Abstract:The stability of two-layer lycopene-microemulsions and the degradation of lycopene in microemulsions subjected to thermal processing and under environmental stress were investigated. The formulation of microemulsions made with different ratios of whey protein isolate (WPI), high-methylester-pectin (HMP), and an oil phase containing lycopene was optimized. The oil volume fractions, concentrations of WPI and HMP, and their interactions significantly influenced the physical stability of lycopene-rich microemulsions. The two-layer microemulsion consisted of WPC and HMP was much more stable under environmental stress compared to the WPC one-layer microemulsions. The one-layer lycopene-microemulsions destabilized at low pH, but the two-layer lycopene-microemulsion became unstable when the pH was in the neutral range of 6.12 to 7.01. The stability of microemulsions declined with increasing NaCl concentrations. About 36.8 g/100 g, 23.2 g/100 g, and 11.1 g/100 g of the total lycopenes were lost in the oil-phase, the one-layer lycopene-microemulsions, and the two-layer lycopene-microemulsions after thermal treatments, respectively. The optimized microemulsions contained 0.2 g/100 g (w/w) whey protein concentrate (WPC), 0.5 g/100 g HMP (w/w), and 5 mL/100 mL oil phase fraction had the highest physical stability. The optimized lycopene-microemulsion was more stable to thermal treatment and changes of pH, but become sensitive to NaCl treatment.
Property and Stability of Astaxanthin Emulsion Based on Pickering Emulsion Templating with Zein and Sodium Alginate as Stabilizer

Abstract:Astaxanthin loaded Pickering emulsion with zein/sodium alginate (SA) as a stabilizer (named as APEs) was developed, and its structure and stability were characterized. The encapsulation efficiency of astaxanthin (Asta) in APEs was up to 86.7 ± 3.8%, with a mean particle size of 4.763 μm. Freeze-dried APEs showed particles stacked together under scanning electronic microscope; whereas dispersed spherical nanoparticles were observed in APEs dilution under transmission electron microscope images. Confocal laser scanning microscope images indicated that zein particles loaded with Asta were aggregated with SA coating. X-ray diffraction patterns and Fourier transform infrared spectra results showed that intermolecular hydrogen bonding, electrostatic attraction and hydrophobic effect were involved in APEs formation. APEs demonstrated non-Newtonian shear-thinning behavior and fit well to the Cross model. Compared to bare Asta extract, APEs maintained high Asta retention and antioxidant activity when heated from 50 to 10 °C. APEs showed different stability at pH (3.0-11.0) and Na+, K+, Ca2+, Cu2+ and Fe2+ conditions by visual, zeta potential and polydispersity index measurements. Additionally, the first order kinetics fit well to describe APEs degradation at pH 3.0 to 9.0, Na+, and K+ conditions. Our results suggest the potential application of Asta-loaded Pickering emulsion in food systems as a fortified additive.

Abstract:In this study, a layer-by-layer (LBL) encapsulated astaxanthin (Ast) nanoemulsion delivery system based on chitosan (CS) and tremella polysaccharide (TP) was successfully developed. The system constructed an Ast-CS-TP emulsion with high encapsulation efficiency and an excellent stability profile by utilizing the opposite charge properties of CS and TP. This study evaluated the effects of different stresses (including temperature, salt addition, pH, UV irradiation, and centrifugal force) on the emulsion's stability. To further investigate the protective mechanism of the emulsions, we performed antioxidant activity experiments after UV treatment. Additionally, an in vitro digestion experiment was conducted to assess the behavior of Ast emulsion under simulated gastrointestinal conditions. The stability correlation coefficients were calculated using the Python database Pandas. The results showed that Ast-CS-TP emulsions exhibited turbidity and enhanced homogeneity with a small particle size of around 400 nm and a high absolute zeta potential of 35 mV and exhibited excellent stability under various stresses. The Ast-CS-TP emulsions also exhibited pH-responsive release at pH ≥ 7, consistent with pH changes in the gastrointestinal tract, and were stable in highly concentrated salt solutions. We found that the CS and TP layers significantly improved the photostability of Ast. CS and TP significantly enhanced Ast's oral bioavailability. The stability correlation coeffcients showed that pH and salt concentration were the largest factors that affected the stability of the emulsion. This study provided important insights into the encapsulation and targeting of Ast, providing a theoretical foundation and technical guidance for the comprehensive utilization of Ast.

