Abstract:A novel electrochemical microbial biosensor for the rapid monitoring of p-nitrophenyl-substituted organophosphates (OPs) compounds based on glass carbon electrode (GCE) modified with both ordered mesopore carbons (OMCs) and cell surface-expressed organophosphorus hydrolase (OPH) (OPH-bacteria/OMCs/GCE) was described in this paper. The genetically engineered Escherichia coli strain surface displayed mutant OPH (S5) with improved enzyme activity and favorable stability was constructed using a newly identified N-terminal of ice nucleation protein as an anchoring motif, which can be used directly without further time-consuming enzyme-extraction and purification, thereafter greatly improved the stability of the enzyme. Compared to OPH-bacteria modified GCE (OPH-bacteria/GCE), the OPH-bacteria/OMCs/GCE not only significantly enhanced the current response but also reduced the oxidation overpotential towards oxidizable p-nitrophenol (p-NP), which was the hydrolysate of p-nitrophenyl-substituted OPs. Under the optimized experimental conditions, at +0.84V (vs. SCE), the current–time curve was performed with varying OPs concentration. The current response was linear with paraoxon concentration within 0.05–25μM. Similarly, linear range of 0.05–25μM was found for parathion, and 0.08–30μM for methyl parathion. The low limits of detection were evaluated to be 9.0nM for paraoxon, 10nM for parathion and 15nM for methyl parathion (S/N=3). Thus, a highly specific, sensitive and rapid microbial biosensor was established, which holds great promise for on-site detection of trace p-nitrophenyl-substituted OPs.

Abstract:The inhibition of acetylcholinesterase (AChE) activity based methods have been widely used for rapid detection of organophosphate (OP) pesticide residues. However, the sensitivity of the enzyme toward pesticides is not desirable and complex purification steps increase production cost. In this study, we optimized and synthesized the cDNA of carp AChE based on the preferred codon using Saccharomyces cerevisiae, which was ligated into expression vector pYD1. The AChE was successfully presented on yeast surface through a-agglutinin mediated display system. In shake-flask after induction with galactose in YPD medium for 24 h, this strain expressed AChE activity at a high level of 0.15 U/OD600/ml. The correct location of AChE was confirmed by immunofluorescence analysis. The whole cell catalyst was applied to visible spectrophotometry detection for OP pesticides based on inhibition rate. The inhibition rate was linear with paraoxon concentration in the range of 0.5 ng/ml-10 mu g/ml and parathion concentration in the range of 5 ng/ml-10 mu g/ml. The low detection limit was 0.136 ng/ml paraoxon and 3.72 ng/ml parathion (S/N = 3). The proposed method is applicable to analyzing OP pesticides in real samples. Therefore, the recombinant strain developed could be an efficient and affordable biomaterial for rapid and accurate detection of OP pesticide residues.

Abstract:A multicolor enzyme-linked immunosorbent sensor was constructed to detect organophosphates pesticides (OPs) on base of TMB2+-mediated AuNRs etching. As the etching progresses, a rainbow-like color responses was generated. The naked eye limit of detection (LOD) of the sensing system is 0.25 ng mL-1. In particular, the longitudinal localized surface plasmon resonances (LSPR) peak shift of AuNRs is linearly correlated with the logarithm of OPs concentration in range of 104 to 5×106 pg mL-1 for parathion-methyl and fenitrothion, 5×104 to 107 pg mL-1 for fenthion, respectively. Compared with reported methods, this sensor displayed satisfactory linear ranges and detection limits. Average recoveries of this method ranged from 73.28% to 98.08% and relative standard deviations (RSDs) were below 8.0% as for food samples analysis without complicated pretreatments. Remarkably, the developed multicolor enzyme-linked immunosorbent sensor is capable of providing accurate and reliable assay for OPs in complex food samples.

