Abstract:As observed by Needleman and co-workers [1], it is still not clear to what extent the initially elastic cohesive surfaces are more appropriate than the initially rigid cohesive surfaces for a given application. In this work, an analysis where the initially elastic model is approached asymptotically to the initially rigid is proposed. The purpose of this analysis is to gain more insight into the numerical mechanisms that lead to different results using different approaches. It was found that convergence is only achieved when the initial cohesive stiffness is much higher than the bulk stiffness. When a relative low value of this cohesive stiffness is chosen, crack tip speed is delayed and branching is artificially increased. GM R&D report # 9650, Aug. 2003. Introduction Cohesive Models have been gaining significant importance in the modeling of crack propagation in recent years. The most commonly used technique to incorporate the cohesive zone model into a finite element analysis is the discrete representation of the crack which is accomplished by introducing cohesive surfaces along inter-element boundaries. Within this framework, these cohesive interfaces can be classified into two approaches: the intrinsic potential-based law used by Xu and Needleman [2], and the extrinsic linear law developed by Camacho and Ortiz [3]. The disti+nction between these two approaches is associated with the way the crack initiation and evolution is modeled. In the intrinsic approach, zero-thickness interface elements are embedded between volumetric elements from the beginning of the analysis. The tensile and shear traction in the interface elements are calculated from the constitutive cohesive law. The interface between two surfaces is intact until the interface traction reaches the maximum value. Once the maximum traction is reached, the interface starts failing and the traction reduces to zero up as the displacement increases up to a critical value according to the traction-displacement relationship. The propagation of a crack can thus be simulated as the consecutive failure of interface elements. These cohesive laws are often called “initially elastic” laws and they can have different shapes, such as exponential [2], trapezoidal [4] and bi-lineal [5,6]. In the extrinsic approach, interface elements are introduced in the mesh only after the corresponding interface is predicted to start failing. Beginning the calculation with a regular mesh, the stress acting along the interface between two volumetric elements is monitored at any time to evaluate where crack will initiate. Once the stress reaches a critical value, a zero-thickness interface element is inserted by duplicating the nodes. Unlike the initially elastic interface elements, the initial cohesive response is rigid and the initial cohesive traction is equal to the critical stress [3]. Then, the interface opens in accordance with a prescribed traction-separation relation called “initially rigid” cohesive law. Recently, Falk et al. [1] have demonstrated that the prediction of crack branching strongly depends on whether the initially rigid or initially elastic approach is used. The reason why the initially rigid approach showed absence of crack branching is unclear. The purpose of this work is to study this issue using a bi-lineal initially elastic cohesive law where the initial stiffness is varied from low to high values without affecting the maximum cohesive traction or the cohesive energy. In this way, the initially elastic model is approached asymptotically to the initially rigid and, therefore, a detailed analysis of the material behavior can be carried out. 2. Approach used by M. Falk, A. Needleman and J. Rice[1] In [1], Falk and co-workers compare the extrinsic approach proposed by Camacho and Ortiz [5] with the intrinsic approach proposed by Xu and Needleman [2]. As explained in [3], using the extrinsic approach, new fracture surfaces are created by splitting nodes according to a brittle fracture criterion. In quadratic triangular elements, mid-side nodes can only be split in one way, namely along the unique element boundary crossing that node. By contrast, interior corner nodes can potentially open up along multiple fracture paths, all of which need to be evaluated in turn. To this end, Camacho and Ortiz [3] begin the process by computing the traction t acting at the node across all potential fracture surfaces. If this traction satisfies the fracture criterion, the nodes involved in that surface are split in order to create two new surfaces. The details of the computation are given in [3]. As previously mentioned, this kind of interface element is called “initially rigid” cohesive element. For the intrinsic approach, potential cohesive surfaces are introduced along all boundaries in a finite element mesh where cracks may initiate and propagate. When these “initially elastic” cohesive surfaces are introduced along all finite element boundaries, the mechanical response clearly depends on the mesh spacing, as well as on physical parameters. In fact, the exponential formulation proposed by Needleman has the feature that the cohesive surfaces contribute to the linear elastic response of the body. Abstract:Objective: The present work was focused on the development and validation of the reversed-phase high-performance liquid chromatography (RP-HPLC) method, which is simple, rapid, precise, accurate, sensitive, economical, and stability-indicating for the quantification of rosuvastatin calcium in bulk and tablet formulation. Methods: The separation was attained on reversed-phase Princeton (C18) column with dimensions (250 mm × 4.6 mm, 5μ) employing buffer, which is a mixture of water (pH 3.0, adjusted with orthophosphoric acid) and methanol in the ratio (20:80) v/v as mobile phase, at flow rate 1.0 ml/min. and detection was carried out at wavelength 240 nm. The retention time under the optimized condition of rosuvastatin calcium was found to be 2.844 min. Results: The linearity of the method was demonstrated in the concentration range of 6-16 μg/ml for rosuvastatin calcium with a correlation coefficient (r2) of 0.9999. The percentage relative standard deviation was ˂2% and percentage recovery was found to be 100.12-101.37% for rosuvastatin calcium. The assay of marketed tablet formulations was found to be 98.99%. Conclusion: The developed RP-HPLC technique was found to be simple, specific, sensitive, rapid, linear, accurate, precise, and economical and will be used for regular quality control of rosuvastatin calcium in bulk and tablet formulations.

