Abstract:Edible and medicinal fungal polysaccharides with excellent biological activity and high viscosity are potential thickeners for food applications. In this study, the rheological properties of aqueous solutions of exopoly-saccharide or fermentation broth polysaccharide (SCFP) and intracellular polysaccharide or mycelial poly-saccharide (SCMP) obtained from liquid submerged fermentation of the fungus Schizophyllum commune (S. commune), and their thickening effects on coffee and fruit tea were investigated. The polysaccharide aqueous solutions behaved as pseudoplastic fluids, and their viscosity was concentration-dependent but not significantly affected by temperature (10-70 degrees C). The critical concentrations (c*) of SCFP and SCMP were 0.275 g/L and 0.356 g/L, respectively. The intensity of the thixotropic properties of the polysaccharide aqueous solutions was positively correlated with polysaccharide concentration. Both polysaccharide solutions exhibited solid-like viscoelastic behavior and formed weak gels, with enhanced gel strength at high concentrations and low temperatures. The viscosity at a shear rate of 50 s(-1) (eta(50)) of the 5 g/L SCFP- and SCMP-thickened coffee was 0.07 Pa.s, while that of SCFP- and SCMP-thickened fruit tea was 0.1 Pa.s and 0.08 Pa.s, respectively, which were significantly higher than that of polysaccharide aqueous solutions (0.05 Pa.s for SCFP and SCMP). The yield stress values increased with increasing polysaccharide concentration, suggesting a potential role of for these polysaccharides in dysphagia management. Overall, SCFP and SCMP have significant potential for application in the food industry.

Abstract:OBJECT The goal of this study was to find which central nervous system (CNS) pathways are involved in volitional control over reinnervated biceps or pectoral muscles. METHODS Intercostal nerves (ICNs) were coapted to the musculocutaneous nerve (MCN) or the medial pectoral nerve (MPN) in 23 patients with root avulsions of the brachial plexus to restore biceps or pectoral muscle function. The facilitatory effects of respiration and voluntary contraction on cortical motor-evoked potentials of biceps or pectoral muscles were used to study CNS control over the reinnervated muscles. The time course of the facilitatory effect of respiration and voluntary contraction differed significantly. In the end stage of nerve regeneration, the facilitatory effect of voluntary contraction was significantly larger than that of respiration, indicating that the CNS control network over the muscle comes to resemble that of the recipient nerve (MCN or MPN) rather than that of the donor nerve (ICN). CONCLUSIONS The strengthening of previously subthreshold synaptic connections in a CNS network connecting ICN to MCN or MPN neurons may underlie changing excitability.
Physicochemical and rheological properties of Tremella fuciformis polysaccharide fractions by ethanol precipitation

Abstract:Four Tremella fuciformis polysaccharides (TPS) fractions named as TPS20, TPS40, TPS60, and TPS80 were obtained by stepwise ethanol precipitation. Their physicochemical and rheological properties were investigated. Results showed TPS60 had the highest contents of polysaccharide (81.12%), protein (0.29%), uronic acid (14.36%), and sulfate (5.92%). Their molecular weights were in a descending order of TPS20, TPS40, TPS60, and TPS80. In a certain range of shear rate, they all presented pseudoplastic fluid and viscosity characteristic at low frequency. Increasing temperature or concentration of salts decreased the viscosity of their solutions. Sucrose concentration less than 6% led to an increasing viscosity of TPS20, TPS40, and TPS60. Both strong acidity and alkalinity could reduce the viscosity of all TPS solutions. TPS80 solution showed the best stability and its viscosity was minimally affected by pH, temperature, salts, and sucrose. These results can provide theoretical references for utilizations of TPS fractions in foods, pharmaceuticals, and cosmetics.

