Abstract:Plants have evolved a set of finely regulated mechanisms to respond to various biotic stresses. Transient changes in intracellular calcium (Ca2+ ) concentration have been well documented to act as cellular signals in coupling environmental stimuli to appropriate physiological responses with astonishing accuracy and specificity in plants. Calmodulins (CaMs) and calmodulin-like proteins (CMLs) are extensively characterized as important classes of Ca2+ sensors. The spatial-temporal coordination between Ca2+ transients, CaMs/CMLs and their target proteins is critical for plant responses to environmental stresses. Ca2+ -loaded CaMs/CMLs interact with and regulate a broad spectrum of target proteins, such as ion transporters (including channels, pumps, and antiporters), transcription factors, protein kinases, protein phosphatases, metabolic enzymes and proteins with unknown biological functions. This review focuses on mechanisms underlying how CaMs/CMLs are involved in the regulation of plant responses to diverse biotic stresses including pathogen infections and herbivore attacks. Recent discoveries of crucial functions of CaMs/CMLs and their target proteins in biotic stress resistance revealed through physiological, molecular, biochemical, and genetic analyses have been described, and intriguing insights into the CaM/CML-mediated regulatory network are proposed. Perspectives for future directions in understanding CaM/CML-mediated signalling pathways in plant responses to biotic stresses are discussed. The application of accumulated knowledge of CaM/CML-mediated signalling in biotic stress responses into crop cultivation would improve crop resistance to various biotic stresses and safeguard our food production in the future.

Abstract:Abiotic stresses are important factors affecting plant growth and development and limiting agricultural production worldwide. Plants have evolved complex regulatory mechanisms to respond and adapt to constantly changing environmental conditions. C2H2 zinc finger proteins form a relatively large family of transcriptional regulators in plants. Recent studies have revealed that C2H2 zinc finger proteins function as key transcriptional regulators in plant responses to a wide spectrum of stress conditions, including extreme temperatures, salinity, drought, oxidative stress, excessive light and silique shattering. Here, we summarize recent functional analysis on C2H2 zinc finger proteins in plant responses to abiotic stresses and discuss their roles as part of a large regulatory network in the perception and responses by plants to different environmental stimuli.

Abstract:Intracellular changes in calcium ions (Ca(2+)) in response to different biotic and abiotic stimuli are detected by various sensor proteins in the plant cell. Calmodulin (CaM) is one of the most extensively studied Ca(2+)-sensing proteins and has been shown to be involved in transduction of Ca(2+) signals. After interacting with Ca(2+), CaM undergoes conformational change and influences the activities of a diverse range of CaM-binding proteins. A number of CaM-binding proteins have also been implicated in stress responses in plants, highlighting the central role played by CaM in adaptation to adverse environmental conditions. Stress adaptation in plants is a highly complex and multigenic response. Identification and characterization of CaM-modulated proteins in relation to different abiotic stresses could, therefore, prove to be essential for a deeper understanding of the molecular mechanisms involved in abiotic stress tolerance in plants. Various studies have revealed involvement of CaM in regulation of metal ions uptake, generation of reactive oxygen species and modulation of transcription factors such as CAMTA3, GTL1, and WRKY39. Activities of several kinases and phosphatases have also been shown to be modulated by CaM, thus providing further versatility to stress-associated signal transduction pathways. The results obtained from contemporary studies are consistent with the proposed role of CaM as an integrator of different stress signaling pathways, which allows plants to maintain homeostasis between different cellular processes. In this review, we have attempted to present the current state of understanding of the role of CaM in modulating different stress-regulated proteins and its implications in augmenting abiotic stress tolerance in plants.
The Mechanism of CBL-CIPK Signal Transduction under Stresses in Plants

Abstract:In plant, intracellular calcium levels altered in response to multiple abiotic stresses result in calcium sig-natures. Although the specific signature of the calcium transients can encode information on its own, an additional level of regulation in calcium signaling is achieved via the action of calcium-binding proteins. Calcineurin B-like proteins, as the plant calcium-binding proteins recently identified, must function by binding and regulating a group of serine-threonine protein kinases called CIPKs (CBL-interacting protein kinases) and provide a signal network to adapt various stresses. The calcium signal pathways have been well studied in plant and it has been identi-fied that the pathways play important roles in multiple abiotic stresses, including salt, drought and low K+.