Abstract:Rosuvastatin calcium is a widely used 3-hydroxy-3-methylglutaryl coenzyme A-reductase inhibitor developed for the treatment of dyslipidemia. To establish a control strategy for the elemental impurities, a new digestion method combined with an inductively coupled plasma-mass spectrometer (ICP-MS) was developed and validated by our team to determine elements Cd, Pb, As, Hg, Co, V, and Ni in rosuvastatin calcium tablets, which digest the sample perfectly even in the presence of a large number of excipients, especially titanium dioxide. The measurement mode was collision cell mode with kinetic energy discrimination (KED). 209Bi+, 115In+, and 89Y+ were chosen as internal standard elements. The recoveries of the limit of quantitation (LOQ) ranged from 90.5% to 106.4%, concentrations of the abovementioned elements in LOQ were 0.25 μg·L−1, 0.25 μg·L−1, 0.75 μg·L−1, 1.5 μg·L−1, 2.5 μg·L−1, 5 μg·L−1, and 8 μg·L−1 , respectively, linear correlation coefficients were above 0.9997, the recoveries in accuracy item ranged from 91.8% to 103.6%, and relative standard deviations (RSDs) of recovery in precision were not more than 1.8%, reflecting a reliable method of high sensitivity, strong anti-interference capacity, and good precision, and that it was suitable for the determination of elemental impurities in drugs.

Abstract:In this study, quantative nuclear magnetic resonance (qNMR) method was used to determine the content of rosuvastatin in tablet. Linearity, range, limit of detection (LOD), limit of quantification (LOQ), accuracy, and precision were determined in validation study of rosuvastatin. Furthermore, validation study of rosuvastatin was performed with high performance liquid chromatography (HPLC). Uncertainties of qNMR and HPLC methods were determined using per EURACHEM/CITAC Guide CG 4 (3th edition), quantifying uncertainty in analytical measurement. qNMR and HPLC methods were linear in the ranges of 0.10 - 5.00 mg/mL and 0.001 - 0.0995 mg/mL, respectively and these lineraties indicate very good linearity performance with regression coefficients (R2 value) above > 0.99. Moreover, LOD and LOQ values using qNMR method were observed as 0.25 mg/mL and 0.80 mg/mL, respectively. These values using HPLC method were found as 0.00051 µg/mL and 0.001695 µg/mL, respectively. The strengths and weaknesses of qNMR method and HPLC method were determined with spectral emphasis on the role of identical reference standards in qualitive and quantitative analyses. It was found that qNMR method is simple, efficient, reliable, and accurate method. Moreover, qNMR method is an easy, practical, and useful method for the validation and optimization of rosuvastatin in the tablet.

