Abstract:This study was conducted to assess efficacy of biological control against bacterial leaf blight (BLB) of rice produced by Xanthomonas oryzae pv. oryzae. Five endophytic strains (A1, A2, A3, A13 and A15) and two rhizospherial Bacilli (D29 and H8) were tested for their antagonistic activities against BLB in vitro and in vivo. All seven strains showed high potential of antagonistic activity against X. oryzae pv. oryzae and three phytopathogenic fungi in vitro. Test of 16SrRNA gene sequence were assigned isolates A1, A3 and A13 as Bacillus amyloliquefaciens while isolates A2 and A15 as B. methylotrophicus and B. subtilis, respectively. In greenhouse, four strains of displayed 50.29%-57.86% inhibition rate against the pathogen and significantly increased plant fresh weight from 50.03% to 73.11% and dry weight from 64.11% to 86.65% in treated rice plants. In addition, these strains demonstrated strong capability to produce indole-3-acetic acid, siderophores, solubilizing phosphate and also colonize roots. Real-time quantitative polymerase chain reaction revealed that expression of defense-related genes including OsAOS2, OsJMT1, OsNPR1 and OsPR1b were significantly up-regulated in leaves of D29-exposed rice plants, suggesting that treatment of rice with D29 suppressed BLB through systemic activation of the plant defense system. Therefore, data suggest that Bacillus isolates A13, A15, D29 and H8 support effective antagonistic activity against BLB under greenhouse conditions in addition to their potential to promote growth of rice plants.
Controlling and Defence‐related Mechanisms of Bacillus Strains Against Bacterial Leaf Blight of Rice

Abstract:Bacillus strains are broadly studied for their beneficial role in plant growth and biological control of plant disease and pest; however, little is known about their underlying mechanisms. In this study, we assessed the controlling and defence-related mechanisms of three Bacillus strains including rice seed-associated strain B.subtilis A15, rhizobacterial strains B.amyloliquefaciens D29 and B.methylotrophicus H8, all of which are against bacterial leaf blight (BLB) caused by Xanthomonas oryzae pv. oryzae. Results indicated that all three strains showed strong biofilm formation ability. The culture filtrates of each strain significantly suppressed the growth and biofilm formation of X.oryzae, while changes in bacterial cell morphology such as cell swell and severe cell wall alterations were observed through the transmission electron microscopy images. PCR analysis revealed that all three strains harbour the antimicrobial-associated genes that are responsible for biosynthesis of bacillomycin, fengycin, iturin and surfactin. Subsequent real-time qPCR analysis revealed the upregulated expression of fenD and srfAA genes in D29 and H8, and fenD and ituC genes in A15 during their invitro interaction with X.oryzae. It suggests that the antibacterial mechanisms of the three strains may be at least partially associated with their ability to secrete corresponding lipopeptides. Interestingly, the applications of the three strains in greenhouse conditions were found to be effective in controlling the BLB disease, which was achieved through the activation of inducing systemic resistance resulted from the enhanced activities of defence-related enzymes. This is the first report of demonstration of the mode of antibacterial effect of Bacillus strains against X.oryzae. Overall, data from the current study provide valuable information for biological control of BLB disease in rice.
Plant Growth Promotion and Suppression of Bacterial Leaf Blight in Rice by Paenibacillus Polymyxa Sx3

Abstract:The effects and mechanisms of Paenibacillus polymyxa Sx3 on growth promotion and the suppression of bacterial leaf blight in rice were evaluated in this study. The results from a plate assay indicated that Sx3 inhibited the growth of 20 strains of Xanthomonas oryzae pv. oryzae (Xoo). Rice seedling experiments indicated that Sx3 promoted plant growth and suppressed bacterial leaf blight. In addition, bacteriologicaltests showed that Sx3 was able to fix nitrogen, solubilize phosphate and produce indole acetic acid, indicating that various mechanisms may be involved in the growth promotion by Sx3. The culture filtrate of P. polymyxa Sx3 reduced bacterial growth, biofilm formation and disrupted the cell morphology of Xoo strain GZ 0005, as indicated by the transmission and scanning electron microscopic observations. In addition, MALDI-TOF MS analysis revealed that Sx3 could biosynthesize two types of secondary metabolites fusaricidins and polymyxin P. In summary, this study clearly indicated that P. polymyxa Sx3 has strong invitro and invivo antagonistic activity against Xoo, which may be at least partially attributed to its production of secondary metabolites.
A Consortium of Rhizobacterial Strains and Biochemical Growth Elicitors Improve Cold and Drought Stress Tolerance in Rice (oryza Sativa L.).