Abstract:The utilization of astaxanthin in food processing is considered to be narrow because of its substandard solubility in aqueous matrices and the instability of chemical compounds during the processing of food and the instability of chemical compounds during the processing of food. The investigation sought to evaluate multilayer emulsions stabilized by ionic interfacial layers of lupin protein isolate (LPI), ι-carrageenan (CA), and chitosan (CHI) on the physical stability of the emulsion as well as the retention of astaxanthin during the spray drying process. Primary emulsion (Pr-E) was prepared by adding LPI on oil droplet surfaces containing astaxanthin. The homogenization pressure and cycles to obtain the Pr-E were investigated. The secondary emulsion (Se-E) and tertiary emulsion (Te-E) were elaborated by mixing CA/Pr-E and CHI/Se-E, respectively. Emulsion stability was assessed under different environmental stresses (pH and NaCl). Astaxanthin retention of emulsions was determined immediately after finishing the spray-drying process. The results showed that Pr-E was stabilized with 1.0% (w/v) of LPI at 50 MPa and three cycles. Se-E and Te-E were obtained with CA/Pr-E and Se-E/CHI of 70/30 and 50/50% (w/w), respectively. The Se-E was the most stable compared to the Pr-E and Te-E when subjected to different pHs; nevertheless, once the NaCl concentration rose, no variations in the ζ-potential of all emulsions studied or destabilization were observed. The Se-E and Te-E derived provided higher astaxanthin retention (>95%) during the spray-drying process compared to Pr-E (around 88%). The results indicated that these astaxanthin multilayer emulsions show considerable potential as a functional ingredient in food products.

Abstract:The purpose of this research was to prepare emulsions stabilized by mixed proteins to achieve excellent storage stability and superior functional properties. The emulsions (O/W) with uniform nanoscale droplets (∼250 nm, polydispersity index < 0.2) were stabilized by mixed sodium caseinate (SC) and soy protein isolate (SPI) and produced through high pressure homogenization. Characteristics of the emulsions were evaluated by Dynamic Light Scattering (DLS), Nanoparticle Tracking Analysis (NTA), Transmission Electron Microscopy (TEM), Rheometer and Turbiscan Lab analyzer. The stabilization mechanism was preliminarily investigated by studying the influences of pH, NaCl and SDS solutions on the droplet size, with the application of sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). It was found that both flexible protein SC and rigid globular protein SPI were adsorbed on the oil-water interface while some free protein molecules dissolved in the aqueous phase. Electrostatic repulsion and steric stabilization were contributed to the system stability. The retention ratio of vitamin A palmitate was above 93% after three months of storage at room temperature, which demonstrated the protection efficiency of the emulsion was excellent. This work showed a significant promise to develop nutritional products with satisfactory storage stability and physiological benefits.
Pickering emulsions stabilized with spirulina protein-chitosan complex for astaxanthin delivery

Abstract:Astaxanthin (AXT) is a lipid-soluble carotenoid with good anti-oxidation, hepatic steatosis reduction, anti-inflammation and intestinal microbiota regulation ability, whose poor stability and pH vulnerability limit its bioavailability. Spirulina protein (SP) derived from Spirulina has good emulsifying ability with potential application in nutraceuticals, medicines and cosmetics. In this study, Pickering emulsions were fabricated using SP-chitosan (CS) complex as an emulsifier. The particle size, zeta potential and three-phase contact angle of SP-CS complex with different SP to CS ratios were investigated. A mass ratio of 1:2.5 SP-CS complex showed good emulsifying ability in preparing Pickering emulsion. Higher storage modulus and viscoelasticity were observed with higher SP-CS complex concentrations and oil fractions. The SP-CS Pickering emulsion significantly improved the stability and bioavailability of AXT. The results indicated that the SP-CS complex could act as a Pickering emulsion stabilizer and had the potential to deliver protective hydrophobic AXT.

The overwinter survival of ovine gastro-intestinal parasites in the Maritime provinces.

Abstract:FOR MANY YEARS parasitism has been recognized as a major cause of unthriftiness and losses in sheep in Eastern Canada (1, 13). Although new parasiticides and treatment regimes have become available, parasitism is still a serious problem in the Maritimes where sheep-raising has been a primary industry for many decades. Since 1960, the authors have been conducting investigations on parasitism in Maritime sheep, particularly on the Nova Scotia community pastures where large numbers of sheep from small farm flocks are grazed together. Originally, sheep were grazed on the same pastures during consecutive grazing seasons. This practice suggested that residual pasture infection might be an important factor in parasitic outbreaks which occurred from time to time. Griffiths (5), and Swales (14), have shown that certain ovine parasites were able to survive overwinter on pastures at Macdonald College, Quebec. Due to the close proximity of the sea Maritime weather conditions may differ somewhat from those encountered in Western Quebec. Experiments were undertaken during 1961 and 1962 to determine what species of ovine parasites survive on the pastures during Maritime winters and the role such overwinter survival might play in the development of parasitic outbreaks during the following grazing season.
Developing biopolymer-stabilized emulsions for improved stability and bioaccessibility of lutein