Abstract:Objective To establish a HPLC method to determine the content and related substance of rosiglitazone hydrochloride tablets.Methods The analysis was achieved on a Diamonsil C18 column(250 mm×4.6 mm,5 μm) using a mobile phase of 0.025 mol/L ammonium acetate-acetonitrile(50∶50).The detection wavelength was at 245 nm.Results There was good resolution among the peak of rosiglitazone,the main related substances and the degradable impurities after being destroyed.There was a good linear relationship between the peak area and the concentration of rosiglitazone in the range of 19.57～195.70 μg/ml(r=1.000 0,n=6).Regression equation could be expressed by A=68 525C+31 152,r=1.000 0(n=6).The precision in a day(RSD﹤0.5%) and during the days(RSD﹤1.0%) were good.The average recovery was 99.8%(RSD=0.3%,n=9) and the limit of detection was 0.29 ng.The solution was stable for 10 hours.Conclusion The method was specific,accurate,sensitive and could be used to determine rosiglitazone.

Abstract:Abstract Background The present work is aimed at development and validation of RP HPLC method which is simple, specific, precise, and accurate for estimation of Sofosbuvir and its process-related impurity in bulk and pharmaceutical dosage forms. Extensive literature survey revealed no method for estimation of the above said. The chromatographic separation was achieved on Agilent Eclipse XDB-C18, 4.6 × 250 mm, 5 μm with mobile phase composed of 0.1% trifluoroacetic acid in 1000 ml of water:acetonitrile (50:50) using an isocratic mode of elution. Detection was made using UV detector at 260.0 nm and LC solution software for analysis of data. The developed method was validated according to ICH guidelines. Results The linearity of calibration curve for Sofosbuvir in concentration range of 160-480 μg/ml was good. The curve was linear for its process related impurity (Phosphoryl) in concentration range of 10-30 μg/ml. There exists a good correlation between peak area and analyte concentration. Retention time for Sofosbuvir was found to be 3.674 min and its impurity was 5.704 min. Relative standard deviation values for Sofosbuvir is 1.741 and its process related impurity is 0.043. LOD for Sofosbuvir and its impurity was found to be 0.01% (0.04 μg) and 0.03% (0.12 μg) respectively. LOQ for Sofosbuvir and its impurity was found to be 0.50% (0.125 μg) and 1.50% (0.375 μg) respectively. Conclusion All the results reveal that the proposed method was found to be highly sensitive, simple, precise, accurate, robust, and fast. Large number of samples can be analyzed in shorter time due to shorter retention times, so it can be successfully applied for routine analysis of Sofosbuvir and related phosphoryl impurity in bulk and pharmaceutical dosage forms.
Development and validation of a stability-indicating, single HPLC method for sacubitril-valsartan and their stereoisomers and identification of forced degradation products using LC-MS/MS

Abstract:The aim of this research work was to develop and validate a stability-indicating, single reversed-phase HPLC method for the separation of five impurities, including enantiomers, diastereomers, and degradation products in sacubitril-valsartan tablets. The method was developed using a Chiralcel OJ-RH column (150 × 4.6 mm, 5 μm) at 45°C with a gradient program of (T/%B) 0.01/25, 10.0/25, 25/38, 37.0/45, 39.0/25, and 45.0/25 at a flow rate of 0.8 ml/min. Mobile phase A consisted of 1 ml of trifluoroacetic acid in 1000 ml of Milli-Q water. Mobile phase B consisted of 1 ml of trifluoroacetic acid in a mixture of acetonitrile and methanol in the ratio of 950:50 (v/v). Sacubitril, valsartan, and their five impurities were monitored at 254 nm. Degradation was not observed when sacubitril-valsartan was subjected to heat, light, hydrolytic, and oxidation conditions. In acid degradation study (1 N HCl/60°C/2 h) impurity 1 (m/z 383.44) was formed, and in base degradation study (0.1 N NaOH/40°C/1 h) impurities 1 and 5 (m/z 265.35) were formed; both impurities were confirmed using LC-MS. The degradation products, enantiomers, and diastereomers were well separated from sacubitril and valsartan, proving the stability-indicating power of the method. The developed method was validated per the International Council for Harmonisation of Technical Requirements for Pharmaceuticals for Human Use guidelines. The inter- and intra-day percentage relative standard deviation for sacubitril, valsartan, and their five impurities was less than 5.2%, recovery of the five impurities was between 93 and 105%, and linearity was ≥0.999. The limit of detection was 0.030-0.048 μg/ml, and the limit of quantification was 0.100-0.160 μg/ml.

