Abstract:Recently, microbial-based iron reduction has been considered as a viable alternative to typical chemical-based treatments. The iron reduction is an important process in kaolin refining, where iron-bearing impurities in kaolin clay affects the whiteness, refractory properties, and its commercial value. In recent years, Gram-negative bacteria has been in the center stage of iron reduction research, whereas little is known about the potential use of Gram-positive bacteria to refine kaolin clay. In this study, we investigated the ferric reducing capabilities of five microbes by manipulating the microbial growth conditions. Out of the five, we discovered that Bacillus cereus and Staphylococcus aureus outperformed the other microbes under nitrogen-rich media. Through the biochemical changes and the microbial behavior, we mapped the hypothetical pathway leading to the iron reduction cellular properties, and found that the iron reduction properties of these Gram-positive bacteria rely heavily on the media composition. The media composition results in increased basification of the media that is a prerequisite for the cellular reduction of ferric ions. Further, these changes impact the formation of biofilm, suggesting that the cellular interaction for the iron(III)oxide reduction is not solely reliant on the formation of biofilms. This article reveals the potential development of Gram-positive microbes in facilitating the microbial-based removal of metal contaminants from clays or ores. Further studies to elucidate the corresponding pathways would be crucial for the further development of the field.

Abstract:This study reports the bacterial properties of mixed Acidophilic microbiota with sulfur as electron donor and Acidithiobacillus ferrooxidans (A. ferrooxidans) with FeS2 as electron donor and conducted microbial desulfurization experiments. The results showed that mixed Acidophilic microbiota and A.ferrooxidans could effectively reduce the sulfur content in iron concentrate, with the desulfurization efficiency of 79.8 % and 45.7 % in 30 days. And the recovery rates were above 90 % and 88 % respectively. The results of the Scanning Electron Microscope (SEM) and Energy Dispersive Spectrometer (EDS) showed that both bacteria oxidized the iron concentrate and removed the sulfur-containing impurity from the surface of the ore sample. According to the enrichment of elemental phosphorus on the sulfur of the ore sample, it can be deduced that the bacteria were oxidized. After the leaching of A.ferrooxidans, it was found that jarosite was generated, which was consistent with the conclusion of Stereomicroscope morphological observation. Based on X-ray photoelectron spectroscopy analysis, it was concluded that mixed Acidophilic microbiota could reduce the deposition on the surface of iron concentrate after the leaching of sulfate and improve the bioleaching efficiency. The conversion route of sulfur was identified to be from S2-/S22- to Sn2-/S0, and finally SO42-. In conclusion, a model for the oxidation of iron concentrate by A.ferrooxidans and Mixed Acidophilic Microbiota during bioleaching has been proposed.

Abstract:Biotechnology has increasing relevance worldwide in the mining sector, either as a response to the recovery of metals (gold, silver, copper, zinc, nickel, among others) as well as an alternative in the bioremediation of contaminated soil and water, frequent problems directly linked to mining activities. Hence, acidophilic microorganisms are of special scientific and industrial interest for the sustainable use of mineral resources. Nowadays, a wide variety of acidophilic chemolithotrophic microorganisms (MOs) are recognized, Acidithiobacillus ferrooxidans , Acidithiobacillus thiooxidans , Acidithiobacillus caldus , and Leptospirillum ferrooxidans, among others; those MOs grow in culture medium at pH ≤ 3 and obtain cellular energy from the oxidation of inorganic compounds, such as sulfur and iron. These microorganisms have different abilities to act on the mineral, converting insoluble metal sulfides into soluble metal sulfates of those species that are of interest, or that prevent optimal recovery of a specific mineral. Such microorganisms have been applied in biomining operations and are internationally known for the recovery of valuable metals from low-grade ores and refractory ores. Likewise, these acidophilic MOs can bioremediate soils contaminated with metals, extract metals from sludge generated as a byproduct in wastewater treatment, detoxify hazardous waste and recover metals from electronic waste; so the main interest of biomining processes lies in the economic impact that has benefited the world, since it is known that 5% of the gold and 20% of the copper that has been extracted worldwide are using this type of bacteria in bioleaching processes. The objective of this review is to expand the knowledge of the characteristics and applications of the main acidophilic microorganisms used in the solubilization/extraction of minerals, whether for the recovery of metals, bioremediation, or reduction of metals in different systems.

