times smaller than those formed by strains devoid of PAD activity.

Path Planning for Altruistically Negotiating Systems: The Near-Sighted Tarzan Algorithm

Abstract:This paper introduces a variant of the Rapidlyexploring Random Leafy Tree, called the Near-sighted Tarzan Algorithm, that performs path planning (for message routing) in a discrete graph simulating the communication network of an Altruistically Negotiating System. The Near-sighted Tarzan Algorithm is designed to operate in environments with similar characterstics as an ad hoc network. These characteristics include an unordered topology, large quantities of agents, network connectivity, and limited knowledge of the network by any given agent. Comparison tests with Dijkstra’s algorithm as a benchmark show the Tarzan algorithm to be superior when searching for paths between large distances. The Tarzan algorithm can be easily run in parallel making it suitable for use in an Altruistically Negotiating System. Functional Characterization of an Aminotransferase Required for Pyoverdine Siderophore Biosynthesis in Pseudomonas aeruginosa PAO1

Abstract:ABSTRACT The fluorescent dihydroxyquinoline chromophore of the pyoverdine siderophore in Pseudomonas is a condensation product of d-tyrosine and l-2,4-diaminobutyrate. Both pvdH and asd (encoding aspartate β-semialdehyde dehydrogenase) knockout mutants of Pseudomonas aeruginosa PAO1 were unable to synthesize pyoverdine under iron-limiting conditions in the absence of l-2,4-diaminobutyrate in the culture media. The pvdH gene was subcloned, and the gene product was hyperexpressed and purified from P. aeruginosa PAO1. PvdH was found to catalyze an aminotransferase reaction, interconverting aspartate β-semialdehyde and l-2,4-diaminobutyrate. Steady-state kinetic analysis with a novel coupled assay established that the enzyme adopts a ping-pong kinetic mechanism and has the highest specificity for α-ketoglutarate. The specificity of the enzyme toward the smaller keto acid pyruvate is 41-fold lower. The enzyme has negligible activity toward other keto acids tested. Homologues of PvdH were present in the genomes of other Pseudomonas spp. These homologues were found in the DNA loci of the corresponding genomes that contain other pyoverdine synthesis genes. This suggests that there is a general mechanism of l-2,4-diaminobutyrate synthesis in Pseudomonas strains that produce the pyoverdine siderophore.

Biochemical evaluation of a parsley tyrosine decarboxylase results in a novel 4-hydroxyphenylacetaldehyde synthase enzyme

Abstract:Plant aromatic amino acid decarboxylases (AAADs) are effectively indistinguishable from plant aromatic acetaldehyde syntheses (AASs) through primary sequence comparison. Spectroscopic analyses of several characterized AASs and AAADs were performed to look for absorbance spectral identifiers. Although this limited survey proved inconclusive, the resulting work enabled the reevaluation of several characterized plant AAS and AAAD enzymes. Upon completion, a previously reported parsley AAAD protein was demonstrated to have AAS activity. Substrate specificity tests demonstrate that this novel AAS enzyme has a unique substrate specificity towards tyrosine (km 0.46mM) and dopa (km 1.40mM). Metabolite analysis established the abundance of tyrosine and absence of dopa in parsley extracts. Such analysis indicates that tyrosine is likely to be the sole physiological substrate. The resulting information suggests that this gene is responsible for the in vivo production of 4-hydroxyphenylacetaldehyde (4-HPAA). This is the first reported case of an AAS enzyme utilizing tyrosine as a primary substrate and the first report of a single enzyme capable of producing 4-HPAA from tyrosine.
Purification, Cloning, Expression, and Biochemical Characterization of a Monofunctional Catalase, KatP, from Pigmentiphaga Sp. DL-8.

