Abstract:The purpose of this research was to prepare emulsions stabilized by mixed proteins to achieve excellent storage stability and superior functional properties. The emulsions (O/W) with uniform nanoscale droplets (∼250 nm, polydispersity index < 0.2) were stabilized by mixed sodium caseinate (SC) and soy protein isolate (SPI) and produced through high pressure homogenization. Characteristics of the emulsions were evaluated by Dynamic Light Scattering (DLS), Nanoparticle Tracking Analysis (NTA), Transmission Electron Microscopy (TEM), Rheometer and Turbiscan Lab analyzer. The stabilization mechanism was preliminarily investigated by studying the influences of pH, NaCl and SDS solutions on the droplet size, with the application of sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). It was found that both flexible protein SC and rigid globular protein SPI were adsorbed on the oil-water interface while some free protein molecules dissolved in the aqueous phase. Electrostatic repulsion and steric stabilization were contributed to the system stability. The retention ratio of vitamin A palmitate was above 93% after three months of storage at room temperature, which demonstrated the protection efficiency of the emulsion was excellent. This work showed a significant promise to develop nutritional products with satisfactory storage stability and physiological benefits.

Abstract:In this research, a simple, green and effective strategy was developed to produce long-term stable oil in water emulsion from soy protein and soy polysaccharide. Soy protein and soy polysaccharide formed dispersible complexes at pH around 3.25 aqueous solution through electrostatic and hydrophobic interactions. A high pressure homogenization produced the protein/polysaccharide complex emulsion having a droplet size about 250nm. A heat treatment of the emulsion resulted in the protein denaturation, forming irreversible oil–water interfacial films composed of soy protein/soy polysaccharide complexes. The droplets of the emulsion were characterized by dynamic light scattering, ζ-potential, transmission electron microscopy, polysaccharide digestion via pectinase, and confocal laser scanning microscopy observation via dual fluorescence probes. As a result of the polysaccharide being fixed on the droplet surface, the emulsions exhibited long-term stability in the media containing pH values of 2–8 and 0.2mol/L NaCl. The stable soy protein/soy polysaccharide complex emulsion is a suitable food-grade delivery system in which lipophilic bioactive compounds can be encapsulated.
Enhanced O/W emulsion stability and betanin protection using yeast protein and chitooligosaccharide: Comparative insights from complex coacervation and layer-by-layer methods

Abstract:Emulsions play a pivotal role in food and related industries, yet oil-in-water (O/W) emulsions, a staple emulsion, are plagued by instability issues like aggregation, flocculation, and gravitational separation during storage. Stabilizing these emulsions has often involved the use of proteins or polysaccharides. Within this context, this research delves into comparing two distinctive stabilization strategies for emulsions. The yeast protein (YP), chitooligosaccharide (COS), and betanin were employed to formulate a complexation emulsion (CE) using a complex coacervation method, and a bilayer emulsion (BE) through a layer-by-layer method. The effects of these divergent methods on the emulsion's physicochemical properties, microscopic structure, and the stability of betanin was assessed. The findings indicate that the yeast protein-chitooligosaccharide-betanin (YCB) emulsion prepared using the layer-by-layer method demonstrated the smallest droplet size and the greatest absolute ζ-potential. After a storage period of seven days, this emulsion showed the lowest emulsification index, indicating excellent storage stability. In tandem, an increase in apparent viscosity was observed, with the emulsion exhibiting weak gel-like behavior. The incorporation of betanin has improved the physical stability of emulsions, and concurrently, the emulsion presented an improved storage stability of betanin. These results provide insightful perspectives for the development of emulsions based on bilayer interfaces, contributing to the expansion of novel fungal protein applications in the fields of emulsion technology, food processing, and the stabilization of bioactive molecules.