Reduction of soluble iron and reductive dissolution of ferric iron-containing minerals by moderately thermophilic iron-oxidizing bacteria

Abstract:Five moderately thermophilic iron-oxidizing bacteria, including representative strains of the three classified species (Sulfobacillus thermosulfidooxidans, Sulfobacillus acidophilus, and Acidimicrobium ferrooxidans), were shown to be capable of reducing ferric iron to ferrous iron when they were grown under oxygen limitation conditions. Iron reduction was most readily observed when the isolates were grown as mixotrophs or heterotrophs with glycerol as an electron donor; in addition, some strains were able to couple the oxidation of tetrathionate to the reduction of ferric iron. Cycling of iron between the ferrous and ferric states was observed during batch culture growth in unshaken flasks incubated under aerobic conditions, although the patterns of oxidoreduction of iron varied in different species of iron-oxidizing moderate thermophiles and in strains of a single species (S. acidophilus). All three bacterial species were able to grow anaerobically with ferric iron as a sole electron acceptor; the growth yields correlated with the amount of ferric iron reduced when the isolates were grown in the absence of oxygen. One of the moderate thermophiles (identified as a strain of S. acidophilus) was able to bring about the reductive dissolution of three ferric iron-containing minerals (ferric hydroxide, jarosite, and goethite) when it was grown under restricted aeration conditions with glycerol as a carbon and energy source. The significance of iron reduction by moderately thermophilic iron oxidizers in both environmental and applied contexts is discussed.
Unveiling the potential of Glutamicbacter nicotiana for enhanced bioleaching of nickel and valuable metals from low- and high-grade nickeline ores

Abstract:This study explores the utilization of three heterotrophic bacteria, isolated from iron mine wastewater and tailings, for the bioleaching of low- and high-grade nickel-bearing arsenic ores. The objective was to assess the leaching potential of these bacteria strains in extracting nickel, cobalt, arsenic, and copper ions from nickeline samples at varying pulp densities (0.5%, 0.8%, 1.1%, and 3%). A comparative analysis was conducted to evaluate the efficiencies of these isolates, including their performance relative to each other, the supernatant (spent medium), culture, and mixed strains. Under carefully controlled experimental conditions, all the bacterial strains exhibited remarkable leaching capabilities. Notably, Glutamicbacter nicotiana emerged as the most effective strain among them. Glutamicbacter nicotiana demonstrated impressive extraction rates, reaching up to 100% for nickel, 16% for cobalt, and 73% for arsenic in the low-grade sample. Similarly, in the high-grade sample, it achieved extraction rates of up to 70% for nickel, 62% for cobalt, and 16% for arsenic. In contrast, the control leaching experiment yielded minimal dissolution of the target elements, with only 0.33% for nickel, 1.2% for cobalt, and 0.4% for arsenic. To comprehensively characterize the untreated and bioleaching residue samples, various analytical techniques were employed, including SEM-EDS, FE-SEM, and XRD. These methods enabled a thorough investigation of the samples' structural and chemical composition. Additionally, HPLC analysis detected the presence of amino acids, such as glutamic acid, which were produced by the microorganisms involved in the bioleaching process. Furthermore, the study incorporated kinetic modeling to understand the factors influencing the bioleaching of both low- and high-grade samples. The results indicated that chemical control constituted the rate-limiting factor in the bioleaching process.

Abstract:The extraction of phosphorus (P) from iron-rich sludge is a critical step for P recovery process. The acidogenic phosphorus release (APR) process was proved as a cost-effective and environmental-friendly method to separate and release Fe–P from iron-rich sludge, including primary sludge and activated sludge. However, the role of bacteria in this microbial-driven metal reductive dissolution approach was not clear. This study aimed to reveal the mechanism of microbial cooperation during APR process. The P release fraction could achieve as high as 63.2% from iron-rich primary sludge. High-throughput sequencing of 16S rRNA gene amplicon was conducted to characterize the functional microorganisms participating in APR process, including hydrolytic bacteria Prevotella, acidogens Selenomonas, Bacteroides, Lactococcus, Tolumonas, and iron-reducing bacteria Aeromonas and Sulfurospirillum. Microbial iron reduction and acidogenic fermentation were identified as two critical biological reactions in APR process. A two-phase temporal succession of bacterial community was characterized in batch APR process. The strong positive relationship between iron-reducing bacteria and acidogens was revealed by network analysis. This study successfully proved that bacteria-mediating metal reductive dissolution approach could efficiently release the reducible metal compounds (such as Fe–P) from complex mixture, which can be extended to many other scenarios regarding resource extraction and recovery.
Enriched Iron(III)-Reducing Bacterial Communities are Shaped by Carbon Substrate and Iron Oxide Mineralogy