Abstract:Developing simple, efficient, and inexpensive method for trace amount organophosphorus pesticides' (OPs) detection with high sensitivity and specificity is of significant importance for guaranteeing food safety. Herein, an Ag/Au bimetallic nanoparticle-based acetylcholinesterase (AChE) surface-enhanced Raman scattering (SERS) biosensor was constructed for in situ simple and sensitive detection of pesticide residues in food. The principle of this biosensor exploited 4-mercaptophenylboronic acid (4-MPBA)-modified Ag/Au bimetallic nanoprobes as SERS signal probe to improve sensitivity and stability. The combination of AChE and choline oxidase (CHO) can hydrolyze acetylcholine (ATCh) to generate H 2 O 2 . The product of H 2 O 2 selectively oxidizes the boronate ester of 4-MPBA, decreasing the Raman intensity of the B-O symmetric stretching. In the presence of OPs, it could inhibit the production of H 2 O 2 by destroying the AChE activity, so the reduction of the SERS signal was also alleviated. Based on the principle, an Ag/Au bimetallic nanoparticle-based AChE SERS sensor was established without any complicated pretreatments. Benefiting from the synergistic effects of Ag/Au bimetallic hybrids, a linear detection range from 5×10 −9 to 5×10 −4 M was achieved with a limit of detection down to 1.7×10 −9 M using parathion-methyl (PM) as the representative model of OPs. Moreover, the SERS biosensor uses readily available reagents and is simple to implement. Importantly, the proposed SERS biosensor was used to quantitatively analyze OP residues in apple peels. The levels of OPs detected in real samples by this method were consistent with those obtained using gas chromatography–mass spectrometry (GC–MS), suggesting the proposed assay has great potential applications for OPs in situ detection in food safety fields.

Abstract:Rapid and sensitive detection methods are in urgent demand for the screening of extensively used organophosphorus pesticides and highly toxic nerve agents for their neurotoxicity. In this study, we developed a novel Fe(3)O(4) magnetic nanoparticle (MNP) peroxidase mimetic-based colorimetric method for the rapid detection of organophosphorus pesticides and nerve agents. The detection assay is composed of MNPs, acetylcholinesterase (AChE), and choline oxidase (CHO). The enzymes AChE and CHO catalyze the formation of H(2)O(2) in the presence of acetylcholine, which then activates MNPs to catalyze the oxidation of colorimetric substrates to produce a color reaction. After incubation with the organophosphorus neurotoxins, the enzymatic activity of AChE was inhibited and produced less H(2)O(2), resulting in a decreased catalytic oxidation of colorimetric substrates over MNP peroxidase mimetics, accompanied by a drop in color intensity. Three organophosphorus compounds were tested on the assay: acephate and methyl-paraoxon as representative organophosphorus pesticides and the nerve agent Sarin. The novel assay displayed substantial color change after incubation in organophosphorus neurotoxins in a concentration-dependent manner. As low as 1 nM Sarin, 10 nM methyl-paraoxon, and 5 μM acephate are easily detected by the novel assay. In conclusion, by employing the peroxidase-mimicking activity of MNPs, the developed colorimetric assay has the potential of becoming a screening tool for the rapid and sensitive assessment of the neurotoxicity of an overwhelming number of organophosphate compounds.

Abstract:An acetylcholinesterase (AChE) biosensor was constructed based on gold nanoparticles (AuNPs) using electroless plating on vertical nitrogen-doped single-walled carbon nanotubes (VNSWCNTs) for detecting organophosphorus pesticides (OPs). AChE was immobilised on AuNPs via Au–S bonding, and VNSWCNTs were produced by spontaneous chemical adsorption of NSWCNTs on gold electrode, also via Au–S bonding. This modified electrode exhibited excellent electron transfer capacity due to the synergy between AuNPs and VNSWCNTs. The developed biosensor showed good linear relations at concentrations of 10−5 – 1 ppb, and the detection limits were 3.04 × 10−6 ppb for methyl parathion, 1.96 × 10−6 ppb for malathion and 2.06 × 10−6 ppb for chlorpyrifos, respectively. The AChE biosensor revealed satisfactory stability, excellent sensitivity and good repeatability. These results suggest that this biosensor has good application prospects and can function as a sensitive device in OPs analysis.

Abstract:Organophosphate nerve agents (OPNAs) are lethal chemical weapons (CWs) which hydrolyze rapidly to alkyl phosphate compounds in the environment. The identification of alkyl phosphate compounds in the environment is critical for the verification of OPNAs. Currently, due to the complex sample preparation and long detection process for organophosphate nerve agent hydrolysis products (OPNAHPs), their high-throughput determination is difficult. To simplify sample preparation and detection of OPNAHPs from environmental samples, a quick, easy, cheap, effective, rugged and safe (QuEChERS) method for the simultaneous purification of the five OPNAHPs in environmental samples combined with detection through direct analysis in real-time ionization source coupled with tandem mass spectrometer (DART-MS/MS) was developed. An internal standard method was applied for the quantification. The method exhibited a good linearity (R2 > 0.991) in the range of 1.00 ng/mL-4.00 μg/mL. The limits of detection and limits of quantification were 1.00–7.00 ng/mL and 10.0–20.0 ng/mL, respectively. The recoveries of OPNAHPs in environmental samples were between 65.3% and 107%, with relative standard deviations ranged from 1.6% to 19%. In addition, the sample preparation and analysis process for OPNAHPs could be completed in less than 10 min. The results demonstrated that the modified QuEChERS method could quickly and efficiently remove the matrix of environmental samples. Combined with DART-MS/MS, an accurate and high-throughput detection method of OPNAHPs in environmental samples was provided. The developed method has great potential in the fields of early warning, clinical treatment and environmental decontamination.

