Abstract:Rice (Oryza sativa L.) is one of the most popular cereal crops, being consumed by almost half of the world’s population. Among several cultivars widely distributed in Thailand, Maled Phai is a Thai pigmented-upland rice with exceptionally high nutritional value and high demand in the local Thai market. This study aimed to investigate the feasibility of producing plant growth-promoting properties (PGP) and enhancing the accumulation of phytochemicals in Maled Phai rice seeds of endophytic fungi isolated from upland black rice. Among a total of 56 isolates, the 4 most effective PGP isolates were identified as Trichoderma zelobreve PBMP16, Talaromyces pinophilus PBMP28, Aspergillus flavus KKMP34, and Trichoderma sp. PBMP4 based on their morphological characteristics and multigene (ITS, rpb2, tef-1, CaM, and BenA) phylogenetic analyses. These four endophytic fungi could promote plant growth parameters under greenhouse conditions. Outstandingly, upland rice inoculated with Tr. zelobreve PBMP16 had a significant increase in total seed weight, root length, phenolic compounds, anthocyanin, antioxidants, and N uptake, which were higher than those of the noninoculated control, and even better than the chemical fertilizer. Overall, this report shows that endophytic fungi efficiently promoted growth and increased the phenolic compounds, anthocyanin, and antioxidants of Maled Phai rice.

Abstract:Utilization of plant-associated beneficial microorganisms, particularly endophytes, is growing, as they occupy a relatively privileged niche within various plant tissues, where they face less competition for food and shelter than in the rhizo-sphere. The plant and these beneficent endophytes form a harmonious interaction that has profound effects on the host plant’s physiological and developmental process in its native habitat. The purpose of this study aims to emphasize the endophytic fungi found on flourishing rice seedlings as well as to investigate the preordained factors affecting the growth of the plant. 56 endophytic fungi were isolated from various rice plant tissues and their percent of dominance was determined. The fungal strains were assessed for in-vitro mechanisms that boost plant growth-promoting activities studies.Out of 14 isolates, 92.85% were able to produce Gibberellic acid, 7.14% ammonia production, 71.42% for phosphate solubilization activity, 92.85% for zinc solubilization activity, 21.42% for potassium solubilization activity, 7.14% for HCN production and 64.28% for siderophore development, indicated the potency to promote crop growth enhancement. Therefore, endophytic fungi isolated from rice plants have the potential to be a fortuitous bio-stimulant; further research into this area is required to aid in bridging the void in agricultural and biotechnological prospects.

Abstract:The present investigation was carried out during 2021-22 at Acharya Narendra Deva University of Agriculture & Technology, Kumarganj, Ayodhya (Uttar Pradesh). Results revealed that phenotypic characterization of plant growth promoting microbial isolates from rice rhizosphere and phyllosphere have significant effects on physicochemical properties of soil. On the other hand, microbial isolates have a significant effect on microbial biomass carbon (MBC) and total protein content of the soil. Applying microbial isolates has a positive impact on microbial population in terms of total rhizospheric bacterial population (0.77 - 8.4×106 cfu g-1 soil). Whereas, fungal population (1.0 – 51.0 ×103 sfu g-1 and actinomycetes (1.7- 97 ×103 cfu g-1 of soil). Whereas, total number of phyllospheric bacterial population of rice plant leaves and clumps were 0.53- 6.4×106 cfu g-1, leaf fungal population 1.0- 73 ×102 sfu g-1, leaf Actinomycetes population 0.67- 1.2 ×103 cfu g-1. A total of 6 bacterial stains, 2 Actinomycetes and 3 fungal strains were isolated from the rhizospheric soil. Similarly, 4 bacterial stains, 2 Actinomycetes and 2 fungal strains were also isolated from the phyllosphere of the rice field by serial dilution effect on plating techniques. Plant growth promoting (PGP) traits were evaluated. The highest IAA (Indole Acetic Acid) production (23.75μg ml-1) was recorded under the RRS5 (Rice Rhizospheric Sample 5) and followed by RRS3 which was produced at 18.76 μgml-1. The isolated microorganisms can be used as an effective bio inoculant either individually or in different combinations for the formulation of different multi potent biofertilizers for plant growth promotion substances as well as control of plant diseases in rice crop.