Abstract:Iron (Fe) oxides exist in a spectrum of structures in the environment, with ferrihydrite widely considered the most bioavailable phase. Yet, ferrihydrite is unstable and rapidly transforms to more crystalline Fe(III) oxides (e.g., goethite, hematite), which are poorly reduced by model dissimilatory Fe(III)-reducing microorganisms. This begs the question, what processes and microbial groups are responsible for reduction of crystalline Fe(III) oxides within sedimentary environments? Further, how do changes in Fe mineralogy shape oxide-hosted microbial populations? To address these questions, we conducted a large-scale cultivation effort using various Fe(III) oxides (ferrihydrite, goethite, hematite) and carbon substrates (glucose, lactate, acetate) along a dilution gradient to enrich for microbial populations capable of reducing Fe oxides spanning a wide range of crystallinities and reduction potentials. While carbon source was the most important variable shaping community composition within Fe(III)-reducing enrichments, both Fe oxide type and sediment dilution also had a substantial influence. For instance, with acetate as the carbon source, only ferrihydrite enrichments displayed a significant amount of Fe(III) reduction and the well-known dissimilatory metal reducer Geobacter sp. was the dominant organism enriched. In contrast, when glucose and lactate were provided, all three Fe oxides were reduced and reduction coincided with the presence of fermentative (e.g., Enterobacter spp.) and sulfate-reducing bacteria (e.g., Desulfovibrio spp.). Thus, changes in Fe oxide structure and resource availability may shift Fe(III)-reducing communities between dominantly metal-respiring to fermenting and/or sulfate-reducing organisms which are capable of reducing more recalcitrant Fe phases. These findings highlight the need for further targeted investigations into the composition and activity of speciation-directed metal-reducing populations within natural environments.

Abstract:Bioleaching of metal sulfides is performed by a diverse group of microorganisms. The dissolution chemistry of metal sulfides follows two pathways, which are determined by the mineralogy and the acid solubility of the metal sulfides: the thiosulfate and the polysulfide pathways. Bacterial cells can effect this metal sulfide dissolution via iron(II) ion and sulfur compound oxidation. Thereby, iron(III) ions and protons, the metal sulfide-attacking agents, are available. Cells can be active either in planktonic state or in forming biofilms on the mineral surface; however, the latter is much more efficient in terms of bioleaching kinetics. In the case of Acidithiobacillus ferrooxidans, bacterial exopolymers contain iron(III) ions, each complexed by two uronic acid residues. The resulting positive charge allows an electrostatic attachment to the negatively charged pyrite. Thus, the first function of complexed iron(III) ions is the mediation of cell attachment, while their second function is oxidative dissolution of the metal sulfide, similar to the role of free iron(III) ions in non-contact leaching. In both cases, the electrons extracted from the metal sulfide reduce molecular oxygen via a redox chain forming a supercomplex spanning the periplasmic space and connecting both outer and inner membranes. In this review, we summarize some recent discoveries relevant to leaching bacteria which contribute to a better understanding of these fascinating microorganisms. These include surface science, biochemistry of iron and sulfur metabolism, anaerobic metabolism, and biofilm formation. The study of microbial interactions among multispecies leaching consortia, including cell-to-cell communication mechanisms, must be considered in order to reveal more insights into the biology of bioleaching microorganisms and their potential biotechnological use.