Abstract:Catalases are essential components of the cellular equipment used to cope with oxidative stress. The monofunctional catalase KatP was purified from Pigmentiphaga sp. using ammonium sulfate precipitation (ASP), diethylaminoethyl ion exchange chromatography (IEC), and hydrophobic interaction chromatography CHIC). The purified catalase formed polymer with an estimated monomer molecular mass of 54 kDa, which were resolved by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and zymogram analysis. KatP exhibited a specific catalytic activity of 73,000 U/mg, which was higher than that of catalase-1 of Comamonas terrigena N3H (55,900 U/mg). Seven short tryptic fragments of this catalase were obtained by electrospray ionization quadrupole time-of-flight mass spectrometry (ESI-Q-TOF MS/MS), and the gene, katP, was cloned by PCR amplification and overexpressed in Escherichia coli BL21 (DE3). Based on the complete amino acid sequence, KatP was identified as a clade 3 monofunctional catalase. The specific activities of recombinant KatP for hydrogen peroxide (690,000 U/mg) increased 9-fold over that of the parent strain. The K-m and V-max of recombinant KatP were 9.48 mM and 81.2 mol/min mg, respectively. The optimal pH and temperature for KatP were 7.0 and 37 degrees C, respectively, and the enzyme displayed abroad pH-stable range of 4.0-11.0. The enzyme was inhibited by Zn2+, Cu2+, Cr2+, and Mn2+, whereas Fe3+ and Mg2+ stimulated KatP enzymatic activity. Interestingly, the catalase activity of recombinant KatP displayed high stability under different temperature and pH conditions, suggesting that KatP is a potential candidate for the production of catalase. Arginine decarboxylase from Lathyrus sativus seedlings. Purification and properites

Abstract:Arginine decarboxylase which makes its appearance in Lathyrus sativus seedlings after 24 h of seed germination reaches its highest level around 5-7 days, the cotyledons containing about 60% of the total activity in the seedlings at day 5. The cytosol enzyme was purified 977-fold from whole seedlings by steps involving manganese chloride treatment, ammonium sulphate and acetone fractionations, positive adsorption on alumina C-gamma gel, DEAE-Sephadex chromatography followed by preparative disc gel electrophoresis. The enzyme was shown to be homogeneous by electrophoretic and immunological criteria, had a molecular weight of 220,000 and appears to be a hexamer with identical subunits. The optimal pH and temperature for the enzyme activity were 8.5 and 45 degrees C respectively. The enzyme follows typical Michaelis-Menten kinetics with a Km value of 1.73 mM for arginine. Though Mn2+ at lower concentrations stimulated the enzyme activity, there was no dependence of the enzyme on any metal for the activity. The arginine decarboxylase of L. sativus is a sulfhydryl enzyme. The data on co-factor requirement, inhibition by carbonyl reagents, reducing agents and pyridoxal phosphate inhibitors, and a partial reversal by pyridoxal phosphate of inhibition by pyridoxal-HCl suggests that pyridoxal 5'-phosphate is involved as a co-factor for the enzyme. The enzyme activity was inhibited competitively by various amines including the product agmatine. Highest inhibition was obtained with spermine and arcain. The substrate analogue, L-canavanine, homologue L-homoarginine and other basic amino acids like L-lysine and L-ornithine inhibited the enzyme activity competitively, homoarginine being the most effective in this respect.
Design phenylalanine dehydrogenase for L-homophenylalanine synthesis by conservation and co-evolution analysis

Abstract:L-homophenylalanine(L-HPA) is a key chiral intermediate for angiotensin-converting medicine. The study utilized the advantages of the enzymes produced by marine strains that are tolerant to extreme environments, and through co-evolution and conservation analysis, studied the mutation of phenylalanine dehydrogenase during the reductive amination synthesis of L-HPA from 2-oxo-4-phenylbutanoic acid ethyl ester (EOPB), and identified several key mutant residues. 24 single mutants were obtained and the result indicated Lys76Ser, Gly60Ser, Gly60Thr, Tyr285Leu and Asn288Glu enhanced the activity of reductive amination. Four double site mutants were constructed by combination of positive mutations. The k cat /K m of PheDH/ Gly60Thr/Lys76Ser and PheDH/Lys76Ser/Tyr285Leu were increased by 1.96 and 2.03-fold, respectively. Molecular docking revealed the expanded volume and enhanced hydrophobicity of the substrate-binding pocket. Furthermore, the effects of metal ions, salt concentration and organic solvents tolerance were studied. Abstract:Pseudomonas fluorescens E118 was isolated from soil as an effective eugenol-degrading organism by a screening using eugenol as enrichment substrate. The first enzyme involved in the degradation of eugenol in this organism, eugenol dehydrogenase, was purified after induction by eugenol, and the purity of the enzyme was shown by SDS-PAGE and gel-permeation HLPC. The enzyme is a heterodimer that consists of a 10-kDa cytochrome c and a 58-kDa subunit. The larger subunit presumably contains flavin, suggesting a flavocytochrome c structure and an electron transfer via flavin and cytochrome c during dehydrogenation. The activity of the purified enzyme depended on the addition of a final electron acceptor such as phenazine methosulfate, 2,6-dichlorophenol-indophenol, cytochrome c, or potassium ferricyanide. The enzyme catalyzed the dehydrogenation of three different 4-hydroxybenzylic structures including the conversion of eugenol to coniferyl alcohol, 4-alkylphenols to 1-(4-hydroxyphenyl)alcohols, and 4-hydroxybenzylalcohols to the corresponding aldehydes. The catalytic and structural similarity between this enzyme and a Penicillium vanillyl-alcohol oxidase and 4-alkylphenol methylhydroxylases from several Pseudomonas species is discussed.