Abstract:In the present study, a consortium of two rhizobacteria Bacillus amyloliquefaciens Bk7 and Brevibacillus laterosporus B4, termed 'BB', biochemical elicitors salicylic acid and β-aminobutyric acid (SB) and their mixture (BBSB) were investigated for cold and drought stress tolerance in rice plants. After withholding water for 16 days, rice plants treated with BBSB showed 100% survival, improved seedling height (35.4 cm), shoot number (6.12), and showed minimum symptoms of chlorosis (19%), wilting (4%), necrosis (6%) and rolling of leaves. Similarly, BB inoculation enhanced plant growth and reduced overall symptoms in rice seedlings subjected to 0 ± 5 °C for 24 h. Our results imply several mechanisms underlying BB- and BBSB-elicited stress tolerance. In contrast to the control, both treatments significantly decreased leaf monodehydroascorbate (MDA) content and electrolyte leakage, and increased leaf proline and cholorophyll content. Moreover, activities of antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT) increased 3.0- and 3.6-fold, respectively. Moreover, expression of OsMYB3R-2, OsDIL, OsDREB1A and OsCDPK13 genes was significantly up-regulated, suggesting that these genes play important roles in abiotic stress tolerance of rice. In addition, bacterial strains Bk7 and B4 were able to produce high amounts of IAA and siderophores, and colonise the plant roots, while only strain Bk7 exhibited the capability to form biofilms and solubilise inorganic phosphate. This study indicates that the BB and BBSB bio-formulations can be used to confer induced systematic tolerance and improve the health of rice plants subject to chilling and drought stress.
Efficacy of Plant Growth-Promoting Bacteria Bacillus cereus YN917 for Biocontrol of Rice Blast

Abstract:Bacillus cereus YN917, obtained from a rice leaf with remarkable antifungal activity against Magnaporthe oryzae , was reported in our previous study. The present study deciphered the possible biocontrol properties. YN917 strain exhibits multiple plant growth-promoting and disease prevention traits, including production of indole-3-acetic acid (IAA), ACC deaminase, siderophores, protease, amylase, cellulase, and β-1,3-glucanase, and harboring mineral phosphate decomposition activity. The effects of the strain YN917 on growth promotion and disease prevention were further evaluated under detached leaf and greenhouse conditions. The results revealed that B. cereus YN917 can promote seed germination and seedling plant growth. The growth status of rice plants was measured from the aspects of rice plumule, radicle lengths, plant height, stem width, root lengths, fresh weights, dry weights, and root activity when YN917 was used as inoculants. YN917 significantly reduced rice blast severity under detached leaf and greenhouse conditions. Genome analysis revealed the presence of gene clusters for biosynthesis of plant promotion and antifungal compounds, such as IAA, tryptophan, siderophores, and phenazine. In summary, YN917 can not only be used as biocontrol agents to minimize the use of chemical substances in rice blast control, but also can be developed as bio-fertilizers to promote the rice plant growth.

Abstract:Aims The aim of this study was to evaluate the antagonistic activity of halotolerant bacteria against rice brown stripe pathogen Acidovorax oryzae. Methods and Results Fifteen of 136 isolates of halotolerant bacteria exhibited strong in vitro and in vivo antagonistic activity against both strains of A. oryzae. The 15 antagonistic isolates were identified as 'operational group Bacillus amyloliquefaciens' based on physiological and biochemical features, fatty acid profiles as well as sequence analysis of 16S rRNA, gyrA and rpoB genes. Furthermore, this result indicated that the most effective antagonistic isolates K5-3 and PPB6 could produce siderophore in iron-limiting medium, and four kinds of secondary metabolites based on MALDI-TOF analysis. In addition, the culture filtrates of isolates K5-3 and PPB6 caused the damage of cell membrane evidenced by the TEM images, and resulted in 73-80% reduction in cell numbers, 55-65% reduction in biofilm formation, and 42-50% reduction in swimming ability of both strains of A. oryzae. Conclusions These isolates in particular K5-3 and PPB6 of halotolerant bacteria markedly inhibited the growth of A. oryzae. Significance and Impact of the Study To our knowledge, this is the first report on biological control of halotolerant bacteria against bacterial brown stripe of rice.
Bacillus spp., a bio-control agent enhances the activity of antioxidant defense enzymes in rice against Pyricularia oryzae