Abstract:Magnetic iron minerals, such as magnetite (Fe 3 O 4 ) and maghemite (γ-Fe 2 O 3 ), are widely distributed in sediments, holding significant potential for reconstructing paleoenvironments through their magnetic properties. However, the impact of microbial iron reduction on the physicochemical and magnetic characteristics of Fe(III)-containing magnetic minerals has recently been recognized as a crucial but often overlooked factor in paleoenvironmental reconstructions. Although magnetic iron minerals are frequently associated with clay minerals in sedimentary settings, our understanding of the influence of clay minerals on the microbial reduction of Fe(III) in magnetic minerals remains limited. To address this knowledge gap, a series of bioreduction experiments were conducted in a non-growth medium, in which lactate acted as the electron donor, magnetite or maghemite nanoparticles (NPs) served as the electron acceptor, and a typical iron-reducing bacterium, Shewanella oneidensis strain MR-1, acted as the reaction mediator, in the presence or absence of montmorillonite (STx-1b). Our results demonstrated that the addition of montmorillonite STx-1b significantly enhanced the dispersion of magnetite and maghemite NPs, thereby increasing the final extent of microbial iron reduction. Moreover, the degree of microbial iron reduction impacted the crystallography and magnetic properties of magnetite and maghemite. Upon bioreduction, the lattice constant, magnetic susceptibility, and the 10-K magnetic coercivity of these magnetic iron minerals exhibited an increase, accompanied by the presence of more structural defects. Specifically, maghemite-magnetite solid solutions were formed in the maghemite-amended biosystems. These findings highlight the influence of clay minerals in natural settings during microbial iron reduction on the magnetic properties of magnetic iron minerals used for paleoenvironmental reconstructions.

Abstract:Acidithiobacillus ferrooxidans is an acidophilic and chemolithotrophic sulfur- and iron-oxidizing bacterium that has been widely used in the bioleaching process for extracting metals. Extracellular polymeric substances (EPS) are essential for bacteria-ore interactions, and the regulation of EPS synthesis could be an important way of influencing the efficiency of the bioleaching process. Therefore, exploring and utilizing the regulatory pathways of EPS synthesis to improve the bacterial bioleaching capability have posed a challenge in the study and application of bioleaching bacteria. Here, several engineering strains were constructed using genetic manipulation methods. And we revealed the regulatory function of the AfeI/R quorum sensing (QS) system in EPS synthesis and biofilm formation of A. ferrooxidans, and the AfeI/R-mediated EPS synthesis could influence bacteria-substrate interactions and the efficiency of bioleaching. Finally, an AfeI/R-mediated bioleaching model was proposed to illustrate the role of QS system in this process. This study provided new insights into and clues for developing highly efficient bioleaching bacteria and modulating the bioleaching process.

Culture-dependent and culture-independent methods reveal microbe-clay mineral interactions by dissimilatory iron-reducing bacteria in an integral oilfield

Abstract:Fe(III) may be reasonably considered as one of the most important electron acceptors in petroleum reservoir ecosystems. The microbial mineralization of clay minerals, especially montmorillonite, is also of great significance to the exploration of petroleum and gas reservoirs. The bioreduction mechanisms of iron-poor minerals in petroleum reservoirs have been poorly investigated. This study investigated the bioreduction of montmorillonite by dissimilatory iron-reducing bacteria (DIRB) in petroleum reservoirs based on culture-independent and culture-dependent methods. Microbial diversity analysis revealed that Halolactibacillus, Bacillus, Alkaliphilus, Shewanella, Clostridium, and Pseudomonas were the key genera involved in the bioreduction of Fe(III). Through the traditional culture-dependent method, most of the key genera were isolated from the samples collected from petroleum reservoirs. Traditional culture-dependent methods can be used to reveal the metabolic characteristics of microorganisms (such as iron-reduction efficiency) to further elucidate the roles of different species (B. subtilis and B. alkalitelluris) in the environment. Moreover, many species with high iron-reduction efficiencies and relatively low abundances in the samples, such as Tessaracoccus and Flaviflexus, were isolated from petroleum reservoirs for the first time. The combination of culture-dependent and culture-independent methods can be used to further the understanding of the microbial communities and the metabolic characteristics of DIRB in petroleum reservoirs. Structural alterations that occurred during the interactions of microorganisms and montmorillonite were revealed through scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), and X-ray powder diffraction (XRD). The physical and chemical analysis results demonstrated that microorganisms from petroleum reservoirs can dissolve iron-poor montmorillonite and promote the release of interlayer water. The secondary minerals illite and clinoptilolite were observed in bioreduced smectite. The formation of secondary minerals was closely related to the dissolution degrees of minerals based on iron reduction.

