Abstract:Stable and well dispersed nickel nanoparticles (NiNPs) were fabricated and embedded in a novel polymer sulfonate and benzimidazole functionalized poly (arylene ether ketone) (S-BI-PAEK) film. After drop-casting the mixed solution of S-BI-PAEK and NiSO4 on glassy carbon electrode (GCE) surface, the uniformly distributed NiNPs were formed and stably embedded in S-BI-PAEK film by in-situ electrochemical reduction method. The embedment and well dispersity of NiNPs in S-BI-PAEK film was probably attributed to the strong chelation of sulfonate and benzimidazole functional groups contained in S-BI-PAEK toward Ni2+ ions, as well as the transferability of Ni2+ ions in S-BI-PAEK film. The NiNPs/S-BI-PAEK composite film was characterized by scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDS), electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). It exhibited good electrocatalytic activity toward glucose oxidation in 0.1mol L-1 NaOH solution with high stability. The NiNPs/S-BI-PAEK/GCE showed a fast amperometric response with a wide linear range from 1M to 4mM and a low detection limit of 200nmolL(-1) (S/N=3) for the determination of glucose by amperometry at a potential of 0.55V. Finally it was successfully employed to determine glucose in human serum. Therefore, the novel fabrication method of nickel nanoparticles was promising for the future development of non-enzymatic glucose sensor.
Stabilization of Bilirubin Oxidase in a Biogel Matrix for High-performance Gas Diffusion Electrodes

Abstract:Enzyme immobilization on solid conducting surfaces faces some challenges for practical applications in technologies such as biosensors and biofuel cells. Short-term stability, poor electrochemical performance, and enzyme inhibition are some issues that remain unsolved. Here, we propose a simple methodology for bilirubin oxidase (BOD) immobilization on carbon-based gas-diffusion electrodes for a four-electron electrochemical oxygen reduction reaction (ORR). The enzyme is incorporated into a Nafion® polymeric matrix and cross-linked with glutaraldehyde by a one-pot reaction in a buffered solution, producing a stable BOD-based biogel. The biogel prevents the formation of enzyme aggregates, producing a homogeneous bioelectrode surface, and allows access to the direct electron-transfer mechanism of multicopper centers buried in the enzyme. A biocatalytic reduction current of -1.52 ± 0.24 mA cm -2 at 0.19 ± 0.06 V was observed under gas-diffusion conditions. Additionally, the bioelectrode showed an unprecedented long-term stability under continuous operation combined with satisfactory catalytic current without redox mediator, demonstrating that the BOD-based biogel provides a suitable microenvironment for long-term enzymatic activity involving a bio-three-phase interfacial reaction. Therefore, the present study contributes new insights into enzyme immobilization to overcome the critical short-term stability issue of enzyme-based electrochemical devices for practical applications.

Abstract:Aggregation of reduced graphene oxide (RGO) due to π-π stacking is a recurrent problem in graphene-based electrochemistry, decreasing the effective working area and therefore the performance. Dispersing RGO on three-dimensional (3D) carbon paper electrode is one strategy towards overcoming this, which partially relieves aggregation. In this report, we describe the graft of negatively charged 4-aminobenzoic acid (4-ABA) onto a graphene functionalized carbon paper electrode surface. 4-ABA functionalization induces separation of the RGO layers, at the same time leading to favorable orientation of the blue multi-copper enzyme Myrothecium verrucaria bilirubin oxidase (MvBOD) for direct electron transfer (DET) in the dioxygen reduction reaction (ORR) at neutral pH. Simultaneous electroreduction of graphene oxide to RGO and covalent attachment of 4-ABA are achieved by applying alternating cathodic and anodic electrochemical potential pulses, leading to a very high catalytic current density (Δjcat:193 ± 4 μA cm−2) under static conditions. Electrochemically grafted 4-ABA not only leads to a favorable orientation of BOD as validated by fitting a kinetic model to the electrocatalytic data, but also acts to alleviate RGO aggregation as disclosed by scanning electron microscopy, most likely due to the electrostatic repulsion between 4-ABA-grafted graphene layers. With a half-lifetime of 55 h, the bioelectrode also shows the highest operational stability for DET-type MvBOD-based bioelectrodes reported to date. The bioelectrode was finally shown to work well as a biocathode of a membrane-less glucose/O2 enzymatic biofuel cell with a maximum power density of 22 μW cm−2 and an open circuit voltage of 0.51 V.