Abstract:Plant growth promoting rhizobacteria (PGPR) are found to control the plant diseases by adopting various mechanisms. Induced systemic resistance (ISR) is an important defensive strategy manifested by plants against numerous pathogens especially infecting at aerial parts. Rhizobacteria elicit ISR by inducing different pathways in plants through production of various metabolites. In the present study, potential of Bacillus spp. KFP-5, KFP-7, KFP-17 was assessed to induce antioxidant enzymes against Pyricularia oryzae infection in rice. The antagonistic Bacillus spp. significantly induced antioxidant defense enzymes i-e superoxide dismutase (1.7-1.9-fold), peroxidase (3.5-4.1-fold), polyphenol oxidase (3.0-3.8-fold), phenylalanine ammonia-lyase (3.9-4.4-fold), in rice leaves and roots under hydroponic and soil conditions respectively. Furthermore, the antagonistic Bacillus spp significantly colonized the rice plants (2.0E+00-9.1E+08) and secreted multiple biocontrol determinants like protease (1.1-5.5 U/mg of soil or U/mL of hydroponic solution), glucanase, (1.0-1.3 U/mg of soil or U/mL of hydroponic solution), siderophores (6.5-42.8 μg/mL or mg) in the rhizosphere of different rice varieties. The results showed that treatment with Bacillus spp. enhanced the antioxidant defense activities in infected rice, thus alleviating P. oryzae induced oxidative damage and suppressing blast disease incidence.

Screening and utilization of antagonistic bacteriae against rice sheath blight

Abstract:Rice seed is one of the most important resources of biocontrol agents.It's necessary for us to understand distribution of the antagonistic bacteria against sheath blight of rice under different culture pat- terns and their characteristics in order to gain satisfied biocontrol agents in Zhejiang Province.A field survey and isolation of the antagonistic bacteria was conducted.It was showed that the isolation frequency of the antagonistic bacteria from rice seeds of direct broadcast-seeding was significantly higher than those from direct row-seeding and transplanting.Thirteen strong antagonistic bacteriae against Rhizoctonia sola- ni were screened in vitro from 1000 bacterial isolates obtained from 3 different culture patterns of rice. Pathogenicity test showed that isolate S270262 produced lesions on rice plant and other 12 strains were non-pathogenicity,which were used for rice seed bacterization against R.solantiv in the screening-house and greenhouse and spraying in paddy field and identified by Biolog.The results showed the 10 strains had growth promoting effect on rice plant and 2 inhibiting the growth.Strain T300363,S300265 and S90132 had better performance in pot and field plot experiments.They were identified by Biolog as Sal- monella subsp.1 G,Pseudomonas aeruginosa and Pseudomonas spp.

Abstract:Rice (Oryza sativa L.) is a staple food crop for more than two billion people around the world. Bacterial leaf blight (BLB) caused by Xanthomonas oryzae pv. oryzae (Xoo) (Ishimiya) Swings et al. is one of the most destructive diseases of rice in all the major rice-growing. Use of antibiotics is the common practice for the management of BLB, but it has detrimental effect on human health and environment. Hence other eco-friendly strategies like biological control and host plant resistance (HPR) should be effectively exploited for BLB management. An attempt was made to manage the disease using actinobacteria isolated from rice rhizosphere. Among sixteen actinobacterial isolates, six were selected for secondary screening based on disc diffusion assay. Out of six isolates AUDT-502 produced maximum zone of inhibition (1.81 cm) followed by AUDT-508 (1.62 cm) and were evaluated under glasshouse condition. AUDT-502 (Streptomyces rimosus) appeared as the most promising bio agent which was able to inhibit the pathogen (Xoo) both in in vitro and in vivo condition. AUDT-502 increased the plant growth parameters like number of tillers (5.73), plant height (56.85 cm), root length (34.33cm) and dry root weight (3.90g) and also reduced the leaf blight lesion length and per cent disease index (4.43 per cent) significantly when compared to other treatments. The current study thus showed that there was a good correlation between in vitro inhibition of the pathogen and the in vivo control of the disease and AUDT-502 (Streptomyces rimosus) appears to be a hopeful bioagent for the management of bacterial leaf blight of rice.