Abstract:In many European regions, both local metallic and non-metallic raw materials are poorly exploited due to their low quality and the lack of technologies to increase their economic value. In this context, the development of low cost and eco-friendly approaches, such as bioleaching of metal impurities, is crucial. The acidophilic strain Acidiphilium sp. SJH reduces Fe(III) to Fe(II) by coupling the oxidation of an organic substrate to the reduction of Fe(III) and can therefore be applied in the bioleaching of iron impurities from non-metallic raw materials. In this work, the physiology of Acidiphilium sp. SJH and the reduction of iron impurities from quartz sand and its derivatives have been studied during growth on media supplemented with various carbon sources and under different oxygenation conditions, highlighting that cell physiology and iron reduction are tightly coupled. Although the organism is known to be aerobic, maximum bioleaching performance was obtained by cultures cultivated until the exponential phase of growth under oxygen limitation. Among carbon sources, glucose has been shown to support faster biomass growth, while galactose allowed highest bioleaching. Moreover, Acidiphilium sp. SJH cells can synthesise and accumulate Poly-β-hydroxybutyrate (PHB) during the process, a polymer with relevant application in biotechnology. In summary, this work gives an insight into the physiology of Acidiphilium sp. SJH, able to use different carbon sources and to synthesise a technologically relevant polymer (PHB), while removing metals from sand without the need to introduce modifications in the process set up.

Abstract:The evolution of iron minerals under buried conditions is one of the most important processes controlling the mineral composition and heavy metal transportation in sediments. Microbial-mediated reduction plays a critical role in iron mineral transformation in natural environment. This study examined the transformation pathways of iron minerals mediated by bacteria and the changes of associated arsenic species in iron-rich mine tailings. Static and column reactions were designed to monitor variations of minerals and released iron and arsenic, a reactive transport model was simulated to support laboratory results. Laboratory experiments showed that major ferric minerals were preferentially dissolved and reduced by dissimilatory iron-reducing bacteria. The released Fe3+ in fluid promoted oxidative dissolution of pyrite and arsenopyrite, and precipitation of oxides and carbonates. The arsenic released to fluid was inferred to be immobilized by both pristine ferrihydrite and newly formed hydrous ferric oxides via surface complexation. The reaction system maintained a steady-state of iron mineral transformation and arsenic (im)mobilization. In the latter stage of column reactor experiments, continuous reaction and removal of dissolved Fe3+ and Fe2+ destabilized the state, leading to arsenic re-location and eventually rising concentration in fluid. The findings implicate that microbial-mediated iron mineral evolution remarkably influence the natural mineral assemblages and the fate of contaminant transport in the environment, and that deposition of iron oxides is essential in environmental protection and pollution recovery.
Reductive dissolution of jarosite by inorganic sulfur compounds catalyzed by Acidithiobacillus thiooxidans

Abstract:The adsorption of jarosite and the resulting passivation of mineral surfaces can negatively influence metal extraction from sulfidic ores as well as the fate of other elements in biohydrometallurgical processes. Some bioleaching microorganisms mediate dissimilatory iron reduction coupled to sulfur oxidation (DIRSO), a process utilized predominantly in continuously enhanced leaching of metals from sulfide and/or oxidized ores. In this study, the reductive dissolution of jarosite (biosynthesized by the iron-oxidizing archaeon Acidianus manzaensis ) catalyzed by the mesophilic acidophilic bacterium Acidithiobacillus ( At .) thiooxidans oxidizing different inorganic sulfur compounds was investigated. Kinetic measurements of pH, ORP, iron concentrations, and planktonic cell counts were performed to describe the reductive dissolution of jarosite. Moreover, the solid leaching residues were analyzed using X-ray absorption near edge structure (XANES), Inductively coupled plasma - optical emission spectrometry (ICP-OES), scanning electron microscopy (SEM), X-ray diffraction (XRD), and Raman spectroscopy. The dissolution rates of jarosite after 34 days of bioleaching by At. thiooxidans with S 0 , Na 2 S 2 O 3 , and Na 2 SO 3 were 41.7, 76.3, and 98.4%, respectively, while negligible jarosite dissolution was detected in abiotic controls. The presence of Na 2 S 2 O 3 and Na 2 SO 3 resulted in structural modifications on the jarosite surfaces, but the dissolution of jarosite was not promoted in the absence of At. thiooxidans . In the biotic assays with Na 2 SO 3 , jarosite was completely dissolved, indicating that Na 2 SO 3 was the most suitable electron donor (out of those tested) for DIRSO by At. thiooxidans . The findings obtained in this study can contribute to designing suitable bioleaching strategies for oxidized ores. They also highlight the potential of microbially catalyzed DIRSO to mitigate jarosite formation that often hinders bioleaching of sulfidic ores.

