Abstract:Plant glutamate receptor-like channels (GLRs) are transmembrane proteins that allow the movement of several ions across membranes. In the model plant Arabidopsis, there are 20 GLR isoforms grouped in three clades and, since their discovery, it was hypothesized that GLRs were mainly involved in signaling processes. Indeed, in the last years, several pieces of evidence demonstrate different signaling roles played by GLRs, related to pollen development, sexual reproduction, chemotaxis, root development, regulation of stomatal aperture, and response to pathogens. Recently, GLRs have gained attention for their role in long-distance electric and calcium signaling. In this review, we resume the evidence about the role of GLRs in signaling processes. This role is mostly linked to the GLRs involvement in the regulation of ion fluxes across membranes and, in particular, of calcium, which represents a key second messenger in plant cell responses to both endogenous and exogenous stimuli.

Abstract:Glutamate receptors like channels (GLRs) are ligand gated non-selective cation channels and are multigenic in nature. They are homologs of mammalian ionic glutamate receptors (iGLRs) that play an important role in neurotransmission. It has been more than 25 years of discovery of plant GLRs, since then, significant progress has been made to unravel their structure and function in plants. Recently, the first crystal structure of plant GLR has been resolved that suggests that, though, plant GLRs contain the conserved signature domains of iGLRs, their unique features enable agonist/antagonist-dependent change in their activity. GLRs exhibit diverse subcellular localization and undergo dynamic expression variation in response to developmental and environmental stress conditions in plants. The combined use of genetic, electrophysiology and calcium imaging using different genetically encoded calcium indicators has revealed that GLRs are involved in generating calcium (Ca2+) influx across the plasma membrane and are involved in shaping the Ca2+ signature in response to different developmental and environmental stimuli. These findings indicate that GLRs influence cytosolic Ca2+ dynamics, thus, highlighting "GLR-Ca2+-crosstalk (GCC)" in developmental and stress-responsive signaling pathways. With this background, the present review summarises the recent developments pertaining to GLR function, in the broader context of regulation of stress tolerance in plants.

Abstract:Plant glutamate receptor-like (GLR) genes encode ion channels with demonstrated roles in electrical and calcium (Ca2+) signaling. The expansion of the GLR family along the lineage of land plants, culminating in the appearance of a multiclade system among flowering plants, has been a topic of interest since their discovery nearly 25 years ago. GLRs are involved in many physiological processes, from wound signaling to transcriptional regulation to sexual reproduction. Emerging evidence supports the notion that their fundamental functions are conserved among different groups of plants as well. In this review, we update the physiological and genetic evidence for GLRs, establishing their role in signaling and cell-cell communication. Special emphasis is given to the recent discussion of GLRs' atomic structures. Along with functional assays, a structural view of GLRs' molecular organization presents a window for novel hypotheses regarding the molecular mechanisms underpinning signaling associated with the ionic fluxes that GLRs regulate. Newly uncovered transcriptional regulations associated with GLRs-which propose the involvement of genes from all clades ofArabidopsis thaliana in ways not previously observed-are discussed in the context of the broader impacts of GLR activity. We posit that the functions of GLRs in plant biology are probably much broader than anticipated, but describing their widespread involvement will only be possible with (a) a comprehensive understanding of the channel's properties at the molecular and structural levels, including protein-protein interactions, and (b) the design of new genetic approaches to explore stress and pathogen responses where precise transcriptional control may result in more precise testable hypotheses to overcome their apparent functional redundancies.

Abstract:Abstract Animal ionotropic glutamate receptors (iGluRs) are ligand-gated channels whose evolution is intimately linked to that of the nervous system, where the agonist glutamate and co-agonists glycine/d-serine act as neurotransmitters or neuromodulators. While iGluRs are specialized in neuronal communication, plant glutamate receptor-like (GLR) homologs have evolved many plant-specific physiological functions, such as sperm signaling in moss, pollen tube growth, root meristem proliferation, innate immune, and wound responses. GLRs have been associated with Ca2+ signaling by directly channeling its extracellular influx into the cytosol. Nevertheless, very limited information on functional properties of GLRs is available, and we mostly rely on structure/function data obtained for animal iGluRs to interpret experimental results obtained for plant GLRs. Yet, a deeper characterization and better understanding of plant GLRs is progressively unveiling original and different functions when compared with their mammalian counterparts. Here, we review the function of plant GLRs comparing their predicted structure and physiological roles with those of the well-documented roles of iGluRs. We conclude that interpreting GLR function based on comparison with their animal counterparts calls for caution, especially when presuming physiological roles and the mode of action for plant GLRs, and when comparing iGluRs in neuronal tissues with those in peripheral, non-neuronal tissues.