Abstract:It is necessary to understand the bacterial populations associated with rice so as to provide more information and natural resources for effective management of major diseases in rice. A survey on screening and identification of gram-positive bacteria was conducted during 1998-2004. Seven hundred and fifty-six rice samples were collected from Zhejiang, Jiangsu, Fujian and Yunnan Provinces, China. Over 1000 bacterial isolates were isolated and tested for colony morphology, pathogenicity, and some characteristics of bacteriology including Gram staining, fluorescent pigment on Kings medium B and microscopic observation for endospore. Together with five standard reference strains, 74 representative gram-positive bacterial isolates were confirmed by Biolog and gas chromatographic analysis of fatty acid methyl esters. Five bacterial species of Bacillus and other three genera were identified and isolates from Bacillus sublitis and Bacillus megaterium, exhibited the most effective inhibition against the pathogens of sheath blight and bakanae disease of rice. A few isolates from Bacillus pumilus and Bacillus megaterium showed weak virulent on rice together with some virulent isolates, risk should be considered when isolates from these species were screened for biocontrol agents.
Revealing plant growth-promoting mechanisms of Bacillus strains in elevating rice growth and its interaction with salt stress

Abstract:Soil salinity is a major environmental stress that has been negatively affecting the growth and productivity of rice. However, various salt-resistant plant growth-promoting rhizobacteria (PGPR) have been known to promote plant growth and alleviate the damaging effects of salt stress via mitigating physio-biochemical and molecular characteristics. This study was conducted to examine the salt stress potential of Bacillus strains identified from harsh environments of the Qinghai-Tibetan plateau region of China. The Bacillus strains NMTD17, GBSW22, and FZB42 were screened for their response under different salt stress conditions (1, 4, 7, 9, 11, 13, and 16%). The screening analysis revealed strains NMTD17, GBSW22, and FZB42 to be high-salt tolerant, moderate-salt tolerant, and salt-sensitive, respectively. The NMTD17 strain produced a strong biofilm, followed by GBSW22 and FZB42. The expression of salt stress-related genes in selected strains was also analyzed through qPCR in various salt concentrations. Further, the Bacillus strains were used in pot experiments to study their growth-promoting ability and antioxidant activities at various concentrations (0, 100, 150, and 200 mmol). The analysis of growth-promoting traits in rice exhibited that NMTD17 had a highly significant effect and GSBW22 had a moderately significant effect in comparison with FZB42. The highly resistant strain NMTD17 that stably promoted rice plant growth was further examined for its function in the composition of rhizobacterial communities. The inoculation of NMTD17 increased the relative abundance and richness of rhizobacterial species. These outcomes propose that NMTD17 possesses the potential of PGPR traits, antioxidants enzyme activities, and reshaping the rhizobacterial community that together mitigate the harmful effects of salinity in rice plants.

Silver Nanoparticles Synthesized by Using Bacillus Cereus SZT1 Ameliorated the Damage of Bacterial Leaf Blight Pathogen in Rice