Abstract:Rapid urbanization and globalization demand increasing agricultural productivity. Soil nutrient supply capacity is continuously decreasing due to soil erosion, degradation, salt deposition, undesired element, metal deposition, water scarcity, and an uneven nutrient delivery system. Rice cultivation requires a large amount of water which is becoming detrimental due to these activities. There is a need to increase its productivity. Microbial inoculants are becoming increasingly important in achieving sustainable agricultural production systems. The current study was conducted to investigate the interaction between the root endophytic fungus Serendipita indica ( S. indica ) and the actinobacterium Zhihengliuella sp. ISTPL4 (Z. sp. ISTPL4) and their synergistic effects on the growth of rice (Oryza sativa L). Both S. indica and Z. sp. ISTPL4 showed positive interactions. Growth of S. indica was observed at different days after Z. sp. ISTPL4 inoculation, and stimulated growth of S. indica was observed when Z. sp. ISTPL4 was inoculated at 5 dafi (days after fungal inoculation). Z. sp. ISTPL4 promoted the growth of S. indica as it increased spore germination. Furthermore, confocal and scanning electron microscopy (SEM) analyses showed a 27% increase in the spore size of S. indica in the presence of Z. sp. ISTPL4. In a liquid chromatography with tandem mass spectrometry (LC-MS/MS) analysis increased production of alanine and glutamic acid was observed in their sequential co-culture as compared with individual cultures. Sequential inoculation of S. indica and Z. sp. ISTPL4 significantly increased the biochemical and physical characteristics of rice as compared with their individual inoculum. Biochemical parameters such as chlorophyll content, total soluble sugar, and flavonoid content in the rice increased by up to 57%, 47%, and 39%, respectively, in the presence of the combined inoculum of S. indica and Z. sp. ISTPL4. This will be the first study, to the best of our knowledge, which shows the fungus and actinobacterium interaction and their synergistic roles in the growth promotion of rice. Furthermore, this novel combination can also be used to boost the growth of other crops to increase the agricultural yield.

Abstract:This study investigated the combined effects of novel plant growth-promoting rhizobacteria (PGPR)—Agrobacterium pusense NC2, Kosakonia oryzae WN104, and Phytobacter sp. WL65—and Ascophyllum nodosum seaweed extract (ANE) as biostimulants (PGPR-ANE) on rice growth, yield, and rhizosphere bacterial communities using the RD79 cultivar. The biostimulants significantly enhanced plant growth, shoot and root length, and seedling vigour; however, seed germination was not affected. In pot experiments, biostimulant application significantly increased the richness and evenness of bacterial communities in the rhizosphere, resulting in improvements in rice growth and yield, with increases in plant height (9.6–17.7%), panicle length (14.3–17.9%), and seeds per panicle (48.0–53.0%). Notably, biostimulant treatments also increased post-harvest soil nutrient levels, with nitrogen increasing by 7.7–19.2%, phosphorus by 43.4–161.4%, and potassium by 16.9–70.4% compared to the control. Principal coordinate analysis revealed distinct differences in bacterial composition between the tillering and harvesting stages, as well as between biostimulant treatments and the control. Beneficial bacterial families, including Xanthobacteraceae, Beijerinckiaceae, Acetobacteraceae, Acidobacteriaceae, and Hyphomicrobiaceae, increased in number from the tillering to harvesting stages, likely contributing to soil health improvements. Conversely, methanogenic bacterial families, such as Methanobacteriaceae and Methanosarcinaceae, decreased in number compared to the control. These findings highlight the dynamic responses of the rhizosphere microbiome to biostimulant treatments and underscore their potential benefits for promoting sustainable and productive agriculture.

Abstract:Biofertilizers are currently considered the only alternative of chemical fertilizers. In addition, improving the effect of biofertilizers can make chemical fertilizers less demanded, thereby leading to the reduction of adverse environmental effects. In the present study, three rice rhizosphere bacteria (Bacillus megaterium DD-2, Bacillus aryabhattai DD-3 and Bacillus subtilis DD-4) were isolated and then identified from the soil of a farm in Northeast China. As revealed from the results, all isolates exhibited the ability to dissolve potassium and phosphorus. To be specific, DD-4 was capable of dissolving potassium to the greatest extent (1.37 mg/L), DD-2 could maximally dissolve inorganic phosphorus (288.33 mg/L) and DD-3 exhibited the maximum solubility of organic phosphorus (31.58 mg/L). All isolates were capable of synthesizing Indole-3-acetic acid (IAA) (55.66 ~ 75.89 mg/L), Gibberellic acid (GA) (16.33 ~ 23.58 mg/L) and siderophores (38.77 ~ 52.88%). As suggested from the results of the pot experiment, the isolates were capable of promoting the shoot and underground part of rice seedlings. With the increase in the concentration of isolates and the time of addition, the overall effect of the isolates to boost rice seedling growth first increased and then leveled off. The mentioned isolates were observed to exhibit great potential for being biologically applied. However, the impact of isolates on rice required more specific assessments under biotic and abiotic stress before being recommended as biofertilizers.