Abstract:Plants rely on systemic signalling mechanisms to establish whole-body defence in response to insect and nematode attacks. GLUTAMATE RECEPTOR-LIKE ( GLR ) genes have been implicated in long-distance transmission of wound signals to initiate the accumulation of the defence hormone jasmonate (JA) at undamaged distal sites. The systemic signalling entails the activation of Ca 2+ -permeable GLR channels by wound-released glutamate, triggering membrane depolarization and cytosolic Ca 2+ influx throughout the whole plant. The systemic electrical and calcium signals rapidly dissipate to restore the resting state, partially due to desensitization of the GLR channels. Here we report the discovery of calmodulin-mediated, Ca 2+ -dependent desensitization of GLR channels, revealing a negative feedback loop in the orchestration of plant systemic wound responses. A CRISPR-engineered GLR3.3 allele with impaired desensitization showed prolonged systemic electrical signalling and Ca 2+ waves, leading to enhanced plant defence against herbivores. Moreover, this Ca 2+ /calmodulin-mediated desensitization of GLR channels is a highly conserved mechanism in plants, providing a potential target for engineering anti-herbivore defence in crops.

Abstract:Calcium (Ca 2+ ) serves as a ubiquitous second messenger by mediating various signaling pathways and responding to numerous environmental conditions in eukaryotes. Therefore, plant cells have developed complex mechanisms of Ca 2+ communication across the membrane, receiving the message from their surroundings and transducing the information into cells and organelles. A wide range of biotic and abiotic stresses cause the increase in [Ca 2+ ] cyt as a result of the Ca 2+ influx permitted by membrane-localized Ca 2+ permeable cation channels such as C YCLIC N UCLEOTIDE- G ATE C HANNELs (CNGCs), and voltage-dependent H YPERPOLARIZATION- A CTIVATED C ALCIUM 2+ PERMEABLE C HANNELs (HACCs), as well as G LUTAMATE RECEPTOR- L IKE R ECEPTORs (GLRs) and T WO- P ORE C HANNELs (TPCs). Recently, resistosomes formed by some N UCLEOTIDE-BINDING L EUCINE-RICH R EPEAT RECEPTORs (NLRs) are also proposed as a new type of Ca 2+ permeable cation channels. On the contrary, some Ca 2+ transporting membrane proteins, mainly Ca 2+ -ATPase and Ca 2+ /H + exchangers, are involved in Ca 2+ efflux for removal of the excessive [Ca 2+ ] cyt in order to maintain the Ca 2+ homeostasis in cells. The Ca 2+ efflux mechanisms mediate the wide ranges of cellular activities responding to external and internal stimuli. In this review, we will summarize and discuss the recent discoveries of various membrane proteins involved in Ca 2+ influx and efflux which play an essential role in fine-tuning the processing of information for plant responses to abiotic and biotic stresses.

Abstract:Tolerance of plants to a number of biotic and abiotic stresses such as pathogen and herbivore attack, drought, salinity, cold and nutritional limitations is ensued by complex multimodule signaling pathways. The outcome of this complex signaling pathways results in adaptive responses by restoring the cellular homeostasis and thus promoting survival. Functions of many plant cation transporter and channel protein families such as glutamate receptor homologs (GLRs), cyclic nucleotide-gated ion channel (CNGC) have been implicated in providing biotic and abiotic stress tolerance. Ion homeostasis regulated by several transporters and channels is one of the crucial parameters for the optimal growth, development and survival of all living organisms. The CNGC family members are known to be involved in the uptake of cations such as Na(+), K(+) and Ca(2+) and regulate plant growth and development. Detail functional genomics approaches have given an emerging picture of CNGCs wherein these protein are believed to play crucial role in pathways related to cellular ion homeostasis, development and as a 'guard' in defense against biotic and abiotic challenges. Here, we discuss the current knowledge of role of CNGCs in mediating stress management and how they aid plants in survival under adverse conditions.