Abstract:Lutein is essential for infant visual and cognitive development but has low stability and solubility. This study aimed to enhance the stability and bioaccessibility of lutein using oil-in-water emulsions stabilized with biopolymers. Commercially available octenylsuccinylated (OS) starches, including capsule TA® (CTA), HI-CAP®100 (HC), and Purity Gum® 2000 (PG), along with gum Arabic (GA) variants Ticaloid acacia Max® (TAM), TICAmulsion® 3020 (TM), and pre-hydrate gum Arabic (PHGA), were chosen as emulsifiers. By screening the effect of biopolymer concentration and oil volume fraction (Φ), emulsions stabilized with CTA, HC, or TM at 20% and 30% ( w / v ) concentration and 70% Φ exhibited a gel-like structure and were selected for further assessments. After a week at 25 °C, emulsions stabilized by CTA and HC showed no significant change in droplet size, while TM emulsion exhibited a 1.58-fold increase. At 45 °C, all emulsions exhibited increase in droplet size. Lutein retention is higher in CTA emulsions at both storage temperatures than free lutein. In vitro bioaccessibility of all lutein emulsions was higher than that of free lutein. These findings highlight the superior stability and bioaccessibility of the lutein emulsion stabilized by OS starch, positioning it as a promising carrier to broaden lutein applications in infant foods.

Abstract:Emulsion is considered to be an excellent system for delivering nutraceuticals, but instability limits the application of the emulsion. Adding stabilizers is an effective method to improve the stability of the emulsion. Due to safety concerns, stabilizers from natural resources are attracting more and more attentions in food industry. Plant polysaccharides are natural biopolymers which are widely distributed in the cells of plants. Many plant polysaccharides have been shown good emulsifying and thickening effects, therefore, they are widely used as stabilizers in emulsions. The application of plant polysaccharides in improving emulsion stability was reviewed in this paper. Firstly, the factors affecting the stability of the emulsion were elaborated in detail. On this basis, the methods of improving the stability of the emulsion were discussed. Furthermore, the effects of the plant polysaccharide structures on emulsion stability were explained, such as the molecular weight of the polysaccharide, the hydrophobic group of the polysaccharide, and the protein content of the polysaccharide. Additionally, the application of several common plant polysaccharides and their derivatives in emulsions were introduced. It is hoped that this review can provide sufficient theoretical basis for application of plant polysaccharides in emulsions in the future.
Stability of high internal-phase emulsions prepared from phycocyanin and small-molecule sugars

Abstract:Background: The use of high internal-phase Pickering emulsions in the food industry is widespread due to their excellent stability and special rheological properties. Proteins are often used as food-grade Pickering stabilizers due to their safety and nutritious properties. Nowadays, the development and efficient utilization of novel proteins as Pickering stabilizers has become a new challenge. Results: Phycocyanin complexes with small-molecule sugars (SMS), formed as a result of non-thermal interactions, can serve as stabilizers for high internal-phase Pickering emulsions. The addition of SMS-enabled gel-like emulsions significantly reduced the amount of emulsifier used. When the SMS was sorbitol, the emulsion had excellent elastic properties and self-supporting ability and was stable during long-term storage, when subjected to centrifugation, and under different temperature conditions. The fluorescent property of phycocyanin was utilized to investigate the formation mechanism of the emulsion. Small-molecule sugars were able to form 'sugar-shell' structures on the surface of proteins to enhance the structural stability of proteins. Phycocyanin-SMS-stabilized emulsions provided superior protection for photosensitive and volatile substances. The retention rates of trans-resveratrol and n-hexane increased by 384.75% and 30.55%, respectively. Conclusion: These findings will encourage the development of proteins that stabilize Pickering emulsions. They will also provide new ideas for protecting photosensitive and volatile substances. © 2023 Society of Chemical Industry.

Abstract:The effects of random accelerations on the measurements of quantities in the vicinity of the liquid-gas critical point are considered when the system is in a microgravity environment. These accelerations couple to the order parameter through the transverse component of the velocity field, whose dynamics are also governed by critical point properties of the liquid-gas system. The action of the accelerations is amplified by the singular static and dynamic response of the gas-liquid system. A general formulation, based on "model H" critical dynamics allows for the calculation of a variety of quantities. It is found that the random accelerations expected in a microgravity environment will not compromise the accuracy of any experiment that is currently envisioned.