Abstract:The formation of a single soybean protein isolate (SPI) gel is limited by the processing conditions, and has the disadvantages of poor gel property, and it is usually necessary to add other biomacromolecules to improve its property. In this study, we investigated the effects of polysaccharide concentration on gel properties and interaction mechanisms of Tremella fuciformis polysaccharide (TFP)-SPI complexes. It was found that (1) the rheological properties, texture properties, water-holding properties, and thermal stability of TFP-SPI composite gels were improved with the addition of TFP (0.25–2.0 %, w / v ) in a concentration-dependent manner; (2) hydrogen bond, the electrostatic interaction, hydrophobic interaction, and disulfide bond in the gel system increased with the increase of TFP concentration; (3) the electrostatic and hydrophobic interactions played an important role in the formation of the TFP-SPI composite gel while hydrogen bond formation was the least contributor to the binary composite gel network. Overall, TFP is not only a critical health food but also a promising structural component for improving the gel properties of SPI.

Abstract:In this paper, emulsions stabilized by Tremella fuciformis polysaccharides (TFP) were prepared and the physiochemical properties were assessed. Results showed that the TFP emulsions illustrated the highest emulsifying activity (EAI) and emulsifying stability (ESI) when the concentration of TFP and oil were 0.8% and 10% (wt%). The higher pH value was in favor of the emulsifying properties, while the addition of NaCl impaired the stability, and the greater the concentration, the lower the EAI and ESI. Besides, the emulsifying and rheological properties and stability analysis were evaluated in comparison with gum arabic, pectin, and carboxymethyl cellulose emulsions. It was discovered that TFP illustrated better storage and freeze-thaw stability, which was proved by the result of zeta-potential and particle size. The rheological measurement revealed that all the emulsions behaved as pseudoplastic fluids, while TFP displayed a higher viscosity. Meanwhile, TFP emulsions tended to form a more stable network structure according to the analysis of the parameters obtained from the Herschel–Bulkley model. FTIR spectra suggested that the O-H bond could be destructed without the formation of new covalent bonds during the emulsion preparation. Therefore, this study would be of great importance for the research of emulsions stabilized by TFP as a natural food emulsifier.

Abstract:The evolution of the starch fine structure during growth and its impact on the gelatinization behavior of cassava starch (CS) was investigated by isolating starch from South China 6068 (SC6068) cassava harvested from the 4th to 9th growth period. During growth, the short-range ordered structure, crystallinity as well as particle size distribution of starch were increased. Meanwhile, the starch molecular size and amylopectin (AP) proportion increased, while the proportion of amylose (AM) exhibited a decreasing tendency. The chains of short-AM (X - 100-1000) were mainly significantly reduced, whereas the short and medium-AP chains (X - 6-24) had the most increment in AP. The solubility, thermal stability, shear resistance, and retrogradation resistance of starch were enhanced after gelatinized under the influence of the results mentioned above. This study presented a deeper insight into the variation of starch fine structure during growth and its influence on gelatinization behavior, which would provide a theoretical basis for starch industrial applications.

Abstract:In the food industry, there has been a debate about the interactions between fish gelatin and polysaccharide with the interaction mechanism in different solute and solvent systems still unclear. This article tries to explore interactions between fish gelatin (FG) and Tremella polysaccharides (AP) by the turbidity titration. Especially, the turbidity was greatly influenced by pH, polymer ratio, total polymer concentration, and ionic strength. Optimally, the most complex coacervates could be electrostatically formed at a mass ratio of 1:1 and pH 4.0, which was collected for subsequent characterization. In addition, there was a positive correlation between the turbidity and the total concentration, while the ionic strength was just reverse. Modern instruments including FTIR, XRD, DSC, SEM and rotational rheometer were adopted to characterize the complex coacervates formed at different pH. Results showed that the FG–AP complex coacervates showed porous network structure and had the strongest apparent viscosity with viscoelastic modulus at pH 3.5. These results suggested important implications in how we make a good alternative for fat in food dairy.