Abstract:Effect of indole acetic acid (IAA)-overproducing mutants of Burkholderia cepacia (RRE25), a member of β-subclass of Proteobacteria and naturally occurring rice endophyte, was observed on the growth of rice (Oryza sativa L.) plants grown under greenhouse conditions. Nine mutants were characterized for altered biosynthesis of IAA after nitrous acid mutagenesis. These mutants were grouped into two classes: class I mutants have reduced production of IAA as compared to the wild type, while class II mutants showed overproduction of IAA. Mutants of both classes and RRE25, the parent (wild type), were inoculated on rice seedlings of two cultivars (Sarjoo-52 and NDR-97). Uptake of nitrogen, phosphorous, and potassium was estimated in these plants. Significant increase in the amount of uptake of all three elements was observed when inoculated with the IAA-overproducing mutants over control as well as in the plants inoculated with the wild type (RRE25). Effect of inoculation of IAA-overproducing mutants was more pronounced on the uptake of phosphorous in cultivar NDR-97 than Sarjoo-52, while it was opposite with respect to potassium uptake. Any significant difference was not observed in nitrogen uptake among the two cultivars. It shows that the host also plays an important role in the beneficial endophytic association. It was concluded from these results that one of the possible mechanisms of growth promotion of rice plants inoculated with bacterial endophytes is their effects on an increase in the capability of nutritional uptake possible through the effect of IAA production which results in proliferation of root system that could mine more nutrients from the soil.

Abstract:Endophytic bacteria, recognized as eco-friendly biofertilizers, have demonstrated the potential to enhance crop growth and yield. While the plant growth-promoting effects of endophytic bacteria have been extensively studied, the impact of weed endophytes remains less explored. In this study, we aimed to isolate endophytic bacteria from native weeds and assess their plant growth-promoting abilities in rice under varying chemical fertilization. The evaluation encompassed measurements of mineral phosphate and potash solubilization, as well as indole-3-acetic acid (IAA) production activity by the selected isolates. Two promising strains, tentatively identified as Alcaligenes faecalis (BTCP01) from Eleusine indica (Goose grass) and Metabacillus indicus (BTDR03) from Cynodon dactylon (Bermuda grass) based on 16S rRNA gene phylogeny, exhibited noteworthy phosphate and potassium solubilization activity, respectively. BTCP01 demonstrated superior phosphate solubilizing activity, while BTDR03 exhibited the highest potassium (K) solubilizing activity. Both isolates synthesized IAA in the presence of L-tryptophan, with the detection of nifH and ipdC genes in their genomes. Application of isolates BTCP01 and BTDR03 through root dipping and spraying at the flowering stage significantly enhanced the agronomic performance of rice variety CV. BRRI dhan29. Notably, combining both strains with 50% of recommended N, P, and K fertilizer doses led to a substantial increase in rice grain yields compared to control plants receiving 100% of recommended doses. Taken together, our results indicate that weed endophytic bacterial strains BTCP01 and BTDR03 hold promise as biofertilizers, potentially reducing the dependency on chemical fertilizers by up to 50%, thereby fostering sustainable rice production.

