Abstract:Polysaccharides from Tremella spp. are known to have various functions and applications. To fully understand the polysaccharide compositions of fruiting body of T. fuciformis, cold water extract (CWE), hot water extract (HWE), dilute alkaline extract (ALE), and dilute acid extract (ACE) were sequentially prepared and characterized. Their physicochemical properties, chemical composition, micromorphology, and functional properties were determined. Firstly, the four extracts showed significant difference in chemical compositions. Secondly, in the structural aspect, liquid chromatographic elution curves of the extracts had only one main peak except that CWE had two peaks. Fucose, galactose (or glucose), xylose, and mannose were detected in CWE and HWE. Differently, ALE had fucose, glucose, and xylose, and ACE had only galactose and glucose, which agreed with the information obtained from Fourier transform infrared spectra. Thirdly, in the physicochemical and nutritional aspects, the thermal stability of the four extracts decreased in the order of: ACE>HWE≈ALE>CWE. HWE and ALE had similar dynamic viscoelasticity, exhibiting viscous characteristics at low frequencies and elastic characteristics at higher vibration frequencies. The four extracts were digestive-resistant after in vitro mimic experiments. This study could provide some theoretical data support for the scientific research and its application in food, medicine, and cosmetics of tremella polysaccharides.

Abstract:A water-soluble polysaccharide was extracted from Tremella fuciformis Berk. Crude Tremella fuciformis polysaccharide was gained by boiling in hot-water, precipitating using ethanol, removing proteins with papain-Sevage method. Then the crude polysaccharide was applied to DEAE-52 cellulose colume and further purified through Sephadex s-400 column, followed by dialysis, lyopilization. The resulting sample was named as TFP1. Total sugar and uronic acid contents of TFP1 were determined by phenol-sulfuric acid method and m-hydroxydiphenyl combined with sulfuric acid method using glucose and glucuronic acid as standard respectively. UV spectrum showed that no protein or nucleic acid presented in TFP1. The results of the PMP precolumn derivatization HPLC analysis and IR spectroscopy showed that TFP1 was an acidic heteropolysaccharide containing mannose, glucuronic acid, xylose and fucose with the molar ratio of 5.78∶0.7∶1.1∶1. I2-KI assay suggested that many branched chains attached to the backbone chain which was composed of mannose residues.

Abstract:Different methods were used to degrade Tremella fuciformis polysaccharides (TFP) and prepare low molecular weight polysaccharides of Tremella fuciformis (TFLP) to improve their bioavailability. It was found that the TFLP prepared by ultrasonic-assisted H 2 O 2 -Vc method showed the highest level of antioxidant activity and stress resistance in C. elegans . The structural characteristics, in vivo antioxidant and stress resistance of TFLP-1 were evaluated after isolation and purification of TFLP, it was found that TFLP-1 was an acid polysaccharide with a molecular weight of 75770 Da, which mainly composed of mannose. Meanwhile, it could regulate the antioxidant activity and stress resistance in C. elegans by upregulating the transcription of fat-5 , fat-7 , acs-2 , glp-1 , hsf-1 , hsp-1 , mtl-1 , nhr-49 , skn-1 and sod-3 mRNA. The improvement effects were closely related to the significant regulation of galactose metabolism, alpha linolenic acid metabolism, and pantothenate and CoA biosynthesis metabolic pathways. These results provided insights into the high value application of Tremella fuciformis in the food industry and the development of antioxidant related functional foods.

Abstract:To investigate the composition, structure, and physical and chemical properties of Tremella fuciformis polysaccharide (TFPS), four new polysaccharide fractions (TFPS1, TFPS2, TFPS3, and TFPS4) were successfully isolated from hot water-extracted crude polysaccharides using a DEAE-52 cellulose column. Their structures were analyzed using FT-IR, SEM, HPGPC, monosaccharide composition, Congo red and I 2 -KI experiments. TFPS1 was found to be a heteropolysaccharide with a molecular weight of 742 Da for the major polysaccharide fragment. TFPS2, TFPS3, and TFPS4 were identified as single-component homogeneous polysaccharides with molecular weights of 350774 Da, 622675 Da, and 624724 Da, respectively. Glucose was the dominant monosaccharide of TFPS1 and TFPS2, while mannose was the major monosaccharide of TFPS3 and TFPS4. The results showed that TFPS1 and TFPS2 displayed a stable triple helix structure in aqueous solution, while TFPS3 and TFPS4 had longer side chains and more branched chains. Additionally, all four TFPSs showed in vitro antioxidant activity, as well as significant cholesterol binding and sodium taurocholate binding abilities.