Abstract:Given the importance of food safety, it is highly desirable to develop a convenient, low-cost, and practical sensor for organophosphorus pesticides (OPs) detection. Here, a fluorescent paper analytical device (FPAD) based on aggregation-induced emission (AIE) nanoparticles (PTDNPs-0.10) and two-dimension MnO2 nanoflakes (2D-MnNFs) was developed for instrument-free and naked-eye analysis of OPs. PTDNP-MnNFs composites were obtained through 2D-MnNFs and PTDNPs-0.10 by electrostatic interaction. and the fluorescence emission of PTDNPs-0.10 was quenched through fluorescence resonance energy transfer (FRET). When acetylcholinesterase (AChE) was present, acetylthiocholine (ATCh) was catalytically hydrolyzed into thiocholine, which reduced MnO2 of PTDNP-MnNFs into Mn2+, subsequently blocking the FRET and enhancing the fluorescence. Upon the addition of OP, AChE activity was depressed and thus the FRET between 2D-MnNFs and PTDNPs-0.10 was not affected, resulting in a slight change in fluorescence. On the basis of the variation in fluorescence intensity, highly sensitive detection of OP was readily achieved with a detection limit of 0.027 ng/mL; on the basis of the variation in brightness of FPAD, instrument-free and visual detection of OP was realized using a smartphone with a detection limit of 0.73 ng/mL. The application of FPAD has significantly simplified the detection procedure and decreased the test cost, supplying a new approach for on-site detection of OPs.

Abstract:A g-C3N4/AgNPs sensor could be used for the determination of organophosphorus pesticides (OPs).The method exhibits ultra-sensitivity and high-selectivity toward OPs.Double-signal OPs probe was developed with low cost, simple interpretation and easy handling.A highly sensitive and selective method was developed for both UV–vis spectrophotometric and fluorimetric determination of organophosphorus pesticides (OPs). This method uses silver nanoparticles (AgNPs) modified with graphitic carbon nitride (g-C3N4). The AgNPs reduce the fluorescence intensity of g-C3N4. Acetylthiocholine (ATCh) can be catalytically hydrolyzed by AChE to form thiocholine, which induces aggregation of the AgNPs. This aggregation leads to the recovery of the blue fluorescence of g-C3N4, with excitation/emission peaks at 310/460 nm. This fluorescence intensity can be reduced again in the presence of OPs because of the inhibitory effect of OPs on the activity of AChE. The degree of reduction was found to be proportional to the concentration of the OPs, and the limit of fluorometric detection was 0.0324 μg/L (S/N = 3). In addition, the absorption of the g-C3N4/AgNPs at 390 nm decreased because of the aggregation of the AgNPs, but was recovered in presence of OPs because of the inhibition of enzyme activity by OPs. Abstract:We have developed a novel optical biosensor device using recombinant methyl parathion hydrolase (MPH) enzyme immobilized on agarose by metal-chelate affinity to detect organophosphorus (OP) compounds with a nitrophenyl group. The biosensor principle is based on the optical measurement of the product of OP catalysis by MPH (p-nitrophenol). Briefly, MPH containing six sequential histidines (6 × His tag) at its N-terminal was bound to nitrilotriacetic acid (NTA) agarose with Ni ions, resulting in the flexible immobilization of the bio-reaction platform. The optical biosensing system consisted of two light-emitting diodes (LEDs) and one photodiode. The LED that emitted light at the wavelength of the maximum absorption for p-nitrophenol served as the signal light, while the other LED that showed no absorbance served as the reference light. The optical sensing system detected absorbance that was linearly correlated to methyl parathion (MP) concentration and the detection limit was estimated to be 4 μM. Sensor hysteresis was investigated and the results showed that at lower concentration range of MP the difference got from the opposite process curves was very small. With its easy immobilization of enzymes and simple design in structure, the system has the potential for development into a practical portable detector for field applications.
Polystyrene/Ag nanoparticles as dynamic surface-enhanced Raman spectroscopy substrates for sensitive detection of organophosphorus pesticides