Abstract:Plant‐growth‐promoting rhizobacteria (PGPR) should effectively colonize along the plant root to enhance the plant and soil health. The present investigation aims to improve the PGPR‐mediated plant health benefits through above‐ground foliar management. A green fluorescent protein‐tagged PGPR strain, Pseudomonas chlororaphis (ZSB15‐M2) was inoculated in a nonautoclaved agricultural soil before rice culturing. Salicylic acid and cell extracts of Corynebacterium glutamicum and Saccharomyces cerevisiae as a supply of hormonal and inducer compounds were applied on the foliage of the 10‐days‐old rice plants and subsequently observed the colonizing ability of ZSB15‐M2. The cell extracts of Corynebacteria and yeast showed a 100‐fold increase in the ZSB15‐M2 population in the rhizosphere of rice, whereas salicylic acid had a 10‐fold increase in relation to mock control. The rice root exudates collected after the spraying of salicylic acid and microbial extracts showed significantly enhanced release of total carbon, total protein, total sugar, total amino nitrogen, total nitrogen, and phenol content. In vitro assays revealed that these root exudates collected after exogenous spray of these chemicals enhanced the chemotactic motility and biofilm formation of ZSB15‐M2 compared to the control plant's root exudate. Metabolomic analysis of root exudates collected from these rice plants by gas chromatography–mass spectrometry revealed that the Corynebacteria and yeast cell extracts enhanced the divergence of metabolites of rice root exudate. Further, due to these cumulative effects in the rice rhizosphere, the total chlorophyll, total protein, total nitrogen, and total phosphorus of rice were significantly improved. These observations provide insights into the rhizosphere functioning of rice plants as modulated by above‐ground treatments with improved colonization of inoculant strains as well as the plant growth.

Abstract:Rice (Oryza sativa L.) is an important crop that is grown worldwide to supply the world’s expanding food demand. In the current study, the effects of plant growth-promoting rhizobacteria (PGPR) and Arbuscular mycorrhizal fungi (AMF) on soil fertility and rice growth were explored. Rice plants were inoculated to evaluate how AMF fungi and PGPR affect various aspects of soil and plants, implicating abiotic stress tolerances. The experiment was carried out in a completely randomized design with three replicates under the controlled conditions. Results depicted that the plants that were inoculated with a mixture of AMF and PGPR had better yields and nutritional concentrations, while both AMF and PGPR lowered soil pH and organic matter differently. Similarly, AMF and PGPR treatments significantly increased the amount of N, P, K, and B in the post-harvest soil. The PGPR-inoculated plants had a 10–40% higher buildup of N in their tissues. Similarly, when they were compared with non-infected plants, AMF-inoculated treatments demonstrated a greater N accumulation in the rice tissue. The maximum P content in plant tissues was 0.149% in PGPR5-infected plants, either alone or in combination with AMF. In T12, AMF + PGPR5 inoculated rice plants, the maximum K uptake was 1.98%, which was 54% higher than the control treatment. The sole application of AMF raised K buildup in rice tissues by 38% compared with the control treatment. The improved productivity of plants with AMF and PGPR (especially with PGPR5) was attributed to the increased availability of nutrients in the soil. As a result, rice plant growth, yield, and essential element uptakes were boosted significantly. The present study’s results suggested using the combined application of AMF + PGPR5 for improving the rice yield and for sustaining the soil health.

Abstract:This study was conducted to assess efficacy of biological control against bacterial leaf blight (BLB) of rice produced by Xanthomonas oryzae pv. oryzae. Five endophytic strains (A1, A2, A3, A13 and A15) and two rhizospherial Bacilli (D29 and H8) were tested for their antagonistic activities against BLB in vitro and in vivo. All seven strains showed high potential of antagonistic activity against X. oryzae pv. oryzae and three phytopathogenic fungi in vitro. Test of 16SrRNA gene sequence were assigned isolates A1, A3 and A13 as Bacillus amyloliquefaciens while isolates A2 and A15 as B. methylotrophicus and B. subtilis, respectively. In greenhouse, four strains of displayed 50.29%-57.86% inhibition rate against the pathogen and significantly increased plant fresh weight from 50.03% to 73.11% and dry weight from 64.11% to 86.65% in treated rice plants. In addition, these strains demonstrated strong capability to produce indole-3-acetic acid, siderophores, solubilizing phosphate and also colonize roots. Real-time quantitative polymerase chain reaction revealed that expression of defense-related genes including OsAOS2, OsJMT1, OsNPR1 and OsPR1b were significantly up-regulated in leaves of D29-exposed rice plants, suggesting that treatment of rice with D29 suppressed BLB through systemic activation of the plant defense system. Therefore, data suggest that Bacillus isolates A13, A15, D29 and H8 support effective antagonistic activity against BLB under greenhouse conditions in addition to their potential to promote growth of rice plants.
A Consortium of Rhizobacterial Strains and Biochemical Growth Elicitors Improve Cold and Drought Stress Tolerance in Rice (oryza Sativa L.).