Abstract:Different methods were used to degrade Tremella fuciformis polysaccharides (TFP) and prepare low molecular weight polysaccharides of Tremella fuciformis (TFLP) to improve their bioavailability. It was found that the TFLP prepared by ultrasonic-assisted H 2 O 2 -Vc method showed the highest level of antioxidant activity and stress resistance in C. elegans . The structural characteristics, in vivo antioxidant and stress resistance of TFLP-1 were evaluated after isolation and purification of TFLP, it was found that TFLP-1 was an acid polysaccharide with a molecular weight of 75770 Da, which mainly composed of mannose. Meanwhile, it could regulate the antioxidant activity and stress resistance in C. elegans by upregulating the transcription of fat-5 , fat-7 , acs-2 , glp-1 , hsf-1 , hsp-1 , mtl-1 , nhr-49 , skn-1 and sod-3 mRNA. The improvement effects were closely related to the significant regulation of galactose metabolism, alpha linolenic acid metabolism, and pantothenate and CoA biosynthesis metabolic pathways. These results provided insights into the high value application of Tremella fuciformis in the food industry and the development of antioxidant related functional foods.

Effects of ultrasonic-enzymatic-assisted ethanol precipitation method on the physicochemical characteristics, antioxidant and hypoglycemic activities of Tremella fuciformis polysaccharides

Abstract:This investigation involved the extraction of a novel polysaccharide from the spore fermentation broth of Tremella fuciformis using a method that combined ultrasonic and enzymatic assistance with ethanol precipitation. It was then compared with enzymatic and ultrasonic extraction methods. The objective of this research is to offer a reference point for expanding the application of ultrasonic-assisted enzymatic extraction technology in T. fuciformis polysaccharides (TFPs). Based on single-factor experiments, Box-Behnken was used to optimize the extraction conditions of TFPs by ultrasonic-enzymatic-assisted ethanol precipitation extraction. The results revealed an optimal combination of enzymes, with a cellulase-to-papain ratio of 2:1, an enzyme addition of 4000U/100 mL, an enzymolysis temperature of 49 °C, ultrasonicpower at 3 W/mL and an ultrasonictime of 20 min. The extraction rate of TFPs and α- amylase inhibition rates were 23.94 % and 61.44 %, respectively. Comparing the physicochemical properties, structural characterization and in vitro activity of TFPs extracted through different methods, the results showed that ultrasonic treatment significantly influences the apparent morphology of polysaccharide and could enhance its in vitro biological activity. However, different extraction techniques exhibit insubstantial impact on the chemical composition, glycosidic bonds or glycosidic ring configurations within the polysaccharides. Among them, ultrasonic-enzymatic-assisted ethanol precipitation extraction of polysaccharide has the highest extraction rate and the lowest viscosity. It has significant effects on ABTS+ scavenging activity, α- amylase inhibition rate and glucose dialysis retardation index, polysaccharide treated with ultrasonic-enzymatic showed the best performance. These findings suggest that ultrasonic-enzymatic-assisted ethanol precipitation extraction can enhance the activities of TFPs, thereby providing a valuable insight for their future development and application.

Abstract:Polysaccharides are used in starch-based product formulations to enhance the final quality of food products. This study examined the interaction mechanisms in Ficus pumila polysaccharide (FPP) and wheat starch (WS) gel systems with varying FPP concentrations using linear and nonlinear rheological analysis. Physicochemical structural analyses showed non-covalent FPP-WS interactions, strengthening hydrogen bonding between molecules and promoting water binding and ordered structure generation during WS gel aging. Small amplitude oscillatory shear analyses revealed that elevated FPP concentrations led to increased storage modulus (G'), loss modulus (G"), critical strains (From 29.02 % to 53.32 %) and yield stresses (From 0.94 Pa to 30.97 Pa) in the WS gel system, along with improved resistance to deformation and short-term regeneration. In the nonlinear viscoelastic region, FPP-WS gels shifted from elastic to viscous behavior. Higher FPP concentrations displayed increased energy dissipation, strain hardening (S>0, e 3 /e 1 > 0) and shear thinning (T<0, v 3 /v 1 <0). FPP contributes more nonlinearity in the dynamic flow field as showed by the high harmonic ratio, with a larger I 3 /I 1 values overall. This study highlights FPP's potential in starch gel food processing, and offers a theoretical basis for understanding hydrocolloid-starch interactions.