Abstract:This article presents the development of a reversed-phase ultra-high-performance liquid chromatographic method for determining process-related impurities in ropinirole hydrochloride drug substance applying the analytical quality by design approach. The current pharmacopeial method suffers from selectivity issues due to two coelutions of two pairs of impurities. The development of a new method began with preliminary experiments, based on which the Acquity UPLC BEH C8 was selected as the most appropriate column. The effects of six different critical method parameters (CMPs) were then investigated using a fractional factorial screening design. Column temperature, the ratio of methanol in mobile phase B, and gradient slope turned out to be highly significant CMPs in achieving critical resolutions, and they were further evaluated using a central composite face-centered response-surface design. Mathematical models were created by applying a multiple linear regression method. Based on the elution order of an unknown degradation impurity and impurity C, two design spaces were established, and for each design space an optimal combination of CMPs was determined. The method developed was validated for precision, accuracy, linearity, and sensitivity, and it was proven suitable for determining nine process-related impurities of ropinirole.

Abstract:An efficient, cost effective, rapid reverse phase high-performance liquid chromatographic method was developed and validated for the determination of Ezetimibe and Simvastatin in pharmaceutical dosage forms. Chromatography was carried out by using X-terra C18, 150 x 4.6mm, 3.5μ or equivalent internal diameter with a mixture of 0.5 mL glacial acetic acid in 2000 mL water as mobile phase A and Acetonitrile as mobile phase B. Analytical method validation parameters such as specificity, linearity, precision, accuracy, solution stability and robustness, limit of detection (LOD) and limit of quantification (LOQ) was done. The correlation coefficient was found to be linear for each analyte in the desired concentration range. The average recovery was found with range of 96.7 and 107.7 for Ezetimibe and Simvastatin and their respective impurities respectively. The proposed method is highly sensitive, precise and accurate, which was evident from the LOD value range of minimum 0.135 ppm for Ezetimibe and 0.356 ppm for Lovastatin & Epilovastatin for Simvastatin impurities. Hence the present method can be applied successfully for the quantification of finished dosage form in the combined formulations of Ezetimibe and Simvastatin.
Stability‐indicating RP‐HPLC method development and validation for the quantification of amlodipine besylate and valsartan tablets in solid oral dosage form

Abstract:The present study discusses the development of simple, rapid, specific, precision, accuracy, stability indicating the HPLC method for the analysis of amlodipine besylate and valsartan tablet dosage form. The chromatographic separation was achieved using phosphate buffer with 1% triethyl amine (pH 3.0) as mobile phase‐A and mixed Methanol and buffer in the ratio of (65:35)(v/v) as mobile phase‐B. The detection of components was made at 237 nm for amlodipine besylate and valsartan. Analytical techniques should enrich sensitivity and specificity for the estimation of pharmaceutical drug products. Evaluated stress studies under different types of ICH conditions. The optimized HPLC method was validated as per the current ICH guidelines. The validated HPLC method was obtained highly specific with linearity ranging between 25 and 200 μgmL−1 of amlodipine besylate and 40–320 μgmL−1 of valsartan and both components correlation coefficient was > 0.999. The method showed high accuracy more than 97%. In stress studies, amlodipine besylate and valsartan were found to be sensitive to acid stress conditions and oxidation stress conditions. The method was found to be suitable for the quality control of amlodipine besylate and valsartan in the tablet as well as in stability‐indicating studies. The method was applied to the analysis of stability samples.