Abstract:Bilirubin oxidase (BOD) molecules were firmly tethered on the surface of graphene oxide (GO) functionalized with 2-hydroxy-4-amino-azobenzene (AZO-1). Further, a BOD-based electrode was prepared through the immobilization of enzyme on the nanocomplex via drop casting. The morphology, structural properties, surface chemistry, and physiochemical features of the prepared nanocomplex were investigated by microscopic, spectrometric, electrochemical, and other characterization techniques. The as-prepared nanocomplex with enzyme incorporation exhibited high loading amount of redox protein molecule, excellent mechanical robustness, and thermal stability. The complicated mutual interaction between the nanocomplex and protein molecules led to a partial ordered conglomeration of enzyme molecules with micelle-like surface characteristics. The combination of cofactors within BOD and AZO-1 formed a composite with significantly suppressed fluorescence emission and inferior capability of electron transportation. Direct charge transfer in BOD-based electrode could be achieved with redox group of GO as the primary electroactive site. A considerable negative impact on the efficiency of electron transport, turnover frequency of substrate conversion, and incident light utilization for this redox-protein-based electrode was verified. However, the as-prepared BOD-based electrode exhibited high affinity to oxygen molecules, and the oxygen binding was not hampered severely in the presence of ligation between adjacent heteroatoms within AZO-1 and T1 site in BOD.

Abstract:A novel hybrid nanomaterial (GO-MWNTs) was explored based on the self-assembly of multiwall carbon nanotubes (MWNTs) and graphene oxide (GO). Compared with pristine MWNTs, such a nanocomposite could be well dispersed in aqueous solution and exhibit a negative charge. Driven by the electrostatic interaction, positively charged horseradish peroxidase (HRP) could then be immobilized onto GO-MWNTs at the surface of a glassy carbon (GC) electrode to form a HRP/GO-MWNT/GC electrode under mild conditions. TEM was used to characterize the morphology of the GO-MWNT nanocomposite. UV-vis and FTIR spectra suggested that HRP was immobilized onto the hybrid matrix without denaturation. Furthermore, the immobilized HRP showed enhanced direct electron transfer for the HRP-Fe(III)/Fe(II) redox center. Based on the direct electron transfer of the immobilized HRP, the HRP/GO-MWNT/GC electrode exhibited excellent electrocatalytic behavior to the reduction of H2O2 and NaNO2, respectively. Therefore, GO-MWNTs could provide a novel and efficient platform for the immobilization and biosensing of redox enzymes, and thus may find wide potential applications in the fabrication of biosensors, biomedical devices, and bioelectronics.

Abstract:A nanohybrid biomaterial was fabricated by mixing Co3O4 nanorods, gold nanoparticles (Au-NPs) and myoglobin (Mb), and depositing it on the surface of a carbon paste electrode containing the ionic liquid N-hexylpyridinium hexafluorophosphate as the binder. UV–vis and FT-IR revealed the Mb in the composite film to have remained in its native structure. A pair of well-defined redox peaks appears in cyclic voltammograms and indicates direct electron transfer from the Mb to the underlying electrode. The results are attributed to the favorable orientation of Mb in the composite film, to the synergistic effects of Co3O4 nanorods and Au-NPs. The modified electrode shows excellent electrocatalytic ability towards the reduction of substrates such as trichloroacetic acid and nitrite, and displays good stability and reproducibility.