Abstract:Recent studies have revealed the physiological roles of glutamate receptor-like channels (GLRs) in Arabidopsis; however, the functions of GLRs in rice remain largely unknown. Here, we show that knockout of OsGLR3.4 in rice leads to brassinosteroid (BR)-regulated growth defects and reduced BR sensitivity. Electrophoretic mobility shift assays and transient transactivation assays indicated that OsGLR3.4 is the downstream target of OsBZR1. Further, agonist profile assays showed that multiple amino acids can trigger transient Ca2+ influx in an OsGLR3.4-dependent manner, indicating that OsGLR3.4 is a Ca2+-permeable channel. Meanwhile, the study of internode cells demonstrated that OsGLR3.4-mediated Ca2+ flux is required for actin filament organization and vesicle trafficking. Following root injury, the triggering of both slow wave potentials (SWPs) in leaves and the jasmonic acid (JA) response are impaired in osglr3.4 mutants, indicating that OsGLR3.4 is required for root-to-shoot systemic wound signaling in rice. Brassinosteroid treatment enhanced SWPs and OsJAZ8 expression in root-wounded plants, suggesting that BR signaling synergistically regulates the OsGLR3.4-mediated systemic wound response. In summary, this article describes a mechanism of OsGLR3.4-mediated cell elongation and long-distance systemic wound signaling in plants and provides new insights into the contribution of GLRs to plant growth and responses to mechanical wounding.

Abstract:In Arabidopsis thaliana, local wounding and herbivore feeding provoke leaf-to-leaf propagating Ca2+ waves that are dependent on the activity of members of the glutamate receptor-like channels (GLRs). In systemic tissues, GLRs are needed to sustain the synthesis of jasmonic acid (JA) with the subsequent activation of JA-dependent signaling response required for the plant acclimation to the perceived stress. Even though the role of GLRs is well established, the mechanism through which they are activated remains unclear. Here, we report that in vivo, the amino-acid-dependent activation of the AtGLR3.3 channel and systemic responses require a functional ligand-binding domain. By combining imaging and genetics, we show that leaf mechanical injury, such as wounds and burns, as well as hypo-osmotic stress in root cells, induces the systemic apoplastic increase of L-glutamate (L-Glu), which is largely independent of AtGLR3.3 that is instead required for systemic cytosolic Ca2+ elevation. Moreover, by using a bioelectronic approach, we show that the local release of minute concentrations of L-Glu in the leaf lamina fails to induce any long-distance Ca2+ waves.
Genome-Wide Identification and Expression Analysis of BraGLRs Reveal Their Potential Roles in Abiotic Stress Tolerance and Sexual Reproduction

Abstract:Glutamate receptors (GLRs) are involved in multiple functions during the plant life cycle through affecting the Ca2+ concentration. However, GLRs in Brassica species have not yet been reported. In this study, 16 glutamate receptor-like channels (GLR) belonged to two groups were identified in the Brassica rapa (B. rapa) genome by bioinformatic analysis. Most members contain domains of ANF_receptor, Peripla_BP_6, Lig_chan, SBP_bac_3, and Lig_chan_Glu_bd that are closely related to glutamate receptor channels. This gene family contains many elements associated with drought stress, low temperature stress, methyl jasmonate (MeJA), salicylic acid (SA), and other stress resistance. Gene expression profiles showed that BraGLR genes were expressed in roots, stems, leaves, flowers, and siliques. BraGLR5 expression was elevated after drought stress in drought-sensitive plants. BraGLR1, BraGLR8, and BraGLR11 expression were significantly upregulated after salt stress. BraGLR3 expression is higher in the female sterile-line mutants than in the wild type. The expression levels of BraGLR6, BraGLR9, BraGLR12, and BraGLR13 were significantly higher in the male sterile-line mutants than in the wild type. The expression of most BraGLRs increased after self-pollination, with BraGLR9 exhibiting the greatest increase. These results suggest that BraGLRs play an important role in abiotic stress tolerance and sexual reproduction.

Abstract:Plant glutamate-like receptor genes (GLRs) are homologous to mammalian ionotropic glutamate receptors genes (iGluRs). Although GLRs have been implicated in plant defenses to biotic stress, the relationship between GLR-mediated plant immunity against fungal pathogens and electrical signals remains poorly understood. Here, we found that pretreatment with a GLR inhibitor, 6,7-dinitriquinoxaline-2,3-dione (DNQX), increased the susceptibility of tomato plants to the necrotrophic fungal pathogen Botrytis cinerea. Assessment of the glr3.3, glr3.5 and glr3.3/glr3.5 double-mutants upon B. cinerea infection showed that tomato GLR3.3 and GLR3.5 are essential for plant immunity against B. cinerea, wherein GLR3.3 plays the main role. Analysis of the membrane potential changes induced by glutamate (Glu) or glycine (Gly) revealed that amplitude was significantly reduced by knocking out GLR3.3 in tomato. While treatment with Glu or Gly significantly increased immunity against B. cinerea in wild-type plants, this effect was significantly attenuated in glr3.3 mutants. Thus, our data demonstrate that GLR3.3- and GLR3.5-mediated plant immunity against B. cinerea is associated with electrical signals in tomato plants.
The effects of cervical spinal cord injury on the pattern of breathing in the rat.