Abstract:A reverse-phase high performance liquid chromatography (RP-HPLC) method was developed and validated for simultaneous estimation of fimasartan (FIM), amlodipine besylate (AML) and rosuvastatin calcium (ROS) in combination for the treatment of hypertension with dyslipidemia presenting a major risk factor for cardiovascular diseases Chromatographic separation was performed on stationary reverse-phase inert sustain-swift C 18 100A° (250 mm × 4.6 mm, 5 μm i.d.) column. The optimized mobile phase acetonitrile – methanol – water (15:15:70, v/v, at pH 3.5 adjusted with 0.1% ortho-phosphoric acid) was pumped at a flow rate of 1.0 mL/min and the effluent spectrum was monitored at 230 nm. Calibration graphs of drugs constructed at their wavelengths of determination were linear in a concentration range of 10 – 100 μg/mL for all three components. The retention times for FIM, ROS calcium, and AML peaks at 230 nm were found to be 4.6, 6.4, and 10.8 min respectively The method suggested usefulness of a unique mobile phase during the estimation of two or more multicomponent dosage forms. Due to its simplicity, rapidness, high precision, and accuracy, the proposed RP-HPLC method can be applied for simultaneous determination of FIM, ROS and AML in bulk mixture.

Abstract:The purpose of this study is to develop an accurate, simple, and fast reverse-phase high-performance liquid chromatographic (RP-HPLC) method and validate for analyzing organic impurities in osimertinib mesylate (OSM) tablets. The developed method separates and validated for chloro impurity, hydroxy impurity, methoxy impurity, des-acrylic acid impurity, N-oxide, OSM dimer and OSM. The separation of all these impurities were achieved on inert sustain C18 (4.6×250) mm, 3 μ column, The mobile phase consists of 0.1% formic acid with pH to 6.50 adjusted with ammonium hydroxide and acetonitrile delivered in gradient elution mode. The analytes were monitored at 268 nm. Stress studies were performed to evaluate the capability of stability-indicating of this method. The OSM was observed to be stable under thermal, photolytic and neutral conditions. However, it is labile under acidic, basic and oxidative conditions. All the degradants are well resolved form the principle peak OSM. All the major degradation products were isolated and characterized through UHPLC-IMS-Q-TOF-MS. The method was validated as per International Conference on Harmonization (ICH) guidelines, and the validation included specificity, limit of detection (LoD), limit of quantification (LoQ), linearity, accuracy, precision, robustness, and solution stability for isolated impurities and OSM. The method was developed for this drug product with the purpose of properly defining the quality specification of the drug product or drug substance. This method can be incorporated in the monographs of USP, EP/BP, IP etc.
The Development and Validation of Fast and Robust Stability Indicating RP-HPLC Method for Simultaneous Estimation of Azilsartan Medoxomil and Cilnidipine in Pharmaceutical Dosage Form

Abstract:Objectives: A selective, precise and accurate RP-HPLC stability indicating assay method has been developed for the simultaneous estimation of Azilsartan medoxomil and Cilnidipine in tablet dosage form. Materials and Methods: The efficient chromatographic separation of drug was achieved by using C18 (150mm×4.6mm, Agilent 5μm) Column at ambient temperature. Mobile phase contains triethylamine buffer (pH 3.5 adjusted with ortho-phosphoric acid) and acetonitrile (40:60 V/V). Flow rate of mobile phase 1.0 ml/min using isocratic mode .Wavelength selected at 249 nm by using photo diode array detector. Results: The retention time of Azilsartan medoxomil peak 1, Azilsartan medoxomil peak 2 and Cilnidipine were noticed to be 2.16 min, 3.90 min and 9.52 min respectively. The linearity range for Azilsartan medoxomil and Cilnidipine were found to be 50 -150 μg/ml and 12.5-37.5 μg/ml and percent recoveries were noticed to be 99.27±0.58 and 98.65±0.49 respectively. Various stress testing conditions such applied to the drug ingredients and drug formulation. The degradants and drugs efficiently separated by using enhanced chromatographic conditions. The developed method was validated as per recommendation parameters of International council on harmonization guideline Q2(R1). Conclusion: The validation parameters stated that the drug substances were efficiently separated from its degradants and developed method can be routinely applied for the simultaneous estimation of Azilsartan medoxomil and Cilnidipine in tablet formulation in a laboratory.