Abstract:The forms of presentation of NPC, as related to the signs and symptoms of the disease, were studied in 215 patients over a 2-year period. A neck mass was the most common presenting symptom. Seventy percent of the patients were in Ho's stages III to V. This evidence emphasizes the need for earlier diagnosis to improve the prognosis of the disease. This earlier diagnosis could be achieved by increasing the awareness of both the patient and the clinician, particularly in countries where the incidence of NPC is high.
Preparation and Characterization of Fish Oil Pickering Emulsions Stabilized by Resveratrol-Loaded Gliadin/Chitin Nanocrystal Composite Nanoparticles

Abstract:Unsaturated fatty acids present in fish oil offer various physiological benefits to the human body. However, their susceptibility to oxidation severely limits their potential applications. The purpose of this study was to develop Pickering emulsions stabilized from a composite of resveratrol-loaded gliadin nanoparticles and oxidized chitin nanocrystals (GR/OC) to protect fish oil from oxidation. The effects of the GR/OC composite on the characterizations of fish oil Pickering emulsions were investigated, including the microstructure, physicochemical properties (stability and rheological behavior), and digestion properties in vitro. The results revealed that an increased concentration of the GR/OC composite significantly reduced the droplet size and improved the ambient stability of the emulsions (in terms of pH, ionic strength, temperature, and storage time). Confocal laser scanning microscopy images depicted that the GR/OC nanoparticles were uniformly dispersed at the interface between water and fish oil (W-O interface). This distribution formed a protective envelope around the droplets. Remarkably, the addition of 2% GR/OC nanoparticles stabilized the Pickering emulsions and showed the most positive effect on the antioxidant capacity compared to that of the control group. These stabilized emulsions maintained lower peroxide values and acid values, which were 1.5 times less than those of the blank control during the 14 day accelerated oxidation experiment. Furthermore, the Pickering emulsions stabilized by GR/OC nanoparticles exhibited the ability to protect fish oil from contamination by gastric juices and facilitate the intestinal absorption of omega-3 polyunsaturated fatty acids. The findings suggest that these GR/OC-stabilized Pickering emulsions offer a promising alternative for delivering fish oils in various industries, including the food industry.

Abstract:W/O/W emulsions can be used to encapsulate both hydrophobic and hydrophilic bioactive as nutritional products. However, studies on protein stabilized gel-like W/O/W emulsions have rarely been reported, compared to the liquid state multiple emulsions. The purpose of this study was to investigate the effect of different oil–water ratios on the stability of W/O/W emulgels fabricated with salt-soluble proteins (SSPs) of Sipunculus nudus. The physical stability, structural characteristics, rheological properties, and encapsulation stability of vitamin C and β-carotene of double emulgels were investigated. The addition of W/O primary emulsion was determined to be 10% after the characterization of the morphology of double emulsion. The results of microstructure and rheological properties showed that the stability of W/O/W emulgels increased with the increasing concentration of SSPs. Additionally, the encapsulation efficiency of vitamin C and β-carotene were more than 87%, and 99%, respectively, and still could maintain around 50% retention of the antioxidant capacity after storage for 28 days at 4 °C. The aforementioned findings demonstrate that stable W/O/W emulgels are a viable option for active ingredients with an improvement in shelf stability and protection of functional activity.

Abstract:This study aimed to prepare fish oil microcapsules by freeze-drying an emulsion co-stabilized by rice bran protein fibrils (RBPFs) and xanthan gum (XG) to improve the oxidation stability and controlled release effect. Emulsions stabilized either solely by RBPFs or unfibrillated rice bran protein (RBP) or by a combination of RBP and XG were also fabricated as microcapsule templates for comparison. The rheological properties, particle size, and zeta potential of the emulsions were examined. In addition, the characteristics of the fish oil microcapsules such as surface oil content, encapsulation efficiency, water activity, moisture content, morphological structure, oxidation stability, and digestive performance were also assessed. The rheological properties revealed that the addition of XG increased the storage modulus of the emulsion and reduced the loss modulus and apparent viscosity. At shear rates of 0-100 s-1, the fish oil emulsion did not exhibit any gel properties or shear thinning. Fibrillation increased the particle size of the fish oil emulsion, whereas adding XG reduced the droplet size. The combination of RBP fibrillation and XG addition provided the highest encapsulation efficiency for fish oil. Fibrillation reduced the water activity and moisture content of the fish oil microcapsules. The anisotropy of the fibrils and the high viscosity of XG produced a layer of wrapping on the continuous heterogeneous surface of the freeze-dried powder particles. RBPF/XG microcapsules stored at 45 °C for 1 month had the lowest peroxide value and thiobarbituric acid value, the lowest surface oil content, and the lightest yellowness. These results suggest that the combination of RBPFs and XG provides better encapsulation and protective effects for fish oil microcapsules. Upon simulated digestion, the microcapsules containing XG and RBPFs exhibited a more favorable controlled release of free fatty acids. These findings indicate that microcapsules formed from emulsions co-stabilized by XG and RBPFs are suitable for encapsulating fish oil in functional foods.