Abstract:Amongst serious biotic factors deteriorating crop yield, the most destructive pathogen of rice is Xanthomonas oryzae pv. oryzae (Xoo), which causes bacterial leaf blight (BLB) disease. This study involved targeted use of biogenic silver nanoparticles (AgNPs) to control BLB in order to cope with the disadvantages of chemical disease control. AgNPs were biologically synthesized from natively isolated Bacillus cereus strain SZT1, which was identified through 16S rRNA gene sequence analysis. Synthesis of AgNPs in bacterial culture supernatant was confirmed through UV-VIS spectroscopy. Fourier transform infrared spectroscopy (FTIR) confirmed that the existence of AgNPs was stabilized with proteins and alcoholic groups. X-ray diffraction (XRD) data revealed the crystalline nature and imaging with scanning electron microscopy (SEM) and transmission electron microscopy (TEM), showing the spherical shape of AgNPs with particle sizes ranging from 18 to 39 nm. The silver presence in AgNPs was further confirmed by energy dispersive spectra. Biogenic AgNPs showed substantial antibacterial activity (24.21 ± 1.01 mm) for Xoo. In a pot experiment, AgNPs were found to be effective weapons for BLB by significantly increasing the plant biomass with a decreased cellular concentration of reactive oxygen species and increased concentration of antioxidant enzyme activity.
Characterization of Streptomyces spp. from Rice Fields as a Potential Biocontrol Agent against Burkholderia glumae and Rice Plant Growth Promoter

Abstract:The usage of plant growth-promoting (PGP) Streptomyces to improve plant growth is an alternative to existing strategies of chemical fertilizers that commonly caused environmental pollution. The aim of this study was to isolate and characterize PGP Streptomyces from Malaysian rice fields with antagonistic activity against Bacterial Panicle Blight disease in rice caused by Burkholderia glumae. A total of 50 bacterial isolates were recovered from the soil, rhizosphere, and endosphere; 22% showed antagonistic activity against B. glumae. Molecular identification using 16S rRNA amplification and phylogenetic tree analyses revealed that the antagonistic isolates belonged to the genus Streptomyces. Among the antagonistic isolates, Streptomyces sp. TBS5 (26.35 ± 0.14 mm) demonstrated the most antagonistic activity (p < 0.05) against B. glumae. Through API® ZYM analysis, the antagonistic isolates were revealed to have phosphoric hydrolase, ester hydrolase, peptide hydrolase, and glycosidase activities that play a crucial role in plant growth promotion. The antagonistic isolates demonstrated the highest (80%) PGP traits including able to fix nitrogen and solubilize phosphate, as well as produce siderophore and indole-3-acetic acid. Plant growth promotion assay under laboratory and greenhouse conditions were analyzed with the treatment of rice, Oryza sativa, seeds with the antagonistic isolates. Seeds treated with Streptomyces sp. TBS5, JAS2, R2-7, and TKR8 showed improvement in vigor index by more than 100% compared to water-treated control plants under both conditions. Augmentation of root length showed an overall increment of more than 101.5% under laboratory condition and 151.9% under greenhouse condition using Streptomyces sp. TBS5, JAS2, R2-7, and TKR8 treatments. Results suggest that Streptomyces sp. TBS5, JAS2, R2-7, and TKR8 are a good candidate to be developed as both biocontrol agent against B. glumae and PGP agent in rice.
Evaluation of Biocontrol Activities of Streptomyces spp. against Rice Blast Disease Fungi

Abstract:Rhizosphere bacteria can positively influence plant growth by direct and indirect mechanisms. A total of 112 bacterial strains were isolated from the rhizosphere of rice and tested for plant beneficial activities such as siderophore production, cell-wall-degrading enzyme production, hydrogen cyanide (HCN) production and antifungal activity against rice blast disease fungus. The actinomycetes count was 3.8 × 106 CFU/g soil. Streptomyces strains PC 12, D 4.1, D 4.3 and W1 showed strong growth inhibition of blast disease fungus, Pyricularia sp. (87.3%, 82.2%, 80.0% and 80.5%) in vitro. Greenhouse experiments revealed that rice plants treated with Streptomyces strain PC 12 recorded maximum plant height, root length and root dry weight compared to the control. Taxonomic characterization of this strain on the basis of 16S rRNA gene sequence led to its identification as Streptomyces palmae PC 12. Streptomyces palmae PC 12 may be used as biofertilizer to enhance the growth and productivity of commercially important rice cultivar RD6 and the biocontrol of blast disease fungus.