Abstract:Combination of sacubitril, a novel neprilysin inhibitor, with valsartan, an angiotensin receptor blocker, was approved by FDA as first line treatment of chronic heart failure resulting from hypertension. A selective, rapid, accurate and precise stability indicating RP-HPLC method was developed for the estimation of sacubitril and valsartan in the presence of their impurities in bulk powder and pharmaceutical dosage form. Experimental design using face-centered composite design was applied during method optimization to achieve best resolution with minimum experimental trials. The two independent variables were buffer pH and organic modifier volume in the mobile phase. Chromatographic separation was accomplished using a Zorbax Eclipse C18 Cyano column (150 mm, 4.6 mm, 5 µm) and a mobile phase composed of ammonium acetate buffer (0.02 M, adjusted to pH 4 with acetic acid): acetonitrile (55:45, v/v) at a flow rate 1 mL min−1. Detection was performed at 254 nm. Sharp and well-resolved peaks for sacubitril and valsartan were obtained with retention times 4.91 and 2.53 min, respectively. A complete forced degradation of sacubitril was achieved and degradation kinetics under basic conditions was studied and was found to follow first-order kinetics. Structure elucidation was performed using mass spectrometry. The method was fully validated in terms of selectivity, linearity, accuracy, precision and robustness in agreement with ICH guidelines Q2 (R1).

Abstract:The research focus of this work was development and validation of an efficient analytical method that can be used for separation and determination of related substances of bisoprolol in finished drug product using reversed-phase HPLC with UV-DAD. In a previous systematic study of various stationary phases and elution conditions it was found that several octadecylsilane phases can be successfully employed for separation of the critical pairs of bisoprolol and its related degradation products. Namely, method development led to conclusions that satisfactory resolution and peak shapes were obtained with gradient elution with water with 0.2% perchloric acid and acetonitrile and the reversed-phase columns: Hypersil 3 BDS C18 (100 x 4 mm, 3 m); Zorbax SB C18 (150 x 4.6 mm, 3.5 m); Acquity UPLC BEH C18 (50 x 2.1 mm, 1.7 m), and Xterra MS C18 (100 x 4.6 mm, 3.5 m). Since regular quality control includes the parameter related substances, which is of great importance for the overall quality of dosage forms, this study was oriented towards widening the available validated analytical methods for determination of this parameter. In this work, the results from the validation of the method for determination of related substances of bisoprolol using Zorbax SB C18 150 x 4.6 mm, 3.5 m, are presented. Validation parameters that were tested (linearity, accuracy, precision, selectivity) confirmed that the method is suitable for its intendance and it was used for characterization of the samples from a forced degradation study of bisoprolol tablets. Determination of Rosuvastatin Concentration in Human Plasma by LC-MS Method

Abstract:OBJECTIVE:To establish the method for the LC-MS determination of rosuvastatin concentration in human plasma.METHODS:After liquid-liquid extraction with aether-dichloromethane(3∶2),LC-MS method was adopted.The determination were separated on a Shim-Pack VP-ODS column with the mobile phase consisted of methanol-5 mmol·L~(-1)ammonium acetate solution(containing 0.5% formic acid)(65∶35)at a flow rate of 0.3 mL·min-1.Detection was carried out by electrospray positive ionization mass spectrometry in the multiple reaction monitoring(MRM)mode.The MRM transitions of m/z 482.2→258.2 and m/z 748.5→158.4 were used to quantify rosuvastatin and I.S.,respectively.RESULTS:Rosuvastatin and I.S.were eluted for 3.49 min and 2.70 min,respectively.The calibration curve was linear in the range of 0.050 0 ng·mL-1～30.0 ng·mL~(-1)(r=0.996 2)with the lower limit of quantitation(LLOQ)of 0.050 0 ng·mL~(-1).Intra-day and inter-day RSD were both less than 11%.Mean extraction recoveries were(89.7±3.6)%.In the stability studies,rosuvastatin in plasma was found to be stable under various storage conditions.CONCLUSION:It is a rapid,sensitive,selective and reliable method for the determination of rosuvastatin concentration in human plasma.The method is suitable for bioequivalence study of rosuvastatin calcium preparations in human subjects.