Abstract:Calcium (Ca2+) ion is a critical ubiquitous intracellular second messenger, acting as a lead currency for several distinct signal transduction pathways. Transient perturbations in free cytosolic Ca2+ ([Ca2+]cyt) concentrations are indispensable for the translation of signals into adaptive biological responses. The transient increase in [Ca2+]cyt levels is sensed by an array of Ca2+ sensor relay proteins such as calmodulin (CaM), eventually leading to conformational changes and activation of CaM. CaM, in a Ca2+-dependent manner, regulates several transcription factors (TFs) that are implicated in various molecular, physiological, and biochemical functions in cells. CAMTA (calmodulin-binding transcription activator) is one such member of the Ca2+-loaded CaM-dependent family of TFs. The present review focuses on Ca2+ as a second messenger, its interaction with CaM, and Ca2+/CaM-mediated CAMTA transcriptional regulation in plants. The review recapitulates the molecular and physiological functions of CAMTA in model plants and various crops, confirming its probable involvement in stress signaling pathways and overall plant development. Studying Ca2+/CaM-mediated CAMTA TF will help in answering key questions concerning signaling cascades and molecular regulation under stress conditions and plant growth, thus improving our knowledge for crop improvement.

Abstract:Calcium is a crucial messenger in many growth and developmental processes in plants. The central mechanism governing how plant cells perceive and respond to environmental stimuli is calcium signal transduction, a process through which cellular calcium signals are recognized, decoded, and transmitted to elicit downstream responses. In the initial decoding of calcium signals, Ca2+ sensor proteins that bind Ca2+ and activate downstream signaling components are implicated, thereby regulating specific physiological and biochemical processes. After calcineurin B-like proteins (CBLs) sense these Ca2+ signatures, these proteins interact selectively with CBL-interacting protein kinases (CIPKs), thereby forming CBL/CIPK complexes, which are involved in decoding calcium signals. Therefore, specificity, diversity, and complexity are the main characteristics of the CBL-CIPK signaling system. However, additional CBLs, CIPKs, and CBL/CIPK complexes remain to be identified in plants, and the specific functions of their abiotic and biotic stress signaling will need to be further dissected. Therefore, a much-needed synthesis of recent findings is important to further the study of CBL-CIPK signaling systems. Here, we review the structure of CBLs and CIPKs, discuss the current knowledge of CBL–CIPK pathways that decode calcium signals in Arabidopsis, and link plant responses to a variety of environmental stresses with specific CBL/CIPK complexes. This will provide a foundation for future research on genetically engineered resistant plants with enhanced tolerance to various environmental stresses.

Abstract:Calcium (Ca2+) signals are a core regulator of plant growth and development and responses to environmental cues and thus highlighted in plant physiological and stress biology. External stimuli trigger specifically intracellular spatial and temporal [Ca2+]cyt variations in plant cells. This [Ca2+]cyt variations will be sensed and decoded by calcium sensors and, in turn, calcium sensor interacting proteins transmit resulting signals to the downstream effectors to activate the expression of early response genes or promote ion channel activities, finally leading to specific stress responses. How the plant cell distinguishes different types or intensity of external stimuli through sensing intracellular spatial and temporal variations of Ca2+ signals is a scientific issue recently highlighted by plant biologists. This review summarized recent advances in the research field of plant calcium sensors, including the structural characteristics, functional roles, and stress signaling path-ways of calcium-dependent protein kinases (CDPKs), calmodulins (CaMs), calmodulin-like proteins (CMLs), and cal-cineurin B-like proteins (CBLs) and their interacting kinases (CIPKs), and moreover provided new insights and perspectives.