Abstract:We report the use of Polystyrene/Ag (PS/Ag) nanoparticles as dynamic surface-enhanced Raman spectroscopy (dynamic-SERS) substrates for sensitive detection of low levels of organophosphorus pesticides. The PS particles clearly observed using Raman microscopy provide the masterplate for in situ growth of Ag NPs, leading to multiple active sites for SERS measurements. Besides obtaining the fingerprints of target molecules and recording time-resolved Raman spectra, this dynamic-SERS method can be used as an ultra-sensitive analytical technique which can enhance 1-2 orders of magnitude the signals of analytes in comparison to that of the traditional methods. On the other hand, importantly, it shows much better correlations between concentration and intensity than does the conventional SERS technique so that it can build the foundation for quantitative analysis of analytes. The as-prepared individual PS/Ag nanoparticle has been demonstrated for the sensitive detection of organophosphorus paraoxon and sumithion. SERS spectra are acquired at different concentrations of each pesticide and linear calibration curves are obtained by monitoring the strongest intensity value of bands arising from stronger stretching mode as a function of analyte concentration. The limits of detection and limits of quantitation are reported for two pesticides. The limit of detection for paraoxon is 96 nM (0.026 ppm) and for sumithion is 34 nM (0.011 ppm). The limits of quantitation are 152 nM (0.042 ppm) and 57 nM (0.016 ppm) for paraoxon and sumithion, respectively. It can be seen that these two organophosphorus pesticides can be detected in the low nM range based on this dynamic-SERS analytical method. Also, in the real sample experiments of paraoxon and sumithion, the results confirm that this dynamic-SERS technique would have potential applicability for quantitative analysis with slight interference.

Abstract:In this work, a convenient ratiometric fluorescent platform was designed to measure organophosphorus pesticides (OPs) based on acetylcholinesterase (AChE), acetylthiocholine (ATCh), manganese dioxide nanosheets (MnO 2 ), near-infrared carbon dots (RCDs) and o-phenylenediamine (OPD). In this platform, a direct oxidation of OPD by MnO 2 generated the luminescent product 2,3-diaminophenolazine (DAP) through intrinsic oxidase activity, while RCDs served as a fluorescent reference indicator. In the presence of AChE and ATCh, the enzymatic hydrolysate thiocholine (TCh) would reduce MnO 2 nanosheets to Mn 2+ , leading to the quenching of DAP fluorescence. On the other hand, OPs can inhibit the catabolism of ATCh by AChE thus acting as a recognizer of OPs. According to these reactions, OPs were quantitatively analyzed by the intensity ratio of fluorescence emitted from RCDs and DAP ( F 560 / F 676 ). The constructed platform can detect OPs with the range of 0.2–0.6 μM with a detection limit of 4.3 nM.

Abstract:A nano-silver electrode immobilizing acetylcholinesterase (AChE) for the detection of organophosphorus (OPPs) pesticides is reported. Scanning electron microscopy (SEM) was used to characterize the surface structure of two kinds of electrodes fabricated with different sizes of silver powders and the interface between chitosan layer and nano-silver powder layer. Cyclic voltammetry was carried out to characterize the response of silver/chitosan electrode in the absence and in the presence of thiocholine (TCh). It was also used to evaluate the insulativity of the chitosan layer. An amperometric method was performed to measure the response of the electrode to TCh, which is the product of the enzymatic reaction for detecting organophosphorus pesticides indirectly. Although there are many kinds of nanoparticles, silver was chosen for its internal advantage in detecting TCh at low potential without further modification. The result shows nano-silver powder has better performance than usual silver powder, and the limit of detection of paraoxon is 4 ppb under optimized conditions. One percent (w/v) chitosan solution was used as binder for the immobilization of nano-silver powder and AChE, which made it possible for independent electrode fabrication at room temperature, whereas 3% (w/v) chitosan solution was used as insulating compound for controlling the electrode area. Unlike traditional organic insulating ink, chitosan is safe and environmentally friendly, and it is used as insulating material for the first time. The flexible nano-silver/AChE/chitosan electrode was evaluated in Chinese chives and cabbage, and the recoveries of standard addition were 105.11 and 96.41%, respectively. Owing to the antibacterial property of nano-silver and the biocompatibility, safety, and biodegradability of chitosan, the proposed method is safe, facile, environmentally friendly, and has great potential in organophosphorus pesticide detection for food safety.