Effects of ultrasonic-enzymatic-assisted ethanol precipitation method on the physicochemical characteristics, antioxidant and hypoglycemic activities of Tremella fuciformis polysaccharides

Abstract:This investigation involved the extraction of a novel polysaccharide from the spore fermentation broth of Tremella fuciformis using a method that combined ultrasonic and enzymatic assistance with ethanol precipitation. It was then compared with enzymatic and ultrasonic extraction methods. The objective of this research is to offer a reference point for expanding the application of ultrasonic-assisted enzymatic extraction technology in T. fuciformis polysaccharides (TFPs). Based on single-factor experiments, Box-Behnken was used to optimize the extraction conditions of TFPs by ultrasonic-enzymatic-assisted ethanol precipitation extraction. The results revealed an optimal combination of enzymes, with a cellulase-to-papain ratio of 2:1, an enzyme addition of 4000U/100 mL, an enzymolysis temperature of 49 °C, ultrasonicpower at 3 W/mL and an ultrasonictime of 20 min. The extraction rate of TFPs and α- amylase inhibition rates were 23.94 % and 61.44 %, respectively. Comparing the physicochemical properties, structural characterization and in vitro activity of TFPs extracted through different methods, the results showed that ultrasonic treatment significantly influences the apparent morphology of polysaccharide and could enhance its in vitro biological activity. However, different extraction techniques exhibit insubstantial impact on the chemical composition, glycosidic bonds or glycosidic ring configurations within the polysaccharides. Among them, ultrasonic-enzymatic-assisted ethanol precipitation extraction of polysaccharide has the highest extraction rate and the lowest viscosity. It has significant effects on ABTS+ scavenging activity, α- amylase inhibition rate and glucose dialysis retardation index, polysaccharide treated with ultrasonic-enzymatic showed the best performance. These findings suggest that ultrasonic-enzymatic-assisted ethanol precipitation extraction can enhance the activities of TFPs, thereby providing a valuable insight for their future development and application.

Abstract:Tremella sanguinea is a traditional Chinese medicinal and edible mushroom. Polysaccharides from Tremella mushrooms have received increasing amounts of research attention due to their diverse pharmacological activities. In this study, via the incubation of basidiospores collected from fresh artificially cultivated basidiocarps of T. sanguinea, a haploid yeast strain of T. sanguinea was obtained, and it was found to be a typical loose-slime-forming yeast capable of producing a large amount of exopolysaccharides (EPS). Using DEAE-52 cellulose column chromatography and Sephadex G-100 gel permeation chromatography, the major polysaccharide, named TSPS-1, was separated and purified from the EPS produced by the haploid yeast strain of T. sanguinea. TSPS-1 was a homogeneous polysaccharide with a molecular weight of 2.5 × 103 kDa and consisted of rhamnose, glucose, xylose, mannose and glucuronic acid at a molar ratio of 1: 0.7: 62.2: 24.6: 11.5. The bioactivity of the TSPS-1 polysaccharide was evaluated. The results show that TSPS-1 exhibited noticeable antioxidant activity by scavenging hydroxyl radicals (EC50 = 1.92 mg/mL) and superoxide radicals (EC50 = 1.33 mg/mL), and prebiotic activity by promoting the growth of different probiotic strains in the genus Lactobacillus and Bifidobacterium. These results suggest that the cultivation of the haploid yeast strain can be a promising alternative for the efficient production of valuable T. sanguinea polysaccharides with antioxidant and prebiotic potential.

Abstract:OBJECT The goal of this study was to find which central nervous system (CNS) pathways are involved in volitional control over reinnervated biceps or pectoral muscles. METHODS Intercostal nerves (ICNs) were coapted to the musculocutaneous nerve (MCN) or the medial pectoral nerve (MPN) in 23 patients with root avulsions of the brachial plexus to restore biceps or pectoral muscle function. The facilitatory effects of respiration and voluntary contraction on cortical motor-evoked potentials of biceps or pectoral muscles were used to study CNS control over the reinnervated muscles. The time course of the facilitatory effect of respiration and voluntary contraction differed significantly. In the end stage of nerve regeneration, the facilitatory effect of voluntary contraction was significantly larger than that of respiration, indicating that the CNS control network over the muscle comes to resemble that of the recipient nerve (MCN or MPN) rather than that of the donor nerve (ICN). CONCLUSIONS The strengthening of previously subthreshold synaptic connections in a CNS network connecting ICN to MCN or MPN neurons may underlie changing excitability.