Abstract:Here, we designed an oxidized starch/β-lactoglobulin (OS/β-lg) complex colloidal particle using a dual-channel microfluidic chip for the stabilization of astaxanthin emulsion. The effect of the mixing ratio, pH, and the degree of substitution (DS) of the oxidized starch on the formation of OS/β-lg complex particles was investigated in detail. The optimal complexation occurred at a pH of 3.6, a mixing ratio of 2:10, and a DS of 0.72%, giving an ideal colloidal particle with near-neutral wettability. With this optimum agent, the astaxanthin-loaded oil-in-water emulsions were successfully prepared. The obtained emulsions showed the typical non-Newton fluid behavior, and the rheological data met the Herschel–Bulkley model. The microscopic images confirmed the dense adsorption of the particle on the oil/water interface. In vitro release and stability studies demonstrated this compact layer contributed to the controlled-release and excellent stability of astaxanthin emulsions facing heat, ultraviolet, and oxidative intervention. This work suggests the potential of microfluidics for the production of food-grade solid emulsifiers.

Abstract:This study aimed to elaborate the combination effect of polysaccharides on physicochemical properties and in vitro digestive behavior of astaxanthin (AST)-loaded Pickering emulsion gel. AST-loaded Pickering emulsion gel was prepared by heating Pickering emulsion with konjac glucomannan (KGM) and κ-carrageenan (CRG). The microstructure revealed that adding the two polysaccharides resulted in Pickering emulsion forming a network structure. It exhibited a denser and more uniform network structure, enhancing its mechanical properties four times and increasing its water-holding capacity by 20 %. In vitro digestion experiments demonstrated that the release of free fatty acids from the Pickering emulsion gel (4.25 %) was notably lower than that from conventional Pickering emulsion (17.19 %), whereas AST bioaccessibility was remarkably low at 0.003 %. It provided a feasible strategy to regulate the bioaccessibility in Pickering emulsion, which has theoretical significance to guide the current eutrophic diet people.

Abstract:The incorporation of lipophilic nutrients, such as astaxanthin (a fat soluble carotenoid) in nanodispersion systems can either increase the water solubility, stability and bioavailability or widen their applications in aqueous food and pharmaceutical formulations. In this research, gelatin and its combinations with sucrose oleate as a small molecular emulsifier, sodium caseinate (SC) as a protein and gum Arabic as a polysaccharide were used as stabilizer systems in the formation of astaxanthin nanodispersions via an emulsification-evaporation process. The results indicated that the addition of SC to gelatin in the stabilizer system could increase the chemical stability of astaxanthin nanodispersions significantly, while using a mixture of gelatin and sucrose oleate as a stabilizer led to production of nanodispersions with the smallest particle size (121.4±8.6 nm). It was also shown that a combination of gelatin and gum Arabic could produce optimal astaxanthin nanodispersions in terms of physical stability (minimum polydispersity index (PDI) and maximum zeta-potential). This study demonstrated that the mixture of surface active compounds showed higher emulsifying and stabilizing functionality compared to using them individually in the preparation of astaxanthin nanodispersions.

Abstract:Lutein is a kind of vital carotenoid with high safety and significant advantages in biological functions. However, poor water solubility and stability of lutein have limited its application. This study selected different weight ratios of sodium caseinate to acetylated mung bean starch (10:0, 9:1, 7:3, 5:5, 3:7, 1:9, and 0:10) to prepare lutein emulsions, and the microcapsules were produced by spray drying technology. The microstructure, physicochemical properties, and storage stability of microcapsules were investigated. The results show that the emulsion systems were typical non-Newtonian fluids. Lutein microcapsules were light yellow fine powder with smooth and relatively complete particle surface. The increase of sodium caseinate content led to the enhanced emulsion effect of the emulsion and the yield and solubility of microcapsules increased, and wettability and the average particle size became smaller. The encapsulation efficiency of lutein microcapsules ranged from 69.72% to 89.44%. The thermal characteristics analysis showed that the endothermic transition of lutein microcapsules occurred at about 125 °C. The microcapsules with sodium caseinate as single wall material had the worst stability. Thus, it provides a reference for expanding the application of lutein in food, biological, pharmaceutical, and other industries and improving the stability and water dispersion of other lipid-soluble active ingredients.