The Interplay of Iron Minerals and Microflora to Accelerate Cr (VI) Reduction

Abstract:Widespread hexavalent chromium (Cr (VI)) in the environment has become a worldwide problem, and economical and efficient treatment is urgently needed. In this paper, the treatment method of Cr (VI) by microorganisms and iron minerals (pyrite and magnetite) under anaerobic conditions was investigated. Furthermore, the influence of Cr (VI) on the microbial community structure was explored. The reduction test demonstrated that the removal rate of Cr (VI) in a single biological group was 54.96%; however, in the pyrite and biological groups and magnetite and biological groups, the removal rates of Cr (VI) increased to 83.06% and 78.23%, respectively. Microorganisms and iron minerals work together to produce a better removal effect on the removal rate of Cr (VI). Mechanistic studies have found that in the process of Cr (VI) reduction, a passivation layer is formed on the surface of the mineral that hinders the progress of the reaction. The addition of bacteria can reduce the negative impact of the passivation layer. At the same time, iron minerals have better electron-receiving and -conducting ability and can be used as electron carriers for bacteria to reduce Cr (VI). In addition, iron minerals and the disappearance of Cr (VI) will change the structure of the community and affect the expression of its functions, which is more conducive to reducing Cr (VI). This work sheds new light on the treatment of heavy metal pollution and the understanding of the synergistic reduction mechanism of Cr (VI).

Abstract:Freshwater lakes are often polluted with various heavy metals in the Anthropocene. The iron-oxidizing microorganisms and their mineralized products can coprecipitate with many heavy metals, including Al, Zn, Cu, Cd, and Cr. As such, microbial iron oxidation can exert a profound impact on environmental remediation. The environmental pH is a key determinant regulating microbial growth and mineralization and then influences the structure of the final mineralized products of anaerobic iron-oxidizing bacteria. Freshwater lakes, in general, are neutral-pH environments. Understanding the effects of varying pH on the mineralization of iron-oxidizing bacteria under neutrophilic conditions could aid in finding out the optimal pH values that promote the coprecipitation of heavy metals. Here, two typical neutrophilic Fe(II)-oxidizing bacteria, the nitrate-reducing Acidovorax sp. strain BoFeN1 and the anoxygenic phototrophic Rhodobacter ferrooxidans strain SW2, were selected for studying how their growth and mineralization response to slight changes in circumneutral pH. By employing focused ion beam/scanning electron microscopy (FIB–SEM) and transmission electron microscopy (TEM), we examined the interplay between pH changes and anaerobic iron-oxidizing bacteria and observed that pH can significantly impact the microbial mineralization process and vice versa. Further, pH-dependent changes in the structure of mineralized products of bacterial iron oxidation were observed. Our study could provide mechanical insights into how to manipulate microbial iron oxidation for facilitating remediation of heavy metals in the environment.

Abstract:Iron-reducing enrichments were obtained from leachate ponds at the U.S. Borax Company in Boron, Calif. Based on partial small-subunit (SSU) rRNA gene sequences (approximately 500 nucleotides), six isolates shared 98.9% nucleotide identity. As a representative, the isolate QYMF was selected for further analysis. QYMF could be grown with Fe(III)-citrate, Fe(III)-EDTA, Co(III)-EDTA, or Cr(VI) as electron acceptors, and yeast extract and lactate could serve as electron donors. Growth during iron reduction occurred over the pH range of 7.5 to 11.0 (optimum, pH 9.5), a sodium chloride range of 0 to 80 g/liter (optimum, 20 g/liter), and a temperature range of 4 to 45degreesC (optimum, approximately 35degreesC), and iron precipitates were formed. QYMF was a strict anaerobe that could be grown in the presence of borax, and the cells were straight rods that produced endospores. Sodium chloride and yeast extract stimulated growth. Phylogenetic analysis of the SSU rRNA gene indicated that the bacterium was a low-G+C gram-positive microorganism and had 96 and 92% nucleotide identity with Alkaliphilus transvaalensis and Alkaliphilus crotonatoxidans, respectively. The major phospholipid fatty acids were 14:1, 16:1w7c, and 16:0, which were different from those of other alkaliphiles but similar to those of reported iron-reducing bacteria. The results demonstrated that the isolate might represent a novel metal-reducing alkaliphilic species. The name Alkaliphilus metalliredigens sp. nov. is proposed. The isolation and activity of metal-reducing bacteria from borax-contaminated leachate ponds suggest that bioremediation of metal-contaminated alkaline environments may be feasible and have implications for alkaline anaerobic respiration.
Arsenic Biomineralization by Iron Oxidizing Strain (ochrobactrum Sp.) Isolated from a Paddy Soil in Hunan, China