Abstract:In their natural environment, plants have to continuously face constraints such as biotic and abiotic stresses. To achieve their life cycle, plants have to perceive and interpret the nature, but also the strength of environmental stimuli to activate appropriate physiological responses. Nowadays, it is well established that signaling pathways are crucial steps in the implementation of rapid and efficient plant responses such as genetic reprogramming. It is also reported that rapid raises in calcium (Ca<sup>2+</sup>) levels within plant cells participate in these early signaling steps and are essential to coordinate adaptive responses. However, to be informative, calcium increases need to be decoded and relayed by calcium-binding proteins also referred as calcium sensors to carry-out the appropriate responses. In a recent study, we showed that CML8, an Arabidopsis calcium sensor belonging to the calmodulin-like (CML) protein family, promotes plant immunity against the phytopathogenic bacteria Pseudomonas syringae pv tomato (strain DC3000). Interestingly, other CML proteins such as CML9 were also reported to contribute to plant immunity using the same pathosystem. In this addendum, we propose to discuss about the specific contribution of these 2 CMLs in stress responses.

Abstract:Plants have a variety of regulatory mechanisms to perceive, transduce, and respond to biotic and abiotic stress. One such mechanism is the calcium-sensing CBL–CIPK system responsible for the sensing of specific stressors, such as drought or pathogens. CBLs perceive and bind Calcium (Ca 2+ ) in response to stress and then interact with CIPKs to form an activated complex. This leads to the phosphorylation of downstream targets, including transporters and ion channels, and modulates transcription factor levels and the consequent levels of stress-associated genes. This review describes the mechanisms underlying the response of the CBL–CIPK pathway to biotic and abiotic stresses, including regulating ion transport channels, coordinating plant hormone signal transduction, and pathways related to ROS signaling. Investigation of the function of the CBL–CIPK pathway is important for understanding plant stress tolerance and provides a promising avenue for molecular breeding.

Abstract:Calcium (Ca 2+ ) is a second messenger in plants growth and development, as well as in stress responses. The transient elevation in cytosolic Ca 2+ concentration have been reported to be involved in plants response to abiotic and biotic stresses. In plants, Ca 2+ -induced transcriptional changes trigger molecular mechanisms by which plants adapt and respond to environment stresses. The mechanism for transcription regulation by Ca 2+ could be either rapid in which Ca 2+ signals directly cause the related response through the gene transcript and protein activities, or involved amplification of Ca 2+ signals by up-regulation the expression of Ca 2+ responsive genes, and then increase the transmission of Ca 2+ signals. Ca 2+ regulates the expression of genes by directly binding to the transcription factors (TFs), or indirectly through its sensors like calmodulin, calcium-dependent protein kinases (CDPK) and calcineurin B-like protein (CBL). In recent years, significant progress has been made in understanding the role of Ca 2+ -mediated transcriptional regulation in different processes in plants. In this review, we have provided a comprehensive overview of Ca 2+ -mediated transcriptional regulation in plants in response to abiotic stresses including nutrition deficiency, temperature stresses (like heat and cold), dehydration stress, osmotic stress, hypoxic, salt stress, acid rain, and heavy metal stress.

Abstract:Calcium signalling, including pulse, amplitude, and duration, is essential for plant development and response to various stimuli. However, the calcium signalling should be decoded and translated by calcium sensors. In plants, three classes of calcium-binding proteins have been identified as calcium sensors, including calcium-dependent protein kinase (CDPK), calcineurin B-like protein (CBL), and calmodulin (CaM). Calmodulin-like proteins (CMLs), which have several EF-hands, also serve as specific calcium sensors and can sense, bind, and interpret the calcium signal during the plant's growth and defense decision-making processes. In recent decades, the function of CMLs in plant development and response to various stimuli has been systematically reviewed, shedding light on the molecular mechanism of plant CML-mediated networks in calcium signal transduction. Here, by providing an overview of CML expression and biological function in plants, we demonstrate that growth-defense trade-offs occur during calcium sensing, an aspect that has not been well studied in recent years.