Abstract:Single emulsifiers exhibit varying degrees of restriction in stabilizing emulsions. Oppositely charged chitin nanoparticles and fucoidan complex particles were used as emulsifiers to stabilize a o/w Pickering emulsion and explore its stability and antioxidant activity under different environmental stresses. The results showed that the emulsion with the smallest mean particle size (1.02 μm) and strongest zeta potential (−29.3 mV) was formed at pH 7. Moreover, at this pH, it presented the highest physical stability and antioxidant activity and the lowest emulsion creaming index. The investigation of the effect of temperature on the stability and antioxidant activity of the emulsion revealed that, after freezing/thawing at −20 °C, the emulsion was unstable, the particle size increased, and the stability and antioxidant activity were low. In contrast, the emulsions treated at 25, 37, and 60 °C displayed no significant differences and exhibited high stabilities and antioxidant activities. Additionally, increasing the salt ion concentration further decreased the emulsion stability and antioxidant activity. Particularly, the emulsion with a salt concentration of 500 mM displayed the lowest stability, and stratification occurred after 30 d of storage. The Pickering emulsion remained stable under different environmental stresses expect for at a temperature of −20 °C and 500 mM salt ion concentration.

Abstract:The cotton-tipped applicator is a widely available and versatile tool for the dermatologist. In addition to the common uses of this inexpensive tool there are many other ways for the dermatologist to use cotton-tipped applicators. From its usefulness in dermatologic examination and potassium hydroxide preparations to its multiple surgical applications, the cotton-tipped applicator is a valuable and underrated tool.
Stabilization of O/W emulsions via interfacial protein concentrating induced by thermodynamic incompatibility between sarcoplasmic proteins and xanthan gum

Abstract:Thermodynamic incompatibility between sarcoplasmic proteins (SPs) and xanthan gum (XG) in aqueous solutions and enhanced emulsion stabilization induced by interfacial concentrating were discovered in this research. The phase diagram was investigated in the dependence of the concentrations of SP and XG at pH 7.2. With XG fixed at 0.5%, as the SP concentration in the solution system was increased, the mean particle size of the aqueous system decreased gradually. This might be attributed to the reduction in the aggregation of XG. The Turbiscan Stability Index and delta backscattering profiles indicted that 1% was the critical SP concentration, thermodynamic incompatibility then enhanced greatly and led to phase separation once exceeding 1%. Particle size distribution results and confocal laser scanning microscopy images revealed that in the emulsion system, the particle size distribution of the droplets and flocculation decreased as the SP concentration reached 2% (w/v). The results of interfacial properties suggested that due to the interfacial concentrating effect by XG, more SPs are adsorbed onto the oil-water interface, resulting in an increase in interfacial pressure. The decrement in ζ-potential in the presence of XG (from −23.42 mV to −21.80 mV at 2% SPs) implied that the charge stabilization effect did not dominate in the stabilization of SP/XG-based emulsions. The emulsion with 0.5% XG showed better physical stability. Our research may have potential implications for the recycle of SPs and the regulation of meat-protein-based emulsions by the interfacial concentrating effect.

Abstract:Environmentally friendly emulsifiers safe for human consumption are urgently needed to stabilize emulsions for applications in the food industry. In this study, we prepared complexes combining modified aggregated insoluble soybean protein hydrolysate (AISPH) mixed with xanthan gum (XG) (0.05-0.3 %, w/v), and further to construct water-in-oil-in-water (W/O/W) emulsions to deliver vitamin C and β-carotene. We observed a decrease in the AISPH α-helix and β-sheet content, surface hydrophobicity, and fluorescence intensity all decreased after binding. In contrast, the particle size and absolute ξ-potential significantly increased, indicating that molecular non-covalent interactions occurred in the solution. The emulsification property of AISPH was also improved by adding XG, and the AISPH-XG-stabilized emulsion showed improved stability, encapsulation efficiency, and rheological properties. Among them, AISPH-XG-0.25-stabilized emulsion exhibited a smaller particle size (8.41 ± 0.49 μm) and the highest encapsulation efficiency for vitamin C (90.03 ± 0.23 %) and β-carotene (70.56 ± 0.06 %). Additionally, simulated gastric digestion indicated that vitamin C and β-carotene bioavailability increased by 3.6 and 5.8 times, respectively. Finally, the emulsion exhibited good pH, ionic, and thermal stability. In general, AISPH-XG-stabilized W/O/W emulsions showed good stability and carrying capacity, providing a theoretical basis for improving their application.