Abstract:In the present study, a consortium of two rhizobacteria Bacillus amyloliquefaciens Bk7 and Brevibacillus laterosporus B4, termed 'BB', biochemical elicitors salicylic acid and β-aminobutyric acid (SB) and their mixture (BBSB) were investigated for cold and drought stress tolerance in rice plants. After withholding water for 16 days, rice plants treated with BBSB showed 100% survival, improved seedling height (35.4 cm), shoot number (6.12), and showed minimum symptoms of chlorosis (19%), wilting (4%), necrosis (6%) and rolling of leaves. Similarly, BB inoculation enhanced plant growth and reduced overall symptoms in rice seedlings subjected to 0 ± 5 °C for 24 h. Our results imply several mechanisms underlying BB- and BBSB-elicited stress tolerance. In contrast to the control, both treatments significantly decreased leaf monodehydroascorbate (MDA) content and electrolyte leakage, and increased leaf proline and cholorophyll content. Moreover, activities of antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT) increased 3.0- and 3.6-fold, respectively. Moreover, expression of OsMYB3R-2, OsDIL, OsDREB1A and OsCDPK13 genes was significantly up-regulated, suggesting that these genes play important roles in abiotic stress tolerance of rice. In addition, bacterial strains Bk7 and B4 were able to produce high amounts of IAA and siderophores, and colonise the plant roots, while only strain Bk7 exhibited the capability to form biofilms and solubilise inorganic phosphate. This study indicates that the BB and BBSB bio-formulations can be used to confer induced systematic tolerance and improve the health of rice plants subject to chilling and drought stress.

Abstract:AimsWe aimed to identify plant growth-promoting rhizobacteria that could be used to develop a biofertilizer for rice.MethodsTo obtain plant growth-promoting rhizobacteria, rhizosphere soils from different crops (rice, wheat, oats, crabgrass, maize, ryegrass, and sweet potato) were inoculated to rice plants. Moreover, genetic analysis was carried out to evaluate their phylogenetic relationships based on 16S rRNA sequence data.ResultsStrains of Bacillus altitudinis, Pseudomonas monteilii, and Pseudomonas mandelii formed associations with rice plants and fixed nitrogen. A strain of Rhizobium daejeonense showed nitrogen fixation activity in an in vitro assay and in vivo. Strains of B. altitudinis and R. daejeonense derived from rice rhizosphere soil, strains of P. monteilii and Enterobacter cloacae derived from wheat rhizosphere soil, and a strain of Bacillus pumilus derived from maize rhizosphere soil significantly promoted rice plant growth.ConclusionsThese methods are effective to identify candidate species that could be developed as biofertilizers for target crops.

Abstract:IntroductionBiological control holds great promise for environmentally friendly and sustainable management of the phytopathogens. The multi-function features of plant growth-promoting rhizobacteria (PGPR) enable to protect the plants from disease infections by replacing the chemical inputs. The interaction between the plant root exudates and the microbes stimulates the production of secondary metabolism and enzymes and induces systemic resistance in the plants.AimThe aim was to identify the potential PGPR which would show an antagonistic effect against basmati rice fungal and bacterial diseases.MethodsIn the study, native originating microbes have been isolated, characterized using 16S rRNA sequencing, and used as potential antagonistic microbial isolates against diseases of rice plants.ResultsRhizobacteria isolated from rhizosphere, endo-rhizosphere, and bulk soil samples of Basmati 370 exhibited promising inhibitory activity against rice pathogens. Molecular characterization of bacterial isolates based on 16S rRNA sequencing classified the bacterial isolates into different genera such as Bacillus, Pseudomonas, Streptomyces, Exiguobacterium, Aeromonas, Chryseobacterium, Enterobacter, and Stenotrophomonas. PGPRs exhibited biocontrol activities against various rice diseases like bacterial leaf blight, leaf blast, brown spot, and sheath blight and boost the plant growth traits.ConclusionIn the study, the potentially identified PGPRs isolates could be used as efficient bioinoculants as bio-fertilizers and biocontrol agents for sustainable rice crop production.

Isolation and Characterization of Bacillus Consortia for Plant Growth Promotion in Rice (Oryza sativa L.)