Abstract:In this study, the stability mechanism of grape seed oil/rice hydrolyzed protein emulsion was studied. The grape seed oil (10% v/v) and rice hydrolyzed protein (2% w/v) were homogenized under high pressure to prepare the emulsion. It was observed by CLSM and Multiple light scatterometer that the emulsion had long-term storage stability, and the average particle size of droplets was 0.984–1.363 μm. ζ-potential ranged from −37.733 mV to −25.633 mV. It is found that the emulsion has strong resistance to temperature, ions and other environmental factors from the macroscopic and microscopic structure, and no emulsion stratification phenomenon occurs. The composite emulsion can be used in the field of food industry and fine chemical industry, which can provide nutrition and functionality of products, its research has certain value and has a wide space for development.

Abstract:In this study, the whey protein isolate–high-methoxyl pectin (WPI-HMP) complex prepared by electrostatic interaction was utilized as an emulsifier in the preparation of docosahexaenoic acid (DHA) algal oils in order to improve their physicochemical properties and oxidation stability. The results showed that the emulsions stabilized using the WPI-HMP complex across varying oil-phase volume fractions (30–70%) exhibited consistent particle size and enhanced stability compared to emulsions stabilized solely using WPI or HMP at different ionic concentrations and heating temperatures. Furthermore, DHA algal oil emulsions stabilized using the WPI-HMP complex also showed superior storage stability, as they exhibited no discernible emulsification or oil droplet overflow and the particle size variation remained relatively minor throughout the storage at 25 °C for 30 days. The accelerated oxidation of the emulsions was assessed by measuring the rate of DHA loss, lipid hydroperoxide levels, and malondialdehyde levels. Emulsions stabilized using the WPI-HMP complex exhibited a lower rate of DHA loss and reduced levels of lipid hydroperoxides and malondialdehyde. This indicated that WPI-HMP-stabilized Pickering emulsions exhibit a greater rate of DHA retention. The excellent stability of these emulsions could prove valuable in food processing for DHA nutritional enhancement.

Abstract:Lutein is essential for infant visual and cognitive development but has low stability and solubility. This study aimed to enhance the stability and bioaccessibility of lutein using oil-in-water emulsions stabilized with biopolymers. Commercially available octenylsuccinylated (OS) starches, including capsule TA® (CTA), HI-CAP®100 (HC), and Purity Gum® 2000 (PG), along with gum Arabic (GA) variants Ticaloid acacia Max® (TAM), TICAmulsion® 3020 (TM), and pre-hydrate gum Arabic (PHGA), were chosen as emulsifiers. By screening the effect of biopolymer concentration and oil volume fraction (Φ), emulsions stabilized with CTA, HC, or TM at 20% and 30% ( w / v ) concentration and 70% Φ exhibited a gel-like structure and were selected for further assessments. After a week at 25 °C, emulsions stabilized by CTA and HC showed no significant change in droplet size, while TM emulsion exhibited a 1.58-fold increase. At 45 °C, all emulsions exhibited increase in droplet size. Lutein retention is higher in CTA emulsions at both storage temperatures than free lutein. In vitro bioaccessibility of all lutein emulsions was higher than that of free lutein. These findings highlight the superior stability and bioaccessibility of the lutein emulsion stabilized by OS starch, positioning it as a promising carrier to broaden lutein applications in infant foods.

Abstract:The aim of this study was to prepare lactoferrin-epigallocatechin-3-gallate (LF–EGCG) conjugates and to determine their ability to protect emulsified algal oil against aggregation and oxidation. LF–EGCG conjugates were formed using an ultrasound-assisted alkaline treatment. The ultrasonic treatment significantly improved the grafting efficiency of LF and EGCG and shortened the reaction time from 24 h to 40 min. Fourier transform infrared spectroscopy and circular dichroism spectroscopy analyses showed that the covalent/non-covalent complexes could be formed between LF and EGCG, with the CO and CN groups playing an important role. The formation of the conjugates reduced the α-helix content and increased the random coil content of the LF. Moreover, the antioxidant activity of LF was significantly enhanced after conjugation with EGCG. LF–EGCG conjugates as emulsifiers were better at inhibiting oil droplet aggregation and oxidation than LF alone. This study demonstrates that ultrasound-assisted formation of protein–polyphenol conjugates can enhance the functional properties of the proteins, thereby extending their application as functional ingredients in nutritionally fortified foods.