Abstract:The recent international scenario highlights the importance to protect human health and environmental quality from toxic compounds. In this context, organophosphorous (OP) Nerve Agents (NAs) have received particular attention, due to their use in terrorist attacks. Classical instrumental detection techniques are sensitive and selective, but they cannot be used in real field due to the high cost, specialized personnel requested and huge size. For these reasons, the development of practical, easy and fast detection methods (smart methods) is the future of this field. Indeed, starting from initial sensing research, based on optical and/or electrical sensors, today the development and use of smart strategies to detect NAs is the current state of the art. This review summarizes the smart strategies to detect NAs, highlighting some important parameters, such as linearity, limit of detection and selectivity. Abstract:A novel single-walled carbon nanotube (SWNT) based biosensor for real-time detection of organophosphate has been developed. Horizontally aligned SWNTs are assembled to desirable electrodes using AC dielectrophoresis technique. Organophosphorus hydrolase (OPH) immobilized on the SWNTs by nonspecific binding triggers enzymatic hydrolysis of organophosphates (OPs), such as paraoxon, consequently causing a detectable change in the conductance of the SWNTs. The conductance change is found to be correlated to the concentration of organophosphate. Our results suggest the novel biosensor has great potential to serve as a simple and reusable platform of sensing organophosphate on a real-time basis.

Abstract:Accurately detect the residues of organophosphate pesticides (OPs) in food and environment is critical to our daily lives. In this study, we developed a novel acetylcholinesterase (AChE) biosensor based on Au–Tb alloy nanospheres (NSs) for rapid and sensitive detection of OPs for the first time. Au–Tb alloy NSs that with good conductivity and biocompatibility were produced with a mild hydrothermal. Under optimal conditions, the AChE biosensor was obtained by a simple assembly process, with a big linear range (10−13–10−7 M) and the limit of detection was 2.51 × 10−14 M for the determination of methyl parathion. Moreover, the determination of methyl parathion with the prepared biosensor presented a high sensitivity, outstanding repeatability and superior stability compared with other reported biosensors. Through the determination of tap water and Yanming lake samples, it was proved that the modified biosensor with satisfactory recoveries (96.76%–108.6%), and are realizable in the determination of OPs in real samples.

Abstract:Nerve agents, including organophosphorus compounds such as paraoxon, are potent and highly toxic chemicals with grave implications for human health and the environment. In this paper, we present the development of a novel enzyme inhibition-based biosensor for the sensitive and selective detection of paraoxon, which is commonly used as a surrogate for nerve agents. The biosensor employs reduced graphene oxide as a screen-printed electrode surface modification nanomaterial, leading to increased surface electroactivity and, thus, more sensitive detection. The biosensor exhibits a low detection limit of 0.56 pg/ml (limit of detection, LOD) and 1.25 pg/ml (limit of quantification, LOQ), highlighting its high sensitivity for trace-level analysis of nerve agents in complex sample matrices. Our biosensor demonstrates remarkable selectivity for paraoxon, with minimal interference from other non-target chemicals. Stability and repeatability tests reveal that the system maintains its performance integrity over a 45-day period and consistently produces readings with a margin of error of only 5%. Real sample testing in river water, wastewater, and tap water further confirms the biosensor's practical utility, with recovery percentages ranging from 84 to 115%. This biosensor represents a significant advancement in biosensor technology, facilitating the rapid, cost-effective, and reliable detection of toxic substances in real-world scenarios.

Abstract:On-site quantitative detection of organophosphorus pesticides (OPs) is crucial for safeguarding food and public safety. This study presents a novel acetylcholinesterase (AChE)-mediated paper-based Au3+-etching of gold nanobipyramids (AuNBPs) system. The system employs a long-term storable AuNBPs-deposited nylon membrane embedded within a portable and temperature-controlled paper-based analytical device. This system, coupled with a colorimeter-based quantitative method, enables the development of a practical paper-based multicolor sensor (PMS) for on-site quantitative detection of three common OPs (paraoxon, dichlorvos, and trichlorfon). In the absence of OPs, AChE hydrolyzes acetylthiocholine to thiocholine, which reduces Au3+ to Au+. The presence of OPs inhibits AChE activity, thereby preserving Au3+ to etch AuNBPs on nylon membranes, accompanied by multicolor changes. These color changes can be simply quantified by measuring the a∗ parameter of the CIELab color space using a portable colorimeter. Under optimal conditions, the PMS displayed eight OPs-corresponding color changes with a minimum detectable concentration of 1.0-10 μg/L (visual observation) and limits of detection of 0.8-7.2 μg/L (colorimeter) and 0.2-3.4 μg/L (UV-vis spectrometry). The PMS successfully determined the OPs in vegetable and rice samples with recoveries of 89.0-109 % and RSDs (n = 5) of <6 %. These results were consistent with those obtained using the HPLC-MS method. The PMS demonstrates excellent portability, AuNBPs stability, detection sensitivity, and reproducibility, making it a promising tool for the on-site quantitative detection of OPs residues in food. Furthermore, the paper-based etching system coupled with the colorimeter-based quantitative method provides a valuable reference to develop practical PMSs for various targets in diverse fields.