Abstract:Plants are exposed to various environmental stresses. The sensing of environmental cues and the transduction of stress signals into intracellular signaling are initial events in the cellular signaling network. As a second messenger, Ca 2+ links environmental stimuli to different biological processes, such as growth, physiology, and sensing of and response to stress. An increase in intracellular calcium concentrations ([Ca 2+ ] i ) is a common event in most stress-induced signal transduction pathways. In recent years, significant progress has been made in research related to the early events of stress signaling in plants, particularly in the identification of primary stress sensors. This review highlights current advances that are beginning to elucidate the mechanisms by which abiotic environmental cues are sensed via Ca 2+ signals. Additionally, this review discusses important questions about the integration of the sensing of multiple stress conditions and subsequent signaling responses that need to be addressed in the future.

Abstract:To cope with environmental stimuli, plants have evolved precise regulatory mechanisms to perceive, transduce and respond to abiotic stresses that can negatively affect growth and development. The CBL–CIPK signaling system is a newly emerging plant-specific and Ca2+-dependent network mediating abiotic stress tolerance. CBLs may sense a Ca2+ signature triggered by abiotic stresses, and have specific interactions with novel CIPK-type kinases after binding Ca2+. The CBL/CIPK complexes may post-translationally phosphorylate downstream target proteins to regulate abiotic stress tolerance in a cell or tissue-specific manner. In some cases transcription factors are induced to activate stress-responsive genes that control adaptation reactions. The CBL–CIPK signaling system exhibits specificity, diversity and complexity. Meanwhile, cross talk also exists in the CBL–CIPK signaling. To date, significant progress has been made in the role of the CBL–CIPK signaling system in responding to salt, low K+ and to high pH, which will provide a fast and efficient method of molecular design breeding combined with the CBL/CIPK engineering of crop plants, for enhanced tolerance to abiotic stresses. However, more CBL/CIPK components remain to be identified, particularly from specific plants that grow in conditions with abiotic stress, and the specificity of their abiotic stress signaling will need to be dissected.

Abstract:The Ca(2+) ion is recognized as a crucial second messenger in signaling pathways coupling the perception of environmental stimuli to plant adaptive responses. Indeed, one of the earliest events following the perception of environmental changes (temperature, salt stress, drought, pathogen, or herbivore attack) is intracellular variation of free calcium concentrations. These calcium variations differ in their spatio-temporal characteristics (subcellular location, amplitude, kinetics) with the nature and strength of the stimulus and, for this reason, they are considered as signatures encrypting information from the initial stimulus. This information is believed to drive a specific response by decoding via calcium-binding proteins. Based on recent examples, we illustrate how individual calcium sensors from the calcium-dependent protein kinase and calmodulin-like protein families can integrate inputs from various environmental changes. Focusing on members of these two families, shown to be involved in plant responses to both abiotic and biotic stimuli, we discuss their role as key hubs and we put forward hypotheses explaining how they can drive the signaling pathways toward the appropriate plant responses.