Improved Oxidative Stability of Fish Oil Emulsion by Grafted Ovalbumin-Catechin Conjugates

Abstract:Conjugates of ovalbumin (OVA) with (+)-catechin (C), (-)-epigallocatechin (EGC) or (-)-epigallocatechin gallate (EGCG) were synthesized through a free-radical grafting approach. The covalent binding between OVA and catechins was confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). OVA-catechin conjugates were further characterized by a combination of steady-state fluorescence spectroscopy, differential scanning calorimetry (DSC), Fourier transform infrared (FTIR) spectroscopy and circular dichroism (CD) analyses. Results suggested that the conjugates contained less α-helix but more β-sheet secondary structure than OVA. The catechin-grafting reaction led to improved surface hydrophobicity and decreased tertiary conformation stability of OVA. Compared with that emulsified by OVA, fish oil emulsion coated by conjugates demonstrated smaller droplet size, better storage stability and less viscosity. Besides, conjugates inhibited the lipid oxidation in fish oil emulsion more effectively than OVA because of their higher antioxidant activity and interfacial accumulation.
Fabrication of algal oil powder from pea protein-EGCG complex-stabilized emulsion template for structured antioxidant oil system

Abstract:In this work, we evaluated the potential of algal oil powders as structured antioxidant oils by stabilizing emulsion templates with pea protein (PP)- Epigallocatechin-3-gallate (EGCG) complexes. Turbidity, fluorescence, isothermal titration calorimetry (ITC) measurements indicated that the formation of PP-EGCG complexes and the interaction between PP and EGCG was caused by non-covalent binding. Furthermore, small-angle X-ray scattering (SAXS) measurement shown that the PP-EGCG complexes have smaller size and partial folding occurs internally. Compared with PP, PP-EGCG complexes have lower interfacial tension at the oil-water interface, an indication of better emulsification capacity. The physical storage stability of the emulsion was improved due to the smaller droplets of the emulsion stabilized by the PP-EGCG complexes. In addition, the agglomeration phenomenon of algal oil powder was weakened with increasing the mass ratio of EGCG/PP. The oxidation stability of the algae oil powder prepared from the PP-EGCG complex stabilized emulsion is improved, as evidenced by the decreased amount of the main oxidation product (hydrogen peroxide) and the by-product (hexanal). This study shown that PP-EGCG complexes can be used for construct stabilized emulsions templated oil powder for structured antioxidant oil system.
Use of olive and sunflower protein hydrolysates for the physical and oxidative stabilization of fish oil-in-water emulsions

Abstract:Background: Olive and sunflower seeds are by-products generated in large amounts by the plant oil industry. The technological and biological properties of plant-based substrates, especially protein hydrolysates, have increased their use as functional ingredients for food matrices. The present study evaluates the physical and oxidative stabilities of 50 g kg-1 fish oil-in-water emulsions where protein hydrolysates from olive and sunflower seeds were incorporated at 20 g kg-1 protein as natural emulsifiers. The goal was to investigate the effect of protein source (i.e. olive and sunflower seeds), enzyme (i.e. subtilisin and trypsin) and degree of hydrolysis (5%, 8% and 11%) on the ability of the hydrolysate to stabilize the emulsion and retard lipid oxidation over a 7-day storage period. Results: The plant protein hydrolysates displayed different emulsifying and antioxidant capacities when incorporated into the fish oil-in-water emulsions. The hydrolysates with degrees of hydrolysis (DH) of 5%, especially those from sunflower seed meal, provided higher physical stability, regardless of the enzymatic treatment. For example, the average D [2, 3] values for the emulsions containing sunflower subtilisin hydrolysates at DH 5% only slightly increased from 1.21 ± 0.02 μm (day 0) to 2.01 ± 0.04 μm (day 7). Moreover, the emulsions stabilized with sunflower or olive seed hydrolysates at DH 5% were stable against lipid oxidation throughout the storage experiment, with no significant variation in the oxidation indices between days 0 and 4. Conclusion: The results of the present study support the use of sunflower seed hydrolysates at DH 5% as natural emulsifiers for fish oil-in-water emulsions, providing both physical and chemical stability against lipid oxidation. © 2024 Society of Chemical Industry.