Abstract:Metabolic syndrome is an associated condition that occurs together and increases the risk of heart disease and diabetes. These conditions include high blood pressure, high blood sugar, and high body mass index (BMI) in terms of cholesterol and triglyceride levels. Most of the elderly population may administer three drugs to control the above conditions. Therefore, this study aims to develop an analytical assay for the precise analysis of three components and to formulate a Self-Nanoemulsifying Drug-Delivery System (SNEDDS) loaded with three drugs: Rosuvastatin Calcium (RC; antilipidemic), Glibenclamide (GB; antidiabetic), and Candesartan Cilexetil (CC; antihypertensive). A design of the experiment was developed at a level of 32, and the influence of column temperature and flow rate was studied in terms of retention time, peak area, peak asymmetry, and resolution. The assay was subjected to several studies to ensure its validation. Under the optimized conditions—column temperature at 50 °C and flow rate at 0.25 mL/min—the three drugs, RC, GB, and CC, are separated. Their retention times are 0.840, 1.800, and 5.803 min, respectively. The assay was valid in terms of linearity, accuracy, and precision. Moreover, the developed assay shows a good tolerance against any change in the condition. The assay was tested also to separate the drugs in a pharmaceutical formulation as SNEDDs. The assay successfully separates the drug with a good resolution.

Abstract:A fast, simple, accurate and robust reversed phase HPLC method for the simultaneous determination of two flavonoids, hesperidin and diosmin, in combined tablets was developed and validated. This method uses a short C8 column with dimensions of 75 mm × 4 mm with 5 μm particles thermostated at 30 °C, and a mobile phase composed of formic acid (pH 4.1 and 0.05%, V/V) and methanol (58:42, V/V), delivered at a flow rate of 1.2 ml/min, with UV detector signal monitoring at 280 nm and an injection volume of 5 μl. These chromatographic conditions yielded chromatograms with symmetric peaks of hesperidin, eluting at a ~2 min retention time, and diosmin, at a ~4.5 min retention time, with a total run time cycle of 6 min. The method validation parameters confirmed excellent values for accuracy, linearity and reproducibility. This method is suitable for routine analysis in pharmaceutical and food quality control laboratories.

Abstract:New, fast, simple and mild conditioned High Performance Liquid Chromatography (HPLC) method for determination of atorvastatin and its 7 main specified impurities, as well as unspecified impurities that might possibly appear, was developed. Chromatographic runs last between 25 and 40 minutes, with simple stepwise gradient elution. The main accent in our method development strategy was focused on mobile phase, composed of simple binary system composed of phosphate buffer and acetonitrile, at pH 4.1, without use of tetrahydrofuran, ion-pair reagents, trifluoroacetic acid and other modifiers with high Ultraviolet (UV) cut-off like absorptive acetate or formiate buffers or amines. With our concept of mobile phase, different columns from myriad were tested, with different efficiency, dimensions and properties, which resulted in different separation efficiency and run time. The best results, concerning essential critical peak resolution, run time length including column preparation and equilibration and column backpressure, were achieved with: YMC C18 Triart 150mm x 4.6mm, 3μm (YMC America, Inc.), afterwards with Nucleodur 100-3-C18ec 250mm x 4.6mm, 3μm (Macherey-Nagel GmbH & Co., Germany), Waters Symmetry C18 250mm x 4.6mm, 5μm (Waters, USA) and Superspher C18e 125mm x 4mm, 4μm (Merck, Darmstadt, Germany). All this columns achieve excellent results regarding obligated critical resolution between atorvastatin impurity B and atorvastatin (according to European Pharmacopoeia),1 or in some cases between atorvastatin impurity B and atorvastatin impurity C, to be minimum about 1.5, in both cases.