Abstract:Sixteen oligosaccharide monomers with the degree of polymerization 3 to 18 (DP 3 to DP 18) and three active fractions (DP 3-9, DP 8-11, and DP 11-17) were separated from Atractylodes lancea (Thunb.) DC. by optimized fast protein liquid chromatography coupled with refractive index detector (FPLC-RID) and preparation hydrophilic interaction chromatography (Pre-HILIC). Gas chromatography-mass spectrometer (GC-MS), liquid chromatography tandem mass spectrometry (LC-MS/MS), nuclear magnetic resonance (NMR) spectroscopy, and methylation analysis showed that the oligosaccharide in A. lancea was 1-kestose [β-D-fructofuranosyl-(2 → 1)-β-D-fructofuranosyl-(2 → 1)-α-D-glucopyranoside] (inulin-type fructooligosaccharides, FOS). Particularly, DP 3-9 showed the best capacity in stimulating phagocytic, NO, and cytokines production on RAW264.7 cells than any other purified oligosaccharide monomers and active fractions. It could also activate T-cells in Peyer's patch cells and enhance the production of colony stimulation factors. Besides, FPLC-RID showed a good capacity for large-scale preparation of DP 3-9 with the recovery of more than 93%. The bioactivity of sixteen FOS monomers (DP 3 to DP 18) and three FOS fractions (DP 3-9, DP 8-11, and DP 11-17) investigated in this study are beneficial for the utilization of FOS as a functional ingredient in novel product development.

Abstract:The edible fungus Tremella fuciformis was shown to have a high molecular weight (1.87 × 103 kDa) bioactive polysaccharide, denoted as TFP-F1. Monosaccharide composition and NMR analysis of the polysaccharide and its derivatives indicated it contained fucose (Fucp), xylose (Xylp), mannose (Manp), and glucuronic acid (GlcAp) in a ratio of 0.9:1.0:3.2:1.2. Using IR, NMR, and GC-MS spectroscopic data, the structure of TFP-F1 was elucidated as {→3)-[β-D-GlcAp-(1→2)]-α-D-Manp-(1→3)-α-D-Manp-(1→3)-[α-L-Fucp-(1→2)-β-D-Xylp-(1→2)]-α-D-Manp-(1→}n, with partial acetylation of C6-OH in mannoses. Furthermore, at a concentration of 1 μg/mL, TFP-F1 was found to stimulate the secretion of TNF-α and IL-6 in J774A.1 macrophage cells in vitro via interaction with toll-like receptor 4 (TLR4). The removal of O-acetyl groups led to the loss of immunomodulatory activities, demonstrating that O-acetyl groups play an essential role in enhancing the production of pro-inflammatory cytokines.

Abstract:Herein, the low-molecular-weight heteropolysaccharide (designated as TABP), with a weight-average Mw of 5408 Da, was produced by the endophytic bacterium Bacillus sp. TAB, which was initially isolated from the fruiting bodies of the wild Tremella aurantialba . A relatively high TABP accumulation was obtained and enhanced to 6.94 g/L in 5 L fed-batch fermentation by high-density cultivation. Monosaccharide composition analysis showed that the TABP comprised arabinose, glucosamine , galactose , glucose, and mannose with a molar ratio of 0.073: 0.145: 0.406: 0.182: 0.195, respectively. Methylation and NMR analyses indicated that TABP contained 1,4-linked β– d –Gal p and 1,4-linked β– d –Man p pyranosyl backbone, extensively substituted at the side chains to form a complex structure. Prebiotic potential analysis exhibited significant growth-promoting effects for various lactic acid bacteria by more than 90 %. Overall, this study initially provides valuable insights into the endophytic exopolysaccharides from T. aurantialba and their biological activity, which provides prospective sources of prebiotics for functional foods and aids in understanding the endophytes symbiosis mechanism in edible mushrooms.

Abstract:Four Tremella fuciformis polysaccharides (TPS) fractions named as TPS20, TPS40, TPS60, and TPS80 were obtained by stepwise ethanol precipitation. Their physicochemical and rheological properties were investigated. Results showed TPS60 had the highest contents of polysaccharide (81.12%), protein (0.29%), uronic acid (14.36%), and sulfate (5.92%). Their molecular weights were in a descending order of TPS20, TPS40, TPS60, and TPS80. In a certain range of shear rate, they all presented pseudoplastic fluid and viscosity characteristic at low frequency. Increasing temperature or concentration of salts decreased the viscosity of their solutions. Sucrose concentration less than 6% led to an increasing viscosity of TPS20, TPS40, and TPS60. Both strong acidity and alkalinity could reduce the viscosity of all TPS solutions. TPS80 solution showed the best stability and its viscosity was minimally affected by pH, temperature, salts, and sucrose. These results can provide theoretical references for utilizations of TPS fractions in foods, pharmaceuticals, and cosmetics.

Abstract:Since ancient times, Tremella aurantialba has been proposed to have medicinal and food benefits. Modern phytochemistry and pharmacological studies have demonstrated that polysaccharides, the main components from T. aurantialba appear to be an all-round talent resisting a variety of chronic inflammatory diseases and protecting against different types of tumors, diabetes and cardiovascular diseases. These health and pharmacological benefits have gained much attention from scholars around the world. Further, more and more methods for polysaccharides extraction, purification, structure identification have been proposed. Significantly, the bioactivity of fungus polysaccharides is affected by many factors such as extraction and purification conditions and chemical structure. This paper provides an overview of recent advances in the isolation, structural features and biological effects of polysaccharides derived from T. aurantialba , covers recent advances in the field and outlines future research and applications of these polysaccharides.