Abstract:The purpose of this study was to examine the relationship between visual perception and visual-motor integration in 30 normal children compared to 30 clumsy children. Difficulty in visual perception, as assessed by the Test of Visual-Perceptual Skills, accounts for about half the variance in the clumsy children's performance in visual-motor integration, as assessed by the Developmental Test of Visual Motor Integration. In contrast, the correlation between scores on these tests for normal control children was low and not significant. These results suggest that visual perception and visual-motor integration may be separate functions in normally developing children. When considering clumsy children, however, these functions cannot be considered as two independent skills.
Purification and Characterization of an Arene cis-Dihydrodiol Dehydrogenase Endowed with Broad Substrate Specificity toward Polycyclic Aromatic Hydrocarbon Dihydrodiols

Abstract:Initial reactions involved in the bacterial degradation of polycyclic aromatic hydrocarbons (PAHs) include a ring-dihydroxylation catalyzed by a dioxygenase and a subsequent oxidation of the dihydrodiol products by a dehydrogenase. In this study, the dihydrodiol dehydrogenase from the PAH-degrading Sphingomonas strain CHY-1 has been characterized. The bphB gene encoding PAH dihydrodiol dehydrogenase (PDDH) was cloned and overexpressed as a His-tagged protein. The recombinant protein was purified as a homotetramer with an apparent Mr of 110,000. PDDH oxidized the cis-dihydrodiols derived from biphenyl and eight polycyclic hydrocarbons, including chrysene, benz[a]anthracene, and benzo[a]pyene, to corresponding catechols. Remarkably, the enzyme oxidized pyrene 4,5-dihydrodiol, whereas pyrene is not metabolized by strain CHY-1. The PAH catechols produced by PDDH rapidly auto-oxidized in air but were regenerated upon reaction of the o-quinones formed with NADH. Kinetic analyses performed under anoxic conditions revealed that the enzyme efficiently utilized two- to four-ring dihydrodiols, with Km values in the range of 1.4 to 7.1 $\mu$M, and exhibited a much higher Michaelis constant for NAD+ (Km of 160 $\mu$M). At pH 7.0, the specificity constant ranged from (1.3 $\pm$ 0.1) x 106 M?1 s?1 with benz[a]anthracene 1,2-dihydrodiol to (20.0 $\pm$ 0.8) x 106 M?1 s?1 with naphthalene 1,2-dihydrodiol. The catalytic activity of the enzyme was 13-fold higher at pH 9.5. PDDH was subjected to inhibition by NADH and by 3,4-dihydroxyphenanthrene, and the inhibition patterns suggested that the mechanism of the reaction was ordered Bi Bi. The regulation of PDDH activity appears as a means to prevent the accumulation of PAH catechols in bacterial cells.

Abstract:Eleven subjects with clinical bronchial hyperreactivity towards hexamethylene diisocyanate used in their work and verified by spirometry before and after a shift were screened for their antitrypsin phenotypes by isoelectric focusing. One third were M1M1 homozygotes (over 50% in controls) while the rest were heterozygotes comprising one case, each, of M1S and M1I (expected frequencies 5 and 0.5%, respectively). It seems that heterozygous alpha 1-antitrypsin carriers may be at a greater risk to develop a pulmonary disease in the organic diisocyanate exposure than the homozygotes.