Abstract:Bacterial leaf blight (BLB) disease caused by Xanthomonas oryzae pv. oryzae ( Xoo ) is one of the most destructive pathogens responsible for severe yield losses in rice. Application of bacterial biocontrol agent (BCA) with plant growth promotion (PGP) abilities is a promising method that alternates current conventional practices to manage BLB disease and increase crop yield. Hence, this study aimed to isolate and identify BCA with PGP abilities from internal plant tissues and rhizosphere soil of healthy and Xoo -infected rice and evaluate their antagonistic and PGP properties under greenhouse and field conditions.

Abstract:The application of beneficial bacteria has recently been used for sustainable agriculture. In current research, 71 bacterial isolates were obtained from rice plant and the rhizosphere soil of different paddy fields in Guilan province, Iran. After primitive investigation, 40 bacteria with typical predominant characteristics were selected. By PCR-RFLP of their 16S r-DNA gene, 8 Operational Taxonomic Units (OTUs) totally consisted of 33 isolates were obtained. From all of them, 8 isolates were selected for rice seed germination experiment, then, effective isolates were used for pot experiment to evaluate their ability for promoting rice growth. All of them were able to increase rice growth and yield, but in different potential. These tested isolates were identified as Alcaligenes faecalis (DEp8, O1R4), Pantoea ananatis (AEn1), Bacillus vietnamensis (MR5), Bacillus idriensis (MR2) and Stenotrophomonas maltophilia by partial sequencing of their 16S r-DNA gene. Among them, AEn1 and MR5 produced indole-3- acetic acid (IAA) in larger amounts than the other isolates and the isolates AEn1 and O1R4 were able to solubilize phosphate in higher amounts. According to the results obtained, it can be concluded that AEn1, O1R4 and MR5 can be considered as bacterial inoculants to use as alternatives for chemical fertilizers.

Abstract:Rice false smut, which is caused by the soil-borne fungal pathogen Ustilaginoidea virens ( U. virens ), is one of the most threatening diseases in most of the rice-growing countries including India that causes 0.5–75% yield loss, low seed germination, and a reduction in seed quality. The assessment of yield loss helps to understand the relevance of disease severity and facilitates the implementation of appropriate management strategies. This study aimed to mitigate biotic stress in rice by employing a rhizobacterial-based bioformulation, which possesses diverse capabilities as both a plant growth promoter and a biocontrol agent against U. virens . Rhizobacteria were isolated from the soil of the rice rhizospheres from the healthy plant of the false smut affected zone. Furthermore, they were identified as Bacillus strains: B. subtilis (BR_4), B. licheniformis (BU_7), B. licheniformis (BU_8), and B. vallismortis (KU_7) via sequencing. Isolates were screened for their biocontrol potential against U. virens under in vitro conditions. The antagonistic study revealed that B. vallismortis (KU_7) inhibited U. virens the most (44.6%), followed by B. subtilis BR_4 (41.4%), B. licheniformis BU_7 (39.8%), and B. licheniformis BU_8 (43.5%). Various biochemical and plant growth promoting attributes, such as phosphate and Zn solubilization, IAA, ammonium, siderophore, and chitinase production, were also investigated for all the selected isolates. Furthermore, the potential of the isolates was tested in both in vitro and field conditions by employing talc-based bioformulation through bio-priming and root treatment. The application of bioformulation revealed a 20% decrease in disease incidence in plants treated with B. vallismortis (KU_7), a 60.5% increase in the biological yield, and a 45% increase in the grain yield. This eco-friendly approach not only controlled the disease but also improved the grain quality and reduced the chaffiness.

Broad-spectrum antagonistic potential of Bacillus spp. volatiles against Rhizoctonia solani and Xanthomonas oryzae pv. oryzae