Abstract:The principle cause of mortality and chronic morbidity after spinal cord injury (SCI) is respiratory failure. Hemisection of the second cervical spinal segment (C2HS) is frequently used as a model of SCI. The effects of this injury on breathing pattern, however, have not been investigated. The pattern of breathing was examined at 1-month (controls, n = 12; C2HS, n = 7) and 2-months postinjury (controls, n = 11; C2HS, n = 5). Measurements were made before and after bilateral vagotomy to determine the role of vagal afferents in the effects of injury. Rats were anesthetized with urethane and allowed to breathe room air spontaneously. Direct arterial blood pressure was measured with a calibrated pressure transducer from a catheter in the femoral artery positioned via cut down. Arterial blood gases were measured using an i-STAT analyzer. Normothermia was maintained throughout the study. A linear pneumotachometer was calibrated by standard methods before and after each study day and attached to the end of a tracheotomy tube and 30 minutes was allowed before measurements were taken. The inspiratory and expiratory airflow was measured before and after bilateral vagotomy. The airflow signal from the pneumotachometer was integrated to give expired tidal volume (VT). Respiratory rate (RR) was measured from the airflow signal. All data are presented as mean ± SD. Means for expired tidal volume were compared between animal groups using the Kruskal–Wallis anova and the Mann–Whitney U-test. All other means were compared using a two factor anova. Multiple comparisons were made using t-tests with Bonferroni's correction. No differences in blood pressure or blood gases were detected between groups. Injured rats maintained the same minute ventilation as controls by utilizing higher RR (minute−1) (1-month; controls 127 ± 19, injured 177 ± 31: 2-months; controls 126 ± 20, injured 168 ± 23) and lower VT (mL kg−1) (1-month; controls 8.6 ± 0.8, injured 6.9 ± 0.2: 2-months; controls 8.3 ± 0.7, injured 5.9 ± 0.2). In addition, the frequency of augmented breaths was greater in injured rats at 2-months postinjury compared to the 2-month control group (minute−1) (controls 0.12 ± 16, injured 0.45 ± 0.20). After bilateral vagotomy, VT increased and RR decreased in all groups and the pattern of breathing was no longer different between injured and control animals. In conclusion, the C2HS alters the pattern of breathing at 1- and 2-months postinjury and increases the frequency of augmented breaths at 2-months postinjury. These results also suggest that the pattern of breathing may be determined by the vagal mechanisms rather than impaired respiratory motor drive.
Calmodulin and calmodulin-like protein-mediated plant responses to biotic stresses

Abstract:Plants have evolved a set of finely regulated mechanisms to respond to various biotic stresses. Transient changes in intracellular calcium (Ca2+ ) concentration have been well documented to act as cellular signals in coupling environmental stimuli to appropriate physiological responses with astonishing accuracy and specificity in plants. Calmodulins (CaMs) and calmodulin-like proteins (CMLs) are extensively characterized as important classes of Ca2+ sensors. The spatial-temporal coordination between Ca2+ transients, CaMs/CMLs and their target proteins is critical for plant responses to environmental stresses. Ca2+ -loaded CaMs/CMLs interact with and regulate a broad spectrum of target proteins, such as ion transporters (including channels, pumps, and antiporters), transcription factors, protein kinases, protein phosphatases, metabolic enzymes and proteins with unknown biological functions. This review focuses on mechanisms underlying how CaMs/CMLs are involved in the regulation of plant responses to diverse biotic stresses including pathogen infections and herbivore attacks. Recent discoveries of crucial functions of CaMs/CMLs and their target proteins in biotic stress resistance revealed through physiological, molecular, biochemical, and genetic analyses have been described, and intriguing insights into the CaM/CML-mediated regulatory network are proposed. Perspectives for future directions in understanding CaM/CML-mediated signalling pathways in plant responses to biotic stresses are discussed. The application of accumulated knowledge of CaM/CML-mediated signalling in biotic stress responses into crop cultivation would improve crop resistance to various biotic stresses and safeguard our food production in the future.
Principles of Calcium Signal Generation and Transduction in Plant Cells