Abstract:Polydopamine (Pdop) has recently been shown to adsorb to a wide variety of surfaces and serves as an adhesion layer to immobilize biological molecules. In this work, the multifunctional carbon nanotube (CNT) composites were prepared though the oxidation of dopamine at room temperature and subsequent electroless silver deposition by mildly stirring. The stable immobilization and direct electron transfer of glucose oxidase were achieved on the composite film modified glassy carbon electrode. The resulting electrode gave a well-defined redox peaks with a formal potential of about −482mV (vs. SCE) in pH 7.0 buffer. The electron transfer rate constant was estimated to be 3.6s−1, due to the combined contribution of Pdop, CNTs and Ag nanoparticles with the help of Nafion. Furthermore, the method for detecting of glucose was proposed based on the decrease of oxygen caused by the enzyme-catalyzed reaction between glucose oxidase (GOD) and glucose. The linear response to glucose ranging from 50.0μM to 1.1mM (R2=0.9958), with a calculated detection limit of 17.0μM at a signal-to-noise ratio of 3. The low calculated apparent Michaelis–Menten constant (KMapp) was 5.46mM, implying the high enzymatic activity and affinity of immobilized GOD for glucose. It can reasonably be expected that this observation might hold true for other noble metal nanostructure-electroactive protein systems, providing a promising platform for the development of biosensors and biofuel cells.
Direct Electrochemistry of Hemoglobin Immobilized in CuO Nanowire Bundles.

Abstract:It is one of main challenges to find the suitable materials to enhance the direct electron transfer between the electrode and redox protein for direct electrochemistry field. Nano-structured metal oxides have attracted considerable interest because of unique properties, well biocompatibility, and good stability. In this paper, the copper oxide nanowire bundles (CuO NWBs) were prepared via a template route, and the bioelectrochemical performances of hemoglobin (Hb) on the CuO NWBs modified glass carbon electrodes (denoted as Hb-CuO NWBs/GC) were studied. TEM and XRD were used to characterize the morphology and structure of the as synthesized CuO NWBs. Fourier transform-infrared spectroscopy (FT-IR) proved that Hb in the CuO NWBs matrix could retain its native secondary structure. A pair of well-defined and quasi-reversible redox peaks at approximately -0.325 V (vs. Ag/AgCl saturated KCl) were shown in the cyclic voltammogram curve for the Hb-CuO NWBs/GC electrode, which indicated the direct electrochemical behavior. The Hb-CuO NWBs/GC electrode also displayed a good electrocatalytic activity toward the reduction of hydrogen peroxide. These results indicate that the CuO NWBs are good substrates for immobilization of biomolecules and might be promising in the fields of (bio) electrochemical analysis.

Abstract:Glyconanoparticles (GNPs) made by self-assembly of carbohydrate-based polystyrene-block-β-cyclodextrin copolymer are used as a building block for the design of nanostructured biomaterials of electrode. The firm immobilization of GNPs is carried out on electrochemically generated polymer, poly(pyrrole-adamantane), and copolymer, poly(pyrrole-adamantane)/poly(pyrrole-lactobionamide) via host-guest interactions between adamantane and β-cyclodextrin. The ability of GNPs for the specific anchoring of biological macromolecules is investigated using glucose oxidase enzyme modified by adamantane groups as a protein model (GOx-Ad). The immobilization of GOx-Ad is carried out by incubation of an aqueous enzyme solution on a coating of GNPs adsorbed on a platinum electrode. The presence of immobilized GOx-Ad is evaluated in aqueous glucose solution by potentiostating the underlying platinum electrode at 0.7 V/SCE for the electro-oxidation of H2 O2 generated by the enzyme. The analytical performance of the bioelectrodes for the detection of glucose is compared to control electrodes prepared without GNPs or without electropolymerized films. The better permeability of copolymer compared to polymer and the possibility to elaborate two alternating layers of GNPs and GOx-Ad are clearly observed. The best amperometric response is recorded with a multilayered bioelectrode displaying a wide linear range linear range of the calibration curve: 68 µmol L-1 to 0.1 mol L-1 .