Abstract:In this study, soil samples from the rhizosphere of various uncultivated weeds were collected from fifteen different locations of Gujarat. Heat treatment was given at 65°C for 20 minutes prior to initial screening for spore-forming Bacillus spp. Among them, 20 nitrogen-fixing (NFB), 27 phosphate solubilising (PSB) and 15 potassium mobilizing (KMB) isolates were screened primarily. After molecular identification only Bacillus isolates were further selected and characterized. Three superior Bacillus isolates were selected from each category by secondary screening. All isolates belonging to different category were compatible in nature and showed significant ammonia production, ARA, phosphate solubilisation, potassium mobilization, siderophore production, IAA production and organic acid producers. The relative expression analysis of three genes NRT2.1 (Nitrate transporter), PT6 (Phosphorus transporter), and AKT1 (Potassium transporter) at transcriptional level were performed in the juvenile root tissues of Rice using qRT-PCR technique at 15th, 30th and 45th days after transplanting with Actin as a internal control. Rice transplants were inoculated with different NPK fixing Bacillus consortia, which significantly improved growth parameters as compared to control in field study. Various growth and yield attributing characters of rice viz., plant height (at 60 DAT, 90 DAT, and at harvesting), number of productive tillers, days to 50 % flowering, 100 seed weight, grain yield, straw yield, as well as dry biomass per hectare were significantly influenced by different treatments. An application of 100 % RDF with NPK consortia (T7) treatment of rice crop resulted in significantly higher plant height (49.25 cm at 60 DAT, 107.00 cm at 90 DAT, 129.50 cm at harvesting), days to 50% flowering (75.63), 100 seed weight (3.53 g), number of productive tillers per plant (12.13), grain yield (6889.00 kg ha-1), straw yield (8754.25 kg ha-1) and dry biomass (7889.25 kg ha-1) followed by the treatments T4 (T2 + N consortia), T5 (T2 + P consortia), T6 (T2 + K consortia), and T2 (RDF (NP) 100%). Nutrient content in grain and straw was differing significantly due to different treatments. However, significantly higher N, P, K, content in straw (1.28 %, 0.15 %, and 1.52 %) and grain (2.06 %, 0.23 %, and 0.25 %) respectively, were recorded under the treatment T7. The spore forming Bacillus consortia was able to survive at a wide range of temperature and pH fluctuations and found to be effective as N-fixers, P-solubilizers, K-mobilizers, siderophore producers, IAA producers with having antagonistic activity against rice pathogen Magnaportha oryzae.

Abstract:Being sessile in nature, plants have to withstand various adverse environmental stress conditions including both biotic and abiotic stresses. Comparatively, abiotic stresses such as drought, salinity, high temperature, and cold pose major threat to agriculture by negatively impacting plant growth and yield worldwide. Rice is one of the most widely consumed staple cereals across the globe, the production and productivity of which is also severely affected by different abiotic stresses. Therefore, several crop improvement programs are directed toward developing stress tolerant rice cultivars either through marker assisted breeding or transgenic technology. Alternatively, some known rhizospheric competent bacteria are also known to improve plant growth during abiotic stresses. A plant growth promoting rhizobacteria (PGPR), Bacillus amyloliquefaciens NBRI-SN13 (SN13) was previously reported by our lab to confer salt stress tolerance to rice seedlings. However, the present study investigates the role of SN13 in ameliorating various abiotic stresses such as salt, drought, desiccation, heat, cold, and freezing on a popular rice cv. Saryu-52 under hydroponic growth conditions. Apart from this, seedlings were also exogenously supplied with abscisic acid (ABA), salicylic acid (SA), jasmonic acid (JA) and ethephon (ET) to study the role of SN13 in phytohormone-induced stress tolerance as well as its role in abiotic and biotic stress cross-talk. All abiotic stresses and phytohormone treatments significantly affected various physiological and biochemical parameters like membrane integrity and osmolyte accumulation. SN13 also positively modulated stress-responsive gene expressions under various abiotic stresses and phytohormone treatments suggesting its multifaceted role in cross-talk among stresses and phytohormones in response to PGPR. To the best of our knowledge, this is the first report on detailed analysis of plant growth promotion and stress alleviation by a PGPR in rice seedlings subjected to various abiotic stresses and phytohormone treatments for 0, 1, 3, 10, and 24 h.