Abstract:Low molecular weight polysaccharides had higher bio-activity and bioavailability compared to ultra-high molecular weight polysaccharides, this study aimed to obtain low molecular weight polysaccharides from Tremella fuciformis (TFLP) by using high-temperature and high-pressure assisted hydrochloric acid method to degrade Tremella fuciformis polysaccharides (TFP), and the structural characteristics, in vivo antioxidant and immune enhancing activities of TFP and TFLP was explored through Caenorhabditis elegans (C. elegans) and mice model. It was found that TFP and TFLP were acidic polysaccharides with molecular weights of 2238 kDa and 3 kDa, respectively. The glycosidic bonding of TFP and TFLP was mainly composed of different configurations of mannopyranose. TFP and TFLP had excellent in vivo antioxidant activity and stress resistance by regulating the mRNA transcription level and metabolites in C. elegans. Results also showed that TFP and TFLP could enhance the antioxidant capacity and immunity of serum, spleen and small intestine tissues in normal mice and cyclophosphamide-induced immunosuppressive mice through regulating the relative transcription and expression levels of anti-inflammatory related signaling factors, and it has found that TFLP showed better immune enhancement and antioxidant activity than TFP. In addition, Akkermansia, Bacteroides and Alloprevotella were characteristic bacteria at the genus level in immunosuppressed mice intervened with TFLP, with a significant increase in relative abundance. The content of SCFAs significantly increased in immunosuppressed mice by TFLP. These results indicated that TFP and TFLP had potential in vivo antioxidant and immune enhancing activities.

T. fuciformis polysaccharides mainly consist of heteropolysaccharides with α-(1→3)-linked D-mannan as the backbone chain, exhibiting diverse activities such as immunomodulation, antitumor, antioxidation, and antiaging. The present paper reviews the structural characteristics, various activities and structure -activity correlation of T. fuciformis polysaccharides.

Abstract:Abstract : This report represents an investigation of the interaction of aldosterone with kidney and liver cell membranes and with kidney subcellular frac tions and to compare this interaction with that of other steroid hormones. Aldosterone is weakly bound to kidney subcellular fractions. Liver and kidney cell membranes were found to either take up the steroids tested according to their solu bility in lipids or to bind them in accordance with the polarity rule of steroid-protein inter action. Spirolactone failed to displace aldo sterone from the kidney cell membranes. The re sults indicate that none of the cell fractions investigated bind aldosterone strongly or preferentially. (Author)

Abstract:Three kinds of water-soluble polysaccharides (PS, PFB and PM) were isolated from the sporocarp, fermentation broth and cultured mycelium of wild mushroom Agaricus bitorquis (Quél.) Sacc. Chaidam (ABSC). The Mw of PS, PFB and PM ranged from 893 Da to 38,340 Da. FTIR and NMR spectroscopy indicated that the skeleton of PS-I in PS contained α-D-mannopyranose with a backbone of (1 → 4)-α-D-glycosidic. The isolated fraction PFB-I in PFB was connected by (1 → 6) or (1 → 4)-α-D-glycosidic linkage. PM-I in PM was connected by (1 → 4)-α-D-glucopyranosyl residues with branched chain connected by (1 → 6)-α-glycosidic linkage. The anti-hypoxia activity results in vivo revealed that the supplementation with PS, PFS and PM could extend the survival time of mice under hypoxia stress, and increase GSH-Px, CAT and SOD activities and protein levels in liver tissues of mice, decrease MDA and NO levels. The findings suggested that such polysaccharides from ABSC showed anti-hypoxia activity.
Structural Characterization of a Polysaccharide from Trametes Sanguinea Lloyd with Immune-Enhancing Activity Via Activation of TLR4

Abstract:A bioactive polysaccharide (TS2-2A) with a molecular weight of 15 kDa was isolated from Trametes sanguinea Lloyd, a medicinal food homologous fungus, by water extraction-alcohol precipitation and chromatographic separation. NMR analysis of polysaccharide and MS/MS analysis of its oligosaccharide indicated that TS2-2A featured a novel straight chain with a backbone of 1,3-alpha-D-glucopyranose and 1,4-beta-D-glucopyranose at a molar ratio of 1:4. Moreover, TS2-2A, recognized by Toll-like receptor 4 (TLR4), stimulated RAW 264.7 macrophages to release related cytokines and contributed to immune-enhancing effects. Briefly, with remarkable immune enhancing activity and noncytotoxicity, TS2-2A was proposed to be a potential immune enhancer for supplementing drugs or functional foods.