Abstract:TiO2 nanoparticles were homogeneously coated on multiwalled carbon nanotubes (MWCNTs) by hydrothermal deposition, and this nanocomposite might be a promising material for myoglobin (Mb) immobilization in view of its high biocompatibility and large surface. The glassy carbon (GC) electrode modified with Mb–TiO2/MWCNTs films exhibited a pair of well-defined, stable and nearly reversible cycle voltammetric peaks. The formal potential of Mb in TiO2/MWCNTs film was linearly varied in the range of pH 3–10 with a slope of 48.65 mV/pH, indicating that the electron transfer was accompanied by single proton transportation. The electron transfer between Mb and electrode surface, ks of 3.08 s−1, was greatly facilitated in the TiO2/MWCNTs film. The electrocatalytic reductions of hydrogen peroxide were also studied, and the apparent Michaelis–Menten constant is calculated to be 83.10 μM, which shows a large catalytic activity of Mb in the TiO2/MWCNTs film to H2O2. The modified GC electrode shows good analytical performance for amperometric determination of hydrogen peroxide. The resultant Mb–TiO2/MWCNTs modified glassy carbon electrode exhibited fast amperometric response to hydrogen peroxide reduction, long term life and excellent stability. Finally the activity of the sensor for nitric oxide reduction was also investigated.

Abstract:In this paper, we compared the direct electron transfer and electrocatalytic properties of bilirubin oxidase (BOD) immobilized at two kinds of carbon nanotubes (CNTs), bamboo-CNTs and uniform-CNTs. X-ray diffraction and X-ray photoelectron spectroscopy results indicated that the ratio of sp2 band to sp3 band and the content of oxygen-containing groups at the surface of uniform-CNTs were higher than that of bamboo-CNTs. Moreover, uniform-CNTs can be well separated at the surface of the electrode. Better electrochemical and electrocatalytic properties of BOD immobilized at uniform-CNTs were shown.

Abstract:A method for immobilizing proteins in a carbon mesoporous material (CMM) containing platinum nanoparticles (Pt-NPs) is demonstrated. Compared to pure CMM or carbon nanotubes, CMM containing Pt-NPs enhances the electron transfer and redox properties of redox enzymes, such as glucose oxidase (GOx), due to a cooperative effect of Pt-NPs and CMM. The quasi-reversible electron transfer of GOx in this system is probed, and the apparent heterogeneous electron transfer rate constants are found to be 66% larger than in pure CMM. The GOx/Pt-CMM based glucose biosensor enables the determination of glucose at a potential of 600 mV (vs. SCE). Its detection limit is 10 times lower, and the sensitivity is 16 times higher than that of the respective biosensor without Pt-NPs.

Abstract:In this paper we have described that multi-walled carbon nanotubes (MWNTs) can be homogeneously dispersed and solubilized in water using non-covalent functionalization. These soluble MWNTs were then used for the immobilization of glucose oxidase (GOx) on glassy carbon electrode. Scanning electron microscopy is used to investigate the nanoscale structure of the film formed by the functionalized MWNTs. Electrochemical measurements were conducted to reveal the electroactivity of the GOx on the MWNT modified electrode. The functionalized MWNTs served as a network carrier for GOx as well as a conductor, enhancing greatly the response current. The GOx immobilized in this way retained its bioelectrocatalytic activity for the oxidation of glucose.
Pt based enzyme electrode probes assembled with Prussian Blue and conducting polymer nanostructures