Polysaccharide from Clitocybe squamulosa: Gel-forming properties and its compound effect with food thickener

Abstract:To study the gel-forming properties of polysaccharide from the fruiting body of Clitocybe squamulosa (CSFP) and its degradation product (UH-CSFP), the changes in steady-state and dynamic rheological properties of CSFP and UH-CSFP under different conditions (polysaccharide mass fraction, temperature, pH, and salt ion concentration) were studied. Polysaccharides with good gel-forming properties were selected and mixed with common edible thickeners (gelatin, guar gum, and locust bean gum), after which the properties of the composite gel were assessed. The steady-state rheological results showed that CSFP and UH-CSFP were pseudoplastic fluids, their apparent viscosity decreased with increasing temperature, the viscosity was greatest when the pH was 7. The addition of Na+ and Ca2+ could increase the viscosity, and the viscosity of UH-CSFP was lower than that of CSFP at the same mass fraction. The results of dynamic rheology indicated that G´ and G´´ of CSFP and UH-CSFP increased with increasing mass fraction, pH, and ion concentration (0.01 M to 1 M), and G´´ was always smaller than G´ indicating weak gel behavior. The thixotropy-related experimental results showed that the thixotropy ring area of CSFP and UH-CSFP increased with increasing mass fraction, the ring area of CSFP was larger than that of UH-CSFP, and the gel strength of CSFP was greater than that of UH-CSFP. The results of CSFP and three types of edible gels showed that the composite gels were pseudoplastic fluids, and their apparent viscosity was ranked (in descending order) as follows: guar bean gum, locust bean gum, and gelatin. The addition of CSFP improved the gel-forming properties of guar gum but did not significantly improve the gel properties of locust bean gum and gelatin. This study provides a theoretical basis for the selection of processing methods and the application of polysaccharides.

Abstract:Polysaccharides from Tremella spp. are known to have various functions and applications. To fully understand the polysaccharide compositions of fruiting body of T. fuciformis, cold water extract (CWE), hot water extract (HWE), dilute alkaline extract (ALE), and dilute acid extract (ACE) were sequentially prepared and characterized. Their physicochemical properties, chemical composition, micromorphology, and functional properties were determined. Firstly, the four extracts showed significant difference in chemical compositions. Secondly, in the structural aspect, liquid chromatographic elution curves of the extracts had only one main peak except that CWE had two peaks. Fucose, galactose (or glucose), xylose, and mannose were detected in CWE and HWE. Differently, ALE had fucose, glucose, and xylose, and ACE had only galactose and glucose, which agreed with the information obtained from Fourier transform infrared spectra. Thirdly, in the physicochemical and nutritional aspects, the thermal stability of the four extracts decreased in the order of: ACE>HWE≈ALE>CWE. HWE and ALE had similar dynamic viscoelasticity, exhibiting viscous characteristics at low frequencies and elastic characteristics at higher vibration frequencies. The four extracts were digestive-resistant after in vitro mimic experiments. This study could provide some theoretical data support for the scientific research and its application in food, medicine, and cosmetics of tremella polysaccharides.

Abstract:In this study, we have investigated the effects of Tremella fuciformis polysaccharide (TP) on the pasting, rheological, structural and in vitro digestive properties of Cyperus esculentus starch (CS). The results showed that the addition of TP significantly changed the pasting characteristics of CS, increased the pasting temperature and pasting viscosity, inhibited pasting, reduced the exudation of straight-chain starch and was positively correlated with the amount of TP added. The addition of the appropriate amount of TP could increase its apparent viscosity and enhance its viscoelasticity. The composite system of CS/TP exhibited higher short-range ordered structure and solid dense structure, which protected the crystal structure of CS, but was related to the amount of TP added. In addition, the introduction of TP not only decreased the in vitro digestion rate of CS and increased the content of slow-digestible starch (SDS) and resistant starch (RS), but also reduced the degree of digestion. Correlation studies established that TP could improve the viscoelasticity, relative crystallinity and short-range order of the CS/TP composite gel, maintain the integrity of the starch granule and crystalline structure, reduce the degree of starch pasting and strengthen the gel network structure of CS, which could help to lower the digestibility of CS.