Abstract:Calmodulins (CaMs) and calmodulin-like proteins (CMLs) belong to families of calcium-sensors that act as calcium ion (Ca2+) signal-decoding proteins and regulate downstream target proteins. As a tropical halophyte, Canavalia rosea shows great resistance to multiple abiotic stresses, including high salinity/alkalinity, extreme drought, heat, and intense sunlight. However, investigations of calcium ion signal transduction involved in the stress responses of C. rosea are limited. The CaM and CML gene families have been identified and characterized in many other plant species. Nevertheless, there is limited available information about these genes in C. rosea. In this study, a bioinformatic analysis, including the gene structures, conserved protein domains, phylogenetic relationships, chromosome distribution, and gene synteny, was comprehensively performed to identify and characterize CrCaMs and CrCMLs. A spatio-temporal expression assay in different organs and environmental conditions was then conducted using the RNA sequencing technique. Additionally, several CrCaM and CrCML members were then cloned and functionally characterized using the yeast heterogeneous expression system, and some of them were found to change the tolerance of yeast to heat, salt, alkalinity, and high osmotic stresses. The results of this study provide a foundation for understanding the possible roles of the CrCaM and CrCML genes, especially for halophyte C. rosea's natural ecological adaptability for its native habitats. This study also provides a theoretical basis for further study of the physiological and biochemical functions of plant CaMs and CMLs that are involved in tolerance to multiple abiotic stresses.

Effects of jen-sheng-yang-yung-tang on cellular immunocompetence of gamma-irradiated mice.

Abstract:This study was conducted to determine the therapeutic effects of Jen-Sheng-Yang-Yung-Tang on mice irradiated by gamma-ray. A total of 160 male ICR strain mice, 6-7 weeks of age, were chosen to receive different treatment of radiation and Jen-Sheng-Yang-Yung-Tang. After the treatments, six to eight mice from each group were sacrificed on days 1, 5, 12, 19, 26 and 33. The body and splenic weights of mice by different treatments were measured and the splenic cells were separated under aseptic conditions, thereafter. The changes of cellular immunocompetence in mice following treatments were measured by 3H-thymidine incorporation in each experimental mouse. The results revealed that 4 Gy of gamma-ray irradiation inhibited the increases of body and splenic weights and exerted an obvious inhibitory effect on the blastogenic responses of the splenic lymphoid cells after being stimulated by mitogens such as PHA, Con A, PWM and LPS. Jen-Sheng-Yang-Yung-Tang administration seemed to help the recovery of cellular immunocompetence in the gamma-ray irradiated mice. Among these treatments, Jen-Sheng-Yang-Yung-Tang administered with the concentration of 20 mg/20 g body weight after irradiation enhanced the recovery of radiation damages and had the highest efficacy of any other kinds of treatments.

Abstract:Plants evolve stress-specific responses that sense changes in their external environmental conditions and develop various mechanisms for acclimatization and survival. Calcium (Ca2+ ) is an essential stress-sensing secondary messenger in plants. Ca2+ sensors, including calcium-dependent protein kinases (CDPKs), calmodulins (CaMs), CaM-like proteins (CMLs), and calcineurin B-like proteins (CBLs), are involved in jasmonates (JAs) signalling and biosynthesis. Moreover, JAs are phospholipid-derived phytohormones that control plant response to abiotic stresses. The JAs signalling pathway affects hormone-receptor gene transcription by binding to the basic helix-loop-helix (bHLH) transcription factor. MYC2 acts as a master regulator of JAs signalling module assimilated through various genes. The Ca2+ sensor CML regulates MYC2 and is involved in a distinct mechanism mediating JAs signalling during abiotic stresses. This review highlights the pivotal role of the Ca2+ sensors in JAs biosynthesis and MYC2-mediated JAs signalling during abiotic stresses in plants.

Abstract:Abstract As sessile organisms, plants have to cope with environmental change and numerous biotic and abiotic stress. Upon perceiving environmental cues and stress signals using different types of receptors, plant cells initiate immediate and complicated signaling to regulate cellular processes and respond to stress. Receptor-like cytoplasmic kinases (RLCKs) transduce signals from receptors to cellular components and play roles in diverse biological processes. Recent studies have revealed the hubbing roles of RLCKs in plant responses to biotic stress. Emerging evidence indicates the important regulatory roles of RLCKs in plant responses to abiotic stress, growth, and development. As a pivot of cellular signaling, the activity and stability of RLCKs are dynamically and tightly controlled. Here, we summarize the current understanding of how RLCKs regulate plant responses to biotic and abiotic stress.