Abstract:Conductive polymer nanotubules of 1,2-diaminobenzene (1,2-DAB) were prepared using a porous polycarbonate membrane template, placed on a Pt foil and used to support the polymer, then, the electropolymerisation was performed by chronocoulometry. The obtained conductive polymer nanostructures were then placed on Pt electrode and used to support highly dispersed prussian blue (PB), which acts as the active component for H2O2 detection. The observed good stability of PB as catalyst of H2O2 was related to the presence of organic non-conventional conducting polymers in a composite nanostructured film. These nanostructured polymer/PB composite films were also characterised by scanning electron microscopy (SEM) and Raman spectroscopy. The non-conventional conducting polymer nanotubules/PB modified Pt electrodes were tested by cyclic voltammeter for stability at different pH values, then, by amperometry, for hydrogen peroxide, ascorbic acid, acetaminophen, uric acid and acetylcholine. Glucose oxidase (GOD), lactate oxidase (LOD), L-amino acid oxidase (L-AAOD), alcohol oxidase (AOD), glycerol-3-phosphate oxidase (GPO), lysine oxidase (LyOx), and choline oxidase (ChOx) were immobilised on PB layer supported on 1,2-diaminobenzene (1,2-DAB) nanotubules onto the Pt electrodes. Different strategies for enzyme immobilisation were performed and used. Analytical parameters such as reproducibility, interference rejection, response time, storage and operational stability of the sensors have been studied and optimised. Results provide a guide to design high sensitive, stable and interference-free biosensors. The glucose biosensors assembled with nanostructured poly(1,2-DAB) showed a detection limit of 5 x 10(-5) mol l(-1), a wide linearity range (5 x 10(-5) to 5 x 10(-3) mol l(-1)), a high selectivity, a stability of 3 months at 4 degrees C, and at least 4 weeks at room temperature. Similar analytical parameters and stability were also studied for L-(+)-lactic acid, L-leucine, ethanol, glycerol-3-phosphate, lysine, and choline biosensors.

Abstract:A novel glassy carbon electrode modified by a gel containing multi-walled carbon nanotubes (MWNTs) and ionic liquid of 1-butyl-3-methylimidazolium hexafluorophosphate (BMIPF6) is reported. The gel is formed by grinding of MWNTs and BMIPF6. Such gel is then coated on the surface of a glassy carbon electrode. We have employed scanning electron microscopy, Fourier transform infrared spectrometry ( FTIR) and cyclic voltammetry to characterize the modified electrode. The direct electron transfers of hemoglobin and catalase on the modified electrode have been observed and studied in detail electrochemically. Hemoglobin is verified to be adsorbed on the modified electrode with the retention of conformation, which has been proved by microscopic FTIR. The electrochemical response of the adsorbed hemoglobin on the modified electrode is very stable, and shows repeated changes in the different pH solutions. It also has shown electrocatalysis to the reduction of oxygen and trichloroacetic acid. Catalase adsorbed on the gel modified electrode still keep activity to hydrogen peroxide. This work provides a simple and easy approach to construct biosensors based on the carbon nanotubes and ionic liquids.
Electrocatalysis Via Direct Electrochemistry of Myoglobin Immobilized on Colloidal Gold Nanoparticles

Abstract:The direct electron transfer between immobilized myoglobin (Mb) and colloidal gold modified carbon paste electrode was studied. The Mb immobilized on the colloidal gold nanoparticles displayed a pair of redox peaks in 0.1 M pH 7.0 PBS with a formal potential of –(0.108 ± 0.002) V (vs. NHE). The response showed a surface‐controlled electrode process with an electron transfer rate constant of (26.7 ± 3.7) s −1 at scan rates from 10 to 100 mV s −1 and a diffusion‐controlled process involving the diffusion of proton at scan rates more than 100 mV s −1 . The immobilized Mb maintained its activity and could electrocatalyze the reduction of both hydrogen peroxide and nitrite. Thus, the novel renewable reagentless sensors for hydrogen peroxide and nitrite were developed, respectively. The activity of Mb with respect to the pseudo peroxidase with a K M app value of 0.65 mM could respond linearly to hydrogen peroxide concentration from 4.6 to 28 μM. The sensor exhibited a fast amperometric response to NO 2 − reduction and reached 93% of steady‐state current within 5 s. The linear range for NO 2 − determination was from 8.0 to 112 μM with a detection limit of 0.7 μM at 3 σ .
Nanobioelectrocatalysis Using Human Liver Microsomes and Cytochrome P450 Bactosomes: Pyrenyl-Nanocarbon Electrodes