Abstract:Bacillus methylotrophicus has been demonstrated to promote growth of various plants, whereas the promoting effect of B. methylotrophicus on rice growth has been rarely reported. In this study, B. methylotrophicus DD-1, capable of efficiently promoting the growth of rice, was isolated from the root soil of rice plants. The isolate exhibited potassium-solubilizing (1.18 mg/L), Indole-3-acetic acid (IAA) (87.26 mg/L), Gibberellic acid (GA) (25.91 mg/L) and Siderophore production activity (52.32%). As indicated from the result, plant growth parameters (e.g., dry weight, tiller number, root and shoot length) of rice seedlings treated with the isolate DD-1 were more effective than those of the control group in pot and soilless culture experiments. Moreover, the adsorption capacity of rice roots which were soaked in the bacterial suspension of isolate increased with the increase in concentration and absorption time. In sterilized and unsterilized soil, conformation of root colonization activity by bacterial isolate established by its nearer existence to the rice root. Thus, the B. methylotrophicus DD-1 enhances plant growth promotion by multifarious growth promoting and root colonization traits, thereby augmenting potassium level in soil. Henceforth, the potential bacterium could be exploited for the development of biological fertilizer, leading towards sustainable agronomy.

Abstract:Eighteen pesticide-degrading endophytic bacteria were isolated from the roots, stems, and leaves of healthy rice plants and identified through 16S rRNA gene sequencing. Furthermore, biochemical properties, including enzyme production, dye degradation, anti-bacterial activities, plant-growth-promoting traits, including N-fixation, P-solubilization, auxin production, and ACC-deaminase activities of these naturally occurring endophytic bacteria along with their four consortia, were characterized. Enterobacter cloacae HSTU-ABk39 and Enterobacter sp. HSTU-ABk36 displayed inhibition zones of 41.5 ± 1.5 mm, and 29 ± 09 mm against multidrug-resistant human pathogenic bacteria Staphylococcus aureus and Staphylococcus epidermidis, respectively. FT-IR analysis revealed that all eighteen isolates were able to degrade chlorpyrifos pesticide. Our study confirms that pesticide-degrading endophytic bacteria from rice plants play a key role in enhancing plant growth. Notably, rice plants grown in pots containing reduced urea (30%) mixed with either endophytic bacterial consortium-1, consortium-2, consortium-3, or consortia-4 demonstrated an increase of 17.3%, 38.6%, 18.2%, and 39.1% yields, respectively, compared to the control plants grown in pots containing 100% fertilizer. GC–MS/MS analysis confirmed that consortia treatment caused the degradation of chlorpyrifos into different non-toxic metabolites, including 2-Hydroxy-3,5,6 trichloropyridine, Diethyl methane phosphonate, Phorate sulfoxide, and Carbonochloridic. Thus, these isolates could be deployed as bio-stimulants to improve crop production by creating a sustainable biological system.

Abstract:Rice is one of the world's most important food crop that is severely affected by abiotic stresses. In our earlier study, inoculation with Bacillus amyloliquefaciens SN13 (SN13) improved the growth of rice seedlings under salt and drought stress as compared to other abiotic stresses was demonstrated. Therefore, the present study was carried out to understand the possible mechanism induced by B. amyloliquefaciens for salt and drought tolerance. Further, the role of SN13 in phytohormone-induced stress tolerance by exogenous application of abscisic acid and ethylene was investigated. The SN13 inoculated rice seedlings showed significantly better performance as demonstrated by physio-biochemical, antioxidant enzyme activities, and nutrient analysis. The expression of positively regulated stress-responsive genes was found under abiotic stresses and phytohormone treatments in inoculated seedlings indicating its multifaceted role in abiotic stresses and phytohormone cross-talk in response to PGPR. Inoculated seedlings also showed altered metabolites primarily related to carbohydrates (glucose, galactose, fructose, ribose, trehalose, turanose, hexapyranose, and xylose) and fatty acid (dodecanoic acid, eicosenoic acid, hexadecanoic acid, linolenic acid, and octadecadienoic acid) metabolism. These findings affirm that B. amyloliquefaciens SN13 positively modulates plant nutrient status, stress-responsive genes, and metabolic pathways related to carbohydrate and fatty acid metabolism supporting its involvement in cross-talk between imposed stresses and phytohormones.