Abstract:The objectives of this study were to investigate the properties and stability of microcapsules containing fish oil co-encapsulated with phytosterol ester and limonene, prepared by spray-drying and freeze-drying methods. Whey protein isolate and soluble corn fiber were used as wall materials in the encapsulation process. The properties of microcapsules, including structure, glass transition, volatile/non-volatiles retention, microencapsulation efficiency, oxidation stability, color measurement, and sensory profiles, were evaluated after drying and during a seven-day accelerated storage trial. The finding reveals that drying methods have an effect on the retention of volatile fraction and the physical structure of the wall matrix consisted of WPI and SCF, consequently influencing the storage stability of the powders. Significantly higher retention of volatile fraction (p < 0.05) and lower surface oil were found in the spray-dried samples, resulting in the higher microencapsulation efficiency. However, samples dehydrated by both methods have good redispersion properties, showing no statistical significance (p > 0.05). The oxidation of the encapsulated oils was comparable for both spray- and freeze-dried samples during the seven-day accelerated storage trial but the loss of limonene flavor was significantly higher in the freeze-dried samples (p < 0.05). Sensory evaluation indicated that the addition of limonene could mask the unpleasant fishy odor in the co-encapsulated microcapsules. Overall, freeze drying did not produce powders with superior properties and did not show better protection towards the core materials than spray drying.

Abstract:The application of many hydrophilic and hydrophobic nutraceuticals is limited by their poor solubility, chemical stability, and/or bioaccessibility. In this study, a novel Pickering high internal phase double emulsion co-stabilized by modified pea protein isolate (PPI) and sodium alginate (SA) was developed for the co-encapsulation of model hydrophilic (riboflavin) and hydrophobic (β-carotene) nutraceuticals. Initially, the effect of emulsifier type in the external water phase on emulsion formation and stability was examined, including commercial PPI (C-PPI), C-PPI-SA complex, homogenized and ultrasonicated PPI (HU-PPI), and HU-PPI-SA complex. The encapsulation and protective effects of these double emulsions on hydrophilic riboflavin and hydrophobic β-carotene were then evaluated. The results demonstrated that the thermal and storage stabilities of the double emulsion formulated from HU-PPI-SA were high, which was attributed to the formation of a thick biopolymer coating around the oil droplets, as well as thickening of the aqueous phase. Encapsulation significantly improved the photostability of the two nutraceuticals. The double emulsion formulated from HU-PPI-SA significantly improved the in vitro bioaccessibility of β-carotene, which was mainly attributed to inhibition of its chemical degradation under simulated acidic gastric conditions. The novel delivery system may therefore be used for the development of functional foods containing multiple nutraceuticals.

Influence of emulsifier on lipid oxidation in spray-dried microencapsulated O/W emulsions

Abstract:Lipid oxidation limits the shelf-life of dried microencapsulated oils (DMOs), such as infant formula. However, it is poorly understood how lipid oxidation is affected by different types of emulsifiers. To improve our understanding, we prepared DMOs with different emulsifiers (whey protein isolate (WPI), pea protein isolate (PPI), and non-proteinaceous CITREM) and studied lipid oxidation in both the free and encapsulated fat. Only a small difference in oxidation rate was observed between these fat fractions for all formulations. We ascribed this to a non-discrete distribution of the fractions and the subsequent low fractionation selectivity as shown by Raman microscopy. The DMO with PPI showed hardly any oxidation during a 7-week incubation at 40 °C, whereas the DMOs with WPI and CITREM both reached significantly higher contents of oxidation products (lipid hydroperoxides, aldehydes, and epoxides). The enhanced stability of DMO-PPI could not be ascribed to the presence of phytic acid. In conclusion, we demonstrate the potential of using PPI to produce oxidatively stable DMOs.

Abstract:To investigate the oxidative stability and microencapsulation of algal oil. Results showed that algal oil had higher oxidative stability than fish oil, and the antioxidant could further enhance its oxidative stability. Significant differences in the microencapsulation of algal oil were observed by using dextrin of various DE in place of modified starch. Oxidative stability of microencapsulated algal oil was improved by using corn syrup of DE42 in place of 30% of modified starch. In addition, flavor cover was effective in covering off the stinking smell from the oxidation of algal oil.