Abstract:Human liver microsomes containing various drug-metabolizing cytochrome P450 (P450) enzymes, along with their NADPH-reductase bound to phospholipid membranes, were absorbed onto 1-pyrene butylamine pi-pi stacked with amine-functionalized multiwalled carbon nanotube-modified graphite electrodes. The interfaced microsomal biofilm demonstrated direct electrochemical communication with the underlying electrode surface and enhanced oxygen reduction electrocatalytic activity typical of heme enzymes such as P450s over the unmodified electrodes and nonenzymatic currents. Similar enhancements in currents were observed when the bioelectrodes were constructed with recombinant P450 2C9 (single isoform) expressed bactosomes. The designed liver microsomal and 2C9 bactosomal bioelectrodes successfully facilitated the electrocatalytic conversion of diclofenac, a drug candidate, into 4'-hydroxydiclofenac. The enzymatic electrocatalytic metabolite yield was several-fold greater on the modified electrodes than on the unmodified bulk graphite electrodes adsorbed with a microsomal or bactosomal film. The nonenzymatic metabolite production was less than the enzymatically catalyzed metabolite yield in the designed microsomal and bactosomal biofilm electrodes. To test the throughput potential of the designed biofilms, eight-electrode array configurations were tested with the microsomal and bactosomal biofilms toward electrochemical 4'-hydroxydiclofenac metabolite production from diclofenac. The stability of the designed microsomal bioelectrode was assessed using nonfaradaic impedance spectroscopy over 40 h, which indicated good stability.

Abstract:An electrochemical biointerface of 3D graphene aerogel (3DGA-AuNPs) decorated with Au nanoparticles (AuNPs) was fabricated to immobilize Cytochrome c (Cyt c) for H2O2 detection. Morphology and surface characterization of 3DGA-AuNPs reveal a successful formation of 3D-networked structure that can enhance conductivity and effectively immobilize enzyme. The immobilized Cyt c retained its bioactivity and realized the direct electron transfer on electrode owing to the biocompatibility of AuNPs and large surface area of 3DGA. The as-prepared 3DGA-AuNPs/Cyt c/GCE exhibited a pair of well-defined redox peaks of FeIII/II redox couple of Cyt c and revealed excellent catalytic activity toward H2O2 with high sensitivity of 351.57 μA·mM−1·cm−2. This biosensor was further successfully used to real-time detection of trace amount of H2O2 released from living cells, indicating the promise for practical application.

Abstract:Catalases decompose hydrogen peroxide (H2O2) into water and oxygen in an effective and environmentally friend way. The decomposition of H2O2 by nanofibrous membrane-supported catalases is reported in this work. Poly(acrylonitrile-co-N-vinyl-2-pyrrolidone) (PANCNVP) and polyacrylonitrile (PAN) blending with or without multi-walled carbon nanotubes (MWCNTs) were electrospun into nanofibrous membranes as enzyme immobilization matrixes and were carefully characterized. To immobilize catalase covalently, the nanofibrous membranes were treated with alkali solution to generate carboxyl groups followed by activation procedure with EDC/NHS. Results indicate that the nanofibrous membranes can bind large amount of catalases due to their superior surface area to volume ratio. The decomposition behavior of H2O2 with these immobilized catalases was explored. The increment of activity retention induced by the NVP moieties and/or MWCNTs is remarkable. Stabilities of the immobilized catalases including the effects of substrate pH and reaction temperature, thermal stability, operational stability and storage stability were also carefully studied. Results elucidate the improved stabilities of the nanofibrous membrane-supported catalases. In conclusion, the nanofibrous membranes electrospun from this biocompatible polymer, especially those filled with MWCNTs, are suitable matrices for catalase immobilization.