Abstract:Abstract Calcium ions exhibit unique properties and a universal ability to transmit diverse signals in plant cells under the primary action of hormones, pathogens, light, gravity, and various abiotic stressors. In the last few years, considerable progress has been achieved in deciphering the mechanisms of Ca 2+ involvement in the regulation of plant responses. Recent studies revealed the genes encoding Ca 2+ -permeable channels that conduct Ca 2+ currents across the membranes during the transduction of the Ca 2+ signal. These proteins comprise the ligand-gated Ca 2+ -permeable channels activated by cyclic nucleotides (CNGC) and amino acids (glutamate receptor-like channels, GLR), the voltage-gated tonoplast channel (two-pore channel, TPC1), mechanosensitive channels (MSL, MCA, OSCA1), and annexins. The role of Ca 2+ -ATPase and Ca 2+ /H + -exchangers in the active extrusion of excess cytoplasmic Ca 2+ into the apoplast or cell organelles was examined in detail. The calmodulins (CaM), CaM-like proteins (CML), Ca 2+ -dependent protein kinases (CDPK), and complexes of calcineurin-B-like proteins (CBL) with CBL-interacting protein kinases (CIPK) were found to produce intricate signaling networks that decode Ca 2+ signals and elicit plant responses to external stimuli. This review analyzes the data accumulated over the past decade on the principles of formation and propagation of the calcium signal in plant cells.

Upregulation of GLRs expression by light in Arabidopsis leaves

Abstract:Abstract Background Glutamate receptor-like (GLR) channels are plant homologs of iGluRs, animal ionotropic glutamate receptors which participate in neurotransmission. GLRs mediate plant adaptive processes and photomorphogenesis. Despite their contribution to light-dependent processes, signaling mechanisms that modulate GLR response to light remain unknown. Here we show that leaf expression of 7 out of 20 Arabidopsis GLRs is significantly up-regulated by monochromatic irradiation. Results Our data indicates that both red and blue light stimulate the expression of selected AtGLRs . Using a photosynthesis inhibitor and different irradiation regimes, we demonstrated that retrograde signaling from photosystem II is unlikely to be involved in light-dependent GLR up-regulation. Analysis of transcriptional patterns in mutants of key photoreceptors allowed us to observe that both phytochromes and cryptochromes are likely to be involved in the control of light-dependent up-regulation of AtGLR expression, with phytochromes playing a clearly dominating role in this process. Conclusions In mature Arabidopsis leaves, phytochromes, assisted by cryptochromes, mediate light-driven transcriptional up-regulation of several genes encoding GLR proteins. Since GLRs are known to be involved in a wide range of plant developmental processes our results provide mechanistic insight into how light may influence plant growth and development.

Abstract:The Arabidopsis genome encodes for 20 members of putative ligand-gated channels, termed glutamate receptors (GLR). Despite the fact that initial studies suggested a role for GLRs in various aspects of photomorphogenesis, calcium homeostasis or aluminium toxicity, their functional properties and physiological role in plants remain elusive. Here, we have focussed on AtGLR3.4, which is ubiquitously expressed in Arabidopsis including roots, vascular bundles, mesophyll cells and guard cells. AtGLR3.4 encodes a glutamate-, touch-, and cold-sensitive member of this gene family. Abiotic stress stimuli such as touch, osmotic stress or cold stimulated AtGLR3.4 expression in an abscisic acid-independent, but calcium-dependent manner. In plants expressing the Ca(2+) -reporter apoaequorin, glutamate as well as cold elicited cytosolic calcium elevations. Upon glutamate treatment of mesophyll cells, the plasma membrane depolarised by about 120 mV. Both glutamate responses were transient in nature, sensitive to glutamate receptor antagonists, and were subject to desensitisation. One hour after eliciting the first calcium signal, a 50% recovery from desensitisation was observed, reflecting the stimulus-induced fast activation of AtGLR3.4 transcription. We thus conclude that AtGLR3.4 in particular and GLRs in general could play an important role in the Ca(2+) -based, fast transmission of environmental stress.

Abstract:Glutamate receptor-like channels (GLRs) play vital roles in various physiological processes in plants, such as wound response, stomatal aperture control, seed germination, root development, innate immune response, pollen tube growth, and morphogenesis. Despite the importance of GLRs, knowledge about their molecular organization is limited. Here we use X-ray crystallography and single-particle cryo-EM to solve structures of the Arabidopsis thaliana GLR3.4. Our structures reveal the tetrameric assembly of GLR3.4 subunits into a three-layer domain architecture, reminiscent of animal ionotropic glutamate receptors (iGluRs). However, the non-swapped arrangement between layers of GLR3.4 domains, binding of glutathione through S-glutathionylation of cysteine C205 inside the amino-terminal domain clamshell, unique symmetry, inter-domain interfaces, and ligand specificity distinguish GLR3.4 from representatives of the iGluR family and suggest distinct features of the GLR gating mechanism. Our work elaborates on the principles of GLR architecture and symmetry and provides a molecular template for deciphering GLR-dependent signaling mechanisms in plants.