Abstract:Rhizoctonia solani and Xanthomonas oryzae pv. oryzae (Xoo) are the two major diseases affecting the quality and quantity of rice production. In the current study, volatile organic compounds (VOCs) of Bacillus spp. were used as green biocontrol agents for plant diseases. In in vitro experiments, Bacillus spp. FZB42, NMTD17, and LLTC93-VOCs displayed strong antimicrobial volatile activity with inhibition rates of 76, 66, and 78% for R. solani and 78, 81, and 76% for Xoo, respectively, compared to control. The synthetic VOCs, namely Pentadecane (PDC), Benzaldehyde (BDH), 1,2-Benz isothiazol-3(2H)-one (1,2-BIT), and mixture (MIX) of VOCs showed high volatile activity with inhibition rates of 86, 86, 89, and 92% against R. solani and 81, 81, 82, and 86%, respectively, against Xoo as compared to control. In addition, the scanning and transmission electron microscopes (SEM and TEM) analyses were performed to examine the effect of Bacillus and synthetic VOC treatments on R. solani and Xoo morphology. The analysis revealed the deformed and irregularized morphology of R. solani mycelia and Xoo cells after VOC treatments. The microscopic analysis showed that the rapid inhibition was due to severe oxidative productions inside the R. solani mycelia and Xoo cells. By using molecular docking, it was determined that the synthetic VOCs entered the active binding site of trehalase and NADH dehydrogenase proteins, causing R. solani and Xoo cells to die prematurely and an accumulation of ROS. In the greenhouse experiment, FZB42, NMTD17, and LLTC93-VOCs significantly reduced the lesions of R. solani 8, 7, and 6 cm, and Xoo 7, 6, and 6 cm, respectively, then control. The synthetic VOCs demonstrated that the PDC, BDH, 1,2-BIT, and MIX-VOCs significantly reduced R. solani lesions on leaves 6, 6, 6, and 5 cm and Xoo 6, 5, 5, and 4 cm, respectively, as compared to control. Furthermore, plant defence-related genes and antioxidant enzymes were upregulated in rice plants. These findings provide novel mechanisms by which Bacillus antimicrobial VOCs control plant diseases.
Isolation and Characterization of Bacillus Consortia for Plant Growth Promotion in Rice (Oryza sativa L.)

Abstract:In this study, soil samples from the rhizosphere of various uncultivated weeds were collected from fifteen different locations of Gujarat. Heat treatment was given at 65°C for 20 minutes prior to initial screening for spore-forming Bacillus spp. Among them, 20 nitrogen-fixing (NFB), 27 phosphate solubilising (PSB) and 15 potassium mobilizing (KMB) isolates were screened primarily. After molecular identification only Bacillus isolates were further selected and characterized. Three superior Bacillus isolates were selected from each category by secondary screening. All isolates belonging to different category were compatible in nature and showed significant ammonia production, ARA, phosphate solubilisation, potassium mobilization, siderophore production, IAA production and organic acid producers. The relative expression analysis of three genes NRT2.1 (Nitrate transporter), PT6 (Phosphorus transporter), and AKT1 (Potassium transporter) at transcriptional level were performed in the juvenile root tissues of Rice using qRT-PCR technique at 15th, 30th and 45th days after transplanting with Actin as a internal control. Rice transplants were inoculated with different NPK fixing Bacillus consortia, which significantly improved growth parameters as compared to control in field study. Various growth and yield attributing characters of rice viz., plant height (at 60 DAT, 90 DAT, and at harvesting), number of productive tillers, days to 50 % flowering, 100 seed weight, grain yield, straw yield, as well as dry biomass per hectare were significantly influenced by different treatments. An application of 100 % RDF with NPK consortia (T7) treatment of rice crop resulted in significantly higher plant height (49.25 cm at 60 DAT, 107.00 cm at 90 DAT, 129.50 cm at harvesting), days to 50% flowering (75.63), 100 seed weight (3.53 g), number of productive tillers per plant (12.13), grain yield (6889.00 kg ha-1), straw yield (8754.25 kg ha-1) and dry biomass (7889.25 kg ha-1) followed by the treatments T4 (T2 + N consortia), T5 (T2 + P consortia), T6 (T2 + K consortia), and T2 (RDF (NP) 100%). Nutrient content in grain and straw was differing significantly due to different treatments. However, significantly higher N, P, K, content in straw (1.28 %, 0.15 %, and 1.52 %) and grain (2.06 %, 0.23 %, and 0.25 %) respectively, were recorded under the treatment T7. The spore forming Bacillus consortia was able to survive at a wide range of temperature and pH fluctuations and found to be effective as N-fixers, P-solubilizers, K-mobilizers, siderophore producers, IAA producers with having antagonistic activity against rice pathogen Magnaportha oryzae.