Abstract:A water-soluble polysaccharide was extracted from Tremella fuciformis Berk. Crude Tremella fuciformis polysaccharide was gained by boiling in hot-water, precipitating using ethanol, removing proteins with papain-Sevage method. Then the crude polysaccharide was applied to DEAE-52 cellulose colume and further purified through Sephadex s-400 column, followed by dialysis, lyopilization. The resulting sample was named as TFP1. Total sugar and uronic acid contents of TFP1 were determined by phenol-sulfuric acid method and m-hydroxydiphenyl combined with sulfuric acid method using glucose and glucuronic acid as standard respectively. UV spectrum showed that no protein or nucleic acid presented in TFP1. The results of the PMP precolumn derivatization HPLC analysis and IR spectroscopy showed that TFP1 was an acidic heteropolysaccharide containing mannose, glucuronic acid, xylose and fucose with the molar ratio of 5.78∶0.7∶1.1∶1. I2-KI assay suggested that many branched chains attached to the backbone chain which was composed of mannose residues.

Abstract:The sol-gel behavior of tapioca starch (TS) plays a crucial role in the processing and quality control of flour-based products. However, natural tapioca starch has low gel strength and poor viscosity, which tremendously limits its application. To solve this problem, this study investigated the effects of κ-carrageenan (KC), konjac gum (KGM), and Mesona chinensis Benth polysaccharide (MCP) on the pasting behavior, rheological, and structural properties of tapioca starch. KC, KGM, and MCP significantly increased the viscosity of TS. With the exception of high-concentration KGM (0.5 %), all other blending systems showed decrease in setback. This may be attributed to the stronger effect of the high-concentration KC (0.5 %) and MCP (0.5 %) functional groups interacting with starch. KC, KGM, and MCP effectively improved the dynamic modulus (G' and G") of TS gel and were effective in increasing the gel strength and hardness of TS. The FT-IR analysis indicated that the short-range order of TS was mainly influenced by polysaccharides through non-covalent bonding interactions. Furthermore, it was confirmed that three polysaccharides could form dense structures by hydrogen bonding with TS. Similarly, more stable structure and pore size were observed in the microstructure diagram.

Abstract:To investigate the composition, structure, and physical and chemical properties of Tremella fuciformis polysaccharide (TFPS), four new polysaccharide fractions (TFPS1, TFPS2, TFPS3, and TFPS4) were successfully isolated from hot water-extracted crude polysaccharides using a DEAE-52 cellulose column. Their structures were analyzed using FT-IR, SEM, HPGPC, monosaccharide composition, Congo red and I 2 -KI experiments. TFPS1 was found to be a heteropolysaccharide with a molecular weight of 742 Da for the major polysaccharide fragment. TFPS2, TFPS3, and TFPS4 were identified as single-component homogeneous polysaccharides with molecular weights of 350774 Da, 622675 Da, and 624724 Da, respectively. Glucose was the dominant monosaccharide of TFPS1 and TFPS2, while mannose was the major monosaccharide of TFPS3 and TFPS4. The results showed that TFPS1 and TFPS2 displayed a stable triple helix structure in aqueous solution, while TFPS3 and TFPS4 had longer side chains and more branched chains. Additionally, all four TFPSs showed in vitro antioxidant activity, as well as significant cholesterol binding and sodium taurocholate binding abilities.

Abstract:The chemical structures of the polysaccharides from fruit body, spore, and submerged culture of T. fuciformis are similar. T. fuciformis polysaccharides mainly consist of heteropolysaccharides with α-(1→3)-linked D-mannan as the backbone chain, exhibiting diverse activities such as immunomodulation, antitumor, antioxidation, and antiaging. The present paper reviews the structural characteristics, various activities and structure -activity correlation of T. fuciformis polysaccharides.