Abstract:The biogeochemistry of iron (Fe) could affect arsenic (As) fate and change its availability. Fe(II) oxidation mediated by Fe oxidizing bacteria (FeOB) has the potential for As contamination remediation due to the formation of biogenic minerals. However, microbial Fe(II) oxidation coupled with the immobilization of As and the effect of FeOB on As remediation in soils has not been thoroughly investigated. In this study, a FeOB was isolated from a paddy soil and identified as Ochrobactrum sp. EEELCW01. The kinetics reactions were used to investigate how biological and chemical reactions were involved in the Fe(II)-NO3--FeOB system. Microbial Fe(II) oxidation coupled As mineralization, as well as its application in As contaminated paddy soils was studied. The results suggested that biological pathway contributed to the Fe(II) oxidation and NO3- reduction in the whole process, in which the chemical Fe(II) oxidation by biogenic NO2- was involved. Meanwhile, Fe(II) facilitated the reduction of NO2- while inhibited the reduction of NO3-. Adding 2% or more bacteria led to a significant removal effect for aqueous As, resulting in the formation of lepidocrocite and two As-containing minerals (angelellite, loellingite). With the simultaneous application of FeOB and NO3-, the available As content in soils decreased by 37.6% and stabilized at 0.80 mg kg(-1) finally. In addition, the immobilization showed long-term effect compared to the treatment with Fe(II) or NO3- alone. The study proved that the isolated FeOB had a significant ability for As mineralization and immobilization, which provided theoretical and application basis for the remediation of As contaminated soils.
Fe(III) bioreduction kinetics in anaerobic batch and continuous stirred tank reactors with acidophilic bacteria relevant for bioleaching of limonitic laterites

Abstract:In the framework of the H2020 project CROCODILE, the recovery of Co from oxidized ores by reductive bioleaching has been studied. The objective was to reduce Fe(III) to Fe(II) to enhance the dissolution of Co from New-Caledonian limonitic laterites, mainly composed of goethite and Mn oxides. This study focused on the Fe(III) bioreduction which is a relevant reaction of this process. In the first step, biomass growth was sustained by aerobic bio-oxidation of elemental sulfur. In the second step, the biomass anaerobically reduced Fe(III) to Fe(II). The last step, which is not in the scope of this study, was the reduction of limonites and the dissolution of metals. This study aimed at assessing the Fe(III) bioreduction rate at 35°C with a microbial consortium composed predominantly of Sulfobacillus ( Sb. ) species as the iron reducers and Acidithiobacillus ( At. ) caldus . It evaluated the influence of the biomass concentration on the Fe(III) bioreduction rate and yield, both in batch and continuous mode. The influence of the composition of the growth medium on the bioreduction rate was assessed in continuous mode. A mean Fe(III) bioreduction rate of 1.7 mg·L −1 ·h −1 was measured in batch mode, i.e., 13 times faster than the abiotic control (0.13 mg·L −1 ·h −1 ). An increase in biomass concentrations in the liquid phase from 4 × 10 8 cells·mL −1 to 3 × 10 9 cells·mL −1 resulted in an increase of the mean Fe(III) bioreduction rate from 1.7 to 10 mg·L −1 ·h −1 . A test in continuous stirred tank reactors at 35°C resulted in further optimization of the Fe(III) bioreduction rate which reached 20 mg·L −1 ·h −1 . A large excess of nutrients enables to obtain higher kinetics. The determination of this kinetics is essential for the design of a reductive bioleaching process.
